Abstract

Effective monitoring of manufacturing processes is crucial for maintaining product quality and operational efficiency. Modern manufacturing environments generate vast amounts of multimodal data, including visual imagery from various perspectives and resolutions, hyperspectral data, and machine health monitoring information such as actuator positions, accelerometer readings, and temperature measurements. However, interpreting this complex, high-dimensional data presents significant challenges, particularly when labeled datasets are unavailable. This paper presents a novel approach to multimodal sensor data fusion in manufacturing processes, inspired by the Contrastive Language-Image Pre-training (CLIP) model. We leverage contrastive learning techniques to correlate different data modalities without the need for labeled data, developing encoders for five distinct modalities: visual imagery, audio signals, laser position (x and y coordinates), and laser power measurements. By compressing these high-dimensional datasets into low-dimensional representational spaces, our approach facilitates downstream tasks such as process control, anomaly detection, and quality assurance. We evaluate the effectiveness of our approach through experiments, demonstrating its potential to enhance process monitoring capabilities in advanced manufacturing systems. This research contributes to smart manufacturing by providing a flexible, scalable framework for multimodal data fusion that can adapt to diverse manufacturing environments and sensor configurations.

2. Effects of Varying Land Coverage, Rotation Period, and Water Vapor on Equatorial Climates that Bridge the Gap between Earth-like and Titan-like

Abstract

Saturn's largest moon, Titan, has an Earth-like volatile cycle, but with methane playing the role of water and surface liquid reservoirs geographically isolated at high latitudes. We recreate Titan's characteristic dry hydroclimate at the equator of an Earth-like climate model without seasons and with water as the condensable by varying a small set of planetary parameters. We use three observationally motivated criteria for Titan-like conditions at the equator: 1) the peak in surface specific humidity is not at the equator, despite it having the warmest annual-mean temperatures; 2) the vertical profile of specific humidity in the equatorial column is nearly constant through the lower troposphere; and 3) the relative humidity near the surface at the equator is significantly lower than saturation (lower than 60%). We find that simply reducing the available water at the equator does not fully reproduce Titan-like conditions. We additionally vary the rotation period and volatility of water to mimic Titan's slower rotation and more abundant methane vapor. Longer rotation periods coupled with a dry equatorial surface meet fewer of the Titan-like criteria than equivalent experiments with shorter rotation periods. Experiments with higher volatility of water meet more criteria than those with lower volatility, with some of those with the highest volatility meeting all three, demonstrating that an Earth-like planet can display Titan-like climatology by changing only a few physical parameters., Comment: 40 pages, 18 figures

Abstract

INTRODUCTION The Colorado River system is facing a crisis. For more than two decades, the basin has faced the driest period on record due to drought and climate change. (1) [...], Native Americans are the first inhabitants of the Colorado River Basin and have relied on its water and other resources since time immemorial. However, tribes were not involved in the shaping the Colorado River Compact and its governing institutions, and they have faced uphill battles to secure, protect, and develop their water rights--including the ability to acquire access to clean water for their members. This article begins by explaining the historic role of tribes in governing the Colorado River system. It then reviews ongoing efforts to better integrate tribal needs, interests, and priorities into management decisions, and to support opportunities for more meaningful engagement of tribes in collaborative problem-solving and decision-making. Finally, the article provides a roadmap for the future role of tribes in governing the Colorado River system. The negotiation and implementation of the post-2026 management framework presents an important inflection point in the history of the basin to enhance the capacity of tribes and to reform the architecture of governance.

9. How far should adjacent pan traps be placed for bee sampling?

Abstract

Among several methods of active and passive sampling bees (Hymenoptera: Apoidea), pan traps, also known as bee bowls, are commonly utilized for their efficiency and as a means of avoiding collector bias. When comparing pan‐trap samples using traditional statistical methods, the assumption of independence among samples needs to be met. To determine the necessary distance between pan traps to obtain independent samples, we investigated spatial dependence in bee catches using pan traps at four sites. At each site, a regular grid of pan traps was laid out, with each sample having two colors of fluorescent trap (i.e. blue and yellow). These trap pairs were separated by 10 m, with 100–109 points in the grid. In addition, five points within the grid were chosen randomly, and an additional 40 traps were placed at 2 m and 4 m from the five sample points to sample bees at a minimum 2 m trap distance. Traps collected bees for five consecutive days and bees were counted for each point and each trap color. Bee count data were analyzed using geostatistics to determine the spatial dependency in trap catches for yellow traps, blue traps, and both colors combined. The highest value of the range parameter of the semivariogram found across the four sites and pan trap colors was 17 m for blue traps. From this, we recommend a conservative minimum distance of 17 m between adjacent pan traps to obtain spatially independent samples for optimizing sampling plans when independent samples are necessary. Abstract

Targeting tumor-specific molecular alterations has shown significant clinical benefit. Molecular tumor boards (MTBs) connect cancer patients with personalized treatments and clinical trials. However, rural cancer centers often have limited access to MTB expertise. We established an academic–community partnership expanding our academic MTB to affiliated rural community cancer centers. We developed a centralized molecular registry of tumors (MRT) to aggregate the comprehensive genomic profiling (CGP) results and facilitate multidisciplinary MTB review. Of the 151 patients included, 87 (58%) had actionable genomic biomarkers, 42 (28%) were eligible for a targeted off-label therapy, and 27 (18%) were matched to a clinical trial. Of those with a clinical trial match, only 1 of 27 (3%) was enrolled in the identified trial. One year into implementation, community oncology providers were anonymously surveyed on persistent barriers to precision treatment utilization. The primary barriers to clinical trial enrollment were the distance to the trial center (70%), lack of transportation (55%), and lack of local trials (50%). This study offers a framework to improve access to molecular expertise, but significant barriers to the equitable use of CGP and trial enrollment persist. [ABSTRACT FROM AUTHOR]

Abstract

"Liquid biopsy" approaches analyzing cell-free DNA (cfDNA) from the blood of patients with cancer are increasingly utilized in clinical practice. However, it is not yet known whether cfDNA sequencing from large cohorts of patients with cancer can detect genomic alterations at frequencies similar to those observed by direct tumor sequencing, and whether this approach can generate novel insights. Here, we report next-generation sequencing data from cfDNA of 1,397 patients with colorectal cancer. Overall, frequencies of genomic alterations detected in cfDNA were comparable to those observed in three independent tissue-based colorectal cancer sequencing compendia. Our analysis also identified a novel cluster of extracellular domain (ECD) mutations in EGFR, mediating resistance by blocking binding of anti-EGFR antibodies. Patients with EGFR ECD mutations displayed striking tumor heterogeneity, with 91% harboring multiple distinct resistance alterations (range, 1-13; median, 4). These results suggest that cfDNA profiling can effectively define the genomic landscape of cancer and yield important biological insights.Significance: This study provides one of the first examples of how large-scale genomic profiling of cfDNA from patients with colorectal cancer can detect genomic alterations at frequencies comparable to those observed by direct tumor sequencing. Sequencing of cfDNA also generated insights into tumor heterogeneity and therapeutic resistance and identified novel EGFR ectodomain mutations. Abstract

Enteropathy-associated T cell lymphoma (EATL) is a lethal, and the most common, neoplastic complication of celiac disease. Here, we defined the genetic landscape of EATL through whole-exome sequencing of 69 EATL tumors. SETD2 was the most frequently silenced gene in EATL (32% of cases). The JAK-STAT pathway was the most frequently mutated pathway, with frequent mutations in STAT5B as well as JAK1, JAK3, STAT3, and SOCS1 We also identified mutations in KRAS, TP53, and TERT Type I EATL and type II EATL (monomorphic epitheliotropic intestinal T cell lymphoma) had highly overlapping genetic alterations indicating shared mechanisms underlying their pathogenesis. We modeled the effects of SETD2 loss in vivo by developing a T cell-specific knockout mouse. These mice manifested an expansion of γδ T cells, indicating novel roles for SETD2 in T cell development and lymphomagenesis. Our data render the most comprehensive genetic portrait yet of this uncommon but lethal disease and may inform future classification schemes.

14. Floods and fire ants, Solenopsis invicta (Hymenoptera: Formicidae): The Australian experience

Abstract

The rafting behaviour of the red imported fire ant, Solenopsis invicta, in response to flooding events is well documented, although studies generally have focussed on the mechanisms of raft assembly and the behaviour of the raft’s occupants. Flooding as a means of dispersal of S. invicta is frequently mentioned in the literature, although there are few data on the distances travelled or how effective it is compared to natural flight. In Australia, S. invicta is a priority invasive species with a national eradication program operating for 23 years, focussed on the population in southeast Queensland, which currently encompasses more than 700 000 ha. Flooding presents a risk to the success of the program through extending the infestation area or recolonising successfully treated areas. We used the program’s extensive spatiotemporal dataset of known fire ant colony locations to assess the effects of two significant flood events on the dispersal or displacement of S. invicta in Queensland. Results indicated that flooding did not spread S. invicta beyond the known boundaries of infestation but contributed to localised spread, particularly for sites with known polygyne infestations. This situation could change if the ant spreads to new river catchments. A novel method developed to assess the risk of S. invicta dispersal through flooding is presented, alongside program actions that can be applied to mitigate this risk.

18. Can an Earth-like Planet have a Titan-like Climate? Exploring the “In-Betweens” of Terrestrial Planetary Climate States

Abstract

The three planets of the Inner Solar System with significant atmospheres, Venus, Earth, and Mars, can be described as representing three “climate archetypes” of terrestrial planets: Venus is hot, dry, and rotates slowly; Mars is cold and dry, with fast rotation similar to Earth; Earth is the “middle ground”, warm enough to sustain liquid water on its surface but not so warm it evaporates away. These archetypes can be placed as endpoints on a spectrum of climates, where adjusting one or more planetary parameters can move a climate from one archetype to another, e.g. drying the surface can move an Earth-like planet towards the Venus and Mars archetypes. In addition to the three inner planets, there is one additional body in the Solar System that has a thick atmosphere and solid surface: Titan, a moon of Saturn. Titan presents a unique opportunity in observable planetary climates because it has a volatile liquid, or condensable, on its surface in the form of methane. This methane is able to evaporate to form clouds (Turtle et al., 2018) and likely rain (Turtle et al., 2011), but is mostly restricted to large polar lakes (Lunine and Lorenz, 2009) with the rest of the surface a vast desert (Mitchell and Lora, 2016). This means Titan’s climate archetype is between the ocean-dominated Earth and the fully-dry Venus/Mars.In this dissertation, we seek to further investigate the “in-betweens” of these climate archetypes, focusing on the transition between an Earth-like planet and a Titan-like one. To accomplish this, we recreate a Titan-like climate using an Earth-like global climate model (GCM) by varying a small set of planetary parameters. We first limit the available water by placing a continental land strip centered on the equator and varying its width. This mimics Titan’s dry tropics and wet poles, and could be similar to past continental arrangements in Earth’s history. Second, we take three of these land strip widths and vary the rotation period, starting with Earth’s rotation and moving towards Titan’s (16 Earth days). Third, for the same three land strip widths and using Earth’s rotation, we vary the volatility of the condensable via a constant multiplied to the saturation vapor pressure. Titan’s condensable, methane, is more volatile under Titan’s surface conditions than water is on Earth, resulting in high specific humidities. By artificially increasing the saturation vapor pressure, we can approximate this effect without changing the properties of the condensable.We find that simply replicating Titan’s parameters in our simulations does not fully reproduce Titan-like conditions. In addition, we find that it is possible to reproduce key Titan-like features by varying only the width of the equatorial land strips. This may indicate that there are many possible “in-between” states an Earth-like planet can have that span the gap between the Earth and Titan climate archetypes. It also suggests Titan’s current climate is primarily dependent on its surface liquid distribution, meaning an Earth-like planet with similar topography is likely to display the same features.

Abstract

Introduction: It has long been established that high-dose methotrexate is an essential part of therapy for primary central nervous system lymphoma. When regimens utilizing high-dose methotrexate were first studied, a dose of 8 g/m2 was used. More recently, reduced dosing strategies have been studied and adopted in attempts to reduce rates of adverse events. Studies utilizing 3.5 g/m2 of methotrexate have shown promising outcomes and improved rates of adverse events but there have never been any randomized head-to-head studies of differing dose levels of high-dose methotrexate. The purpose of this study was to compare efficacy and safety of different dosing strategies of high-dose methotrexate (HD-MTX) for primary central nervous system lymphoma (PCNSL). Methods: This single center retrospective review was conducted between 07/01/2013 to 6/3/2020. The patient population was separated into two arms based upon dose of methotrexate. The high intensity (HiHD) arm was defined as patients who received doses > 3.5 g/m2, while the low intensity (LiHD) arm received ≤ 3.5 g/m2. The primary endpoint was overall response rate (ORR) and secondary endpoints include efficacy via 2-year overall survival (OS), progression to transplant, and utilization of consolidation or salvage therapy. Safety was assessed through monitoring of relevant laboratory studies. Results: A total of 92 patients were included in this analysis. Baseline demographics were similar between groups, with the LiHD group trending toward older age. There were 78 patients eligible for assessment for ORR; there was no significant difference between the two groups (42.0% LiHD vs. 44.4% HiHD; p = 1.0). Rates of OS, progression to transplant and progression to consolidation chemotherapy were not different between groups. There were statistically significantly higher rates of renal and/or hepatic dysfunction with the first dose in the HiHD group compared with the LiHD group (11.5% LiHD vs. 64.3% HiHD; p ≤ 0.01). Conclusions: In this PCNSL patient cohort, there is no difference in terms of efficacy between HiHD LiHD methotrexate, but patients in the HiHD group had higher rates of renal and hepatic dysfunction.
