PGE2 is produced at high levels during inflammation, and its suppressive effects are caused by the ligation of the E prostanoid receptors EP2 and EP4, which results in the production of cyclic AMP. However, PGE(2) also exhibits immunostimulatory properties due to binding to EP3, which results in decreased cAMP levels. The various guanine nucleotide-binding proteins (G proteins) that are coupled to the different EP receptors account for the pleiotropic roles of PGE(2) in different disease states. Here, we discuss the production of PGE(2) and the actions of this prostanoid in phagocytes from different tissues, the relative contribution of PGE(2) to the modulation of innate immune responses, and the novel therapeutic opportunities that can be used to control inflammatory responses. O processamento e apresentação de peptídeos próprios oriundos das células apoptóticas via MHC, associado a presença do patógeno, permite a indução de um fenótipo imunogênico nessas CD, e, portanto, propiciando o ambiente favorável para ativação de clones autorreativos. A infecção pelo vírus Zika (ZIKV) vem sendo amplamente estudada devido às complicações neuronais no desenvolvimento fetal e em adultos, como a microcefalia e síndrome de Guillain-Barré, respectivamente. Considerando que a infecção pelo ZIKV induz intensa morte celular por apoptose em diferentes tipos celulares, a hipótese deste estudo fundamenta-se que a fagocitose de células apoptóticas infectadas pelo ZIKV levaria a ativação de CD com um perfil imunogênico. Para investigar o efeito da eferocitose na ativação de células dendríticas, inicialmente duas cepas viras e diferentes linhagens celulares foram testadas para obtenção de células infectadas por ZIKV. O potencial de infecção das cepas virais ZIKV PE243 (linhagem asiática isolada no Brasil) e ZIKV MR766 (linhagem africana isolada em Uganda) foram testadas utilizando cinco linhagens celulares: HaCaT, A549, Huh7.5, C6 e SH-SY5Y. As linhagens HaCaT, A549 e C6 apresentaram baixa infectividade, enquanto as linhagens Huh7.5 e SH-SY5Y mostraram alta taxa de infecção pela cepa ZIKV PE234. A eferocitose de células Huh7.5 infectadas pelo ZIKV não foi capaz de induzir um aumento na expressão de moléculas de MHC-II em células dendríticas derivadas de medula óssea (BMDC) e apresentou uma produção mista de Il-6 e IL-10, sugerindo um perfil não definido. No entanto, a eferocitose de células da linhagem neuronal SH-SY5Y infectadas pelo ZIKV levou ao aumento na expressão das moléculas CD86 e MHC-II nas BMDC, além de uma maior produção das citocinas IL-1β, IL-6 e TGF-β quando comparada ao controle de células SH-SY5Y não infectadas. Esse perfil de BMDC e de citocinas inflamatórias após a eferocitose de SH-SY5Y infectadas pelo ZIKV reforça nossa hipótese inicial, ou seja, que apresenta potencial para ativar células T autorreativas. No entanto, novos experimentos utilizando células T isoladas de animais transgênicos OT-II serão necessários para confirmar essa hipótese. Recent studies show that phagocytosis of infected apoptotic cells by dendritic cells (CD) results in activation of self-reactive CD4+ T lymphocytes clones and the development of autoimmune diseases. The processing of self-peptides associated with the presence of the pathogen inside the apoptotic cells allows the induction of an immunogenic phenotype in these dendritic cells, thus providing a favorable environment for activation of self-reactive T CD4+ clones. Zika virus (ZIKV) infection has been widely studied due to neuronal complications in fetal development and in adults such as microcephaly and Guillain-Barré syndrome, respectively. Considering that ZIKV infection induces intense apoptotic cell death in different cell types, the hypothesis of this study was that the phagocytosis of ZIKV infected apoptotic cells would lead to activation of dendritic cells in an immunogenic profile. To investigate the effect of efferocytosis of infected cells on dendritic cell activation, two strains and different cell lineages were tested to obtain ZIKV infected cells. Infection potential of viral strains ZIKV PE243 (Asian strain isolated in Brazil) and ZIKV MR766 (African strain isolated in Uganda) were tested using five cell lineages: HaCaT, A549, Huh7.5, C6 e SH-SY5Y. HaCaT, A549 and C6 cell lineages showed low susceptibility to the ZIKV PE243 strain while Huh7.5 and SH-SY5Y showed high infection rate. Efferocytosis of ZIKV infected Huh7.5 cells was not able to induce an increase in MHCII molecule expression in bone marrow-derived dendritic cells (BMDC) and a mixed cytokine production of IL-6 and IL-10, indicating non defined profile. However, the eferocytosis of cells from the ZIKV infected SH-SY5Y neuronal lineage caused a higher expression of CD86 and MHCII molecules in BMDC, as well as a higher production of IL-1β, IL-6 and TGF-β cytokines when compared to uninfected SH-SY5Y cells. This immunogenic profile of BMDC and inflammatory cytokines produced after the coculture of ZIKV-infected SH-SY5Y confirms our initial hypothesis, that there is potential to activate autoreactive T cells. Esse prostanoide vem sendo descrito como um importante mediador envolvido no agravamento da Encefalomielite Autoimune Experimental (EAE). A EAE é uma doença mediada por células Th1/Th17 autorreativas, responsáveis pela intensa resposta inflamatória contra antígenos do sistema nervoso central (SNC). Alguns estudos descrevem que a inibição da síntese desse prostanoide, ou o bloqueio de seus receptores EP, reduzem os níveis de IL-17A e IFN- e atenuam drasticamente o desenvolvimento da doença. A coexistência de linfócitos Th1 e Th17 na EAE, assim como a presença de células Th17 produtoras de IFN-γ (Th1-like) no SNC sugerem uma possível plasticidade destas subpopulações de linfócitos. No entanto, até o momento, não há relatos na literatura se a presença de PGE2, presente no SNC, estaria envolvida na plasticidade de linfócitos Th17 em Th1 nessa autoimunidade. Portanto, a hipótese desse estudo é que as células Th17 migrariam para o SNC e desencadeariam o recrutamento de células inflamatórias e o aparecimento dos primeiros sinais clínicos da doença. A presença de PGE2, associado esse microambiente inflamatório, favoreceria a plasticidade das células Th17 para um padrão Th1, resultando na diferenciação de células T CD4+ patogênicas (IL17+IFN+) e células Th1-like. Os resultados obtidos demonstram que as células Th17, quando cultivadas em condições polarizantes para Th1, foram hábeis em se converterem em linfócitos Th1-like, porém, a presença de PGE2 aumenta a porcentagem de células T CD4+IFN-γ+, via EP4 e, possivelmente, pela via de adenilato ciclase/PKA. Além disso, o tratamento com indometacina, em animais Il17cre/mTmG imunizados com MOG, durante a fase aguda da doença, resultou na diminuição de células Th17 produtoras de IFN-γ no SNC de camundongos que desenvolveram a EAE, sem causar a alteração no score clínico da doença e no infiltrado celular. Prostaglandin E2 (PGE2) is a lipid mediator which participates in both differentiation and expansion of T helper (Th) Th1 and Th17. This prostanoid has been described as an important mediator involved in the worsening of EAE, an experimental model for Multiple Sclerosis in humans. EAE is a disease mediated for autoreactive Th1/Th17 cells, responsible for an intense inflammatory response against central nervous system (CNS) antigens. Some studies have described that inhibition of PGE2 synthesis or blockage of its receptors decrease IL-17A and IFN- levels and attenuate drastically the disease development. The coexistence of Th17 and Th1 lymphocytes together with IFN-γ-producing Th17 cells (Th1-like) at CNS suggests a possible plasticity between these subtypes of lymphocytes in EAE. However, until now, there are no reports in the literature if the presence of PGE2 in the CNS could be involved in Th17 to Th1 plasticity during EAE. Therefore, the hypothesis of this study is that Th17 cells could migrate to the central nervous system and trigger the recruitment of inflammatory cells and the appearance of EAE first clinical symptoms. The presence of PGE2, associated with the inflammatory microenvironment, could induce Th17 plasticity to Th1 lymphocytes, resulting in the differentiation of pathogenic T CD4+ (IL17+IFN+) and Th1-like cells, responsible for tissue damage. Recent data from our research group showed that Th17 cells, when cultured in Th1 polarizing condition, were capable to differentiate into Th1-like lymphocytes, however, in the presence of PGE2, T CD4+IFN-γ+ cells percentage is increased, through EP4 signaling, possible through cyclase adenylate/PKA activation pathway. Furthermore, the treatment of Il17cre/mTmG mice immunized with MOG with indomethacin, during the acute phase of the disease, impairs IFN-γ producing Th17 cells inside the CNS of mice which developed EAE, however, no change in the clinical score of the disease and cellular infiltrate was observed. Como consequência deste processo, há a produção dos mediadores anti-inflamatórios, fator de transformação de crescimento (TGF-β), prostaglandina E2 (PGE2) e a interleucina-10 (IL-10). No entanto, atualmente sabe-se que a fagocitose de células apoptóticas infectadas direciona a produção de mediadores inflamatórios interleucina-6 (IL-6), interleucina-23 (IL-23) e TGF-β, Resultados obtidos recentemente por nosso grupo demonstram que a fagocitose de células apoptóticas infectadas (iAC) com Escherichia coli por células dendríticas promove, além da produção de TGF-β, interleucina-1β (IL-1β) e IL-6, a síntese de altos níveis de PGE2. Além disso, observamos que a PGE2, via receptor de prostaglandina E2 4 (EP4), inibe a diferenciação de linfócitos Th17 por meio da modulação da expressão do receptor de IL-1 (IL-1R) em linfócitos Th. Desta forma, o objetivo deste trabalho é elucidar o mecanismo pelo qual PGE2, oriunda da eferocitose de células apoptóticas infectadas, inibe a expressão de IL-1R e a diferenciação de linfócitos Th17. Os resultados obtidos demonstram que PGE2, via receptor EP4, induz a ativação de adenilato ciclase/PKA/EPAC, e que esta via de sinalização promove além da inibição de IL-1R, a inibição de um importante fator de transcrição envolvido na diferenciação de linfócitos Th17, STAT3. Sabe-se que a PGE2 pode exercer suas funções supressoras na diferenciação de células Th17 pela ativação de SOCS, bem como promover a ativação de PI3K. No entanto, apesar da presença de PGE2 aumentar a expressão de socs1, a inibição da fosforilação de STAT3 não parece ser mediada por SOCS1, tão pouco PI3K está envolvida na inibição da diferenciação de células Th17. O conjunto de resultados sugere que a inibição na diferenciação de linfócitos Th17 por PGE2, durante a eferocitose de células infectadas, ocorre através da ativação do eixo EP4/adenilato ciclase/PKA/EPAC, que resulta na inibição direta ou indireta da forforilação de STAT3. The expression of inflammatory reactions is characterized by phagocytosis of dead cells, a process called eferocytosis. As a consequence of this process, there is the production of anti-inflammatory mediators, transforming growth factor (TGF-β), prostaglandin E2 (PGE2) and an interleukin-10 (IL-10). However, it is now known that phagocytosis of infected apoptotic cells is one of the responsible for the production of interleukin-6 (IL-6), interleukin-23 (IL-23) and TGF-β lung mediators. The results obtained by our group demonstrate that the phagocytosis of infected apoptotic cells (iCA) with Escherichia coli by dendritic cells promotes, in addition to the production of TGF-β, interleukin-1β (IL-1β) and IL-6, high levels of PGE2. In addition, PGE2, via the prostaglandin E2 receptor (EP4), inhibits differentiation of Th17 lymphocytes by modulating IL-1 receptor (IL-1R) expression in Th lymphocytes. Thus, the objective of this work is to elucidate the mechanism by which PGE2, derived from the ectocytosis of infected apoptotic cells, inhibits the expression of IL-1R and differentiation of Th17 lymphocytes. The results obtained demonstrate that PGE2, via the EP4 receptor, induces the activation of adenylate cyclase / PKA / EPAC, and that this signaling pathway promotes in addition to the inhibition of IL-1R, the inhibition of an important transcription factor involved in the differentiation of Th17 lymphocytes, the STAT3. PGE2 is known to exert its suppressor functions in the differentiation of Th17 cells by the activation of SOCS, as well as to promote the activation of PI3K. However, despite the presence of PGE2 enhancing socs1 expression, inhibition of STAT3 phosphorylation does not appear to be mediated by SOCS1, so little PI3K is involved in inhibiting Th17 cell differentiation. The set of results suggests that the inhibition in differentiation of Th17 lymphocytes by PGE2 during the eferocytosis of infected cells occurs through the activation of the EP4 / adenylate cyclase / PKA / EPAC axis, which results in the direct or indirect inhibition of STAT3 forforylation. 2016/10964-5

Há diferentes terapias disponíveis, como os antifúngicos da classe dos poliênicos, azóis e sulfonamidas. O desenvolvimento de novos antifúngicos com eficácia superior é necessário para prevenir recaídas e diminuir os efeitos secundários, o período de tratamento e sequelas. A molécula de ácido gálico com diferentes modificações (galatos de alquila) apresentou atividade antifúngica contra um painel representativo de fungos patogênicos. O galato de decila (G14) foi objeto de estudo deste trabalho, para o qual a interação Paracoccidioides¬-hospedeiro foi relacionada ao mecanismo de ação. Para tanto, o fungo Paracoccidioides lutzii (Pl01) foi desafiado com o G14 para análise das alterações no desenvolvimento de brotamentos e na viabilidade celular pelo ensaio de MTT. Foi avaliada a influência do G14 na taxa de interação de Pl01 com a linhagem de macrófago alveolar murino AMJ2-C11, com as linhagens pulmonares de mamífero A549 e MRC-5, e hemócitos da larva de Galleria mellonella utilizando ensaios por citometria de fluxo. O G14 ainda foi avaliado quanto à taxa de sobrevivência da larva de G. mellonella infectada e a capacidade na modulação da produção de óxido nítrico em macrófago RAW. O estudo do mecanismo de ação foi realizado pelo ensaio de interação químico-genética seguida da interação genética, que consiste na comparação de uma coleção de mutantes da levedura Saccharomyces cereviseae com perfil hipersensível ao G14, a um banco de dados de duplos mutantes para predizer a via afetada. Em P. lutzii (Pl01) e P. brasiliensis (Pb18) foi avaliada a taxa de atividade de enzimas n-glicosiladas (α-(1,3)-glucanase, N-Acetyl-β-D-glucosaminidase e α-amilase) ligadas à formação da parede celular; assim como a expressão relativa dos genes envolvidos através de PCR em tempo real. No ensaio de interação químico-genética, cerca de 50 mutantes foram hipersensíveis e com a análise da interação genética deduziu-se influência sobre a via de glicosilação e/ou na resposta a proteínas não enoveladas. A concentração de 0,5 µg/mL de G14 apresentou redução significativa de 42 % da atividade metabólica e de 38,5 % na formação de novos brotos do Pl01 com 24 horas de contato, indicando forte influência sobre o desenvolvimento da célula fúngica. As enzimas n-glicosiladas testadas para Pl01 e Pb18 apresentaram baixa atividade quando desafiadas com G14, porém os genes referentes a estas enzimas não apresentaram expressão relativa baixa, inferindo a influência nas modificações pós-traducionais destas enzimas. A influência do G14 na atividade funcional destas enzimas resulta na instabilidade da parede celular, a qual é um elemento crucial para a virulência e para a interação Paracoccidioides-célula hospedeira. Assim, foi observada redução na taxa de interação do Pl01 de 1,6 vezes com a linhagem A549, e de 1,2 com a linhagem AMJ2-C11. Além disso, foi observado um aumento no tempo de sobrevivência da larva infectada com Pl01 e tratada, indicando que G14 tem propriedades benéficas ao hospedeiro, o que foi observado com a redução de 20 % na quantificação de óxido nítrico quando macrófagos RAW estimulados por LPS foram desafiados com G14, o que sugere uma atividade antioxidante. The fungus of genus Paracoccidioides is the etiological agent of paracoccidioidomycosis (PCM), an important mycosis in Brazil and Latin America. There are different available therapies, as antifungals belong to the class of polyenes, azoles and sulfonamides. The development of new antifungal agents with greater effectiveness is required to prevent relapse and reduce side effects, the period of treatment and sequelae. The gallic acid molecule with different modifications (alkyl gallates) showed antifungal activity against a representative panel of pathogenic fungi. The decyl gallate (G14) was the study object of this work, for which the Paracoccidioides-host interaction was connected with the mechanism of action. Therefore, the fungus Paracoccidioides lutzii (Pl01) was challenged with the G14 to analyze the changes in the development of budding and cell viability by MTT assay. Then, it was assessed the influence of the G14 in the interaction rate of Pl01 with murine alveolar macrophage lineage AMJ2-C11, with mammalian lung lineage A549 and MRC-5, and hemocytes of Galleria mellonella larvae using assays by flow cytometry. The compound G14 was evaluated for survival rate of infected G. mellonella larvae and to capacity of modulation of nitric oxide production in RAW macrophages. The mechanism of action was performed by chemical-genetic interaction assay followed by genetic interaction, which involves comparing a mutant collection, of Saccharomyces cerevisiae yeast, with hypersensitive profile to G14 to double mutant database to predict via affected. In P. lutzii (Pl01) and P. brasiliensis (Pb18) was evaluated the rate of activity of n-glycosylated enzymes (α- (1,3) -glucanase, N-Acetyl-β-D-glucosaminidase and α-amylase) linked to cell wall formation; as well as the relative expression of the genes involved by real time PCR. In the test chemical-genetic interaction, about 50 mutants were hypersensitive to G14 and with the genetic interaction analysis was deduced that it influence the glycosylation pathway and/or in the response unfolded proteins. The concentration of 0.5 µg/ml of G14 after 24 hours of contact showed a significant reduction of 42% in metabolic activity and 38.5% in the formation of new buds of Pl01, indicating a strong influence on the development of the fungal cell. The N-glycosylated enzymes tested for Pl01 and Pb18 after 72 hours of contact with 0.5 µg/ml of G14 showed low activity when challenged with G14, but the genes related to these enzymes did not show low relative expression, inferring the influence on the post-translational modification of these enzymes. The influence of the G14 in the functional activity of these enzymes results in the instability of the cell wall, which is crucial to the virulence and the host Paracoccidioides-cell interaction. Thus, it was observed a reduction in the interaction rate of Pl01 in 1.6 times with lineage A549 and in 1.2 times with lineage AMJ2-C11. Besides that, it was observed an increase of survival time of Pl01infected larvae and treated, indicating G14 has properties that benefit the host, what was observed with a reduction of 20% in the quantification of nitric oxide when RAW macrophages stimulated by LPS were challenged with G14, which showed an antioxidant activity.

Recentes resultados obtidos por nosso grupo demonstram que a fagocitose, por CD, de células apoptóticas infectadas resulta também na produção de altos IL- 1 e PGE2. A presença destes mediadores solúveis foi capaz de induzir a diferenciação de células Th17, in vitro, no entanto, a inibição da síntese de PGE2, pelo tratamento com inibidor não seletivo da COX (Indometacina), resultou no aumento da porcentagem destes linfócitos. Neste estudo foi avaliado o efeito da fagocitose de células apoptóticas infectadas com Escherichia coli (CA+Ec) na diferenciação de células Th17 in vivo e o efeito de PGE2 neste processo. A instilação de animais com CA+Ec, induziu a produção das citocinas IL-6, IL-1β e TGF-β no pulmão, o que sugere que a presença de CA+Ec é capaz de gerar um microambiente favorável para induzir a diferenciação Th17. A instilação de CA+Ec promoveu a diferenciação de células Th17, assim como a produção de IL-17. Porém, o tratamento com inibidor de COX resultou no aumento na porcentagem e no número de células Th17 no pulmão. A presença destas células resultou no aumento na porcentagem de neutrófilos no tecido pulmonar que parece refletir na diminuição da carga bacteriana recuperada do pulmão, quando comparado com animais que não receberam o tratamento. Assim, o conjunto destes resultados sugere que a fagocitose de CA+E. coli promove a diferenciação de células Th17. No entanto, a secreção de PGE2, pela fagocitose destas CA+Ec suprime a diferenciação e/ou migração de células Th17 para o pulmão, levando a inibição do recrutamento de neutrófilos e aumento da suscetibilidade às infecções pulmonares. The phagocytosis of infected-apoptotic cells (iAC) by dendritic cells (DC) induces the production of TGF-β, IL-6 and IL-23, providing a microenvironment to Th17 cell differentiation. Recent data obtained in our group showed that the phagocytosis of infected- apoptotic cells by CD also results in high levels of IL-1β and PGE2. The presence of these soluble mediators was able to induce the differentiation of Th17 cells, in vitro. However, the inhibition of PGE2 synthesis, by non-selective COX inhibitor (indomethacin), result the increase of percentage of Th17. In this study, it was evaluated the effect of phagocytosis of infected-apoptotic cells (AC+E. coli) during the Th17 cell differentiation, in vivo, and the effect of PGE2 in this process. The inoculation of AC+ Ec (in.) into lung induced the production of IL-6, IL-1β and TGF-β in lung, suggesting that the presence of AC+Ec is able to induce a microenvironment favorable to Th17 cell differentiation. The instillation of AC+Ec induced the differentiation of Th17 cells. However, the treatment of the animals with COX inhibitor resulted in an increase in the percentage, and number, of Th17 cells in the lung. The presence of these cells increased in the percentage of neutrophils in the lung and that apparently reflect in bacterial clearance from the lung, compared to animals that received no treatment. Taken all together, the phagocytosis of AC+Ec promotes the Th17 cell differentiation. However, the production of PGE2 by efferocytosis suppresses the differentiation and / or migration of Th17 cells to the lungs, impair the recruitment of neutrophils and increase the susceptibility to lung infections. Os macrófagos além de atuarem na remoção de células apoptóticas, também colaboram na defesa contra microrganismos. Existem ao menos duas populações de macrófagos classificadas como macrófagos M1 (pró-inflamatórios) e macrófagos M2 (anti-inflamatórios). Estes leucócitos diferem quanto ao fenótipo e funções efetoras dependendo, primordialmente, do microambiente e dos microrganismos com que interagem no tecido. Em uma situação de injúria pulmonar estéril (inalação de agentes tóxicos), há acúmulo de células apoptóticas sem a presença de agente microbiano. Por outro lado, durante uma infecção pulmonar bacteriana, ocorre intensa migração de células para o local da infecção na tentativa de conter a proliferação bacteriana, resultando em intenso acúmulo de células apoptóticas infectadas neste tecido. Portanto, nesse trabalho foi avaliado, in vitro e in vivo, o efeito da fagocitose de células apoptóticas infectadas (AC-Sp) ou estéreis (AC) na polarização de macrófagos M1/M2, bem como suas funções efetoras, e o efeito da PGE2 nesse contexto de eferocitose. Macrófagos M0 co-cultivados com AC ou AC-Sp apresentam um fenótipo intermediário entre M1 e M2, com secreção de altos níveis de IL-6, TNF-α, PGE2, TGF-β e NO e a expressão de genes relacionados ao perfil M1, iNOS, e ao perfil M2, Arginase 1. Além disso, macrófagos M0 cultivados com AC ou AC-Sp, apresentam supressão da função microbicida quando desafiados com S. pneumoniae. A presença de PGE2 dirige a polarização de macrófagos M0 a um perfil M2 e a presença de inibidores de COX promove a reversão do fenótipo de polarização destes macrófagos. The phagocytosis of apoptotic cells is dynamic and crucial for homeostasis after injury. Macrophages act on the clearance of apoptotic cells and collaborate with defense against microorganisms. There are at least two distinct macrophage populations classified as M1 macrophages (pro-inflammatory) and M2 macrophages (anti-inflammatory). Both cells differ on the state of polarization and effectors function depending primarily on the microenvironment and microorganisms that interact into the tissue. In a situation of sterile lung injury (inhalation of toxic agents), there are accumulation of apoptotic cells without the presence of pathogen. On the other hand, during a bacterial pulmonary infection there is intense cell migration to the infection site in an attempt to impair bacterial growth, resulting in a large accumulation of infected-apoptotic cells in the tissue. Therefore, this study has evaluated in vitro and in vivo, the effect of the phagocytosis of infected-apoptotic cells (AC-Sp) or sterile cells (AC) on the polarization of M1/M2 macrophages, as well as in their effector functions and the effect of PGE2 in the context of efferocytosis. M0 macrophages co-cultured with AC or AC-Sp show an intermediate phenotype between M1 and M2 with high secretion levels of IL-6, TNF-α, PGE2, NO and TGF-β, and gene expression profile related to M1, iNOS, and M2, arginase 1. Furthermore, M0 macrophages cultured with AC or AC-Sp show strong suppression of the macrophage microbicidal function when challenged with S. pneumonia. The presence of PGE2 drives the polarization of M0 macrophages to a M2 profile, and in the presence of COX inhibitors, it reverses the polarization of this macrophage phenotype. Sabe-se que indivíduos acometidos por doenças pulmonares obstrutivas crônicas (DPOC) são susceptíveis a infecções bacterianas recorrentes e apresentam um intenso acúmulo de células apoptóticas (ACs) no parênquima pulmonar. A hipótese deste estudo fundamenta-se em que o acúmulo de ACs e a fagocitose destas por AMs poderia suprimir as funções efetores destas células, através da liberação de mediadores anti-inflamatórios, tais como TGF- e PGE2. No entanto, nada se sabe quanto aos mecanismos pelos quais PGE2 poderia suprimir esses mecanismos efetores de macrófagos alveolares contra S. pneumoniae, via SR. Nossos resultados demonstram que a presença de ACs promove a inibição da fagocitose e a atividade microbicida de AMs contra S. pneumoniae. A inibição da fagocitose de S. pneumoniae mediada pela eferocitose foi revertida pela inibição da síntese de PGE2 endógena, assim como pelo tratamento com antagonistas deEP2 e pela inibição da enzimaadenililciclase. No entanto, estes efeitos supressores de PGE2 decorrentes da eferocitose por AMs foram independentes da ação de PKA. Desta forma, podemos sugerir que a presença de ACs no pulmão de indivíduos com DPOC poderia desencadear a síntese exacerbada de PGE2 e contribuir na supressão das funções efetoras de AMs contra infecções bacterianas, como S. pneumoniae. Portanto, o tratamento com inibidores da COX, concomitante à terapia antimicrobiana, poderia levar a uma melhora no quadro de imunossupressão desencadeado pela ação de PGE2 no microambiente pulmonar e, portanto, restauração das funções efetoras de AMs The scavenger receptors (SR) are the major receptors involved in the Streptococcus pneumonia phagocytosis by alveolar macrophages (AMs) that act as the first line of defense in the lung. The increase of susceptibility to bacterial infections has been demonstrated in chronic inflammatory pulmonary disease in which there is an intense accumulation of apoptotic cells (ACs). Our hypothesis is that the uptake of ACs by macrophages could suppress immune responses by releasing anti-inflammatory mediators, such as TGF- and PGE2. However, the way in which PGE2 suppress the effector mechanisms against Streptococcus pneumonia by SR in alveolar macrophages is unclear. We found that the preincubation with AC inhibited the ingestion and killing of S. pneumonia by AMs. The inhibition of S. pneumonia phagocytosis by efferocytosis was partially reverted when endogenous PGE2 production was repressed with a COX inhibitor, EP2 antagonist and likewise with an adenylate cyclase inhibitor. However, this suppressive effect was PKA independent. Moreover, we demonstrated that the inhibition of S. pneumonia phagocytosis by efferocytosis was more pronounced in scavenger receptors class B (SR-B). Thus, we suggest that the presence of ACs in the lungs of patients with COPD could trigger the synthesis of PGE2 and promote the suppression of effector functions of AMs against bacterial infections, such as S. pneumoniae. Therefore, treatment with COX inhibitors, concomitant-microbial therapy, could lead to an improvement in the immunosuppression triggered by the action of PGE2 in the lung microenvironment and thus restoration of effector functions of AMs
