Phosphoinositide-specific phospholipase C proteins mediate environmental stress responses in many plants. However, the potential of PI-PLC genes involved with abiotic stress tolerance in wheat remains un-explored. To study TaPLC1 genetic relation with wheat drought and heat resistance. The seedlings were treated with PI-PLC inhibitor U73122 at the single leaf stage. The seedlings were treated with drought and heat stress at the two leaf stage, and some physiological indexes and the expression profile of TaPLC1 gene were determined. And the TaPLC1 overexpression vector was transferred to Arabidopsis and selected to T3 generation for drought and heat stress treatment. After 4 h of drought and heat stress, the SOD activity, MDA and soluble sugar content of the two cultivars with inhibitor were higher than those without inhibitor, the chlorophyll content decreased. CS seedlings showed significant wilting phenomenon, and TAM107 showed slight wilting. After the elimination of drought and heat stress, all seedling wilting gradually recovered, while the leaf tips of the two varieties treated with inhibitors began to wilt and turn yellow, which was more significant 5 days after the drought and heat stress, while the degree of spring wilting and yellow in CS was earlier than that in TAM107. The expression patterns of TaPLC1 gene were different in the two cultivars, but the expression levels reached the maximum at 30 min of heat stress. The change of TaPLC1 expression in TAM107 without inhibitor treatment was significantly greater than that in CS. The expression level of TaPLC1 in the two cultivars under stress was significantly different between the two cultivars treated with inhibitor and untreated, and was lower than that of the normal plants under normal conditions. These results indicated that inhibition of TaPLC1 gene expression could enhance the sensitivity of seedlings to stress. In Arabidopsis, the root lengths of transgenic and wild-type seedlings were shortened after drought stress treatment, but the root lengths of transgenic plants decreased slightly. And the expression of TaPLC1 gene was significantly increased after drought and heat stress. This indicated that overexpression of TaPLC1 improved drought resistance of Arabidopsis. The results of this study suggest that TaPLC1 may be involved in the regulation mechanism of drought and heat stress in wheat.

Phenazine-1-carboxylic acid (PCA) is a broad-spectrum antibiotic produced by rhizobacteria in the dryland wheat fields of the Columbia Plateau. PCA and other phenazines reductively dissolve Fe and Mn oxyhydroxides in bacterial culture systems, but the impact of PCA upon Fe and Mn cycling in the rhizosphere is unknown. Here, concentrations of dithionite-extractable and poorly crystalline Fe were approximately 10% and 30-40% higher, respectively, in dryland and irrigated rhizospheres inoculated with the PCA-producing (PCA+) strain Pseudomonas synxantha 2-79 than in rhizospheres inoculated with a PCA-deficient mutant. However, rhizosphere concentrations of Fe(II) and Mn did not differ significantly, indicating that PCA-mediated redox transformations of Fe and Mn were transient or were masked by competing processes. Total Fe and Mn uptake into wheat biomass also did not differ significantly, but the PCA+ strain significantly altered Fe translocation into shoots. X-ray absorption near edge spectroscopy revealed an abundance of Fe-bearing oxyhydroxides and phyllosilicates in all rhizospheres. These results indicate that the PCA+ strain enhanced the reactivity and mobility of Fe derived from soil minerals without producing parallel changes in plant Fe uptake. This is the first report that directly links significant alterations of Fe-bearing minerals in the rhizosphere to a single bacterial trait.

Plants live in association with microorganisms that positively influence plant development, vigor, and fitness in response to pathogens and abiotic stressors. The bulk of the plant microbiome is concentrated belowground at the plant root-soil interface. Plant roots secrete carbon-rich rhizodeposits containing primary and secondary low molecular-weight metabolites, lysates, and mucilages. These exudates provide nutrients for soil microorganisms and modulate their affinity to host plants, but molecular details of this process are largely unresolved. We addressed this gap by focusing on the molecular dialogue between eight well-characterized beneficial strains of thePseudomonas fluorescensgroup andBrachypodium distachyon, a model for economically important food, feed, forage, and biomass crops of the grass family. We collected and analyzed root exudates ofB. distachyonand demonstrated the presence of multiple carbohydrates, amino acids, organic acids and phenolic compounds. The subsequent screening of bacteria by Biolog Phenotype MicroArrays revealed that many of these metabolites provide carbon and energy for thePseudomonasstrains. RNA-seq profiling of bacterial cultures amended with root exudates revealed changes in the expression of genes encoding numerous catabolic and anabolic enzymes, transporters, transcriptional regulators, stress response, and conserved hypothetical proteins. Almost half of the differentially expressed genes mapped to the variable part of the strains’ pangenome, reflecting the importance of the variable gene content in the adaptation ofP. fluorescensto the rhizosphere lifestyle. Our results collectively reveal the diversity of cellular pathways and physiological responses underlying the establishment of mutualistic interactions between these beneficial rhizobacteria and their plant hosts.

Phenazine-1-carboxylic acid (PCA) is produced by rhizobacteria in dryland but not in irrigated wheat fields of the Pacific Northwest, USA. PCA promotes biofilm development in bacterial cultures and bacterial colonization of wheat rhizospheres. However, its impact upon biofilm development has not been demonstrated in the rhizosphere, where biofilms influence terrestrial carbon and nitrogen cycles with ramifications for crop and soil health. Furthermore, the relationships between soil moisture and the rates of PCA biosynthesis and degradation have not been established. In this study, expression of PCA biosynthesis genes was upregulated relative to background transcription, and persistence of PCA was slightly decreased in dryland relative to irrigated wheat rhizospheres. Biofilms in dryland rhizospheres inoculated with the PCA-producing (PCA+ ) strain Pseudomonas synxantha 2-79RN10 were more robust than those in rhizospheres inoculated with an isogenic PCA-deficient (PCA- ) mutant strain. This trend was reversed in irrigated rhizospheres. In dryland PCA+ rhizospheres, the turnover of 15 N-labelled rhizobacterial biomass was slower than in the PCA- and irrigated PCA+ treatments, and incorporation of bacterial 15 N into root cell walls was observed in multiple treatments. These results indicate that PCA promotes biofilm development in dryland rhizospheres, and likely influences crop nutrition and soil health in dryland wheat fields.

Scientists, industry, and farmers must work closely together to develop, adapt, and apply new technologies to a wide range of cropping systems. Consumer education is needed help grow public awareness that 'plant probiotics' offer a safe and environmentally friendly alternative to dependence on the use of chemical pesticides.

Plant nutrient uptake is affected by environmental stress, but how plants respond to cation-nutrient stress is poorly understood. We assessed the impact of varying degrees of cation-nutrient stress on cation uptake in an experimental plant-mineral system. Column experiments, with red pine (Pinus resinosa Ait.) seedlings growing in sand/mineral mixtures, were conducted for up to 9 months. The Ca and K were supplied from both minerals and nutrient solutions with varying Ca and K concentrations. Cation nutrient stress had little impact on carbon allocation after 9 months of plant growth and K was the limiting nutrient for biomass production. Measurement of Ca/Sr and K/Rb ratios allowed independent estimation of dissolution incongruency and discrimination against Sr and Rb during cation uptake processes. The fraction of K in biomass from biotite increased with decreasing K supply from nutrient solutions. The mineral anorthite was consistently the major source of Ca, regardless of nutrient treatment. Red pine seedlings exploited more mineral K in response to more severe K deficiency. This did not occur for Ca since Ca was not limiting plant growth. Plant discrimination factors must be carefully considered to accurately identify nutrient sources using cation tracers.
