18. Multiwalled Carbon Nanotubes Interact with Macrophages and Influence Tumor Progression and Metastasis

Abstract

Macrophages are one of the most important types of immune effector cells and are closely associated with tumor progression and metastasis. In this work, we investigated the influences of oxidized multiwalled carbon nanotubes (o-MWCNT) on macrophages that are resting in the normal subcutis tissue or in the tumor microenvironment in vivo as well as on the macrophage cell line of RAW 264.7 treated with combination of IL4, IL10 and IL13 in vitro. The o-MWCNT were characterized with SEM, DLS, FTIR, TGA, and UV-vis-NIR spectroscopy, and their effects on the RWA 264.7 cell line and breast cancer tumor-bearing mice were analyzed using the MTS assay, flow cytometry analysis, and histological and immunohistochemical observations. Our experimental results showed that subcutaneously injected o-MWCNT not only induced phagocytosis of the local resident macrophages, but also competitively recruited macrophages from other tissues. These interactions resulted in macrophage reduction and decreased vessel density around the tumor mass, which together inhibited tumor progression and metastasis in the lung. In the cell line model, the o-MWCNT inhibited the ability of the interleukin treated RAW macrophages to promote tumor cell migration as well as decreased their proliferation rate.

Abstract

1-Alkylpyridinium bromide [Cnpy]Br is a common intermediate in chemical synthesis. With the discharge of industrial wastewater, it enters the environment and is toxic to plants. In this study, the impacts of two pyridine-based ionic liquids (ILs), [C3py]Br and [C5py]Br, on the growth and physiology of rape seedlings were investigated at concentrations of 10, 50, 100, 200, 300, and 400 mg/L. Within the concentration range (10–400 mg/L) of [C3py]Br and [C5py]Br treatment, the root length, plant height, activities of antioxidant enzymes (SOD, POD, and CAT), and the contents of Chla and Chlb showed an increase at low concentrations and a decrease at high concentrations. [C3py]Br and [C5py]Br increased MDA content in rape seedlings leaves in a concentration-dependent manner. It was also found that [C5py]Br was more toxic to rape seedlings than [C3py]Br. The toxicity of pyridine ILs such as [C3py]Br and [C5py]Br to plants should be highly concerned. [ABSTRACT FROM AUTHOR]

Abstract

While the functions of STING1 (stimulator of interferon response cGAMP interactor 1) during DNA virus infection had been well documented, the roles STING1 plays during RNA viruses infection is obscure. Infection with foot-and-mouth disease virus (FMDV), a well-known picornavirus, induces endoplasmic reticulum (ER) stress response and autophagy. Here, we found that the FMDV-induced integrated stress response originates from the cellular pattern recognition receptor DDX58/RIG-I (DExD/H-box helicase 58). DDX58 transmits signals to the ER-anchored adaptor protein STING1, which specifically activates the EIF2AK3/PERK (eukaryotic translation initiation factor 2A)-dependent integrated stress response and finally leads to reticulophagy and degradation of STING1 itself. Knockdown/knockout of STING1 or EIF2AK3 suppresses FMDV genome replication and viral protein expression. Reticulophagy induction by STING1 does not require its translocation to the Golgi or IFN response activation. However, STING1 polymerization is necessary for the FMDV-induced integrated stress response and reticulophagy. Our work illustrated the signaling cascades that mediate the cellular stress response to FMDV infection and indicated that induction of autophagy in response to both DNA and RNA virus infection may be an evolutionarily conserved function of STING1. Abstract

Due to the migration or transboundary spread of domestic and wild animals, peste des petits ruminants virus posed a high potential threat to them. In this study, we initially detected that a class of animal named Procapra przewalskii was infected with peste des petits ruminants virus (PPRV ChinaGS2018) in Gansu province. According to phylogenetic relationships analysis, we found that ChinaGS2018 comprised of 15,954 nucleotides and was classified into IV genotypes. In addition, indirect immunofluorescence assay (IFA) showed that ChinaGS2018 could infect isolated primary goat tracheal epithelium cells (GTC). Comparing with full-length genome sequences revealed that ChinaGS2018 strain has high identity to the reference complete genomes (87.16-99.55%) at the nucleotide level. Multiple sequence alignment showed that F protein has the highest identity of 99.8%, and H protein has the highest nucleotide substitution ratio. Our study also suggested this strain may be transmitted from Xinjiang, China. Along with the migratory of Procapraprzewalskii, this wild ruminant infected with PPRV can pose a huge threat to other wild ruminants and domestic ones. This is the first report describing infected with PPRV which will provide insights into the epidemiology and pathogenesis of this important virus.

Abstract

Coastal Louisiana hosts 37% of the coastal wetland area in the conterminous US, including one of the deltaic coastal regions more susceptible to the synergy of human and natural impacts causing wetland loss. As a result of the construction of flood protection infrastructure, dredging of channels across wetlands for oil/gas exploration and maritime transport activities, coastal Louisiana has lost approximately 4900 km(2) of wetland area since the early 1930s. Despite the economic relevance of both wetland biomass and net primary productivity (NPP) as ecosystem services, there is a lack of vegetation simulation models to forecast the trends of those functional attributes at the landscape level as hydrological restoration projects are implemented. Here, we review the availability of peer-reviewed biomass and NPP wetland data (below and aboveground) published during the period 1976-2015 for use in the development, calibration and validation of high spatial resolution (<200 m x 200 m) vegetation process-based ecological models. We discuss and list the knowledge gaps for those species that represent vegetation community associations of ecological importance, including the long-term research issues associated to limited number of paired belowground biomass and productivity studies across hydrological basins currently undergoing different freshwater diversions management regimes and hydrological restoration priorities.

Abstract

The density and surface tension values of aqueous 1-propylpyridinium bromide [C3py]Br at concentrations between 0.0099 and 1.0337 mol·kg–1were measured at T= (288.15–318.15) K with an interval of 5 K, and the molar surface quasi-Gibbs energy model of aqueous [C3py]Br was established, and the relationships among the molar surface quasi-Gibbs energy, molality, and temperature were also explored. Moreover, the Eötvös equation was improved in terms of the molar surface quasi-Gibbs energy model.
