Abstract

Background: The NORMAN Association (https://www.norman-network.com/) initiated the NORMAN Suspect List Exchange (NORMAN-SLE; https://www.norman-network.com/nds/SLE/) in 2015, following the NORMAN collaborative trial on non-target screening of environmental water samples by mass spectrometry. Since then, this exchange of information on chemicals that are expected to occur in the environment, along with the accompanying expert knowledge and references, has become a valuable knowledge base for “suspect screening” lists. The NORMAN-SLE now serves as a FAIR (Findable, Accessible, Interoperable, Reusable) chemical information resource worldwide. Results: The NORMAN-SLE contains 99 separate suspect list collections (as of May 2022) from over 70 contributors around the world, totalling over 100,000 unique substances. The substance classes include per- and polyfluoroalkyl substances (PFAS), pharmaceuticals, pesticides, natural toxins, high production volume substances covered under the European REACH regulation (EC: 1272/2008), priority contaminants of emerging concern (CECs) and regulatory lists from NORMAN partners. Several lists focus on transformation products (TPs) and complex features detected in the environment with various levels of provenance and structural information. Each list is available for separate download. The merged, curated collection is also available as the NORMAN Substance Database (NORMAN SusDat). Both the NORMAN-SLE and NORMAN SusDat are integrated within the NORMAN Database System (NDS). The individual NORMAN-SLE lists receive digital object identifiers (DOIs) and traceable versioning via a Zenodo community (https://zenodo.org/communities/norman-sle), with a total of > 40,000 unique views, > 50,000 unique downloads and 40 citations (May 2022). NORMAN-SLE content is progressively integrated into large open chemical databases such as PubChem (https://pubchem.ncbi.nlm.nih.gov/) and the US EPA’s CompTox Chemicals Dashboard (https://comptox.epa.gov

Abstract

The NORMAN Association (https://www.norman-network.com/) initiated the NORMAN Suspect List Exchange (NORMAN-SLE; https://www.norman-network.com/nds/SLE/) in 2015, following the NORMAN collaborative trial on non-target screening of environmental water samples by mass spectrometry. Since then, this exchange of information on chemicals that are expected to occur in the environment, along with the accompanying expert knowledge and references, has become a valuable knowledge base for "suspect screening" lists. The NORMAN-SLE now serves as a FAIR (Findable, Accessible, Interoperable, Reusable) chemical information resource worldwide.

10. Multiple-tracer based characterisation of a river catch-ment in the Pamir Mountains

Das Pamirgebirge in Tadschikistan ist Teil einer semi- bis kaltariden Bergregion in Zentralasien. Seine Schnee- und Gletscherablagerungen sind die Quelle eines der größten zentralasiatischen Flüsse - des Amu Darya. Die Entwicklung der weiteren Verfügbarkeit dieser wichtigen Süßwasserressourcen in Hinblick auf bevorstehende Veränderungen durch den Klimawandel steht im Mittelpunkt dieser Studie. Dazu wird der tatsächliche hydrologische Zustand eines für den Pamir exemplarischen hochalpinen Einzugsgebietes - des Flusses Gunt - beschrieben. Durch den Einsatz von hydrochemischen Tracern (Hauptionen und physikalisch-chemische Parameter) und isotopenhydrologischen Methoden (δ2H, 3H, δ18O, δ7Li, 87Sr/86Sr, Edelgase) werden grundlegende hydrologische Prozesse identifiziert und dargestellt. Dieser Multi-Tracer-Ansatz ermöglicht es, ein detailliertes Bild über das Abflussregime und die chemische Entwicklung des Flusswassers über zwei bis drei hydrologische Jahre zu erhalten., The Pamir Mountains in Tajikistan are part of a semi- to cold-arid mountain region in Central Asia. Its snow and glacier deposits are the source of one of the largest Central Asian rivers - the Amu Darya. The development of the further availability of these important fresh water resources depending on high impact from factors such as climate change is in the central focus of this study. For this, the actual hydrological condition of the high-alpine catchment of the Gunt river is investigated as exemplary catchment for the Pamir. By application of hydrochemical tracers (major ions and physico-chemical parameters) and isotope hydrological methods (δ2H, 3H, δ18O, δ7Li, 87Sr/86Sr, noble gases) basic hydrological processes are identified and described. This multi-tracer approach allows to get a detailed picture on the flow regime and chemical evolution of the river water of two to three circles of season.

Abstract

Air moisture of oceanic origin can be subject to long-range transport and could contribute to precipitation at distant places. With ongoing climate change the relationship between water vapour sources and sinks is under constant evolution and plays an important role for water budget assessments. This study analysed monthly integrated and event-based precipitation samples from two field sites in the Western Pamir Mountains (Tajikistan) for stable isotope compositions of oxygen (δ18O) and hydrogen (δ2H) of water. The aim was to investigate water vapour sources and, in particular, to evaluate contributions of water vapour from the Mediterranean region. The latter has been often postulated as a potential moisture source region for Central Asia. Deuterium excess values (d), that serve as a fingerprint of moisture origin, were below 13‰ in monthly integrated samples, for most of the year. This indicates a smaller contribution of high Mediterranean moisture (∼20‰) to the Western Pamir Mountains than originally expected. A maximum d value of 19‰ was observed in March, which is not in agreement with the common observation of increased Mediterranean contribution during winter (DJF). A Lagrangian backward trajectory model (HYSPLIT) was applied to infer the general air mass origin for sampled precipitation events. A maximum contribution of ‘western’ moisture of 40% was also detected in March, while 40%–60% of moisture that contributed to precipitation events in winter was transported by trajectories that originated from the Northern Indian Ocean.

Fachärztinnen und Fachärzte für «Prävention und Gesundheitswesen» oder schlichtweg für «Public Health» befassen sich traditionell mit der Epidemiologie von Erkrankungen, deren Determinanten und Verbreitung in Bevölkerungen. Gesundheitsförderung, Prävention oder Gesundheitsmonitoring und Systementwicklung, all diese Prozesse brauchen Daten, die erhoben, analysiert und interpretiert werden müssen, um eine nachhaltige und ganzheitliche Bevölkerungsgesundheit zu gewährleisten. Im digitalen Zeitalter stehen auf einmal sehr viele Gesundheitsdaten elektronisch zur Verfügung, die Auskunft geben über den Gesundheitszustand der Bevölkerung. Die Fachärzteschaft «Prävention und Gesundheitswesen» hat sich anlässlich einer gemeinsamen Fortbildung mit der Vereinigung der Kantonsärztinnen und -ärzte der Schweiz am 17. November 2016 mit Experten dazu ausgetauscht (Antoine Flahault, Institute of Global Health; Séverine Rion Logean, Swiss Re; Marcel Salathé, EPFL; Dr. Olivia Woolley, EPFZ). Die Anzahl an Veranstaltungen zum Thema, die in den darauffolgenden Monaten allein in der Schweiz stattfanden, weisen ebenso wie die steigende Anzahl an Publikationen von 84 im Jahr 2000 auf fast 500 im Jahr 2016 darauf hin: digitale Epidemiologie ist ein «hot topic». Doch was bedeutet digitale Epidemiologie? Und welche Konsequenzen hat sie für «Public Health»?

Abstract Background The common cold, the most prevalent contagious viral disease in humans still lacks a safe and effective antiviral treatment. Iota-Carrageenan is broadly active against respiratory viruses in-vitro and has an excellent safety profile. This study investigated the efficacy and safety of an Iota-Carrageenan nasal spray in patients with common cold symptoms. Methods In a randomized, double-blind, placebo-controlled exploratory trial, 35 human subjects suffering from early symptoms of common cold received Iota-Carrageenan (0.12%) in a saline solution three times daily for 4 days, compared to placebo. Results Administration of Iota-Carrageenan nasal spray reduced the symptoms of common cold (p = 0.046) and the viral load in nasal lavages (p = 0.009) in patients with early symptoms of common cold. Pro-inflammatory mediators FGF-2, Fractalkine, GRO, G-CSF, IL-8, IL-1α, IP-10, IL-10, and IFN-α2 were reduced in the Iota-Carrageenan group. Conclusions Iota-Carrageenan nasal spray appears to be a promising treatment for safe and effective treatment of early symptoms of common cold. Larger trials are indicated to confirm the results.

Abstract Background Type I hypersensitivity is characterized by the overreaction of the immune system against otherwise innocuous substances. It manifests as allergic rhinitis, allergic conjunctivitis, allergic asthma or atopic dermatitis if mast cells are activated in the respective organs. In case of systemic mast cell activation, life-threatening anaphylaxis may occur. Currently, type I hypersensitivities are treated either with glucocorticoids, anti-histamines, or mast cell stabilizers. Although these drugs exert a strong anti-allergic effect, their long-term use may be problematic due to their side-effects. Results In the course of a routine in vitro screening process, we identified beta-escin as a potentially anti-allergic compound. Here we tested beta-escin in two mouse models to confirm this anti-allergic effect in vivo. In a model of the early phase of allergic reactions, the murine passive cutaneous anaphylaxis model, beta-escin inhibited the effects of mast cell activation and degranulation in the skin and dose-dependently prevented the extravasation of fluids into the tissue. Beta-escin also significantly inhibited the late response after antigen challenge in a lung allergy model with ovalbumin-sensitized mice. Allergic airway inflammation was suppressed, which was exemplified by the reduction of leucocytes, eosinophils, IL-5 and IL-13 in the bronchoalveolar lavage fluid. Histopathological examinations further confirmed the reduced inflammation of the lung tissue. In both models, the inhibitory effect of beta-escin was comparable to the benchmark dexamethasone. Conclusions We demonstrated in two independent murine models of type I hypersensitivity that beta-escin has potent anti-allergic properties. These results and the excellent safety profile of beta-escin suggest a therapeutic potential of this compound for a novel treatment of allergic diseases.

Abstract

Abstract Background Human rhinoviruses (HRVs) are the predominant cause of common cold. In addition, HRVs are implicated in the worsening of COPD and asthma, as well as the loss of lung transplants. Despite significant efforts, no anti-viral agent is approved for the prevention or treatment of HRV-infection. Results In this study we demonstrate that Iota-Carrageenan, a sulphated polysaccharide derived from red seaweed, is a potent anti-rhinoviral substance in-vitro. Iota-Carrageenan reduces HRV growth and inhibits the virus induced cythopathic effect of infected HeLa cells. In addition, Iota-Carrageenan effectively prevents the replication of HRV1A, HRV2, HRV8, HRV14, HRV16, HRV83 and HRV84 in primary human nasal epithelial cells in culture. The data suggest that Iota-Carrageenan acts primarily by preventing the binding or the entry of virions into the cells. Conclusion Since HRV infections predominately occur in the nasal cavity and the upper respiratory tract, a targeted treatment with a product containing Iota-Carrageenan is conceivable. Clinical trials are needed to determine whether Iota-Carrageenan-based products are effective in the treatment or prophylaxis of HRV infections.

Abstract

Despite being the main drinking water resource for over five million people, the water balance of the Eastern Mountain Aquifer system on the western side of the Dead Sea is poorly understood. The regional aquifer consists of fractured and karstified limestone – aquifers of Cretaceous age and can be separated in Cenomanian aquifer (upper aquifer) and Albian aquifer (lower aquifer). Both aquifers are exposed along the mountain ridge around Jerusalem, which is the main recharge area. From here, the recharged groundwater flows in a highly karstified aquifer system towards the east, to discharge in springs in the Lower Jordan Valley and Dead Sea region. We investigated the Eastern Mountain Aquifer system on groundwater flow, groundwater age and potential mixtures, and groundwater recharge. We combined 36Cl/Cl, tritium and the anthropogenic gases SF6, CFC-12 and CFC-11, CFC-113 as dating tracers to estimate the young water components inside the Eastern Mountain Aquifer system. By application of lumped parameter models, we verified young groundwater components from the last 10 to 30 years and an admixture of a groundwater component older than about 70 years. Concentrations of nitrate, Simazine® (Pesticide), Acesulfame K® (artificial sweetener) and Naproxen® (drug) in the groundwater were further indications of infiltration during the last 30 years. The combination of multiple environmental tracers and lumped parameter modelling helped to understand the groundwater age distribution and to estimate recharge despite scarce data in this very complex hydrogeological setting. Abstract

Background: The common cold, the most prevalent contagious viral disease in humans still lacks a safe and effective antiviral treatment. Iota-Carrageenan is broadly active against respiratory viruses in-vitro and has an excellent safety profile. This study investigated the efficacy and safety of an Iota-Carrageenan nasal spray in patients with common cold symptoms. Methods: In a randomized, double-blind, placebo-controlled exploratory trial, 35 human subjects suffering from early symptoms of common cold received Iota-Carrageenan (0.12%) in a saline solution three times daily for 4 days, compared to placebo. Results: Administration of Iota-Carrageenan nasal spray reduced the symptoms of common cold (p = 0.046) and the viral load in nasal lavages (p = 0.009) in patients with early symptoms of common cold. Pro-inflammatory mediators FGF-2, Fractalkine, GRO, G-CSF, IL-8, IL-1a, IP-10, IL-10, and IFN-α2 were reduced in the Iota-Carrageenan group. Conclusions: Iota-Carrageenan nasal spray appears to be a promising treatment for safe and effective treatment of early symptoms of common cold. Larger trials are indicated to confirm the results. Abstract

Background: Type I hypersensitivity is characterized by the overreaction of the immune system against otherwise innocuous substances. It manifests as allergic rhinitis, allergic conjunctivitis, allergic asthma or atopic dermatitis if mast cells are activated in the respective organs. In case of systemic mast cell activation, life-threatening anaphylaxis may occur. Currently, type I hypersensitivities are treated either with glucocorticoids, anti-histamines, or mast cell stabilizers. Although these drugs exert a strong anti-allergic effect, their long-term use may be problematic due to their sideeffects. Results: In the course of a routine in vitro screening process, we identified beta-escin as a potentially anti-allergic compound. Here we tested beta-escin in two mouse models to confirm this anti-allergic effect in vivo. In a model of the early phase of allergic reactions, the murine passive cutaneous anaphylaxis model, beta-escin inhibited the effects of mast cell activation and degranulation in the skin and dose-dependently prevented the extravasation of fluids into the tissue. Beta-escin also significantly inhibited the late response after antigen challenge in a lung allergy model with ovalbumin-sensitized mice. Abstract

CA-125 is an antigenic determinant expressed on ovarian cancer cells and serves as serum marker in patients with ovarian cancer. It has also been detected, however, in serum of healthy women and in patients with benign gynecologic diseases. Previous studies in patients with ovarian hyperstimulation syndrome suggested that the production of CA-125 is correlated with proliferative ovarian activity. In this study, we describe the pattern of CA-125 secretion in 13 healthy women during menstrual cycle and during hormonal contraceptive treatment. That pattern is characterized by an increase of CA-125 serum concentrations parallel to the growth of the dominant follicle. It was not observed during hormonal contraceptive treatment. We conclude that CA-125 serum levels during menstrual cycle seem to be dependent upon the cyclic changes in the female genital tract.
