12. Questions of the right and the good: metaethics and anthropology

Abstract

Questions of the right and the good have been variously recognised by anthropologists as key to understanding human action and behaviour since the early days of the discipline, with the influential recent ‘ethical turn’ perhaps the most obvious example (Mattingly and Throop, 2018). Here, the authors – who together founded the Social Science and Ethics Group at the University of Sussex – reflect on how their own fieldwork led them to engage with metaethics, and how doing so advanced their understanding of their ethnographic material, anthropological theory, and their own professional ethics. The three cases draw on fieldwork in Nicaragua, Bolivia, and China.

Abstract

Acute Myeloid Leukemia (AML) is a fatal hematological cancer. The genetic abnormalities underlying AML are extremely heterogeneous among patients, making prognosis and treatment selection very difficult. While clinical proteomics data has the potential to improve prognosis accuracy, thus far, the quantitative means to do so have yet to be developed. Here we report the results and insights gained from the DREAM 9 Acute Myeloid Prediction Outcome Prediction Challenge (AML-OPC), a crowdsourcing effort designed to promote the development of quantitative methods for AML prognosis prediction. We identify the most accurate and robust models in predicting patient response to therapy, remission duration, and overall survival. We further investigate patient response to therapy, a clinically actionable prediction, and find that patients that are classified as resistant to therapy are harder to predict than responsive patients across the 31 models submitted to the challenge. The top two performing models, which held a high sensitivity to these patients, substantially utilized the proteomics data to make predictions. Using these models, we also identify which signaling proteins were useful in predicting patient therapeutic response., Author Summary Acute Myeloid Leukemia (AML) is a hematological cancer with a very low 5-year survival rate. It is a very heterogeneous disease, meaning that the molecular underpinnings that cause AML vary greatly among patients, necessitating the use of precision medicine for treatment. While this personalized approach could be greatly improved by the incorporation of high-throughput proteomics data into AML patient prognosis, the quantitative methods to do so are lacking. We held the DREAM 9 AML Outcome Prediction Challenge to foster support, collaboration, and participation from multiple scientific communities in order to solve this problem. The outcome of the challenge yielded several accurate methods (AUROC >0.78, BAC > 0.69) capable of predicting whether a patient would respond to therapy. Moreover, this study also determined aspects of the methods which enabled accurate predictions, as well as key signaling proteins that were informative to the most accurate models.

35. Stable lines of transgenic zebrafish exhibit reproducible patterns of transgene expression

Abstract

To study the frequency of germ-line transformation and to examine the reproducibility of tissue-specific trans gene expression, we produced several lines of transgenic zebrafish expressing a recombinant chloramphenicol acetyltransferase (CAT) gene. Supercoiled plasmids containing both Rous sarcoma virus and SV-40 pro moter sequences upstream of the CAT coding region were injected into zebrafish embryos prior to first cleavage. CAT activity could be detected in batches of injected embryos as early as 8 h and up to at least 12 days post-fertilization. Approximately 18 % of injected fish raised to maturity exhibited CAT activity in their fins, and approximately 5 % of injected fish became stable germ-line transformants. Breeding studies indicated that although transgenic founder fish were frequently germ-line mosaics, transgenic individuals of subsequent generations were fully hemizygous for the transgene marker. The transgenes present in the F] progeny of four independent lines were relatively well expressed in fin and skin, while lower levels of expression were observed in heart, gill and muscle. Little or no CAT expression was observed in the brain, liver and gonad. A monoclonal antibody directed against the CAT gene product consistently revealed variegated patterns of CAT expression in ectodermally derived fin epidermal cells in three of these lines. These results show that it is possible to efficiently produce stable germ-line trans formants of the zebrafish and to observe reproducible tissue-specific patterns of transgene expression in this organism. Possible mechanisms for the variegated ex pression observed within tissues are also considered.

36. Replication, integration and stable germ-line transmission of foreign sequences injected into early zebrafish embryos

Abstract

To generate stable lines of transgenic fish, early zebrafish embryos were injected with high concentrations of a linear bacterial plasmid. After injection, the foreign DNA was converted into a high molecular weight form and then amplified approximately tenfold during the initial rapid cleavages characteristic of the early embryo prior to gastrulation. While most of this DNA was subsequently degraded during gastrulation, some of the foreign sequences survived the gastrula stage and could be found in most of the injected fish at 3 weeks of age. Only about 5 % of fish analysed 4 months after the injection retained foreign DNA in their fins, usually at less than one copy per cell. One of these fish was also found to contain about 100 copies per cell of foreign DNA in a fraction of its germ cells. Approximately 20% of the F, offspring from this germ-line-positive parent inherited the foreign DNA, whereas 50 % of F2 progeny obtained from an identified F1 individual inherited these sequences. The 50 % transmission rate in F2 progeny was as expected for a single, heterozygous genomic insert. These observations indicate that injected DNA can be integrated into the fish genome, that the resulting transgenic fish are mosaic and that some of these mosaic individuals give rise to stable lines of transgenic fish.

Abstract

The microfilm rapid selector -- a dream 30 years ago and a laboratory instrument as recently as five years ago -- is today the basis of an operating information retrieval system saving time and money for the Navy Department's Bureau of Ships. The present-day Rapid Selector is a device for searching, selecting, and copying documents stored on microfilm. Readily adapted to the use of film made in any commercially available 35 mm microfilm camera, the Selector searches and selects at the rate of 100 document images per second. Copies of selected images are made on microfilm without any reduction in the speed of the film being searched. (A predictive search, in which copies are not made, is used to evaluate the question.) Present coding capability is 240 bits, arranged in 60 hexidecimal fields. A single field or combination of fields can be used as the basis for a search. Binary notation and superimposed random-number coding are used extensively. Coded information includes usual bibliographic information, major subject-matter designations, and secondary subject-matter designations. Average time for indexing a document is 20 minutes. Average time for complete search, including processing, is 12 minutes. Average number of searches per month is 10. Studies of development rate and use rate of indexing terms are being conducted continually. Convinced that high-speed search and selection without high-speed delivery of the results of the search creates an anomaly, the Bureau of Ships is presently testing newly-developed fascimile equipment designed to deliver Rapid Selector output to remote destinations by microwave linkages.
