1. Modified Coulomb potential and the S-state wavefunction of heavy quarkonia

Abstract: The combination of relativistic kinematics together with Coulomb or Yukawa potentials is a common place in atomic, nuclear and meson mass phenomenology. In meson spectroscopy, linear and Coulomb-like potentials are the most commonly used potentials and decays rates are also calculated using the value of the wavefunction at the origin. But, it has been known for sometime that the use of relativistic kinematics together with Coulomb or Coulomb-like potentials produces wavefunction singularity at the origin for S-wave \cite{DuranFriarPolyzou}. Therefore, there is a need to address this problem somehow. In this letter, we show how to overcome this problem without destroying the phenomenological success of the use of relativistic kinematics in meson mass spectroscopy. Abstract: Generative artificial intelligence (GenAI) and large language models (LLMs) have simultaneously opened new avenues for enhancing human learning and increased the prevalence of poor-quality information in student response - termed Botpoop. This study introduces Professor Leodar, a custom-built, Singlish-speaking Retrieval Augmented Generation (RAG) chatbot designed to enhance educational while reducing Botpoop. Deployed at Nanyang Technological University, Singapore, Professor Leodar offers a glimpse into the future of AI-assisted learning, offering personalized guidance, 24/7 availability, and contextually relevant information. Through a mixed-methods approach, we examine the impact of Professor Leodar on learning, engagement, and exam preparedness, with 97.1% of participants reporting positive experiences. These findings help define possible roles of AI in education and highlight the potential of custom GenAI chatbots. Abstract: Recent advances in machine learning techniques have made it possible to use high-throughput screening to identify novel materials with specific properties. However, the large number of potential candidates produced by these techniques can make it difficult to select the most promising ones. In this study, we develop the oxidation state probability (OSP) method which evaluates ternary compounds based on the probability (the OSP metric) of each element to adopt the required oxidation states for fulfilling charge neutrality. We compare this model with Roost and the Fourier-transformed crystal properties (FTCP)-based synthesizability score. Among the top 1000 systems with the most database entries in Materials Project (MP), more than 500 systems exhibit an attested compound among the top 3 compositions when ranked by the OSP metric. We find that the OSP method shows promising results for certain classes of ternary systems, especially those containing nonmetals, s-block, or transition metals. When applied to the Cu-In-Te ternary system, an interesting system for thermoelectric applications, the OSP method predicted the synthesizability of CuIn$_3$Te$_5$ without prior knowledge, and we have successfully synthesized CuIn$_3$Te$_5$ in experiment. Our method has the potential to accelerate the discovery of novel compounds by providing a guide for experimentalists to easily select the most synthesizable candidates from an arbitrarily large set of possible chemical compositions., Comment: 11 pages, 3 figures

Abstract: Human immunodeficiency virus-1 (HIV-1) infects the central nervous system (CNS) and causes HIV-associated neurocognitive disorders (HAND) in about half of the population living with the virus despite combination anti-retroviral therapy (cART). HIV-1 activates the innate immune system, including the production of type 1 interferons (IFNs) α and β. Transgenic mice expressing HIV-1 envelope glycoprotein gp120 (HIVgp120tg) in the CNS develop memory impairment and share key neuropathological features and differential CNS gene expression with HIV patients, including the induction of IFN-stimulated genes (ISG). Here we show that knocking out IFNβ (IFNβKO) in HIVgp120tg and non-tg control mice impairs recognition and spatial memory, but does not affect anxiety-like behavior, locomotion, or vision. The neuropathology of HIVgp120tg mice is only moderately affected by the KO of IFNβ but in a sex-dependent fashion. Notably, in cerebral cortex of IFNβKO animals presynaptic terminals are reduced in males while neuronal dendrites are reduced in females. The IFNβKO results in the hippocampal CA1 region of both male and female HIVgp120tg mice in an ameliorated loss of neuronal presynaptic terminals but no protection of neuronal dendrites. Only female IFNβ-deficient HIVgp120tg mice display diminished microglial activation in cortex and hippocampus and increased astrocytosis in hippocampus compared to their IFNβ-expressing counterparts. RNA expression for some immune genes and ISGs is also affected in a sex-dependent way. The IFNβKO abrogates or diminishes the induction of MX1, DDX58, IRF7 and IRF9 in HIVgp120tg brains of both sexes. Expression analysis of neurotransmission related genes reveals an influence of IFNβ on multiple components with more pronounced changes in IFNβKO females. In contrast, the effects of IFNβKO on MAPK activities are independent of sex with pronounced reduction of active ERK1/2 but also of active p38 in the HIVgp120tg brain. In summary, our findings show that the absence of IFNβ impairs memory dependent behavior and modulates neuropathology in HIVgp120tg brains, indicating that its absence may facilitate development of HAND. Moreover, our data suggests that endogenous IFNβ plays a vital role in maintaining neuronal homeostasis and memory function.

Abstract: Macrophages (MΦ) infected with human immunodeficiency virus (HIV)-1 or activated by its envelope protein gp120 exert neurotoxicity. We found previously that signaling via p38 mitogen-activated protein kinase (p38 MAPK) is essential to the neurotoxicity of HIVgp120-stimulated MΦ. However, the associated downstream pathways remained elusive. Here we show that cysteinyl-leukotrienes (CysLT) released by HIV-infected or HIVgp120 stimulated MΦ downstream of p38 MAPK critically contribute to neurotoxicity. SiRNA-mediated or pharmacological inhibition of p38 MAPK deprives MΦ of CysLT synthase (LTC4S) and, pharmacological inhibition of the cysteinyl-leukotriene receptor 1 (CYSLTR1) protects cerebrocortical neurons against toxicity of both gp120-stimulated and HIV-infected MΦ. Components of the CysLT pathway are differentially regulated in brains of HIV-infected individuals and a transgenic mouse model of NeuroHIV (HIVgp120tg). Moreover, genetic ablation of LTC4S or CysLTR1 prevents neuronal damage and impairment of spatial memory in HIVgp120tg mice. Altogether, our findings suggest a novel critical role for cysteinyl-leukotrienes in HIV-associated brain injury.

9. Comparing observed occurrence of mistreatment during childbirth with women's self-report: a validation study in Ghana, Guinea and Nigeria.

Abstract: BackgroundThere has been substantial progress in developing approaches to measure mistreatment of women during childbirth. However, less is known about the differences in measurement approaches. In this study, we compare measures of mistreatment obtained from the same women using labour observations and community-based surveys in Ghana, Guinea and Nigeria.MethodsExperiences of mistreatment during childbirth are person-centred quality measures. As such, we assessed individual-level and population-level accuracy of labour observation relative to women's self-report for different types of mistreatment. We calculated sensitivity, specificity, percent agreement and population-level inflation factor (IF), assessing prevalence of mistreatment in labour observation divided by 'true' prevalence in women's self-report. We report the IF degree of bias as: low (0.75Results1536 women across Ghana (n=779), Guinea (n=425) and Nigeria (n=332) were included. Most mistreatment items demonstrated better specificity than sensitivity: observation of any physical abuse (44% sensitive, 89% specific), any verbal abuse (61% sensitive, 73% specific) and presence of a labour companion (19% sensitive, 93% specific). Items for stigma (IF 0.16), pain relief requested (IF 0.38), companion present (IF 0.32) and lack of easy access to fluids (IF 0.46) showed high risk of bias, meaning labour observations would substantially underestimate true prevalence. Other items showed low or moderate bias.ConclusionUsing self-report as the reference standard, labour observations demonstrated moderate-to-high specificity (accurately identifying lack of mistreatment) but low-to-moderate sensitivity (accurately identifying presence of mistreatment) among women. For overall prevalence, either women's self-report or observations can be used with low-moderate bias for most mistreatment items. However, given the dynamicity, complexity, and limitations in 'objectivity', some experiences of mistreatment (stigma, pain relief, labour companionship, easy access to fluids) require measurement via women's self-report. More work is needed to understand how subjectivity influences how well a measure represents individual's experiences.

11. Factors affecting infant feeding choices with a focus on barriers to exclusive breastfeeding in Western Jamaica: a qualitative study

Abstract: Abstract Background Despite the many benefits of exclusive breastfeeding to infants and mothers, only 33% of Jamaican infants are exclusively breastfed up to the recommend six months. This study was conducted to identify factors affecting mothers’ feeding choices focusing on barriers to exclusive breastfeeding of infants six weeks to less than six months old. Methods A qualitative study consisting of four focus group discussion sessions was conducted among 22 mothers attending postnatal clinics in western Jamaica from May to August 2016. The transcripts were coded by three independent coders and content analysis conducted to generate themes. Results Four themes were identified namely, perceived advantages of breastfeeding centered mainly on the benefits of breastfeeding for the infant and mother, perceived barriers of breastfeeding highlighting physical pain and fatigue, supplementing culturally acceptable complementary foods and herbal remedies, and cultural norms including perception of how breastfeeding affects a woman’s body, societal sources of breastfeeding information, satiation of infants, and family and other support. Mothers overwhelmingly agreed that breastfeeding was inexpensive, allowed them to bond with their infants and was good for the overall health and intellectual development of the infants. They identified painful nipples, engorged breasts, lack of sleep, physical exhaustion and pressure to return to work as barriers to breastfeeding. Mothers named a number of complementary foods, such as pumpkin, carrots, potato, banana, and chocho (Chayote), that were culturally accepted for feeding infants in Jamaica and discussed herbs that were considered to aid in infants’ nutrition and overall health. Other cultural factors that were noted to influence exclusive breastfeeding were mothers feeling that breastfeeding would help their bodies, especially their bellies, go back to their pre-maternity figure, sources of breastfeeding information in the society including the internet, belief that breast milk alone does not satisfy babies, and family and other support. Conclusion Mothers in this study identified unique challenges to exclusive breastfeeding that if addressed, would help to increase exclusive breastfeeding so that the World Health Organization’s exclusive breastfeeding recommendations can be achieved.

Abstract: Abstract Background SARS-CoV-2 remains rapidly evolving, and many biologically important genomic substitutions/indels have characterised novel SARS-CoV-2 lineages, which have emerged during successive global waves of the pandemic. Worldwide genomic sequencing has been able to monitor these waves, track transmission clusters, and examine viral evolution in real time to help inform healthcare policy. One school of thought is that an apparent greater than average divergence in an emerging lineage from contemporary variants may require persistent infection, for example in an immunocompromised host. Due to the nature of the COVID-19 pandemic and sampling, there were few studies that examined the evolutionary trajectory of SARS-CoV-2 in healthy individuals. Methods We investigated viral evolutionary trends and participant symptomatology within a cluster of 16 SARS-CoV-2 infected, immunocompetent individuals with no co-morbidities in a closed transmission chain. Longitudinal nasopharyngeal swab sampling allowed characterisation of SARS-CoV-2 intra-host variation over time at both the dominant and minor genomic variant levels through Nimagen-Illumina sequencing. Results A change in viral lineage assignment was observed in individual infections; however, there was only one indel and no evidence of recombination over the period of an acute infection. Minor and dominant genomic modifications varied between participants, with some minor genomic modifications increasing in abundance to become the dominant viral sequence during infection. Conclusions Data from this cohort of SARS-CoV-2-infected participants demonstrated that long-term persistent infection in an immunocompromised host was not necessarily a prerequisite for generating a greater than average frequency of amino acid substitutions. Amino acid substitutions at both the dominant and minor genomic sequence level were observed in immunocompetent individuals during infection showing that viral lineage changes can occur generating viral diversity.

Abstract: Myanmar is located within an important geographic corridor of prehistoric demographic and technological exchange, yet relatively few archaeological sites have been securely dated. Here, the authors present a new radiocarbon chronology for Halin, a UNESCO-listed complex in the north-central Sagaing Division of Myanmar, which contributes to the generation of nuanced regional chronologies and to improving the temporal resolution of Southeast Asia more generally. Discussion of 94 radiocarbon determinates, together with site stratigraphy and pottery traditions, provides a chronological sequence from the early third millennium BC to the early second millennium AD. Corroboration of the beginning of this sequence would place Halin as the oldest currently dated Neolithic site in Mainland Southeast Asia and would provide support for the two-layer model of Neolithic migration. Abstract: Surface mobility control is a fundamental challenge in robotics, and its applications range from autonomous vehicles to industrial automation. The main aim of the paper is to analyze the PID controller parameters for the speed control of a surface mobility platform mobile robot. MATLAB simulation toolbox and mathematical model of the DC motor are used to find the optimal PID controller parameters to complete the DC motor speed control system. For the SMP motion control system, the mobile robot kinematic model is supported as a differential drive system. In this paper, the PID controller parameters for the DC motor control system are analyzed with simulation results using various KP, KI, KD gains. The experimental results of the SMP mobile robot with optimal PID controller parameters are analyzed by driving on the rough, slope and even terrains. According to simulation results and experimental tests, the proposed system effectively controls the speed of the SMP mobile robot, demonstrating satisfactory performance in settling time, rise time, and system overshoot. Among PI and PID controllers, the designed PID controller can quickly control to reach the desired DC motor speed. Using the optimal PID coefficient parameters, all motors reached their desired speed in 0.12 seconds during simulations. In the experimental tests on various terrain (even, slope and rough), all motors achieved their desired speed simultaneously within 25 seconds.

Abstract: Abstract Background Intrauterine inflammation is considered a major cause of brain injury in preterm infants, leading to long-term neurodevelopmental deficits. A potential contributor to this brain injury is dysregulation of neurovascular coupling. We have shown that intrauterine inflammation induced by intra-amniotic lipopolysaccharide (LPS) in preterm lambs, and postnatal dopamine administration, disrupts neurovascular coupling and the functional cerebral haemodynamic responses, potentially leading to impaired brain development. In this study, we aimed to characterise the structural changes of the neurovascular unit following intrauterine LPS exposure and postnatal dopamine administration in the brain of preterm lambs using cellular and molecular analyses. Methods At 119–120 days of gestation (term = 147 days), LPS was administered into the amniotic sac in pregnant ewes. At 126-7 days of gestation, the LPS-exposed lambs were delivered, ventilated and given either a continuous intravenous infusion of dopamine at 10 µg/kg/min or isovolumetric vehicle solution for 90 min (LPS, n = 6; LPSDA, n = 6). Control preterm lambs not exposed to LPS were also administered vehicle or dopamine (CTL, n = 9; CTLDA, n = 7). Post-mortem brain tissue was collected 3–4 h after birth for immunohistochemistry and RT-qPCR analysis of components of the neurovascular unit. Results LPS exposure increased vascular leakage in the presence of increased vascular density and remodelling with increased astrocyte “end feet” vessel coverage, together with downregulated mRNA levels of the tight junction proteins Claudin-1 and Occludin. Dopamine administration decreased vessel density and size, decreased endothelial glucose transporter, reduced neuronal dendritic coverage, increased cell proliferation within vessel walls, and increased pericyte vascular coverage particularly within the cortical and deep grey matter. Dopamine also downregulated VEGFA and Occludin tight junction mRNA, and upregulated dopamine receptor DRD1 and oxidative protein (NOX1, SOD3) mRNA levels. Dopamine administration following LPS exposure did not exacerbate any effects induced by LPS. Conclusion LPS exposure and dopamine administration independently alters the neurovascular unit in the preterm brain. Alterations to the neurovascular unit may predispose the developing brain to further injury.

Abstract: Abstract Background To fight the current coronavirus disease (COVID-19) pandemic, many countries have implemented various mitigation measures to contain the spread of the disease. By engaging with health service providers, the community’s participation in adherence to preventive measures is certainly required in the implementation of COVID-19 mitigation strategies. Therefore, this study aimed to assess the level of adherence to COVID-19 preventive measures and its associated factors among the residents, Yangon Region, Myanmar. Methods A community-based cross-sectional study was carried out among 636 residents in Yangon Region, Myanmar, from October to December 2021. A multistage non-probability sampling method, purposively selected for three townships in Yangon Region and convenience sampling for 212 participants from each township, was applied and the data were collected by face-to-face interviews using structured and pretested questionnaires. Data were entered, coded, and analyzed using IBM SPSS version 25.0. Simple and multiple logistic regression analysis were performed to identify the significant variables of adherence to COVID-19 preventive measures. Results As a level of adherence to COVID-19 preventive measures, the proportion of residents who had good adherence was 39.3% (95% CI 35.5–43.2%), moderate adherence was 37.6% (95% CI 33.8–41.5%), and poor adherence was 23.1% (95% CI 19.9–26.6%). The age group of 31–40 years (AOR: 3.13, 95% CI 1.62–6.05), 30 years and younger (AOR: 3.22, 95% CI 1.75–5.92), Burmese ethnicity (AOR: 2.52, 95% CI 1.44–4.39), own business (AOR: 3.19, 95% CI 1.15–8.87), high school education level and below (AOR: 1.64, 95% CI 1.02–2.69), less than 280.90 USD of monthly family income (AOR: 1.51, 95% CI 1.01–2.29), low knowledge about COVID-19 (AOR: 1.90, 95% CI 1.26–2.88) were significantly associated with poor adherence to COVID-19 preventive measures. Conclusions In this study, nearly one-fourth of the residents were experiencing poor adherence to COVID-19 preventive measures. Therefore, building up the risk communication through the community using widely used mainstream media, the continuation of disease surveillance and announcement of updated information or advice for the public to increase awareness towards COVID-19, and enforcement to follow the recommended directions and regulations of health institutions are vital to consider for improving the adherence to preventive measures against COVID-19 among the residents.

Abstract: Abstract Background Excessive or inappropriate use of social media has been linked to disruptions in regular work, well-being, mental health, and overall reduction of quality of life. However, a limited number of studies documenting the impact of social media on health-related quality of life (HRQoL) are available globally. Aim This study aimed to explore the perceived social media needs and their impact on the quality of life among the adult population of various selected countries. Methodology A cross-sectional, quantitative design and analytical study utilized an online survey disseminated from November to December 2021. Results A total of 6689 respondents from ten countries participated in the study. The largest number of respondents was from Malaysia (23.9%), followed by Bangladesh (15.5%), Georgia (14.8%), and Turkey (12.2%). The prevalence of social media users was over 90% in Austria, Georgia, Myanmar, Nigeria, and the Philippines. The majority of social media users were from the 18–24 age group. Multiple regression analysis showed that higher education level was positively correlated with all four domains of WHOQoL. In addition, the psychological health domain of quality of life was positively associated in all countries. Predictors among Social Media Needs, Affective Needs (β = -0.07), and Social Integrative Needs (β = 0.09) were significantly associated with psychological health. Conclusion The study illuminates the positive correlation between higher education levels and improved life quality among social media users, highlighting an opportunity for policymakers to craft education-focused initiatives that enhance well-being. The findings call for strategic interventions to safeguard the mental health of the global social media populace, particularly those at educational and health disadvantages.

Abstract: IntroductionUnlike white adipose tissue depots, bone marrow adipose tissue (BMAT) expands during caloric restriction (CR). Although mechanisms for BMAT expansion remain unclear, prior research suggested an intermediary role for increased circulating glucocorticoids. MethodsIn this study, we utilized a recently described mouse model (BMAd-Cre) to exclusively target bone marrow adipocytes (BMAds) for elimination of the glucocorticoid receptor (GR) (i.e. Nr3c1) whilst maintaining GR expression in other adipose depots. ResultsMice lacking GR in BMAds (BMAd-Nr3c1-/-) and control mice (BMAd-Nr3c1+/+) were fed ad libitum or placed on a 30% CR diet for six weeks. On a normal chow diet, tibiae of female BMAd-Nr3c1-/- mice had slightly elevated proximal trabecular metaphyseal bone volume fraction and thickness. Both control and BMAd-Nr3c1-/- mice had increased circulating glucocorticoids and elevated numbers of BMAds in the proximal tibia following CR. However, no significant differences in trabecular and cortical bone were observed, and quantification with osmium tetroxide and μCT revealed no difference in BMAT accumulation between control or BMAd-Nr3c1-/- mice. Differences in BMAd size were not observed between BMAd-Nr3c1-/- and control mice. Interestingly, BMAd-Nr3c1-/- mice had decreased circulating white blood cell counts 4 h into the light cycle.DiscussionIn conclusion, our data suggest that eliminating GR from BMAd has minor effects on bone and hematopoiesis, and does not impair BMAT accumulation during CR.

Abstract: This paper presents a novel and robust hybrid controller that combines the backstepping controller with the nonsingular fast terminal sliding mode controller (NFTSMC) for DC-DC boost converters (DDBCs) operating alongside constant power loads (CPLs). The primary aim of the controller is to enhance the stability of the DC-bus voltage while consistently delivering power to the CPL. To achieve this objective, the exact feedback linearization approach is applied to transform the nonlinear dynamical model of the DDBC into Brunovsky’s canonical forms. The incorporation of matched uncertainties into the dynamical model illustrates the robustness of the proposed control approach. The hybrid controller is formulated based on this model, leveraging Lyapunov stability theory to ensure large signal stability under the designated control law. Significantly, the NFTSMC effectively addresses the singularity problem associated with sliding mode controllers (SMCs). The effectiveness and resilience of the proposed controller are validated under variations in input voltage, reference voltage, and power demand. Comparative analysis is conducted by contrasting simulation results with those obtained using conventional SMC (CSMC) and a typical PI controller (PIC). The results unequivocally highlight the superiority of the proposed controller in terms of ensuring system stability and providing a rapid response across various operating modes. Furthermore, to authenticate the practical applicability of the proposed controller, processor-in-loop (PIL) validation is employed, confirming its effectiveness in real-world scenarios.

Abstract: Objective: Exposure of adipocytes to ‘cool’ temperatures often found in the periphery of the body induces expression of Stearoyl-CoA Desaturase-1 (Scd1), an enzyme that converts saturated fatty acids to monounsaturated fatty acids. The goal of this study is to further investigate the roles of Scd in adipocytes. Method: In this study, we employed Scd1 knockout cells and mouse models, along with pharmacological Scd1 inhibition to dissect the enzyme's function in adipocyte physiology. Results: Our study reveals that production of monounsaturated lipids by Scd1 is necessary for fusion of autophagosomes to lysosomes and that with a Scd1-deficiency, autophagosomes accumulate. In addition, Scd1-deficiency impairs lysosomal and autolysosomal acidification resulting in vacuole accumulation and eventual cell death. Blocking autophagosome formation or supplementation with monounsaturated fatty acids maintains vitality of Scd1-deficient adipocytes. Conclusion: This study demonstrates the indispensable role of Scd1 in adipocyte survival, with its inhibition in vivo triggering autophagy-dependent cell death and its depletion in vivo leading to the loss of bone marrow adipocytes.

Abstract: Radiographic imaging is the current standard for monitoring progression of tumor-burden and therapeutic resistance in patients with metastatic melanoma. Plasma circulating tumor DNA (ctDNA) has shown promise as a survelience tool, but longitudinal data on the dynamics between plasma ctDNA concentrations and radiographic imaging is lacking. We evaluated the relationship between longitudinal radiographic measures of tumor burden and ctDNA concentrations in plasma on 30 patients with metastatic melanoma on systemic treatment. In 9 patients with no radiographic evidence of disease over a total of 15 time points, ctDNA concentrations were undetectable. In 21 patients with radiographic tumor burden, ctDNA was detected in 81 % of 58 time points. Plasma ctDNA concentrations demonstrated a modest positive correlation with total tumor burden (TTB) measurements (R2= 0.49, p < 0.001), with the greatest degree of correlation observed under conditions of progressive disease (PD) (R2 = 0.91, p = 0.032). Plasma ctDNA concentrations were significantly greater at times of RECIST v1.1 progression (PD; 22.1 % ± 5.7 %) when compared to samples collected during stable disease (SD; 4.99 % ± 3.0 %) (p = 0.012); this difference was independent of total tumor burden (p = 0.997). 44. Consolidation of Calcium Carbonate Using Polyacrylamides with Different Chemistries

Abstract: In this work, the consolidation of calcium carbonate (CaCO3) by polyacrylamide (PAM) of different molecular weights, charge densities, and functional groups was investigated via oscillatory rheology and unconfined compressive strength (UCS) analysis. Oscillatory rheology showed that the storage modulus G′ was approximately 10 times higher than the loss modulus G″, indicating a highly elastic CaCO3 sample upon consolidation via PAM. Both oscillatory rheology and UCS analysis exhibited similar trends, wherein the mechanical values (G′, G″, and UCS) first increased with increasing polymer dosage, until they reached a peak value (typically at 3 mgpol/gCaCO3), followed by a decrease in the mechanical values. This indicates that there is an optimum polymer dosage for the different PAM-CaCO3 colloidal systems, and that exceeding this value induces the re-stabilisation of the colloidal system, leading to a decreased degree of consolidation. Regarding the effect of the PAM molecular weight, the peak G′ and UCS values of CaCO3 consolidated by hydrolysed PAM (HPAM) of different molecular weights are very similar. This is likely due to the contour length of the HPAMs being either almost the same or longer than the average distance between two CaCO3 particles. The effect of the PAM charge density revealed that the peak G′ and UCS values decreased as the charge density of the PAM increased, while the optimum PAM dosage increased with decreasing PAM charge density. The higher likelihood of lower-charge PAM bridging between the particles contributes to higher elastic energy and mechanical strength. Finally, regarding the PAM functional group, CaCO3 consolidated by sulfonated polyacrylamide (SPAM) typically offers lower mechanical strength than that consolidated with HPAM. The bulky sulfonate side groups of SPAM interfere with the surface packing, reducing the number of polymers able to adsorb onto the surface and, eventually, reducing the degree of consolidation of CaCO3. The zeta potential of the PAM-CaCO3 samples became more negative with increasing PAM concentration due to the saturation of the particle surface. Good agreement between oscillatory rheology and UCS analysis could accelerate PAM screening for optimum CaCO3 consolidation.

45. Arresting calcium-regulated sperm metabolic dynamics enables prolonged fertility in poultry liquid semen storage

Abstract: Abstract The preservation of liquid semen is pivotal for both industrial livestock production and genetic management/conservation of species with sperm that are not highly cryo-tolerant. Nevertheless, with regard to poultry semen, even brief in vitro storage periods can lead to a notable decline in fertility, despite the in vivo capacity to maintain fertility for several weeks when within the hen’s sperm storage tubules. For fertility in sperm, intracellular calcium ions ([Ca2+]i) play a key role in signaling towards modifying energy metabolism. While reducing [Ca2+]i has been found to enhance the preservation of sperm fertility in some mammals, the connection between semen fertility and calcium availability in avian sperm has received limited attention. In this study, we demonstrate that the use of extracellular and intracellular calcium chelators in liquid semen extenders, specifically EGTA and EGTA-AM, has distinct effects on prolonging the fertility of chicken sperm. These results were validated through in vivo fertility tests. Mechanistically, the effects observed were linked to coordination of mitochondrial metabolism and ATP catabolism. Despite both calcium chelators inducing hypoxia, they differentially regulated mitochondrial respiration and ATP accumulation. This regulation was closely linked to a bimodal control of dynein ATPase activity; a direct initial activation with reduction in [Ca2+]i, and subsequent suppression by cytoplasmic acidification caused by lactic acid. These findings not only contribute to advancing poultry liquid semen preservation techniques, but also elucidates biologically relevant mechanisms that may underlie storage within the female reproductive tract in birds.

Abstract: Abstract Contaminated food with antibiotic-resistant Enterococcus spp. could be the vehicle for transmitting Enterococcus to humans and accordingly cause a public health problem. The accumulation of biogenic amines produced by Enterococcus faecalis (E. faecalis) in food may have cytological effects. Bacteriophages (phage in short) are natural antimicrobial agents and can be used alone or in combination with other food preservatives to reduce food microbial contaminants. The aim of this study was to isolate a novel phage against E. faecalis and determine its host range to evaluate its potential application. Bacteriophage, vB_EfKS5, with a broad host range, was isolated to control the growth of E. faecalis. The vB_EfKS5 genome is 59,246 bp in length and has a GC content of 39.7%. The computational analysis of phage vB_EfKS5 genome confirmed that it does not contain any lysogenic, toxic, or virulent genes. Phage vB_EfKS5 exhibited lytic activity against most E. faecalis isolates with different multiplicities of infections and it infected 75.5% (22/29) of E. faecalis isolates and 42.3% (3/7) of E. faecium isolates. It was also able to destroy the biofilm formed by E. faecalis with different MOIs. Phage vB_EfKS5 alone or in combination with nisin could control the growth of E. faecalis in broth and milk. Based on its high productivity, stability, short latent period, and large burst size, phage vB_EfKS5 has a high potential for applications both in food and medical applications.

47. Probabilistic seismic hazard assessments for Myanmar and its metropolitan areas

Abstract: Abstract Although Myanmar is an earthquake-prone country, there has not been proposed an official national seismic hazard map. Thus, this study conducted a probabilistic seismic hazard assessment for Myanmar and some of its metropolitan areas. Performing this assessment required a set of databases that incorporates both earthquake catalogs and fault parameters. We obtained seismic parameters from the International Seismological Centre, and the fault database includes fault parameters from paper reviews and the database. Based on seismic activities, we considered three categories of seismogenic sources—active fault source, shallow area source, and subduction zone source. We evaluated seismic activity of each source based on the earthquake catalogs and fault parameters. Evaluating the ground-shaking behaviors for Myanmar requires evaluation of ground-shaking attenuation; therefore, we validated existing ground motion prediction equations (GMPEs) by comparing instrumental observations and felt intensities for recent earthquakes. We then incorporated the best fitting GMPEs into our seismic hazard assessments. By incorporating the V s 30 (the average shear velocity down to 30 m depth) map from an analysis of topographic slope, we utilized site effect and assessed national probabilistic seismic hazards for Myanmar. The assessment shows highest seismic hazard levels near those faults with high slip rates, including the Sagaing Fault and along the Western Coast of Myanmar. We also assessed seismic hazard for some metropolitan cities, including Bagan, Bago, Mandalay, Sagaing, Taungoo and Yangon, in the forms of hazard curves and disaggregation by implementing detailed V s 30 maps from micro-tremor surveys. The city-scale assessments show higher hazards for sites close to an active fault or/and with a low V s 30, demonstrating the importance of investigating site conditions. The outcomes of this study will be beneficial to urban planning on a city scale and building code legislation on a national scale.

Abstract: Bacillus cereus sensu stricto is a foodborne pathogen that causes food poisoning. Their spore and biofilm-forming abilities persist in various environments and foods. This study investigated the prevalence, virulence, antibiotic resistance, and genetic diversity of B. cereus s. s. strains isolated from various food samples. Of 179 samples, 22.34% were positive for B. cereus s. s., with significantly high detection rates in milk products and raw chicken meat. Forty strains were isolated from positive samples. Matrix-assisted laser desorption ionization/time of flight mass spectrometry analysis revealed nine distinct clusters and multi-locus sequence typing revealed 34 sequence types including 23 novel sequences, demonstrating high genetic diversity among the isolates. PCR analysis revealed that all the strains contained at least one toxin gene, but none contained the cytK gene. Antibiotic resistance tests revealed that all isolates were classified as multidrug-resistant, with high resistance levels, particularly to β-lactam antibiotics and vancomycin, but were susceptible to gentamicin. All isolates showed variations in biofilm formation. This study highlights the significant public health risk due to B. cereus s. s. and underscores the need for stringent monitoring and control measures in food production to manage antimicrobial resistance and ensure food safety.

49. Agro Active Potential of Bacillus subtilis PE7 against Didymella bryoniae (Auersw.), the Causal Agent of Gummy Stem Blight of Cucumis melo

Abstract: Microbial agents such as the Bacillus species are recognized for their role as biocontrol agents against various phytopathogens through the production of diverse bioactive compounds. This study evaluates the effectiveness of Bacillus subtilis PE7 in inhibiting the growth of Didymella bryoniae, the pathogen responsible for gummy stem blight (GSB) in cucurbits. Dual culture assays demonstrate significant antifungal activity of strain PE7 against D. bryoniae. Volatile organic compounds (VOCs) produced by strain PE7 effectively impede mycelial formation in D. bryoniae, resulting in a high inhibition rate. Light microscopy revealed that D. bryoniae hyphae exposed to VOCs exhibited abnormal morphology, including swelling and excessive branching. Supplementing a potato dextrose agar (PDA) medium with a 30% B. subtilis PE7 culture filtrate significantly decreased mycelial growth. Moreover, combining a 30% culture filtrate with half the recommended concentration of a chemical fungicide yielded a more potent antifungal effect than using the full fungicide concentration alone, inducing dense mycelial formation and irregular hyphal morphology in D. bryoniae. Strain PE7 was highly resilient and was able to survive in fungicide solutions. Additionally, B. subtilis PE7 enhanced the nutrient content, growth, and development of melon plants while mitigating the severity of GSB compared to fungicide and fertilizer treatments. These findings highlight B. subtilis PE7 as a promising biocontrol candidate for integrated disease management in crop production.

Abstract: BackgroundMyanmar is one of the six countries in the Greater Mekong Subregion (GMS) of Southeast Asia. Malaria vectors comprise many Anopheles species, which vary in abundance and importance in malaria transmission among different geographical locations in the GMS. Information about the species composition, abundance, and insecticide resistance status of vectorial systems in Myanmar is scarce, hindering our efforts to effectively control malaria vectors in this region.MethodsDuring October and November 2019, larvae and adult females of Anopheles mosquitoes were collected in three sentinel villages of Banmauk township in northern Myanmar. Adult female mosquitoes collected by cow-baited tent collection (CBTC) and adults reared from field-collected larvae (RFCL) were used to determine mortality rates and knockdown resistance (kdr) against deltamethrin using the standard WHO susceptibility test. Molecular species identification was performed by multiplex PCR and ITS2 PCR, followed by DNA sequencing. The kdr mutation at position 1014 of the voltage-gated sodium channel gene was genotyped by DNA sequencing for all Anopheles species tested.ResultsA total of 1596 Anopheles mosquitoes from seven morphologically identified species groups were bioassayed. Confirmed resistance to deltamethrin was detected in the populations of An. barbirostris (s.l.), An. hyrcanus (s.l.), and An. vagus, while possible resistance was detected in An. annularis (s.l.), An. minimus, and An. tessellatus. Anopheles kochi was found susceptible to deltamethrin. Compared to adults collected by CBTC, female adults from RFCL had significantly lower mortality rates in the four species complexes. A total of 1638 individuals from 22 Anopheles species were molecularly identified, with the four most common species being An. dissidens (20.5%) of the Barbirostris group, An. peditaeniatus (19.4%) of the Hyrcanus group, An. aconitus (13.4%) of the Funestus group, and An. nivipes (11.5%) of the Annularis group. The kdr mutation L1014F was only detected in the homozygous state in two An. subpictus (s.l.) specimens and in a heterozygous state in one An. culicifacies (s.l.) specimen.ConclusionsThis study provides updated information about malaria vector species composition and insecticide resistance status in northern Myanmar. The confirmed deltamethrin resistance in multiple species groups constitutes a significant threat to malaria vector control. The lack or low frequency of target-site resistance mutations suggests that other mechanisms are involved in resistance. Continual monitoring of the insecticide resistance of malaria vectors is required for effective vector control and insecticide resistance management.
