Abstract

Performance during perceptual decision-making exhibits an inverted-U relationship with arousal, but the underlying network mechanisms remain unclear. Here, we recorded from auditory cortex (A1) of behaving mice during passive tone presentation, while tracking arousal via pupillometry. We found that tone discriminability in A1 ensembles was optimal at intermediate arousal, revealing a population-level neural correlate of the inverted-U relationship. We explained this arousal-dependent coding using a spiking network model with a clustered architecture. Specifically, we show that optimal stimulus discriminability is achieved near a transition between a multi-attractor phase with metastable cluster dynamics (low arousal) and a single-attractor phase (high arousal). Additional signatures of this transition include arousal-induced reductions of overall neural variability and the extent of stimulus-induced variability quenching, which we observed in the empirical data. Our results elucidate computational principles underlying interactions between pupil-linked arousal, sensory processing, and neural variability, and suggest a role for phase transitions in explaining nonlinear modulations of cortical computations.

Abstract

BACKGROUND: In Spring 2021, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) B.1.1.7 (Alpha) became the predominant variant in the United States. Research suggests that Alpha has increased transmissibility compared with non-Alpha lineages. We estimated household secondary infection risk (SIR), assessed characteristics associated with transmission, and compared symptoms of persons with Alpha and non-Alpha infections. METHODS: We followed households with SARS-CoV-2 infection for 2 weeks in San Diego County and metropolitan Denver, January to April 2021. We collected epidemiologic information and biospecimens for serology, reverse transcription-polymerase chain reaction (RT-PCR), and whole-genome sequencing. We stratified SIR and symptoms by lineage and identified characteristics associated with transmission using generalized estimating equations. RESULTS: We investigated 127 households with 322 household contacts; 72 households (56.7%) had member(s) with secondary infections. SIRs were not significantly higher for Alpha (61.0% [95% confidence interval, 52.4-69.0%]) than non-Alpha (55.6% [44.7-65.9%], P = .49). In households with Alpha, persons who identified as Asian or Hispanic/Latino had significantly higher SIRs than those who identified as White (P = .01 and .03, respectively). Close contact (eg, kissing, hugging) with primary cases was associated with increased transmission for all lineages. Persons with Alpha infection were more likely to report constitutional symptoms than persons with non-Alpha (86.9% vs 76.8%, P = .05). CONCLUSIONS: Household SIRs were similar for Alpha and non-Alpha. Comparable SIRs may be due to saturation of transmission risk in households due to extensive close contact, or true lack of difference in transmission rates. Avoiding close contact within households may reduce SARS-CoV-2 transmission for all lineages among household members.

Abstract

OBJECTIVE: To assess the household secondary infection risk (SIR) of B.1.1.7 (Alpha) and non-Alpha lineages of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) among children. STUDY DESIGN: During January to April 2021, we prospectively followed households with a SARS-CoV-2 infection. We collected questionnaires, serial nasopharyngeal swabs for reverse transcription polymerase chain reaction testing and whole genome sequencing, and serial blood samples for serology testing. We calculated SIRs by primary case age (pediatric vs adult), household contact age, and viral lineage. We evaluated risk factors associated with transmission and described symptom profiles among children. RESULTS: Among 36 households with pediatric primary cases, 21 (58%) had secondary infections. Among 91 households with adult primary cases, 51 (56%) had secondary infections. SIRs among pediatric and adult primary cases were 45% and 54%, respectively (OR, 0.79; 95% CI, 0.41-1.54). SIRs among pediatric primary cases with Alpha and non-Alpha lineage were 55% and 46%, respectively (OR, 1.52; 95% CI, 0.51-4.53). SIRs among pediatric and adult household contacts were 55% and 49%, respectively (OR, 1.01; 95% CI, 0.68-1.50). Among pediatric contacts, no significant differences in the odds of acquiring infection by demographic or household characteristics were observed. CONCLUSIONS: Household transmission of SARS-CoV-2 from children and adult primary cases to household members was frequent. The risk of secondary infection was similar among child and adult household contacts. Among children, household transmission of SARS-CoV-2 and the risk of secondary infection was not influenced by lineage. Continued mitigation strategies (eg, masking, physical distancing, vaccination) are needed to protect at-risk groups regardless of virus lineage circulating in communities.

Abstract

Recent studies in mice reveal widespread cortical signals during task performance; however, the various task-related and task-independent processes underlying this activity are incompletely understood. Here, we recorded wide-field neural activity, as revealed by GCaMP6s, from dorsal cortex while simultaneously monitoring orofacial movements, walking, and arousal (pupil diameter) of head-fixed mice performing a Go/NoGo visual detection task and examined the ability of task performance and spontaneous or task-related movements to predict cortical activity. A linear model was able to explain a significant fraction (33-55% of variance) of widefield dorsal cortical activity, with the largest factors being movements (facial, walk, eye), response choice (hit, miss, false alarm), and arousal and indicate that a significant fraction of trial-to-trial variability arises from both spontaneous and task-related changes in state (e.g., movements, arousal). Importantly, secondary motor cortex was highly correlated with lick rate, critical for optimal task performance (high d'), and was the first region to significantly predict the lick response on target trials. These findings suggest that secondary motor cortex is critically involved in the decision and performance of learned movements and indicate that a significant fraction of trial-to-trial variation in cortical activity results from spontaneous and task-related movements and variations in behavioral/arousal state.

Abstract

A Lagrangian numerical scheme for solving nonlinear degenerate Fokker-Planck equations in space dimensions $d\ge2$ is presented. It applies to a large class of nonlinear diffusion equations, whose dynamics are driven by internal energies and given external potentials, e.g. the porous medium equation and the fast diffusion equation. The key ingredient in our approach is the gradient flow structure of the dynamics. For discretization of the Lagrangian map, we use a finite subspace of linear maps in space and a variational form of the implicit Euler method in time. Thanks to that time discretisation, the fully discrete solution inherits energy estimates from the original gradient flow, and these lead to weak compactness of the trajectories in the continuous limit. Consistency is analyzed in the planar situation, $d=2$. A variety of numerical experiments for the porous medium equation indicates that the scheme is well-adapted to track the growth of the solution's support., Comment: 32 pages, 7 figures

Abstract

Borrelia miyamotoi, transmitted by Ixodes spp. ticks, was recognized as an agent of hard tick relapsing fever in the United States in 2013. Nine state health departments in the Northeast and Midwest have conducted public health surveillance for this emerging condition by using a shared, working surveillance case definition. During 2013-2019, a total of 300 cases were identified through surveillance; 166 (55%) were classified as confirmed and 134 (45%) as possible. Median age of case-patients was 52 years (range 1-86 years); 52% were male. Most cases (70%) occurred during June-September, with a peak in August. Fever and headache were common symptoms; 28% of case-patients reported recurring fevers, 55% had arthralgia, and 16% had a rash. Thirteen percent of patients were hospitalized, and no deaths were reported. Ongoing surveillance will improve understanding of the incidence and clinical severity of this emerging disease. This paper establishes the local-in-time existence and uniqueness of solutions to the viscous, non-resistive magnetohydrodynamics (MHD) equations in $\mathbb{R}^d$, $d=2,3$, with initial data $B_0\in H^s(\mathbb{R}^d)$ and $u_0\in H^{s-1+\varepsilon}(\mathbb{R}^d)$ for $s>d/2$ and any $0<\varepsilon<1$. The proof relies on maximal regularity estimates for the Stokes equation. The obstruction to taking $\varepsilon=0$ is explained by the failure of solutions of the heat equation with initial data $u_0\in H^{s-1}$ to satisfy $u\in L^1(0,T;H^{s+1})$; we provide an explicit example of this phenomenon.

If $u$ is a smooth solution of the Navier--Stokes equations on ${\mathbb R}^3$ with first blowup time $T$, we prove lower bounds for $u$ in the Sobolev spaces $\dot H^{3/2}$, $\dot H^{5/2}$, and the Besov space $\dot B^{5/2}_{2,1}$, with optimal rates of blowup: we prove the strong lower bounds $\|u(t)\|_{\dot H^{3/2}}\ge c(T-t)^{-1/2}$ and $\|u(t)\|_{\dot B^{5/2}_{2,1}}\ge c(T-t)^{-1}$, but in $\dot H^{5/2}$ we only obtain the weaker result $\limsup_{t\to T^-}(T-t)\|u(t)\|_{\dot H^{5/2}}\ge c$. In this paper we prove the existence of solutions to the viscous, non-resistive magnetohydrodynamics (MHD) equations on the whole of $\mathbb{R}^{n}$, $n=2,3$, for divergence-free initial data in certain Besov spaces, namely $\boldsymbol{u}_{0} \in B^{n/2-1}_{2,1}$ and $\boldsymbol{B}_{0} \in B^{n/2}_{2,1}$. The a priori estimates include the term $\int_{0}^{t} \| \boldsymbol{u}(s) \|_{H^{n/2}}^{2} \, \mathrm{d} s$ on the right-hand side, which thus requires an auxiliary bound in $H^{n/2-1}$. In 2D, this is simply achieved using the standard energy inequality; but in 3D an auxiliary estimate in $H^{1/2}$ is required, which we prove using the splitting method of Calder\'on (Trans. Amer. Math. Soc. 318(1), 179--200, 1990). By contrast, we prove that such solutions are unique in 3D, but the proof of uniqueness in 2D is more difficult and remains open.

Abstract

Cortical feedback pathways are hypothesized to distribute context-dependent signals during flexible behavior. Recent experimental work has attempted to understand the mechanisms by which cortical feedback inputs modulate their target regions. Within the mouse whisker sensorimotor system, cortical feedback stimulation modulates spontaneous activity and sensory responsiveness, leading to enhanced sensory representations. However, the cellular mechanisms underlying these effects are currently unknown. In this study we use a simplified neural circuit model, which includes two recurrent excitatory populations and global inhibition, to simulate cortical modulation. First, we demonstrate how changes in the strengths of excitation and inhibition alter the input-output processing responses of our model. Second, we compare these responses with experimental findings from cortical feedback stimulation. Our analyses predict that enhanced inhibition underlies the changes in spontaneous and sensory evoked activity observed experimentally. More generally, these analyses provide a framework for relating cellular and synaptic properties to emergent circuit function and dynamic modulation.

Abstract

Unit recordings in behaving animals have revealed the transformation of sensory to motor representations in cortical neurons. However, we still lack basic insights into the mechanisms by which neurons interact to generate such transformations. Here, we study cortical circuits related to behavioral control in mice engaged in a sensory detection task. We recorded neural activity using extracellular and intracellular techniques and analyzed the task-related neural dynamics to reveal underlying circuit processes. Within motor cortex, we find two populations of neurons that have opposing spiking patterns in anticipation of movement. From correlation analyses and circuit modeling, we suggest that these dynamics reflect neural ensembles engaged in a competition. Furthermore, we demonstrate how this competitive circuit may convert a transient, sensory stimulus into a motor command. Together, these data reveal cellular and circuit processes underlying behavioral control and establish an essential framework for future studies linking cellular activity to behavior.

Abstract

The state of the brain and body constantly varies on rapid and slow timescales. These variations contribute to the apparent noisiness of sensory responses at both the neural and the behavioral level. Recent investigations of rapid state changes in awake, behaving animals have provided insight into the mechanisms by which optimal sensory encoding and behavioral performance are achieved. Fluctuations in state, as indexed by pupillometry, impact both the "signal" (sensory evoked response) and the "noise" (spontaneous activity) of cortical responses. By taking these fluctuations into account, neural response (co)variability is significantly reduced, revealing the brain to be more reliable and predictable than previously thought.

Abstract

We prove existence, uniqueness and regularity of weak solutions of a coupled parabolic-elliptic model in two dimensions; we consider the standard equations of magnetohydrodynamics with the advective terms removed from the velocity equation. Despite the apparent simplicity of the model, the proof requires results that are at the limit of what is available, including elliptic regularity in $L^{1}$ and a strengthened form of the Ladyzhenskaya inequality \[ \| f \|_{L^{4}} \leq c \| f \|_{L^{2,\infty}}^{1/2} \| \nabla f \|_{L^{2}}^{1/2}, \] which we derive using the theory of interpolation. The model has applications to the method of magnetic relaxation, introduced by Moffatt (J. Fluid. Mech. 159, 359-378, 1985), to construct stationary Euler flows with non-trivial topology.

Abstract

Using elementary arguments based on the Fourier transform we prove that for $1 \leq q < p < \infty$ and $s \geq 0$ with $s > n(1/2-1/p)$, if $f \in L^{q,\infty}(\R^n) \cap \dot{H}^s(\R^n)$ then $f \in L^p(\R^n)$ and there exists a constant $c_{p,q,s}$ such that \[ \|f\|_{L^p} \leq c_{p,q,s} \|f\|_{L^{q,\infty}}^\theta \|f\|_{\dot H^s}^{1-\theta}, \] where $1/p = \theta/q + (1-\theta)(1/2-s/n)$. In particular, in $\R^2$ we obtain the generalised Ladyzhenskaya inequality $\|f\|_{L^4}\le c\|f\|_{L^{2,\infty}}^{1/2}\|f\|_{\dot H^1}^{1/2}$. We also show that for $s=n/2$ the norm in $\|f\|_{\dot H^{n/2}}$ can be replaced by the norm in BMO. As well as giving relatively simple proofs of these inequalities, this paper provides a brief primer of some basic concepts in harmonic analysis, including weak spaces, the Fourier transform, the Lebesgue Differentiation Theorem, and Calderon-Zygmund decompositions.

Abstract

How the brain takes in information, makes a decision, and acts on this decision is strongly influenced by the ongoing and constant fluctuations of state. Understanding the nature of these brain states and how they are controlled is critical to making sense of how the nervous system operates, both normally and abnormally. While broadly projecting neuromodulatory systems acting through metabotropic pathways have long been appreciated to be critical for determining brain state, more recent investigations have revealed a prominent role for fast acting neurotransmitter pathways for temporally and spatially precise control of neural processing. Corticocortical and thalamocortical glutamatergic projections can rapidly and precisely control brain state by changing both the nature of ongoing activity and by controlling the gain and precision of neural responses.

This thesis establishes the existence and uniqueness of solutions to certain systems of equations connected to magnetohydrodynamics (MHD). The models have potential applications to the method of magnetic relaxation introduced by Moffatt (J. Fluid. Mech. 159, 359-378, 1985) to construct stationary Euler ows with non-trivial topology. Firstly, we prove existence, uniqueness and regularity of weak solutions of a coupled parabolic-elliptic model in 2D, and existence of weak solutions in 3D; we consider the standard equations of MHD with the advective terms removed from the velocity equation. Despite the apparent simplicity of the model, the proof in 2D requires results that are at the limit of what is available, including elliptic regularity in L1 and a strengthened form of the Ladyzhenskaya inequality ||f||L3≤c||f||1/2 L2,∞||∇f||1/2 L2 Secondly, we establish the local-in-time existence and uniqueness of strong solutions in Hs for s>n=2 to the viscous, non-resistive MHD equations in Rn, n = 2; 3, as well as for a related model where the advection terms are removed from the velocity equation (the above parabolic-elliptic system with zero resistivity). The uniform bounds required for proving existence are established by means of a new estimate, which is a partial generalisation of the commutator estimate of Kato & Ponce (Comm. Pure Appl. Math. 41(7), 891-907, 1988). Finally, we generalise the results of the previous chapter to prove the localin-time existence of strong solutions in the Besov space Bn/2 2;1 (ℝn) to the viscous, non-resistive MHD equations in ℝn.
