Local livestock breeds are pivotal to ensure sustainable agriculture and represent a real genetic resource in the context of genetic variability reduction. This study aimed at broadening the knowledge about hematological values in female Verzaschese and Camosciata delle Alpi goats (an Italian local and a cosmopolitan goat breed, respectively) and investigating factors affecting them (i.e., breed, age, season). Blood samples were collected monthly from 34 Verzaschese and 37 Camosciata delle Alpi female goats kept under the same semi-extensive farming system for a whole year. The main hematological parameters were evaluated, and descriptive as well as inferential statistical analyses were performed. Reference intervals for complete blood cell count were established and several inter-breed differences were found. Traditional health care practices and ethnoveterinary medicine are still common in camels. This study aimed at describing camel health issues reported by caretakers including their management and treatment practices. Associations between the health issues and caretakers’ background, management practices, and facilities were also investigated. Camel caretakers (n=49) at a permanent market in Qatar were interviewed and data related to the camel pens (e.g. presence of shelters, water, and feeding points) collected. During the interview, questions sought to elicit the following information; i) caretaker’s background (age, level of education, experience, and how they identify when a camel is sick), ii) caretaker’s management practices (feeding, watering, and health care practices), iii) caretaker’s description of the last health problem (camel details, recalled diagnosis/clinical signs, possible cause, treatment, and outcome). The majority of the caretakers were middle-aged (31-50 years old) and they used to identify a sick camel mainly observing its behavioural modifications (p < 0.001). Treatment for endo- and ectoparasites were usually administered by caretakers, health checks routinely performed by veterinarians, while vaccinations were not conducted in most of the pens (p < 0.001). A total of 38 health problems were described and the high environmental temperature was the most reported possible cause (43.2%; p < 0.001). Among the recalled health problems, sunstrokes were the most common (11/38; p = 0.046), followed by fever (9/38). Sunstroke was more likely to affect males (OR: 5.04; CI95%: 1.87-23.38; p = 0.039), while fever younger animals (OR: 0.68, CI95%: 0.47-1.00; p = 0.050). Sunstrokes were more likely managed by non-veterinarians (OR: 4.75; CI95%: 1.05-21.34; p = 0.042). The majority of the cases were early identified and had a full recovery, but 11/38 camels died. Interestingly, drugs were the most common treatment (73.7%, p < 0.001), but often bought and administered directly by caretakers. Duty of care, infectious disease prevention, early and appropriate treatments could be recommended to guarantee the principle of good health and consequently animal welfare. Education on how to identify a sick camel and how to manage it should be also promoted.

Inflammatory bowel diseases (IBD) are characterized by inflammatory conditions of the intestine. Probiotic bacteria (PB) can have beneficial effects in several gastrointestinal disorders. The objectives of this study were: (i) to provide an acute experimental IBD model induced by 2,4,6-trinitrobenzenesulfonic acid (TNBS) in CD-1 mice, and (ii) to assess the preventive effects of Citogenex (Lactobacillus casei and Bifidobacterum lactis) supplementation on intestinal tissues and microbiota. Mice were inoculated intrarectally with saline, ethanol or different TNBS solutions. 1%TNBS induced clinical signs of colitis (P less than 0.01) and histological damage (P less than 0.01). Based on these results, mice were pre-treated with Citogenex or saline for 1, 2 or 3 weeks before 1%TNBS treatment. Probiotic pre-treatment determined a reduction of clinical signs (P less than 0.05), histological alterations of colitis (P less than 0.05) and increased beneficial bacteria (P less than 0.05). This study confirms that TNBS-induced colitis in CD-1 mice is useful for studying the mechanisms involved in IBD pathogenesis, and pre-treatment with Citogenex prevents the intestinal damage induced by TNBS.

Leptin regulates body weight and several physiological processes including reproduction. We evaluated the circulating levels of leptin in pregnant and non-pregnant bitches as well as their correlation with body weight, food intake and number of foetuses. Infections of the genital tract can perturb the fertility in humans and animals. Pathogen recognition and activation of innate immunity onset through the pattern recognition receptor activation, such as Toll-like receptor 4 (TLR4), leading to the production of proinflammatory cytokines and mediators. TLR4 is expressed both on leukocytes and nonimmune cells. Rabbit TLR4 shows great similarity to its human counterpart. Moreover, the TLR4 signalling pathway could be modulated by long-chain polyunsaturated fatty acids (LC-PUFA). The objectives of this study were (i) to determine the expression levels of TLR4 and proinflammatory cytokines in the reproductive hypothalamic-gonadal axis of the male rabbit and (ii) to evaluate if the n-3 PUFA-enriched diets can modify their expression levels in the tissues and LC-PUFA profiles in seminal plasma. Fifteen rabbit bucks (n=5/experimental group) were fed with different diets: commercial standard (group C), rich in extruded linseed (10%, group L), and in fish oil (3%, group FO) for 110 days. TLR4, TNF-α, and IL-1βmRNA were ubiquitously expressed throughout the hypothalamic-gonadal axis. However, TLR4 mRNA expression was lower in the hypothalamus than the epididymis (P<0.01), seminal vesicles (P<0.01), and pituitary gland (P<0.05). Dietary enrichment in PUFA did not modify the gene expression profile nor the histological characteristics of the tissues. Conversely in seminal plasma, rabbits fed with L and FO had lower n-6 (P<0.05), LC-PUFA n-6 (P<0.05), and n-6/n-3 ratio (P<0.05) but higher n-3 (P<0.001) and LC-PUFA n-3 (P<0.01) compared to the control group. Our study builds a map of the gene expression of TRL4 and proinflammatory cytokines in the reproductive hypothalamic-gonadal axis of the male rabbit, fundamental step for understanding the immune defence mechanisms. Diets enriched in LC-PUFA did not affect basal gene expression but modulated sperm fatty acid composition. Finally, rabbit may be an excellent animal model to study the relationship between inflammation and infertility, and the nutritional modulation of immune functions.

Pre-slaughter conditions and their effects on carcass quality have been largely addressed for pigs of 90–100 kg live weight, while few studies consider the effects of pre-slaughter conditions on the quality of the carcasses obtained from heavy pigs intended for Protected Designation of Origin (PDO) production. A total of 1680 heavy pigs were transported in 72 batches from a farm to a commercial abattoir on 16 different days, avoiding mixing unfamiliar animals. Slaughterhouse conditions, animal behaviors, and human–animal interactions were annotated at unloading and during the race toward the stunning cage. Carcass lesions on the rear, middle, and shoulder parts of the carcasses were scored. The prevalence of carcasses with severe lesions was 6.92%, 11.87%, and 6.83%, for the rear, middle, and shoulder parts, respectively. Among the pre-slaughter events, waiting before unloading and improper handling practices at the abattoir were the major factors affecting carcass lesion severity. Lairage pen space allowance was also found to affect severe rear and shoulder lesions, and the batches that were transported in the trailer had an increased prevalence of severe shoulder lesions. Our results suggest waiting time before unloading should be shortened as much as possible, and educational programs to train operators for more careful management of animals in the abattoir are greatly required to avoid improper animal handling practices.

Simple Summary In Europe, young lambs can be transported long distances for slaughter. While transport is regulated by E.U. law, there is a lack of research investigating the optimal transport conditions specifically for young lambs. For example, while the regulations set a minimum space allowance for lambs above 26 kg, no minimum is specified for young lambs meaning they can be transported in overcrowded conditions. Further, while the temperature within the vehicle must be maintained between 5–30 °C, this is well above the 21 °C said to be the upper end of the comfortable range for lambs. This study aimed to investigate how variation in space allowance and temperature can affect the welfare of young lambs. Three groups of lambs were created where either the density of individuals (0.27 vs. 0.20 m2 per head) or the temperature range (12–18 °C vs. 19–30 °C) varied. Lambs housed in the higher density and heat stress groups showed more discomfort and higher body temperatures. In addition, lambs kept in the heat stress group lost weight over the course of the study, and showed signs of heat stress when the temperature exceeded 25 °C. These results indicate that the regulations for the transport of young lambs need to be refined. Abstract Current European animal transportation law contains only a few and vague indications concerning how to move lambs of less than 26 kg. Moreover, little information is available in the literature about factors affecting these lambs’ welfare. We investigated the effect of space allowance and ambient temperature on the welfare of unweaned Lacaune lambs during a simulation of long-distance transportation (19 h). Three groups of lambs (N = 130) were housed in equally sized pens for 19 h, Control (C; n = 39; 0.27 m2 per head), Low Space Allowance (LSA; n = 52; 0.20 m2 per head), and Heat Stress (HS; n = 39; 0.27 m2 per head) groups. LSA lambs had lower space allowance than C but were tested at the same temperature, within their Thermoneutral zone (range = 12–18 °C). The HS lambs were, instead, subjected to higher temperatures (range = 19–30 °C). Scan sampling of behavior was conducted, eye temperature and body weight were also recorded. LSA and HS lambs showed more discomfort behaviors (p < 0.05) and higher eye temperatures (p < 0.001) compared to C lambs, while HS lambs additionally showed a decrease in body weight over the experimental period (p < 0.001). This study indicates that lower space allowances and higher temperatures impact negatively the welfare of lambs transported for slaughter suggesting that the regulation should be implemented taking these factors into account.

Reliable and measurable animal-based measures (ABMs) are essential for assessing animal welfare. This study aimed at proposing ABMs for dromedary camels identifying their possible associations with management. Data were collected at a permanent camel market; a total of 76 pens and 528 camels were evaluated. ABMs were collected for each welfare principle (i.e., good feeding, good housing, good health, appropriate behavior), while resources or management-based measures were collected at three levels of investigations (animal, herd, or caretakers). Recent foodborne pathogen outbreaks associated with dry pet food and treats have focused the attention on these products as vehicles of pathogens for both pets and their owners. This study investigates the purchasing habits of dog owners and determines if and in what form pet snacks can be potentially dangerous for humans, especially for children. For these purposes, questionnaires collected from 406 dog owners were evaluated and microbiological analyses were performed on 120 dry pet snacks. The shape of the pet snack affects the purchase (median score, Mdn = 6.4) and the star bone was the preferred one (Mdn = 7.1). Most of the participants (76.0%; p < 0.001) stated that snacks are attractive to their children and that the possibility for children to get in contact with pet snacks is rare, but not implausible (Mdn = 3.1). Indeed, more than 6% of respondents admitted that there had been incidents of involuntary ingestion of pet snacks, with fever and gastrointestinal manifestations. Microbiological analyses showed that 119 dry pet snacks analyzed of a total of 120 (99.2%) were in good sanitary condition except for one sample where the presence of Listeria ivanovii was confirmed. The upward trend in the presence of pets in households and the strong and continuous growth in value and volume of the pet food market led researchers to analyze possible public health issues. Children and infants in particular are the most exposed subjects as they are more likely to come into contact with pet snacks, attracted by a shape similar to that of a toy. For this reason, although our findings did not highlight important microbiological contamination of pet snacks, it would be useful to standardize food safety criteria with those for human food from a One Health perspective.

Movements of livestock across the Member States is monitored using the Trade Control and Expert System (TRACES), a web-based veterinarian certification tool. This study aimed to evaluate the criticalities of this tracing system and the most frequent non-conformities in their compilation. TRACES of cattle (n = 320) departed from France and arriving or transiting across Puglia (Italy) in 2019 (n = 199) and 2020 (n = 121) were analysed. The distribution arriving in Italy and Greece did not vary between the 2 years. The majority of the bovine (53.4%) were medium-sized cattle transported for fattening with a median body weight of 410 kg. However, almost half of the TRACES did not report correctly the total expected loaded weight or the available space, so for them, it was impossible to calculate and double-check whether the minimal space allowance was respected. Long journeys must include a resting stop after the first 14 h, however, 20.9% of the examined and declared stops did not comply with it. Finally, comparing the expected declared journey duration with a calculated one (dividing travel distance by an average speed of 70 km/h, adding 1 h for loading and unloading, and 1 h or 24 h for resting stops), we found that only 30.6% of the journey durations were congruent. The aim of this study was to develop and describe a protocol for assessing welfare in camels reared in intensive or semi-intensive systems. A literature review was conducted searching for scientific papers on assessment of animal welfare and camel behavior, management, physiology, and pathology. The paradigms of Five Freedoms, the Five Domains Model, and the welfare principles and criteria applied by the Welfare Quality® and AWIN methods were then adapted to camels. A combination of animal-, resource- and management-based indicators were selected and categorized according to three levels of assessment: (i) Caretaker, (ii) Herd, and (iii) Animal. The Caretaker level is an interview of 23 questions exploring the caretaker's background, experience, and routine management practices. The Herd level is a check of the herd and of the place (i.e., box/pen) where camels are kept. The Animal level is a visual inspection aiming at evaluating individual camel behavior and health status. The selected indicators are presented for each welfare principle and level; for instance for the principle of “Appropriate nutrition,” feeding management is investigated at Caretaker level; feed availability and quality, the number of feeding points, and camel feeding behavior are recorded at Herd level, while body condition score (BCS) is evaluated at Animal level. In this study recording sheets for the assessment at the three levels are proposed and how to conduct the assessment is described. Limitations of the proposed protocol are also discussed. Further applications of this protocol for assessing camel welfare on a large number of farms is needed to validate the proposed indicators and identify the thresholds for their acceptability as well as to develop overall welfare indices and welfare standards in camels.

This work aimed to evaluate the antimicrobial susceptibility of 87 Salmonella Infantis strains isolated in Italy from 2016 to 2019 along the food chain of broiler meat production and in humans and to determine the genetic profiles of the strains in order to establish a possible correlation with the antimicrobial pattern. All isolates were tested by the disk diffusion method to evaluate antimicrobial susceptibility toward sixteen antimicrobials, and the broth microdilution method was used to confirm extended spectrum &beta, lactamase (ESBL) production. PCR and pulsed field gel electrophoresis (PFGE) were applied to characterize ESBL-encoding and AmpC &beta, lactamase genes and to analyze the S. Infantis strains genetic profiles respectively. S. Infantis isolates showed high prevalence of resistance, in particular toward nalidixic acid (97.7%), tetracycline (96.5%), sulphamethoxazole/trimethoprim (91%) and cefepime (72.4%). The 80.5% of isolates were ESBL, cefotaxime-resistant, carrying the blaCTX-M1 gene. The most prevalent PFGE profile was XbaI.0126 (35.6%). The remaining strains had a genetic homology from 81% to 97% with the XbaI.0126 profile. The strains belonging to these profiles were isolated from different matrices collected along the broiler food chain independently on the year and from the region and there was no correlation between the PFGE profiles and resistance patterns. We found two ESBL-producing S. Infantis strains with the same XbaI.2621 profile isolated from humans and from poultry feces, not yet reported in Italy. Our findings confirmed the diffusion of ESBL-multi drug resistant (MDR) S. Infantis along the broiler food chain and in humans and underlined the importance of continuous monitoring to control and to reduce the prevalence of this bacterium, applying a global One Health approach.
