Abstract

A non-perturbative approach to the solution of the time-dependent, two-center Dirac equation is presented with a special emphasis on the proper treatment of the potential of the nuclei. In order to account for the full multipole expansion of this potential, we express eigenfunctions of the two-center Hamiltonian in terms of well-known solutions of the "monopole" problem that employs solely the spherically-symmetric part of the interaction. When combined with the coupled-channel method, such a wavefunction-expansion technique allows for an accurate description of the electron dynamics in the field of moving ions for a wide range of internuclear distances. To illustrate the applicability of the proposed approach, the probabilities of the K- as well as L- shell ionization of hydrogen-like ions in the course of nuclear alpha-decay and slow ion-ion collisions have been calculated.

Abstract

Mouse embryos bearing hypomorphic and conditional null Fgf8 mutations have small and abnormally patterned telencephalons. We provide evidence that the hypoplasia results from decreased Foxg1 expression, reduced cell proliferation and increased cell death. In addition, alterations in the expression of Bmp4, Wnt8b, Nkx2.1 and Shh are associated with abnormal development of dorsal and ventral structures. Furthermore, nonlinear effects of Fgf8 gene dose on the expression of a subset of genes, including Bmp4 and Msx1, correlate with a holoprosencephaly phenotype and with the nonlinear expression of transcription factors that regulate neocortical patterning. These data suggest that Fgf8 functions to coordinate multiple patterning centers, and that modi. cations in the relative strength of FGF signaling can have profound effects on the relative size and nature of telencephalic subdivisions.

5. Studies on the molecular biology of sex determination in the tilapias

Abstract

This study set out to develop molecular biological techniques for the study of sex determination in the tilapias, and, in particular, in Oreochromis niloticus (Linnaeus), Lake Manazala strain. Two heterospecific probes, pDP1007 (supplied by Dr. D. Page) which gives a Y-chromosome specific hybridization pattern in humans, and pUGD0600 (supplied by Dr. S. Mizuno) which is W chromosome specific in chickens, were tested for sex specificity in Oreochromis niloticus. Results from hybridizations of these probes to Southern blotted digests of O.niloticus DNA are reported. Both probes hybridized to O.niloticus genomic DNA but neither gave sex specific patterns. Four short repeat oligonucleotide sequences were synthesized. Two sequences were labelled both non-radioactively, with dioxygenin and with 32P and were used to probe Southern blots of O.niloticus DNA and dried agarose gels containing O.niloticus DNA. The probes (GATA)4(GACA)1 and (GATA)3(GACA)2 both gave fingerprint like patterns but no sex specific banding patterns were observed. The implications of these results for the organization of repetitive sequences in the O.niloticus genome are discussed. A partial DNA library, enriched for male specific sequences was created by subtractive hybridization using the Phenol Emulsion Reassociation Technique (PERT). Thirteen recombinant colonies containing inserts were isolated. Inserts from the library were screened for sex specificity. Two inserts were partially sequenced and PCR primers for them were constructed which were used to amplify the sequences in genomic DNA. Both sequences were successfully amplified from genomic DNA but neither was male specific. The amplified sequences were restricted with a number of restriction enzymes to identify sex specific restriction fragment length polymorphisms (RFLPs) but, again, none were observed. Sequences were also tested for similarity to sequences in the EMBL/GENBANK nucleotide databases using the suite of programs available on SEQNET (Daresbury laboratory). Future approaches to the isolation of sex specific DNA sequences in the Tilapias are discussed in detail.

8. Early Literacy Development: Skill Growth and Relations between Classroom Variables for Preschool Children

Abstract

Promoting the development of early literacy skills might prevent later reading achievement problems. This longitudinal study investigated rates of early literacy growth using Individual Growth and Development Indicators (IGDIs) for 69 preschool children classified as belonging to 1 of 4 groups with or without risk for developing early literacy skills. Group differences were obtained for 3 IGDIs (Picture Naming, Rhyming, and Alliteration) and rate of growth over time varied across groups for Picture Naming and Rhyming. Percent time spent in various classroom situations, as measured by the Ecobehavioral System for the Complex Assessment of Preschool Environments (ESCAPE), was correlated with each child's rate of IGDI growth. Correlations between IGDI growth rates and ESCAPE variables suggest classroom areas in which interventions could be concentrated for specific groups of children. Implications for practice and future research are discussed.

Abstract

Background The oncolytic virus, coxsackievirus A21 (CVA21), has shown promise as a single agent in several clinical trials and is now being tested in combination with immune checkpoint blockade. Combination therapies offer the best chance of disease control; however, the design of successful combination strategies requires a deeper understanding of the mechanisms underpinning CVA21 efficacy, in particular, the role of CVA21 anti-tumor immunity. Therefore, this study aimed to examine the ability of CVA21 to induce human anti-tumor immunity, and identify the cellular mechanism responsible. Methods This study utilized peripheral blood mononuclear cells from i) healthy donors, ii) Acute Myeloid Leukemia (AML) patients, and iii) patients taking part in the STORM clinical trial, who received intravenous CVA21; patients receiving intravenous CVA21 were consented separately in accordance with local institutional ethics review and approval. Collectively, these blood samples were used to characterize the development of innate and adaptive anti-tumor immune responses following CVA21 treatment. Results An Initial characterization of peripheral blood mononuclear cells, collected from cancer patients following intravenous infusion of CVA21, confirmed that CVA21 activated immune effector cells in patients. Next, using hematological disease models which were sensitive (Multiple Myeloma; MM) or resistant (AML) to CVA21-direct oncolysis, we demonstrated that CVA21 stimulated potent anti-tumor immune responses, including: 1) cytokine-mediated bystander killing; 2) enhanced natural killer cell-mediated cellular cytotoxicity; and 3) priming of tumor-specific cytotoxic T lymphocytes, with specificity towards known tumor-associated antigens. Importantly, immune-mediated killing of both MM and AML, despite AML cells being resistant to CVA21-direct oncolysis, was observed. Upon further examination of the cellular mechanisms responsible for CVA21-induced anti-tumor immunity we have identified the importance of type I IFN for NK cell activation, and demonstrated that both ICAM-1 and plasmacytoid dendritic cells were key mediators of this response. Conclusion This work supports the development of CVA21 as an immunotherapeutic agent for the treatment of both AML and MM. Additionally, the data presented provides an important insight into the mechanisms of CVA21-mediated immunotherapy to aid the development of clinical biomarkers to predict response and rationalize future drug combinations. 18. The complete mitochondrial genome of the Australian Common Rock Rat, Zyzomys argurus

Abstract

The Common Rock Rat Zyzomys argurus is an abundant small- to medium-sized Murid rodent that is endemic to Australia. It is a nocturnal mammal with a mostly herbivorous diet. This species is native to the wet/dry tropics of Northern Australia and can be identified from other rock rats on the basis of its small size and its tail length (which is at least equivalent to its head-body length). Here, we describe the complete mitochondrial genome of Z. argurus and compare it to other Rodentia. The Z. argurus circular mitogenome was 16,261 bp and contained 13 protein-coding genes, two rRNA genes, 22 tRNAs and a control region (D-loop) of 859 bp. Phylogenetic analysis of selected, published sequenced mitogenomes reveal it is most closely related to the Lakeland Downs mouse Leggadina lakedownensis in the order Rodentia.

Abstract

The Thermal Control Subsystem (TCS) is a key part of the Advanced Plant Habitat (APH) for the International Space Station (ISS). The purpose of this subsystem is to provide thermal control, mainly cooling, to the other APH subsystems. One of these subsystems, the Environmental Control Subsystem (ECS), controls the temperature and humidity of the growth chamber (GC) air to optimize the growth of plants in the habitat. The TCS provides thermal control to the ECS with three cold plates, which use Thermoelectric Coolers (TECs) to heat or cool water as needed to control the air temperature in the ECS system. In order to optimize the TCS design, pressure drop and heat transfer analyses were needed. The analysis for this system was performed in Siemens NX Thermal/Flow software (Version 8.5). NX Thermal/Flow has the ability to perform 1D or 3D flow solutions. The 1D flow solver can be used to represent simple geometries, such as pipes and tubes. The 1D flow method also has the ability to simulate either fluid only or fluid and wall regions. The 3D flow solver is similar to other Computational Fluid Dynamic (CFD) software. TCS performance was analyzed using both the 1D and 3D solvers. Each method produced different results, which will be evaluated and discussed.

27. Parentage assignment using microsatellite DNA typing for the endangered numbat (Myrmecobius fasciatus)

Abstract

The numbat (Myrmecobius fasciatus) is an endangered and peculiar marsupial with a diet that consists almost exclusively of termites. This study developed a parentage-testing system for numbats using microsatellite markers. Nineteen loci detected 143 alleles, with 4-13 alleles/locus and average expected heterozygosity of 77% (range 0.665-0.855). The total parentage exclusion probability was >0.9999 (given only the genotype of the offspring), >0.9999 for excluding a candidate parent from the parentage of an arbitrary offspring (given the genotype of the offspring and parent) and the probability of identity for full-sibs was 4.6 × 10-9. Overall, these microsatellites offer a simple and highly informative marker-set for parentage identification in numbats.

Abstract

A number of novel and known genes whose spatiotemporal expression patterns suggest that they may play important roles in the ontogeny of the brain have been isolated. Using in situ hybridization, we present the expression pattern in the developing rat brain of one of these molecules--a gene previously reported by Kang et al. (1998, J. Cell Biol. 143, 403) and named CDO. cdo appears to be a receptor, with an extracellular domain of 967 residues consisting of five immunoglobulin domains, three fibronectin-like domains, one transmembrane of 26 residues, and a cytoplasmic domain of 255 residues. Functional studies of CDO in immortalized myoblast cell lines have implicated a role for CDO in the decision process of myoblasts to undergo differentiation. The temporal onset and restricted expression of cdo in the CNS suggest an important role for cdo in the developing nervous system.
