BACKGROUND:Spinocerebellar ataxia type 28 (SCA28) is a dominantly inherited neurodegenerative disease caused by pathogenic variants in AFG3L2. The AFG3L2 protein is a subunit of mitochondrial m-AAA complexes involved in protein quality control. Objective of this study was to determine the molecular mechanisms of SCA28, which has eluded characterisation to date. METHODS:We derived SCA28 patient fibroblasts carrying different pathogenic variants in the AFG3L2 proteolytic domain (missense: the newly identified p.F664S and p.M666T, p.G671R, p.Y689H and a truncating frameshift p.L556fs) and analysed multiple aspects of mitochondrial physiology. As reference of residual m-AAA activity, we included SPAX5 patient fibroblasts with homozygous p.Y616C pathogenic variant, AFG3L2+/- HEK293 T cells by CRISPR/Cas9-genome editing and Afg3l2 -/- murine fibroblasts. RESULTS:We found that SCA28 cells carrying missense changes have normal levels of assembled m-AAA complexes, while the cells with a truncating pathogenic variant had only half of this amount. We disclosed inefficient mitochondrial fusion in SCA28 cells caused by increased OPA1 processing operated by hyperactivated OMA1. Notably, we found altered mitochondrial proteostasis to be the trigger of OMA1 activation in SCA28 cells, with pharmacological attenuation of mitochondrial protein synthesis resulting in stabilised levels of OMA1 and OPA1 long forms, which rescued mitochondrial fusion efficiency. Secondary to altered mitochondrial morphology, mitochondrial calcium uptake resulted decreased in SCA28 cells. CONCLUSION:Our data identify the earliest events in SCA28 pathogenesis and open new perspectives for therapy. By identifying similar mitochondrial phenotypes between SCA28 cells and AFG3L2+/- cells, our results support haploinsufficiency as the mechanism for the studied pathogenic variants.

Sectors of the Campania region (southern Italy), where pyroclastic airfall deposits cover carbonate slopes, have frequently experienced debris avalanches and debris flows. These landslides are triggered by heavy rainfall and in the last century caused severe destruction and hundreds of deaths. Their occurrence is controlled by the interaction of several factors, both natural and human induced. The initial failure is linked with the presence of morphological discontinuities such as rocky scarps and road cuts, which interrupt the stratigraphical continuity of an “infinite slope”. Here the main criteria for susceptibility analysis are defined trying to develop a procedure for identify highly-susceptible sources, runout and invasion areas.

Coastal cliff erosion is a significant hazard for the safety of people, buildings, utilities and infrastructure, given the sudden and episodic nature of the retreat process in time and space. Thus, understanding past retreat trends and a regular diagnosis of the cliff face condition is fundamental for risk management. In this study we show preliminary results of a project aimed at the definition of the coastal erosion (in terms of temporal and spatial scales) of a small portion of cliff located along the Maronti beach in Ischia, Italy. Drone-based and terrestrial acquisition approaches offered the opportunity to exploit photogrammetric techniques to estimate the spatial and temporal rate of change of the investigated portion of cliff. In this study, cliff face topographical evolution is evaluated out by comparing the results obtained from three photographic datasets (variable approach either boat and UAV-based) obtained in 2011, 2021 and 2023. Spatial distribution of two main instability events dated back to 2020 and 2022 were well represented in the maps of change generated with the M3C2 algorithm. The comparison of point clouds for the period 2011-2021 and 2021-2023 produced max retreat rate of the cliff face of approximately 12 and 25 m, respectively. Results highlighted the need for robust co-registration methods to accurately estimate erosion rates. Critical issues are discussed to highlight potentials and limitations encountered with the abovementioned multi-temporal cliff evolution assessment techniques. [ABSTRACT FROM AUTHOR]

- Abstract
Objective: Trained immunity (TI) is a de facto memory program of innate immune cells, characterized by immunometabolic and epigenetic changes sustaining enhanced production of cytokines. TI evolved as a protective mechanism against infections; however, inappropriate activation can cause detrimental inflammation and might be implicated in the pathogenesis of chronic inflammatory diseases. In this study, we investigated the role of TI in the pathogenesis of giant cell arteritis (GCA), a large-vessel vasculitis characterized by aberrant macrophage activation and excess cytokine production. Methods: Monocytes from GCA patients and from age- and sex-matched healthy donors were subjected to polyfunctional studies, including cytokine production assays at baseline and following stimulation, intracellular metabolomics, chromatin immunoprecipitation-qPCR, and combined ATAC/RNA sequencing. Immunometabolic activation (i.e. glycolysis) was assessed in inflamed vessels of GCA patients with FDG-PET and immunohistochemistry (IHC), and the role of this pathway in sustaining cytokine production was confirmed with selective pharmacologic inhibition in GCA monocytes. Results: GCA monocytes exhibited hallmark molecular features of TI. Specifically, these included enhanced IL-6 production upon stimulation, typical immunometabolic changes (e.g. increased glycolysis and glutaminolysis) and epigenetic changes promoting enhanced transcription of genes governing pro-inflammatory activation. Immunometabolic changes of TI (i.e. glycolysis) were a feature of myelomonocytic cells in GCA lesions and were required for enhanced cytokine production. Conclusions: Myelomonocytic cells in GCA activate TI programs sustaining enhanced inflammatory activation with excess cytokine production.

Spinocerebellar ataxia type 28 (SCA28) is a neurodegenerative disease caused by mutations of the mitochondrial protease AFG3L2. The SCA28 mouse model, which is haploinsufficient for Afg3l2, exhibits a progressive decline in motor function and displays dark degeneration of Purkinje cells (PC-DCD) of mitochondrial origin. Here, we determined that mitochondria in cultured Afg3l2-deficient PCs ineffectively buffer evoked [Ca.sup.2+] peaks, resulting in enhanced cytoplasmic [Ca.sup.2+] concentrations, which subsequently triggers PC-DCD. This [Ca.sup.2+]-handling defect is the result of negative synergism between mitochondrial depolarization and altered organelle trafficking to PC dendrites in Afg3l2-mutant cells. In SCA28 mice, partial genetic silencing of the metabotropic glutamate receptor mGluR1 decreased [Ca.sup.2+] influx in PCs and reversed the ataxic phenotype. Moreover, administration of the β-lactam antibiotic ceftriaxone, which promotes synaptic glutamate clearance, thereby reducing [Ca.sup.2+] influx, improved ataxia-associated phenotypes in SCA28 mice when given either prior to or after symptom onset. Together, the results of this study indicate that ineffective mitochondrial [Ca.sup.2+] handling in PCs underlies SCA28 pathogenesis and suggest that strategies that lower glutamate stimulation of PCs should be further explored as a potential treatment for SCA28 patients., Introduction SCA28 is a form of dominant cerebellar ataxia characterized by unbalanced standing, gait incoordination, hyperreflexia of the lower limbs, nystagmus, and ophthalmoparesis (1). Functionalized silica-based nanocarriers (SiNPs) can be labeled for in vivo imaging applications and loaded with chemotherapy drugs, making possible the simultaneous noninvasive diagnosis and treatment (theranostic) for HER2-positive BC.Methods: Firstly, FITC-filled SiNPs, were engineered with two different amounts of Hc-TZ (trastuzumab half-chain) per single nanoparticle (1:2 and 1:8, SiNPs to Hc-TZ ratio), which was 99mTc-radiolabeled at histidine residues for ex vivo and in vivo biodistribution evaluations. Secondly, nanoparticles were loaded with DOX and their in vitro and ex vivo/in vivo delivery was assessed, in comparison with liposomal Doxorubicin (Caelyx).
