3. Informal Learning in the Workplace: A Study of a Municipal Utility in the Southeast

Abstract

Employees engage in informal learning to solve a workplace problem and to gain knowledge at the time the need arises without waiting to attend a scheduled training session (Jacobs & Park, 2009; Livingstone, 1999; Schugurensky, 2000; Schurmann & Beausaert, 2016). The definition employed in this study is the self-guided pursuit of knowledge, as an intentional (explicit) behavior, when the employee is cognizant of the learning while it is taking place. The organization of study was a municipal (government) utility organization in the southeast United States with combination of onsite and remote office staff, and skilled field/plant workers. The purpose of the study was to examine employees' informal learning behaviors (ILBs) and tools that support their job performance, and to explain why they used those informal learning strategies and tools for their job performance. 172 employees completed a questionnaire reporting on their frequency of use of the following ILBs: talking with others, observing others, searching the internet, using job aids, using social media, reviewing policies and procedures, manuals, or standards, participating in coaching/mentoring, using shared materials, resources, and knowledge, using resources from professional organizations, and engaging in trial and error. Interviews with 10 employees representing both office and field employees, explained why employees engaged in ILBs and tools at work. Employees responded to the survey and the findings revealed that nine out of the ten strategies were currently being used by 70% or more of the respondents. Results also show that five out of the eight informal learning tools listed on the questionnaire were being used by 70% or more of the respondents. While office and field workers ILBs and tools usage results were similar, their explanations of their specific work context revealed some important points for the organization to consider. Workers in the field could benefit from access to organizational information and applications on their mobile device because the nature of field work required them to learn on-the-go. Other unique work environments, such as remote office workers, depended on reliable access to collaboration tools and videoconferencing. The findings can help this organization and others provide a work environment that encourages ILBs and supports their employees' job performance by providing necessary resources and tools for informal learning. 4. Graduation Profiles of Pedagogy Programmes According to the Current Educational Policies

Abstract

The research analyzes the graduate profiles of pedagogical careers belonging to the universities of the Valparaíso region, Chile. For this purpose, a metrically consistent instrument has been proposed and created, according to the main Education Policies of the country that is called as an integral profile, which includes aspects with which Chilean teachers are evaluated, plus indicators of inclusion and citizen education. These Policies are not always measured, that's why universities give such importance to these three educational policies. For these reasons, the objective of this article is to analyze, based on quantitative techniques, the relation of similarity between graduation profiles and educational policies, contributing to empower the descended aspects and to visualize the importance that the universities grant to train students with an integral profile. The results of this research invite to promote the Educational Policies related to the exit profiles of the pedagogical careers, since having integral profiles contributes to form a model of teacher training for the country, especially in those with emerging economies, such as those of Latin America.

Abstract

DNA-based therapeutics have emerged as a revolutionary approach for addressing the treatment gap in rare inherited conditions by targeting the fundamental genetic causes of disease. Charcot–Marie–Tooth (CMT) disease, a group of inherited neuropathies, represents one of the most prevalent Mendelian disease groups in neurology and is characterized by diverse genetic aetiology. Axonal forms of CMT, known as CMT2, are caused by dominant mutations in >30 different genes that lead to degeneration of lower motor neuron axons. Recent advances in antisense oligonucleotide therapeutics have shown promise in targeting neurodegenerative disorders. Here, we elucidate pathomechanistic changes contributing to variant specific molecular phenotypes in CMT2E, caused by a single nucleotide substitution (p.N98S) in the neurofilament light chain gene (NEFL). We used a patient-derived induced pluripotent stem cell-induced motor neuron model that recapitulates several cellular and biomarker phenotypes associated with CMT2E. Using an antisense oligonucleotide treatment strategy targeting a heterozygous gain-of-function variant, we aimed to resolve molecular phenotypic changes observed in the CMT2E p.N98S subtype. To determine the therapeutic potential of antisense oligonucleotide, we applied our treatment strategy in induced pluripotent stem cell-derived motor neurons and used both established and new biomarkers of peripheral nervous system axonal degeneration. Our findings demonstrated a significant decrease in clinically relevant biomarkers of axonal degeneration, presenting the first clinically viable genetic therapeutic for CMT2E. Abstract

Background/Objectives: Standard-of-care influenza vaccines contain antigens that are typically derived from components of wild type (WT) influenza viruses. Often, these antigens elicit strain-specific immune responses and are susceptible to mismatch in seasons where antigenic drift is prevalent. Thanks to advances in viral surveillance and sequencing, influenza vaccine antigens can now be optimized using computationally derived methodologies and algorithms to enhance their immunogenicity. Methods: Mice and ferrets that had been previously exposed to historical H1N1 and H3N2 influenza viruses were vaccinated intramuscularly with bivalent mixtures of H1 and H3 recombinant hemagglutinin (rHA) proteins, which were generated using a computationally optimized broadly reactive antigen (COBRA) design methodology. The vaccine antigens were mixed with a cationic lipid nanoparticle adjuvant, Infectimune®, which promotes both humoral and cellular immune responses. Results: Mice and ferrets vaccinated with Infectimune® and COBRA rHAs elicited protective antibody titers against panels of H1N1 and H3N2 influenza viruses isolated over the past 10 years. These animals also had antibodies that neutralized numerous modern H1N1 and H3N2 influenza viruses in vitro. When challenged with the A/Victoria/2570/2019 H1N1 influenza virus, the COBRA rHA vaccinated animals had minimal weight loss, and no detectable virus was present in their respiratory tissues on day 3 post-infection. Conclusions: These results demonstrate that COBRA rHA vaccines formulated with Infectimune® elicit protective antibody responses against influenza strains, which were isolated across periods of time when standard-of-care vaccines were frequently reformulated, thus reducing the need to update vaccines on a nearly annual basis. Abstract

Biallelic SORD mutations cause one of the most frequent forms of recessive hereditary neuropathy, estimated to affect ∼10 000 patients in North America and Europe alone. Pathogenic SORD loss-of-function changes in the encoded enzyme sorbitol dehydrogenase result in abnormally high sorbitol levels in cells and serum. How sorbitol accumulation leads to peripheral neuropathy remains to be elucidated. A reproducible animal model for SORD neuropathy is essential to illuminate the pathogenesis of SORD deficiency and for preclinical studies of potential therapies. Therefore, we have generated a Sord knockout (KO), Sord −/−, Sprague Dawley rat, to model the human disease and to investigate the pathophysiology underlying SORD deficiency. We have characterized the phenotype in these rats with a battery of behavioural tests as well as biochemical, physiological and comprehensive histological examinations. Sord −/− rats had remarkably increased levels of sorbitol in serum, CSF and peripheral nerve. Moreover, serum from Sord −/− rats contained significantly increased levels of neurofilament light chain, an established biomarker for axonal degeneration. Motor performance significantly declined in Sord −/− animals starting at ∼7 months of age. Gait analysis evaluated with video motion-tracking confirmed abnormal gait patterns in the hindlimbs. Motor nerve conduction velocities of the tibial nerves were slowed. Light and electron microscopy of the peripheral nervous system revealed degenerating myelinated axons, de- and remyelinated axons, and a likely pathognomonic finding—enlarged 'ballooned' myelin sheaths. These findings mainly affected myelinated motor axons; myelinated sensory axons were largely spared. In summary, Sord −/− rats develop a motor-predominant neuropathy that closely resembles the human phenotype. Our studies revealed novel significant aspects of SORD deficiency, and this model will lead to an improved understanding of the pathophysiology and the therapeutic options for SORD neuropathy. Abstract

The research studied the incidence of the internal control system in the logistics management of a gastronomic company in the city of Pilar in the year 2022. Variables such as the internal control system used, the logistics system implemented and the correlation between both variables were analyzed. A correlational methodology of non-experimental, cross-sectional design based on an exclusively quantitative method was used. The population included administrative and general services personnel working in the company chosen for the study. The survey technique was used as the data collection technique. The results obtained show that the internal control system affects the logistics area for its efficient operation and for the optimization of the company's resources., La investigación estudió la incidencia del sistema de control interno en la gestión de logística de una empresa gastronómica de la ciudad de Pilar en el año 2022. Se analizaron variables como sistema de control interno utilizado, sistema de logística implementado y correlación entre ambas variables. Se utilizó una metodología de nivel correlacional de diseño no experimental, de corte transversal basado en el método exclusivamente cuantitativo. La población se encuentra comprendida entre el personal administrativo y de servicios generales de empresa elegida para el estudio. Como técnica de recolección de datos se ha empleado la encuesta. Los resultados obtenidos muestran que el sistema de control interno incide en el área de logística para su funcionamiento eficiente y para la optimización de los recursos de la empresa.

Abstract

Aquest TFG està dins el projecte europeu LifeSPOT, Groundwater, as the primary source of freshwater globally, represents approximately 97% of the total freshwater supply on Earth. However, this vital resource is exposed to contamination resulting from human activities such as agriculture, livestock farming, and industry. In particular, the use of fertilizers, application of pesticides, and indiscriminate use of antibiotics can infiltrate the soil, reach aquifers, and lead to the presence of contaminants such as antibiotics, pesticides, and antibiotic resistance genes. Within the scope of this study, the efficiency of a green technology system, based on microalgae and bacteria along with wood and cork pellets, in the removal of antibiotics, pesticides, and antibiotic resistance genes in groundwater contaminated by agricultural exploitation was investigated. The abundance of three antibiotic resistance genes (ARGs) (sul1, qnrS1, and tetM), as well as the integrase gene of class 1 integrons (intI1), related to gene mobilization among bacteria, was determined in the untreated groundwater and at different points in the pilot plant using qPCR. Additionally, the 16S rRNA gene, used as an indicator of bacterial abundance in the samples under study, was also analyzed. Antibiotics and pesticides were analyzed in the same samples using liquid chromatography-mass spectrometry (HPLC-MS and/or GC-MS) technologies. The results obtained indicate that the technology based on microalgae and bacteria, in conjunction with wood and cork pellets, could be optimal for the removal of ARGs. However, it is important to ensure that the bacteria introduced into the system, responsible for contaminant removal in water, are not carriers of these genes. Furthermore, the green technology used in this study proved to be effective in the removal of antibiotics and pesticides in contaminated waters. These findings highlight the importance of addressing these issues and demonstrate the efficacy of green technologies in the removal of these contaminant, L'aigua subterrània, com a principal font d'aigua dolça a nivell mundial, representa aproximadament el 97% del subministrament total d'aigua dolça a la Terra. No obstant això, aquest recurs vital està exposat a la contaminació derivada d'activitats humanes com l'agricultura, la ramaderia i la indústria. En particular, l'ús de fertilitzants, l'aplicació de plaguicides i l'ús indiscriminat d'antibiòtics poden infiltrar-se al sòl, arribar als aqüífers i donar lloc a la presència de contaminants, com ara antibiòtics, plaguicides i gens de resistència als antibiòtics. En el marc d'aquest estudi, s'ha investigat l'eficiència d'un sistema de tecnologia verda, basat en microalgues i bacteris juntament amb pèl·lets de fusta i suro, en l'eliminació d'antibiòtics, plaguicides i gens de resistència als antibiòtics en aigües subterrànies contaminades per explotació agrícola. L'abundància de tres gens de resistència als antibiòtics (ARGs) (sul1, qnrS1 i tetM), així com el gen d'integrassa d'integrons de classe 1 (intI1), relacionat amb la mobilització de gens entre bacteris, es va determinar en aigües subterrànies a tractar i en diferents punts de la planta pilot mitjançant qPCR. A més, també es va analitzar el gen 16S RNAr, utilitzat com a indicador de l'abundància bacteriana en les mostres d'estudi. Tanmateix en les mateixes mostres es van analitzar antibiòtics i plaguicides utilitzant tecnologies d'espectrometria de masses amb cromatografia líquida (HPLC-MS i/o GC-MS). Els resultats obtinguts indiquen que la tecnologia basada en microalgues i bacteris, juntament amb pèl·lets de fusta i suro, podria ser òptima per a l'eliminació d'ARGs. No obstant això, és important garantir que els bacteris introduïts en el sistema, encarregats de l'eliminació de contaminants a l'aigua, no siguin portadors d'aquests gens. D'altra banda, la tecnologia verda utilitzada en aquest estudi va mostrar-se eficaç en l'eliminació d'antibiòtics i plaguicides en aigües contaminades. Els resultats destaque, El agua subterránea, como principal fuente de agua dulce a nivel mundial, constituye aproximadamente el 97% del suministro total de agua dulce en la Tierra. Sin embargo, este recurso vital está expuesto a la contaminación derivada de actividades humanas como la agricultura, la ganadería y la industria. En particular, el uso de fertilizantes, la aplicación de pesticidas y el uso indiscriminado de antibióticos pueden infiltrarse en el suelo, llegar a los acuíferos y dar lugar a la presencia de contaminantes, como antibióticos, pesticidas y genes de resistencia a los antibióticos. En el marco de este estudio, se investigó la eficiencia de un sistema de tecnología verde, basado en microalgas i bacterias junto con pellets de madera y corcho, en la eliminación de antibióticos, pesticidas y genes de resistencia a los antibióticos en aguas subterráneas contaminadas por explotación agrícola. La abundancia de tres genes de resistencia a antibióticos (ARGs) (sul1, qnrS1 y tetM), así como el gen de integrasa de integrones de clase 1 (inlt1), relacionado con la movilización de genes entre bacterias, se determinó en aguas subterráneas a tratar y en diferentes puntos de la planta piloto mediante qPCR. Además, también se analizó el gen 16S rRNA, utilizado como indicador de la abundancia bacteriana en las muestras de estudio. En las mismas muestras también se analizaron antibióticos y pesticidas utilizando tecnologías de espectrometría de masas con cromatografía líquida (HPLC-MS y/o GC-MS). Los resultados obtenidos indican que la tecnología basada en microalgas y bacterias en conjunto con pellets de madera y corcho podría ser óptima para la eliminación de ARGs, sin embargo, es importante garantizar que las bacterias introducidas en el sistema, encargadas en la eliminación de contaminantes en el agua, no sean portadoras de dichos genes. Abstract

The macrocyclic tetrapeptide CJ-15,208 (cyclo[Phe-D-Pro-Phe-Trp]) and its D-Trp isomer exhibit kappa opioid receptor (KOR) antagonism which prevents stress-induced reinstatement of extinguished cocaine-conditioned place preference. Here, we evaluated the effects of substitution of Trp and D-Trp on the peptides' opioid activity, antinociceptive tolerance, and the ability to prevent relapse to extinguished drug-CPP. Six analogs were synthesized using a combination of solid-phase peptide synthesis and cyclization in solution. The analogs were evaluated in vitro for opioid receptor affinity in radioligand competition binding assays, efficacy in the [35S]GTPγS assay, metabolic stability in mouse liver microsomes, and for opioid activity and selectivity in vivo in the mouse 55 °C warm-water tail-withdrawal assay. Potential liabilities of locomotor impairment, respiratory depression, acute tolerance, and conditioned place preference (CPP) were also assessed in vivo, and the ameliorating effect of analogs on the reinstatement of extinguished cocaine-place preference was assessed. Substitutions of other D-amino acids for D-Trp did not affect (or in one case increased) KOR affinity, while two of the three substitutions of an L-amino acid for Trp decreased KOR affinity. In contrast, all but one substitution increased mu opioid receptor (MOR) affinity in vitro. The metabolic stabilities of the analogs were similar to those of their respective parent peptides, with analogs containing a D-amino acid being much more rapidly metabolized than those containing an L-amino acid in this position. In vivo, CJ-15,208 analogs demonstrated antinociception, although potencies varied over an 80-fold range and the mediating opioid receptors differed by substitution. KOR antagonism was lost for all but the D-benzothienylalanine analog, and the 2′-naphthylalanine analog instead demonstrated significant delta opioid receptor (DOR) antagonism. Introduction of DOR antagonism coincided with reduced acute opioid antinociceptive tolerance and prevented stress-induced reinstatement of extinguished cocaine-CPP. [ABSTRACT FROM AUTHOR]

In the present comparative analysis of a subject that will serve for reflection, between the expense that is made in a person deprived of his liberty in the spaces of social reintegration of the State of Jalisco and what a worker who lives respecting the legally established norms earns. by the State, but emphasizing that he works a working day of at least eight hours and is paid a unit of measure for carrying out this work, that is, what was previously known as the minimum wage, with a comparative methodology of what the institutions legally established report workers and spaces of deprivation of liberty as salary, finding a difference between the salary earned by a worker with an honest way of living and a person who lives from committing crimes, therefore, workers in Mexico earn an insufficient minimum wage to meet their needs and those of their family despite spending most of the day on their chores work and do it honestly, so the other members of the family are forced to get a job in order to cover household expenses, En el presente artículo se hace un análisis comparativo de un tema que servirá para la reflexión, entre el gasto que se hace en una persona privada de su libertad en los espacios de reinserción social del Estado de Jalisco y lo que gana un trabajador que vive respetando las normas legalmente establecidas por el Estado, pero resaltando que trabaja una jornada laboral de mínimo ocho horas y le pagan por realizar este trabajo una unidad de medida, es decir, lo que se conocía con anterioridad como salario mínimo, con una metodología comparativa de lo que las instituciones legalmente establecidas reportan como salario a los trabajadores y los espacios de privación de la libertad, encontrando una diferencia entre el salario que gana un trabajador con una forma honesta de vivir y una persona que vive de la comisión de delitos, por lo que los trabajadores en México ganan un salario mínimo insuficiente para satisfacer sus necesidades y las de su familia a pesar de que dediquen la mayor parte del día a sus tareas laborales y lo hagan de forma honesta, por lo que, los demás miembros de la familia se ven forzados a conseguir un empleo para poder cubrir los gastos del hogar

Abstract

En esta investigación etnográfica, mi objetivo principal fue analizar las transformaciones de las dinámicas familiares producto del encarcelamiento y de la pandemia por Covid-19, a través del lente de las mujeres (compañeras y familiares) de los hombres privados de la libertad en la cárcel La Modelo de Bogotá. Esto, a través de técnicas etnográficas como la observación participante y las entrevistas semiestructuradas para vislumbrar las diferentes lógicas y relaciones que se dan entre ellas como familiares de los PPL (Personas privadas de la libertad) y la institución penitenciaria (INPEC) como institución que media la relación con sus seres queridos. Esta es una investigación que se inscribe en las etnografías de prisiones, además de ser un análisis desde la antropología jurídica y los estudios de género que muestra como la realidad carcelaria transforma la vida familiar, la cual se agudizó con la llegada de la pandemia. Con esto, afirmo que los fenómenos carcelarios van más allá de los muros y las rejas de las prisiones, afectando sistemáticamente a un número significativo de personas que sufren consecuencias del encierro al verse de cara al sistema penal sin haber cometido ningún delito.

Abstract

La presente investigación plantea un programa de capacitación para desarrollar las competencias didácticas de los docentes de Comprensión y Redacción de textos I de una universidad privada de Lima. La investigación realizada es de tipo aplicada educacional, la cual corresponde al paradigma sociocrítico, con enfoque cualitativo y diseño no experimental. De igual manera, las técnicas para la recolección de datos son la observación, la encuesta, la entrevista y el análisis documental, cuyos instrumentos son, una guía de observación de clase estructurada, dos cuestionarios, una guía de entrevista semiestructurada y dos guías de análisis documental. El diagnóstico demuestra que las subcategorías menos desarrolladas de las competencias didácticas son la competencia metodológica, diferenciadora, curricular y tecnológica. Asimismo, existe la necesidad de repotenciar y mejorar la competencia evaluadora, a través de la evaluación formativa con retroalimentaciones pertinentes y la competencia planificadora, para realizar una programación contextualizada. Del mismo modo, es importante considerar dentro de la competencia comunicativa y relacional, los contratos didácticos, para afianzar y mejorar las relaciones entre docentes y estudiantes. A partir del diagnóstico, se plantea una propuesta de programa de capacitación fundamentada en teorías educativas, tales como enfoque constructivista, enfoque holístico, enfoque por competencias, diseño instruccional, perspectiva laboral y modelo administrativo, así como fundamentos socioeducativos y curriculares. De igual manera, el programa de capacitación considera el entorno E-learning como medio eficaz para desarrollar el programa. Finalmente, el programa de capacitación representa un aporte práctico para la organización, ya que permite desarrollar las competencias didácticas de los docentes y con ello contribuir en la calidad educativa. Abstract

La investigación tiene por finalidad diseñar un programa de capacitación metodológica para perfeccionar el desempeño de los docentes en la clase virtual del curso de Gestión y Liderazgo en la Maestría en Educación con mención en educación superior de la escuela de postgrado de una universidad privada de Lima. La metodología utilizada se fundamenta en el paradigma socio – crítico interpretativo, el enfoque es cualitativo encaminado a una investigación educacional de tipo aplicada en donde el investigador delimito el problema en su praxis pedagógica de su aula. La muestra seleccionada por conveniencia la conforman tres docentes y veinticinco estudiantes que reciben el curso de Gestión y Liderazgo. Se emplean diversas técnicas e instrumentos tales como: observación a clases, entrevista semiestructurada a los docentes y un cuestionario a los estudiantes, así como el criterio de expertos para evaluar la efectividad de la propuesta didáctica fruto de la investigación. El resultado de los instrumentos aplicados devela las categorías emergentes que intervienen en la problemática de estudio que fueron analizadas integralmente. Como conclusión principal se presenta la modelación del programa de capacitación que se apoya en el fundamento científico, principios y categorías para optimizar el desempeño virtual docente mediante el uso adecuado de herramientas digitales.
