IntroductionChemical fertilizer reduction combined with organic fertilizer (organic fertilizer substitution) has a positive impact on crop productivity and sustainable development. However, the effects of short-term organic fertilizer substitution on microbial community structure and functions of sorghum rhizosphere soil and on sorghum yield remain unclear. Herein, this study investigated the short-term effects of organic fertilizer substitution on sorghum soil physicochemical properties, microbial community structure and enzyme activities using Metagenomic sequencing technology.MethodsThe fertilization treatment included no fertilization (CK), pure chemical fertilizer N (CF), substitution of 25% chemical fertilizer N with organic N (NF25), substitution of 50% chemical fertilizer N with organic N (NF50), substitution of 75% chemical fertilizer N with organic N (NF75), and pure organic fertilizer N (NF100); soil samples were collected and analyzed in the flowering period of sorghum.Results and DiscussionThe results showed that the suitable organic fertilizer substitution rate of sorghum field was 50%, and its yield was the highest among all treatments (9789.9 kg/hm2). Compared with the CF treatment, a medium ratio (50%) of organic fertilizer substitution significantly reduced soil alkalization (by 3.05%), improved soil nutrients, enhanced soil enzyme activities, and increased sorghum yield (P < 0.05). After organic fertilizer substitution treatment, higher protein, fat, and total starch levels accumulated in sorghum grains, and the tannin content of grains decreased. The effect of organic fertilizer substitution on bacterial diversity was greater than that on fungal diversity. Among the dominant bacterial phyla, the medium ratio of organic substitution treatment significantly increased the relative abundances of Proteobacteria (by 3.57%) and Actinomycetes (by 14.94%), and decreased the relative abundances of Acidobacteria (by 5.18%) and Planctomycetes (by 7.76%) compared with no fertilization, while the dominant fungal phyla did not respond significantly to the addition of organic fertilizer. Organic fertilizer substitution also improved soil microbial metabolic pathways, biosynthesis of secondary metabolites, and carbon metabolism. The biomarkers enriched in inorganic fertilizer treatment and organic fertilizer substitution treatments had similar relevant environmental elements but reversed correlation trends. Moreover, soil Alkali-hydrolyzable nitrogen and L-leucine aminopeptidase were important environmental factors influencing the structure of bacterial and fungal communities in sorghum soils, respectively. Soil nutrient levels and microbial communities together explained the variation in annual sorghum yield. The results of this study provide evidence that short-term organic fertilizer substitution increases sorghum yield by improving soil properties and regulating microbial community structure.

Justify that the use of selenium has been shown to promote seed germination and seedling growth of some crops under salt stress but the physiological mechanisms in sorghum remain poorly understood. Herein, grain sorghum [Sorghum bicolor (L.) Moench] variety Jinza 2002 was used to explore the effects and mechanisms of seed priming with 25 μmol/L sodium selenite [Se(IV)] on seed germination, seedling growth, antioxidant enzyme activity and osmotic regulatory substances under 120 mmol/L NaCl. The results showed that soaking sorghum seeds in Se(IV) effectively alleviated the harm of salt stress. Compared with the Na group, Se(IV) application increased germination percentage (by 23.8%), while facilitating the elongation of radicles (by 4.2%) and plumules (by 18.4%) under salt stress. Salt-stressed seedlings showed increased levels of leaf chlorophyll, soluble sugar, soluble protein and proline contents, and antioxidant enzyme activities, as well as root activity, and total biomass following Se(IV) application, along with decreased relative electrical conductivity and malondialdehyde content in the leaves. Findings of this study indicate that seed priming with Se(IV) facilitates sorghum seed germination and seedling growth under salt stress by improving root activity, enhancing leaf photosynthesis and antioxidant responses, and reducing cell membrane damage.

The effects of balanced fertilization with nitrogen, phosphorus, and potassium (NPK) on foxtail millet productivity and the soil environment under the same conditions of total nutrients have received limited research attention. Therefore, in this study, three balanced fertilization patterns of 27-14-10 (T1), 27-17-7 (T2), and 30-10-11 (T3), and one no fertilization treatment (CK), a total of four treatments, were set up through a two-year field experiment to study the effects of balanced fertilization patterns on foxtail millet yield and soil environment. Mantel analysis was conducted to reveal the correlation between soil environmental factors and the community and their contribution to productivity. The results showed that: (1) all balanced fertilization treatments significantly increased foxtail millet yield, with the highest yield in the T1 treatment. (2) The contents of EC, available K, available P, and alkaline-hydrolyzable nitrogen in the soil of the two-year TI treatments were higher than those of the other treatments and increased by 7.20–9.36%, 24.87–52.35%, 55.83–56.38%, and 21.05–43.95%, respectively, compared with CK. (3) Soil urease activity in the T1 treatment increased significantly by 26.67% and 9.00% compared with the control over the two years. Sucrase activity increased by 36.27% and 23.88% in the T1 treatment compared to CK, and glutaminase activity increased by 33.33% and 19.23% in the T1 treatment compared to CK. (4) T1 treatment significantly increased the OUT number and diversity index of the soil bacterial community. (5) Mantel analysis and principal component analysis showed that available soil nutrients and soil enzymes were positively correlated, and soil enzymes and soil nutrients contributed more to foxtail millet productivity. In this study, the 27-14-10 balanced fertilization pattern was more effective, providing a theoretical basis for the research and development of special fertilizers for foxtail millet and offering technical guidance for realizing the light simplified cultivation of foxtail millet and sustainable development of cost–saving and increased efficiency.

Conventional fertilizer management can destroy the structure of soil. Replacing chemical fertilizers with organic fertilizers can improve soil quality and nitrogen use efficiency. We aimed to study the effects of organic fertilizer substitutions for chemical nitrogen fertilizer on soil fertility and nitrogen use efficiency in order to clarify the effectiveness of the available nutrient management measures in improving soil quality and increasing foxtail millet yield. A field experiment was carried out over two consecutive years, and a total of six treatments were set up: no fertilizer (CK), chemical nitrogen fertilizer alone (N), the substitution of 25% of chemical nitrogen fertilizer with bio-organic fertilizer (N25A1), the substitution of 25% of chemical nitrogen fertilizer with fermented mealworm manure (N25B1), the substitution of 50% of chemical nitrogen fertilizer with bio-organic fertilizer (N50A2), and the substitution of 50% of chemical nitrogen fertilizer with fermented mealworm manure (N50B2). The results of this study show the following: (1) Compared with chemical nitrogen fertilizer, the substitution of organic fertilizer for nitrogen fertilizer reduced the bulk density and solid phase of the soil, and it increased the total porosity, water content, liquid phase, and gas phase of the soil. (2) Compared with nitrogen fertilizer, the use of an organic fertilizer increased the contents of nitrate nitrogen, ammonium nitrogen, and total nitrogen in the soil by 13.59~52.56%, 4.47~18.27%, and 4.40~12.09%, respectively. The content of alkaline nitrogen increased by 1.70~32.37%, and the contents of soil available potassium, available phosphorus, and organic matter also increased. (3) The activities of sucrase, urease, glutaminase, and asparaginase were improved by replacing chemical nitrogen fertilizer with organic fertilizer. The N25 treatments performed better than the N50 treatments, and fermented mealworm manure performed better than biological organic fertilizer. (4) A moderate application of organic fertilizer (N25) can increase the grain yield, ear weight, grain weight, and 1000-grain weight of foxtail millet, whereas excessive application of organic fertilizer (N50) can reduce foxtail millet yield. (5) Replacing chemical nitrogen fertilizer with organic fertilizer can improve the agronomic use efficiency, physiological efficiency, biased productivity, harvest index, and apparent use efficiency of nitrogen fertilizer. In this study, the substitution of 25% of chemical nitrogen fertilizer with fermented mealworm manure was the best combination for restoring crop productivity and soil quality.

Foxtail millet (Setaria italica L.) is of high nutritious value, which is an important crop in arid and semi-arid regions. The objective of this experiment was to explore the effects of the synthetic auxin naphthalene acetic acid (NAA) on the physiological processes of foxtail millet, and to provide a theoretical basis and technical approaches for its efficient use in millet cultivation. Two foxtail millet varieties (‘Jingu 21’ and ‘Zhangzagu 5’) were treated with six concentrations of NAA from 0-144 mg L-1 at the grain-filling stage in field experiments. The photosynthetic pigment contents, gas exchange parameters, chlorophyll fluorescence parameters, and grain yield were measured in foxtail millet. The results showed that low concentrations of NAA (18-36 mg L-1) increased the contents of photosynthetic pigments, and increased the activities of antioxidant enzymes, the photosynthetic rate, and the activity of photosystem system II (PS II). At higher NAA concentrations, the facilitation effect of the treatments diminished, showing a clear concentration effect. In this study, yield was significantly and positively correlated with PS II effective quantum yield (Y(II)) and the PSII electron transport rate (ETR), and the net photosynthetic rate (Pn) was significantly and positively correlated with chlorophyll content, stomatal conductance (Gs), Y(II), and ETR. These results also indicated that exogenous NAA application promotes the production of ATP and NADPH by increasing the efficiency of electron transfer within the photosystems and also improved photochemical utilization, which facilitates the fixation and reduction of carbon, ultimately leading to an increase in Pn and increasing grain yield in foxtail millet.

Sorghum bicolor (L.) is one of the oldest crops cultivated by human beings which has been used in food and wine making. To understand the genetic diversity of sorghum breeding resources and further guide molecular-marker-assisted breeding, six yield-related traits were analyzed for 214 sorghum germplasm from all over the world, and 2,811,016 single-nucleotide polymorphisms (SNPs) markers were produced by resequencing these germplasms. After controlling Q and K, QTLs were found to be related to the traits using three algorisms. Interestingly, an important QTL was found which may affect multiple traits in this study. It was the most likely candidate gene for the gene SORBI_3008G116500, which was a homolog of Arabidopsis thaliana gene-VIP5 found by analyzing the annotation of the gene in the LD block. The haplotype analysis showed that the SORBI_3008G116500hap3 was the elite haplotype, and it only existed in Chinese germplasms. The traits were proven to be more associated with the SNPs of the SORBI_3008G116500 promoter through gene association studies. Overall, the QTLs and the genes identified in this study would benefit molecular-assisted yield breeding in sorghum.
