Introdução: A fibrose cística (FC) é uma doença genética de transmissão autossômico-recessiva, cujas manifestações clínicas resultam da disfunção de uma proteína denominada proteína reguladora da condutância transmembrana na fibrose cística ou Cystic Fibrosis Transmembrane Condutance Regulator (CFTR). Ademais, ela é uma doença crônica, progressiva e altamente letal. Metodologia: Trata-se de um estudo exploratório de Revisão Narrativa da Literatura. Utilizou-se os Descritores em Ciências da Saúde (DeCS) de forma associada: “climatério”, “estratégias”, “sinais”, "sintomas”. Os critérios de inclusão foram: artigos disponibilizados na íntegra, disponíveis nos idiomas português, espanhol e inglês e publicados entre os anos de 2017 e 2022. Foram selecionados 7 estudos para esta revisão. Resultado e Discussão: Pelo fato de a FC atingir variados sistemas do corpo humano, ela exige um cuidado complexo e uma atenção contínua para promoção de uma condição clínica estável do paciente; para tanto, é imprescindível tanto envolvimento da equipe de enfermagem quanto da família, sob as mais diferentes maneiras. O contato dos profissionais da saúde com a FC torna-se bastante diversificado devido às mudanças no seu crescimento e desenvolvimento, e também está relacionado às limitações da condição crônica que acompanham as crianças em sua vida diária. Conclusão: observou-se que estabelecer uma parceria entre equipe de enfermagem-adentado-família, baseada em confiança mútua, é essencial para os que vivenciam a FC e as dificuldades que a doença traz no cotidiano. Destaca-se a importância que a equipe de saúde tem no processo saúde-doença crônica, e o papel da enfermagem é apoiar o paciente e a família deste e ter a sensibilidade para perceber que essas pessoas se deparam com o inesperado em suas vidas e necessitam receber apoio de todas as partes.

Due to its chemical properties, glyphosate [N-(phosphonomethyl)glycine] is one of the most commonly used agricultural herbicides globally. Due to risks associated with human exposure to glyphosate and its potential harmfulness, the need to develop specific, accurate, online, and sensitive methods is imperative. In accordance with this, the present review is focused on recent advances in developing nanomaterial-based sensors for glyphosate detection. Reported data from the literature concerning glyphosate detection in the different matrices using analytical methods (mostly chromatographic techniques) are presented; however, they are expensive and time-consuming. In this sense, nanosensors’ potential applications are explained to establish their advantages over traditional glyphosate detection methods. Zero-dimensional (0D), one-dimensional (1D), two-dimensional (2D), and three- dimensional (3D) materials are reviewed, from biomolecules to metallic compounds. Bionanomaterials have generated research interest due to their selectivity with respect to using enzymes, DNA, or antibodies. On the other hand, Quantum Dots also are becoming relevant for their vast surface area and good limit of detection values (in the range of pM). This review presents all the characteristics and potential applications of different nanomaterials for sensor development, bearing in mind the necessity of a glyphosate detection method with high sensitivity, selectivity, and portability.

In this work, we sought to obtain a more stable laccase with higher operational stability for the oxidation of phenols. During this reaction, phenoxy free radicals are produced that gradually inactivate the enzyme; the inactivation rate depends on the phenol chemical nature. In order to predict residues prone to oxidize within the active site, we simulated activated states of the catalytic region of a fungal laccase using QM-MM tools (Quantum Mechanics-Molecular Mechanics). After simulating the electron distribution in both the basal and activated state (plus or minus one electron) of several conformations of Coriolopsis gallica laccase, residues that could be susceptible to oxidation were identified, according to the values of spin density obtained from calculations. Three targets were selected (F357, F413, and F475) to be replaced by site-directed mutagenesis with less oxidizable residues such as leucine, alanine, and isoleucine. The resulting variants displayed a higher specific activity (from 1.5-to 4-fold) than the parental enzyme. Catalyst depletion during phenol oxidation was 2.5-fold lower for the variants, reflecting a higher operational stability.

In this work we communicate the heterologous expression of a laccase from Coriolopsis gallica in Pichia pastoris . This enzyme has been reported to efficiently degrade a variety of pollutants such as industrial dyes. The expression strategy included using a previously reported modified α-factor preproleader for enhanced secretion and p AOX1 , a methanol-responsive promoter. Methanol concentration, copper salts concentration and temperature were varied in order to enhance laccase expression in this heterologous system. A volumetric activity of 250 U/L was achieved after 12-day culture in Fernbach flasks. The protein was recovered from the supernatant and purified, obtaining a preparation with 90% electrophoretic purity. The catalytic constants of the recombinant enzyme are almost identical to the fungal enzyme, thus rendering this system a useful tool for protein engineering of laccase from C. gallica .

A spectrophotometric method for the determination of glyphosate based on the monitoring of a complex formation between bis 5-phenyldipyrrinate of nickel (II) and the herbicide was developed. The method showed a short response time (10 s), high selectivity (very low interference from other pesticides and salts), and high sensitivity (LOD 2.07 &times, 10&minus, 7 mol/L, LOQ 9.87 &times, 7 mol/L, and a Kd from 1.75 &times, 6 to 6.95 &times, 6 mol/L). The Job plot showed that complex formation occurs with a 1:1 stoichiometry. The method was successfully applied in potable, urban, groundwater, and residual-treated water samples, showing high precision (0.34&ndash, 2.9%) and accuracy (87.20&ndash, 119.04%). The structure of the complex was elucidated through theoretical studies demonstrating that the nickel in the bis 5-phenyldipyrrinate forms a distorted octahedral molecular geometry by expanding its coordination number through one bond with the nitrogen and another with the oxygen of the glyphosate&rsquo, carboxyl group, at distances between 1.89&ndash, 2.08 &Aring

The management of protected areas in karstic regions is a challenge because flooded cave systems form there and provide underground hydrological conducts that may link different zones. As a consequence, affectations to the protected areas can possibly occur as a consequence of human activities in remote areas and may therefore pass undetected. Thus, the monitoring of possible contaminants in these regions is becoming imperative. In this work, we analyze the concentration of essential (iron) and non-essential metals (cadmium and chromium) in the seagrass Thalassia testudinum that grows in Yalahau Lagoon, located in a near-to-pristine protected area of the Yucatan Peninsula, close to the rapidly developing touristic belt of the Mexican Caribbean. Salinity and silicate patterns show that Yalahau is an evaporation lagoon, where groundwater discharge is important. High iron (>400 μg/g), cadmium (>4 μg/g), and chromium (≈1 μg/g) concentrations were found in the area of highest groundwater input of the lagoon. High levels (5.1 μg/g) were also found near the town dump. In the rest of the sampling sites, metal concentrations remained near to background levels as estimated from other works. Temporal changes of concentrations in the seagrass tissues show also a local input and an input from the groundwater that could provoke an environmental problem in the Yalahau Lagoon in the near future.

In this work, we report the synthesis and characterization of a novel series of first and second generation Fréchet type dendrons bearing amino-nitro substituted azobenzene units and tetra(ethylene glycol) spacers. These compounds were fully characterized by FTIR, 1H and 13C-NMR spectroscopies, and their molecular weights were determined by MALDI-TOF-MS. The thermal properties of the obtained dendrons were studied by TGA and DSC and their optical properties by absorption spectroscopy in solution and cast film. Molecular calculations were performed in order to determine the optimized geometries of these molecules in different environments. Besides, Langmuir and Langmuir Blodgett films were prepared with the first generation dendrons that were shown to be amphiphilic. Finally, some of the dendrons showed a liquid crystalline behaviour, which was studied by light polarized microscopy as a function of the temperature in order to determine the transition temperatures and the structure of the mesophase.

The main goal of this investigation is to study the possible mechanisms of Casiopeinas® as anticancer agents. Electrodonating (χ-) and electroaccepting (χ+) electronegativity were calculated applying Density Functional Theory. Two different anticancer mechanisms of Casiopeínas® are proposed. There might be antiradical molecules preventing the formation of cancer cells or these molecules could reduce the amount of GSH and as a result over-produce free radicals, increasing the oxidative stress which in turn kills the cancer cells.
