BACKGROUND RESULTS CONCLUSION Pseudostellaria heterophylla, known for its significant bioactive ingredients, offers potential health benefits. Amounts of bioactive compounds of the tuberous root of cultivated Pseudostellaria heterophylla are sensitive to environmental conditions. We selected 22 sampling sites in Guizhou Province, China, a primary Pseudostellaria heterophylla planting area. We analyzed polysaccharides, water‐soluble extractives, total ash and inorganic elements (Fe, Mn, Zn, Mg and Ca) in Radix Pseudostellariae, and pH, organic carbon (OC), available nitrogen (AN), available phosphorus (AP), available potassium (AK) and inorganic elements in the soil.Our study revealed a substantial presence of polysaccharides (85.00–181.00 mg g−1), water‐soluble extractives (47.52–57.63%) and total ash (1.87–3.39%) in Radix Pseudostellariae. Polysaccharides and total ash showed no sensitivity to soil pH. Radix Pseudostellariae collected from soil with pH > 7 exhibited slightly higher levels of water‐soluble extractives, Mg and Ca than that from soil with pH < 5. Conversely, soil with a pH less than 5 had higher OC, AN, AP and AK contents. Water‐soluble extractives in Radix Pseudostellariae were negatively correlated with soil pH but positively correlated with OC and AN.The results imply that the sequestration of soil nutrients over long‐term Pseudostellaria heterophylla cultivation could negatively impact the accumulation of some bioactive ingredients in Radix Pseudostellariae. Background: Plants that contain brewing microorganisms are used in traditional fermentation starters, which are an essential part of local diet, nutrition, life, and health. Regionally, the plant species used and the microorganisms included in traditional fermentation starters are diverse, endowing local fermented drinks with different flavors and health benefits. However, related traditional knowledge has been scarcely documented or revealed. Methods: An ethnobotanical survey was conducted in five towns of Nayong County in northwestern Guizhou, China. Snowball sampling, semi-structured interviews, free lists, and participatory observation were used to collect information on Jiuqu Plants (JPs) and jiuqu-making techniques. The PacBio platform was used to study the microbial community structure and diversity in the Chuanqing people's jiuqu. Results: In total, 225 informants were interviewed, including 116 who provided plants and technological processes for making Chinese baijiu jiuqu (CBJ) and 139 who provided information about making fermented glutinous rice jiuqu (FGRJ). This study found that older people have more abundant knowledge about CBJ plants. Poaceae was found to be the dominant family used in making CBJ and FGRJ (7 species each). Compared to individual plant parts, the whole plant is most commonly used in two kinds of jiuqu (19.5% in CBJ and 22.6% in FGRJ). The Chuanqing people's jiuqu is used to treat dietary stagnation and indigestion. The highest relative frequency of citation of the CBJ plant was Ficus tikoua Bureau, and the counterpart of the FGRJ plant was Buddleja macrostachya Benth. The dominant bacterial species in jiuqu were Gluconobacter japonicus (YQ1, YQ4) and Pediococcus pentosaceus (YQ2, YQ3), and the dominant fungal species was Rhizopus oryzae. Conclusion: For the first time, this study documents the unique traditional jiuqu knowledge and reveals the microbial mystery behind the FGRJ of the Chuanqing people. Therefore, this study encourages the use of online social media platforms in order to spread Jiuqu culture, the use of the new media wave in order to create multimedia databases, and also suggests that local communities should develop preservation intervention programs, in addition to nurturing the inheritors in order to prevent the disappearance of traditional Jiuqu knowledge. This research contributes to the conservation and demystification of the traditional jiuqu knowledge of the Chuanqing people and lays the foundation for further research on its microbiology, nutrition, and metabolomics. Introduction: Triple-negative breast cancer (TNBC) represents a significant global health crisis due to its resistance to conventional therapies and lack of specific molecular targets. This study explored the potential of Eriocephalus racemosus (E. racemosus) as an alternative treatment for TNBC. The cytotoxic properties and high-resolution respirometry mitochondrial activities of E. racemosus against the MDA-MB 231 TNBC cell line were evaluated. Methods: Hexane solvent and bioactive fraction extractions of E. racemosus were performed, while mass spectrometry-based metabolite profiling was used to identify the phytochemical constituents of the extracts. The extracts were further tested against MDA-MB 231 TNBC cells to determine their cytotoxicity. The mode of cell death was determined using flow cytometry. The activities of caspases 3, 8, and 9 were assessed using a multiplex activity assay kit. Glycolytic activity and High-resolution respirometry measurements of mitochondrial function in the MDA-MB 231 cell line were conducted using the Seahorse XFp and Oroboros O2K. Results: Metabolite profiling of E. racemosus plant crude extracts identified the presence of coumarins, flavonoids, sesquiterpenoids, triterpenoids, and unknown compounds. The extracts demonstrated promising cytotoxic activities, with a half maximal inhibitory concentration (IC50) of 12.84 µg/mL for the crude hexane extract and 15.49 µg/mL for the bioactive fraction. Further, the crude hexane and bioactive fraction extracts induced apoptosis in the MDA-MB-231 TNBC cells, like the reference drug cisplatin (17.44%, 17.26% and 20.25%, respectively) compared to untreated cells. Caspase 3 activities confirmed the induction of apoptosis in both cisplatin and the plant crude extracts, while caspase 8 and 9 activities confirmed the activation of both the intrinsic and extrinsic apoptosis pathways. Increased levels of glycolytic activity were observed in the hexane crude extract. High-resolution respiratory measurements showed elevated mitochondrial activities in all mitochondrial states except for complex-IV activity. Conclusion: These findings support further exploration of E. racemosus as a potential therapeutic agent for TNBC, offering a promising avenue for the development of targeted treatments with minimal adverse effects. Psidium guajava: is a tropical tree that is widely used in traditional medicine, especially for treating diarrhea. While P. guajava has been the subject of numerous reviews, none have specifically examined its ethnobotany, pharmacology, and phytochemistry in relation to its antidiarrheal activity. This review aims to summarize the evidence of effectiveness and safety of P. guajava in the treatment of diarrhea. Literature searches were conducted through Web of Science, PubMed, and ScienceDirect by using keywords "Psidium guajava" and "diarrhea" in October 2022. A total of 189 studies were included in this review. P. guajava is widely used in traditional medicine in 44 countries. Decoction and oral were the most represented method of preparation and administration, respectively, while leaves represented the most frequently cited part of the plant. Around 27 antidiarrheal or antibacterial compounds have been isolated and identified, including benzophenone glycosides, terpenes, polysaccharides, phenols, and flavonoids. This article presents ethnobotanical and pharmacological evidence for the efficacy of P. guajava leaves in the treatment of diarrhea and provides reference information for further investigation of this plant. However, despite the large number of publications on the topic, there are still some questions to answer: are quercetin and its glycosides the only ones to act as antidiarrheal agents? What is the mechanism of action of P. guajava antidiarrheal compounds? are the use of guava leaves safe in all types of populations including children, and at what dosage? Coronary heart disease is a prevalent cardiovascular ailment globally, with myocardial infarction (MI) being one of its most severe manifestations. The morbidity and mortality of MI are escalating, showing an increasing trend among younger, highly educated individuals, thereby posing a serious threat to public health. Currently, thrombolysis, percutaneous coronary intervention, and coronary artery bypass grafting are the primary clinical treatments for MI. Although these methods significantly reduce patient mortality, complications often result in poor prognoses. Due to limitations in chemical synthetic drug research, the focus has shifted towards developing herbs based on natural substances. Natural medicines represent a novel approach for safer and more effective MI management and treatment. They can control multiple pathogenic variables by targeting various pathways and systems. This paper investigates the molecular mechanisms of MI and evaluates the application of natural products and medicinal plants in MI treatment over the past 5 years, demonstrating their specific good therapeutic potential and superior tolerance. These natural therapies have been shown to mitigate myocardial cell damage caused by MI through mechanisms such as oxidative stress, inflammation, apoptosis, angiogenesis, myocardial fibrosis, autophagy, endoplasmic reticulum stress, mitophagy, and pyroptosis. Background: Cancer continues to pose a significant threat to human well-being due to the overwhelming rate of morbidity and mortality associated with it. Hence, the quest for newer, effective and safer anticancer agents has become more crucial. Over the years, some medicinal plants have been used to treat abnormal tissue growths (tumours) in Ghana. Even though sufficient literature points out that people found some relief in their use, there is limited scientific evidence of their antiproliferative activities. Method: Ethanolic extracts of nine medicinal plant materials from seven plant species, including the stem bark of Terminalia superba, Talbotiella gentii and Ceiba pentandra and the leaves of Morinda lucida, Dracaena arborea, Dioscorea dumetorum, Thaumatococcus danielli, Ceiba pentandra and Talbotiella gentii, were evaluated for antiproliferative activities against four human cancer cell lines (hepatocellular carcinoma, colorectal adenocarcinoma, cervical carcinoma, and mammary adenocarcinoma) using an MTT-based assay. Results: The extract of C. pentandra leaves, exhibited generally higher antiproliferative activity, which was particularly substantial against human hepatocellular carcinoma (HepG2) cells (IC50 = 16.3 µg/mL) and human colorectal adenocarcinoma (RKO) cells (IC50 = 18.7 µg/mL). All the other plant materials demonstrated weak (IC50: 201–500 µg/mL) to moderate (IC50: 21–200 µg/mL) antiproliferative activities against the four cancer cell lines. Conclusion: The extracts of the plant materials demonstrated varied antiproliferative activities. Extract of C. pentandra leaves exhibited the highest antiproliferative activity. The IC50 values of C. pentandra leaves met the benchmark to be considered effective against HepG2 and RKO cancer cell lines in particular. Therefore, there is the need to further undertake fractionation work on C. pentandra leaves. The antiproliferative effect of extract of C. pentandra leaves against other cancer cell lines and normal cell line could also be explored in the future to ascertain the anticancer potential of this plant material. Humans have been using plants in the treatment of various diseases for millennia. Currently, even with allopathic medicines available, numerous populations globally still use plants for therapeutic purposes. Although plants constitute a safer alternative compared to synthetic agents, it is well established that medicinal plants might also exert adverse effects. Thus, the present investigation aimed to assess the phytotoxic, cytotoxic, and genotoxic potential of two plants from the Brazilian Cerrado used in popular medicine, Davilla nitida (Vahl) Kubitzki, and Davilla elliptica (A. St.-Hil.). To this end, germination, growth, and cell cycle analyses were conducted using the plant model Lactuca sativa. Seeds and roots were treated with 0.0625 to 1 g/L for 48 hr under controlled conditions. The germination test demonstrated significant phytotoxic effects for both species at the highest concentrations tested, while none of the extracts produced significant effects in the lettuce growth test. In the microscopic analyses, the aneugenic and cytotoxic action of D. elliptica was evident. In the case of D. nitida greater clastogenic action and induction of micronuclei, (MN) were noted suggesting that the damage initiated by exposure to these extracts was not repaired or led to apoptosis. These findings indicated that the observed plant damage was transmitted to the next generation of cells by way of MN. These differences in the action of the two species may not be attributed to qualitative variations in the composition of the extracts as both are similar, but to quantitative differences associated with synergistic and antagonistic interactions between the compounds present in these extracts. Background: The field of traditional medicine encompasses a wide range of knowledge, skills, and practices that are deeply rooted in the theories, beliefs, and experiences of different cultures. The research aimed to identify traditional medicinal plants used in Guraferda District and assess the threats they face. Method: A total of 96 individuals, 80 males and 16 females, were interviewed to gather ethnobotanical data. Statistical tests like independent t tests, ANOVA, correlation, and regression were conducted using R software version 4.3.2 to compare informant groups. Result: The study found 81 medicinal plant species in the district from 71 genera and 38 families, with Asteraceae and Solanaceae families having the most species. Leaves were the most commonly used plant part for medicine. Significant differences in plant knowledge were observed across genders, age groups, education levels, and experiences. The highest ICF value was for Dermal and Cutaneous ailments, and Cissampelos mucronata A. Rich and Bidens pilosa L. had the highest fidelity levels. Conclusion: The study highlighted the importance of traditional medicinal plants in treating ailments but noted threats like overharvesting, habitat destruction, and climate change. Conservation efforts and sustainable harvesting practices are crucial to ensure the availability of these plants for future generations. The Climate Suitability Index (CSI) can increase agricultural efficiency by identifying the high-potential areas for cultivation from the climate perspective. The present study develops a probabilistic framework to calculate CSI for rainfed cultivation of 12 medicinal plants from the climate perspective of precipitation and temperature. Unlike the ongoing frameworks based on expert judgments, this formulation decreases the inherent subjectivity by using two components: frequency analysis and Particle Swarm Optimization (PSO). In the first component, the precipitation and temperature layers were prepared by calculating the occurrence probability for each plant, and the obtained probabilities were spatially interpolated using geographical information system processes. In the second component, PSO quantifies CSI by classifying a study area into clusters using an unsupervised clustering technique. The formulation was implemented in the Lake Urmia basin, which was distressed by unsustainable water resources management. By identifying clusters with higher CSI values for each plant, the results provide deeper insights to optimize cultivation patterns in the basin. Background: Scutellaria, a sub-cosmopolitan genus, stands as one of the Lamiaceae family's largest genera, encompassing approximately 500 species found in both temperate and tropical montane regions. Recognized for its significant medicinal properties, this genus has garnered attention as a research focus, showcasing anti-cancer, anti-inflammatory, antioxidant, and hepatoprotective qualities. Additionally, it finds application in agriculture and horticulture. Comprehending Scutellaria's taxonomy is pivotal for its effective utilization and conservation. However, the current taxonomic frameworks, primarily based on morphological characteristics, are inadequate. Despite several phylogenetic studies, the species relationships and delimitations remain ambiguous, leaving the genus without a stable and reliable classification system. Results: This study analyzed 234 complete chloroplast genomes, comprising 220 new and 14 previously published sequences across 206 species, subspecies, and varieties worldwide. Phylogenetic analysis was conducted using six data matrices through Maximum Likelihood and Bayesian Inference, resulting in a robustly supported phylogenetic framework for Scutellaria. We propose three subgenera, recommending the elevation of Section Anaspis to subgeneric rank and the merging of Sections Lupulinaria and Apeltanthus. The circumscription of Subgenus Apeltanthus and Section Perilomia needs to be reconsidered. Comparative analysis of chloroplast genomes highlighted the IR/SC boundary feature as a significant taxonomic indicator. We identified a total of 758 SSRs, 558 longer repetitive sequences, and ten highly variable regions, including trnK–rps16, trnC–petN, petN–psbM, accD–psaI, petA–psbJ, rpl32–trnL, ccsA–ndhD, rps15–ycf1, ndhF, and ycf1. These findings serve as valuable references for future research on species identification, phylogeny, and population genetics. Conclusions: The phylogeny of Scutellaria, based on the most comprehensive sample collection to date and complete chloroplast genome analysis, has significantly enhanced our understanding of its infrageneric relationships. The extensive examination of chloroplast genome characteristics establishes a solid foundation for the future development and utilization of Scutellaria, an important medicinal plant globally. The aim of the current research is to develop different Pharmacognostic and phytochemical analysis for the evaluation of leaf of this plant. Microscopy, Phytochemical analysis of leaf drug was performed and results were maintained. Phytochemical tests indicated the occurrence of flavonoids, tannins, steroids and phenols etc. The results obtained from standardization of leaf drug recognized the macro and microscopical parameters, physicochemical parameters. ttarakhand has a rich plant biodiversity, many of which have high medicinal properties. Medicinal plants have been used against insects, diseases, and herbivorous mammals through the production of chemical compounds. Various reasons like overexploitation, overgrazing, unsustainable development, etc. lead to their extinction and make them threatened species. The International Union for Conservation of Nature (IUCN) was established to prioritize species conservation, assess threat status, and develop suitable strategies due to the vast diversity of species. The genetic diversity of these threatened species can be determined using various markers. Molecular marker technology aids in analyzing genetic diversity, population structure, and species adaptation, enabling conservation measures and assessing genetic stability in conserved species. In this article, we have reviewed the genetic diversity of six threatened plants of Uttarakhand (Picrorhiza kurroa, Aconitum heterophyllum, Nardostachys jatamansi, Dactylorhiza hatagirea, Zanthoxylum armatum, Saussurea costus) in India as well as in other countries. A review highlighting the use of recent molecular markers like ISSR, SSR, RAPD, etc. to determine genetic diversity. Most of the medicinal plants show diversity less than 0.5, which indicates that the plants are at extreme risk of extinction. The diversity is calculated based on polymorphism percentage. The analysis of genetic diversity between species using ISSR, nextgeneration sequencing, AFLP, and microsatellite markers reveals low genetic diversity in A. heterophyllum, P. kurroa, N. jatamansi, Z. armatum, D. hatagirea, and S. costus. The global pandemic of SARS-CoV-2, or severe acute respiratory syndrome coronavirus, in 2020 claimed millions of lives globally. The clinical manifestation of this virus is called COVID-19, and it can cause everything from flu-like symptoms to severe clinical consequences and even death. The global health organization (WHO) created awareness campaigns to reduce infection rates and slow the virus's rapid spread because there was no proven medication that could treat this infection or lessen its problems with little to no negative effects on the health of the patients. Despite the fact that vaccinations have been created as preventative measures, people still prefer to use conventional herbal drugs because it offers exceptional health benefits and, with comparatively minor side effects, can either prevent a viral infection or restrict the development of serious symptoms through various mechanistic pathways. This thorough analysis offers empirical data clarifying the protective effect of various plants against SARS-CoV-2, opening the door for additional research to reevaluate plant-based extracts—rich in bioactive compounds—in more sophisticated clinical evaluations to determine their effect on COVID-19 patients. Metal complexes were synthesized and characterized using infra-red spectra, magnetic susceptibility and X-ray diffraction analysis. The combined application of these metal complexes with extracts from medicinal plants (Tamarindus indica, Emblica officinalis, Vitex negundo and Mangifera indica L.) with antimicrobial properties, showed synergistic effect. Comparative analysis revealed that the antimycotic activity of medicinal plant extracts combined with metal complexes was higher than individual metal complexes or plant extracts alone. Notably, cadmium ferrocyanide + E. officinalis extract and nickel ferrocyanide + T. indica extract exhibited highest antimycotic activities, respectively, when integrated with metal hexacyanoferrate (II) compounds against Alternaria solani. These findings showed the enhanced antimicrobial efficacy through synergistic interactions between metal complexes and medicinal plant extracts, highlighting their potential for sustainable disease management. We did a preliminary investigation of the allelopathic activity of 60 invasive and medicinal plants in Sri Lanka on Lactuca sativa (lettuce) and Raphanus sativus (radish). Out of 60 plants, 14 significantly reduced lettuce seedling lengths than distilled water, and 55 plants reduced radish seedling growth. In consideration of all the parameters tested, Cardiospermum halicacabum and Canarium zeylanicum Blume showed the highest phytotoxicity. C. zeylanicum (family Burseraceae), an endemic plant in Sri Lanka was selected for the isolation of phytotoxic compounds. The whole tree has a distinct fragrance bears edible seeds and has potential health benefits. This plant is used for medicinal purposes and its bark contains 3-hydroxy-12-methoxy-8,11,13-podocarpatrien-3-one and acetyl aleuritolic acid. The microspectrophotometer/microspectrofluorimeter was used to find the azulenes on the intact surface of 30 allelopathic and medicinal plant species growing in moderate or drought climatic conditions. Here in the maxima range of 580-620 nm, peculiar to blue pigments (azulenes), were clearly observed. The pigments were found in blue or silver leaves (needles) or plant parts (petioles, flowers and pollen). The peaks 590-610 nm, characteristic to azulenes were also observed in the absorbance spectra of ethanolic 10 min -extracts from the leaf surface, and 24 h - extracts from whole leaves. Among plants studied, the pigments were first observed in 14 terrestrial species (Anthriscus sylvestris, Lolium perenne, Petasites spurius, Phlomis tuberosa, Crambe maritima, Seseli gummiferum, Filipendula ulmaria, Echinocystis lobata, Colutea cilica, Rhus coriaria, Quercus pubescens, Pinus brutia and 2 algae (Chara vulgaris and Spirogyra sp.) use of spectral methods is recommended to search new azulene-containing species for pharmacy, agriculture and ecology. Opium poppy, coca and cannabis are raw materials for three notorious illicit drugs. For a long time, drug lords have been growing and smuggling these drugs in a variety of ways and channels and are continually finding new ways of trafficking their wares, which has led to the increasing difficulty of global drug enforcement. In the present paper, we propose an innovative pollen identification system for these important drug plants, which provides a tool for screening and detection of the drugs to aid in drug enforcement. By utilizing the characteristics of these fine particles, their abundant production, and high resistance to decay, we believe this tool could be applied in the following scenarios: detecting and dynamically monitoring drug cultivation activities; determining whether a suspect has been to fields of drug plants and determining whether the site has ever been planted with a drug plant and/or was involved in drug production. In the future, combined with microscope automatic image acquisition technology and intelligent image recognition technology, this pollen identification system is expected to be used to screen three notorious illicit drug plants, thus enhancing the efficiency of drug related crime investigations. Medicinal plants of Fritillaria are widely distributed in numerous countries around the world and possess excellent antitussive and expectorant effects. In particular, Fritillariae Bulbus (FB) as a precious traditional medicine has thousands of years of medical history in China. Herbs of Fritillaria have a high market value and demand while limited by harsh growing circumstances and scarce wild resources. As a consequence, fraudulent behaviors are regularly engaged by the unscrupulous merchants in an attempt to reap greater profits. It is of an urgent need to evaluate the quality of Fritillaria herbs and their products using various analytical instruments and techniques. This review has scrutinized approximately 160 articles from 1995 to 2022 published on the investigation of Fritillaria herbs and related herbal products. The botanical classification of genus Fritillaria, types of counterfeits, technologies applied for differentiating Fritillaria species were comprehensively summarized and discussed in the current review. Molecular and chromatographic identification were the dominant technologies in the authentication of Fritillaria herbs. Additionally, we brought some potential and promising technologies and analytical strategies into attention, which are worthy attempting in the future researches. Magnolia biondii flower buds are a well‐known medicinal plant in China, and it is important to determine the differences in aroma, flavour components, and biological activity of the essential oils of M. biondii flower buds (EOM) from different origins. In this study, the essential oils of M. biondii flower buds from Sichuan (EOM‐SC), Anhui (EOM‐AH) and Henan (EOM‐HN) provinces were extracted by hydrodistillation. The condition of the extraction process was optimized. The differences in the flavour compounds were further confirmed by GC–MS and relative odour activity value (ROAV) analyses. In addition, the antibacterial and antioxidant activity was also evaluated. The results showed that the optimized condition for steam distillation was water: flower powder ratio = 10:1 with extraction time for more than 3.0 h. Interestingly, special floral, fruity and freshness flavours were found for EOM‐SC, and the medicinal herb and sweetness odours of EOM‐HN were more prominent. The numbers and contents of flavour compounds in EOM‐SC and EOM‐HN were higher than those of EOM‐AH. Eucalyptol, β‐pinene, linalool, α‐terpineol and sabinene could be proposed as the key mutual flavour compounds for three essential oils. In addition, the strong floral, fruity and freshness aroma of EOM‐SC could be associated with the high ROAVs of α‐terpineol, linalool and sabinene. Achalypha indica is a weed that has medicinal properties. This study aims to determine the metabolite profile and evaluate the antioxidant activity of A. indica from areas with different altitudes, Tuban (20 to 50 masl; TBN), Banyuwangi (60 to 90 masl; BB), Magetan (300 to 600 masl; MGT), and Malang (500 to 800 masl; MLG). The ethanol extract of A. indica was analyzed for total phenolics and antioxidant activity. Separation and identification of samples were carried out using liquid chromatography-high-resolution mass spectrometry (LC HRMS), then classified by Principal Component Analysis (PCA). The total phenol value ranged from 6.83 to 10.67 mg GAE /g of dry powder, with an antioxidant IC 50 of 86.09 to 118.96 µg/mL. LC HRMS successfully identified 83 metabolites. PCA analysis was able to clearly distinguish each sample based on the altitude of the growing location with a total variance value reaching 82 % (PC-1: 57 %, and PC-2: 25 %). Heat map analysis that observes variations in metabolite composition in each sample shows an increase in the content of flavonoid compounds in the lowlands under certain geographical conditions. Therefore, the height of the growing location has a significant influence on the metabolite profile and antioxidant activity. These findings are very important for assessing the quality, safety, and efficacy of medicinal plant raw materials. The use of medicinal plants for managing diabetes and related conditions has been an integral part of traditional healthcare systems throughout the world including Nagaland, India. Authentic identification of these medicinal plants is crucial for their safe and effective use. A study was aimed for authentic identification of some anti-diabetic potential ethnomedicinal plants from Nagaland, India using DNA sequence based molecular characterization by amplifying and sequencing of ITS, rbcL and matK barcode regions. DNA barcode loci from the nuclear and chloroplast genomes from 15 different ethnomedicinal anti-diabetic plants of Nagaland, India, were studied for molecular characterisation. Based on the ITS, rbcL and matK region sequences, three main discrimination methods, i.e., BLAST, Phylogenetic tree and genetic distance, were adopted to identify the species. Utilising DNA markers for identification is both highly advantageous and efficient due to its reliability and rapidity. Successful amplification across species and high blast hits (100 % identity) highlighted their potential for molecular characterisation. Phylogenetic trees constructed using various models showed distinct clades and subclades, with matK revealing hypervariability and rbcL demonstrating consistent matches. Genetic distances revealed varying levels of similarity, with certain species establishing close relationships based on pairwise comparisons. GC% variations were detected, potentially indicating nucleotide composition differences and functional constraints among species genomes. The findings of this study highlight that the ITS, rbcL , and matK regions serve as effective markers for verifying the authenticity of anti-diabetic medicinal plants indigenous to Nagaland, India. Cyperus sesquiflorus is commonly called fragrant kyllinga, fragrant spikesedge, or fragrant flatsedge. It is a cespitose perennial herb with a Pantropical distribution. In Brazil, it is a native species, being commonly named fragrant grass due to its aromatic content. In this study, the essential oil (EO) of C. sesquiflorus was extracted by hydrodistillation with a Clevenger apparatus. Yields of the samples ranged between 0.12% and 0.27% (v/w) oils from fresh and dehydrated whole flowering plants (with roots) and the oil was analyzed by Gas Chromatography/-Mass spectrometry/flame ionization detection (GC-MS/FID) and Gas chromatography/Mass spectroscopy (GC/MS). The main constituent of the oil, neopetasan (3S,4aR,5S)-4a,5-dimethyl-3-(prop-1-en-2-yl)-4,4a,5,6,7,8 hexahydronaphthalen-2(3 H)-one, was isolated by preparative thin layer chromatography (PTLC) and its structure was elucidated by extensive one- and two-dimensional nuclear magnetic resonance (1D/2D-NMR) (COSY, HSQC, HMBC, and NOESY) and Fourier transform infrared (FTIR) spectroscopy in combination with MS and comparison with literature data. Summary: In this study, the effect of different levels (1%–5%) of whole and defatted Moringa oleifera leaf powder (MOLP) on the nutritional composition, physicochemical properties, protein digestibility and consumer acceptability of sorghum mahewu was investigated. The MOLP was boiled and added to the fermented sorghum mahewu. The results demonstrated that sorghum mahewu enriched with whole and defatted MOLP had higher amounts (P < 0.05) of protein, ash and fibre content compared to the control sample. Moreover, minerals, amino acid contents and protein digestibility of the enriched sorghum mahewu samples were significantly improved compared to the control sample. The pH decreased with an increasing proportion of MOLP ranging from 4.36 to 4.09 (mahewu enriched with whole MOLP) and from 4.36 to 4.17 (mahewu enriched with defatted MOLP). Sorghum mahewu samples enriched with whole and defatted MOLP had higher contents of total titratable acid compared to the control sample, while the total soluble solids and viscosity decreased. In terms of colour profile, the b*, and chroma values of sorghum mahewu increased with an increasing percentage of whole and defatted MOLP while L* and a* values decreased. Regarding overall liking, sorghum mahewu enriched with 1% whole and defatted MOLP did not differ (P > 0.05) from the control sample. Therefore, sorghum mahewu must be enriched with low percentages of MOLP to improve its acceptance among consumers. These findings suggest that incorporating medicinal plants such as boiled MOLP to basic foods produced from sorghum flour could improve their nutritional profile and protein digestibility. Finding methods, which may alleviate drought stress on the growth and biochemical properties of safflower (Carthamus tinctorius L.), as one of the important medicinal plants, is of significance. The objective was to alleviate drought stress on seed germination, seedling growth, and biochemical properties of different genotypes of safflower using 24-epibrassinosteroid (EBR). The experiments, conducted in the greenhouse in 2014 and 2015, were factorial on the basis of a complete randomized block design with three replicates. Drought stress (85 (S1), 65 (S2) and 45% (S3) of field capacity), genotype (Goldasht, Faraman and Sina), and the foliar application of 24-epibrassinosteroid (EBR) (0 and 10−7 M) were tested. Seed germination indices, seedling growth and biochemical properties were determined. The results indicated the significant effects of drought stress on the measured parameters including seed germination, which was the most sensitive stage. The use of EBR significantly increased seed germination and seedling growth, nutrient uptake and grain protein and oil contents, at different levels of stress. EBR significantly enhanced grain N (7.3%) and P (0.5%) uptake, as well as protein percentage (38.7%), even at the highest level of stress (S3), to the levels which were comparable with S1 and S2. The use of EBR may improve seed germination and the growth of safflower seedlings under drought stress conditions by affecting the physiological properties, mainly related to the mechanisms of protein production. Stevia plant is one of the most important medicinal plants used to control diabetes due to its sweetening properties and low calories. Stevia is cultivated in many parts of the world, and to increase its sweetening properties, the effects of many different factors have been tested on this plant. In this research, we investigated the effect of elements related to metal oxidants on the induction of molecular levels and transcription. Thus, the activity of 3 key genes named CPPS, HDS, and GGDPS in response to six different metal oxidants named Cro3, Pbo, Fe2O3, Ag2O, Bao, and Tio2 was carried out in this research. The results showed that the increased concentration of metal oxides, especially Fe2O3 and TiO2, escalates gene expression in the biosynthesis of sweeteners extracted from stevia leaves. Also, related to all treatments, the higher the concentration, the higher the gene expression. Among all metal oxide treatments, Pbo and Bao resulted in low gene expression for CPPS, HDS, and GGDPS genes. On the other side, control showed the lowest expression regarding all three surveyed genes, indicating that using metal oxides can achieve higher production of sweeteners in stevia plants. The results of this research determined that physiological characteristics are affected by metal oxide treatments. Also, the expression of genes effective in the production of steviol glycosides, which is one of the important sweetening factors of this plant in the leaves, increases under the influence of these treatments. Appraised for being one of the oldest staple nutritive foods mainly in the Arabian Peninsula, the date palm tree (Phoenix dactylifera L.), is a crop native to the subtropical and tropical regions of Southern Asia and Africa. Different parts of the date tree have been extensively studied for their nutritional and therapeutic properties. Despite an array of publications on the date tree, there has been no attempt to compile in a single study the traditional uses, nutritive value, phytochemical profile, the medicinal properties as well as the potential of the different plant parts as a functional food. Therefore, this review endeavors to systematically review the scientific literature to highlight the traditional uses of date fruit and parts around the world, the nutritional profile of several parts and the medicinal properties. A total of 215 studies was retrieved (traditional uses (n = 26), nutritional (n = 52), and medicinal (n = 84)). Scientific articles were further categorized as in vitro (n = 33), in vivo (n = 35), and clinical (n = 16) evidences. Date seeds were found to be effective against E. coli and Staphylococcus aureus. Aqueous date pollen was used to manage hormonal problems and boost fertility. Palm leaves showed anti-hyperglycemic effects via inhibition of α-amylase and α-glucosidase. Unlike previous studies, this study highlighted the functional roles of all the plant parts of the palm tree and provided insights into the various mechanism of action of their bioactive compounds. Although scientific shreds of evidence have been growing over the years, there is still a dearth of studies concerning the clinical validation of the date fruit and other plant parts to provide strong evidence on their medicinal uses. In conclusion, P. dactylifera can be regarded as a potent medicinal plant with prophylactic potential and should be further explored to alleviate the burden of both communicable and non-communicable diseases. A pair of atropisomers secofumitremorgins C (1a) and D (1b), together with fifteen known alkaloids (2-16), were isolated from a saltern-derived fungus Aspergillus fumigatus GXIMD00544. The structures of atropisomers 1a and 1b were elucidated by the detailed spectroscopic data, chemical reaction and quantum chemical calculations. Compounds 1 and 8 displayed antifungal spore germination effects against plant pathogenic fungus associated with sugarcane Fusarium sp. with inhibitory rates of 53% and 77% at the concentration of 100 µM, repectively. Atropisomers 1 also exhibited antifouling potential against Balanus amphitrite larval settlement with an inhibitory rate of 96% at the concentration of 100 µM. [ABSTRACT FROM AUTHOR]

Vascular calcification is a crucial risk factor that affects the incidence and mortality of cardiovascular disease in chronic kidney disease patients. Modern medicine relies on calcium-phosphorus binding agents, calcium mimetics, active vitamin D, and hemodialysis to prevent and treat vascular calcification, however, their efficacy is unsatisfactory and adverse reactions often occur. Medical plant therapy can act as an integrative regulator in patients with chronic kidney disease-associated vascular calcification, which can significantly improve patients' symptoms, but its specific mechanism has not been fully elucidated yet. In this paper, we reviewed the domestic and international theoretical studies on the pathogenesis mechanism of chronic kidney disease-associated vascular calcification in recent years, summarized eight active ingredients of medicinal plants as well as four compound formulas for improving chronic kidney disease-associated vascular calcification, and explored the mechanism of action of herbal medicine, which will provide a new strategy for promoting the prevention and treatment of vascular calcification. Oral squamous cell carcinoma (OSCC) is the most common malignant cancer of the head and neck, with high morbidity and mortality, ranking as the sixth most common cancer in the world. The treatment of OSCC is mainly radiotherapy, chemotherapy and surgery, however, the prognosis of patients is still poor and the recurrence rate is high. This paper reviews the range of effects of natural medicinal plant active ingredients (NMPAIs) on OSCC cancer, including the types of NMPAIs, anti-cancer mechanisms, involved signaling pathways, and clinical trials. The NMPAIs include terpenoids, phenols, flavonoids, glycosides, alkaloids, coumarins, and volatile oils. These active ingredients inhibit proliferation, induce apoptosis and autophagy, inhibit migration and invasion of OSCC cells, and regulate cancer immunity to exert anti-cancer effects. The mechanism involves signaling pathways such as mitogen-activated protein kinase, phosphatidylinositol 3 kinase/protein kinase B, nuclear factor kappa B, miR-22/WNT1/β-catenin and Nrf2/Keap1. Clinically, NMPAIs can inhibit the growth of OSCC, and the combined drug is more effective. Natural medicinal plants are promising candidates for the treatment of OSCC. [ABSTRACT FROM AUTHOR]

As ethnic medicine, the whole grass of plants in Cirsium was used as antimicrobial. This review focuses on the antimicrobial activity of plants in Cirsium, including antimicrobial components, against different types of microbes and bacteriostatic mechanism. The results showed that the main antimicrobial activity components in Cirsium plants were flavonoids, triterpenoids and phenolic acids, and the antimicrobial ability varied according to the species and the content of chemicals. Among them, phenolic acids showed a strong antibacterial ability against Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterococcus faecium. The antibacterial mechanisms include: (1) damaging the cell membrane, cell walls, mitochondria and nucleus of bacteria; (2) inhibiting the synthesis of proteins and nucleic acids; (3) suppressing the synthesis of enzymes for tricarboxylic acid cycle pathways and glycolysis, and then killing the bacteria via inhibition of energy production. Totally, most research results on antimicrobial activity of Cirsium plants are reported based on in vitro assays. The evidence from clinical data and comprehensive evaluation are needed. Cosmos caudatus is a widely used traditional medicinal plant in Indonesia for cancer treatment. However, the mechanism of action of plants in treating breast neoplasms remains unclear. Thus, the active ingredients of C. caudatus and possible molecular pathways against breast neoplasms were investigated using pharmacological networks. The active compounds were screened using Lipinski and ADME parameters. Targets for the compounds were obtained using SwissTargetPrediction. Target diseases were identified using the Therapeutic Target Database, whereas disease pathways were analyzed using the Kyoto Encyclopedia of Genes and Genomes. Network pharmacology was constructed using Cytoscape. Protein-protein interactions were constructed using STRING. Gene ontology and KEGG were analyzed using DAVID. Molecular docking confirmed that the compounds and targets exhibited the best interactions. The compounds, targets, and pathways with the highest degrees in the breast neoplasm network were eriodictyol, ABCC1, and signal transduction, respectively. EGFR is the key target of PPIs. GO enrichment in BP, MF, CC, and KEGG regulated NF-KB transcription factor activity, chromatin, transcription coactivator binding, and pathways in cancer, respectively. Molecular docking with the best score on interaction 5-O-methylvisammioside-SLC2A1 (PDB ID: 6THA) (-10.3 kcal mol-1). Bisphenol-A (BPA) is a synthetically manufactured phenolic substance that is very widely employed in producing polycarbonate polymers and epoxy resins which further can be found in numerous items of daily use such as foodstuff and their containers, beverages, milk, audit rolls, dental fillings and so forth. Additionally, it is present in the air we breathe-in. The studies have reported that BPA produces toxic impacts on reproductive, cardiovascular, endocrine, renal, gastrointestinal, immune, respiratory, nervous and skeletal system. In the current literature review, we have targeted to highlight the protective role of herbs in opposition to BPA toxicity. The gift of nature-'Plants' seem to be very efficient and potential agents to overcome the BPA induced toxicity as they themselves are devoid of adverse effects. Plants contain a mixture of phyto-molecules which together act to provide a synergistic effect. Further, we have explained the signaling mechanisms implicated in BPA toxicity. Even though, multiple mechanisms involved in BPA toxicity have been examined, no particular target therapy for this toxicity is available at present. Astragali radix (AR, namded Huangqi in Chinese) is the dried root of Astragalus membranaceus (Fisch.) Bge. var. mongholicus (Bge.) Hsiao or Astragalus membranaceus (Fisch.) Bge. As a widely used ethnomedicine, the biological activities of AR include immunomodulatory, anti-hyperglycemic, anti-oxidant, anti-aging, anti-inflammatory, anti-viral, anti-tumor, cardioprotective, and anti-diabetic effects, with minimum side effects. Currently, it is known that polysaccharides, saponins, and flavonoids are the indispensable components of AR. In this review, we will elaborate the research advancements of AR on ethnobotany, ethnopharmacological practices, phytochemicals, pharmacological activities, clinical uses, quality control, production developments, and toxicology. Medicinal plants are rich in a variety of secondary metabolites with therapeutic value. However, the yields of these metabolites are generally very low, making their extraction both time-consuming and labour-intensive. Transcription factor-targeted secondary metabolic engineering can efficiently regulate the biosynthesis and accumulation of secondary metabolites in medicinal plants. v-Myb avian myeloblastosis viral oncogene homolog (MYB) transcription factors are involved in regulating various morphological and developmental processes, responses to stress, and the biosynthesis of secondary metabolites in plants. This review discusses the biological functions and transcription regulation mechanisms of MYB transcription factors and summarizes research progress concerning MYB transcription factors involved in the biosynthesis of representative active components. In the transcriptional regulatory network, MYB transcription factors regulate multiple synthase genes to mediate the biosynthesis of active compounds. The selection of appropriate solvents in the extraction process plays a crucial role in determining the nature and quantity of secondary metabolites extracted from medicinal plants. This study investigates the impact of solvent polarity on the extraction yield, phenolic content, and antioxidant properties of seeds from four plants belonging to the Malvaceae family: Abutilon fruticosum, Abutilon pannosum, Sida ovata and Malvastrum coromendelianum. The seed powder of each species was subjected to extraction using solvents with varying polarities, including acetone, chloroform, methanol, and water. Physicochemical analysis, extraction yield, quantitative phenolic content, and antioxidant activity were assessed. Total phenolic content (TPC) was determined using the Folin–Ciocalteu reagent, and antioxidant activity was measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Analysis of variance (ANOVA) revealed that the choice of extracting solvents significantly influenced (p < 0.05) extraction yield, phenolic content, and antioxidant properties. In conclusion, water and methanol emerged as the most effective solvents for achieving high extraction yields, phenolic compound content, and free radical scavenging activities. The results demonstrated a robust correlation between total phenolic content (mg of Gallic acid equivalent/g of sample) and free radical scavenging activity (IC50). The study highlights the substantial impact of solvent type, suggesting the potential benefits of using a combination of polar and nonpolar solvents to enhance the extraction efficiency of phytochemicals, phenolic content and antioxidant quality from medicinal plant seeds. Objectives: Roscoea is a Sino-Himalayan alpine genus in pantropical family Zingiberaeae. As traditional Tibetan medicinal plants, many species of this genus are threatened by digging, logging, land clearance, grazing and climate change. Roscoea debilis is an endemic species in the Hengduan Mountains with a narrow distribution range. In this study, the assembled and annotated genome of Roscoea was presented in order to furnish significant resources for comparative and functional genomic investigations. The first complete reference genome of Roscoea is expected to shed light on research on conservation and evolutionary biology. Data description: A chromosome-level genome of 1601.04 Mb was obtained for R. debilis by combining Illumina short reads (107.28 Gb) and PacBio Hi-Fi reads (64.08 Gb), achieving high-quality sequencing coverage of roughly 67 × and 40 ×. The assembly was additionally assisted by 271.65 Gb Hi-C data (169 ×), which resulted in a contig N50 of 136.17 Mb and a scaffold N50 of 90.48 Mb. Benchmarking Universal Single-Copy Orthologs (BUSCO) assessment results revealed that most of the core embryophyta genes (98.7%) in the BUSCO dataset (embryophyta_odb10) were successfully identified. Background: Traditional ecological knowledge (TEK) and practice associated with wild edible plants (WEPs) is an important part of local culture of the Yao people in Jianghua Yao Autonomous County, which possesses the biggest population of Yao ethnic group in China. Economic development, urbanization, and transition of lifestyle and changing interests of the younger generations risk loss of such valuable knowledge. However, no record had been reported about WEPs from the Yao communities of Jianghua County. It is urgent to assemble data on the major WEPs and their uses in Jianghua. This can be used to educate and stimulate new interest in these WEPs, to aid inheritance and improvement of cultural identity and confidence, to enhance local resilience to various changes and to suggest pathways for novel and value-added applications to create new local business opportunities. Methods: To record WEPs associated with TEK and practice, we conducted field surveys in 2018, 2019 and 2023. The ethnobotanical methods such as free-listing, participatory observation, semi-structured interview, and market survey were adopted in the field investigations. Information about WEPs including vernacular names, parts used, ways of use, and collection time were recorded. Voucher specimens were collected, identified, and deposited in the herbarium at Minzu University of China, in Beijing. Results: Totally, 81 Yao people from 12 villages were interviewed. WEPs traditionally play a crucial role in Yao peoples' daily lives. A total of 130 plant species from 89 genera and 49 families were recorded. The plants collected showed great diversity and reflected unique local culture. Most (54.5%) WEPs were also used as medicines. Baba can be defined as a cultural identity food, and WEPs are commonly used as tea substitutes for their health benefits. Conclusions: We conclude that WEPs use is facing great challenges in a rapidly changing era. Preservation of WEPs related practice and knowledge is not only important for conserving local biocultural diversity, but also valuable in providing potential functional and healthy materials for food security and future economic development. Don commonly known as 'Himalayan horsetail', has been traditionally used in the treatment of back pain, bone fracture and dislocation, and arthritis by various tribal communities of India. Our previous study confirmed the anti-inflammatory efficacy of the plant through in silico, in vitro, and in vivo model studies. Therefore, the current research is focused on safety dose evaluation for the first-time of the whole-plant methanol extract (EDME) of E. diffusum through appropriate in silico, in vitro, and in vivo approaches. Method: The whole plant, along with its rhizomes, was collected, and the methanol extract was prepared. The in silico ADMET study was performed to predict the pharmacokinetics profile and toxicity of all the identified phyto-compounds of EDME previously screened by GC–MS study. In vitro cytotoxicity study of EDME was performed using two cell lines: kidney (HEK293) and liver (Huh7) cell lines. The in vivo toxicity study of EDME was validated by the acute toxicity (OECD 423, 2002) and sub-acute toxicity assays (OECD 407, 2008) in the Wistar Albino rat model. Results: The in silico ADMET study of all 47 bioactives predicted good pharmacokinetic and low toxicity profiles. In vitro cytotoxicity showed higher IC50 values of EDME viz., 672 ± 15.7 μg/mL and 1698 ± 6.54 μg/mL for both kidney (HEK293) and liver (Huh7) cell lines, respectively, which were considered as low-toxic. Based on acute oral toxicity, the LD50 value of the extract was considered "non-toxic" up to a feeding range of 2000 mg/kg of body weight. The regular consumption of the extract for an extended period (28 days) was also qualified as safe based on the body and organ weight, hematological, biochemical, and histoarchitecture results in the sub-acute toxicity assay. Conclusion: The detailed in silico, in vitro, in vivo (acute and sub-acute oral toxicity) studies gave us a new insight to the safety dose evaluation of Equisetum diffusum, which may serve as a reliable documentation for undertaking the experimental validation of the ethnobotanical uses of the plant which would help in the field of drug development for the treatment of inflammation related complications. This study evaluated and compared the phytochemical and antioxidant properties of the solvent extracts of Azadirachta indica A. Juss and Vernonia amygdalina Del leaves. Methanolic and aqueous extracts showed high (P ≤ 0.05) extract yields (in %), compared to chloroform and ethyl acetate extracts from both V. amygdalina and A. indica leaves. The study exhibited high phytochemical content in methanol and aqueous extracts compared to chloroform and ethyl acetate extracts, confirming the potential for medicinal use. V. amygdalina methanol and aqueous extracts had higher (P ≤ 0.05) total phenolic content (TPC), in mg GAE/gDW, (158.810±0.846 and 217.883±0.265, respectively) than chloroform (37.574±0.118) and ethyl acetate (104.758±0.236) but higher ethyl acetate content in A. indica extracts. Low polar solvents extracted high (P ≤ 0.05) total flavonoids, in mgQE/gDW, (367.051±0.858 and 149.808±0.009) compared to high polar solvents (14.863±0.071 and 54.226±0.014) in V. amygdalina while as in A. indica leaf extracts, low polar solvents showed high TFC (658.469±3.451 and 275.288±10.490) compared to high polar solvents (26.312±0.063 and 48.858±0.063) respectively. In vitro total antioxidant capacity, in mg/g, was higher in polar solvents than in low-polar solvents, ranging from 34.300±1.784 to 121.015±6.839 for A. indica ethyl acetate and methanolic extracts. A strong correlation between TPC and tannic acid content was observed, except in A. indica methanolic extracts of A. indica. Ferric reducing power was high, except for V. amygdalina chloroform and methanol leaf extracts, which were lower (P≤ 0.05) than that of the standard ascorbic acid. The study revealed that high polar solvents, such as methanol and water, are more efficient in the extraction of antioxidants from A. indica but lower in V. amygdalina extracts. Ethnomedicinal plants are thought to have better prospects of harboring endophytes that produce natural products with pharmacological activities. This study aimed to investigate the antiplasmodial and anticancer properties of secondary metabolites of endophytic fungi from three medicinal plants. The endophytic fungi included Lasiodiplodia theobromae isolated from Cola acuminata, Curvularia lunata Bv4 isolated from Bambusa vulgaris, and Curvularia lunata Eg7 isolated from Elaeis guineensis. The identification of the fungi was based on the internal transcribed spacer (ITS-rDNA) sequence. The fungi were subjected to solid-state fermentation and the secondary metabolites were extracted with ethyl acetate. In vitro antiplasmodial screening of extracts was performed using the SYBR green I-based fluorescence assay on the chloroquine-resistant Plasmodium falciparum strain DD2. The cytotoxicity of the extracts on human red blood cells and Jurkat (leukemia) cells was assessed using the tetrazolium-based colorimetric MTT assay. Gas chromatography-mass spectrometry (GC–MS) analysis was used to identify the constituents of the fungal extracts. The extract of L. theobromae showed the best antiplasmodial activity against chloroquine-resistant P. falciparum (IC50 = 5.4 µg/mL) and was not harmful to erythrocytes (CC50 > 100 µg/mL). All three fungal extracts showed a weak cytotoxic effect against Jukart cell lines (CC50 > 100 µg/mL). GC–MS analysis of the three endophytic fungal extracts revealed the presence of forty major bioactive compounds, including: oxalic acid, isobutyl nonyl ester, 2,4-di-tert-butylphenol, and hexadecanoic acid, among others. The endophytic fungi from the medicinal plants in this study were promising sources of bioactive compounds that could be further evaluated as novel drugs for the treatment of malaria caused by P. falciparum-resistant strains. SUMMARY Astragaloside IV is a significant chemical component derived from the medicinal plant Astragalus membranaceus. Despite the characterization of several glycosyltransferases from A. membranaceus, the complete biosynthetic pathway of astragaloside IV has not been fully elucidated. In this study, we propose a biosynthetic pathway for astragaloside IV that involves a sequence of oxidation–reduction reactions. The biosynthesis pathway from cycloastragenol to astragaloside IV encompasses four key steps: C‐3 oxidation, 6‐O‐glucosylation, C‐3 reduction, and 3‐O‐xylosylation. We identified a hydroxysteroid dehydrogenase AmHSD1 from A. membranaceus. AmHSD1 catalyzes the C‐3 oxidation of cycloastragenol, yielding cycloastragenol‐3‐one, and the C‐3 reduction of cycloastragenol‐3‐one‐6‐O‐glucoside, resulting in cycloastragenol‐6‐O‐glucoside. Additionally, the glycosyltransferases AmGT8 and AmGT1, previously reported by our groups, were identified as catalyzing the 6‐O‐glucosylation and 3‐O‐xylosylation steps, respectively. Astragaloside IV was successfully synthesized in transient expression in Nicotiana benthamiana using the combination of AmHSD1, AmGT8 and AmGT1. These results support the proposed four‐step biosynthetic pathway and suggest that AmHSD1 probably plays a crucial role in the biosynthesis of astragaloside IV within A. membranaceus. AbstractThe present study aims to document and analyze information related to significant ethnomedicinal plants in rural communities of Northeastern Madhya Pradesh, India. Data collection involved the use of open-ended and semi-structured questionnaires to gather information on ethnomedicinal plant usage, including specific plants, applications, and traditional healing practices. Quantitative indices such as Relative frequency of citation (RFC), Use value (UV), Family use value (FUV), Informant consensus factor (ICF), and Jaccard’s coefficient of similarity (JCS) were employed to quantify and analyze the gathered information. A total of 201 medicinal plants, belonging to 170 genera and 76 families, were recorded for treating various diseases, with trees (43%), herbs (33%), and climbers (13%) being predominantly used. Plants such as Hedychium coronarium and Cordia macleodii exhibited high RFC and high UV, particularly for treating eye and blood pressure problems. Vanda tessellata showed a high informant consensus factor for treating ear-related problems. Positive correlations were found between RFC and UV (0.631) and RFC and FUV (0.159). The study highlights the prevalence of traditional healing practices using medicinal plants in the area. Documentation of ethnomedicinal species and their therapeutic applications could stimulate further exploration into phytochemical and pharmacological investigations, potentially leading to the development of novel pharmaceuticals. Background: Snakebites are considered a neglected tropical disease responsible for many accidents, some fatal, and are related to poverty. The Brazilian Amazon has the highest incidence of snakebites per inhabitant, with the state of Pará having the most reported cases. For those who have difficult access to hospitals and pharmacies, this issue is even more urgent. Methods: In this research, we worked together with the population of five riverine communities in Aritapera (Santarém-PA), a fluvial island located in the Lower Amazon floodplain (várzeas), in order to identify the species of venomous snakes and create a record of snakebites in the region and treatments carried out. Results: Dwellers reported a high frequency of encounters throughout the year and mentioned five venomous ethnospecies, although we identified only Bothrops atrox. Approximately 28.7% of the participants had already been bitten, and in 15.8% of the interviews, they mentioned deaths from snakebites. The treatments varied between hospitalization (42.8%), home treatments (23.8%), both together (25.4%) and healers (7.9%). There were cases where no treatment, or just religious treatment, was performed. In general, no serious sequelae were reported. Although home treatments were more common in the past, many people maintained the practice of using them before going to the hospital. Among the most used are Pau X and the fat of the Amazon River Dolphin. The latter appears to be a recent discovery by locals and is considered very efficient both for humans and animals. Conclusion: Difficult access to health centers, a lack of energy to store antivenom and a high rate of encounters with snake place Aritapera dwellers in a vulnerable situation regarding snakebite accidents. In this context, they discovered treatments that improved their well-being until hospitalization. As the Amazon River dolphin is an endangered species, the use of its fat requires attention. In this sense, the dissemination of this knowledge is important to encourage studies that investigate which properties of this fat act as counterpoisons. By discovering substitutes that can be incorporated in other rural and remote communities, an economic and ecologically viable option for the health of residents can be promoted, in addition to valuing traditional knowledge. AbstractLitsea costalis is a well-known medicinal plant in tropical and subtropical areas. Four significant flavonoids were extracted and characterized from the plant leaf extract. One component was discovered for the first time in flavonoid chemistry, flavan-4-ol, (3′, 4′-methylenedioxy-5,7-dimethoxy chroman-4-ol). Three flavonoids were discovered for the first time in this plant. The detected products are (+)-pinostrobin, (+)-onysilin, and (+)-pinocembrin. The fourth component was characterized for the first time in this plant and introduced in flavonoid chemistry. Societal Impact Statement Summary As the second‐leading cause of mortality worldwide, cancer is a major focus of drug discovery research. Traditional plant knowledge can guide the search for undiscovered compounds, but the efficacy of this approach for cancer, a highly complex disease affecting diverse tissues, is unknown. We investigated the patterns underlying plant selection for cancer treatment globally, finding certain lineages are repeatedly targeted. While this indicates therapeutic value, their relatedness with plants used for unrelated ethnobotanical uses suggests that plants are probably selected to treat cancer‐associated symptoms, rather than addressing tumour growth. Careful re‐examination and scoring of ethnobotanical reports may make the prediction of lineages for drug discovery more informative. Cancer is a highly diverse disease and as the second‐leading cause of death worldwide is a focus of drug discovery research. Natural products have been shown to be a useful source of novel molecules for treating cancer. It is likely there are many plants with undiscovered molecules of therapeutic value, however identifying new leads from the vast diversity of plants is very challenging. Traditional knowledge might inform bioprospecting by predicting lineages of plants rich in therapeutically useful molecules. We characterise the phylogenetic diversity of plants used in traditional cancer management using a comprehensive genus‐level phylogeny of angiosperms, and a list of 597 genera used globally to treat different cancers. We phylogenetically predict which lineages may have elevated potential for drug discovery and assess the quality of the prediction. We demonstrate the independent and repeated targeting of specific lineages of plants by different peoples in different parts of the world. However, the lineages we report here as rich in plants used in traditional cancer management coincide with those for other ethnobotanical applications and contain few plants with proven anti‐cancer activity. That the same lineages are used to treat different cancers is suggestive of independent discovery of therapeutic value. However, it is likely that the traditional knowledge explored here is shaped by the selection of plants conferring milder effects for treating wider symptoms, such as tiredness or nausea, rather than for halting tumour growth. Background: cultivated and wild plants are used to treat different ailments. The Astragalus genus is found in temperate and dry climates; thus, it is found in Egypt and the arab world. Astragalus caprinus has a good amount of bioactive chemicals, which may help explain its therapeutic effects in reducing the risk of consequences from disease. Method: The phytochemical investigation of the herb and roots of Astragalus caprinus L. included the analytical characterization for the petroleum ether components by GC/MS, unsaponifiable matter (unsap. fraction), and fatty acids (FAME) investigation by GLC analysis. Main flavonoids were chromatographically isolated from ethyl acetate and n-butanol extracts. In vitro antimicrobial activity has been tested against the Gram-positive bacteria Staphylococcus aureus and Streptococcus mutans for different plant extracts, the Gram-negative bacteria Pseudomonas aeruginosa and Klebsiella pneumonia, the fungus Candida albicans and Aspergillus niger, and the Escherichia coli bacterium. Metabolite cytotoxicity was examined using the MTT assay against HepG-2 (human liver carcinoma) and MCF-7 (breast carcinoma). Results: Identifying the important components of the herb and root petroleum ether extracts was achieved. Using column chromatography, luteolin, cosmosiin (apigenin-7-O-glucoside), and cynaroside (luteolin-7-O-glucoside) were separated and identified using UV, NMR, and Mass Spectroscopy. Root extracts displayed potential antimicrobial activity against most of the tested pathogens. Both extracts (herb and roots) were active against the MCF-7 cell line and HepG-2 cell line with IC50 62.5 ± 0.64 and 72.4 ± 2.3 µg/ml, and 75.9 ± 2.5 and 96.8 ± 4.2 µg/ml, respectively. Conclusion: Astragalus caprinus seems to be a promising source of bioactive compounds that could potentially aid in preventing disease complications and address common health issues in developing countries. Moreover, the various parts of this plant could be utilized as natural raw materials for producing health-boosting products that could address common health issues in developing countries.
