1. Developing Culturally Relevant Math and Science Items: Lessons Learned and Student Reactions. Research Report 2023-12

Abstract

For this project, ACT developed math and science items that attempted to represent unique aspects of cultures, raise awareness of social justice issues, promote cultural learning, be authentic and relatable, be comprehensible, and represent people in positive, nonstereotypical ways. Creating culturally relevant items was a new challenge for ACT test developers, and it involved many rounds of review by diverse content experts. The developers learned that creating culturally relevant items is a more demanding process, but they all reported that it was a positive, enriching experience. During focus groups, high school students viewed the culturally relevant items alongside non--culturally relevant items measuring the same skills. The students expressed interest in seeing culturally relevant items on the ACT, and they reported learning about cultures from the items. However, many students expressed concern about the additional time required to answer longer items during a timed high-stakes assessment like the ACT.

2. Examining the Relationship between Social and Emotional Skills and ACT Composite Scores in Students with Disabilities. ACT Research. Data Byte

Abstract

Numerous studies have demonstrated that social and emotional skills (SE skills) can predict academic achievement, including grades and test scores, and prior research at ACT has shown that stronger SE skills are associated with higher ACT scores. However, despite the abundant evidence that SE skills are beneficial for students in general, less is known about the relationship between SE skills and academic achievement among students with disabilities, who make up approximately 15% of all public school students (National Center for Education Statistics, 2023). In this study, the relationship between SE skills and ACT scores among students with and without disabilities was examined. Using data from the Mosaic™ by ACT® Social and Emotional Learning assessment, two matched groups of 410 students each were created, one whose members had disabilities and one whose members did not. Findings show a positive relationship between SE skills and ACT Composite scores in both students with disabilities and students without disabilities.

3. The 'Assessment of Bullying Experiences Questionnaire' ('ABE') for Neurodivergent Youth: Establishing Scoring Criteria and Clinical Thresholds

Abstract

Neurodivergent youth, such as those with diagnoses of autism spectrum disorder (ASD) or attention-deficit/hyperactivity disorder (ADHD), are frequently victimized by their peers. This bullying victimization may occur in unique ways that are not captured by existing questionnaires. The "Assessment of Bullying Experiences" ("ABE") is a questionnaire specific to neurodivergent youth with acceptable psychometric properties in autistic and non-autistic samples. This study sought to establish scoring criteria and clinical cutoffs for the "ABE." Participants were 335 parents of youth (ages 6-17) characterized as autistic, having ADHD, or from a community comparison sample. Receiver operating characteristic (ROC) analyses were used to (1) compare accuracy of the "ABE" total score in predicting bullying victimization experiences and (2) identify optimal questionnaire cutoffs. Clinical utility of these cutoffs was confirmed using ANCOVA analyses. The "ABE" had good-to-excellent discrimination for bullying victimization across groups. Bullied youth had greater symptoms of anxiety/depression compared to youth who were not bullied; this relationship was partially explained by autism characteristics. "ABE" clinical cutoffs generated bullying victimization prevalence rates consistent with existing literature (50% autism, 28% non-autism). The "ABE" is a useful measure to streamline bullying victimization assessment in neurodivergent youth and inform group-specific intervention.

4. Trajectory Research in Children with an Autism Diagnosis: A Scoping Review

Abstract

Researchers increasingly employ longitudinal trajectory methods to understand developmental pathways of people on the autism spectrum across the lifespan. By assessing developmental or health-related outcome domains at three or more timepoints, trajectory studies can characterize their shape and varying rates of change over time. The purpose of this scoping review was to identify and summarize the published breadth of research that uses a trajectory study design to examine development in children (to age 18 years) diagnosed with autism. Using a systematic search and screening procedure, 103 studies were included. This review summarizes methodological characteristics across studies including the varying statistical approaches used. A series of figures maps where published research is available across 10 outcome domains and the ages over which children have been followed. Evidence gaps, informed by the perspectives of the autistic and caregiver stakeholders that were engaged in this review, are discussed. We recommend that future trajectory research addresses the absence of studies from low- and middle-income countries, considers longitudinal assessment of outcome domains that caregivers and autistic people consider meaningful, and plans follow-up periods with assessment timepoints that cover the gaps in ages where more outcome-specific data are needed.

6. Health challenges of rangers—a planetary health workforce

Abstract

Rangers safeguard the balance between humans and nature by protecting and managing biodiversity and natural resources. The challenging working conditions that rangers face make them vulnerable to wildlife attacks and exposure to zoonotic and vector-borne diseases. Despite all of these work-related challenges and threats to their health, a vast majority of rangers lack access to adequate medical treatment facilities. This research has used data from the one of the largest and most comprehensive surveys of rangers across multiple countries, collected as part of the Global Ranger Perception Survey, to examine the relationship between the precarious working conditions of rangers and their health outcomes. By comparing data from the 2020 World Malaria Report, our study highlights the severe malaria burden carried by rangers around the world. Malaria prevalence in rangers working in Central Africa, East Africa, and South America was estimated to be four times higher than in the general population. The results of this study are valuable because they show that rangers are a vulnerable, high-risk population of professionals and their working conditions are highly associated with their respective health outcomes. The research also makes it imperative that improving working conditions is essential for maintaining a professional ranger workforce that protects the planet’s natural areas and biodiversity.

8. Examining the Efficacy of Career & College Clubs: Documenting Increases in Skills and Postsecondary Education Knowledge. ACT Research Report. 2022-09

Abstract

This study was done in collaboration with the National Council for Community and Education Partnerships (NCCEP) and Alliance College-Ready Public Schools (Alliance). This report, the second in the series, Examining the Efficacy of Career & College Clubs, explores how changes in the college readiness areas of social and emotional skills, knowledge of postsecondary education, and education intentions were related to growth in cumulative student grade point average (GPA) in 11th-grade students. The students in this study come from a cohort of over 900 Alliance students enrolled in Gaining Early Awareness and Readiness for Undergraduate Programs (GEAR UP) who have taken the Career & College Clubs (CCC) curriculum beginning in 9th grade. The results of this study demonstrate that the college readiness skills learned in the CCC curriculum increased significantly in students during their first three years of high school and predicted student grade point average (GPA). 9. Educators' Social and Emotional Skills and Indicators of School Climate Predict Burnout. ACT Research. Data Byte

Abstract

In this report, the authors examined the relationship between social and emotional (SE) skills, school climate, educator variables, and educator burnout. Mosaic Educator, an assessment designed for teachers and school staff, was completed by a sample of educators in several regions of the country during the spring of 2022. This assessment measured educators' SE skills, perceptions of school climate, and experiences of burnout symptoms. Additionally, demographic and work variables were collected, including gender, race/ethnicity, hours worked, and job tenure. The final sample in this analysis included 329 educators (82.2% female, 17.8% male; 62.5% White, 19.2% Black/African American, 16.3% Hispanic/Latino, 2.0% Asian) who had an average tenure of 15.6 years. Prior research has demonstrated that teachers who receive professional development related to SE skills tend to have lower levels of burnout. Based on this study, cultivating staff relationships, providing a safe working environment, and offering professional development related to SE skills like stress management could all provide support toward reaching this goal.

Abstract

Flare frequency distributions represent a key approach to addressing one of the largest problems in solar and stellar physics: determining the mechanism that counter-intuitively heats coronae to temperatures that are orders of magnitude hotter than the corresponding photospheres. It is widely accepted that the magnetic field is responsible for the heating, but there are two competing mechanisms that could explain it: nanoflares or Alfv\'en waves. To date, neither can be directly observed. Nanoflares are, by definition, extremely small, but their aggregate energy release could represent a substantial heating mechanism, presuming they are sufficiently abundant. One way to test this presumption is via the flare frequency distribution, which describes how often flares of various energies occur. If the slope of the power law fitting the flare frequency distribution is above a critical threshold, $\alpha=2$ as established in prior literature, then there should be a sufficient abundance of nanoflares to explain coronal heating. We performed $>$600 case studies of solar flares, made possible by an unprecedented number of data analysts via three semesters of an undergraduate physics laboratory course. This allowed us to include two crucial, but nontrivial, analysis methods: pre-flare baseline subtraction and computation of the flare energy, which requires determining flare start and stop times. We aggregated the results of these analyses into a statistical study to determine that $\alpha = 1.63 \pm 0.03$. Social and emotional (SE) skills are related to many positive outcomes, including degree attainment, improved academic performance, and increased life satisfaction, and are often taught through social and emotional learning (SEL) programs. A positive school climate is important for students' social and emotional wellness and ability to learn. For example, students who have positive relationships with their teachers and feel safe at school tend to show higher academic performance. The purpose of this study was to examine how SE skills and students' perceptions of school climate relate to college readiness in English, as measured by the ACT® English test and ACT's College Readiness Benchmark for English. Mosaic™ by ACT® Social Emotional Learning assessment data were collected from 6,747 students (53% female and 47% male; 67% White, 12% Hispanic, 6% Black, and 3% Asian; 74% 11th graders and 23% 12th graders at the time they took the ACT test) and matched to their ACT scores. Their SEL assessment data were captured during the school years 2018-2019 and 2019-2020, while the ACT tests occurred from 2018 to 2021. The SEL assessment measures five social and emotional skills and two school climate scales. Social and Emotional Skills: (1) Sustaining Effort; (2) Getting Along with Others; (3) Maintaining Composure; (4) Keeping an Open Mind; and (5) Social Connection. School Climate Scales: (1) Relationships with School Personnel; and (2) School Safety. The data show that a larger percentage of students in the high category of each SE skill or school climate scale met or exceeded the ACT English Benchmark.

Abstract Vascular smooth muscle cell (VSMC) proliferation, migration, and apoptosis play important roles in many physiological processes and pathological conditions. To identify genetic influences on VSMC behavior, we measured these traits and undertook genome-wide association studies in primary umbilical artery-derived VSMCs from >2000 individuals. Although there were no genome-wide significant associations for VSMC proliferation or migration, genetic variants at two genomic loci (7p15.3 and 7q32.3) showed highly significant associations with VSMC apoptosis (P = 1.95 × 10−13 and P = 7.47 × 10−9, respectively). The lead variant at the 7p51.3 locus was associated with increased expression of the GSDME and PALS2 genes in VSMCs. Knockdown of GSDME or PALS2 in VSMCs attenuated apoptotic cell death. A protein co-immunoprecipitation assay indicated that GSDME complexed with PALS2. PALS2 knockdown attenuated activated caspase-3 and GSDME fragmentation, whilst GSDME knockdown also reduced activated caspase-3. These findings provide new insights into the genetic regulation of VSMC apoptosis, with potential utility for therapeutic development.

Background: Nonunion after ankle or hindfoot arthrodesis is associated with poor outcomes. Cellular bone allograft is an alternative to autograft for use in these procedures. The purpose of this study was to prospectively evaluate the early efficacy and safety of cellular bone allograft use in hindfoot and ankle arthrodesis procedures. Methods: Fourteen patients undergoing hindfoot or ankle arthrodesis supplemented with cellular bone allograft were prospectively enrolled. Computed tomography (CT) scans were obtained postoperatively at set time points and reviewed by 3 fellowship-trained foot and ankle surgeons as well as 1 musculoskeletal radiologist. Primary outcome was CT-verified union, defined as >25% of joint surface. Complications were recorded and revision procedures offered as indicated. Results: CT-verified union rate during the study period was 76.7% (23 of 30 joints). Union was 100% for the ankle joint (2 of 2), 50% for the talonavicular joint (5 of 10), 100% for the calcaneocuboid joint (8 of 8), and 80.0% for the subtalar joint (8 of 10). One patient underwent revision fusion procedure, and 1 patient underwent hardware removal during the study period. Conclusion: Our initial experience suggests that use of cellular bone allograft augmentation in hindfoot and ankle arthrodesis may offer an alternative to autograft without potential of donor site morbidity. ACT, the National Council for Community and Education Partnerships (NCCEP), and Alliance College-Ready Public Schools (Alliance) collaborated to examine the efficacy of NCCEP's Career & College Clubs (CCC) curriculum for increasing college readiness skills in high school students participating in Gaining Early Awareness and Readiness for Undergraduate Programs (GEAR UP). This report explores the development of college readiness skills in over 900 Alliance students enrolled in GEAR UP who were taking the CCC curriculum during their first three years of high school, beginning in the 2018-2019 school year. To measure the impact of the CCC curriculum, a series of surveys to explore the developmental trajectory of students' social and emotional skills, knowledge of postsecondary education, and education intentions.

Abstract Osteoarthritis (OA) is increasing in prevalence and has a severe impact on patients’ lives. However, our understanding of biomarkers driving OA risk remains limited. We developed a model predicting the five-year risk of OA diagnosis, integrating retrospective clinical, lifestyle and biomarker data from the UK Biobank (19,120 patients with OA, ROC-AUC: 0.72, 95%CI (0.71–0.73)). Higher age, BMI and prescription of non-steroidal anti-inflammatory drugs contributed most to increased OA risk prediction ahead of diagnosis. We identified 14 subgroups of OA risk profiles. These subgroups were validated in an independent set of patients evaluating the 11-year OA risk, with 88% of patients being uniquely assigned to one of the 14 subgroups. Individual OA risk profiles were characterised by personalised biomarkers. Omics integration demonstrated the predictive importance of key OA genes and pathways (e.g., GDF5 and TGF-β signalling) and OA-specific biomarkers (e.g., CRTAC1 and COL9A1). In summary, this work identifies opportunities for personalised OA prevention and insights into its underlying pathogenesis.

Abstract

Abstract African Swine Fever (ASF) is a highly lethal viral disease in swine, with mortality rates approaching 100%. The disease has spread to many swine-producing countries, leading to significant economic losses and adversely impacting global food security. Extensive efforts have been directed toward developing effective ASF vaccines. Among the vaccinology approaches tested to date, live-attenuated virus (LAV) vaccines produced by rational deleting virulence genes from virulent African Swine Fever Virus (ASFV) strains have demonstrated promising safety and efficacy in experimental and field conditions. Many gene-deleted LAV vaccine candidates have been generated in recent years. The virulence genes targeted for deletion from the genome of virulent ASFV strains can be categorized into four groups: Genes implicated in viral genome replication and transcription, genes from the multigene family located at both 5′ and 3′ termini, genes participating in mediating hemadsorption and putative cellular attachment factors, and novel genes with no known functions. Some promising LAV vaccine candidates are generated by deleting a single viral virulence gene, whereas others are generated by simultaneously deleting multiple genes. This article summarizes the recent progress in developing and characterizing gene-deleted LAV vaccine candidates.

Abstract

We present initial results of an ongoing engineering study on the feasibility of a space mission to the focal region of the solar gravitational lens (SGL). The mission goal is to conduct exoplanet imaging operations at heliocentric distances in the range ~548-900 astronomical units (AU). Starting at 548 AU from the Sun, light from an exoplanet located behind the Sun is greatly amplified by the SGL. The objective is to capture this light and use it for multipixel imaging of an exoplanet up to 100 light years distant. Using a meter-class telescope one can produce images of the exoplanet with a surface resolution measured in tens of kilometers and to identify signs of habitability. The data are acquired pixel-by-pixel while moving an imaging spacecraft within the image. Given the long duration of the mission, decades to 900 AU, we address an architecture for the fastest possible transit time while reducing mission risk and overall cost. The mission architecture implements solar sailing technologies and in-space aggregation of modularized functional units to form mission capable spacecraft. Abstract

Research shows elevated gender variance among autistic people and more autistic traits among gender diverse people, each of which is related to mental health concerns. Little work has explored broad features of these presentations in a non-clinical sample. College students (n = 174) ages 18-22 years completed questionnaires assessing the broader autism phenotype (BAP), autistic features, nonconformity to gender norms, and internalizing symptoms. Those with more BAP features or autistic communication reported more nonconformity to gender norms. Higher levels of internalizing symptoms were related to more gender nonconformity, BAP, and autistic features. Gender nonconformity marginally moderated the effect of BAP on depression but not anxiety. The BAP, autistic features, and gender nonconformity are important in understanding mental well-being.

Abstract

When replication forks encounter damaged DNA, cells utilize damage tolerance mechanisms to allow replication to proceed. These include translesion synthesis at the fork, postreplication gap filling, and template switching via fork reversal or homologous recombination. The extent to which these different damage tolerance mechanisms are utilized depends on cell, tissue, and developmental context-specific cues, the last two of which are poorly understood. To address this gap, we have investigated damage tolerance responses in Drosophila melanogaster. We report that tolerance of DNA alkylation damage in rapidly dividing larval tissues depends heavily on translesion synthesis. Furthermore, we show that the REV1 protein plays a multi-faceted role in damage tolerance in Drosophila. Larvae lacking REV1 are hypersensitive to methyl methanesulfonate (MMS) and have highly elevated levels of γ-H2Av (Drosophila γ-H2AX) foci and chromosome aberrations in MMS-treated tissues. Loss of the REV1 C-terminal domain (CTD), which recruits multiple translesion polymerases to damage sites, sensitizes flies to MMS. In the absence of the REV1 CTD, DNA polymerases eta and zeta become critical for MMS tolerance. In addition, flies lacking REV3, the catalytic subunit of polymerase zeta, require the deoxycytidyl transferase activity of REV1 to tolerate MMS. Together, our results demonstrate that Drosophila prioritize the use of multiple translesion polymerases to tolerate alkylation damage and highlight the critical role of REV1 in the coordination of this response to prevent genome instability.

Abstract

BackgroundThe intimal hyperplasia (IH) and vascular remodelling that follows endovascular injury, for instance after post-angioplasty re-stenosis, results in downstream ischaemia and progressive end organ damage. Interferon gamma (IFNγ) is known to play a critical role in this process. In mouse models we have previously shown that fibrocytes expressing tissue factor (TF) are recruited early to the site of injury. Through thrombin generation and protease activated receptor-1 (PAR-1) activation, fibrocytes secrete angiopoietin-2, stimulate neointimal cell proliferation, inhibit apoptosis and induce CXCL-12 production, all of which contribute to the progressive IH that then develops. In this study we investigated the relationship between TF, angiopoietin-2 and IFNγ.Methods and resultsIH developing in carotid arteries of wild-type mice 4 weeks after endoluminal injury contained a significant proportion of IFNγ+ fibrocytes and macrophages, which we show, using a previously defined adoptive transfer model, were derived from circulating CD34+ cells. IH did not develop after injury in IFNγ-deficient mice, except after transplantation of WT bone marrow or adoptive transfer of WT CD34+ cells. In vitro, CD34+ cells isolated from post-injury mice did not express IFNγ, but this was induced when provided with FVIIa and FX, and enhanced when prothrombin was also provided: In both cases IFNγ secretion was TF-dependent and mediated mainly through protease activated PAR-1. IFNγ was predominantly expressed by fibrocytes. In vivo, all IFNγ+ neointimal cells in WT mice co-expressed angiopoietin-2, as did the small numbers of neointimal cells recruited in IFNγ-/- mice. Adoptively transferred WT CD34+ cells treated with either an anti-TIE-2 antibody, or with siRNA against angiopoetin-2 inhibited the expression of IFNγ and the development of IH.ConclusionTF-dependent angiopoietin-2 production by newly recruited fibrocytes, and to a lesser extent macrophages, switches on IFNγ expression, and this is necessary for the IH to develop. These novel findings enhance our understanding of the pathophysiology of IH and expose potential targets for therapeutic intervention.

Abstract

Social and emotional skills, such as Sustaining Effort and Maintaining Composure, are related to many positive outcomes, including academic performance and life satisfaction, and are often taught through social and emotional learning (SEL) programs. The purpose of this study was to examine the relationship between these skills and ACT® test scores. Mosaic™ by ACT® Social Emotional Learning assessment data was collected from 6,747 students (53% female, 47% male; 67% White, 12% Hispanic, 6% Black, 3% Asian; 74% 11th graders, 23% 12th graders at the time of the ACT test) and matched to their ACT scores. Their Mosaic by ACT SEL assessment data were captured during the school years 2018-2019 and 2019-2020, while the ACT tests occurred from 2018 to 2021. The assessment measures five social and emotional skills: (1) Sustaining Effort; (2) Getting Along with Others; (3) Maintaining Composure; (4) Keeping an Open Mind; and (5) Social Connection. The data show that there is a clear positive relationship between social and emotional skills and ACT Composite scores.

A positive school climate is important for students' social and emotional wellness and ability to learn. For example, students with positive relationships with their teachers and who feel safe at school tend to show higher academic performance (Korpershoek et al., 2020). The purpose of the current study was to examine the relationship of school climate to ACT® test scores. Mosaic™ by ACT® Social Emotional Learning (SEL) assessment data was collected from 6,747 students (53% female, 47% male; 67% White, 12% Hispanic, 6% Black, 3% Asian; 74% 11th graders, 23% 12th graders at the time of the ACT test) and matched to their ACT scores. Their SEL assessment data were captured during the 2018-2019 and 2019-2020 school years, while the ACT tests occurred from 2018 to 2021. The assessment includes two measures of school climate, which are defined as follows: (1) Relationships with School Personnel: the extent to which students relate to school personnel as part of their connection to school; and (2) School Safety: students' perception of the school qualities related to security at school. The data show that there is a positive relationship between perceived school climate and ACT scores.

Abstract

Abstract Background Eating disorders have one of the highest mortality rates among psychiatric illnesses. Timely intervention is crucial for effective treatment, as eating disorders tend to be chronic and difficult to manage if left untreated. Clinical practice guidelines play a vital role in improving healthcare delivery, aiming to minimize variations in care and bridge the gap between research and practice. However, research indicates an active guideline implementation approach is crucial to effective uptake. Methods Mixed methods will be used to inform and evaluate our guideline implementation approach. Semi-structured focus groups will be conducted in each of the eight provinces in Canada. Each focus group will comprise 8–10 key stakeholders, including clinicians, program administrators, and individuals with lived experience or caregivers. Qualitative data will be analyzed using conventional content analysis and the constant comparison technique and the results will be used to inform our implementation strategy. The study will then evaluate the effectiveness of our implementation approach through pre- and post-surveys, comparing changes in awareness, use, and impact of the guidelines in various stakeholder groups. Discussion Through a multifaceted implementation strategy, involving the co-creation of educational materials, tailored training, and context-specific strategies, this study intends to enhance guideline uptake and promote adherence to evidence-based practices. Our study will also contribute valuable information on the impact of our implementation strategies.

Abstract Vagus nerve signaling is a key component of the gut-brain axis and regulates diverse physiological processes that decline with age. Gut to brain vagus firing patterns are regulated by myenteric intrinsic primary afferent neuron (IPAN) to vagus neurotransmission. It remains unclear how IPANs or the afferent vagus age functionally. Here we identified a distinct ageing code in gut to brain neurotransmission defined by consistent differences in firing rates, burst durations, interburst and intraburst firing intervals of IPANs and the vagus, when comparing young and aged neurons. The aminosterol squalamine changed aged neurons firing patterns to a young phenotype. In contrast to young neurons, sertraline failed to increase firing rates in the aged vagus whereas squalamine was effective. These results may have implications for improved treatments involving pharmacological and electrical stimulation of the vagus for age-related mood and other disorders. For example, oral squalamine might be substituted for or added to sertraline for the aged.

Abstract

Long non-coding RNAs (lncRNAs) are emerging as important players in gene regulation and cardiovascular diseases. However, the roles of lncRNAs in atherosclerosis are poorly understood. In the present study, we found that the levels of NIPA1-SO were decreased while those of NIPA1 were increased in human atherosclerotic plaques. Furthermore, NIPA1-SO negatively regulated NIPA1 expression in human umbilical vein endothelial cells (HUVECs). Mechanistically, NIPA1-SO interacted with the transcription factor FUBP1 and the NIPA1 gene. The effect of NIPA1-SO on NIPA1 protein levels was reversed by the knockdown of FUBP1. NIPA1-SO overexpression increased, whilst NIPA1-SO knockdown decreased BMPR2 levels; these effects were enhanced by the knockdown of NIPA1. The overexpression of NIPA1-SO reduced while NIPA1-SO knockdown increased monocyte adhesion to HUVECs; these effects were diminished by the knockdown of BMPR2. The lentivirus-mediated-overexpression of NIPA1-SO or gene-targeted knockout of NIPA1 in low-density lipoprotein receptor-deficient mice reduced monocyte-endothelium adhesion and atherosclerotic lesion formation. Collectively, these findings revealed a novel anti-atherosclerotic role for the lncRNA NIPA1-SO and highlighted its inhibitory effects on vascular inflammation and intracellular cholesterol accumulation by binding to FUBP1 and consequently repressing NIPA1 expression.
