Sea surface temperatures are rising at unprecedented rates, leading to a progressive degradation of complex habitats formed by coral reefs. In parallel, acute thermal stress can lead to physiological challenges for ectotherms that inhabit coral reefs, including fishes. Warming and habitat simplification could push marine fishes beyond their physiological limits in the near future. Specifically, questions remain on how warming and habitat structure influence the brain of marine fishes. Here we evaluated how thermal stress and habitat loss are acting independently and synergistically as stressors in a damselfish of the Western Atlantic, Abudefduf saxatilis. For this experiment, 40 individuals were exposed to different combinations of temperature (27°C or 31°C) and habitat complexity (complex vs simple) for 10 days, and changes in brain gene expression and oxidative stress of liver and muscle were evaluated. The results indicate that warming resulted in increased oxidative damage in the liver (p=0.007) and changes in gene expression of the brain including genes associated with neurotransmission, immune function, and tissue repair. Individuals from simplified habitats showed higher numbers of differentially expressed genes, and changes for genes associated with synaptic plasticity and spatial memory. In addition, a reference transcriptome of A. saxatilis is presented here for the first time, serving as a resource for future molecular studies. Human activity has increased concentrations of endocrine-disrupting contaminants (EDCs) in many ecological systems. Many EDCs are xenoestrogens, which imitate naturally derived estrogen like estradiol 17-β (E2). These pollutants can critically affect a broad range of biological functions, particularly in organisms inhabiting aquatic environments. E2 and associated receptors are involved in regulating innate immune responses, where documentation of exogenous E2 on immune parameters is important for understanding health consequences. In this study, we explore the impact of environmentally relevant concentrations of E2 on circulating glucocorticoid levels and several innate immune parameters in hatchling American alligators (Alligator mississippiensis). Twenty-three hatchling alligators were randomly placed in one of three groups that differed in dietary E2 concentration: control (no E2 exposure), low E2 (0.5 μg/kg E2), or high E2 (1 μg/kg E2) for 10 weeks. Following this period, several biomarkers were quantified to monitor the impact of E2: growth, change in body condition, white blood cell (WBC) counts, glucocorticoid levels, and general antibody response. Blood E2 concentrations were greater in individuals exposed to E2, but plasma corticosterone levels were reduced among the experimental groups. Morphology, growth, and immune parameters of E2 exposed animals did not differ from controls. These results suggest that acute exposure to increased environmental estrogen concentrations may alter plasma hormone concentrations but have little to no impact on immediate morphology or immune responses. Birds appear to have the ability to bias offspring sex ratios in response to environmental and social conditions. The mechanisms responsible remain unknown, however one previous study indicated a link between rates of ovarian follicle growth and the sexes of resulting eggs. This could indicate that either follicles destined to retain male or female follicles grow at different rates, or the rate of ovarian follicle growth determines the sex chromosome that will be retained to influence the sex of the resulting offspring. We tested for evidence of both possibilities by staining yolk rings that are indicative of daily growth. First, we tested for a correlation between the number of yolk rings and the sexes of the resulting germinal discs collected from each egg, and second, we tested whether experimentally decreasing follicle growth rates with a dietary yolk supplement impacts the sexes of the resulting germinal discs. There was no significant correlation between the number of yolk rings and the sexes of the resulting embryos, and decreasing follicle growth rates did not impact sexes of resulting germinal discs. Endocrine disrupting chemicals are present in the environment and/or in consumer products. These agents have the capacity to mimic and/or antagonize endogenous hormones and thus perturb the endocrine axis. The male reproductive tract expresses steroid hormone (androgen and estrogen) receptors at high levels and is a major target for endocrine disrupting chemicals. In this study, Long-Evans male rats were exposed to dichlorodiphenyldichloroethylene, a metabolite of dichlorodiphenyltrichloroethane and a chemical present in the environment, in drinking water at 0.1 and 10 μg/L for 4 weeks. At the end of exposure, we measured steroid hormone secretion and analyzed steroidogenic proteins, including 17β-hydroxysteroid dehydrogenase, 3β-hydroxysteroid dehydrogenase, steroidogenic acute regulatory protein, aromatase, and the LH receptor. We also analyzed Leydig cell apoptosis (poly-(ADP-ribose) polymerase) and caspase-3 in the testes. Testicular testosterone (T) and 17β-estradiol (E2) were both affected by exposure to dichlorodiphenyldichloroethylene by displaying altered steroidogenic enzyme expression. Dichlorodiphenyldichloroethylene exposure also increased the expression of enzymes mediating the pathway for programmed cell death, including caspase 3, pro-caspase 3, PARP, and cleaved PARP. Altogether, the present results demonstrate that dichlorodiphenyldichloroethylene directly and/or indirectly can target specific proteins involved in steroid hormone production in the male gonad and suggest that exposure to environmentally relevant dichlorodiphenyldichloroethylene levels has implications for male reproductive development and function., (© The Author(s) 2023. Generation of egg yolk by birds requires the synthesis and deposition of large amounts of protein and lipid, and is often accompanied by the incorporation of additional physiological mediators. While there has been much work examining the relative quantities of yolk components, as well as potential adaptive patterns of their allocation, we still do not have a full understanding of what controls yolk formation and composition. Once ovarian follicles are recruited into the preovulatory hierarchy, the yolk is deposited in concentric rings, with one ring deposited per day. Previous studies have shown that there is substantial interspecific and intraspecific variation in the number of rings in yolks, and thus the number of days it took those yolks to grow. We hypothesized that the ability to grow follicles to maturity quickly is limited by the availability of materials to make yolk precursors in the female, either in body reserves or in dietary access. To test this, we supplemented the diets of Japanese quail with hard-boiled chicken yolk and examined the influences of treatment and female body condition on follicle growth rates. Contrary to predictions, females with higher body condition indices produced yolks that grew more slowly, and yolks from supplemented birds grew more slowly than controls. These results indicate that females can modulate the rate of yolk incorporation into developing follicles, and that an energy balance that is too high may not be optimal for the fast growth of developing ovarian follicles., (© 2022 The Authors. Environmental oestrogens pose serious concerns for ecosystems through their effects on organismal survival and physiology. The gut microbiome is highly vulnerable to environmental influence, yet the effects of oestrogens on gut homeostasis are unknown because they are poorly studied in wildlife populations. To determine the influence of environmental oestrogens (i.e., xenoestrogens) on the diversity and abundance of gut microbiota, we randomly assigned 23 hatchling American alligators (Alligator mississippiensis) to three ecologically relevant treatments (control, low, and high oestrogen concentrations) for 10 weeks. We predicted that xenoestrogen exposure would decrease microbial diversity and abundance within the digestive tract and that this effect would be dose-dependent. Microbial samples were collected following diet treatments and microbial diversity was determined using 16S rRNA gene-sequencing. Individuals in oestrogen-treatment groups had decreased microbial diversity, but a greater relative abundance of operational taxonomic units than those in the control group. In addition, this effect was dose-dependent; as individuals were exposed to more oestrogen, their microbiome became less diverse, less rich and less even. Avian egg yolks provide critical nutrients and energy reserves for a growing embryo. Once a follicle is selected into the ovulatory hierarchy, rings of yolk precursors are deposited daily until the yolk is large enough to ovulate. The composition of hen egg yolk and the mechanisms responsible for deposition in the ovarian follicle are well-studied. However, the factors that affect the rate of yolk deposition, and the number of days it takes for follicles to grow to the optimal size for ovulation, are less well-known. Social interactions appear to impact the rate of yolk deposition in many avian species and testosterone is a key hormone involved in regulating those interactions. Therefore, we hypothesized that testosterone would influence the rate of follicle growth. We tested the influences of exogenous testosterone treatment by rubbing either testosterone propionate cream (1.25 %) or a control cream to hen combs daily for 7 days to increase plasma testosterone concentrations. We then collected eggs and measured egg and yolk weight, yolk diameters, and stained and counted the number of concentric rings in each yolk. Testosterone-treated hens laid fewer eggs and grew their yolks more slowly than control hens. Testosterone also caused an increase in yolk weight but not yolk diameter. In complex terrestrial environments, chemical signals can be the most important sensory modality for locating conspecifics for potential mating opportunities, especially in spatially segregated populations or habitats. Organisms must evolve chemical signals to maximize the efficacy of conveying information, particularly in creating trails or mate-choice cues. Long-distance transmission of chemical signals may be an increasingly important management concern for small and fractured populations or potentially threatened species, such as gopher tortoises in the southeastern U.S. Mental gland secretions have been shown to have pheromonal function in gopher tortoises, suggesting a potential role as trail or marking pheromones, allowing males to track females or other males to find females. In this study, male gopher tortoises were given paired presentations of a negative control (distilled water) with serial dilutions (1:4, 1:20, 1:100, and 1:500) of male mental gland secretions. Male tortoises were able to discern treatment differences up to 1:20 diluted secretions, responding with an array of social behaviors (e.g. for the 1:20 dilution trial, carapace alignment and head bobbing occurred more frequently for the mental gland secretion relative to the control; p < 0.01). Multivariate principal components analysis yielded PC1 (including, approach, carapace alignment, head bobbing, tasting air, sniffing, and doubleback) that differed by treatment (p = 0.0007) and also was higher for the 1:20 diluted presentation relative to the 1:500 diluted presentation (p = 0.04). Chronic exposure to low doses of anthropogenic chemicals in the environment continues to be a major health issue. Due to concerns about the effects in humans and wildlife, use of persistent organic pollutants, such as dichlorodiphenyltrichloroethane (DDT), is prohibited. However, their ubiquitous nature and persistence allows them to remain in the environment at low levels for decades. Dichlorodiphenyldichloroethylene (DDE) is the most persistent metabolite of DDT and has been shown to cause hepatotoxicity, nephrotoxicity, hormonal disorders, and induce oxidative stress in many organisms. Although the effects of acute exposure to DDT and its metabolite DDE have been extensively studied, the chronic effects of sub-lethal DDE exposure at levels comparable to those found in the environment have not been well documented. Long-Evans male rats were used to determine the effect of relatively chronic and short term DDE (doses ranged from 0.001 to 100 μg/L) exposure on endocrine function and oxidative stress at different developmental time points. We found that circulating serum testosterone (T) levels were significantly decreased and T secretion in testicular explants were significantly influenced in a dose dependent manner in both pre-pubertal and pubertal male rats after DDE exposure, with pubertal rats being the most affected contrary to our original prediction. Additionally, exposure to DDE increased expression of protein oxidation indicating a possible increase in cellular damage caused by oxidative stress. Persistent contaminants are ubiquitous in our water and soil, and thus chronic exposure to environmentally relevant levels of these contaminants may pose a risk to humans and wildlife. Two species of small mammals (deer mouse, Peromyscus maniculatus and cotton rat, Sigmodon hispidus) were collected from historically dichlorodiphenyltrichloroethane (DDT)-contaminated and reference areas at the Redstone Arsenal (Huntsville, AL, USA), a US Environmental Protection Agency (USEPA)-designated Superfund site. Soil samples taken concurrently with mammal collection from the DDT abatement site exhibited DDT and dichlorodiphenyldichloroethylene (DDE) levels >1 ppm (USEPA total threshold limit concentration for DDT and DDE <1 ppm), whereas soil samples from the reference area had undetectable levels. Liver samples from individuals of both species collected in the contaminated areas also had significantly elevated DDT/DDE levels, with P. maniculatus averaging an approximately 20× greater load than that documented for S. hispidus. Both rodent species collected from the contaminated site had significantly lower total leukocyte counts and total differential blood cell counts (p = 0.01 and p = 0.04, respectively) than those collected from the nearby (~7000-m) reference site. Cellular stress indices also trended higher in both species, suggesting a potential for chronic exposure to DDT to act as a mediator of oxidative damage. The present study provides support to the idea that environmental exposure to low levels of contaminants can cause physiological consequences that may influence immune responsiveness and initiate cellular stress in resident mammals. Background: Fingolimod, an oral sphingosine 1-phosphate receptor modulator, is approved by EMA for relapsing-remitting multiple sclerosis (RRMS)., Objectives: To assess the effectiveness and safety of fingolimod in patients with RRMS in real-world clinical practice in Portugal., Methods: Retrospective, multicentre, non-interventional study, reporting 3 years follow-up of data collected from October 2015 to July 2016. Sociodemographic data and previous treatments at baseline and data regarding disease evolution, including number of relapses, annualised relapse rates (ARR) and Expanded Disability Status Scale (EDSS), were collected., Results: Two-hundred and seventy-five participants were enrolled in the REALMS study. Results showed that the main reason to switch to fingolimod was failure of previous treatment (56.7%) and only 3.6% were naïve patients. In the total population, there was a significant decrease in ARR of 64.6% in the first year of treatment, 79.7% in the second year and 82.3% in the third year, compared with baseline. More than 67.0% of patients had no relapses during the 3 years after switching to fingolimod. EDSS remained stable throughout the study., Conclusions: Therapy with fingolimod showed a sustained effectiveness and safety over the 3 years, particularly on patients switched from first-line drugs (BRACE). No new safety issues were reported.

AbstractIn birds, incubation temperature is critically deterministic for a range of traits. When parents leave the nest to forage, developing embryos can be exposed to cooling events that represent thermal stress. To investigate the consequences of periodic cooling on offspring development and physiology, we exposed zebra finch embryos to cooling events throughout the incubation period. We then compared embryonic survival, egg mass change, incubation duration, posthatch growth, and adrenocortical response of these individuals with embryos reared at a constant optimal temperature of 37.4°C and embryos reared at a constant suboptimal temperature of 36.4°C, the mean incubation temperature of periodically cooled embryos. There were no differences in embryonic survival or egg mass change during incubation, but individuals exposed to periodic cooling had longer incubation periods than those from the 37.4°C treatment and shorter incubation periods than those from the 36.4°C treatment. Periodically cooled individuals showed slower posthatch growth in comparison with both constant-temperature treatments, but this did not impact adult body size. Treatment groups did not differ in their adrenocortical response, but embryos exposed to periodic cooling and a constant temperature of 37.4°C were able to habituate to repeated capture and restraint stress, while individuals exposed to the constant temperature of 36.4°C were not. These results point to the differential impacts of cooling events versus constant low temperatures during incubation on posthatch growth and physiology and may represent a way for parents to devote less energy toward incubation while still ensuring offspring success.

With a growing number of species of conservation concern, understanding the physiological effects of routine sampling of vertebrate species remains a priority to maintain the welfare status of wildlife and ensure such activities are not counter to conservation goals. The gopher tortoise ( Gopherus polyphemus ) is a species of conservation concern throughout its range and is among the most frequently trapped turtles globally (for both research and conservation activities). Several studies have found equivocal results on the effects of trapping and handling on the glucocorticoid stress response. In this study, we tested how multiple physiological biomarkers (i.e. plasma lactate, corticosterone (cort), heterophil:lymphocyte ratio (HLR) and bactericidal ability (BA)) respond to four different combinations of trapping conditions in comparison to baseline reference sampling. We found that trapping and handling of gopher tortoises yielded a rapid rise in plasma lactate concentration followed by elevations of cort and stress-associated immune changes. In visibly distressed animals that were in traps for fewer than 2 hours, lactate, cort, HLR and BA were all elevated, and generally more so than animals that remained calm in traps for a similar amount of time. Animals that had been trapped and then held for a 3-hour restraint showed similar degrees of physiological alteration as those that showed outward signs of distress. This study demonstrates that trapping may yield physiological disturbances in gopher tortoises, although the intensity of this response is highly variable between individuals and the duration of such alterations remains unknown. Chemical communication is important for mate choice, especially at long distances in fragmented populations. The gopher tortoise is a social species that is threatened in the southeast U.S. due to habitat fragmentation and decline. One consequence of habitat loss is reduced mating opportunities, yet chemical signalling in gopher tortoises is relatively under-studied. Here, we investigated chemoreception of tortoise discrimination of chin secretions, or mental gland (MG) secretions. To assess conspecific recognition of male MG secretions, we conducted two paired-choice experiments: one with a neutral odorant control (NC; distilled water) and one with a pungent odorant control (PC; acetone) vs. male MG secretions. Behaviours were defined a priori, and their durations were quantified relative to treatments. Each sex spent significantly more time with MG secretions vs. acetone control during the PC study (p= 0.001). Each sex also sniffed MG swabs more frequently in both studies (PC study: p=0.0003; NC study: p=0.001). A principal components analysis of behavioural durations from the PC study identified one component with a significant treatment effect performed to MG secretions (p=0.0003), including the behaviours sniffing, head bobbing, biting, and eating near a swab. Our study provides the first chemical-behavioural bioassay of MG secretions from male gopher tortoises, suggesting MG secretions may be a source of pheromones., (Copyright © 2021 Elsevier B.V. Cane toads are highly toxic bufonids invasive in several locations throughout the world. Although physiological changes and effects on native predators for Australian populations have been well documented, Florida populations have received little attention. Cane toads were collected from populations spanning the invaded range in Florida to assess relative toxicity, through measuring morphological changes to parotoid glands, likelihood of secretion, and the marinobufagenin (MBG) content of secretion. We found that residual body indices increased in individuals from higher latitude populations, and relative parotoid gland size increased with increasing toad size. There was no effect of latitude on the allometric relationship between gland size and toad size. We observed an increase in likelihood of secretion by cane toads in the field with increasing latitude. Individuals from southern and northern populations did not vary significantly in the quantity of MBG contained in their secretion. Laboratory-acclimated cane toads receiving injections of epinephrine were more likely to secrete poison with increasing dose, although there was no difference in likelihood of secretion between southern and northern populations. This suggests that differences between populations in the quantities of epinephrine released in the field, due to altered hypothalamic sensitivity upon disturbance, may be responsible for the latitudinal effects on poison secretion. Cane toads (Rhinella marina) were introduced worldwide and have become invasive in multiple locations, representing a major driver of biodiversity loss through competition (food, shelter, territory), predation, and the poisoning of native species. These toads have been used in Australia as a model for studies concerning invasion biology and ecoimmunology, as longer-established (core) and invasion front (edge) populations show altered stress and immune response profiles. Although cane toads were also introduced into the United States in the 1950s, these patterns have yet to be evaluated for the populations spanning Florida. Toads introduced into Florida have dispersed primarily northward along a latitudinal gradient, where they encounter cooler temperatures that may further impact stress and immune differences between core and edge populations. In this study, we sampled cane toads from nine different locations spanning their invasion in Florida. Cane toads from southern populations showed higher plasma bacterial killing ability and natural antibody titers than the toads from the northern populations, indicating they have a better immune surveillance system. Also, southern toads were more responsive to a novel stressor (1 hr restraint), showing a higher increase in corticosterone levels. While the expansion of anthropogenic noise studies in aquatic habitats has produced conservation-based results for a range of taxa, relatively little attention has been paid to the potential impacts on stream fishes. Recent work has shown responses to road noise in single species of stream fish; however, assemblage-wide effects of anthropogenic noise pollution have not yet been investigated. By examining five metrics of disturbance across four ecologically and evolutionarily disparate species of stream fishes, a series of laboratory experiments aimed to describe the effects of and species susceptibility to anthropogenic noise playback. Each species studied represented a unique combination of hearing sensitivity and water column position. Physiological and behavioral metrics were compared across the presence and absence of rail-noise noise playback in four target species. Through repeated subsampling, the temporal dynamics of cortisol secretion in response to noise in two target species were additionally described. Rail-noise playback had no statistically significant effect on blood glucose or water-borne cortisol levels, with the exception of decreased cortisol in noise-exposed largescale stoneroller ( Campostoma oligolepis ). Time-course cortisol experiments revealed rapid secretion and showed minimal effects of noise at most observation points. The presence of noise produced significant changes in ventilation rate and swimming parameters in a portion of the four species observed representing the most conserved responses. Overall, effects of noise were observed in species contrary to what would be hypothesized based on theoretical hearing sensitivity and water column position demonstrating that predicting susceptibility to this type of stressor cannot be accomplished based off these course considerations alone. More importantly, we show that anthropogenic noise can disrupt a variety of behavioral and physiological processes in certain taxa and should be further investigated via measures of fitness in the wild., (© The Author(s) 2020. Decades of work indicate that female birds can control their offspring sex ratios in response to environmental and social cues. In laying hens, hormones administered immediately prior to sex chromosome segregation can exert sex ratio skews, indicating that these hormones may act directly on the germinal disc to influence which sex chromosome is retained in the oocyte and which is discarded into an unfertilizable polar body. We aimed to uncover the gene pathways involved in this process by testing whether treatments with testosterone or corticosterone that were previously shown to influence sex ratios elicit changes in the expression of genes and/or gene pathways involved in the process of meiotic segregation. We injected laying hens with testosterone, corticosterone, or control oil 5h prior to ovulation and collected germinal discs from the F1 preovulatory follicle in each hen 1.5h after injection. We used RNA-sequencing (RNA-seq) followed by DESeq2 and gene set enrichment analyses to identify genes and gene pathways that were differentially expressed between germinal discs of control and hormone-treated hens. Corticosterone treatment triggered downregulation of 13 individual genes, as well as enrichment of gene sets related to meiotic spindle organization and chromosome segregation, and additional gene sets that function in ion transport. Testosterone treatment triggered upregulation of one gene, and enrichment of one gene set that functions in nuclear chromosome segregation. This work indicates that corticosterone can be a potent regulator of meiotic processes and provides potential gene targets on which corticosterone and/or testosterone may act to influence offspring sex ratios in birds., Competing Interests: The authors have declared that no competing interests exist.

Physiological tradeoffs occur in organisms coping with their environments, which are likely to increase as populations reach peripheries of established ranges. Invasive species offer opportunities to study tradeoffs that occur, with many hypotheses focusing on how immune responses vary during dispersal. The cane toad (Rhinella marina) is a well-known invasive species. Populations near the expanding edge of the Australian invasion have altered immune responses compared to toads from longer-established core populations, although this has not been well-documented for Florida populations. In this study, cane toads from a northern edge [New Port Richey (NPR)] and southern core (Miami) population in Florida were collected and injected with lipopolysaccharide (LPS) to compare immune responses. Core population individuals injected with LPS showed greater metabolic increases compared to their baseline rates that were higher compared to those from the edge population. In addition, LPS-injected core individuals had different circulating leukocyte profiles compared to saline-injected cane toads while edge individuals did not. There was a significant interaction between plasma bacteria-killing capability (BKA) and treatment, such that BKA decreased with time in saline compared to LPS-injected individuals, and saline-injected toads from the edge population had lower BKA compared to LPS-injected edge toads at 20 h post-injection. There was also a significant interaction between location and time on circulating corticosterone (CORT) levels following injections with saline or LPS, with CORT decreasing more with time in core population toads. The differential CORT response indicates that differential stress responses contribute to the tradeoffs observed with immunity and dispersal.

Upper respiratory tract disease (URTD) in North American tortoises (Gopherus) has been the focus of numerous laboratory and field investigations, yet the prevalence and importance of this disease remains unclear across many tortoise populations. Furthermore, much research has been focused on understanding diagnostic biomarkers of two known agents of URTD, Mycoplasma agassizii and Mycoplasma testudineum, yet the reliability and importance of these diagnostic biomarkers across populations is unclear. Gopher Tortoises (Gopherus polyphemus) have experienced significant declines and are currently protected range wide. Geographically, Alabama represents an important connection for Gopher Tortoise populations between the core and periphery of this species' distribution. Herein, we systematically sampled 197 Gopher Tortoises for URTD across seven sites in south-central and south-eastern Alabama. Plasma samples were assayed for antibodies to M. agassizii and M. testudineum; nasal lavage samples were assayed for the presence of viable pathogens as well as pathogen DNA. Lastly, animals were scored for the presence of external symptoms and nasal scarring consistent with URTD. External symptoms of URTD were present in G. polyphemus in all sites sampled in Alabama. There was no relationship between active symptoms of URTD and Mycoplasma antibodies, however the presence of URTD nasal scarring was positively related to M. agassizii antibodies (P = 0.032). For a single site that was sampled in three sequential years, seroprevalence to M. agassizii significantly varied among years (P < 0.0001). Mycoplasma agassizii DNA was isolated from four of the seven sites using quantitative PCR, yet none of the samples were culture positive for either of the pathogens. An analysis of disease status and condition indicated that there was a significant, positive relationship between the severity of URTD symptoms and relative body mass (P < 0.05). This study highlights the need for continued monitoring of disease in wild populations. Specifically, focus must be placed on identifying other likely pathogens and relevant biomarkers that may be important drivers of URTD in North American tortoises. When organisms possess chemical defenses, their predators may eventually evolve resistance to their toxins. Eastern indigo snakes ( Drymarchon couperi ; EIS) prey on pitvipers and are suspected to possess physiological resistance to their venom. In this study, we formally investigated this hypothesis using microassays that measured the ability of EIS blood sera to inhibit (A) hemolytic and (B) snake venom metalloproteinase (SVMP) activity of copperhead ( Agkistrodon contortrix ) venom. To serve as controls, we also tested the inhibitory ability of sera from house mice ( Mus musculus ) and checkered gartersnakes ( Thamnophis marcianus ), a snake that does not feed on pitvipers. Sera from both EIS and gartersnakes inhibited over 60% of SVMP activity, while only EIS sera also inhibited venom hemolytic activity (78%). Our results demonstrate that EIS serum is indeed capable of inhibiting two of the primary classes of toxins found in copperhead venom, providing the first empirical evidence suggesting that EIS possess physiological resistance to venom upon injection. Because we documented resistance to hemolytic components of pitviper venom within EIS but not gartersnakes, we speculate this resistance may be driven by selection from feeding on pitvipers while resistance to SVMP may be relatively widespread among snakes., Competing Interests: Competing interestsThe authors declare no competing or financial interests., (© 2019. As infectious diseases in ectothermic vertebrates increasingly threaten wild populations, understanding how host immune systems are affected by the environment is key to understanding the process of infection. In this study, we investigated how temperature change and simulated bacterial infection (via lipopolysaccharide [LPS] injection) interacted to regulate innate immunity, as measured by bactericidal ability (BA), phagocytosis rate, and heterophil:lymphocyte ratio (HLR) in common musk turtles (Sternotherus odoratus). We found that LPS stimulated an acute immune response, as measured by an increase in BA, phagocytosis rate, and HLR. When exposed to a 5 or 10°C temperature change for 48 hr, turtles rapidly acclimated to the new temperature by adjusting their immune output. This acclimation was compensatory as seen by elevated rates of immune output in colder animals and decreased rates of immune output in warmer animals. These results indicate that while temperature change may be a constraint on some animals, S. odoratus have the ability to rapidly adjust immunity to match environmental thermal demand. This rapid ability to adjust immunity may be related to the broad geographic distribution of musk turtles. The cane toad (Rhinella marina) is an invasive amphibian in several parts of the world. Much of the research performed on assessing the dispersal potential of invasive species has focused immunity. Invaders are predicted to rely less on pro-inflammatory immunity, allowing them to allocate energy to dispersal. Elevated stress may play a role in regulation of immune responses used by invasive species. RNA sequencing of spleen tissue from cane toads subjected to an acute LPS challenge revealed genes coding for cytokines involved in typical innate responses such as phagocytic cell recruitment, extravasation, inflammation, and lymphocyte differentiation were significantly upregulated, while toads receiving transdermal application of corticosterone in addition to an LPS injection showed downregulation of genes involved with cell mediated immunity. These results indicate hormonal changes associated with acute stress may alter investment into mounting cell-mediated or humoral responses while allowing for prolonged phagocytic innate responses in this invasive species., (Copyright © 2018 Elsevier Ltd. During geographic range expansion, populations of non-indigenous species at the invasion front may benefit from directing resources away from immune defense. To test this hypothesis, we investigated the strength of two innate immune components in populations of invasive Cuban Treefrogs (Osteopilus septentrionalis) in a long-colonized area (core region) and at the invasion front (leading-edge region). First, we compared the region-specific metabolic response of frogs injected with an endotoxin that induces systemic inflammation (lipopolysaccharide, LPS) to sham-injected control frogs pooled from both regions. Males and females were analyzed independently because we detected a sex-related difference in mass-independent metabolism of control frogs, with males exhibiting a significantly higher metabolic rate (F 1, 21  = 29.02, P < 0.001) than females. We observed a significantly higher metabolic rate in LPS-injected core frogs compared with control frogs for both males (P = 0.041) and females (P = 0.007). Conversely, in leading-edge populations, there was no significant difference in the metabolic rate of LPS-injected and control frogs (males, P  = 0.195; females, P  = 0.132). Second, we directly compared bacterial killing ability of frog blood plasma between regions. Bactericidal ability of plasma was significantly greater in frogs from the core region in comparison with those at the leading edge (F 1, 26   = 28.67, P < 0.001). For both immune components that we examined, populations from the core exhibited stronger immune responses. Complement-an immune protein cascade involved in pathogen lysis-was discovered as a temperature-labile component of vertebrate plasma, yet since that time the thermal performance of complement has not received much attention from a comparative or ecological perspective. We investigated two thermal hypotheses involving the complement system of the cottonmouth snake (Agkistrodon piscivorus). We tested whether complement performance would conform to optimal thermal reaction norms commonly observed in ectotherm ecophysiological studies, predicting that complement efficiency would be maximal at or near the cottonmouth's field body temperatures. We also tested thermal acclimatization of complement performance, by comparing temperature/performance curves from samples collected in three different seasons. Complement efficiency exhibited the same significant positive correlation with temperature in all three seasons. Organisms possess a range of thermoregulatory strategies that may vary in response to sickness, thereby driving important life-history consequences. Because the immune system is vital to maintaining organism function, understanding the suite of immune responses to infection indicates basic costs and benefits of physiological strategies. Here, we assessed consequences of thermoregulation and seasonality on immune function in both immunologically stimulated and nonstimulated gopher tortoises (Gopherus polyphemus). An ectothermic vertebrate was used as an experimental model because the effects of thermoregulation on immunity remain understudied and are of increasing importance in light of anthropogenic alterations to thermal environments. We found that G. polyphemus increased body temperature (T b ) at 1 h after injection with lipopolysaccharide (LPS) when compared with saline controls (P = 0.04), consistent with behavioral fever. LPS increased plasma bactericidal ability (BA; P = 0.006), reduced plasma iron concentration (P = 0.041), and increased heterophil∶lymphocyte ratios (P < 0.001). In nonstimulated animals, thermoregulatory strategy had a strong effect on innate immunity, which demonstrated that individuals have the ability to facultatively adjust immune function when infection burden is low; this relationship was not present in LPS-injected animals, which suggested that animals stimulated with LPS maximize bactericidal ability independently of temperature. Seasonal acclimation state did not influence responses to LPS, although baseline plasma iron was significantly lower in animals acclimated to winter. These results support that a trade-off exists between immunity and other conflicting physiological interests. Moreover, these results clearly demonstrate the ability of individuals to modulate immune function as a direct result of thermoregulatory decisions.

Disease outbreaks are of increasing importance to ectothermic vertebrates as one of numerous results of global change. Anthropogenic climate change is predicted to increase climatic instability, thereby altering natural thermal environments. In this study, we evaluated the direct effects of rapid temperature change on immunity in Gopher Tortoises (Gopherus polyphemus). Specifically, we tested the lag hypothesis, which predicts significant misalignment of optimal and realized immunity when temperature rapidly changes. We assayed constitutive innate immunity, B-cell humoral responses, and heterophil: lymphocyte ratios in response to rapid temperature changes corresponding to realistic changes in body temperature between winter and summer. We found that during summer, rapid temperature reduction caused a series of changes in immunity, including reduced bactericidal ability (P = 0.002), reduced humoral response (P < 0.0001), and increased heterophil:lymphocyte ratios (P < 0.0001). During winter, we found that a temperature increase provided no benefit to immunity. Specifically, there was no increase in bactericidal ability as was predicted by the lag hypothesis. In winter, humoral responses were significantly reduced as a result of rapid warming (P = 0.011) and the rapid warming caused a significant reduction in heterophil:lymphocyte ratios (P < 0.0001). Independent of temperature, we found a significant acclimation effect of winter relative to summer conditions in humoral response (P < 0.001), which showed an overall increase in this parameter during winter. Studies have suggested a role for natural seasonal change to drive patterns of disease, especially within ectothermic vertebrates. In light of recent climate change, it is important to understand baseline disease resistance in a seasonal context to further understand the role that changes in seasonal weather patterns may have in increasing disease frequency. Herein we found support for the seasonal acclimation hypothesis in Gopherus polyphemus (gopher tortoise), which indicated that natural seasonal variation causes differences in baseline immune function across seasonal acclimation states. We found that an innate immune parameter, bactericidal ability (BA), was significantly elevated in the summer (P < 0.00001). Circulating leukocyte profiles varied significantly among seasons, with heterophils and monocytes increased (P = 0.00019 and P = 0.0001, respectively) and lymphocytes decreased (P < 0.00001) during winter. We assayed baseline glucocorticoid concentration (e.g., corticosterone [CORT]) across seasons and sampling conditions to test whether CORT drove the seasonal pattern in immunological acclimation. CORT was significantly lowest during winter and in animals temporarily maintained in seminatural conditions. These changes in CORT occurred independently of the immunological adjustments, suggesting that the seasonal pattern of immunity was not mediated by CORT secretion. The reduction in lymphocytes and BA and also BA during winter suggest that seasonal acclimation is likely a restraint on energetic output when temperature is low and physiological performance is thermally constrained. While these parameters were reduced in winter, the increase in heterophils and monocytes may indicate a compensatory immune adjustment to increase the number of innate phagocytic cells.

Noise pollution from anthropogenic sources is an increasingly problematic challenge faced by many taxa, including fishes. Recent studies demonstrate that road traffic noise propagates effectively from bridge crossings into surrounding freshwater ecosystems; yet, its effect on the stress response and auditory function of freshwater stream fishes is unexamined. The blacktail shiner (Cyprinella venusta) was used as a model to investigate the degree to which traffic noise impacts stress and hearing in exposed fishes. Fish were exposed to an underwater recording of traffic noise played at approximately 140 dB re 1 μPa. Waterborne cortisol samples were collected and quantified using enzyme immunoassay (EIA). Auditory thresholds were assessed in control and traffic exposed groups by measuring auditory evoked potentials (AEPs). After acute exposure to traffic noise, fish exhibited a significant elevation in cortisol levels. Individuals exposed to 2 hours of traffic noise playback had elevated hearing thresholds at 300 and 400 Hz, corresponding to the most sensitive bandwidth for this species.

Rabies in bats is considered enzootic throughout the New World, but few comparative data are available for most countries in the region. As part of a larger pathogen detection program, enhanced bat rabies surveillance was conducted in Guatemala, between 2009 and 2011. A total of 672 bats of 31 species were sampled and tested for rabies. The prevalence of rabies virus (RABV) detection among all collected bats was low (0.3%). Viral antigens were detected and infectious virus was isolated from the brains of two common vampire bats (Desmodus rotundus). RABV was also isolated from oral swabs, lungs and kidneys of both bats, whereas viral RNA was detected in all of the tissues examined by hemi-nested RT-PCR except for the liver of one bat. Sequencing of the nucleoprotein gene showed that both viruses were 100% identical, whereas sequencing of the glycoprotein gene revealed one non-synonymous substitution (302T,S). The two vampire bat RABV isolates in this study were phylogenetically related to viruses associated with vampire bats in the eastern states of Mexico and El Salvador. Additionally, 7% of sera collected from 398 bats demonstrated RABV neutralizing antibody. The proportion of seropositive bats varied significantly across trophic guilds, suggestive of complex intraspecific compartmentalization of RABV perpetuation.

Testosterone has been implicated as a developmental mechanism involved in the organization and expression of sexually dimorphic traits, such as plumage coloration, in birds. Although research findings relating testosterone levels to plumage expression is equivocal, few studies have investigated how testosterone may influence the expression of structurally based plumage coloration. Here, we use experimental and correlational evidence to test the hypothesis that testosterone influences the development and maintenance of structurally based plumage coloration in a wild-breeding population of eastern bluebirds (Sialia sialis). First, we experimentally manipulated yolk testosterone and measured the effect on the development of plumage coloration of nestlings. Second, we implanted juvenile bluebirds with testosterone and measured the effect on nestling growth, body condition, and plumage coloration of nestlings. Third, we measured covariation between circulating testosterone and plumage coloration of breeding males. Yolk testosterone injections had no significant effect on nestling plumage coloration. Testosterone implantation, however, caused a reduction in plumage brightness, elevated corticosterone, and slower growth in nestlings. Finally, in breeding adult males we found no significant relationship between structural coloration and testosterone; however, males with higher testosterone levels exhibited duller chestnut (melanin-based) plumage. Our observations lead us to reject the hypothesis that testosterone increases structural plumage coloration in male eastern bluebirds.

Chytridiomycosis, a disease caused by Batrachochytrium dendrobatidis, has contributed to worldwide amphibian population declines; however, the pathogenesis of this disease is still somewhat unclear. Previous studies suggest that infection disrupts cutaneous sodium transport, which leads to hyponatremia and cardiac failure. However, infection is also correlated with unexplained effects on appetite, skin shedding, and white blood cell profiles. Glucocorticoid hormones may be the biochemical connection between these disparate effects, because they regulate ion homeostasis and can also influence appetite, skin shedding, and white blood cells. During a laboratory outbreak of B. dendrobatidis in Australian Green Tree Frogs, Litoria caerulea, we compared frogs showing clinical signs of chytridiomycosis to infected frogs showing no signs of disease and determined that diseased frogs had elevated baseline corticosterone, decreased plasma sodium and potassium, and altered WBC profiles. Diseased frogs also showed evidence of poorer body condition and elevated metabolic rates compared with frogs showing no signs of disease. Prior to displaying signs of disease, we also observed changes in appetite, body mass, and the presence of shed skin associated with infected but not yet diseased frogs. Collectively, these results suggest that elevated baseline corticosterone is associated with chytridiomycosis and correlates with some of the deleterious effects observed during disease development.

The possible trade-off between the roles of glucocorticoids as facilitators of energy substrate mobilization and neural inhibitors of sexual behavior during breeding season is under debate. We studied the relationship between calling and territorial behavior with plasma levels of corticosterone (CORT) and plasma levels of testosterone (T) across the breeding season of Hypsiboas faber, a large and territorial Neotropical treefrog. We investigated these relationships through focal observations of males calling naturally, followed by blood sampling for hormonal radioimmunoassay. We additionally used an experimental approach, which consisted of broadcasting recorded advertisement calls for 10 min to simulate an invasion in the territory of the focal subjects, followed by behavioral observation and blood sampling for hormonal radioimmunoassay. Results showed a pattern of co-variation between CORT and T across the breeding season. Furthermore, individual variation in CORT and T was related to different aspects of behavior: individuals with higher CORT showed higher calling rates, and individuals with higher steroid levels, mainly T, showed higher responsivity to social stimulation by other males in the chorus. Squamates (lizards and snakes) have independently evolved viviparity over 100 times, and exhibit a wide range of maternal investment in developing embryos from the extremes of lecithotrophic oviparity to matrotrophic viviparity. This group therefore provides excellent comparative opportunities for studying endocrine and immune involvement during pregnancy, and their possible interactions. We studied the cottonmouth (Agkistrodon piscivorus), since they exhibit limited placentation (e.g., ovoviviparity), allowing comparison with squamate species hypothesized to require considerable maternal immune modulation due to the presence of a more extensive placental connection. Furthermore, the cottonmouth's biennial reproductive cycle provides an opportunity for simultaneously comparing pregnant and non-pregnant females in the wild. We document significantly elevated concentrations of progesterone (P4) and significantly lower concentrations of estradiol (E2) in pregnant females relative to non-pregnant females. Pregnant females had lower plasma bacteria lysis capacity relative to non-pregnant females. This functional measure of innate immunity is a proxy for complement performance, and we also determined significant correlations between P4 and decreased complement performance in pregnant females. These findings are consistent with studies that have determined P4's role in complement modulation during pregnancy in mammals, and thus this study joins a growing number of studies that have demonstrated convergent and/or conserved physiological mechanisms regulating viviparous reproduction in vertebrates., (Copyright © 2011. Bats are known reservoirs for numerous emerging infectious diseases, occupy unique ecological niches, and occur globally except for Antarctica. Given their impact on human and agricultural health, it is critical to understand the mechanisms underlying immunocompetence in this reservoir host. To date, few studies have examined immune function in the Order Chiroptera, particularly among natural colonies of bats. The phytohemagglutinin (PHA) skin test has been widely used to measure delayed-type cellular immune response in a wide variety of vertebrates, and has been routinely employed in immunoecological studies. Although this test is frequently described as a measure of T cell proliferation, recent studies indicate it may represent a combination of immune responses. In mammals, the immune response is differentially, temporally and spatially regulated, therefore, we characterized the infiltrating leukocyte response to the PHA skin test in bats by examining a time-series of histological sections from PHA and saline injection areas in 41 Brazilian free-tailed bats (Tadarida brasiliensis). Results suggest that bats exhibit diverse leukocyte traffic within 6 h, and up to 24 h following subcutaneous PHA injection. There was a significant presence of lymphocytes and neutrophils, as well as eosinophils, basophils, and macrophages observed in the PHA-injected tissues, compared with saline-injected control tissues. We observed a highly significant negative correlation between the number of lymphocytes and neutrophils in PHA-injected tissue, with peak lymphocyte response at 12 h, and peak neutrophil response at 24 h post-injection. These results indicate substantial variation in the immune response of individuals, and may aid our understanding of disease emergence in natural populations of bats.

Carotenoid pigments produce yellow, orange, and red integumentary color displays that can serve as reliable signals of health and condition. In many birds and fish, individuals gain competitive or mating advantages by ingesting and utilizing large quantities of carotenoid pigments. Carotenoid pigments serve as antioxidants, performing important functions as free-radical scavengers. The beneficial effects of carotenoid pigments are well documented, but rarely have researchers considered potential detrimental effects of high-level accumulation of carotenoids. We maintained American goldfinches (Carduelis tristis) on high- or low-carotenoid diets through molt and tested for damage to the liver and skeletal muscle. High intake of carotenoids had no measurable effect on liver enzymes but caused an increase in creatine kinase, an indicator of skeletal muscle breakdown, and a reduction in vertical flight performance, a measure of skeletal muscle integrity. The detrimental effects of high-level carotenoid accumulation were approximately equivalent to the negative effects of removing carotenoids from the diet. The adverse effects observed in this study have important implications for theories of the function and evolution of colorful plumage.

Graptemys flavimaculata, the yellow-blotched map turtle, is a long-lived, threatened, species, endemic to the Pascagoula River drainage in Mississippi. During the 1980s, one branch of the drainage (i.e. the Leaf River) was impacted by effluent from a wood pulp processing plant known to contain endocrine disrupters. A decade later, we examined seasonal reproductive parameters (i.e. monthly plasma estradiol-17beta (E(2)), testosterone (T), vitellogenin (VTG) and follicular development) in adult female turtles from historically polluted and reference sites in the drainage to determine if legacy exposure to pollution impacts reproduction . We found no seasonal patterns in E(2) or T and these patterns did not differ between sites. However, E(2) differed significantly among ovarian stages for the reference, but not pollutant exposed females. A significantly greater percentage of reference site females were able to produce a second clutch than females from the historically polluted site (50% and 17%). Additionally, there was a significant positive correlation between E(2) with VTG levels for reference, but not pollutant exposed females. Body and yolk tissue contaminant analysis indicated that exposure to pollutants is presently minimal and unlikely the cause of the reproductive differences observed between sites; instead, differences are potentially due to exposure history.

Bats have recently been implicated as reservoirs of important emerging diseases. However, few studies have examined immune responses in bats, and even fewer have evaluated these responses in an ecological context. We examined aspects of both innate and adaptive immune response in adult female Brazilian free-tailed bats (Tadarida brasiliensis) at four maternity roosts (two natural caves and two human-made bridges) in south-central Texas. Immune measurements included in vitro bactericidal ability of whole blood and in vivo T cell mediated response to mitogenic challenge. Bactericidal activity in T. brasiliensis varied with roosting ecology, but appears to be sensitive to colony-level effects. Blood from females living at one cave had significantly lower bactericidal ability than blood from females at three other sites. T cell mediated response in this species was associated with variation in roost ecology, with females from two caves having greater responses than females from two bridges. T cell mediated response and bactericidal activity were negatively correlated with one another within individuals that were tested for both. Variation in immunological response of T. brasiliensis is important for understanding the influence of the environment on the frequency and distribution of immunologically competent individuals and for understanding disease-host dynamics in this and other colonial species.

Coal combustion residues (CCRs) are documented to negatively impact oral morphology, growth, and development in larval amphibians. It is currently unclear what physiological mechanisms may mediate these effects. Corticosterone, a glucocorticoid hormone, is a likely mediator because when administered exogenously it, like CCRs, also negatively influences oral morphology, growth, and development in larval amphibians. In an attempt to identify if corticosterone mediates these effects, we raised larval Southern Leopard Frogs, Rana sphenocephala, on either sand or CCR substrate and documented effects of sediment type on whole body corticosterone, oral morphology, and time to and mass at key metamorphic stages. Coal combustion residue treated tadpoles contained significantly more corticosterone than controls throughout metamorphosis. However, significantly more oral abnormalities occurred early in metamorphosis when differences in corticosterone levels between treatments were minimal. Overall, CCR-treated tadpoles took significantly more time to transition between key stages and gained less mass between stages than controls, but these differences between treatments decreased during later stages when corticosterone differences between treatments were greatest. Our results suggest endogenous increase in corticosterone content and its influence on oral morphology, growth and development is more complex than previously thought.

Avian egg yolks are made up of complex mixtures of physiologically relevant substances. Androgens, in particular, accumulate in yolks in variable amounts based on social and environmental conditions experienced by laying females, and using experimental elevations of yolk androgen content, researchers have unveiled potent physiological and behavioral effects on offspring. These patterns and effects are exciting in an adaptive context, as the transfer of endogenously-produced substances such as androgens to egg yolks may allow fine manipulation of offspring phenotype to maximize reproductive success. However, to gain an in-depth understanding of how yolk androgens function in an adaptive context, we must first understand the complex entanglement of physiological and endocrine interrelationships that change after exposure to yolk androgens. Here, we take a comparative approach towards a discussion of how yolk androgens can simultaneously affect multiple body systems within developing birds, ultimately resulting in the large-scale phenotypic endpoints that may represent adaptive consequences of exposure to elevated levels of yolk androgens.

Batrachochytrium dendrobatidis, an aquatic fungus, has been linked to recent amphibian population declines. Few surveys have assessed B. dendrobatidis infections in areas where the disease is suggested to be less virulent and population declines have not been observed, such as southeastern North America. Although adult Rana catesbeiana and Rana sphenocephala from the Savannah River Site, South Carolina collected in 1979 and 1982 were identified as having B. dendrobatidis, it is unknown whether the fungus is currently present at the site or if susceptibility to infection varies among species or wetlands with different histories of environmental contamination. From 15 May through 15 August 2004, we collected R. catesbeiana and R. sphenocephala tadpoles from three wetlands with differing contamination histories on the Savannah River Site, South Carolina. We found B. dendrobatidis in only one of the wetlands we surveyed. Batrachochytrium dendrobatidis infection was identified in 64% of the R. catesbeiana tadpoles sampled and histologically assessed (n=50) from a wetland contaminated with mercury, copper, and zinc. No R. sphenocephala tadpoles from this site (n=50) were infected. In combination with a recently published report, our data suggest that B. dendrobatidis has been present at the Savannah River Site for over 25 yr but has not caused any apparent population declines. This time period is similar to the known presence of 30 yr of B. dendrobatidis in northeastern North America. Our data suggest that R. sphenocephala larvae might be resistant to infection, even when occupying the same wetland as the infected R. catesbeiana. Our survey did not clarify the effects of environmental contamination on infection severity, but our study stresses the importance of additional field surveys to document how this pathogen is affecting amphibians globally.

Corticosteroid-binding globulin (CBG) is a plasma protein that binds corticosterone and may regulate access of hormone to tissues. The role of CBG during a stress response is not clear. At least two hypotheses have been proposed: 1) CBG levels may increase in response to a stressor, thereby decreasing the amount of circulating free corticosterone, or 2) CBG levels may decline, making corticosterone available for its role in increased metabolic needs during stress. In this study, southern toads, Bufo terrestris, were exposed to a chronic pollutant (coal-combustion-waste), to determine changes in CBG and free corticosterone levels. Since toads exposed to chronic pollutants in previous studies did not exhibit the predicted changes in metabolic rate and mass, but did experience a significant elevation in total corticosterone, we hypothesized that CBG would likewise increase and thus, mitigate the effects of a chronic (i.e. 2 months) pollutant stressor. To conduct this study, we first characterized the properties of CBG in southern toads. Toad CBG has a K(d)=20.6+/-1.0 nM and a B(max)=332.2+/-5.1 nmol/L plasma. The rank order potencies for steroid inhibition of tritiated corticosterone are: dihydrotestosterone > corticosterone >> progesterone=testosterone >>> estrogen=dexamethasone. After characterization, we monitored the changes in CBG, total corticosterone, and free corticosterone in male toads that were exposed to either coal-combustion-waste or control conditions. CBG increased in all groups throughout the experiment. Total corticosterone, on the other hand, was only significantly elevated at four weeks of exposure to coal-combustion-waste. The increase in CBG did not parallel the increase in total corticosterone; as a result, free corticosterone levels were not buffered by CBG, but showed a peak at four weeks similar to total corticosterone. This finding indicates that, in this species, CBG may not provide a protective mechanism during long-term pollution exposure.

The manipulation of egg content is one of the few ways by which female birds can alter offspring quality before hatch. Lipid-soluble vitamins and carotenoids are potent antioxidants. Female birds deposit these antioxidants into eggs in variable amounts according to environmental and social conditions, and the quantities deposited into eggs can have effects on offspring health and immunological condition. Allocation theory posits that females will alter the distribution of resources according to mate quality, sometimes allocating resources according to the differential allocation hypothesis (DAH), investing more in offspring sired by better-quality males, and other times allocating resources according to a compensatory strategy, enhancing the quality of offspring sired by lower-quality males. It is unknown, however, whether antioxidants are deposited into eggs according to the DAH or a compensatory strategy. We examined deposition patterns of yolk antioxidants (including vitamin E and three carotenoids) in relation to laying order, mate attractiveness, female condition, and yolk androgen content in the house finch (Carpodacus mexicanus). Female house finches deposited significantly more total antioxidants into eggs sired by less attractive males. Additionally, yolk antioxidant content was significantly positively correlated with female condition, which suggests a cost associated with the deposition of antioxidants into eggs. Finally, concentrations of antioxidants in egg yolks were positively correlated with total yolk androgen content. We suggest that yolk antioxidants are deposited according to a compensatory deposition strategy, enabling females to improve the quality of young produced with less attractive males. Additionally, yolk antioxidants may act to counter some of the detrimental effects associated with high levels of yolk androgens in eggs and, thus, may exert a complementary effect to yolk androgens.

More than 50% of the electricity in the United States is produced by coal-burning power plants. The byproduct of coal-burning plants is coal fly ash, which contains increased concentrations of trace metals and is disposed of in collection basins. Southern toads (Bufo terrestris) frequently use these basins for reproduction. Male toads were collected in spring 2001 and 2002 from an ash basin and a reference site and divided into four groups: toads collected at the control site and maintained on (1) control substrate and food or (2) ash and contaminated food and toads collected at the ash site and maintained in (3) control or (4) ash conditions. Blood was collected periodically during 5 months to determine testosterone and corticosterone concentrations. Reference to ash toads exhibited a significant, transient increase in corticosterone at 4 weeks, but neither corticosterone nor testosterone continued to increase beyond this time. In contrast, toads caught and maintained on ash did not exhibit increased corticosterone. Testosterone in these toads appeared to be unrelated to ash exposure. This unexpected lack of a corticosterone response and no effect on testosterone suggests that toads chronically exposed to trace metals can acclimate to a polluted environment, but they may still experience subtle long-term consequences.

Overlap in growth of offspring should constrain the opportunity for sex-biased maternal effects, yet sex-specific allocation of maternal resources among simultaneously growing ova is often observed in vertebrates. In birds, such allocation can be accomplished either by temporal clustering of ova that become the same sex, resulting in sex-biased egg-laying order, or by follicle-specific delivery of maternal resources. Two house finch populations at the northern and southern boundaries of the species range have opposite ovulation sequences of male and female eggs, and thus, in the absence of sex differences in ova growth or sex-specific maternal strategies, would be expected to have opposite sex-specific accumulation of maternal products. We found that the populations had strong and similar gradients of steroid distribution in relation to ovulation order, whereas distribution of carotenoids and vitamins correlated with each follicle's accumulation of steroids. In both populations, temporal bias in production of sons and daughters within a clutch enabled strongly sex-specific acquisition of maternal products, and oocytes of the same sex were highly interdependent in their accumulation of steroids. Moreover, in nests where the sex-bias in relation to ovulation order deviated from population-specific patterns, eggs had highly distinct concentrations of steroids, carotenoids and vitamins. These results and previous findings of sex-specific yolk partitioning among oocytes suggest that oocytes that become males and females are temporally or spatially clustered during their ovarian growth. We discuss the implication of these findings for the evolution of sex-specific maternal resource allocation.

Southern toads (Bufo terrestris) are found in coal fly ash collection basins associated with coal-burning electrical power plants. These basins contain large amounts of trace metals and organisms found in these basins are known to accumulate large quantities of metals. Studies on a variety of organisms exposed to trace metals found that they experience a significant increase in standard metabolic rate. We experimentally exposed southern toads to metal-contaminated sediment and food and measured changes in standard and exercise metabolic rates as well as changes in body, liver and muscle mass, blood glucose, and corticosterone. We found that toads exposed to trace metal contamination gained significantly less mass (18.3 %) than control toads (31.3%) when food was limited and experienced significantly decreased RQ after exercise. However, contaminated toads did not experience changes in standard (x control = 0.114 +/- 0.016 mL O2 g(-1) h(-1); x (ash)=0.109 +/- 0.013 mL O2 g(-1) h(-1), p = 0.08) or exercise metabolic rates (x control = 0.53 +/- 0.06 mL O2 g(-1) h(-1); x ash =0.44 +/- 0.04 mL O2 g(-1) h(-1), p = 0.47) plasma glucose levels (p = 0.6), and hepatic or muscle percentage indices (p > 0.2 in all cases) whether food was limited or not.

In the female red-sided garter snake (Thamnophis sirtalis parietalis), the loss of receptivity following intromission during mating can be prevented by injection of a local anesthetic (tetracaine) in the cloacal region prior to courtship and mating. Females that were courted and then mated had significantly higher uptake of radio-labeled [14C]2-fluoro-2-deoxyglucose (2-DG) in the preoptic area (25%) and significantly lower uptake in the ventromedial hypothalamus (-20%) compared with females that were courted but not mated. Tetracaine-treated females had accumulation patterns similar to courted but unmated females and to females exposed only to other females. These results suggest that in the female red-sided garter snake, sensory input from the cloaca during mating alters patterns of metabolism in those brain areas most often associated with female sexual behavior.
