Abstract

This review paper describes the energy-upgraded CEBAF accelerator. This superconducting linac has achieved 12 GeV beam energy by adding 11 new high-performance cryomodules containing eighty-eight superconducting cavities that have operated CW at an average accelerating gradient of 20 MV/m. After reviewing the attributes and performance of the previous 6 GeV CEBAF accelerator, we discuss the upgraded CEBAF accelerator system in detail with particular attention paid to the new beam acceleration systems. In addition to doubling the acceleration in each linac, the upgrade included improving the beam recirculation magnets, adding more helium cooling capacity to allow the newly installed modules to run cold, adding a new experimental hall, and improving numerous other accelerator components. We review several of the techniques deployed to operate and analyze the accelerator performance, and document system operating experience and performance. In the final portion of the document, we present much of the current planning regarding projects to improve accelerator performance and enhance operating margins, and our plans for ensuring CEBAF operates reliably into the future. For the benefit of potential users of CEBAF, the performance and quality measures for beam delivered to each of the experimental halls is summarized in the appendix., Comment: 66 pages, 73 figures, 21 tables

Abstract

The Polarized Electrons for Polarized Positrons experiment at the injector of the Continuous Electron Beam Accelerator Facility has demonstrated for the first time the efficient transfer of polarization from electrons to positrons produced by the polarized bremsstrahlung radiation induced by a polarized electron beam in a high-$Z$ target. Positron polarization up to 82\% have been measured for an initial electron beam momentum of 8.19~MeV/$c$, limited only by the electron beam polarization. This technique extends polarized positron capabilities from GeV to MeV electron beams, and opens access to polarized positron beam physics to a wide community., Comment: 5 pages, 4 figures

Abstract

Habitat protection is a key component of endangered species conservation, but critical habitat designations are often based on limited data or habitat use during only a portion of a species' life cycle. Protected habitat around breeding pools for the endangered Jefferson Salamander (Ambystoma jeffersonianum (Green, 1827)) and their unisexual dependents (Ambystoma laterale--(2) jeffersonianum) is based upon limited movement data from primarily spring and summer seasons. Furthermore, despite their treatment as distinct species under Canada's Species at Risk Act, differences in habitat use have not been directly compared inareas where they co-occur. We used radiotelemetry to track A. jeffersonianum (JJ) and A. laterale--jeffersonianum (LJJ and LJJJ) during fall migrations to overwintering sites. We used these data and summarized available movement data from past studies that tracked movements in other periods of the annual cycle to estimate year-round critical habitat. Ambystoma jeffersonianum travelled significantly longer distances to overwintering locations than unisexuals. Individuals were more likely to overwinter next to a similar genomotype individual than not. Critical habitat encompassing the entire annual cycle of A. jeffersonianum extends up to 400-450 m from breeding ponds indicating existing regulatory habitat protections in Canada do not currently protect sufficient habitat. Si la protection des habitats est un element cle de la conservation des especes menacees, la designation d'habitats essentiels repose souvent sur des donnees limitees ou l'utilisation d'habitats durant une partie seulement du cycle biologique de l'espece. Les habitats proteges autour d'etangs de reproduction de salamandres de Jefferson (Ambystoma jeffersonianum (Green, 1827)) et de leurs dependants unisexues (Ambystoma laterale--(2) jeffersonianum) sont bases sur des donnees limitees sur leurs deplacements principalement pour le printemps et l'ete. En outre, malgre le fait qu'elles soient traitees comme des especes distinctes en vertu de la Loi sur les especes en peril du Canada, les differences d'utilisation de l'habitat des deux especes dans des lieux ou elles cooccurrent n'ont pas ete examinees. Nous avons utilise la radiotelemetrie pour suivre des A. jeffersonianum (JJ) et des A. laterale--jeffersonianum (LJJ et LJJJ) durant des migrations automnales vers des sites d'hivernage. Nous utilisons ces donnees et un condense des donnees sur les deplacements disponibles tirees d'etudes anterieures qui ont suivi les deplacements pendant d'autres periodes du cycle annuel, afin d'estimer les habitats essentiels a l'echelle annuelle. Les A. jeffersonianum parcourent des distances significativement plus longues vers les lieux d'hivernage que les ambystoma unisexues. Les individus sont plus susceptibles d'hiverner a cote d'un individu de genomotype semblable que de genomotype different. Abstract

The X-33 is a flying testbed to evaluate technologies and designs for a reusable Single Stage To Orbit (SSTO) production vehicle. Although it is sub-orbital, it is trans-atmospheric. This paper will discuss the abort capabilities, both commanded and autonomous, available to the X-33. The cornerstone of the abort capabilities is the Performance Monitor (PM) and it's supporting software. PM is an on-board 3-DOF simulation, which evaluates the vehicle ability to execute the current trajectory. The Abort Manager evaluates the results from PM, and, when indicated, computes and implements an abort trajectory.

Abstract

This paper presents a new marine exposure site being developed on the North-west Atlantic coastline of Ireland in Co. Donegal by the Centre for Built Environment Research at Queen’s University Belfast. The site will initially contain a number of large precast concrete stems, each 1.5m high, 1.5m wide and 1m thick placed on concrete plinths poured in-situ. The concrete stems will be placed at three levels to achieve different exposure conditions outlined in EN 206, namely atmospheric (XS1), a splash or spray zone (XS3) and a tidal zone (XS3) where the stems will be submerged by the incoming tide twice daily. The concrete will consist of different cements and appropriate w/b ratios suitable for this type of exposure. The permeability and diffusion properties will be measured using non-destructive tests developed at Queen’s University Belfast, namely the Autoclam permeability system and the Permit ion migration test. Other tests to measure the corrosion of the embedded rebar will also be undertaken. In order to monitor the internal concrete properties, electrical sensors will be attached to the rebar and embedded in concrete. The information from these sensors will be relayed back to the office using remote wireless technology. Such integrated monitoring systems for concrete structures can reduce assessment and repair costs by continuously profiling the covercrete and corrosion for the ingress of various deleterious substances, such as chlorides, in the reinforcing steel in real-time. This approach permits an informed assessment of the performance of the structure throughout its service life. This site will form part of a world-wide exposure study, including similar sites in Scotland, India and China.

32. Probability of satellite collision

Abstract

A method is presented for computing the probability of a collision between a particular artificial earth satellite and any one of the total population of earth satellites. The collision hazard incurred by the proposed modular Space Station is assessed using the technique presented. The results of a parametric study to determine what type of satellite orbits produce the greatest contribution to the total collision probability are presented. Collision probability for the Space Station is given as a function of Space Station altitude and inclination. Collision probability was also parameterized over miss distance and mission duration.

Abstract

We have prepared a series of achiral aminoacetonitriles, bearing tri-ring benzamide moieties and an aminocyclohexanecarboxylate residue at P2. This combination of binding elements resulted in sub-250 pM, reversible, selective, and orally bioavailable cathepsin K inhibitors. Lead compounds displayed single digit nanomolar inhibition in vitro (of rabbit osteoclast-mediated degradation of bovine bone). The best compound in this series, 39n (CRA-013783/L-006235), was orally bioavailable in rats, with a terminal half-life of over 3 h. 39n was dosed orally in ovariectomized rhesus monkeys once per day for 7 days. Collagen breakdown products were reduced by up to 76% dose-dependently. Plasma concentrations of 39n above the bone resorption IC50 after 24 h indicated a correlation between functional cellular and in vivo assays. Inhibition of collagen breakdown by cathepsin K inhibitors suggests this mechanism of action may be useful in osteoporosis and other indications involving bone resorption.

47. Identification of Glu-268 as the catalytic nucleophile of human lysosomal beta-galactosidase precursor by mass spectrometry.

Abstract

Human lysosomal beta-galactosidase catalyzes the hydrolysis of beta-galactosides via a double displacement mechanism involving a covalent glycosyl enzyme intermediate. By use of the slow substrate 2,4-dinitrophenyl-2-deoxy-2-fluoro-beta-D-galactopyranoside, a glycosyl enzyme intermediate has been trapped on the enzyme. This has allowed the catalytic nucleophile to be identified as Glu-268 by peptic and tryptic digestion of the inactivated enzyme followed by high performance liquid chromatography-electrospray ionization tandem mass spectrometry of the peptide mixture. This glutamic acid is fully conserved in a sequence-related family of enzymes (Family 35), consistent with its essential role.

Abstract

Synopsis This paper examines the relationship between mix proportions, electrical properties of the constituents of concrete and the over-all electrical resistivity for concrete. The mechanisms for the conduction of electricity through the heterogeneous medium of concrete are discussed and an electrical model is proposed. Analysis from a purely theoretical standpoint is also described and the values of electrical resistivity obtained experimentally are shown to compare favourably with the theoretical model.

49. Unequivocal identification of Asp-214 as the catalytic nucleophile of Saccharomyces cerevisiae alpha-glucosidase using 5-fluoro glycosyl fluorides.

Abstract

Yeast alpha-glucosidase is a member of a sequence-related family of alpha-glycosidases (Family 13) that includes important digestive alpha-amylases and alpha-glucosidases. These enzymes catalyze the hydrolysis of alpha-linked oligosaccharides by a two-step mechanism involving a glycosyl-enzyme intermediate. This intermediate can be trapped by use of 5-fluoro-alpha-D-glucosyl fluoride or 5-fluoro-beta-L-idosyl fluoride, members of a new class of mechanism-based glycosidase inactivators. Both of these trapped 5-fluoro glycosyl enzyme intermediates are catalytically competent, turning over when freed of excess inactivator and releasing free enzyme. Two glycosylated peptides in proteolytic digests of these trapped glycosyl enzyme intermediates were identified by use of neutral loss scans on an electrospray ionization triple quadrupole mass spectrometer. Further tandem mass spectrometric analysis in daughter ion scan mode allowed identification of Asp-214 as the catalytic nucleophile in yeast alpha-glucosidase, and this identification was confirmed by aminolysis of the labeled peptide and high resolution mass spectrometry. This residue is one of three active site carboxylates that are completely conserved in this family, thus confirming the role of Asp-214 and the equivalent residues in other family members as the catalytic nucleophile. The other two conserved carboxylates are likely involved in acid/base catalysis.

50. Genetic studies of the ploidy of Moloney murine leukemia virus

Abstract

An assay for Moloney murine leukemia virus was developed that made use of the production of morphologically altered foci in nonproducer mouse cells (15F) carrying murine sarcoma virus. Wild-type (wt) virus gave a ratio of titers at 39 degrees C/34degrees C = 1.05 +/- 0.45 (standard deviation;n = 20). A spontaneous, thermosensitive (ts) mutant of Moloney murine leukemia virus, ts3, defective in a late viral function, gave 39 degrees C/34degrees C = 0. A murine cell line (TB) was mixedly infected with ts3 and wt (multiplicities of infection, 7.8:4.3), cloned after infection, and shown to be infected by both viruses. At 34 degrees C it produced wt, ts, and particles of mixed parentage. The heterozygotes (hz) had ratios of assays 39 degrees C/34 degrees C = 0.06 to 0.84 (mean, 0.36). To eliminate possible interference by multiploid particles with determination of the proportions of the three types of particles, the virus produced by the mixedly infected, cloned cell line at 34 degrees C was distributed by velocity sedimentation in a sucrose gradient, and virus was picked from the lightest part of the gradient. The proportions of ts, wt, and hz were 0.27, 0.26, and 0.47. Those particles identified as hz segreated ts, wt, and hz in the proportions 0.24, 0.27, and 0.49, respectively. These values were not significantly different from those predicted from a diploid model of the genome.
