The application of overhead evaporative cooling (EC) with untreated surface water is common in Washington State to decrease sunburn on apples. While this practice reduces crop and economic losses, applying untreated surface water to produce introduces potential risks of foodborne pathogen contamination. This study examined EC and the survival of inoculated generic Escherichia coli for two apple varieties over three growing seasons. Factors examined included EC treatment (conventional, untreated), canopy location (high, low), and apple variety (Gala, Golden Delicious). Fruit maturity (mature, immature) was also examined for one year. A rifampicin-resistant generic E. coli cocktail was applied to apples at 7.3 ± 0.4 log CFU/apple, and apples were enumerated for E. coli levels at 0, 2, 10, 18, 34, 42, 58, 82, 106, and 154 h postinoculation. Log-linear, Weibull, and Biphasic models were utilized to characterize the die-off pattern of E. coli. For most treatments, inoculated E. coli decreased by almost or over 6 log CFU/apple over the study duration; however, E. coli remained detectable at low levels on apples at 154 h postinoculation. EC treatment and canopy location did not significantly impact daily linear E. coli die-off rates. Apple variety and maturity demonstrated small but statistically significant impacts on daily linear die-off rates. Nonlinear models (i.e., biphasic and Weibull) best captured E. coli die-off on apples, showing a rapid initial decrease followed by a slower decline over time. Overall, results demonstrate that EC, a useful fruit quality practice, did not impact the survival of inoculated generic E. coli on apple surfaces. The study provides valuable insights for the apple industry regarding E. coli die-off rates on in-field apples, guiding practical decisions to mitigate risk.

Inflammatory bowel disease (IBD) is characterized by disturbances in intestinal flora and metabolomic profiles. Building on previous findings that purple potato (PP) supplementation suppresses inflammation and restores intestinal barrier in dextran sulfate sodium (DSS)-treated mice, this study examined changes in the fecal metabolome in the same cohort. Mice were fed 0 % or 10 % PP for 7 weeks, with DSS administered in the fifth week of dietary treatment followed by one-week normal drinking water recovery. A total of 68 metabolites, including amino acids, fatty acids, organic acids, carbohydrates, and polyamines, were significantly altered across the groups. PP supplementation reversed changes in 13 of the 25 DSS-altered metabolites. Six microbial metabolites derived from PP polyphenols were identified and positively correlated with M2 macrophage polarization. Data suggest that dietary PP is effective in reducing inflammation and improves gut metabolites in DSS-induced colitis mice, offering a potential dietary therapy for managing IBD.

Summary: Background: Over 40% of pregnant women in the USA are obese which negatively affects fetal development and offspring health. Maternal obesity (MO) leads to fibrotic infiltration in multiple tissues and organs of offspring during their adulthood although the origin and mechanisms are unclear. Methods: C57BL/6J female mice were fed a control and high-fat diet to mimic MO condition. Embryonic somatic tissues were obtained at E9.5, E11.5, and E13.5 (equivalent to 6 weeks of human pregnancy) from control (CON) and MO mice for single-cell RNA-sequencing (scRNA-seq). To explore the role of AMP-activated protein kinase (AMPK), AMPK was activated by metformin and A769662, and knocked out in embryonic mesenchymal cells (EMC) using AMPKα1 floxed mice. Findings: Using unsupervised clustering, we identified three major cell populations with fibrogenic capacity. Compared to CON, the population of fibrogenic cells increased dramatically (by ∼125%) due to MO, supporting an embryonic origin of fibrosis in the offspring. MO induced inflammatory response and elevated expression of transforming growth factor β (TGFβ) signalling and fibrogenic genes in embryos. MO inhibited AMPK and its activation by metformin and A769662 inhibited TGFβ signalling and fibrogenesis. Interpretation: MO profoundly enhances embryonic fibrogenesis, explaining the origin of fibrosis in the offspring of mothers living with obesity. Our data underscore the importance of early intervention, before 5–6 weeks of pregnancy, in improving embryonic development, and AMPK is an amiable target for suppressing excessive fibrogenesis in MO embryos to assist increasing populations of obese mothers having healthy children. This study explored the benefits of purple potatoes against colitis in IL-10-deficient mice. Mice were fed either a standard diet or one supplemented with 10 % purple potato for 11 weeks. The results demonstrated that purple potato intake notably improved colitis symptoms and pathobiological scores. It induced significant changes in the gut bacteria, reducing harmful phyla associated with colitis, Proteobacteria and Deferribacteres, while increasing beneficial phyla, Firmicutes and Tenericutes. It increased the abundance of Bifidobacteriaceae, Porphyromonadaceae, and Lachnospiraceae-Ruminococcaceae, a major butyrate-producing group while decreasing bacterial families commonly elevated in IBD patients, including Bacteroidaceae, Sutterellaceae, Coriobacteriaceae, Enterobacteriaceae, and Peptostreptococcaceae. Additionally, purple potatoes boosted intestinal integrity, evidenced by increased goblet cell density and enhanced expression of key markers in maintaining gut health. Furthermore, they promoted mitochondrial function and reduced levels of inflammatory cytokines. Collectively, purple potatoes show promise in protecting against colitis through their effects on gut microbiota, intestinal integrity, and oxidative function.

Abstract Due to the exclusive maternal transmission, oocyte mitochondrial dysfunction reduces fertility rates, affects embryonic development, and programs offspring to metabolic diseases. However, mitochondrial DNA (mtDNA) are vulnerable to mutations during oocyte maturation, leading to mitochondrial nucleotide variations (mtSNVs) within a single oocyte, referring to mtDNA heteroplasmy. Obesity (OB) accounts for more than 40% of women at the reproductive age in the USA, but little is known about impacts of OB on mtSNVs in mature oocytes. It is found that OB reduces mtDNA content and increases mtSNVs in mature oocytes, which impairs mitochondrial energetic functions and oocyte quality. In mature oocytes, OB suppresses AMPK activity, aligned with an increased binding affinity of the ATF5‐POLG protein complex to mutated mtDNA D‐loop and protein‐coding regions. Similarly, AMPK knockout increases the binding affinity of ATF5‐POLG proteins to mutated mtDNA, leading to the replication of heteroplasmic mtDNA and impairing oocyte quality. Consistently, AMPK activation blocks the detrimental impacts of OB by preventing ATF5‐POLG protein recruitment, improving oocyte maturation and mitochondrial energetics. Overall, the data uncover key features of AMPK activation in suppressing mtSNVs, and improving mitochondrial biogenesis and oocyte maturation in obese females.

Recent foodborne outbreaks and recalls involving Listeria monocytogenes and Salmonella-contaminated peaches have caused significant economic losses to the peach industry. This study evaluated the effectiveness of chlorine, a commonly used sanitizer in the fresh produce industry, against L. monocytogenes and Salmonella and its ability to control cross-contamination in fresh peaches. Peaches inoculated with L. monocytogenes or Salmonella (~6 log10 CFU/peach) were treated with 50–150 mg/L of free chlorine (FC, pH6.8) 24 h post-inoculation. The results revealed that chlorine had similar efficacy against L. monocytogenes and Salmonella on peaches (p > 0.05). A 30 s treatment at 50, 100, and 150 mg/L FC resulted in dose-dependent reductions (p < 0.05), achieving reductions of 0.88–0.92, 1.54–1.61, and 1.73–1.79 log10 CFU/peach, respectively. Extending the contact time to 2 min slightly but significantly enhanced the chlorine efficacy (p < 0.05). Additionally, a 30 s to 2 min exposure to chlorine with 50–150 mg/L FC resulted in a 1.05–1.43 log10 CFU/peach reduction in yeasts and molds. Tap water exposure led to substantial cross-contamination between inoculated and uninoculated fruits and processed water, with Salmonella exhibiting higher transfer rates than L. monocytogenes. The application of chlorine mitigated the cross-contamination of both pathogens but did not entirely prevent it. These findings offer valuable insights for the peach and other stone fruit industries to verify process controls.

The persistence of Listeria monocytogenes biofilms on equipment surfaces poses a significant risk of cross-contamination, necessitating effective surface decontamination strategies. This study assessed the effectiveness of hurdle treatments combining peroxyacetic acid (PAA) and saturated steam against 7-day-old L. innocua (a non-pathogenic surrogate for L. monocytogenes) biofilms on stainless steel (SS), polyester (PET), and rubber surfaces. Results demonstrated >6 log10 CFU/coupon L. innocua reductions on SS and PET surfaces after PAA (40 ppm, 1 min) followed by steam treatment (100 °C, 6 s). On rubber surfaces, PAA (80 ppm, 1 min) followed by steam treatment (100 °C, 6 s) resulted in ~5 log10 CFU/coupon L. innocua reduction. The presence of apple juice soil reduced the efficacy of hurdle treatments, with PAA (40 ppm, 1 min) and steam exposure (6 s) resulting in 5.6, 5.8, and 4.2 log10 CFU/coupon reductions of L. innocua on SS, PET, and rubber, respectively. The efficacy of this antimicrobial combination was further reduced by surface defects, especially in the presence of organic matter. Nevertheless, the treatment still achieved >5 log10 CFU/coupon reductions of L. innocua on worn SS and PET soiled with apple juice and ~4.5 log10 CFU/coupon reduction on worn, soiled rubber surfaces. These findings highlight that PAA treatments followed by a brief steam exposure are effective strategies for controlling Listeria on food-contact surfaces.

Food-contact surfaces showing signs of wear pose a substantial risk of Listeria monocytogenes contamination and may serve as persistent sources of cross-contamination in fresh produce packinghouses. This study offers a comprehensive exploration into the influence of surface defects on the efficacies of commonly used sanitizers against L. monocytogenes biofilms on major food-contact surfaces. Abstract Background Following muscle injury, fibro‐adipogenic progenitors (FAPs) are rapidly activated and undergo apoptosis at the resolution stage, which is required for proper muscle regeneration. When excessive FAPs remain, it contributes to fibrotic and fatty infiltration, impairing muscle recovery. Mechanisms controlling FAP apoptosis remain poorly defined. We hypothesized that AMP‐activated protein kinase (AMPK) in FAPs mediates their apoptosis during the muscle regeneration. Methods To test, AMPKα1fl/fl PDGFRαCre mice were used to knock out AMPKα1 in FAPs. Following AMPKα1 knockout, the mice were injected with phosphate‐buffered saline or glycerol to induce muscle injury. Abstract Over the past few decades, genetic selection and refined nutritional management have extensively been used to increase the growth rate and lean meat production of livestock. However, the rapid growth rates of modern breeds are often accompanied by a reduction in intramuscular fat deposition and increased occurrences of muscle abnormalities, impairing meat quality and processing functionality. Early stages of animal development set the long-term growth trajectory of offspring. However, due to the seasonal reproductive cycles of ruminant livestock, gestational nutrient deficiencies caused by seasonal variations, frequent droughts, and unfavorable geological locations negatively affect fetal development and their subsequent production efficiency and meat quality. Therefore, enrolling livestock in nutritional intervention strategies during gestation is effective for improving the body composition and meat quality of the offspring at harvest. These crucial early developmental stages include embryonic, fetal, and postnatal stages, which have stage-specific effects on subsequent offspring development, body composition, and meat quality. This review summarizes contemporary research in the embryonic, fetal, and neonatal development, and the impacts of maternal nutrition on the early development and programming effects on the long-term growth performance of livestock. Understanding the developmental and metabolic characteristics of skeletal muscle, adipose, and fibrotic tissues will facilitate the development of stage-specific nutritional management strategies to optimize production efficiency and meat quality.

Large, renowned outbreaks associated with low-moisture foods (LMFs) bring to light some of the potential, inherent risks that accompany foods with long shelf lives if pathogen contamination occurs. Subsequently, in 2013, Beuchat et al. (2013) noted the increased concern regarding these foods, specifically noting examples of persistence and resistance of pathogens in low-water activity foods (LWAFs), prevalence of pathogens in LWAF processing environments, and sources of and preventive measures for contamination of LWAFs. For the last decade, the body of knowledge related to LMF safety has exponentially expanded. This growing field and interest in LMF safety have led researchers to delve into survival and persistence studies, revealing that some foodborne pathogens can survive in LWAFs for months to years. Research has also uncovered many complications of working with foodborne pathogens in desiccated states, such as inoculation methods and molecular mechanisms that can impact pathogen survival and persistence. Moreover, outbreaks, recalls, and developments in LMF safety research have created a cascading feedback loop of pushing the field forward, which has also led to increased attention on how industry can improve LMF safety and raise safety standards. Scientists across academia, government agencies, and industry have partnered to develop and evaluate innovate thermal and nonthermal technologies to use on LMFs, which are described in the presented review. The objective of this review was to describe aspects of the extensive progress made by researchers and industry members in LMF safety, including lessons-learned about outbreaks and recalls, expansion of knowledge base about pathogens that contaminate LMFs, and mitigation strategies currently employed or in development to reduce food safety risks associated with LMFs.

Intestinal epithelial differentiation plays an important role in maintaining gut health and is impaired due to many diseases. Purple potatoes are rich in polyphenols that promote the differentiation of intestinal epithelial cells. In this study, we found that purple potato (PP) extract up-regulated the level of peroxisome proliferator-activated receptor γ coactivator (PGC)-1α, the expression of mitochondrial-related enzymes, the content of mitochondrial DNA, and the levels of tricarboxylic acid cycle metabolites in human intestinal epithelial Caco-2 cells. Inhibition of PGC-1α by either PGC-1α inhibitor SR-18292 or siRNA-mediated knockdown eliminated the enhancing effects of PP extract on the expression of differentiation markers, mitochondrial metabolite contents and mitochondrial DNA copy number, as well as the expression of mitochondrial DNA-encoded enzymes. In summary, PP extract promoted mitochondrial biogenesis and the differentiation of intestinal epithelial cells, and these promotion effects are dependent on the activation of PGC-1α.

Summary: Background: Sarcolipin and uncoupling protein 3 (UCP3) mediate muscle-based non-shivering thermogenesis (NST) to improve metabolic homeostasis. The impacts of maternal obesity (MO) and maternal exercise (ME) on NST in offspring muscle remain unexamined. Methods: Female mice were fed with a control diet or high fat diet to induce obesity. Then, obese mice were further separated into two groups: obesity only (OB) and OB plus daily exercise (OB/Ex). Fetal muscle was collected at embryonic day 18.5 and offspring mice at 3-month-old. Apelin administration during pregnancy and apelin receptor (APJ) knockout mouse were further used for investigating the mediatory role of APJ on muscle-based thermogenesis. To explore the direct effects of exercise on AMP-activated protein kinase (AMPK) downstream targets, AMPK knockout mouse was used. Findings: MO inhibited while ME activated AMPK and peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α) in fetal muscle. AMPK activation increased sarcolipin expression, which inhibited the uptake of calcium ions into sarcoplasmic reticulum, thereby activating CaMKK2. Consistently, the expression of UCP3 and sarcolipin was suppressed due to MO but activated in ME fetal muscle. Importantly, changes of UCP3 and sarcolipin maintained in offspring muscle, showing the transgenerational effects. Furthermore, apelin administration during pregnancy mimicked the effects of ME on AMPK and CaMKK2 activation, and UCP3 and sarcolipin expression, underscoring the mediatory roles of apelin-AMPK signaling in improving fetal muscle development. Interpretation: ME, via activation of apelin signaling-AMPK axis, enhances NST gene expression in fetal and offspring muscle impaired due to MO, which intergenerationally protects offspring from diet-induced obesity and metabolic disorders. Abstract Obesity during pregnancy leads to adverse health outcomes in offspring. However, the initial effects of maternal obesity (MO) on embryonic organogenesis have yet to be thoroughly examined. Using unbiased single‐cell transcriptomic analyses (scRNA‐seq), the effects of MO on the myogenic process is investigated in embryonic day 9.5 (E9.5) mouse embryos. The results suggest that MO induces systematic hypoxia, which is correlated with enhanced BMP signaling and impairs skeletal muscle differentiation within the dermomyotome (DM). The Notch‐signaling effectors, HES1 and HEY1, which also act down‐stream of BMP signaling, suppress myogenic differentiation through transcriptionally repressing the important myogenic regulator MEF2C. Moreover, the major hypoxia effector, HIF1A, enhances expression of HES1 and HEY1 and blocks myogenic differentiation in vitro. In summary, this data demonstrate that MO induces hypoxia and impairs myogenic differentiation by up‐regulating BMP signaling within the DM, which may account for the disruptions of skeletal muscle development and function in progeny.

As the prevalence of inflammatory bowel diseases (IBD) rises, the etiology of IBD draws increasing attention. Glucoraphanin (GRP), enriched in cruciferous vegetables, is a precursor of sulforaphane, known to have anti-inflammatory and antioxidative effects. We hypothesized that dietary GRP supplementation can prevent mitochondrial dysfunction and oxidative stress in an acute colitis mouse model induced by dextran sulfate sodium (DSS). Eight-week-old mice were fed a regular rodent diet either supplemented with or without GRP. After 4 weeks of dietary treatments, half of the mice within each dietary group were subjected to 2.5% DSS treatment to induce colitis. Dietary GRP decreased DSS-induced body weight loss, disease activity index, and colon shortening. Glucoraphanin supplementation protected the colonic histological structure, suppressed inflammatory cytokines, interleukin (IL)-1β, IL-18, and tumor necrosis factor-α (TNF-α), and reduced macrophage infiltration in colonic tissues. Consistently, dietary GRP activated AMP-activated protein kinase (AMPK), peroxisome proliferator-activated receptor-gamma coactivator (PGC)-1α, and nuclear factor erythroid 2-related factor 2 (NRF2) pathways in the colonic tissues of DSS-treated mice, which was associated with increased mitochondrial DNA and decreased content of the oxidative product 8-hydroxydeoxyguanosine (8-OHDG), a nucleotide oxidative product of DNA. In conclusion, dietary GRP attenuated mitochondrial dysfunction, inflammatory response, and oxidative stress induced by DSS, suggesting that dietary GRP provides a dietary strategy to alleviate IBD symptoms.

This study aimed to investigate the effects of low-dose continuous ozone gas in controlling Listeria innocua and quality attributes and disorders of Red Delicious apples during long-term commercial cold storage. Red Delicious apples were inoculated with a three-strain L. innocua cocktail at ∼6.2 log10 CFU/apple, treated with or without 1-methylcyclopropene, and then subjected to controlled atmosphere (CA) storage with or without continuous gaseous ozone in a commercial facility for 36 weeks. Uninoculated Red Delicious apples subjected to the above storage conditions were used for yeast/mold counts and quality attributes evaluation. The 36 weeks of refrigerated air (RA) or CA storage caused ∼2.2 log10 CFU/apple reduction of L. innocua. Ozone gas application caused an additional > 3 log10 CFU/apple reduction of L. innocua compared to RA and CA storage alone. During the 36-week CA storage, low-dose continuous gaseous ozone application significantly retarded the growth of yeast/mold, delayed apple firmness loss, and had no negative influence on ozone burn, lenticel decay, russet, CO2 damage, superficial scald, and soft scald of Red Delicious apples compared to CA-alone storage. In summary, the application of continuous low-dose gaseous ozone has the potential to control Listeria on Red Delicious apples without negatively influencing apple quality attributes.

Raspberry extract is known for its anti-inflammatory effects, but its effectiveness against pathogenic Escherichia coli-induced inflammasome activation in epithelial cells is largely unknown. This study showed that raspberry extract treatment prevented oxidative stress in Caco-2 cells infected by E. coli O157:H7 and had prophylactic effects against E. coli O157:H7-induced NLRP3 inflammasome activation. In addition, raspberry extract mitigated reduction of nuclear translocation of nuclear factor erythroid 2-related factor 2 (NRF2), a key transcription factor regulating antioxidant protein expression and nitric oxide production in Caco-2 cells upon E. coli O157:H7 infection. However, raspberry extract failed to restore NRF2 nuclear content or nitric oxide production in Caco-2 cells in the presence of aminoguanidine hydrochloride, an iNOS inhibitor. Collectively, raspberry extract treatment suppressed NLRP3 mediated inflammatory response and oxidative stress in Caco-2 cells induced by E. coli O157:H7 infection. These protective effects were partially mediated by enhanced NRF2 signaling pathway and nitric oxide production.

Chronic gut inflammation is an important risk factor for colorectal cancer (CRC). Our recent study suggests that dietary Goji reduces gut inflammation and increases Bifidobacteria and butyrate-producing bacteria in interleukin-10-deficient mice. This study profiled faecal metabolomic changes of the same cohort of mice. Thirty-four compounds were altered due to Goji supplementation, including polyunsaturated long-chain fatty acids (PUFAs), sugar and sugar alcohols, benzene derivatives, pyridine derivatives, dicarboxylic acids and short-chain hydroxy acids. Pathway analysis showed that α-linolenic acid and linoleic acid metabolism were strongly affected by Goji supplementation. Four ω-6 PUFAs, including linoleic acid and arachidonic acid were reduced due to Goji supplementation. Faecal levels of most amino acids, positively correlated with gut inflammation and CRC risk, tended to decrease in the Goji supplemented group along with decreased urease activity. Data provide valuable metabolic clues for unravelling the beneficial effects of Goji supplementation against gut inflammation and related disease.

Apples are naturally coated with a water-repelling hydrophobic wax layer, which may limit the antimicrobial efficacies of surface sanitizer solutions. Lauric arginate (LAE) is a cationic surfactant with antimicrobial efficacy against Listeria monocytogenes. In this study, we investigated the antimicrobial and the wettability effects of LAE in enhancing anti-L. monocytogenes efficacy of peracetic acid (PAA) and further verified the optimized treatment combinations in a pilot spray-bar brush bed system. Apples after 48 h of inoculation were treated with PAA surface sanitation in combination with different concentrations of LAE at 22 or 46°C. The effectiveness of PAA with LAE solutions in decontaminating L. monocytogenes significantly increased with the increased concentration of PAA (60–80 ppm) or LAE (0.01–0.05%) or the treatment temperature (from 22 to 46°C). A 30–120-sec wash by 80 ppm PAA with 0.01 and 0.05% LAE at 22°C reduced L. monocytogenes on apples by 2.10–2.25 and 2.48–2.58 log10 CFU/apple, respectively. Including LAE in the PAA solution decreased contact angles on apple surfaces. However, the increased wettability of the sanitizer solution may not be the main contributor to the enhanced antimicrobial efficacy of the PAA solution, given that the addition of Tween 80 or Tween 85 only slightly boosted the anti-L. monocytogenes efficacy of PAA solutions though both increased the wettability of the PAA solutions. The synergistic effects of PAA and LAE were further validated in a pilot spray-bar brush bed packing system, where a 30-sec spray wash with 80 ppm PAA and 0.05% LAE at 22 and 46°C caused 1.68 and 2.08 log reduction of Listeria on fresh apples, respectively. This study provides an improved PAA process/preventive strategy for ensuring microbial food safety of fresh apples that is applicable to commercial apple packing lines.

Summary: Although maternal exercise (ME) becomes increasingly uncommon, the effects of ME on offspring muscle metabolic health remain largely undefined. Maternal mice are subject to daily exercise during pregnancy, which enhances mitochondrial biogenesis during fetal muscle development; this is correlated with higher mitochondrial content and oxidative muscle fibers in offspring muscle and improved endurance capacity. Apelin, an exerkine, is elevated due to ME, and maternal apelin administration mirrors the effect of ME on mitochondrial biogenesis in fetal muscle. Importantly, both ME and apelin induce DNA demethylation of the peroxisome proliferator-activated receptor γ coactivator-1α (Ppargc1a) promoter and enhance its expression and mitochondrial biogenesis in fetal muscle. Such changes in DNA methylation were maintained in offspring, with ME offspring muscle expressing higher levels of PGC-1α1/4 isoforms, explaining improved muscle function. In summary, ME enhances DNA demethylation of the Ppargc1a promoter in fetal muscle, which has positive programming effects on the exercise endurance capacity and protects offspring muscle against metabolic dysfunction.

Epidemiological studies robustly show the beneficial effects of maternal exercise in reducing maternal birth complications and improving neonatal outcomes, though underlying mechanisms remain poorly understood. To facilitate mechanistic exploration, a protocol for maternal exercise of mice is established, with the regimen following the exercise guidelines for pregnant women. Compared to volunteer wheel running, treadmill running allows precise control of exercise intensity and duration, dramatically reducing variations among individual mouse within treatments and facilitating translation into maternal exercise in humans. Based on the maximal oxygen consumption rate (VO2max) before pregnancy, the treadmill exercise protocol is separated into three stages: early stage (E1.5 to E7.5 at 40% VO2max), mid stage (E8.5 to E14.5 at 65% VO2max), and late stage of pregnancy (E15.5 to birth at 50% VO2max), which demonstrated persistent beneficial effects on maternal health and fetal development. This protocol can be useful for standardizing maternal treadmill exercise using mice as an experimental model.

Background: During muscle regeneration, excessive formation of adipogenic and fibrogenic tissues, from their respective fibro/adipogenic progenitors (FAPs), impairs functional recovery. Intrinsic mechanisms controlling the proliferation and differentiation of FAPs remain largely unexplored. Methods: Here, we investigated the role of retinoic acid (RA) signalling in regulating FAPs and the subsequent effects on muscle restoration from a cardiotoxin-induced injury. Blockage of retinoic acid receptor (RAR) signalling was achieved through dominant negative retinoic acid receptor α (RARα403) expression specific in PDGFRα+ FAPs in vivo and by BMS493 treatment in vitro. Effects of RAR-signalling on FAP cellularity and muscle regeneration were also investigated in a high-fat diet-induced obese mice model. Findings: Supplementation of RA increased the proliferation of FAPs during the early stages of regeneration while suppressing FAP differentiation and promoting apoptosis during the remodelling stage. Loss of RAR-signalling caused ectopic adipogenic differentiation of FAPs and impaired muscle regeneration. Furthermore, obesity disrupted the cellular transition of FAPs and attenuated muscle regeneration. Supplementation of RA to obese mice not only rescued impaired muscle fibre regeneration, but also inhibited infiltration of fat and fibrotic tissues during muscle repair. These beneficial effects were abolished after blocking RAR-signalling in FAPs of obese mice. Interpretation: These data suggest that RAR-signalling in FAPs is a critical therapeutic target for suppressing differentiation of FAPs and facilitating the regeneration of muscle and other tissues. Summary: Chitin, a long-chain polymer of N-acetyl-D-glucosamine (NAG) and the most abundant natural nitrogen-containing organic material in the world, is far under-utilized than other biomass resources. Herein, we demonstrate a highly efficient deoxygenation process to convert chitin monomer, i.e., NAG, into various amines, which are the ubiquitous platform chemicals in chemical industry. In the presence of H2 and Ru/C catalyst, the oxygen atoms in the glucosamine molecules are removed in the form of H2O and/or CO/CO2, whereas CO is hydrogenated to CH4. By optimizing the reaction conditions, ∼50% yield of various amines was obtained via the selective deoxygenation of NAG. The reaction mechanism has been proposed. These findings not only promote shell biorefinery in green chemistry and fishery industry but also provide chemicals for material science, resulting in expanding cooperation in new areas such as clean energy, energy conservation, environment protection, and infrastructure.

Abstract Background Vitamin A and its metabolite, retinoic acid (RA), are important regulators of cell differentiation and organ morphogenesis. Its impact on beef cattle muscle growth remains undefined. Method Angus steer calves were administrated with 0 (control) or 150,000 IU vitamin A (retinyl palmitate in glycerol, i.m.) per calf at birth and 1 month of age. At 2 months of age, a biopsy of the Biceps femoris muscle was obtained to analyze the immediate effects of vitamin A injection on myogenic capacity of muscle cells. The resulting steers were harvested at 14 months of age. Results Vitamin A administration increased cattle growth at 2 months. At 2 months of age, Vitamin A increased PAX7 positive satellite cells and the expression of myogenic marker genes including PAX7, MYF5, MYOD and MYOG. Muscle derived mononuclear cells were further isolated and induced myogenesis in vitro. More myotubes and a higher degree of myogenesis was observed in vitamin A groups. Consistently, vitamin A increased Latissimus dorsi (LD) muscle fiber size at harvest. In addition, vitamin A increased the ratio of oxidative type I and type IIA fibers and reduced the glycolic type IIX fibers. Furthermore, we found that RA, a key bioactive metabolite of vitamin A, activated PPARGC1A promoter, which explains the upregulated expression of PPARGC1A in skeletal muscle. Conclusion Vitamin A administration to neonatal calves enhanced postnatal muscle growth by promoting myogenesis and increasing satellite cell density, accompanied with a shift to oxidative muscle fibers.

Maternal vitamin A intake varies but its impact on offspring metabolic health is unknown. Here we found that maternal vitamin A or retinoic acid (RA) administration expanded PDGFRα+ adipose progenitor population in progeny, accompanied by increased blood vessel density and enhanced brown-like (beige) phenotype in adipose tissue, protecting offspring from obesity. Blockage of retinoic acid signaling by either BMS493 or negative RA receptor (RARαDN) over-expression abolished the increase in blood vessel density, adipose progenitor population, and beige adipogenesis stimulated by RA. Furthermore, RA-induced beige adipogenesis was blocked following vascular endothelial growth factor receptor (VEGFR) 2 knock out in PDGFRα+ cells, suggesting its mediatory role. Our data reveal an intrinsic link between maternal retinoid level and offspring health via promoting beige adipogenesis. Thus, enhancing maternal retinoids is an amiable therapeutic strategy to prevent obesity in offspring, especially for those born to obese mothers which account for one third of all pregnancies.

Dandelion (Taraxacum spp.) has received increasing attention in western countries due to its health beneficial properties and pharmaceutical functions. In the current study, we evaluated compounds extracted from different parts of dandelion. Dandelion leaf had the highest, while root had the lowest level of total phenolic and flavonoid contents. Using HPLC, eleven phenolic acids and flavonoids were identified, among which chicoric acid was the main component in all parts of dandelion. Consistent with the total content of phenolics and flavonoids, leaf extract had the highest total antioxidant and 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity. Dandelion leaf extract at 400 μg/ml suppressed reactive oxygen species (ROS) in HT-29 cells induced by hydrogen peroxide, equivalent to the ROS scavenging activity of 20 μg/ml of pure chicoric acid. Importantly, both leaf extract and chicoric acid mitigated inflammatory signaling NF-κB p65 and cyclooxygenase-2 activity induced by lipopolysaccharide, demonstrated by suppressed LRR and PYD domains-containing protein 3 (NLRP3) inflammasome activation and caspase-1 activity, as well as NLRP3 inflammasome–mediated interleukin (IL)-1β and IL-18 secretion. In conclusion, dandelion extract inhibited inflammasome activation through ROS scavenging and anti-inflammatory activity; and chicoric acid, a major component of dandelion, is mainly responsible for the beneficial effects.

Blueberry is an important commodity in Washington State, which was one of the leading blueberry producers in the United States. As a ready-to-eat fruit, blueberry has no or limited post-harvest processing, highlighting an imperative need to evaluate its microbial safety during pre-harvest practice. This study accessed the microbiological safety of blueberry produced in a commercial blueberry field applied with or without manure-derived ammonium sulfate (AS) fertilizer in a 2-year study. Indicator microorganisms of total coliforms and generic E. coli, Shiga toxin-producing Escherichia coli (STEC), Salmonella, and Listeria monocytogenes were monitored in fertilizer, soil, foliar, and blueberry fruit samples by culture methods for each production season. The population of total coliforms in soils was 3.17–3.82 Log10 CFU/g, which was stable throughout the production season and similar between two cropping seasons. Generic E. coli in soils remained at very low levels throughout the 2018 production season. Total coliforms or generic E. coli was not detected in fertilizer, foliar, and blueberry fruit samples collected in both 2017 and 2018 production seasons. STEC and L. monocytogenes were below the detection limit in fertilizer, soil, foliar, and blueberry fruit samples collected in both production seasons. Salmonella was not detected except for soil samples collected pre- and post-fertilizer application in the 2018 cropping season. Collectively, data indicated, under good agricultural practices, blueberry fruits produced in the field with or without manure-derived AS fertilizers had no microbiological safety concern.

Recent multistate outbreaks and recalls of fresh apples due to Listeria monocytogenes contamination have increased consumer concerns regarding fresh and processed apple safety. This study aimed to evaluate the antimicrobial efficacy of two sanitizers, mineral oxychloride (JC9450) and neutral electrolyzed water (NEW), for inactivation of L. monocytogenes on fresh apples. A 2-min treatment of 0.125% (v/v) JC9450 with 100 ppm free available chlorine (FAC) or NEW with 110 ppm FAC caused 0.9–1.2 log10 CFU/apple reduction of L. monocytogenes on both Granny Smith and Fuji apples 24 h post-inoculation. Increasing JC9450 concentration to 0.25 and 0.50% significantly improved its bactericidal effect and reduced L. monocytogenes on Granny Smith apples by ~2.0 and 3.8 log10 CFU/apple, respectively, after a contact time of 2 min. At a shorter contact time of 30 sec, the inactivation efficacy of chlorine and 0.25–0.50% JC9450 against L. monocytogenes on apples was significantly reduced compared with the respective 2-min wash. Furthermore, no L. monocytogenes was recovered in deionized water prepared antimicrobial wash solution or on non-inoculated apples post-NEW with 110 ppm FAC or 0.125–0.5% JC9450 washes, indicating their ability to prevent cross-contamination. In addition, a 2-min exposure to NEW with 110 ppm FAC and 0.50% JC9450 reduced apple native microbiota including total plate count by 0.14 and 0.65 log10 CFU/apple, respectively, and yeast and mold counts by 0.55 and 1.63 log10 CFU/apple, respectively. In summary, L. monocytogenes attached on apples was difficult to eliminate. JC9450 and NEW demonstrated a dose-dependent reduction in L. monocytogenes on apples and successfully prevented cross-contamination, indicating their application potential in post-harvest washes of apples.

Raspberry promotes adipose tissue browning and increases lipid oxidation, but its effect on intramyocellular lipid accumulation and insulin sensitivity is unclear. This study aimed to evaluate the protective role of raspberry on high-fat diet (HFD)-induced skeletal muscle lipid accumulation and insulin resistance in mice. Mice were fed a control diet or a HFD diet with or without 5% raspberry for 12 weeks. Dietary raspberry alleviated skeletal muscle inflammatory response by decreasing the expression of pro-inflammatory cytokines and p65 phosphorylation, a key mediator of inflammatory nuclear factor κB (NF-κB) signaling. Meanwhile, raspberry reduced muscle triglyceride content and improved insulin signaling in HFD mice. Raspberry intake increased mitochondrial biogenesis and phosphorylation of AMP-activated protein kinase (AMPK) α impaired due to HFD. Our results suggest that raspberry reduces lipid accumulation and improves insulin sensitivity in skeletal muscle of HFD-fed mice, which is associated with suppressed inflammation and increased mitochondrial biogenesis and AMPKα activity.

Contaminated food-contact surfaces are recognized as the primary reason for recent L. monocytogenes outbreaks in caramel apples and cantaloupes, highlighting the significance of cleaning and sanitizing food-contact surfaces to ensure microbial safety of fresh produce. This study evaluated efficacies of four commonly used chemical sanitizers at practical concentrations against L. monocytogenes biofilms on major food-contact surfaces including stainless steel, low-density polyethylene (LDPE), polyvinyl chloride (PVC), polyester (PET), and rubber. In general, efficacies against L. monocytogenes biofilms were enhanced by increasing concentrations of quaternary ammonium compound (QAC), chlorine, and chlorine dioxide, or extending treating time from 1 to 5 min. The 5-min treatments of 400 ppm QAC, 5.0 ppm chlorine dioxide, and 200 ppm chlorine reduced 3.0–3.7, 2.4–2.7, and 2.6–3.8 log10 CFU/coupon L. monocytogenes biofilms depending on surfaces. Peroxyacetic acid (PAA) at 160 and 200 ppm showed similar antimicrobial efficacies against biofilms either at 1- or 5-min contact. The 5-min treatment of 200 ppm PAA caused 4.0–4.5 log10 CFU/coupon reduction of L. monocytogenes biofilms on tested surfaces. Surface material had more impact on the efficacies of QAC and chlorine, less influence on those of PAA and chlorine dioxide, while organic matter soiling impaired sanitizer efficacies against L. monocytogenes biofilms independent of food-contact surfaces. Data from this study provide practical guidance for effective disinfection of food-contact surfaces in food processing/packing facilities.

Dairy manure, a by-product in the dairy industry, is also a potential source of nutrients for crops. However, improper application of biological soil amendments of animal origin can be a source of contamination with enteric foodborne pathogens. A 2-year field study was conducted to evaluate impacts of dairy manure fertilizer application on the microbial safety of red raspberry (Rubus idaeus L) production. Fertilizers, including a standard synthetic fertilizer (CON), straight lagoon raw manure (SL), anaerobically digested liquid effluent (DLE), compost (COM) and dairy manure-derived refined fertilizers including ammonium sulfate (AS) and phosphorous solid (PS), were randomly applied in quadruplicate to raspberry plots. Soil, fertilizer, foliar, and raspberry fruit samples were collected during the cropping season for the quantification of indicator microorganisms (total coliform and generic Escherichia coli) and detection of important foodborne pathogens (Shiga toxin-producing E. coli (STEC), Salmonella, and Listeria monocytogenes). Counts of total coliforms in soil were stable over the 2017 cropping season and were not impacted by fertilizer application. In 2018, total coliforms increased with season and soils treated with COM had a significantly higher coliform number than those treated with CON. Both total coliform and generic E. coli in raspberry fruit samples were below the detectable level (3 most probable number/g) regardless of fertilizer types. In both years, no STEC or L. monocytogenes was detected from any of the collected samples regardless of fertilizer treatments. However, Salmonella were detected in some of the fertilizers, including PS (2017), DLE (2018), and SL (2018), which were transferred to soil samples taken directly after application of these fertilizers. Salmonella were not detected in soil samples 2 or 4 months post fertilizer application, foliar, or raspberry fruit samples regardless of fertilizer applications. In summary, one-time application of raw dairy manure or dairy manure-derived fertilizers more than 4 months prior to harvest has no major impact on food safety of red raspberry (6 ft. tall) production in Lynden sandy loam under good agricultural practices.

Peroxyacetic acid (PAA) is the most commonly used antimicrobial in spray bar antimicrobial treatment during fresh apple packing and processing. However, there are limited data regarding its practical efficacy against Listeria monocytogenes on fresh apples. This study evaluated the antimicrobial activity of PAA against L. monocytogenes on fresh apples applicable to current industry practice, and further examined practical parameters impacting its efficacy to maximize the biocidal effects. Apples were inoculated with a three-strain L. monocytogenes cocktail at ~6.0 Log10 CFU/apple and then subjected to comparative antimicrobial treatments after 48 h post-inoculation. An 80 ppm PAA treatment, at 30-s and 2-min exposure, reduced L. monocytogenes on fresh apples by ~1.3 or 1.7 Log10 CFU/apple, respectively. The anti-Listeria efficacy of PAA was not affected by the water hardness and pH of PAA solution, while it improved dramatically when applied at elevated temperature. A 2-min exposure of 80 ppm PAA at 43 and 46°C resulted in a 2.3 and 2.6 Log10 CFU/apple reduction, respectively. A 30-s contact time of 80 ppm PAA at 43–46°C reduced L. monocytogenes on apples by 2.2–2.4 Log10 CFU/apple. Similarly, PAA intervention at elevated temperatures significantly strengthened its effectiveness against naturally occurring apple microbiota. PAA treatment at 43–46°C can provide a vital method to improve antimicrobial efficacy against both L. monocytogenes and indigenous microbiota on fresh apples. Our data provide valuable information and reference points for the apple industry to further validate or verify process controls.

The attachment of Escherichia coli O157:H7 to intestinal epithelial cells is indispensable for its pathogenesis. Besides translocated-intimin receptor (Tir), E. coli O157:H7 interacts with host cell surface receptors to promote intimate adhesion. This study showed that integrin β1 was increased in Caco-2 cells upon E. coli O157:H7 infection, while Caco-2 cells subjected to integrin β1 antibody blocking or CRISPR/Cas9 knockout had reduced bacterial attachment. Infection of E. coli O157:H7 inactivated focal adhesion kinase (FAK) and paxillin, increased focal adhesion (FA) and actin polymerization, and decreased cell migration in Caco-2 cells, which were rescued by integrin β1 antibody blocking or knockout. Pre-treatment with quercetin, known for its anti-oxidant and anti-inflammatory activity, reduced bacterial infection to Caco-2 cells, which might be partially via interfering integrin β1 and FAK association augmented by E. coli O157:H7. In addition, quercetin decreased FA formation induced by bacterial infection and recovered host cell motility. Taken together, data showed that E. coli O157:H7 interacts with integrin β1 to facilitate its adhesion to host cells. Quercetin inhibits bacterial infection possibly by blocking the interaction between E. coli O157:H7 and integrin β1. Collectively, these data indicate that quercetin provides an alternative antimicrobial to mitigate and control E. coli O157:H7 intestinal infection, and suggest potential broad benefits of quercetin and related polyphenols in fighting other enteric pathogen infections.

Listeria monocytogenes contaminated processing equipment and the general packing environment have been implicated in deadly foodborne listeriosis outbreaks, highlighting the significance of proper sanitization and disinfection of food contact surfaces. This study aims to comprehensively evaluate antimicrobial efficacy of commercially available, economical sanitizers at practical concentrations against L. monocytogenes biofilm formed on polystyrene surfaces under different conditions. Ozonated water 1-min treatment at 1.0, 2.0, and 4.0 ppm resulted in ∼0.9, 3.4, and 4.1 log reduction of L. monocytogenes single strain biofilm grown on polystyrene surfaces, respectively. However, its efficacy was dramatically diminished in multi-strain L. monocytogenes biofilm and was further compromised by aged biofilm and in the presence of organic matter. Quaternary ammonium compounds (QAC) at 100/400 ppm, chlorine at 100/200 ppm, chlorine dioxide at 2.5/5.0 ppm and peroxyacetic acid (PAA) at 80/160 ppm resulted in 2.4/3.6, 2.0/3.1, 2.4/3.8, and 3.6/4.8 log reduction of L. monocytogenes single strain biofilm, respectively. Antimicrobial efficacies of all tested sanitizers against 7-day-old biofilm were much lower when compared to 2-day-old biofilm, with PAA being the least influenced by the age of the biofilm. Organic matter conditioning with diluted milk or apple juice dramatically impacted the antimicrobial efficacy of all sanitizers. PAA treatment of 1 min at 160–200 ppm resulted in a 3.2–3.5 log reduction against 7-day-old biofilm in the presence of organic matter, thus showing its effectiveness in eradicating L. monocytogenes biofilm on polystyrene surface. Collectively, data highlight the importance of timely and thoroughly cleaning food contact surfaces before disinfection and provides practical information and guidance for the food industry in selecting the most effective sanitizer in their sanitizing regimes to eliminate L. monocytogenes biofilm.

Epithelial cultures are commonly used for studying gut health. However, due to the absence of mesenchymal cells and gut structure, epithelial culture systems including recently developed three-dimensional organoid culture cannot accurately represent in vivo gut development, which requires intense cross-regulation of the epithelial layer with the underlying mesenchymal tissue. In addition, organoid culture is costly. To overcome this, a new culture system was developed using mouse embryonic small intestine. Cultured intestine showed spontaneous peristalsis, indicating the maintenance of the normal gut physiological structure. During 10 days of ex vivo culture, epithelial cells moved along the gut surface and differentiated into different epithelial cell types, including enterocytes, Paneth cells, goblet cells and enteroendocrine cells. We further used the established ex vivo system to examine the role of AMP-activated protein kinase (AMPK) on gut epithelial health. Tamoxifen-induced AMPKα1 knockout vastly impaired epithelial migration and differentiation of the developing ex vivo gut, showing the crucial regulatory function of AMPK α1 in intestinal health.

The effects of dietary grape seed extract (GSE) on protecting jejunum epithelial were investigated using interlukin-10-deficient mice. GSE supplementation increased villus length, mucosal thickness and enhanced goblet cell density associated with enhanced expression of villin and two key differentiation transcription factors, Hairy/Enhancer of Split 1 (Hes1) and Krüppel-Like Factor-4 (Klf4), and increased p21 (CIP1/WAF1) content. Additionally, expression of nutrient transporters, alanyl aminopeptidase and glucose transporter-2 were increased in GSE supplemented jejunum. Consistent with its anti-inflammatory effects, GSE supplementation decreased tumor necrosis factor-α and interferon-γ expression and down-regulated NF-κB signalling. Furthermore, GSE decreased total alkaline phosphatase activity with a concomitant increase in intestinal alkaline phosphatase protein. Thus, GSE improves jejunum health in interlukin10-deficient mice likely through suppressing inflammation and regulating alkaline phosphatase.

Fresh apples are typically stored for up to 1 year commercially; different apple varieties require different storage temperatures to maintain their quality characteristics. There is sparse information available about Listeria monocytogenes survival on fresh apples under various storage temperatures. The objective of this study was to comprehensively evaluate the effect of storage temperature on apple fruit decay and L. monocytogenes survival. Unwaxed apple fruits of selected varieties (Fuji and Granny Smith) were dip inoculated in a three-strain L. monocytogenes cocktail to establish ∼3.5 and 6.0 Log10 CFU/apple. Twenty-four hours post-inoculation, apples were subjected to 1, 4, 10, or 22°C storage for up to 3 months. Apples under the different storage treatments were sampled at 1-, 4-, 7- and 14-day for short-term storage under all four tested temperatures, and 2-, 4-, 8-, and 12-week for long-term storage at 1, 4, and 10°C. A set of uninoculated and unwaxed apples were simultaneously subjected to the previously mentioned storage temperatures and sampled biweekly for their total bacterial count (TPC) and yeasts/molds (Y/M) count. During the 2-week short-term storage, L. monocytogenes population on organic Granny Smith apples stored at 1, 4, or 10°C was reduced by 0.2–0.3 Log. When apples were stored at 22°C, there was a 0.5–1.2 Log10 CFU/apple reduction 14-day post storage dependent on the initial inoculation level. During the 12-week cold storage under 1, 4, and 10°C, L. monocytogenes count on organic Granny Smith apples decreased by 0.5–1.5 Log10 CFU/apple for both inoculation levels. L. monocytogenes had similar survival pattern on conventional Granny Smith and Fuji apples with 0.8–2.0 Log10 CFU/apple reduction over a 3-month cold storage period. Interestingly, both TPC and Y/M count were stable regardless of apple variety or cultivation practice during the 12-week storage at all tested temperatures. In summary, while L. monocytogenes did not proliferate on apple surfaces during 12 weeks of refrigerated storage, only a limited reduction of L. monocytogenes was observed in this study. Therefore, the apple industry cannot rely on cold storage alone to control this pathogen. Additional interventions are needed to eradicate Listeria on fresh apples during long-term cold storage.

Adenosine monophosphate (AMP)-activated protein kinase (AMPK), a highly conserved energy sensor, has a crucial role in cardiovascular, neurodegenerative and inflammatory diseases, as well as in cancer and metabolic disorders. Accumulating studies have demonstrated that AMPK activation enhances paracellular junctions, nutrient transporters, autophagy and apoptosis, and suppresses inflammation and carcinogenesis in the intestine, indicating an essential role of AMPK in intestinal health. AMPK inactivation is an aetiological factor in intestinal dysfunctions. This review summarizes the favourable outcomes of AMPK activation on intestinal health, and discusses AMPK as a potential therapeutic target for intestinal diseases.

The impairment of intestinal epithelial barrier is the main etiologic factor of inflammatory bowel disease. The proper intestinal epithelial proliferation and differentiation is crucial for maintaining intestinal integrity. Metformin is a common anti-diabetic drug. The objective is to evaluate the protective effects of metformin on ileal epithelial barrier integrity using interleukin-10 deficient (IL10KO) mice.Wild-type and IL10KO mice were fed with/without metformin for 6 weeks and then ileum was collected for analyses. The mediatory role of AMP-activated protein kinase (AMPK) was further examined by gain and loss of function study in vitro.Compared to wild-type mice, IL10KO mice had increased proliferation, reduced goblet cell and Paneth cell lineage differentiation in the ileum tissue, which was accompanied with increased crypt expansion. Metformin supplementation mitigated intestinal cell proliferation, restored villus/crypt ratio, increased goblet cell and Paneth cell differentiation and improved barrier function. In addition, metformin supplementation in IL10KO mice suppressed macrophage pro-inflammatory activity as indicated by reduced M1 macrophage abundance and decreased pro-inflammatory cytokine IL-1β, TNF-α and IFN-γ expressions. As a target of metformin, AMPK phosphorylation was enhanced in mice treated with metformin, regardless of mouse genotypes. In correlation, the mRNA level of differentiation regulator including bmp4, bmpr2 and math1 were also increased in IL10KO mice supplemented with metformin, which likely explains the enhanced epithelial differentiation in IL10KO mice with metformin. Consistently, in Caco-2 cells, metformin promoted claudin-3 and E-cadherin assembly and mitigated TNF-α-induced fragmentation of tight junction proteins. Shiga toxin (stx) genes have been transferred to numerous bacteria, one of which is E. coli O157:H7. It is a common belief that stx gene is transferred by bacteriophages, because stx genes are located on lambdoid prophages in the E. coli O157:H7 genome. Both E. coli O157:H7 and non-pathogenic E. coli are highly enriched in cattle feedlots. We hypothesized that strong UV radiation in combination with high temperature accelerates stx gene transfer into non-pathogenic E. coli in feedlots.E. coli O157:H7 EDL933 strain were subjected to different UV irradiation (0 or 0.5 kJ/m(2)) combination with different temperature (22, 28, 30, 32, and 37 °C) treatments, and the activation of lambdoid prophages was analyzed by plaque forming unit while induction of Stx2 prophages was quantified by quantitative real-time PCR. Data showed that lambdoid prophages in E. coli O157:H7, including phages carrying stx2, were activated under UV radiation, a process enhanced by elevated temperature. Consistently, western blotting analysis indicated that the production of Shiga toxin 2 was also dramatically increased by UV irradiation and high temperature. In situ colony hybridization screening indicated that these activated Stx2 prophages were capable of converting laboratory strain of E. coli K12 into new Shiga toxigenic E. coli, which were further confirmed by PCR and ELISA analysis.These data implicate that high environmental temperature in combination with UV irradiation accelerates the spread of stx genes through enhancing Stx prophage induction and Stx phage mediated gene transfer. Cattle feedlot sludge are teemed with E. coli O157:H7 and non-pathogenic E. coli, and is frequently exposed to UV radiation via sunlight, which may contribute to the rapid spread of stx gene to non-pathogenic E. coli and diversity of shiga toxin producing E. coli.

In mice, very recent studies show that adipocytes and fibroblasts share a common pool of progenitor cells, with Zinc finger protein 423 (Zfp423) as a key initiator of adipogenic differentiation. To evaluate the role of Zfp423 in intramuscular adipogenesis and marbling in beef cattle, we sampled beef muscle for separation of stromal vascular cells. These cells were immortalized with pCI neo-hEST2 and individual clones were selected by G418. A total of 288 clones (3×96 well plates) were isolated and induced to adipogenesis. The presence of adipocytes was assessed by Oil-Red-O staining. Three clones with high and low adipogenic potential respectively were selected for further analyses. Listeria monocytogenes biofilms formed on food-contact surfaces within foodprocessing facilities pose a significant challenge, serving as persistent sources of cross-contamination. In this review, we examined documented cases of foodborne outbreaks and recalls linked to L. monocytogenes contamination on equipment surfaces and in the food production environment, provided an overview of the prevalence and persistence of L. monocytogenes in different foodprocessing facilities, and discussed environmental factors influencing its biofilm formation. We further delved into antimicrobial interventions, such as chemical sanitizers, thermal treatments, biological control, physical treatment, and other approaches for controlling L. monocytogenes biofilms on food-contact surfaces. This review provides valuable insights into the persistent challenge of L. monocytogenes biofilms in food processing, offering a foundation for future research and practical strategies to enhance food safety.
