4. The extra ciliary roles of Meckel-Gruber syndrome proteins

Abstract

Meckel-Gruber syndrome (MKS) is a recessive genetic disease that is uniformly lethal in affected children due to resultant developmental defects in the kidney and brain. 13 MKS genes have been identified, and further candidate genes have been linked to this disease, all encoding unrelated proteins. Their role is believed to be in generation and compartmentalisation of the primary cilium, a microtubule-based organelle that functions in signal transduction of developmentally-crucial pathways. However, recent evidence indicates that these proteins are also likely involved in regulation of the actin cytoskeleton. Furthermore, research is beginning to uncover roles of other ciliopathy proteins in regulation of additional subcellular structures, such as the microtubule cytoskeleton, focal adhesions and the Golgi. To begin to understand the roles of the MKS proteins beyond the cilium, I examined a number of cellular features of patient fibroblasts carrying mutations in TMEM216 (MKS2) and TMEM67 (MKS3). In this thesis, I describe the temporal appearance and nature of prominent actin bundles observed in these cells, and analyse the dependency of these on the Rho/ROCK signalling pathway. Furthermore, I identify novel alterations to the microtubule cytoskeleton and organisation of the Golgi complex in MKS patient cells, and subsequently establish a temporal order of these phenotypes, demonstrating microtubule defects as the first to occur in these cells. Finally, I connect these phenotypic defects to Rho/ROCK signalling. In contrast to the prevailing view in the ciliopathy field, I believe that a diffusion barrier at the transition zone is not the primary role of MKS proteins. Instead I propose, supported by these data, that MKS protein complexes play a dual role as effectors of Rho signalling in addition to performing a structural role with particular importance in tethering the cytoskeleton to membranes. I therefore conclude that these, and other ciliopathy protein complexes, may act as important signal transduction and structural components at multiple locations throughout the cell.

Abstract

Three authors of the study have received financial support during the preparation of this manuscript. Kate McIntosh received summer internship funding from the University of St Andrews (funded internally by the University of St Andrews where Kate was an undergraduate student) to conduct analyses and visualise the data. Konstantinos Liverakos is in receipt of a PhD studentship from the AD Links Trust (funding external to the University of St Andrews where Konstantinos is a PhD student) and has conducted analyses, visualised data, and helped to prepared the manuscript during the period of his PhD. Christopher J.A. Moulin is a senior member of the Institut Universitaire de France. Metacognition is a domain which has illuminated our understanding of the regulation of cognition, but has yet to be applied in detail to more physical activities. We used half marathon finish time predictions from 7211 runners to investigate the factors that influence running performance metacognitive accuracy. In particular, we were concerned with the effects of experience, gender, and age on calibration. We expected more experienced runners to be better calibrated than less experienced ones. Given analogous findings in the domain of metacognition, we expected women to be less overconfident in their predictions, and better calibrated than male runners. Based on the metacognition literature, we expected that if older runners have effectively learned from previous experience, they would be as well-calibrated as younger runners. In contrast, uninformed inferences not based on performance feedback would lead to overestimating performance for older compared to younger runners. As expected, experience in terms of both club membership and previous race completion improved calibration. Unexpectedly though, females were more overconfident than males, overestimating their performance and demonstrating poorer calibration. A positive relationship was observed between age and prediction accuracy, with older runners showing better calibration. The present study demonstrates that data, collected before a test of physical activity, can inform our understanding of how participants anticipate their performance, and how this ability is affected by a number of demographic and situational variables. Athletes and coaches alike should be aware of these variables to better understand, organise, plan, and predict running performance, potentially leading to more appropriate training sessions and faster race finish times. Abstract

Abstract: A number of N 6-substituted adenosine-5′-N-methylcarboxamides were synthesised and their pharmacology, in terms of their receptor affinity, selectivity and cardioprotective effects, were explored. The first series of compounds, 4a–4f and 5a–5f, showed modest receptor affinity for the A3AR with K i values in the low to mid μM range. However, the incorporation of a 4-(2-aminoethyl)-2,6-di-tert-butylphenol group in the N 6-position (in compounds 4g and 5g) significantly improved the affinity with K i values of 30 and 9nM, respectively. Improvements in affinity, as well as selectivity were seen when a functionalised linker was introduced. The N 6-phenyl series, compounds 7a–7d, demonstrated low to mid nanomolar receptor affinities (K i =2.3–45.0nM), with 7b displaying 109-fold selectivity for the A3AR (vs A1). The N 6-benzyl series 9a–9c also proved to be potent and selective A3AR agonists and the longer chain length linker 13 was tolerated at the A3AR without abrogation of affinity or selectivity. Cardioprotection was demonstrated by a simulated ischaemia cell culture assay, whereby 7b, 7c, 9a, 9b and 9c all showed cardioprotective effects at 100nM comparable or better than the benchmark A3AR agonist IB-MECA, but which were indistinguishable by statistical analysis.
