Human organic anion transporter 2 (hOAT2[SLC22A7]) is a Na+-independent multispecific organic solutes (SLC) transporter. To date, several findings regarding the function and substrates of hOAT2[SLC22A7] have been reported; there are still limited data concerning the genetic variants of the hOAT2[SLC22A7] gene in Japanese, specifically, in patients with non-viral liver carcinoma (LC). In the present study, we isolated genomic DNA from a normal portion of liver cancer and screened comprehensively 88 coding single nucleotide polymorphisms (cSNPs) chosen from a database of SNPs (dbSNPs). We found genotype and allele frequencies for 1 synonymous SNP [rs2270860 (1324C > T, Ser425Ser)] to be statistically significant when compared with the SNP database ( http://www.ncbi.nlm.nih.gov/snp/ ). Our present findings suggest that, at least partly, within the entire sequence of coding region of the hOAT2[SLC22A7] gene, rs2270860 (1324C > T, Ser425Ser) might be useful as a biomarker for susceptibility for non-viral LC in Japanese.

O aumento dos índices de obesidade e a ineficácia do tratamento clínico fazem com que a busca por tratamentos cirúrgicos deva ser considerada. Ademais, tanto anemias quanto deficiências nutricionais são consequências comuns da cirurgia bariátrica. O presente estudo tem por objetivo explanar os possíveis tratamentos que buscam minimizar as alterações nutricionais e de que forma o tema vem sendo abordado em diferentes análises. A busca de artigos foi através das bases de dados PubMed e Scientific Electronic Library Online (SciELO) com a utilização dos termos “anemia and gastrectomy” e “anemia and bariatric surgery” e seus correspondentes em português. Como critérios de inclusão foram considerados: a) artigos b) estar disponível em texto completo c) estudos disponíveis nos idiomas português, inglês ou espanhol d) pesquisa clínica e) terem sido publicados entre 2015 e 2020 f) artigos que contemplem anemias carenciais e consequências clinicamente significativas da cirurgia bariátrica. Foram excluídos os textos que não tinham relação com a temática e as duplicatas. Assim, dos 22 artigos apresentados pela busca nas bases de dados, 11 foram selecionados por afinidade à temática de busca e 11 artigos foram excluídos por não serem relacionados com o assunto. A partir desse processo, percebeu-se que as anemias e as deficiências nutricionais são uma consequência comum em pacientes pós-gastrectomia. Assim, a suplementação pré-operatória e o monitoramento nutricional pós-cirúrgico são intervenções para evitar as carências nutricionais. Importante destacar o acompanhamento de pacientes gastrectomizados por equipe multiprofissional para garantir uma melhor qualidade de vida ao paciente.

Human organic solute carrier protein 1 (hOSCP1) is a Na(+)-independent multispecific organic solute transporter. To date, several studies have revealed that gene mutations of the transporters are likely to be associated with some diseases; however, there are no data concerning the genetic polymorphism of the hOSCP1 gene in Japanese patients with non-viral liver carcinoma (LC). In the present study, we isolated genomic DNA from a normal portion of LC, and analyzed 41 single nucleotide polymorphisms (SNPs) chosen from a database of SNPs (dbSNPs). We found genotype frequencies for 2 non-synonymous SNPs [rs34409118 (Thr(131) → Ala) and rs1416840 (Ile(219) → Thr)] and 1 synonymous SNP [rs16822954 (Ser(193) → Ser)] to be statistically significant when compared with dbSNPs. No statistical significance was observed in rs2275477 (Gly(307) → Arg) in the hOSCP1 gene. With respect to the allele frequency, we also observed rs34409118 to be statistically significant. Interestingly, we found that non-viral LC patients do not carry heterozygous mutations in rs1416840 (A/G) and rs16822954 (A/G), suggesting that a non-carrier of heterozygous mutations in these two SNPs might be a biomarker for susceptibility for non-viral LC in Japanese. Further analyses of patients with hOSCP1 variants may elucidate the relationship between the hOSCP1 gene and susceptibility of non-viral LC in Japanese patients.
