Chronic hepatitis B virus (HBV) infection can significantly increase the incidence of cirrhosis and liver cancer, and there is no curative treatment. The persistence of HBV covalently closed circular DNA (cccDNA) is the major obstacle of antiviral treatments. cccDNA is formed through repairing viral partially double-stranded relaxed circular DNA (rcDNA) by varies host factors. However, the detailed mechanisms are not well characterized. To dissect the biogenesis of cccDNA, we took advantage of an in vitro rcDNA repair system to precipitate host factors interacting with rcDNA and identified co-precipitated proteins by mass spectrometry. Results revealed the MRE11-RAD50-NBS1 (MRN) complex as a potential factor. Transiently or stably knockdown of MRE11, RAD50 or NBS1 in hepatocytes before HBV infection significantly decreased viral markers, including cccDNA, while reconstitution reversed the effect. Chromatin immunoprecipitation assay further validated the interaction of MRN complex and HBV DNA. However, MRN knockdown after HBV infection showed no effect on viral replication, which indicated that MRN complex inhibited the formation of cccDNA without affecting its stability or transcriptional activity. Interestingly, Mirin, a MRN complex inhibitor which can inhibit the exonuclease activity of MRE11 and MRN-dependent activation of ATM, but not ATM kinase inhibitor KU55933, could decrease cccDNA level. Likewise, the MRE11 endonuclease activity inhibitor PFM01 treatment decreased cccDNA. MRE11 nuclease assays indicated that rcDNA is a substrate of MRE11. Furthermore, the inhibition of ATR-CHK1 pathway, which is known to be involved in cccDNA formation, impaired the effect of MRN complex on cccDNA. Similarly, inhibition of MRE11 endonuclease activity mitigated the effect of ATR-CHK1 pathway on cccDNA. These findings indicate that MRN complex cooperates with ATR-CHK1 pathway to regulate the formation of HBV cccDNA. In summary, we identified host factors, specifically the MRN complex, regulating cccDNA formation during HBV infection. These findings provide insights into how HBV hijacks host enzymes to establish chronic infection and reveal new therapeutic opportunities.

The photosystem II (PSII) Subunit P (PsbP) protein is a component of its oxygen-evolving complex, which can oxidize water to produce oxygen using light energy and is critical to the core components and stability of PSII. Using the whole-genome information, the PsbP genes of 10 plant species were comprehensively identified. The expression patterns of wheat PsbPs under Blumeria graminis f. sp. tritici (Bgt) infection were assessed using qRT-PCR, and the functions of TaPsbPs in wheat powdery mildew resistance were studied using barley stripe mosaic virus-induced gene silencing. In total, 122 PsbP genes were divided into 8 classes with similar gene structures. No tandem repeat events were identified in wheat PsbP, suggesting that the PsbP genes in common wheat were donated by its diploid progenitor species. The expression levels of TaPsbP2A-1, TaPsbP3A-1, TaPsbP4A-1, TaPsbP4A-2, and TaPsbP7A-2 were induced by Bgt. The silencing of TaPsbP4A-1 increased the resistance of common wheat ‘Bainong AK58’ to Bgt. This study provides valuable information for functional and evolutionary research on the PsbP gene family.

Hepatocellular carcinoma (HCC) is the third leading cause of cancer-related deaths worldwide and presents a significant threat to human health. Despite its prevalence, the underlying regulatory mechanisms of HCC remain unclear. In this study, we integrated RNA-seq datasets, proteome dataset and survival analysis and unveiled Stratifin (SFN) as a potential prognostic biomarker for HCC. SFN knockdown inhibited HCC progression in cell cultures and mouse models. Conversely, ectopic expression of Sfn in primary mouse HCC model accelerated HCC progression. Mechanistically, SFN acted as an adaptor protein, activating AKT1 signaling by fostering the interaction between PDK1 and AKT1, with the R56 and R129 sites on SFN proving to be crucial for this binding. In the syngeneic implantation model, the R56A/R129A mutant of SFN inhibited Akt signaling activation and impeded HCC growth. Additionally, peptide inhibitors designed based on the binding motif of AKT1 to SFN significantly inhibited HCC progression. In summary, our findings establish that SFN promotes HCC progression by activating AKT signaling through the R56 and R129 binding sites. This discovery opens new avenues for a promising therapeutic strategy for the treatment of HCC.

Abstract Background LATS1/2 are frequently mutated and down-regulated in endometrial cancer (EC), but the contributions of LATS1/2 in EC progression remains unclear. Impaired antigen presentation due to mutations or downregulation of the major histocompatibility complex class I (MHC-I) has been implicated in tumor immune evasion. Herein, we elucidate the oncogenic role that dysregulation of LATS1/2 in EC leads to immune evasion through the down-regulation of MHC-I. Methods The mutation and expression as well as the clinical significance of LATS1/2 in EC was assessed in the TCGA cohort and our sample cohort. CRISPR-Cas9 was used to construct knockout cell lines of LATS1/2 in EC. Differentially expressed genes were analyzed by RNA-seq. The interaction between LATS1/2 and STAT1 was verified using co-immunoprecipitation and GST pull-down assays. Mass spectrometry, in vitro kinase assays, ChIP-qPCR, flow cytometry, immunohistochemistry, immunofluorescence and confocal microscopy were performed to investigate the regulation of LATS1/2 on MHC-I through interaction with and phosphorylate STAT1. The killing effect of activated PBMCs on EC cells were used to monitor anti-tumor activity. Results Here, we demonstrate that LATS1/2 are frequently mutated and down-regulated in EC. Moreover, LATS1/2 loss was found to be associated with a significant down-regulation of MHC-I, independently of the Hippo-YAP pathway. Instead, LATS1/2 were found to directly interact with and phosphorylate STAT1 at Ser727, a crucial transcription factor for MHC-I upregulation in response to interferon-gamma (IFN-γ) signaling, to promote STAT1 accumulating and moving into the nucleus to enhance the transcriptional activation of IRF1/NLRC5 on MHC-I. Additionally, the loss of LATS1/2 was observed to confer increased resistance of EC cells to immune cell-mediated killing and this resistance could be reversed by over-expression of MHC-I. Conclusion Our findings indicate that dysregulation of LATS1/2 in EC leads to immune evasion through the down-regulation of MHC-I, leading to the suppression of infiltrating activated CD8 + T cells and highlight the importance of LATS1/2 in IFN-γ signaling-mediated tumor immune response, suggesting that LATS1/2 is a promising target for immune checkpoint blockade therapy in EC.

Abstract Accurate determination of human tumor malignancy is important for choosing efficient and safe therapies. Bioimaging technologies based on luminescent molecules are widely used to localize and distinguish active tumor cells. Here, we report a human cancer grade probing system (GPS) using a water-soluble and structure-changeable Eu(III) complex for the continuous detection of early human brain tumors of different malignancy grades. Time-dependent emission spectra of the Eu(III) complexes in various types of tumor cells were recorded. The radiative rate constants (k r), which depend on the geometry of the Eu(III) complex, were calculated from the emission spectra. The tendency of the k r values to vary depended on the tumor cells at different malignancy grades. Between T = 0 and T = 3 h of invasion, the k r values exhibited an increase of 4% in NHA/TS (benign grade II gliomas), 7% in NHA/TSR (malignant grade III gliomas), and 27% in NHA/TSRA (malignant grade IV gliomas). Tumor cells with high-grade malignancy exhibited a rapid upward trend in k r values. The cancer GPS employs Eu(III) emissions to provide a new diagnostic method for determining human brain tumor malignancy.

Abstract Background There are many predictions about the progression of natural collapse course of osteonecrosis of the femoral head. Here, we aimed to combine the three classical prediction methods to explore the progression of the natural collapse course. Methods This retrospective study included 127 patients admitted to our hospital from October 2016 to October 2017, in whom the femoral head had not collapsed. Logistic regression analysis was performed to determine the collapse risk factors, and Kaplan–Meier survival curves were used for femoral head survival analysis. The collapse rate of the femoral head was recorded within 5 years based on the matrix model. The specificity of the matrix model was analyzed using the receiver operating characteristic curve. Results A total of 127 patients with a total of 202 hips were included in this study, and 98 hips collapsed during the follow-up period. Abstract Objective This aimed to evaluate the status of return to work (RTW) in patients with osteonecrosis of the femoral head (ONFH) after total hip arthroplasty (THA). Methods The baseline characteristics of all patients in this retrospective study were obtained from the hospital patient database. The relevant changes in patients' working conditions, as well as the numerical rating scale (NRS), Harris Hip Score (HHS), self-assessment of work ability, and Likert scale satisfaction assessment were obtained through video call follow-ups. Results 118 patients (response rate: 83%) were ultimately included in this study. The average length of time for the patients to stop working preoperatively was 20.7 weeks. Ninety-four patients (24 women and 70 men) who underwent THA had RTW status, with a mean RTW time of 21.0 weeks. Men had a significantly higher proportion of final RTW and a significantly faster RTW than women. Significant differences in smoking, drinking, cardiovascular diseases, changes in working levels, variations in the types of physical work, changes in working hours, and pain symptoms were observed between the RTW and Non-RTW populations. The patients with a positive RTW status had higher postoperative HHS scores, lower postoperative NRS scores, and higher self-assessment of work ability than patients who had a negative RTW status. Conclusion Ultimately, 80% of patients achieved RTW status. Drinking, sex, change in working level, variation in the type of physical work, change in working hours, post-surgery HHS score and self-assessment of work ability can serve as predictive factors for RTW.

In last decade, a lot of road detection algorithms are researched in remote sensing and aerial images, but they are not valuable for very dark and vague road images, hence, a new algorithm for road detection in a vaguer image is studied in this paper. It firstly uses a Gaussian filter to enhance the original image; then, an new edge detection algorithm is utilized to result in a gradient magnitude image by using different orientation information; subsequently, the road segments in the magnitude image is detected by applying a Watershed based algorithm, and a binary image is obtained; and last, in the binary image, the roads are finalized by removing noise in the image and filling gaps between the segments. To validate the proposed algorithm, the testing images were selected from both public dataset and our own dataset, and the testing results show that the new algorithm results are better than several traditional algorithms and recent semantic methods for the road detection, and the road detection accuracy between 92% and 95%. It is satisfactory for the road detection not only for normal images and also for the dark and vague road images. The new algorithm can be also applied into other applications, such as river detection and image enhancement, etc.

Abstract Our objective was to provide a method for selecting reference beam model and evaluating the dosimetric accuracy of volumetric modulated arc therapy (VMAT) plans delivered on three Elekta beam-matched linacs during radiation oncology. Beam data was measured on three beam-matched linacs including Synergy1, Synergy2 and VersaHD. For eighteen lung and esophagus cases, fifty-four plans were generated using VMAT technique with three linac beam models respectively for point dose measurement and three-dimensional dose measurement. Each VMAT plan was executed sequentially on three linacs respectively. Measurement results were compared with treatment planning system (TPS) calculation results for all VMAT plans. Among three beam-matched linacs, discrepancy in beam output factor, percentage depth dose at 5 cm, 10 cm, 20 cm depth and MLC leaf offset are all within 1% except 20 × 20 cm2 and 30 × 30 cm2 field sizes, and discrepancy in beam profile is all within 2%. With comparison between measurement result and TPS calculation result, the absolute dose deviations are within the range of ± 3%, and the gamma passing rates are all over 95% for all VMAT plans, which are within the tolerance of clinical acceptability. Compared with all plans delivered on Synegy1 and VersaHD, the point dose discrepancy between measured results and TPS calculated results for plans delivered on Synergy2 is smallest, and the gamma passing rate between measured results and TPS calculated results for plans delivered on Synergy2 is highest. The beam-matched linacs demonstrate good agreement between measurement result and TPS calculation result for VMAT plans. The method can be used for selecting reference beam model for VMAT plans.

Hepatitis B virus (HBV) chronically infects 296 million people worldwide, posing a major global health threat. Export of HBV RNAs from the nucleus to the cytoplasm is indispensable for viral protein translation and genome replication, however the mechanisms regulating this critical process remain largely elusive. Here, we identify a key host factor embryonic lethal, abnormal vision, Drosophila-like 1 (ELAVL1) that binds HBV RNAs and controls their nuclear export. Using an unbiased quantitative proteomics screen, we demonstrate direct binding of ELAVL1 to the HBV pregenomic RNA (pgRNA). ELAVL1 knockdown inhibits HBV RNAs posttranscriptional regulation and suppresses viral replication. Further mechanistic studies reveal ELAVL1 recruits the nuclear export receptor CRM1 through ANP32A and ANP32B to transport HBV RNAs to the cytoplasm via specific AU-rich elements, which can be targeted by a compound CMLD-2. Moreover, ELAVL1 protects HBV RNAs from DIS3+RRP6+ RNA exosome mediated nuclear RNA degradation. Notably, we find HBV core protein is dispensable for HBV RNA-CRM1 interaction and nuclear export. Our results unveil ELAVL1 as a crucial host factor that regulates HBV RNAs stability and trafficking. By orchestrating viral RNA nuclear export, ELAVL1 is indispensable for the HBV life cycle. Our study highlights a virus-host interaction that may be exploited as a new therapeutic target against chronic hepatitis B.

This study investigates the enhancement of lateral damage effects after armor penetration by utilizing grooved shaped-charge liners. Traditional methods often fail to achieve penetration and damage comparable to copper warheads when engaging distant targets or targets with interlayer materials. By modifying the liner structure, the jet can be redirected off-axis after penetrating the target, improving the overall damage effectiveness. Smooth Particle Hydrodynamics (SPH) simulations were employed to model the fragment cloud damage effects of high-velocity metal jets from internally grooved liners, and comparisons were made with nongrooved liners. The results show that the fragment clouds generated by grooved liners exhibit greater dispersion angles and velocities than those from nongrooved liners, with groove spacing positively correlated with the divergence angle. SPH simulations align with experimental data, demonstrating that groove spacing and depth significantly influence the primary and secondary perforation areas. Overall, the grooved liner design significantly enhances lateral damage effects, and the consistency between experimental and simulation data further validates its effectiveness. The findings provide new insights and references for improving the postpenetration damage design of shaped-charge warheads.

Abstract To detect lanes at night, each detecting image is the fusion of the multiple images in a video sequence. The valid lane line detection region is identified on region merging. Then, the image preprocessing algorithm based on the Fragi algorithm and Hessian matrix is applied to enhance lanes; to extract the lane line center feature points, the image segmentation algorithm based on Fractional differential is proposed; and according to the possible lane line positions, the algorithm detects the centerline points in four directions. Subsequently, the candidate points are determined, and the recursive Hough transformation is applied to obtain the possible lane lines. Finally, to obtain the final lane lines, we assume that one lane line should have an angle between 25 and 65 degrees, while the other should have an angle between 115 and 155 degrees, if the detected line is not in the regions, the Hough line detection will be continued by increasing the threshold value until the two lane lines are got. By testing more than 500 images and comparing deep learning methods and image segmentation algorithms, the lane detection accuracy by the new algorithm is up to 70%.

Sodium ion batteries (SIBs) have been widely studied because of their low cost, low standard redox potential, and abundant sodium availability. However, the structural rupture during the Na+ insertion/extraction processes and the poor conductivity of the anode material limit its cycling stability and rate capability. Herein, SnSe@C was prepared by high-temperature annealing with dopamine hydrochloride as the carbon source, while SnSe was prepared by a protein reduction method. The carbon layer not only works as a protective layer to limit the volume expansion of SnSe and reduce the dissolution of Na2Se and poly-selenides generated during the discharge process in the electrolyte, but also as a conductive matrix to expedite electron transfer, thereby boosting the cycling stability and rate capability of SnSe@C. Benefiting from the above advantages, SnSe@C exhibits a specific capacity of 211.3 mAh g−1 at 0.1 A g−1 after 110 cycles and outstanging rate capability (210.1 mAh g−1 at 5.0 A g−1 and capacity retention rate of 63.2% from 0.1 to 1.0 A g−1). This study not only proposes an idea for promoting the cycling stability and rate capability of SnSe, but also paves the way for providing anodic materials with a stable structure for SIBs.

Si-based thermoelectric (TE) materials are exhibiting remarkable perspectives in self-energized applications with their special advantages. However, the relatively high total thermal conductivity (κ) prevents their TE enhancement. Here, a strategy of co-compositing dual oxides was implemented for enhancing the TE properties of p-type Si80Ge20 bulks. Composited Ga2O3 was demonstrated to enhance the power factor (PF) due to the crystallization-induced effect of produced Ga by decomposition on SiGe matrix. Associating with compositing SiO2 aerogel (a-SiO2) powder, not only introduced the fine amorphous inclusions and decreased the grain size of host matrix, but also various nano morphologies were formed, i.e., nano inclusions, precipitations, twin boundaries (TBs), and faults. Combining with the eutectic Ge, hierarchical scattering centers impeded the phonon transport comprehensively (decreasing the phonon group velocity (va) and relaxation time) for reducing the lattice-induced thermal conductivity (κl). As a result, a minimum κ of 2.38 W·m−1·K−1 was achieved, which is significantly dropped by 32.6% in contrast with that of the pristine counterpart. Ultimately, a maximal dimensionless figure of merit (ZT) of 0.9 was achieved at 600 ℃, which is better than those of most corresponding oxide-composited Si-based bulks.

Cracks are a major sign of aging transportation infrastructure. The detection and repair of cracks is the key to ensuring the overall safety of the transportation infrastructure. In recent years, due to the remarkable success of deep learning (DL) in the field of crack detection, many researches have been devoted to developing pixel-level crack image segmentation (CIS) models based on DL to improve crack detection accuracy, but as far as we know there is no review of DL-based CIS methods yet. To address this gap, we present a comprehensive thematic survey of DL-based CIS techniques. Our review offers several contributions to the CIS area. First, more than 40 papers of journal or top conference most published in the last three years are identified and collected based on the systematic literature review method. Second, according to the backbone network architecture of the models proposed in them, they are grouped into 10 topics: FCN, U-Net, encoder-decoder model, multi-scale, attention mechanism, transformer, two-stage detection, multi-modal fusion, unsupervised learning and weakly supervised learning, to be reviewed. Meanwhile, our survey focuses on discussing strengths and limitations of the models in each topic so as to reveal the latest research progress in the CIS field. Third, publicly accessible data sets, evaluation metrics, and loss functions that can be used for pixel-level crack detection are systematically introduced and summarized to facilitate researchers to select suitable components according to their own research tasks. Finally, we discuss six common problems and existing solutions to them in the field of DL-based CIS, and then suggest eight possible future research directions in this field.

Intelligent vehicle trajectory tracking exhibits problems such as low adaptability, low tracking accuracy, and poor robustness in complex driving environments with uncertain road conditions. Therefore, an improved method of adaptive model predictive control (AMPC) for trajectory tracking was designed in this study to increase the corresponding tracking accuracy and driving stability of intelligent vehicles under uncertain and complex working conditions. First, based on the unscented Kalman filter, longitudinal speed, yaw speed, and lateral acceleration were considered as the observed variables of the measurement equation to estimate the lateral force of the front and rear tires accurately in real time. Subsequently, an adaptive correction estimation strategy for tire cornering stiffness was designed, an AMPC method was established, and a dynamic prediction time-domain adaptive model was constructed for optimization according to vehicle speed and road adhesion conditions. The improved AMPC method for trajectory tracking was then realized. Finally, the control effectiveness and trajectory tracking accuracy of the proposed AMPC technique were verified via co-simulation using CarSim and MATLAB/Simulink. From the results, a low lateral position error and heading angle error in trajectory tracking were obtained under different vehicle driving conditions and road adhesion conditions, producing high trajectory-tracking control accuracy. Thus, this work provides an important reference for improving the adaptability, robustness, and optimization of intelligent vehicle tracking control systems.

Hepatitis B virus (HBV) chronically infects 296 million individuals and there is no cure. As an important step of viral life cycle, the mechanisms of HBV egress remain poorly elucidated. With proteomic approach to identify capsid protein (HBc) associated host factors and siRNA screen, we uncovered tumor susceptibility gene 101 (TSG101). Knockdown of TSG101 in HBV-producing cells, HBV-infected cells and HBV transgenic mice suppressed HBV release. Co-immunoprecipitation and site mutagenesis revealed that VFND motif in TSG101 and Lys-96 ubiquitination in HBc were essential for TSG101-HBc interaction. In vitro ubiquitination experiment demonstrated that UbcH6 and NEDD4 were potential E2 ubiquitin-conjugating enzyme and E3 ligase that catalyzed HBc ubiquitination, respectively. PPAY motif in HBc and Cys-867 in NEDD4 were required for HBc ubiquitination, TSG101-HBc interaction and HBV egress. Transmission electron microscopy confirmed that TSG101 or NEDD4 knockdown reduces HBV particles count in multivesicular bodies (MVBs). Our work indicates that TSG101 recognition for NEDD4 ubiquitylated HBc is critical for MVBs mediated HBV egress.

IntroductionParkinson’s disease (PD) is the second most prevalent neurodegenerative disorder. However, effective preventative or therapeutic agents for PD remain largely limited. Marigold Calendula officinalis L. (CoL) has been reported to possess a wide range of biological activities, but its neuroprotective activity including anti-neurodegenerative diseases is unclear. Here, we aim to investigate whether the extract of CoL (ECoL) has therapeutic activity on PD.MethodsWe identified the chemical composition of flavonoid, an important active ingredient in ECoL, by a targeted HPLC-Q-TOF-MS analysis. Subsequently, we evaluated the anti-PD effect of ECoL by using zebrafish PD model induced by 1-methyl-4-phenyl-1-1,2,3,6-tetrahydropyridine (MPTP). After ECoL+MPTP co-treatments, the changes of dopaminergic neurons, neural vasculature, nervous system, and locomotor activity were examined, respectively. The expressions of genes related to neurodevelopment and autophagy were detected by RT-qPCR. Further, the interaction between autophagy regulators and ECoL flavonoids was predicted using molecular docking method.ResultsAs a result, 5 kinds of flavonoid were identified in ECoL, consisting of 121 flavones and flavonols, 32 flavanones, 22 isoflavonoids, 11 chalcones and dihydrochalcones, and 17 anthocyanins. ECoL significantly ameliorated the loss of dopaminergic neurons and neural vasculature, restored the injury of nervous system, and remarkably reversed the abnormal expressions of neurodevelopment-related genes. Besides, ECoL notably inhibited the locomotor impairment in MPTP-induced PD-like zebrafish. The underlying anti-PD effect of ECoL may be implicated in activating autophagy, as ECoL significantly upregulated the expressions of genes related to autophagy, which contributes to the degradation of α-synuclein aggregation and dysfunctional mitochondria. Molecular docking simulation showed the stable interaction between autophagy regulators (Pink, Ulk2, Atg7, and Lc3b) and 10 main compounds of flavonoid in ECoL, further affirming the involvement of autophagy activation by ECoL in anti-PD action.ConclusionOur results suggested that ECoL has the anti-PD effect, and ECoL might be a promising therapeutic candidate for PD treatment.

Objective. This study is aimed at discussing the value of RNA interference technology on inhibiting lacrimal gland fibrosis in IgG4-related ocular disease (IgG4-ROD). Methods. Six patients with IgG4-ROD who came to the hospital for surgical treatment from October 2018 to August 2019 were selected, and their diseased lacrimal glands were taken for primary cell culture and fibroblast identification. High efficiency and specificity small interference RNA (siRNA) plasmid vector was constructed, its inhibitory effect on fibroblast proliferation was determined by CCK-8 assay, and the appropriate concentration was selected as the siRNA concentration for subsequent experiments. RT-PCR and Western blot detected the relative expression levels of Fibulin-5 mRNA and protein in the cells 48 hours after transfection. The apoptosis rate of each group of cells at 24 hours, 48 hours, and 72 hours after transfection was detected by flow cytometry, and the proliferation and apoptosis of cells after silencing Fibulin-5 were analyzed and compared. Results. 24 hours after transfection, there was no significant difference in the proliferation rate among the four groups (P>0.05); 48 hours and 72 hours after Fibulin-5 siRNA transfection, the proliferation activity of the transfected cells was significantly decreased compared with the 0 nM group, and the inhibitory effect of 75 nM siRNA was the strongest. Abstract Foveated near-eye display is one of the most promising approaches to deliver immersive experience of mixed reality. However, it is challenged to conceive a compact optical system. Here, we introduce a method to use polarization optics via liquid crystal photonics to improve the foveated display performance. We demonstrate a benchtop prototype of this idea. We implement and combine two display modules for peripheral and foveal visions. A peripheral display consists of a polarization selective lens (PSL) module, a polarization selective diffuser (PSD), and a slanted projection system. An 80 $$^\circ$$ ∘ diagonal field of view is achieved by on-axis optical configuration of the PSL module and the PSD. A foveal holographic display is composed of a spatial light modulator (SLM), a volume grating lens, and a microelectromechanical system mirror possibly in combination with a switchable polarization selective grating module. The holographic reconstruction using the SLM enables accurate focus cue generation and high resolution above 30 cycles per degree within 15 $$^\circ$$ ∘ by 15 $$^\circ$$ ∘ field of view. We explore and discuss the liquid crystal photonics in the prototype that has a novel optical design using volume gratings with polarization selectivity.

Abstract Background Sugar-feeding provides energy for mosquitoes. Facilitated glucose transporters (GLUTs) are responsible for the uptake of glucose in animals. However, knowledge of GLUTs function in Anopheles spp. is limited. Methods Phylogenetic analysis of GLUTs in Anopheles stephensi was performed by the maximum likelihood and Bayesian inference methods. The spatial and temporal expression patterns of four Asteglut genes were analyzed by qPCR. The function of Asteglut1 was examined using a dsRNA-mediated RNA interference method. Transcriptome analysis was used to investigate the global influence of Asteglut1 on mosquito physiology. Results We identified 4 glut genes, Asteglut1, Asteglutx, Asteglut3 and Asteglut4 in An. stephensi. Asteglut1, Asteglut3 and Asteglut4 were mainly expressed in the midgut. Plasmodium berghei infection differentially regulated the expression of Asteglut genes with significant downregulation of Asteglut1 and Asteglut4, while upregulation of Asteglutx. Only knocking-down Asteglut1 facilitated Plasmodium berghei infection in An. stephensi. This might be due to the accumulation of glucose prior to blood-feeding in dsAsteglut1-treated mosquitoes. Our transcriptome analysis revealed that knockdown of Asteglut1 differentially regulated expression of genes associated with multiple functional clusters, especially those related to detoxification and immunity. The dysregulation of multiple pathways might contribute to the increased P. berghei infection. Conclusions Our study shows that Asteglut1 participates in defense against P. berghei in An. stephensi. The regulation of Asteglut1 on vector competence might through modulating multiple biological processes, such as detoxification and immunity.

Aggregate measurement and analysis are critical for civil engineering. Multiple entropy thresholding (MET) is inefficient, and the accuracy of related optimization strategies is unsatisfactory, which results in the segmented aggregate images lacking many surface roughness and aggregate edge features. Thus, this research proposes an autonomous segmentation model (i.e., PERSSA-MET) that optimizes MET based on the chaotic combination strategy sparrow search algorithm (SSA). First, aiming at the characteristics of the many extreme values of an aggregate image, a novel expansion parameter and range-control elite mutation strategies were studied and combined with piecewise mapping, named PERSSA, to improve the SSA’s accuracy. This was compared with seven optimization algorithms using benchmark function experiments and a Wilcoxon rank-sum test, and the PERSSA’s superiority was proved with the tests. Then, PERSSA was utilized to swiftly determine MET thresholds, and the METs were the Renyi entropy, symmetric cross entropy, and Kapur entropy. In the segmentation experiments of the aggregate images, it was proven that PERSSA-MET effectively segmented more details. Compared with SSA-MET, it achieved 28.90%, 12.55%, and 6.00% improvements in the peak signal-to-noise ratio (PSNR), the structural similarity (SSIM), and the feature similarity (FSIM). Finally, a new parameter, overall merit weight proportion (OMWP), is suggested to calculate this segmentation method’s superiority over all other algorithms. The results show that PERSSA-Renyi entropy outperforms well, and it can effectively keep the aggregate surface texture features and attain a balance between accuracy and speed.

Abstract Previous studies have shown that air pollutants have a significant impact on cardiovascular and cerebrovascular diseases (CCD), but these studies have focused more on provincial capitals or large cities. This study was the first time to explore the effects of air pollutants on CCD in Luoyang. In this study, the generalized additive model (GAM) was used to establish a single‐pollutant model, a multi‐pollutant model and stratified modes of age, sex and season to evaluate the effects of PM2.5, PM10, SO2, NO2, CO and O3 on outpatient visits for CCD within a week. The results of the single‐pollutant model showed that PM2.5, PM10, SO2, NO2, CO and O3 all had significant effects on the outpatient visits for CCD with a lag effect. The multi‐pollutant model showed that there might be complex interactions among pollutants. The results of the stratified model showed that there was no significant difference in the effects of different pollutants on different genders and ages, and the results of seasonal stratification showed that PM2.5, PM10, SO2 and CO had a greater influence on the outpatient visits for CCD in spring and summer, while NO2 and O3 had a greater influence on the outpatient visits for CCD in winter and summer, respectively. The results showed that air pollutants significantly affected outpatient visits for CCD, among which NO2 had the greatest influence, and seasonal effects and the combined effects of various pollutants should be considered in strategies for the prevention of CCD.

The extraction of pavement cracks is always a hard task in image processing. In airport and road construction, cracking is the main factor for pavement damage, which can decrease the quality of pavement and affect transportation seriously. Cracks also exist in other artificial or natural objects, such as buildings, bridges, tunnels, etc. Among all the object images, pavement crack images are the most complex, so the image processing and analysis for them is harder than other crack images. From the early image acquisition based on photography technology to the current 3D laser scanning technology, the pavement crack image acquisition technology is becoming more convenient and efficient, but there are still challenges in the automatic processing and recognition of cracks in images. From the early global thresholding to deep learning algorithms, the research for crack extraction has been developed for about 40 years. There are many methods and algorithms that are satisfactory in pavement crack applications, but there is no standard until today. Therefore, in order to know the developing history and the advanced research, we have collected a number of literature in this research topic for summarizing the research artwork status, and giving a review of the pavement crack image acquisition methods and 2D crack extraction algorithms. Also, for image acquisition methods and pavement crack image segmentation, more detailed comparison and discussions are made. Summary: Plant-nectar-derived sugar is the major energy source for mosquitoes, but its influence on vector competence for malaria parasites remains unclear. Here, we show that Plasmodium berghei infection of Anopheles stephensi results in global metabolome changes, with the most significant impact on glucose metabolism. Feeding on glucose or trehalose (the main hemolymph sugars) renders the mosquito more susceptible to Plasmodium infection by alkalizing the mosquito midgut. The glucose/trehalose diets promote proliferation of a commensal bacterium, Asaia bogorensis, that remodels glucose metabolism in a way that increases midgut pH, thereby promoting Plasmodium gametogenesis. We also demonstrate that the sugar composition from different natural plant nectars influences A. bogorensis growth, resulting in a greater permissiveness to Plasmodium. Altogether, our results demonstrate that dietary glucose is an important determinant of mosquito vector competency for Plasmodium, further highlighting a key role for mosquito-microbiota interactions in regulating the development of the malaria parasite.

Objective: Severe acute pancreatitis (SAP) can lead to acute lung injury (ALI). This study investigated the therapeutic effect of emodin and its molecular mechanisms in a rat model of SAP-ALI.Methods: Forty male Sprague-Dawley rats were randomly divided into the groups: Control (CON), SAP (SAP), emodin (EMO), and C23 (C23). The latter three groups of rats were induced for SAP-ALI by retrograde injection of 5% sodium taurocholate into the biliary-pancreatic duct and were treated with vehicle, emodin or C23, respectively. One day post induction, their pancreatic and lung injury was assessed by histology and arterial blood gas analysis. In vitro, rat alveolar macrophages (NR8383 cells) were treated with recombinant rat CIRP in the presence or absence of TAK242 (a TLR4 inhibitor), C23 or emodin. The CIRP-mediated activation of the NLRP3/IL-1β/CXCL1 signaling in rat lungs and NR8383 cells was determined. Similarly, the role of IL-1β in the CIRP-induced CXCL1 expression was investigated.Results: Emodin treatment significantly reduced inflammation and tissue damages in the pancreatic and lung tissues in rats with SAP-ALI, accompanied by decreasing serum amylase, CIRP and IL-1β levels and improving lung function. Furthermore, emodin significantly mitigated the SAP-up-regulated CIRP expression in the pancreatic islets and lung tissues, and attenuated the SAP-activated NF-κB signaling, NLRP3 inflammasome formation and CXCL1 expression in lung resident macrophages as well as neutrophil infiltration in the lungs of rats. In addition, treatment with CIRP significantly activated the NF-κB signaling and NLRP3 inflammasome formation and induced IL-1β and CXCL1 expression and pyroptosis in NR8383 cells, which were abrogated by TAK242 and significantly mitigated by C23 or emodin. Moreover, CIRP only induced very lower levels of CXCL1 expression in IL-1β-silencing NR8383 cells and treatment with IL-1β induced CXCL1 expression in NR8383 cells in a dose and time-dependent manner.Conclusion: Emodin may inhibit the CIRP-activated NLRP3/IL-1β/CXCL1signaling to decrease neutrophil infiltration and ameliorate the SAP-ALI in rats.

Nano lanthanum oxide particles (La2O3 NPs) are important nanoparticle materials which are widely used in photoelectric production, but their potential health hazards to the respiratory system are not clear. The purpose of this study was to explore the possible mechanism of lung injury induced by La2O3 NPs. In this study, 40 SPF male SD rats were randomly divided into low-, medium-, and high-dose groups and control groups, with 10 animals in each group. Rats were poisoned by tracheal injection. The low-, medium-, and high-dose groups were given La2O3 NPs suspension of 25, 50, and 100 mg/kg, respectively, and the control group was given an equal volume of high-temperature sterilized ultrapure water. The rats in each group were exposed once a week for 12 consecutive times. The gene transcription and protein expression levels of PINK1 and parkin in rat lung tissue were mainly detected. Compared with the control group, the gene transcription and protein expression levels of PINK1 and Parkin in the exposed group were significantly higher (p < 0.05). La2O3 NPs may activate PINK1/parkin-induced mitochondrial autophagy.

Two modal decomposition techniques, including proper orthogonal decomposition (POD) and dynamic mode decomposition (DMD), were used to identify the wake patterns past single and two crossing cylinders in 60° and 90° arrangements with gap ratio G = 4. The flow was simulated using direct numerical simulations (DNS) for Reynolds numbers Re = 100. From modal analysis, the flow’s spatial scale decreased with increasing modal frequency. Two main modes were identified in the wake of the cylinders, namely spatially antisymmetric and symmetric modes. Antisymmetric and symmetric modes were related to the cylinders’ vortex shedding and shedding vortices’ shift motion, respectively, whose frequencies were odd and even multiples of the cylinders’ lift force frequency. In addition, a low-frequency mode concerning the shadowing effect of the downstream cylinder (DC) in 90° arrangement was found in the wake of the DC centre.

Disclination lines are topological defects in molecular orientation widely found in liquid crystals. Here Wang et al. use a surface patterning technique to produce a very stable freestanding 3D array of ½ twist disclinations, which could be exploited in a variety of nanometre scale applications.

Peptidoglycan recognition proteins (PGRPs) and commensal microbes mediate pathogen infection outcomes in insect disease vectors. Although PGRP-LD is retained in multiple vectors, its role in host defense remains elusive. Here we report that Anopheles stephensi PGRP-LD protects the vector from malaria parasite infection by regulating gut homeostasis. Specifically, knock down of PGRP-LD (dsLD) increased susceptibility to Plasmodium berghei infection, decreased the abundance of gut microbiota and changed their spatial distribution. This outcome resulted from a change in the structural integrity of the peritrophic matrix (PM), which is a chitinous and proteinaceous barrier that lines the midgut lumen. Reduction of microbiota in dsLD mosquitoes due to the upregulation of immune effectors led to dysregulation of PM genes and PM fragmentation. Elimination of gut microbiota in antibiotic treated mosquitoes (Abx) led to PM loss and increased vectorial competence. Recolonization of Abx mosquitoes with indigenous Enterobacter sp. restored PM integrity and decreased mosquito vectorial capacity. Silencing PGRP-LD in mosquitoes without PM didn't influence their vector competence. Our results indicate that PGPR-LD protects the gut microbiota by preventing hyper-immunity, which in turn promotes PM structurally integrity. The intact PM plays a key role in limiting P. berghei infection.
