1. Anticancer Activity of Rhizophora mucronata Leaves Extract on Sprague–Dawley Rats: In Vivo Model

Abstract

Medicinal plants are now used to treat cancer due to the presence of bioactive compounds. Apart from the plants, mangroves also possess rich bioactive compounds with high medicinal activity. Based on the ethnobotanical attributes of Rhizophora mucronata, we are keen to work with its anticancer activity. The aim of the study is to assess the anticancer activity of methanolic extract of Rhizophora mucronata leaves against breast cancer. Its safety profile for anticancer investigations was therefore confirmed through an acute toxicity assessment. In accordance with OECD guiding principles, the study was approved to evaluate the toxicity, including acute and subacute effects and anticancer activities of methanolic extract of Rhizophora mucronata leaves on Sprague–Dawley rats. In acute toxicity trials, the dose of 1000 mg/kg body weight was determined to be safe and nontoxic even at high dose levels and did not result in any indicators of toxicity or death in the tested groups compared to controls for 14 days. In contrast, rats in a subacute toxicity study were given consistent doses of 100 mg/kg, 200 mg/kg, and 300 mg/kg for a total of 28 days along with a control group. Haematological, biochemical, and histological tests conducted in advance revealed that methanolic extract of Rhizophora mucronata leaves (MERML) at repeated doses of 200 mg/kg and 300 mg/kg was normal and had no significant effects on the treated groups. Rhizophora mucronata extract (250 mg/kg) was successfully used in in vivo trials to stop the growth of breast cancer cells in groups that had been given DMBA. Based on these findings, it has been concluded that methanolic extract of Rhizophora mucronata leaves (MERML) was safe at both higher and lower dosages and could be assessed for pharmacological study.

Abstract

In this current work, the herbicidal activity of fungal metabolites stacked chitosan nanocomposite against significant aquatic invasive weed Eichhornia crassipes (water hyacinth) was examined. Herbicidal metabolites from the fungal strain Allophoma oligotrophica were extracted and purified under standard condition. Altered ionic gelation technique was received for the amalgamation of chitosan nanocomposite fabricated with herbicidal metabolites. Synthesized nanocomposite incited checked herbicidal impact on the leaflets of water hyacinth. Synthesized nanocomposite induced marked herbicidal effect on the tested leaflets of water hyacinth. Necrotic development on the tested leaflets at earlier incubation period followed by progressive enhancement of necrotic lesion reveals the noteworthy herbicidal activity of the synthesized nanocomposite. Parenchymal, mesenchymal tissue disintegration, reduction of total chlorophyll content, elevated anti oxidative enzymes and changes in qualitative protein profiling of tested leaflets reveals the nanocomposite induced noteworthy morphometric and functional effects. Effect of solvents on the release profile demonstrates that ethyl acetate treatment brought about controlled or sustained release of metabolites. No sign of toxic effect on the zebra fish embryonic developmental stages revealed biocompatibility of the nanocomposite.

49. Enhanced Antibacterial Activity of Highly Biocompatible Polymeric Core-Shell Levofloxacin Gold Nanocomposite Formulation Against Pseudomonas aeruginosa

Abstract

Using natural and synthetic polymers as the components for the core-shell nanocomposite preparation has received recent attention in biomedicine due to their high biocompatibility, high efficacy, and biodegradability. In this present investigation, chitosan-polyvinyl alcohol core-shell gold nanocomposite was synthesised adopting green science principles followed by fabrication with fluoroquinolone antibiotic levofloxacin (LE-CS-PVA-AuNC). Core-shell nanocomposite was prepared from biogenic gold nanoparticles, chitosan, polyvinyl alcohol polymer mixture, and levofloxacin under optimum conditions, and the synthesised nanocomposite exhibited a highly stable nanoarchitecture. Enhancement of antibacterial activity of the nanocomposite was evaluated against the clinical strain of Pseudomonas aeruginosa by determination of growth inhibition, survival rate parameters, and biofilm inhibition rate. Levofloxacin-fabricated core-shell nanocomposite brought about higher growth inhibition than the free levofloxacin, which was confirmed by a notable zone of inhibition, growth inhibition at a lower concentration, rapid biofilm inhibitory rate, and changes in survival growth parameters. In vitro drug release pattern was studied by continuous dialysis, which reveals that the nanocomposite exhibited controlled, sustained release pattern and cumulative release reached almost 98.0% at 72 h. Biocompatibility was studied with human keratinocytes (HaCaT cell line), which was studied by measuring cell viability, oxidative stress marker protein, and genotoxicity. The tested nanocomposite was not inducing any sign of toxicity which was confirmed by no marked impact on cell viability, intracellular reduced glutathione, lipid peroxidase, and lactate dehydrogenase activity. In addition, the nanocomposite has not shown any toxic effect on DNA, and all findings indicate that the synthesised nanocomposite was compatible with human keratinocytes. LE-CS-PVA-AuNC synthesised in the present system adopting green science principles can be used in modern biomedicine as an effective and safe antimicrobial agent due to its high antimicrobial action against wound infection pathogens and its best compatibility with human keratinocytes.
