Biosensors are common analytical devices, capable of sensing a myriad of biological analytes, including cancer biomarkers. Although biosensors have different transducer types, electrochemical biosensors provide fast analysis time, high sensitivity, and the ability to perform complex measurements such as multiplexed analysis or screening tests for early diagnosis and prognosis of cancer. This chapter describes the background and theory of electrochemical sensors and introduces the main readout techniques. Innovative electrochemical biosensing strategies for analysis and quantification of important early cancer biomarkers, which include circulating nucleic acids (e.g., circulating tumour DNA, gene mutations, and microRNA) proteins, circulating tumour cells, and extracellular vesicles are discussed with the recent developments to provide an overview of the possible academic and clinical approaches

The process of biological aging or senescence refers to the gradual loss of homeostasis and subsequent loss of function – leading to higher chances of mortality. Many mechanisms and driving forces have been suggested to facilitate the evolution of a molecular circuit acting as a trade-off between survival and proliferation, resulting in senescence. A major observation on biological aging and longevity in humans and model organisms is the prevalence of significant sexual divergence in the onset, mechanisms and effects of aging associated processes. In the current account, we describe possible mechanisms by which aging, sex and reproduction are evolutionarily intertwined in order to maintain systemic energy homeostasis. We also interrogate existing literature on the sexual dimorphism of genetic, cellular, metabolic, endocrine and epigenetic processes driving cellular and systemic aging. Subsequently, based on available evidence, we propose a hypothetic model of sex-limited decoupling of female longevity from sirtuins, a major family of regulator proteins of the survival-proliferation trade-off. We also provide necessary considerations to be made in order to test the hypothesis and explore the physiological and therapeutic implications of this decoupling event in male and female longevity after reaching reproductive maturity. Hypothesis Statement Sirtuins provide survival benefits in a sex-nonspecific manner but the dependency on sirtuins in driving metabolic networks after reaching reproductive maturity is evolutionarily decoupled from female longevity.

Municipal solid waste (MSW) represents an inevitable by-product of human activity and a major crisis for communities across the globe. In recent times, the recycling of MSW has drawn attention as the process can add value through resources from the recovered waste materials and facilitates the process of circular economy. However, during the unprecedented coronavirus (COVID-19) outbreak, the risk of infection with the highly contagious virus has proven detrimental to the continuation of MSW as a valuable resource. The volume of waste, especially household waste, is higher; face masks, PPE (personal protective equipment), and hazardous materials such as batteries and empty chlorine bottles are examples of extra waste that have arisen during the pandemic. Various countries have set up initiatives for MSW management, including safety measurements for employees in the MSW management sector. The use of disinfectant prior to sorting waste, as well as storing waste for 9 days, may help to inactivate the COVID-19 virus, ensuring an appropriate safety level for MSW management. This work aimed at studying different MSW management strategies, specific challenges, and possible solutions for better understanding for those involved in waste management, in addition to providing a possible management strategy during and post-COVID-19 pandemic. Endophytic fungi reside in the intercellular space of plant nourished by the plant. In return, they provide bioactive molecules which can play critical roles on plant defense system. Fifty six endophytes were isolated from the leaves, root, bark and fruits of Sonneratia apetala, a pioneer mangrove plant in the Sundarbans, Bangladesh. A total of 56 isolates were obtained and 12 different species within 8 genera were identified using morphological and molecular characteristics. Antimicrobial activity of Ethyl Acetate (EtOAc) and Methanolic (MeOH) extracts of these 12 different species were analyzed by resazurin assay and the Minimum Inhibitory Concentrations (MICs) were determined. The fungal extracts showed antimicrobial activities against more than one tested bacterium or fungus among 5 human pathogenic microbes, i.e. Escherichia coli NCTC 12241, Staphylococcus aureus NCTC 12981, Micrococcus lutus NCTC 7508, Pseudomonas aeruginosa NCTC 7508 and Candida albicans ATCC 90028. Overall, Methanolic extracts showed greater activity than that of Ethyl Acetate extracts. Of the isolates identified, Colletotrichum gloeosporioides, Aspergillus niger and Fusarium equiseti were the most active isolates and showed activity against microorganisms under investigation. Methanolic extracts of C. gloeosporioides and A. niger showed the lowest MIC (0.0024 mg/mL) against Pseudomonas aeruginosa. The study indicates that endophytic fungi isolated from S. apetala species posses potential antimicrobial properties, which could be further investigated.

Knowledge of heterogeneous etiology and pathophysiology of schizophrenia (SZP) is reasonably inadequate and non-deterministic due to its inherent complexity and underlying vast dynamics related to genetic mechanisms. The evolution of large-scale transcriptome-wide datasets and subsequent development of relevant, robust technologies for their analyses show promises toward elucidating the genetic basis of disease pathogenesis, its early risk prediction, and predicting drug molecule targets for therapeutic intervention. In this research, we have scrutinized the genetic basis of SZP through functional annotation and network-based system biology approaches. We have determined 96 overlapping differentially expressed genes (DEGs) from 2 microarray datasets and subsequently identified their interconnecting networks to reveal transcriptome signatures like hub proteins (FYN, RAD51, SOCS3, XIAP, AKAP13, PIK3C2A, CBX5, GATA3, EIF3K, and CDKN2B), transcription factors and miRNAs. In addition, we have employed gene set enrichment to highlight significant gene ontology (e.g., positive regulation of microglial cell activation) and relevant pathways (such as axon guidance and focal adhesion) interconnected to the genes associated with SZP. Finally, we have suggested candidate drug substances like Luteolin HL60 UP as a possible therapeutic target based on these key molecular signatures.

Apart from boosting flavor, herbs and spices are also known for their preservative, antioxidant, antimicrobial and other medicinal properties. Bioactive metabolites with aspiring prospect have guided the present study to screen for antibacterial and antioxidant activity of three Zingiber plants available in Bangladesh. For that purpose, rhizomes of Zingiber officinale, Z. montanum and Z. zerumbet were extracted with ethanol as solvent system for bioactivity analysis. Antioxidant activity was assessed by employing DPPH free radical scavenging assay, reducing power assay and ferric reducing antioxidant power (FRAP) assay. Z. officinale extract demonstrated the highest free radical scavenging activity (IC50: 43.65 µg/ml), reducing power (EC50: 189.026 µg/ml) and FRAP value (137.18 ± 0.116 µM Fe(II)/100 µg dry weight of extract). Moreover, total phenolic, flavonoid and tannin content were determined which significantly correlated (Pearson correlation) with the antioxidant activities. For the evaluation of antibacterial activity, zone of inhibition (ZOI) was measured by disc diffusion bioassay against four Gram positive bacteria Bacillus megaterium, Staphylococcus aureus, Micrococcus spp., and S. epidermidis as well as ten Gram negative bacteria Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi, S. paratyphi, Vibrio cholera, Shigella dysenteriae, S. flexneri, Proteus spp., Mycobacterium spp., and Campylobacter spp. Z. montanum extract exhibited significant activity against E. coli (13.67 ± 0.577 mm ZOI) and Micrococcus spp. (14.33 ± 0.763 mm ZOI) if compared to standard erythromycin with ZOI 15.33 ± 0.577 mm and 15.67 ± 0.577 mm, respectively. Furthermore, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of Z. montanum extract were determined against B. megaterium, Micrococcus spp., E. coli and S. paratyphi. Thus, Z. officinale and Z. montanum extracts could be utilized as source and template for the synthesis of potential drugs and food supplements as well.

The antioxidant and anti-inflammatory potential of Heritiera fomes ethanolic extract was evaluated.Query Ethanolic extraction of dried and grinded plant leaves and bark was performed. Antioxidant activity was evaluated by 1, 1-diphenyl-2-picryl-hydrazil (DPPH) free radical scavenging assay, ferric reducing antioxidant power (FRAP) and β-carotene bleaching inhibition assay. Total phenolic and flavonoid content were also determined. Anti-inflammatory activity was evaluated by in vitro human RBC membrane stabilizing assay and in vivo mice paw edema test. Ethanolic bark and leaf extracts of H. fomes (S1 and S2, respectively) showed significant scavenging effect on DPPH scavenging 68.51 µg/ml for S1 and 108.64 µg/ml for S2. Potent reduction potential (FRAP) was observed in S1 (83.75 mM of ferrous equivalent) while the FRAP of standard ascorbic acid was found 86.66 μM of ferrous equivalent. S1 also exhibited 87.43% β-carotene bleaching inhibition as comparable to standard butylated hydroxytoluene with 98.85% inhibition. Substantial phenolic and flavonoid content was also observed in S1 (136.83 μg gallic acid equivalent/g of extract and 76.67 μg quercetin equivalent/g of extract, respectively). S1 could provide 61.73% and 53.27% protection of RBC membrane in hypotonicity and heat induced lysis of RBC, respectively while the standard diclofenac sodium provided 91.66% and 86.66% protection, respectively. Furthermore, 44.16% reduction in mice paw edema at 40 μg/kg bw concentration was observed in S1 as compared to standard indomethacine (53.54% in 10 μg/kg bw concentration). The results revealed that the bark extract (S1) of H. fomes possesses significant antioxidant and anti-inflammatory activity.

Free radical production from different biological and environmental sources is due to an imbalance of natural antioxidants, which further leads to inflammation. Antioxidant metabolites are often characterized by anti-inflammatory and anticoagulation activity. Mangrove plants synthesize different classes of metabolites, including antioxidants, to minimize the devastating effect of oxidation resulting from the elevated salinity, UV, and other unique geochemical components. Accordingly, this study aimed at investigating the antioxidant, anti-inflammatory, and anticoagulation properties, as well as polyphenol content of the two selected mangrove plant species: Aegiceras corniculatum and Acanthus ilicifolius. We used the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, reducing power, ferric reducing antioxidant power (FRAP) assay, β-carotene-linoleic acid bleaching assay (BCB), total phenolic content (TPC), total flavonoid content (TFC) and total tannin content (TTC) to determine antioxidant activity of the ethanol extract of A. corniculatum bark and leaves and A. ilicifolius leaves. Furthermore, human red blood cell (HRBC) membrane stabilization assay, lipoxygenase (LOX) inhibition assay, and prothrombin time (PT) test were performed for determining anti-inflammatory activity of the samples. Not only has it cost hundreds of thousands of lives but also severely impacted livelihoods and placed enormous strain on community healthcare and welfare services. In this review, we explore the events associated with SARS-CoV-2 pathogenesis and host immunopathological reactivity due to the clinical manifestations of this coronavirus infection. We discuss that the metallopeptidase enzyme ADAM17, also known as tumor necrosis factor-α-converting enzyme, TACE, is responsible for shedding of angiotensin-converting enzyme 2 and membrane-bound interleukin (IL)-6 receptor. This leads to elevated pro-inflammatory responses that result in cytokine storm syndrome. We argue that cytokine balance may be restored by recovering an IL-6 trans-signaling neutralizing buffer system through the mediation of recombinant soluble glycoprotein 130 and recombinant ADAM17/TACE prodomain inhibitor. This cytokine restoration, possibly combined with inhibition of SARS-CoV-2 entry as well as replication and coagulopathy, could be introduced as a novel approach to treat patients with severe COVID-19. In cases of co-morbidity, therapies related to the management of associated disease conditions could ameliorate those clinical manifestations.

A mangrove medicinal plant Cynometra ramiflora (Family: Leguminosae) was selected to investigate the bioactivities namely antioxidant, antimicrobial and preliminary cytotoxic activity using methanol and chloroform extracts of the leaves and stems, respectively. In 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, 50% inhibitory concentration (IC50) of the methanolic stem extract was found to be 31.62 µg.mL-1. Reducing power of the same extract demonstrated consistent increase in a concentration-dependent manner and was comparable with quercetin while ferric reducing antioxidant power (FRAP) assay revealed potential total antioxidant capacity (84.0 mM Fe (II)/g of extract). In addition, the presence of total phenolics (96.2 mg GAE/g of extract), total flavonoids (166.4 mg QE/g of extract) and tannins content (80.4 mg GAE/g of extract) were determined in the methanolic stem extract. The chloroformic stem extract exhibited moderate antimicrobial activity against a number of bacterial strains while the MIC values of extracts were in the range from 62.5 to 500µg.mL-1. The methanolic stem and leaf extracts demonstrated strong lethality in preliminary cytotoxicity assay using brine shrimp nauplii where the 50% lethal concentration (LC50) values were 1.596 and 4.613 µg.mL-1 respectively. It can be therefore concluded that the methanolic extracts of C. ramiflora possess potential antioxidant, antimicrobial and strong preliminary cytotoxic activity and could be further exploited for prospective scientific exploration towards bioactive principles.

Objective: To determine antioxidant, antibacterial and brine shrimp lethality bioassay of Amoora cucullata using bark and leaf extracts of petroleum ether, chloroform and methanol. Methods: Bark and leaf parts were separately soaked into petroleum ether, chloroform and methanol respectively. To determine the presence of antioxidants; DPPH free radical scavenging assay, total phenolics and flavonoids determination and reducing power assay were carried out. Disk diffusion and brine shrimp lethality bioassay were conducted for the preliminary screening of antibacterial activity and cytotoxicity. Results: IC50 values in DPPH free radical scavenging assay for the extracts of petroleum ether, chloroform and methanol were found to be 316.23, 1192.42 and 128.82 in bark and 1106.32, 1330 and 25.12 μg/mL in leaf extracts respectively. Total phenolic content determined by Folin-Ciocalteu method in bark were 0, 0 and 58.41 mg gallic acid/gm of dry plant material in petroleum ether, chloroform and methanol while 8.11, 0 and 0 mg in leaf respectively. Total flavonoid content in bark and leaf was found to be 663.60, 549.47 and 46.25 mg and 1.64, 0 and 14.09 mg QE/gm of dried plant material in petroleum ether, chloroform and methanol respectively. In reducing power assay,methanolic extracts exhibited good reducing capacity. In antibacterial activity, extracts showed significant inhibition against enteropathogenic bacteria. Besides in brine shrimp lethality bioassay LC50 value was found to be 10, 2.301 and 7.28 in bark and 1.308, 1.94 and 2.14 μg/mL in leaf extracts of petroleum ether, chloroform and methanol respectively. Conclusion: From the present research work it can be concluded that A. cucullata leaf possess moderate antioxidant properties; strong antibacterial activity that correlates with their folkloric uses and potential cytotoxic properties. Meyim spinosa Roxb., a medicinai plant enjoys it itse in the traditional medicine in Bangladesh for the treatment of a number of ailnıents. The present stıtdy was undertaken to investigate the antibaeterial and cvtotoxic activitv of the ethanol extract of Mevna spinosa stem. Antibaeterial activity was investigated against Staphylococcus aureus, Streptococcus pvogenes, Escherichia coli and Shigella dysenteriae by disc diffıtsion and broth maerodilution assay. İn disk diffusion assav, the extract inhibited ali the microorganisms except E. coli. Minimum inhibitorv concentration (MIC§ of the extract w as 1000 ptg/ml for S. aureus, S. pvogenes and E. coli, whereas 500 jug/mL for S. dysenteriae. For cytotoxicity test, the extract w as subjected to brine shrimp lethality bioassav. The LD50 of M. spinosa stem extract was found to be 40 pg/mL. Findings of the stıtdy jııstify the itse of the plant in traditional medicine and suggests for further investigation

The successful construction of an immunosensor depends on having an effective procedure for immobilising the bio-recognition element to the transducer surface. In the present study, an amino-terminated 4-aminothiophenol (ATP) self-assembled monolayer (SAM) was modified with heterobifunctional crosslinker sulfosuccinimidyl 4-[N-maleimidomethyl] cyclohexane-1-carboxylate to couple reduced anti-myoglobin half-antibody fragments. The disulphide groups present in the hinge region of IgG molecules were selectively cleaved by 2-mercaptoethylamine to produce reduced half-antibody fragments with free sulphydryl groups. The maleimide terminated 4-ATP SAM modified surface was coupled to these reduced antibody fragments to produce highly oriented immobilization of the half-antibody via its Fc domain and to allow free access to the Fv bindings sites. This represents an improvement by comparison with biotin/avidin mediated IgG attachment which is essentially randomly oriented. Functional immunosensors were able to detect myoglobin in both phosphate buffered saline and whole serum over the range of concentrations from 10(-13)M to 10(-6)M, and order of magnitude better than avidin/biotin linked immunosensors. In addition, atomic force microscopy (AFM) was carried out to elucidate the nanotopology of the immunosensor surface at different stages of fabrication; the images demonstrate that half antibodies bind as described and show structural changes on subsequent antigen binding.

Objective: To isolate and evaluate the antimicrobial activity of the active principle(s) from the ethyl acetate (EtOAc) extract of endophytic fungus Colletotrichum gloeosporioides (C. gloeosporioides) isolated from Sonneratia apetala. Methods: Water agar technique was used to isolate the fungus, and both microscopic and molecular techniques were used for identification of the strain. Potato dextrose broth was used to grow the fungus in large-scale. Reversed-phase preparative HPLC analysis was performed to isolate the major active compound, kojic acid. The EtOAc extract and kojic acid were screened for their antimicrobial activity against two Gram-positive and two Gram-negative bacteria as well as a fungal strain using the resazurin 96-well microtitre plate antimicrobial assay. Results: The fungus C. gloeosporioides was isolated from the leaves of Sonneratia apetala. Initial identification of the fugal isolate was carried out using spore characteristics observed under the microscope. Subsequently, the ITS1-5.8S-ITS2 sequencing was employed for species-level identification of the fungus C. gloeosporioides. Five litres of liquid culture of the fungus produced approximately 610 mg of a mixture of secondary metabolites. Kojic acid (1) was isolated as the main secondary metabolite present in the fungal extract, and the structure was confirmed by 1D, 2D NMR and mass spectrometry. The EtOAc extract and compound 1 exhibited considerable antimicrobial activity against all tested microorganisms. Whilst the minimum inhibitory concentration (MIC) values from the EtOAc extract ranged between 2.4× 10-4 mg/mL and 2.5 mg/mL, those of kojic acid (1) were between 0.125 mg/mL and 1 mg/mL. The EtOAc extract and kojic acid (1) were most active against Pseudomonas aeruginosa (MIC = 2.4×10-4 mg/mL) and Micrococcus luteus (MIC = 0.125 mg/mL), respectively. Conclusions: The results revealed that the endophytic fungus C. gloeosporioides could be a good source of commercially important kojic acid, which exhibited antimicrobial properties.

The methanol extract of Diospyros peregrina bark was studied for its effect on the central nervous system (CNS) using the pentobarbitone induced sleeping time test, the open field test and the hole cross test in Swiss albino mice. The present investigation revealed that the extract, at the doses of 250 and 500 mg/kg, significantly prolonged the pentobarbitone induced sleeping time in mice though the onset of sleep was delayed as compared to the control. In open field test, the depressing effect was prominent from the second observation period (30 min) and persisted throughout the entire experimental period (240 min). In the hole cross test, the depressing effect was observed from the second observation period (30 min) and persisted up to fifth observation period (120 min) for 250 mg dose group and up to sixth observation period (180 min) for 500 mg dose group. These results support the finding that D. peregrina bark extract at the above doses has CNS depressing effects and indicate that D. peregrina bark may contain biologically active constituent(s) having CNS depressant activity.

Newcastle disease virus (NDV) is the infectious agent of Newcastle disease in poultry. This virus can grow within different animal cells including primary cell culture and established cell line. In order to adapt NDV on African green monkey kidney (Vero) cell line, five consecutive passages were done. Eagle's minimum essential medium (EMEM) with supplements was used for both culturing Vero cells and maintaining NDV on Vero cells. During first and second passage, wild NDV didn't produce any clear evidence of cytopathic effect (CPE), but in third passage changes in the characteristics of cell monolayer were observed. During fourth and fifth passages, clear and consistent CPE were observed within 50 to 60 hours of infection. CPE was characterized by formation of syncytium, giant cell, dendritic-shaped cell and finally plaque. The titer of passage 5 (P5) virus was 10 TCID , whose purity was tested by serum neutralization -3.9 50 test (SNT) and the result was 1.6 × 10 units/ml. 4

Induced pluripotent stem cells (iPSCs) are considered patient-specific counterparts of embryonic stem cells as they originate from somatic cells after forced expression of pluripotency reprogramming factors Oct4, Sox2, Klf4 and c-Myc. iPSCs offer unprecedented opportunity for personalized cell therapies in regenerative medicine. In recent years, iPSC technology has undergone substantial improvement to overcome slow and inefficient reprogramming protocols, and to ensure clinical-grade iPSCs and their functional derivatives. Recent developments in iPSC technology include better reprogramming methods employing novel delivery systems such as non-integrating viral and non-viral vectors, and characterization of alternative reprogramming factors. Concurrently, small chemical molecules (inhibitors of specific signalling or epigenetic regulators) have become crucial to iPSC reprogramming; they have the ability to replace putative reprogramming factors and boost reprogramming processes. Moreover, common dietary supplements, such as vitamin C and antioxidants, when introduced into reprogramming media, have been found to improve genomic and epigenomic profiles of iPSCs. In this article, we review the most recent advances in the iPSC field and potent application of iPSCs, in terms of cell therapy and tissue engineering.

Since the beneficial effects of viable probiotic bacteria as dietary supplements have gained huge research interest, Lactobacillus spp. with probiotic characteristics are widely used to prepare fermented dairy products such as yoghurts, milk-shakes etc. In this study, eight (08) homemade yoghurt samples were collected from different regions in the country for isolation of probiotic Lactobacillus spp. Among the samples, four (04) isolates were identified as Lactobacillus plantarum based on their growth and biochemical characteristics. The Isolates were resistant to NaCl (1-9%) and bilesalt (0.05-0.3%) and showed good growth in the acidic condition, while maximum growth was observed at pH around 6.0. The isolates were examined for their antibacterial activity against nine (09) different test pathogens and found all pathogens are inhibited their growth to some extent but maximum zone of inhibition was observed against Bacillus cereus (53.20 mm) and minimum was against Staphylococcus aureus (19 mm) after 72 hour incubation. The results of the present study indicate that, homemade yoghurts in Bangladesh are potential source of probiotic Lactobacillus spp. Further extensive research on isolation and characterization of probiotic organisms from local fermented foods and their growth optimization might be required for development of probiotic enriched food supplements in our country.

Heritiera fomes Buch.-Ham., a mangrove plant from the Sundarbans, has adapted to a unique habitat, muddy saline water, anaerobic soil, brackish tidal activities, and high microbial competition. Endophytic fungal association protects this plant from adverse environmental conditions. This plant is used in Bangladeshi folk medicine, but it has not been extensively studied phytochemically, and there is hardly any report on investigation on endophytic fungi growing on this plant. In this study, endophytic fungi were isolated from the surface sterilized cladodes and leaves of H. fomes. The antimicrobial activities were evaluated against two Gram-positive and two Gram-negative bacteria and the fungal strain, Candida albicans. Extracts of Pestalotia sp. showed activities against all test bacterial strains, except that the ethyl acetate extract was inactive against Escherichia coli. The structures of the purified compounds, oxysporone and xylitol, were elucidated by spectroscopic means. The anti-MRSA potential of the isolated compounds were determined against various MRSA strains, that is, ATCC 25923, SA-1199B, RN4220, XU212, EMRSA-15, and EMRSA-16, with minimum inhibitory concentration values ranging from 32 to 128 μg/ml. This paper, for the first time, reports on the anti-MRSA property of oxysporone and xylitol, isolation of the endophyte Pestalotia sp. from H. fomes, and isolation of xylitol from a Pestalotia sp.
