Establishment and maintenance of pregnancy in the pig is a complex process that relies on conceptus regulation of the maternal proinflammatory response to endometrial attachment. Following elongation, pig conceptuses secrete interferon gamma (IFNG) during attachment to the endometrial luminal epithelium. The objective here was to determine if conceptus production of IFNG is important for early development and establishment of pregnancy. CRISPR/Cas9 gene editing and somatic cell nuclear transfer technologies were used to create an IFNG loss-of-function study in pigs. Wild-type (IFNG+/+) and null (IFNG−/−) fibroblast cells were used to create embryos through somatic cell nuclear transfer. IFNG expression was not detected in IFNG−/− conceptuses on either day 15 or day 17 of pregnancy. Ablation of conceptus IFNG production resulted in the reduction of stromal CD3+ and mast cells, which localized to the site of conceptus attachment on day 15. The uteri of recipients with IFNG−/− conceptuses were inflamed, hyperemic and there was an abundance of erythrocytes in the uterine lumen associated with the degenerating conceptuses. The endometrial stromal extracellular matrix was altered in the IFNG−/− embryo pregnancies and there was an increased endometrial mRNA levels for collagen XVII (COL17A1), matrilin 1 (MATN1), secreted phosphoprotein 1 (SPP1), and cysteine-rich secretory protein 3 (CRISP3), which are involved with repair and remodeling of the extracellular matrix. These results indicate conceptus IFNG production is essential in modulating the endometrial proinflammatory response for conceptus attachment and survival in pigs.

Uterine and cervical size of Holstein dairy cows is reported among reasons for a decline in dairy cow fertility. Therefore, the objectives of this study were to (a) determine whether size of the cervix and uterus at 4 weeks postpartum impacted subsequent fertility at first service in Jersey cattle, (b) determine whether progesterone level at 4 weeks postpartum impacted cyclicity and (c) the association of the presence of corpus luteum and uterus and cervix size. Body condition scores at calving, presence of postpartum diseases, parity number and milk weights were taken from lactating Jersey dairy cows (N = 147) for 28 days postpartum. During the fourth week postpartum, a blood sample was obtained for progesterone concentration, and transrectal ultrasonography was performed by a high-resolution ultrasound machine to determine cervical and uterine horn diameter, as well as ovarian structures measurements. Correcting for parity number, BCS at calving, presence of diseases and milk yield, cows with a cervix >2.54 ± 0.63 cm and uterine horn >2.25 ± 0.59 cm were less likely to become pregnant at first service (p = .04 and p = .003, respectively). The cows with larger cervix had a trend to be less likely to have a corpus luteum present at the 4th week of lactation (p = .067). Cows with larger uterine horn size were less likely to have a corpus luteum present at the 4th week of lactation (p = .015). It is concluded that a larger cervix and/or uterus during the postpartum was associated negatively with fertility and cyclicity in Jersey cows.

Stress affects the productivity and fertility of cattle. Stress causes strain and individual animals experience different amounts of strain in response to the same amount of stress. The amount of strain determines the impact of stress on fertility. Typical stresses experienced by cattle include environmental, disease, production, nutritional, and psychological. The effect of stress on the reproductive system is mediated by body temperature (heat stress), energy metabolites and metabolic hormones (production and nutritional stresses), the functionality of the hypothalamus-pituitary-gonadal (HPG) axis and (or) the activation of the hypothalamus-pituitary-adrenal (HPA) axis. The strain that occurs in response to stress affects uterine health, oocyte quality, ovarian function, and the developmental capacity of the conceptus. Cows that have less strain in response to a given stress will be more fertile. The goal for future management and genetic selection in farm animals is to reduce production stress, manage the remaining strain, and genetically select cattle with minimal strain in response to stress.

In utero heat stress (IUHS) increases postnatal energy requirements of pigs, and this may exacerbate weaning and transport stress. The study objective was to evaluate the impact of providing a nutrient dense (ND) diet, which met the greater energy requirements of IUHS pigs, on biomarkers of post-absorptive metabolism and stress following weaning and transport. Twenty-four pregnant gilts were assigned to thermoneutral (n = 12; 17.5 ± 2.1°C) or heat stress (n = 12; cyclical 26°C to 36°C) conditions from gestation d 1 to 58, and then TN conditions until farrowing. At weaning (16.2 ± 0.4 d), mixed-sex piglets (n = 160; 4.78 ± 0.15 kg BW) were transported (loading + transport + unloading) for 11 h 40 min and then blocked into pens (n = 4 piglets/pen) by in utero and dietary treatments fed for 14 d (P1): in utero thermoneutral (IUTN) + control diet (C; n = 10 pens), IUTN + ND (n = 10 pens), IUHS + C (n = 10 pens), IUHS + ND (n = 10 pens). The C diet was fed to all pigs from d 15 to 35 post-weaning and transport (P2). One pig per pen was bled immediately pre-transport (Pre-T) and immediately post-transport (Post-T), and on d 2, 7, 14, 28, and 35 post-weaning and transport to analyze cortisol, glucose, insulin, and non-esterified fatty acids (NEFA). Overall, cortisol was decreased (P = 0.03; 25.8%) during transport in IUHS versus IUTN pigs. On d 2, glucose was decreased overall (P = 0.01; 13.5 mg/dL) in IUHS versus IUTN pigs, but no d 2 insulin (P = 0.76; 0.079 ± 0.011 ng/mL) or NEFA (P = 0.65; 810.3 ± 61.1 mEq/L) differences were detected. Insulin and insulin:glucose ratio were increased (P < 0.04; 0.076 ng/mL and 0.05, respectively) during P1 in ND versus C pigs. In summary, the physiological stress response and glucose levels were altered in IUHS versus IUTN pigs post-weaning and transport, but dietary treatment did not interact with in utero treatment.

In utero heat stress (IUHS) reduces swine productivity and welfare but little is known about the long-term impact on immune function. The study objective was to determine the effects of IUHS on the immune and metabolic response of pigs subjected to an 8 h lipopolysaccharide (LPS) challenge during postnatal life. Twenty-four pregnant gilts were exposed to thermoneutral (TN; n = 12; 17.5 ± 2.1⁰C) or heat stress (HS; n = 12; cyclical 27°C-37°C) conditions from d 1 to 58 of gestation, and TN conditions from d 59 of gestation to farrowing. After farrowing, all piglets were housed under the same conditions. At 12 weeks post-farrowing, 16 IUHS and 16 in utero thermoneutral (IUTN) pigs were selected, balanced by sex and given an intravenous injection of LPS (2 µg/kg BW) or saline (SAL). Treatment combinations were: IUTN-SAL, IUTN-LPS, IUHS-SAL, IUHS-LPS. Body temperature was monitored in 30-min intervals and blood samples were obtained at 0, 1, 2, 3, 4, 6, and 8 h. Blood was analyzed for glucose, insulin, non-esterified fatty acids (NEFA), and cytokine concentrations. Body temperature increased (P < 0.01; 1.05°C) in LPS versus SAL pigs, regardless of in utero treatment. Glucose concentrations were reduced overall (P = 0.05; 5.9%) in IUHS versus IUTN pigs. Non-esterified fatty acid concentrations tended to be greater (P = 0.07; 143.4%) in IUHS-LPS pigs compared to all other treatments, and IUTN-LPS pigs tended to have greater circulating NEFA concentrations (127.4%) compared to IUTN-SAL and IUHS-SAL pigs. At 1 h, TNFα was increased (P = 0.01; 115.1%) in IUHS-LPS compared to IUTN-LPS pigs. Overall, IL-1β and IL-6 were greater (P < 0.04; 56.0 and 46.8%, respectively) in IUHS-LPS compared IUTN-LPS pigs. In summary, IUHS altered the postnatal immune and metabolic response of pigs during postnatal life, which has negative implications towards future disease susceptibility.

The effects of in utero heat stress (IUHS) range from decreased growth performance to altered behavior, but the long-term impact of IUHS on postnatal innate immune function in pigs is unknown. Therefore, the study objective was to determine the effects of early gestation IUHS on the immune, metabolic, and stress response of pigs subjected to an 8 hr lipopolysaccharide (LPS) challenge during postnatal life. Twenty-four pregnant gilts were exposed to thermoneutral (TN; n = 12; 17.5 ± 2.1 °C) or heat stress (HS; n = 12; cyclic 26 to 36 °C) conditions from days 6 to 59 of gestation, and then TN conditions (20.9 ± 2.3 °C) from day 60 of gestation to farrowing. At 12 wk of age, 16 IUHS and 16 in utero thermoneutral (IUTN) pigs were selected, balanced by sex and given an intravenous injection of LPS (2 µg/kg BW mixed with sterile saline [SAL] and injected at 2 µL/kg BW) or SAL (2 µL/kg BW). Body temperature was monitored every 30 min, and blood was obtained at 0, 1, 2, 3, 4, 6, and 8 hr following the LPS challenge. Blood samples were analyzed for glucose, insulin, non-esterified fatty acids (NEFA), cortisol, and cytokine concentrations. In addition, white blood cell counts were determined at 0 and 4 hr. Hour 0 data were used as covariates. Body temperature was increased (P < 0.01) in LPS (40.88 ± 0.08 °C) vs. SAL (39.83 ± 0.08 °C) pigs. Eosinophils tended to be decreased overall (P = 0.09; 43.9%) in IUHS vs. IUTN pigs. Glucose concentrations were reduced overall (P = 0.05; 5.9%) in IUHS vs. IUTN pigs. The NEFA concentrations tended to be greater (P = 0.07; 143.4%) in IUHS-LPS pigs compared with all other treatments, and IUTN-LPS pigs tended to have greater (127.4%) circulating NEFA concentrations compared with IUTN-SAL and IUHS-SAL pigs. Cortisol was increased (P = 0.04) in IUHS-LPS compared with IUTN-LPS pigs at 3 hr (21.5%) and 4 hr (64.3%). At 1 hr, tumor necrosis factor α was increased (P = 0.01; 115.1%) in IUHS-LPS compared with IUTN-LPS pigs. Overall, interleukin-1β (IL-1β) and interleukin-6 (IL-6) were greater (P < 0.04; 281.3% and 297.8%, respectively) in IUHS-LPS pigs compared with all other treatments, and IUTN-LPS pigs had increased IL-1β and IL-6 concentrations compared with IUTN-SAL and IUHS-SAL pigs. In summary, IUHS altered the postnatal cytokine, metabolic, and physiological stress response of pigs during postnatal life, which may have negative implications toward the innate immune response of IUHS pigs to pathogens.

The primary objective was to assess the development of fetal gonads and measure the subsequent reproductive capacity of boars and gilts whose mother was either subjected to gestational heat stress (GHS) or thermoneutral (GTN; control) conditions during pregnancy. Gilts were subjected to either GHS (28 to 38 °C; 65% to 88% relative humidity [RH]; n = 30) or GTN (17 to 22 °C; 56% to 65% RH; n = 29) for the second month of gestation (a period that coincides with a critical window of gonadal development). A subset of GHS (n = 12) and GTN (n = 11) gilts was sacrificed immediately following treatment for the collection of pregnancy data. The remaining gilts (n = 18 GHS and n = 18 GTN) were allowed to farrow. Female offspring from the farrowed gilts were studied through puberty, first insemination, and early pregnancy when fetal tissues were again collected. During the treatment period, GHS gilts had greater (P < 0.001) rectal temperature and respiration rate at both measurement time points (morning and afternoon) compared with GTN gilts. When assessed at the end of the second month of gestation, the total number of viable fetuses did not differ (P > 0.10) for GHS vs. GTN. Likewise, the weight of the fetus, placenta, fetal testes, and fetal ovaries were similar (P > 0.10) for GHS and GTN pregnancies. There was a tendency for an effect of treatment (63.3 ± 2.3 vs. 70.1 ± 2.6; GHS vs. GTN; P < 0.073) on the number of oogonia per histological section in the fetal ovaries. There was no effect of treatment on the number of prespermatogonia per histological section in the fetal testis. For gilts farrowing after treatment, litter size, piglet birth weight, and weaning weight were similar (P > 0.10) for the GHS and GTN gilts. Testes collected from castrated GHS boars had fewer prespermatogonia per seminiferous tubule cross section (P < 0.049). Female offspring from the GHS (n = 30) or GTN (n = 37) sows reached puberty at a similar age, and their pregnancies (ninth week of gestation) had fewer corpora lutea (15.6 ± 0.5 vs. 17.1 ± 0.4; GHS vs. GTN; P < 0.038) but the number of fetuses was similar for GHS and GTN. In summary, compared with GTN, GHS during a critical window of gonadal development tended to reduce the number of oogonia in the fetal ovary, reduced the number of prespermatogonia in the neonatal testes, and reduced ovulation rate at first pregnancy in gilts.

In utero heat stress (IUHS) increases the energy requirements of pigs during postnatal life, and this may compound weaning and transport stress. The study objective was to evaluate and mitigate the negative effects of IUHS following weaning and transport through the provision of a nutrient-dense (ND) nursery diet formulated to meet the greater energy requirements of IUHS pigs during the first 14 d postweaning and transport. Twenty-four pregnant gilts were exposed to thermoneutral (TN; n = 12; 17.5 ± 2.1 °C) or heat stress (HS; n = 12; cycling 26 to 36 °C) conditions for the first half of gestation (day 6 to 59) and then TN conditions (20.9 ± 2.3 °C) until farrowing. Nine TN gilts and 12 HS gilts produced litters. At weaning (16.2 ± 0.4 d), mixed-sex piglets (N = 160; 4.78 ± 0.15 kg body weight [BW]) were transported (loading + transport + unloading) for 11 h 40 min. Following transport, piglets were blocked into pens (n = 4 pigs/pen) by in utero and dietary treatments: in utero thermoneutral (IUTN) + control (C) diet (n = 10 pens), IUTN + ND (n = 10 pens), IUHS + C (n = 10 pens), and IUHS + ND (n = 10 pens). Treatment diets were fed from day 1 to 14 postweaning and transport (period 1), and the C diet was fed to all pigs from day 14 to 35 postweaning and transport (period 2). Production measures were taken in 7 d intervals to calculate average daily gain (ADG), average daily feed intake (ADFI), average daily net energy intake (ADEI), gain:feed, and gain:net energy intake. Blood samples were collected prior to transport, following transport, and on days 2, 7, 14, 28, and 35 postweaning and transport to analyze cortisol, glucose, insulin, and nonesterified fatty acids. Behavior was assessed through video-recording on days 3, 5, 8, 11, and 13 postweaning and transport. In period 1, ADG was reduced (P = 0.04; 20.0 g/d) in IUHS vs. IUTN pigs. Pigs fed ND diets had reduced ADFI (P = 0.02; 9.3%) compared with C diet-fed pigs during period 1, which resulted in similar ADEI (P = 0.23; 1,115 ± 35 kcal/d). During transport, cortisol was decreased (P = 0.03; 25.8%) in IUHS vs. IUTN pigs. On day 2, glucose was decreased (P = 0.01; 13.8%) in IUHS vs. IUTN pigs. No in utero treatment-related behavior differences were observed but lying was reduced (P = 0.03; 6.5%) and standing was increased (P = 0.04; 14.1%) in ND vs. C pigs overall. In summary, IUHS reduced growth performance in pigs following weaning and transport, and providing an ND diet did not rescue the lost performance.

This chapter provides a basic understanding of reproductive physiology and endocrinology of the pig. It begins by providing background of the reproductive anatomy of male and female pigs and followed by developing a fundamental knowledge of spermatogenesis and fertility in the boar, and follicle development, the estrous cycle, fertilization, conceptus development, maternal recognition of pregnancy, parturition and lactation in the sow. Spermatogenesis and sperm quality in boars, as well as the endocrinology of the estrous cycle, early elongation of conceptus trophectoderm and attachment of trophectoderm to the uterine surface for epitheliochorial placentation of pig and the importance of colostrum during lactation are specifically described and illustrated.

In utero stress has been shown to negatively affect intact male rats and mice, though very little research has been conducted in boars. The objectives of the present studies were to determine the effects of in utero heat stress (IUHS) on postnatal development and the response to postnatal heat stress of boars. Ten boars were selected at weaning from litters subjected to IUHS or in utero thermoneutral (IUTN) during 30–60 days of gestation. The boars were evaluated for reproductive performance from birth through 57 weeks of age (WOA). Testicular area tended to be smaller for IUHS boars compared to IUTN boars at 24 WOA (P = 0.080). Libido did not differ for IUHS or IUTN (P = 0.818). Total sperm production was reduced in IUHS boars compared to IUTN boars (P ≤ 0.038). Semen volume and semen concentration did not differ (P ≥ 0.469 and P ≥ 0.664, respectively). Total motility and progressive motility did not differ for IUHS and IUTN boars (P ≥ 0.430 and P ≥ 0.652, respectively). In utero heat stressed boars had a greater incidence of sperm with tail abnormalities than IUTN (P ≤ 0.042). In utero heat stressed boars had a lower incidence of sperm with proximal droplets following mild, postnatal heat stress compared with IUTN (P = 0.005). In utero heat stress resulted in significant reductions in sperm production and increased sperm abnormalities in boars. The IUHS boars may be slightly more tolerant to postnatal heat stress, though more research is needed.

Establishment and maintenance of pregnancy in the pig involves activating many physiological, cellular, and molecular signaling pathways between the developing conceptus and hormonally regulated maternal endometrium. Rapid elongation of the pig trophoblast allows for the establishment of sufficient placental surface area for the transport of nutrients to the fetus throughout pregnancy. Estrogens secreted by the conceptus during elongation act on uterine epithelia to induce secretion of uterine factors required for conceptus development and for preventing endocrine secretion of prostaglandin F2α, which would cause luteolysis. Thus, trophoblast expansion within the uterine lumen during early gestation is an essential process for implantation and maintenance of pregnancy in species with an epitheliochorial form of placentation. In the pig, rapid conceptus elongation involves the unique expression of interleukin-1 beta 2 (IL1B2), which establishes pro-inflammatory effects that may be tempered by the spatiotemporal secretion of estrogen from the conceptuses. The present review provides current information on pig conceptus remodeling and signaling via estrogen and IL1B2 pathways, as well as endometrial responses to those conceptus factors leading to establishment of pregnancy.

Background Fertility in dairy cows depends on ovarian cyclicity and on uterine involution. Ovarian cyclicity and uterine involution are delayed when there is uterine dysbiosis (overgrowth of pathogenic bacteria). Fertility in dairy cows may involve a mechanism through which the uterine microbiota affects ovarian cyclicity as well as the transcriptome of the endometrium within the involuting uterus. The hypothesis was that the transcriptome of the endometrium in postpartum cows would be associated with the cyclicity status of the cow as well as the microbiota during uterine involution. The endometrium of first lactation dairy cows was sampled at 1, 5, and 9 weeks postpartum. All cows were allowed to return to cyclicity without intervention until week 5 and treated with an ovulation synchronization protocol so that sampling at week 9 was on day 13 of the estrous cycle. The endometrial microbiota was measured by 16S rRNA gene sequencing and principal component analysis. The endometrial transcriptome was measured by mRNA sequencing, differential gene expression analysis, and Ingenuity Pathway Analysis. Results The endometrial microbiota changed from week 1 to week 5 but the week 5 and week 9 microbiota were similar. The endometrial transcriptome differed for cows that were either cycling or not cycling at week 5 and cyclicity status depended in part on the endometrial microbiota. Compared with cows cycling at week 5, there were large changes in the transcriptome of cows that progressed from non-cycling at week 5 to cycling at week 9. There was evidence for concurrent and longer-term associations between the endometrial microbiota and transcriptome. The week 1 endometrial microbiota had the greatest effect on the subsequent endometrial transcriptome and this effect was greatest at week 5 and diminished by week 9. Conclusions The cumulative response of the endometrial transcriptome to the microbiota represented the combination of past microbial exposure and current microbial exposure. The endometrial transcriptome in postpartum cows, therefore, depended on the immediate and longer-term effects of the uterine microbiota that acted directly on the uterus. There may also be an indirect mechanism through which the microbiome affects the transcriptome through the restoration of ovarian cyclicity postpartum. The study objective was to determine whether in utero heat stress (IUHS) affects piglet physiology and behavior following common production practices. A total of 12 gilts were confirmed pregnant and allocated to either heat stress (HS, n = 6) or thermoneutral (TN, n = 6) conditions on day 30&ndash, 60 of gestation. At weaning (22.5 &plusmn, 2.3 days of age), 1 boar and 1 barrow of median weight were selected from each litter and transported for approximately 7 h. Piglets were then blocked into pens (n = 2/pen) by in utero treatment (IUHS (n = 12) or in utero thermoneutral (IUTN, n = 12)) and sexual status (boar (n = 6/in utero treatment) or barrow (n = 6/in utero treatment)). Plasma cortisol, non-esterified fatty acids (NEFA), insulin and glucose were evaluated 1 day prior to transport (pre-transport) and immediately after transport (post-transport). Behavioral data were collected on day 1&ndash, 7 for 60 min at four different time points each day. In utero heat stressed piglets exhibited reduced cortisol concentrations compared to IUTN piglets immediately post-transport (p = 0.04). Glucose concentrations were not affected by in utero treatment. Insulin concentrations were reduced in IUTN piglets post-transport compared to pre-transport (p = 0.002), but no differences were detected for IUHS pigs. Non-esterified fatty acids tended to be reduced overall for IUHS vs. IUTN pigs (p = 0.08). Overall, IUHS piglets performed more drinking behaviors (p = 0.02) and tended to perform more aggressive behaviors (p = 0.07) than IUTN piglets in the 7 days post-transport. In summary, there was some evidence for altered physiological and behavioral responses among IUHS piglets compared to IUTN piglets following weaning and transport.

Abundant evidence from the medical, veterinary, and animal science literature demonstrates that there is substantial room for improvement of the clarity, completeness, and accuracy of reporting of intervention studies. More rigorous reporting guidelines are needed to improve the quality of data available for use in comparisons of outcomes (or meta-analyses) of multiple studies. Because of the diversity of factors that affect reproduction and the complexity of interactions between these, a systematic approach is required to design, conduct, and analyze basic and applied studies of dairy cattle reproduction. Greater consistency, clarity, completeness, and correctness of design and reporting will improve the value of each report and allow for greater depth of evaluation in meta-analyses. Each of these benefits will improve understanding and application of current knowledge and better identify questions that require additional modeling or primary research. The proposed guidelines and checklist will aid in the design, conduct, analysis, and reporting of intervention studies. We propose an adaptation of the REFLECT (Reporting Guidelines for Randomized Controlled Trials for Livestock and Food Safety) statement to provide guidelines and a checklist specific to reporting intervention studies in dairy cattle reproduction. Furthermore, we provide recommendations that will assist investigators to produce studies with greater internal and external validity that can more often be included in systematic reviews and global meta-analyses. Such studies will also assist the development of models to describe the physiology of reproduction.

The estrous cycle, breeding, establishment of pregnancy, early embryonic and placental development, parturition and lactation are described to provide a fundamental understanding of reproduction in the pig. Endocrinology of the estrous cycle, early conceptus elongation and attachment to the uterine surface to form the epitheliochorial type of placentation of pig are specifically illustrated to compare differences in pregnancy establishment compared to other mammalian species. The article also provides standard methods of breeding and managing reproduction in the swine industry.

Infertility in lactating dairy cows is explained partially by the metabolic state associated with high milk production. The hypothesis was that lactating and nonlactating cows would differ in endometrial and placental transcriptomes during early pregnancy (day 28 to 42) and this difference would explain the predisposition for lactating cows to have embryonic loss at that time. Cows were either milked or not milked after calving. Reproductive [endometrium (caruncular and intercaruncular) and placenta] and liver tissues were collected on day 28, 35, and 42 of pregnancy. The hypothesis was rejected because no effect of lactation on mRNA abundance within reproductive tissues was found. Large differences within liver demonstrated the utility of the model to test an effect of lactation on tissue gene expression. Major changes in gene expression in reproductive tissues across time were found. Greater activation of the transcriptome for the recruitment and activation of macrophages was found in the endometrium and placenta. Changes in glucose metabolism between day 28 and 42 included greater mRNA abundance of rate-limiting genes for gluconeogenesis in intercaruncular endometrium and evidence for the establishment of aerobic glycolysis (Warburg effect) in the placenta. Temporal changes were predicted to be controlled by CSF1, PDGFB, TGFB1, and JUN. Production of nitric oxide and reactive oxygen species by macrophages was identified as a mechanism to promote angiogenesis in the endometrium. Reported differences in pregnancy development for lactating vs. nonlactating cows could be explained by systemic glucose availability to the conceptus and appeared to be independent of the endometrial and placental transcriptomes.

Seasonal infertility is a significant problem in the swine industry, and may be influenced by photoperiod and heat stress. Heat stress during gestation in particular affects pregnancy, resulting in long-term developmental damage to the offspring. This review summarizes what is known about how heat stress on the pregnant sow affects lactation and her offspring. Sows responded to heat stress during gestation with increased rectal temperature, respiration rate, and skin temperature, and tended to reduce their activity-which may have changed their body composition, increasing the adipose-to-muscle ratio. Heat stress during gestation caused temporary insulin resistance during lactation, but this metabolic state did not seem to affect health, lactation, or rebreeding performance of the sow. Heat-stressed sows also presented with a shorter gestation period and reduced litter birth weight, although weaning weights are not affected when these sows are moved to thermoneutral conditions for lactation. The offspring of gestational heat-stressed sows, however, possessed unique phenotypes, including elevated body temperature, greater fat deposition, and impaired gonad development. Thus, gestational heat stress may significantly impact a herd through its effects on sows and their offspring. Further work is necessary to determine the magnitude of the effects across fa cilities and breeds.

Short-term changes to milking frequency can alter the metabolic status of dairy cows depending on the duration, magnitude, and stage of lactation at which the milking frequency changes occur. Additionally, effects of altered milking frequency that are subsequent to cows returning to a normal twice-daily (2×) milking regimen are not well established. This study tested the hypothesis that plasma concentrations of key hormones and metabolites and transcription of genes involved in the somatotropic axis and lipid metabolism would be altered in liver and subcutaneous adipose tissue from cows milked with different frequencies. Multiparous Holstein-Friesian dairy cows were allocated to 2× milking for the whole lactation, or once-(1×) or 3 times-(3×) daily milking for 3 or 6 wk, immediately postpartum, and then 2× milking for the remainder of the lactation. Liver and subcutaneous fat were biopsied at wk 1 (liver only), 3, 6, and 9 postpartum, and transcription of genes involved in the somatotropic axis and lipid metabolism were measured. At wk 3, cows milked 3× had lower hepatic expression of growth hormone receptor (GHR1A) compared with cows milked 2× or 1×, and lower IGF1 expression compared with cows milked 1×, indicating greater uncoupling of the somatotropic axis. At wk 6, reduced transcription of total GHR and GHR1B occurred in the adipose tissue of cows milked 3×. Cows milked 1× had greater transcription in adipose tissue of lipogenesis genes at wk 3 and 6, and lipolysis genes at wk 6, compared with cows milked 2×, indicating a period of increased fatty acid storage, followed by increased fatty acid reesterification. At wk 9, cows previously milked 3× for 6 wk maintained lower transcription of genes involved in lipogenesis, lipolysis, and ketolysis in adipose tissue compared with cows milked 2×, indicating that the effects of 3× milking persist for at least 3 wk after switching to 2× milking. Results indicate that alterations to milking frequency affect the transcription of genes involved in lipid mobilization and storage, enabling the animal to manage the energy demands associated with the change in milk production. Some of these gene transcription changes were maintained in cows previously milked 3×, indicating that the adipose tissue gene expression changes were still required even after 3 wk of the less-demanding 2× milking regimen.

Milk and milk solids production per cow is increasing annually in dairy systems. Peak milk production is in early lactation when the uterus and ovary are recovering from the previous pregnancy. The competing processes of milk production and restoration of reproductive function can be at odds, particularly if unique homeorhetic mechanisms that typify early lactation become imbalanced and cows experience metabolic disease. Homeorhesis leads to an increase in the synthesis of glucose that is irreversibly lost to milk lactose. Irreversible loss of glucose during lactation can invoke an endocrine and metabolic state that impinges upon postpartum uterine health, oestrous cyclicity and subsequent establishment of pregnancy. The first 30 days postpartum may be most critical in terms of the impact that metabolites and metabolic hormones have on reproduction. Depressed immune function caused in part by the postpartum metabolic profile leads to a failure in uterine involution and uterine disease. Oestrous cyclicity (interval to first ovulation and subsequent periodicity) is affected by the same hormones and metabolites that control postpartum immune function. Slower growth of the embryo or foetus perhaps explained by the unique metabolic profile during lactation may predispose cows to pregnancy loss. Understanding homeorhetic mechanisms that involve glucose and collectively affect postpartum uterine health, oestrous cyclicity and the establishment of pregnancy should lead to methods to improve postpartum fertility in dairy cows.

A method for timed artificial insemination (AI) that is used for beef cows, beef heifers, and dairy heifers employs progesterone-releasing inserts, such as the controlled internal drug release (CIDR; Zoetis, New York, NY, USA) that are left in place for 14 days. The 14-day CIDR treatment is a method of presynchronization that ensures that cattle are in the late luteal phase of the estrous cycle when PGF2α is administered before timed AI. The objective of this study was to test the effectiveness of the 14dCIDR-PGF2α program in postpartum dairy cows by comparing it with the traditional "Presynch-Ovsynch" (2xPGF2α-Ovsynch) program. The 14dCIDR-PGF2α cows (n = 132) were treated with a CIDR insert on Day 0 for 14 days. At 19 days after CIDR removal (Day 33), the cows were treated with a luteolytic dose of PGF2α, 56 hours later were treated with an ovulatory dose of GnRH (Day 35), and 16 hours later were inseminated. The 2xPGF2α-Ovsynch cows were treated with a luteolytic dose of PGF2α on Day 0 and again on Day 14. At 12 days after the second PGF2α treatment (Day 26), the cows were treated with GnRH. At 7 days after GnRH, the cows were treated with PGF2α (Day 33), then 56 hours later treated with GnRH (Day 35), and then 16 hours later were inseminated. There was no effect of treatment or treatment by parity interaction on pregnancies per AI (P/AI) when pregnancy diagnosis was performed on Day 32 (115/263; 43.7%) or Days 60 to 90 (99/263; 37.6%) after insemination. There was an effect of parity (P < 0.05) on P/AI because primiparous cows had lesser P/AI (35/98; 35.7%) than multiparous cows (80/165; 48.5%) on Day 32. Cows observed in estrus after the presynchronization step (within 5 days after CIDR removal or within 5 days after the second PGF2α treatment) had greater P/AI than those not observed in estrus (55/103; 53.4% vs. 60/160; 37.5%; observed vs. not observed; P < 0.01; d 32 pregnancy diagnosis). When progesterone data were examined in a subset of cows (n = 208), 55.3% of cows had a "prototypical" response to treatment (i.e., the cow had an estrous cycle that was synchronized by the presynchronization treatment and then the cow responded appropriately to the subsequent PGF2α and GnRH treatments before timed AI). Collectively, cows with a prototypical response to either treatment had 52.2% P/AI that was greater (P < 0.001) than the P/AI for cows that had a nonprototypical response (19%) (P/AI determined at 60-90 days of pregnancy). In conclusion, we did not detect a difference in P/AI when postpartum dairy cows were treated with 14dCIDR-PGF2α or 2xPGF2α-Ovsynch before timed AI. The primary limitation to the success of either program was the failure of the cow to respond appropriately to the sequence of treatments.

Low blood glucose concentrations after calving are associated with infertility in postpartum dairy cows perhaps because glucose is a master regulator of hormones and metabolites that control reproductive processes. The hypothesis was that low blood glucose postpartum is caused by inadequate glucose entry rate relative to whole-body demand as opposed to the alternative possibility that postpartum cows have a lower regulatory set point for blood glucose. Eight early postpartum (10 to 25 d) dairy cows (5 Holstein and 3 Guernsey) were jugular catheterized. During the first 24 h, cows were infused with physiological saline at 83.3 mL/h. After 24 h, the infusion solution was switched to 50% dextrose that was infused at a rate of 41.7 mL/h (total daily glucose dose=500 g). On d 3 and d 4, the rate of glucose infusion was increased to 83.3 mL/h (daily dose=1,000 g) and 125 mL/h (daily dose=1,500 g), respectively. On d 5, physiological saline was infused at 83.3 mL/h. Blood was sampled hourly through a second jugular catheter (contralateral side) and analyzed for glucose, nonesterified fatty acids, β-hydroxybutyrate, insulin-like growth factor 1, and insulin. Blood glucose concentrations on d 1 (saline infusion) averaged 53.4±1.7 mg/dL. Blood glucose concentrations increased on d 2 when cows were infused with 500 g/d and increased further on d 3 when cows were infused with 1,000g of glucose/d. Increasing the infusion rate to 1,500 g/d on d 4 did not cause a further increase in blood glucose concentrations. Based on a segmented regression analysis, the upper physiological set point for blood glucose was 72.1 mg/dL. Both insulin and insulin-like growth factor 1 concentrations increased in response to glucose infusion and decreased when cows were infused with saline on d 5. Serum nonesterified fatty acids and β-hydroxybutyrate concentrations decreased in response to glucose infusion and rebounded upward on d 5 (saline infusion). In conclusion, early postpartum cows had circulating blood glucose concentrations that were well below the upper set point defined in this study (72.1 mg/dL). Infusing approximately 1,000 g of glucose daily increased blood glucose to the physiological set point and rapidly changed the hormonal and metabolic profile that typifies postpartum cows. The inability of the early postpartum cow to achieve an adequate entry rate for glucose relative to whole-body demand is a possible mechanism that links postpartum physiology and nutrition to reproduction in dairy cows.

Fertility to timed AI (TAI) is profoundly affected by progesterone (P4) levels during hormonal synchronization protocols. Holstein-Friesian dairy cows managed in a seasonal-calving, pasture-based production system were randomly assigned to 2 treatments to manipulate P4 before TAI during growth of the preovulatory follicle. Cows in the first treatment (High P4; n=30) were submitted to a Double-Ovsynch protocol {Pre-Ovsynch [GnRH; 7 d, PGF2α; 3 d, GnRH] followed 7 d later by Breeding-Ovsynch [GnRH (G1); 7 d PGF2α; 24 h, PGF2α; 32 h, GnRH (G2); 16 h, TAI]}. Cows in the second treatment (n=30; Low P4) received the same Double-Ovsynch protocol but with an additional PGF2α treatment 24 h after G1. Overall, synchronization rate did not differ between treatments and was 92% (55/60). Unexpectedly, 37% of Low P4 cows were detected in estrus ~24 h before scheduled TAI and were inseminated ~16 h before scheduled TAI. Overall, P4 did not differ between treatments at G1, whereas High P4 cows had greater P4 concentrations at PGF2α and G2 than Low P4 cows. High P4 cows had the smallest mean follicle diameter at G2, whereas Low P4 cows with no estrus before TAI had intermediate mean follicle diameter at G2, and Low P4 cows with estrus before TAI had the largest mean follicle diameter. Low P4 cows with estrus before TAI had larger corpora lutea 15 d after TAI than Low P4 cows without estrus before TAI or High P4 cows. In accordance with corpus luteum size on d 15, High P4 cows and Low P4 cows without estrus before TAI had lower P4 from 4 to 46 d after TAI than Low P4 cows with estrus before TAI. Relative mRNA levels of the interferon-stimulated genes ISG15, MX1, MX2, and OAS1 were greater for Low P4 than for High P4 cows, whereas relative mRNA levels of RTP4 were greater for High P4 than for Low P4 cows 18 d after TAI. Treatment did not affect plasma pregnancy-associated glycoprotein concentrations after TAI; however, pregnancy-associated glycoprotein concentrations were affected by pregnancy status and parity. Treatment did not affect pregnancy per artificial insemination at 29, 39, or 60 d after TAI, and no pregnancy losses were observed from 39 to 60 d after TAI. We concluded that (1) Low P4 cows were more likely to express estrus than High P4 cows; (2) the subpopulation of Low P4 cows that expressed estrus had larger preovulatory follicles and greater P4 concentrations after TAI; and (3) regardless of estrus before TAI, all Low P4 cows had greater mRNA expression for 5 of 6 interferon-stimulated genes than High P4 cows 18 d after TAI.

Glucose is an essential nutrient for the conceptus. The objective was to determine if lactation affected the amount of glucose crossing the placenta by measuring glucose and fructose in placental fluids in lactating and nonlactating cows. Holstein cows were assigned to one of 2 treatments immediately after parturition [lactating (n=23) or nonlactating (dried off immediately after calving; n=20)]. Pregnant cows were slaughtered at one of 3 d of pregnancy (d 28, 35, or 42) and tissues were collected. Plasma glucose and insulin were less in lactating cows. Pregnancies collected from lactating cows had less glucose and fructose in placental fluids compared with those from nonlactating cows. Relative to endometrium, the placenta expressed greater amounts of the glucose transporters SLC2A1 (Glut1), SLC2A3 (Glut3) and SLC2A4 (Glut4) mRNA. The mRNA for SLC2A1 decreased whereas the mRNA for SLC2A4 increased from d 28 to d 42 of pregnancy. Stepwise regression analyses for fetal and placental weight (dependent variable) retained day of pregnancy and maternal plasma insulin concentrations in the final model. The conclusion is that lower blood glucose and insulin in lactating cows may lead to less glucose crossing the placenta and slower fetal development during lactation. The slower fetal development may predispose lactating cows to fetal loss if developmental milestones are not reached.

Previously, we determined that a primary cause of proportional stunted growth in a line of Brahman cattle was related to an apparent refractoriness in metabolic response to GH in young animals. The objective of this study was to determine the effect of administration of GH, insulin (INS), and GH plus INS to mature miniature Brahman cows (n = 6; 9.7 ± 2.06 y; 391 ± 48.6 kg) and bulls (n = 8; 9.4 ± 2.00 y; 441 ± 54.0 kg) on circulating concentrations of metabolic hormones and metabolites, primarily IGF-I and IGF-I binding proteins. We hypothesized that IGF-I secretion could be enhanced by concomitant administration of exogenous GH and INS, and neither alone would be effective. Animals were allotted to a modified crossover design that included four treatments: control (CON), GH, INS, and GH + INS. At the start of the study, one-half of the cattle were administered GH (Posilac; 14-d slow release) and the other one-half served as CON for 7 d. Beginning on day 8, and for 7 d, INS (Novolin L) was administered (0.125 IU/kg BW) twice daily (7:00 AM and 7:00 PM) to all animals; hence, the INS and GH + INS treatments. Cattle were rested for 14 d and then were switched to the reciprocal crossover treatments. Blood samples were collected at 12-hour intervals during the study. Compared with CON, GH treatment increased (P < 0.01) mean plasma concentrations of GH (11.1 vs 15.7 ± 0.94 ng/mL), INS (0.48 vs 1.00 ± 0.081 ng/mL), IGF-I (191.3 vs 319.3 ± 29.59 ng/mL), and glucose (73.9 vs 83.4 ± 2.12 mg/dL) but decreased (P < 0.05) plasma urea nitrogen (14.2 vs 11.5 ± 0.75 mg/dL). Compared with INS, GH + INS treatment increased (P < 0.05) mean plasma concentration of INS (0.71 vs 0.96 ± 0.081 ng/mL), IGF-I (228.7 vs 392.3 ± 29.74 ng/mL), and glucose (48.1 vs 66.7 ± 2.12 mg/dL), decreased (P < 0.01) plasma urea nitrogen (13.6 vs 10.4 ± 0.76 mg/dL), and did not affect GH (13.5 vs 12.7 ± 0.95 ng/mL). In the miniature Brahman model, both the GH and GH + INS treatments dramatically increased circulating concentrations of IGF-I in mature cattle, suggesting that this line of Brahman cattle is capable of responding to bioactive GH.

Establishment of pregnancy in the pig depends on down-regulation of progesterone receptor (PGR) in the uterine luminal and glandular epithelium during the first week after breeding. The present study evaluated the regulation of endometrial PGR by progesterone and the possible role of endometrial tumor necrosis factor (ligand) superfamily member 11 (TNFSF11) and nuclear factor-kappa B (NFKB) activation in PGR expression. Mature, cycling gilts were inseminated (Day 0) and assigned to either untreated control (n = 9) or one of two treatments that employed RU 486 to block progesterone action either before (treatment 1 [T1]) or after (treatment 2 [T2]) the initiation of PGR down-regulation. The T1 gilts were treated with RU 486 (400 mg/day) on Days 3-5 of pregnancy (n = 9), and T2 gilts were treated with RU 486 on Days 6 and 7 of pregnancy (n = 9). Uteri and ovaries were collected on Day 8 or 12 of gestation. The diameter of the conceptuses in T1 gilts was approximately half that in controls by Day 8, and normal conceptuses were not collected from any T1 gilts on Day 12. Endometrial PGR mRNA was more abundant in T1 and T2 gilts compared with control gilts. The PGR-B protein decreased from Day 8 to Day 12 in the luminal epithelium and, to some extent, in superficial glandular epithelium in control and T2 gilts. In T1 gilts, the PGR-B protein remained elevated (i.e., failed to undergo down-regulation) on Day 12. Blocking PGR action early in the cycle (i.e., on or before Day 5), therefore, prevented normal conceptus development, caused elevated PGR mRNA, and prevented the decrease in PGR protein that typically occurs in pigs. We could not confirm a role for NFKB activation in PGR down-regulation, because pigs with extreme differences in PGR and TNFSF11 expression (T1 and controls) had similar NFKB activation on Day 8. Activated NFKB within the luminal epithelium and glandular epithelium (both superficial and deep) was observed in T2 and control pigs on Day 12 when elongating conceptuses (presumably releasing interleukin 1 beta to activate NFKB) were recovered. Gilts treated with RU 486 had greater ovarian follicular growth and greater plasma estradiol concentrations. We conclude that the mechanisms controlling PGR down-regulation are progesterone-dependent and occur between Day 3 and Day 6 of pregnancy. NFKB activation did not appear to have a role in PGR down-regulation within the period that we studied. Blocking progesterone action after Day 6 did not reverse the process of PGR down-regulation, nor did it appear to affect the development of conceptuses collected on Day 12.

The objective was to compare two resynchronization programs; one that used a blood-based ELISA for pregnancy-associated glycoproteins (PAG) for pregnancy diagnosis so that non-pregnant cows were re-inseminated at 28 d after first TAI, and another that used transrectal ultrasonography for pregnancy diagnosis so that non-pregnant cows were re-inseminated at 35 d after first TAI. The PAG_resynch cows (n = 103) began CIDR-Ovsynch resynchronization on Day 18 after first TAI (Day 0). On Day 25, the CIDR was removed and pregnancy diagnosis with a PAG ELISA was performed. If a cow was not pregnant on Day 25, she was treated with PGF 2α , treated with GnRH 2 d later (Day 27), and TAI on Day 28. Control cows (n = 99) were observed for estrus until Day 25, when they began an identical CIDR-Ovsynch program with pregnancy diagnosis by transrectal ultrasonography on Day 32. If a cow was not pregnant on Day 32, then she was treated with PGF 2α , treated with GnRH 2 d later (Day 34), and TAI on Day 35. There was no difference in pregnancy per AI (P/AI) for either group at first or second insemination. For cows without pregnancy loss, the interval between first and second (P 0.001) or second and third (P 0.016) TAI was shorter for PAG_resynch cows compared with Control cows. The interval between first and second or second and third TAI was not different if pregnancy loss cows were included in the analysis. Plasma progesterone concentrations were similar at PGF 2α treatment, and plasma estradiol concentrations increased similarly after PGF 2α treatment for PAG_resynch and Control cows. In conclusion, the 28 d CIDR-Ovsynch resynchronization protocol was comparable to a 35 d CIDR-Ovsynch resynchronization protocol that also included estrus detection. Shortened resynchronization protocols that do not require estrus detection may improve reproductive efficiency in dairy cattle.

Cattle that are not pregnant to first fixed-time artificial insemination (TAI) may be resynchronized for a second TAI if they are found nonpregnant at pregnancy diagnosis. The specific interval between first and second TAI ranges from 4 to 8 wk. The selected interval depends on the available method of pregnancy diagnosis and the efficiency of the resynchronization program. The objective of this experiment was to evaluate a pregnancy diagnosis and resynchronization system that achieved a 21-d interval between TAI. This 21-d interval approximates the natural return-to-service interval. It also enables resynchronization to be implemented within the same estrous cycle in which cattle are first inseminated. Holstein heifers were randomly assigned to a 21-d resynchronization program (21d_resynch; n=40) or a control group in which estrus was observed for the purpose of re-insemination (control; n=29). The 21d_resynch heifers were diagnosed for pregnancy on d 18 after a TAI (d 0) by using predetermined cut-off values for 2′-5′ oligoadenylate synthetase 1 (Oas1) gene expression in leukocytes and plasma progesterone concentration. Heifers that were not pregnant to first TAI had greater expression of Oas1 at the time of PGF 2α (d −3) than pregnant heifers, but this relationship was reversed on d 18 after TAI: the heifers that were pregnant to first TAI had almost 5-fold greater expression of Oas1 compared with nonpregnant heifers. Nonpregnant heifers in the 21d_resynch group were injected with a luteolytic dose of PGF 2α on d 19 and were injected with GnRH on d 21 and submitted to TAI. The pregnancy per AI after first insemination was similar for 21d_resynch (50.0%; pregnancy diagnosis on d 18) and control (51.7%; pregnancy diagnosis on d 27). Likewise, no difference was detected in second insemination pregnancy per AI for 21d_resynch (36.8%; nonpregnant heifers TAI on d 21) and control (35.7%; nonpregnant heifers inseminated at return to estrus or after nonpregnant diagnosis on d 27). The interval between first and second insemination was shorter for 21d_resynch compared with control (21.0±0 and 27.5±2.1 d). The conclusion is that a TAI resynchronization can be programmed within 21 d of previous TAI when a d 18 pregnancy test and a rapid resynchronization are used.

Ghrelin, an orexigenic hormone, is the endogenous ligand for the growth hormone secretagogue receptor (GHSR). Obestatin is produced from the same precursor peptide as ghrelin, and although obestatin was initially thought to promote actions opposite to those of ghrelin, many studies have failed to confirm this hypothesis. In the current study, multiparous cows were continuously infused with ghrelin (n = 10) or obestatin (n = 10) for 8 wk and compared to an untreated group (n = 10) to examine the effects of these hormones on somatotropic and liporegulatory gene expression. The expression of key genes was measured by quantitative real-time polymerase chain reaction. Growth hormone secretagogue receptor mRNA expression was altered in ghrelin- and obestatin-infused cows in a similar manner, as expression was increased at 4 wk, however it had decreased by 8 wk. Obestatin-infused cows presented with a significant decrease in the expression of ATP-binding cassette A1 (ABCA1) in adipose tissue, suggesting changes in cholesterol transport. Liver insulin-like growth factor (IGF) binding protein-3 mRNA displayed a week-by-treatment interaction, as expression was increased in control and obestatin-infused cows; however, expression decreased in ghrelin-infused cows. Adipose expression of hormone sensitive lipase (LIPE) mRNA was not altered by treatment or time, suggesting hormone infusion is not initiating lipolysis. The expression of lipogenic genes in adipose tissue increased with time in all groups, consistent with the general lactational profile of lipogenesis in dairy cows. These data indicate that continuous infusion of ghrelin or obestatin does not alter the expression of key somatotropic or liporegulatory genes in the lactating dairy cow, although obestatin infusion may alter cholesterol transport.

The somatotropic axis [including growth hormone (GH), GH receptor, and insulin-like growth factor (IGF)-I] is uncoupled in high-producing cows in early lactation so that the liver fails to respond to GH and produces less IGF-I. This uncoupling was implicated in the process of nutrient partitioning, enabling high milk production. Different genetic selection goals may affect functional components of the somatotropic axis. Thus, the somatotropic axis was examined in diverse genetic strains of dairy cows [North American Holstein 1990 (NA90), New Zealand Holstein-Friesian 1990 (NZ90), and New Zealand Holstein-Friesian 1970 (NZ70)] that were managed similarly within a pasture-based system but were offered feed allowances commensurate with their genetic ability to produce milk. The NA90 cows produced more milk (26.2 +/- 0.3, 24.1 +/- 0.3, and 20.1 +/- 0.4 kg/d, for NA90, NZ90, and NZ70, respectively), but had lower milk fat percentages (4.28 +/- 0.03, 4.69 +/- 0.03, and 4.58 +/- 0.04 kg/d for NA90, NZ90, and NZ70, respectively) compared with both NZ strains. Milk protein percentages (3.38 +/- 0.02, 3.52 +/- 0.02, and 3.29 +/- 0.03 kg/d for NA90, NZ90, and NZ70, respectively) were greater for NZ90 cows. During early lactation (wk 2 to 6), the total net energy produced in milk was greater in NA90 compared with NZ90 or NZ70 cows, but total net energy in milk after wk 6 was equivalent for NA90 and NZ90 cows. The greater milk production in early lactation in NA90 cows was associated with lower body condition scores (BCS; 1 to 10 scale; 4.0 +/- 0.1) elevated blood GH concentrations (1.6 +/- 0.1 ng/mL), and low blood IGF-I concentrations (14.8 +/- 1.1 ng/mL), indicating an uncoupled somatotropic axis. In comparison, the NZ70 cows retained a coupled somatotropic axis during early lactation, maintaining greater BCS (4.6 +/- 0.1), lower blood GH (0.7 +/- 0.1 ng/mL), and greater blood IGF-I (21.9 +/- 1.2 ng/mL). The degree of uncoupling in NZ90 cows was intermediate between the other 2 strains. Additional feed allowance failed to change blood IGF-I concentrations in NA90 cows but increased IGF-I concentrations in NZ90 cows (20.9 +/- 1.4 and 13.2 +/- 1.4 ng/mL for the high and low feed allowance, respectively). Furthermore, additional feed allowance in NZ90 cows lessened BCS loss in early lactation, but did not affect BCS loss in NA90 cows. Functional components of the somatotropic axis differed for the respective strains and were consistent with strain differences in milk production, BCS, and feed allowance.

The objective of the experiment was to compare follicular dynamics, ovulatory response to GnRH, and synchrony of estrus and ovulation among estrous-cycling and prepubertal beef heifers synchronized with a controlled internal drug-release (CIDR)- based or GnRH-PGF(2alpha) (PG) protocol. Estrous-cycling beef heifers were randomly assigned to 1 of 4 treatments (C1, C2, C3, C4), and prepubertal beef heifers were randomly assigned to 1 of 2 treatments (P1, P2) by age and BW. Blood samples were taken 10 and 1 d before treatment to confirm estrous cyclicity status (progesterone > or =0.5 ng/mL estrous cycling). The CIDR Select (C1, n = 12; P1, n = 14)-treated heifers received a CIDR insert (1.38 g of progesterone) from d 0 to 14, GnRH (100 microg, i.m.) on d 23, and PG (25 mg, i.m.) on d 30. Select Synch + CIDR (C2, n = 12; P2, n = 11)-treated heifers received a CIDR insert and GnRH on d 23 and PG at CIDR removal on d 30. The CIDR-PG (C3, n = 12)-treated heifers received a CIDR insert on d 23 and PG at CIDR removal on d 30. Select Synch (C4, n = 12)-treated heifers received GnRH on d 23 and PG on d 30. HeatWatch transmitters were fitted at CIDR removal (C1, C2, C3, P1, and P2) or at GnRH administration (C4) for estrus detection. Ultrasound was used to determine the response to GnRH and the timing of ovulation after estrus. Among the estrous-cycling heifers, ovulatory response to GnRH and estrous response did not differ (P > 0.05). Among the prepubertal heifers, more (P = 0.02) P1 heifers responded to GnRH than P2 heifers, but estrous response did not differ (P > 0.05). Among the estrous-cycling heifers, variance for interval to estrus after PG was reduced (P 0.05) in variance for interval to estrus or ovulation. Results from C1 and P1 (T1) and C2 and P2 (T2) were combined to compare T1 and T2 among mixed groups of estrous-cycling and prepubertal heifers. Response to GnRH was greater (P < 0.01; 81% T1 and 39% T2), and variances for interval to estrus and ovulation for T1 were reduced (P < 0.01) compared with T2. In summary, CIDR Select improved (P < 0.01) the synchrony of estrus and ovulation compared with Select Synch + CIDR.

After parturition, the somatotropic axis of the dairy cow is uncoupled, partly because of reduced concentration of liver-specific GH receptor (GHR) 1A. Estradiol-17 beta(E(2)) concentrations increase at parturition and E(2) upregulates suppressors of cytokine signaling-2 (SOCS-2) mRNA expression, potentially inhibiting GH signaling. Therefore, we hypothesized that SOCS-2 mRNA is upregulated after parturition. Multiparous Holstein cows (n = 18) were dried off 45 d before expected parturition and fed diets to meet nutrient requirements at ad libitum or limited dry matter intake during the dry period. All cows were fed the same diet ad libitum from calving until 4 wk after parturition. Blood samples were collected weekly and more frequently near parturition. Liver biopsies obtained at - 21, - 7, 2, and 28 d relative to parturition were assessed for SOCS-2 and GHR 1A mRNA by quantitative real-time reverse-transcription PCR. The relative amount of SOCS-2 mRNA increased after parturition with both treatments and was greater on d 2 for cows limit-fed during the dry period compared with cows fed at ad libitum dry matter intake. Plasma E(2) concentrations increased on d - 13, - 5 and 1 relative to parturition and the increases were greater in limit-fed cows. Plasma GH concentration was greater for limit-fed cows and increased after parturition in all cows. The amount of GHR 1A mRNA did not differ between diets but decreased on d 2. In addition to reduced GHR 1A, increased SOCS-2 mRNA after parturition, perhaps because of increased E(2), may further uncouple GH signaling in the liver of the transition dairy cow.

The somatotropic axis consisting of growth hormone, the growth hormone receptor (GHR) insulin-like growth factor (IGF)-I, and IGF binding proteins changes with the stage of lactation and nutrition of the cow and may be 1 mechanism through which lactation and nutrition affect the establishment of pregnancy. The objective of this study was to quantify GHR, IGF-I, and IGF binding protein-2 (IGFBP-2) mRNA in liver and uterine endometrial tissue at 4 stages of lactation (40, 80, 120, and 160 days in milk) and around the time of artificial insemination. Estrus was synchronized with GnRH and PGF 2α , and cows were inseminated 12h after estrus. Uterine biopsies were collected immediately before the second injection of PGF 2α (before estrus), at the initiation of standing estrus, and 4 d after estrus. Liver biopsies were collected once on 4 d after estrus. The abundance of GHR, IGF-I, and IGFBP-2 mRNA in liver and uterus was determined by real-time quantitative PCR. The amount of liver IGF-I mRNA was positively correlated with plasma IGF-I concentrations. Cows that became pregnant after AI had more GHR and IGFBP-2 mRNA in their liver than cows that did not become pregnant. There was no effect of DIM or pregnancy status on abundance of uterine mRNA; however, uterine GHR and IGF-I mRNA was most abundant at estrus. In summary, cows at different stages of lactation or with different pregnancy statuses had similar quantities of uterine mRNA. In contrast, liver quantities of mRNA differed relative to pregnancy status. These data provide evidence that liver indices of metabolic state may be indicative of pregnancy success.

Intervals to estrus and ovulation in weaned sows depend partially on the diameter of ovarian follicles at weaning. The objective was to determine if follicular diameter in sows could be increased by a 48h period of GnRH infusion before weaning and whether this pre-weaning growth would advance follicular development after weaning. The posterior vena cava was cannulated in eight sows at 10+/-1 day after farrowing. Sows were randomly assigned to receive intravenous treatment with either 2mL of GnRH (1microg/mL; n=4) or 2mL of saline (n=4) every 0.5h for 48h beginning 94h before weaning. The average follicular diameter and the number of follicles within diameter classes were determined daily by ultrasonography. Serum LH concentrations increased on the first infusion day but serum LH was equal to control on the last infusion day (P0.077). The GnRH infusion increased the average diameter of ovarian follicles (P0.001). Serum estradiol increased (P0.001) and serum FSH decreased (P0.016) coincident with GnRH-induced follicular development but these changes were reversed within 24h after the end of the infusion period. Follicles that grew in response to GnRH regressed and were replaced by a new population of follicles within 4 days after weaning. Within the experimental model for the present study, a GnRH infusion increased follicular growth in lactating sows but follicles could not be sustained beyond the end of GnRH infusion.
