Spinal cord injury (SCI), a debilitating medical condition that can cause irreversible loss of neurons and permanent paralysis, currently has no cure. However, regenerative medicine may offer a promising treatment. Given that numerous regenerative strategies aim to deliver cells and materials in the form of tissue-engineered therapies, understanding and characterising the mechanical properties of the spinal cord tissue is very important. In this study, we have systematically characterised the spatiotemporal changes in elastic stiffness (elastic modulus, Pa) and viscosity (drop in peak force, %) of injured rat thoracic spinal cord tissues at distinct time points after crush injury using the indentation technique. Our results demonstrate that in comparison with uninjured spinal cord tissue, the injured tissues exhibited lower stiffness (median 3281 Pa versus 9632 Pa; P 0.001) but demonstrated elevated viscosity (median 80% versus 57%; P 0.001) at 3 days postinjury. Between 4 and 6 weeks after SCI, the overall viscoelastic properties of injured tissues returned to baseline values. At 12 weeks after SCI, in comparison with uninjured tissue, the injured spinal cord tissues displayed a significant increase in both elasticity (median 13698 Pa versus 9920 Pa; P 0.001) and viscosity (median 64% versus 58%; P 0.001). This work constitutes the first quantitative mapping of spatiotemporal changes in spinal cord tissue elasticity and viscosity in injured rats, providing a mechanical basis of the tissue for future studies on the development of biomaterials for SCI repair. STATEMENT OF SIGNIFICANCE: Spinal cord injury (SCI) is a devastating disease often leading to permanent paralysis. While enormous progress in understanding the molecular pathomechanisms of SCI has been made, the mechanical properties of injured spinal cord tissue have received considerably less attention. This study provides systematic characterization of the biomechanical evolution of rat spinal cord tissue after SCI using a microindentation test method. We find spinal cord tissue behaves significantly softer but more viscous immediately postinjury. As time passes, the lesion site gradually returns to baseline values and then displays pronounced increased viscoelastic properties. As host tissue mechanical properties are a crucial consideration for any biomaterial implanted into central nervous system, our results may have important implications for further studies of SCI repair.

Spinal cord injury (SCI) is a global problem that brings a heavy burden to both patients and society. Recent investigations indicated degenerative disease is taking an increasing part in SCI with the growth of the aging population. However, little insight has been gained about the effect of cervical degenerative disease on the spinal cord during dynamic activities. In this work, a dynamic fluid-structure interaction model was developed and validated to investigate the effect of anterior and posterior encroachment caused by degenerative disease on the spinal cord during normal extension and flexion. Maximum von-Mises stress and maximum principal strain were observed at the end of extension and flexion. The abnormal stress distribution caused by degenerative factors was concentrated in the descending tracts of the spinal cord. Our finding indicates that the excessive motion of the cervical spine could potentially exacerbate spinal cord injury and enlarge injury areas. Stress and strain remained low compared to extension during moderate flexion. This suggests that patients with cervical degenerative disease should avoid frequent or excessive flexion and extension which could result in motor function impairment, whereas moderate flexion is safe. Besides, encroachment caused by degenerative factors that are not significant in static imaging could also cause cord compression during normal activities.

This study aims to establish and validate a poroelastic L4-L5 finite element model to evaluate the effect of different sitting postures and their durations on the mechanical responses of the disc. During the sustained loading conditions, the height loss, fluid loss and von-Mises stress gradually increased, but the intradiscal pressure decreased. The varying rates of aforementioned parameters were more significant at the initial loading stage and less so at the end. The predicted values in the flexed sitting posture were significantly greater than other postures. The extended sitting posture caused an obvious von-Mises stress concentration in the posterior region of the inter-lamellar matrix. From the biomechanical perspective, prolonged sitting may pose a high risk of lumbar disc degeneration, and therefore adjusting the posture properly in the early stage of sitting time may be useful to mitigate that. Additionally, upright sitting is a safer posture, while flexed sitting posture is more harmful.

The present study preliminarily evaluated neurotoxicity injuries induced by chlorpyrifos in PC12 ​cell, which were used as a model for nervous cell system. In cultured PC12 ​cell, application of soy isoflavones (genistein, daidzein, monomer and mixture) significantly reduced chlorpyrifos induced toxicity, a widely used pesticide, and resulted in a better cell survival rate. Treatments with isoflavones reduced malondialdehyde content, reactive oxygengeneration and acetylcholine level in medium, and maintained mitochondrial membrane potential integrity. Daidzein enhanced endogenous antioxidant system in PC12 ​cell with an increasing in superoxide dismutase per-unit activity. Genistein reduced acetylcholine content in the medium. Daidzein and genistein showed different effects, and their combined effect were greater than individual. In conclusion, soy isoflavones as an antioxidant and neuroprotectant, enhanced choline metabolism, which effectively mitigated disadvantageous influence of PC12 ​cell caused by chlorpyrifos.

Food safety and quality have been gaining increasing attention in recent years. Gas chromatography coupled to tandem mass spectrometry (GC–MS/MS), a highly sensitive technique, is gradually being preferred to GC–MS in food safety laboratories since it provides a greater degree of separation on contaminants. In the analysis of food contaminants, sample preparation steps are crucial. The extraction of multiple target analytes simultaneously has become a new trend. Thus, multi-residue analytical methods, such as QuEChERs and adsorption extraction, are fast, simple, cheap, effective, robust, and safe. The number of microorganic contaminants has been increasing worldwide in recent years and are considered contaminants of emerging concern. High separation in MS/MS might be, in certain cases, favored to sample preparation selectivity. The ideal sample extraction procedure and purification method should take into account the contaminants of interest. Moreover, these methods should cooperate with high-resolution MS, and other sensitive full scan MSs that can produce a more comprehensive detection of contaminants in foods. In this review, we discuss the most recent trends in preparation methods for highly effective detection and analysis of food contaminants, which can be considered tools in the control of food quality and safety.

Objective Preexisting severe cervical spinal cord compression is a significant risk factor in cervical hyperextension injury, and the neurological function may deteriorate after a slight force to the forehead. There are few biomechanical studies regarding the influence of pathological factors in hyperextension loading condition. The aim of this study is to analyze the effects of preexisting different types of cervical disc herniation and different degrees of compression on the spinal cord in cervical hyperextension. Method A 3D finite element (FE) model of cervical spinal cord was modeled. Local type with median herniation, local type with lateral herniation, diffuse type with median herniation, and diffuse type with lateral herniation were simulated in neutral and extention positions. The compressions which were equivalent to 10%, 20%, 30%, and 40% of the sagittal diameter of the spinal cord were modeled. Results The results of normal FE model were consistent with those of previous studies. The maximum von Mises stresses appeared in the pia mater for all 32 loading conditions. The maximum von Mises stresses in extension position were much higher than in neutral position. In most cases, the maximum von Mises stresses in diffuse type were higher than in local type. Conclusion Cervical spinal cord with preexisting disc herniation is more likely to be compressed in hyperextension situation than in neutral position. Diffuse type with median herniation may cause more severe compression with higher von Mises stresses concentrated at the anterior horn and the peripheral white matter, resulting in acute central cord syndrome from biomechanical point of view.

The cervical spine injury is a complicated procedure in the combination of different injury loads and postures. The aim of this study is to investigate the injury mechanism considering different types of cervical curvatures subjected to contusion loads. A finite element model of a cervical spinal cord was constructed. Gray matter, white matter and pia matter were modeled and hyperelastic material properties were assigned. Convergence analysis and validation analysis were carried out. The model was simulated in 3 different spinal curvatures and loaded by 2 directions with 4 compression degrees. The maximum von Mises stress in the whole model was concentrated in the pia matter in all loading cases. When investigating spinal cord injury, the pia matter must be considered. For all three curvatures, the stress in the gray matter and white matter was higher in front-to-back loading condition than that in back-to-front loading condition. The front-to-back impact may cause a larger damage. A back-to-front load damaged the structure around the central canal and a front-to-back contusion load damaged the anterior horn of the spinal cord at most time. From the view of the maximum stress, the lordotic curvature did not show significant buffering effect. However, the pathological curvature had large areas affected and the lordotic curvature showed some benefits to some degree from the view of stress distribution.

Glyphosate is one of the most commonly used and non-selective herbicides in agriculture, which may directly pollute the environment and threaten human health. A simple and effective approach to its detection is thus quite necessary. Surface-enhanced Raman scattering (SERS) spectroscopy was shown to be a very effective method to approach the problem. However, sensitivity in SERS experiments is quite low, caused by different orientation/conformation of the adsorbed molecules on the metal surface, which limit its detection by using SERS. In this paper, 2‒13C‒glyphosate (hereafter: 13&ndash, GLP) was designed as a model molecule for theoretical and experimental studies of the molecule structure. Vibrational modes were assigned based on the modeling results obtained at the B3LYP/6-311++G** level by density functional theory (DFT) calculations, which were performed to predict the FT‒IR and Raman spectra. Band downshifts were caused by 13C atom isotopic substitution with mass changed. Moreover, SERS spectra of 13&ndash, GLP by combining ninhydrin reaction on Ag NPs were obtained. Isotopic Raman shifts are helpful in identifying the components of each Raman band through vibrations across the molecular system. They are coupled by probe molecules and thus bind to the substrates, indirectly offering the opportunity to promote interactions with Ag NPs and reduce the complex equilibrium between different orientation/conformation of glyphosate molecules on the metal surface.

Edible insects are ideal food sources, which contain important nutrients and health-promoting compounds. With a rapid development of industrial insect farming, insect-derived food is a novel and emerging food industry. Edible insects have been traditionally consumed in various communities, while continuously gaining relevance in today's society; however, they currently remain underutilized. Although there are a large number of literature on edible insects, these literature primarily focus on the nutritional value edible insects. The toxicity assessment data of edible insects remain incomprehensive, especially for the new national standard that is currently in effect; and many data and conclusions are not accurately specified/reported. Therefore, we performed a literature review and summarized the data on the toxicological assessment of edible insects in China. The review first describes the research progress on safety toxicological assessment, and then offers references regarding the development of 34 edible insect species in China. These data can be a platform for the development of future toxicological assessment strategies, which can be carried out by a multidisciplinary team, possibly consisting of food engineers, agronomists, farmers, and so on, to improve the acceptability of edible insects.

Glyphosate is one of the most commonly-used and non-selective herbicides in agriculture, which may directly pollute the environment and threaten human health. A simple and effective approach to assessment of its damage to the natural environment is thus quite necessary. However, traditional chromatography-based detection methods usually suffer from complex pretreatment procedures. Herein, we propose a simple and sensitive method for the determination of glyphosate by combining ninhydrin reaction and surface-enhanced Raman scattering (SERS) spectroscopy. The product (purple color dye, PD) of the ninhydrin reaction is found to SERS-active and directly correlate with the glyphosate concentration. The limit of detection of the proposed method for glyphosate is as low as 1.43×10-8mol·L-1 with a relatively wider linear concentration range (1.0×10-7-1.0×10-4mol·L-1), which demonstrates its great potential in rapid, highly sensitive concentration determination of glyphosate in practical applications for safety assessment of food and environment.

Insects provide a series of ecological services vital to human survival. Over 1000 insect species have been used for human consumption in Asia, Latin America, and Africa for more than 2000 years. Among them, the soybean hawkmoth, Clanis bilineata tsingtauica (CBT), is a traditional edible insect. CBT, known for its high nutritional value, is considered safe with a long consumption history in East Asia. The present review provides an overview of the rearing technology and utilization prospects of CBT. It has been extensively cultivated on live soybean plants under field and glasshouse conditions. However, an efficient rearing technology has not been reported. The mass production of CBT is still under investigation, and more advanced technology is required to develop high-quality food ingredients to meet consumer needs on a large scale. In addition, food derived from the soybean hawkmoth is prevalent in the farm product market. It is used as freeze-dried, fried, fresh meat, and canned meat. CBT-derived food, a potential dietary supplement used to retard aging in humans, would be a novel and emerging product in the food industry. The development of CBT-derived food will generate more economic and social value if the market demand can be met. This review will provide an insight into CBT mass production and its potential application in the food industry.

We planned to investigate the protective effect of soy isoflavone (SIF) genistein (GEN), and daidzein (DAI), and their mixed solution against carbaryl-induced toxicity in PC12 cells. PC12 cells were exposed to carbaryl, pretreated, co-treatment, or posttreatment with three different SIF solutions for indicated times. Exposure to SIF resulted a better survival and cell morphology which were subjected to carbaryl toxicity. Treatment with SIF was also found to reduce malondialdehyde content, superoxide dismutase inhibitory rate, reactive oxygen species generation, acetylcholine levels in medium, and maintain the integrity of mitochondrial membrane potential. It was also found to increase GSH level. In conclusion, SIF can effectively mitigate adverse effects induced by carbaryl. GEN and DAI have a combined effect that is greater than the effect obtained with each isoflavone alone, although they have different effects against carbaryl-induced damage. We review the recent literature on the protective effects of soy isoflavones and other bioactive components. We review the effect of combinations of three soy isoflavones, daidzein, genistein, and glycitein and examine the interactions of individual soy isoflavones with other compounds such as vitamins, trace elements, chemotherapeutics, and phytoestrogens. We further review the effect of whole isoflavones and other compounds and discuss these effects when assessing the risks associated with various environmental and food compounds.

Pesticides directly pollute the environment and contaminate foods ultimately being absorbed by the human body. Their residues contain highly toxic substances that have been found to cause serious problems to human health even at very low concentrations. The gold standard method, gas/liquid chromatography combined with mass spectroscopy, has been widely used for the detection of pesticide residues. However, these methods have some drawbacks such as complicated pretreatment and cleanup steps. Recent technological advancements of surface-enhanced Raman spectroscopy (SERS) have promoted the creation of alternative detection techniques. SERS is a useful detection tool with ultrasensitivity and simpler protocols. Present SERS-based pesticide residue detection often uses standard solutions of target analytes in conjunction with theoretical Raman spectra calculated by density functional theory (DFT) and actual Raman spectra detected by SERS. SERS is quite a promising technique for the direct detection of pesticides at trace levels in liquid samples or on the surface of solid samples following simple extraction to increase the concentration of analytes. In this review, we highlight recent studies on SERS-based pesticide detection, including SERS for pesticide standard solution detection and for pesticides in/on food samples. Moreover, in-depth analysis of pesticide chemical structures, structural alteration during food processing, interaction with SERS substrates, and selection of SERS-active substrates is involved.

Spray drying is a useful method for developing egg process and utilization. The objective of this study was to evaluate effects on spray drying condition of egg white. The optimized conditions were spraying flow 22 mL/min, feeding temperature 39.8 °C and inlet-air temperature 178.2 °C. Results of sulfydryl (SH) groups measurement indicated conformation structure have changed resulting in protein molecule occur S-S crosslinking phenomenon when heating. It led to free SH content decreased during spray drying process. There was almost no change of differential scanning calorimetry between fresh egg white and spray-drying egg white powder (EWP). For a given protein, the apparent SH reactivity is in turn influenced by the physico-chemical characteristics of the reactant. The phenomenon illustrated the thermal denaturation of these proteins was unrelated to their free SH contents. Color measurement was used to study browning level. EWP in optimized conditions revealed insignificant brown stain. Swelling capacity and scanning electron micrograph both proved well quality characteristic of spray-drying EWP. Results suggested spray drying under the optimized conditions present suitable and alternative method for egg processing industrial implementation. Egg food industrialization needs new drying method to extend shelf-life. The purpose of the study was to provide optimal process of healthy and nutritional instant spray-drying EWP and study quality characteristic of spray-drying EWP.

We present a review concerning the determination and control of pesticide residues and transformation products in beverages. We review preparation methods using microextraction techniques, which are time-consuming, inexpensive, simple, and endeavor to be environment friendly. Mass spectrometry (MS) and tandem mass spectrometry (MS/MS) technologies are able to detect and quantify matrix-induced chromatographic response effects (matrix effects) accurately, even with the low milligram per kilogram levels. Rapid detection technology is an important tool in food product analysis. We review the technology to analyze the persistence of pesticide residues in beverages in order to effectively control their quality and safety.

Lead ions were coupled to protein carrier bovine serum albumin (BSA) and keyhole limpet hemocyanin (KLH) respectively. Diethylene Triamine Pentacetate Acid (DTPA) was used as bifunctional chelator. Pb-DTPA-Carrier protein, DTPA-Carrier protein and carrier protein were detected qualitatively with ICP-MS, UV spectrophotometer and electrophoresis. It turns out that the Pb-DTPA-Carrier protein contains certain concentration of heavy metal ion, the maximum ultraviolet absorption peak value of Pb-DTPA-Carrier protein significantly higher than that of DTPA-Carrier protein and carrier protein, and the electrophoresis reveals more vague bands of Pb-DTPA-Carrier protein with slower migrations speed comparing to the bands of the DTPA-Carrier protein and carrier protein. The result indicated that the artificial antigen for lead was successfully synthesized.
