Abstract

Friction between the contact lens (CL) and the corneal or conjunctival surfaces is considered one of key factors in triggering CL‐associated adverse effects. However, the relationship between friction properties and these effects remains unclear. Traditional measurement methods often fail to replicate real‐life conditions, thereby highlighting the need for more effective apparatus. In this study, the authors developed an optimised pendulum apparatus integrated with an inclinometer to enhance the measurement of CL friction coefficients, thereby improving its precision and relevance to clinical settings. This new design allows for faster and easier calculation of the friction coefficient based on the amplitude decay per libration cycle, surpassing the accuracy of previous video‐based methods. The pendulum's hemisphere component was made from ethylene–propylene–diene monomer rubber (EPDM) 30, which has an elastic modulus similar to that of a human eyeball, creating a measurement environment that closely mimics real‐world usage. The authors optimised the apparatus by evaluating the effects of hemisphere stiffness and saline volume on the friction coefficient. Measurements of multiple lenses recorded by the authors, particularly Lens A, made of narafilcon A, revealed significant consistency across different hemisphere materials with an optimal saline volume of 150 μL yielding a friction coefficient of 0.026 ± 0.003. No statistically significant differences in the friction coefficients were found across variations in the lens base curve, diameter, centre thickness, or power. This improved apparatus demonstrates the capability of effectively measuring friction coefficients under conditions that simulate clinical usage, providing rapid and reliable results. The findings validate the apparatus and suggest its potential for broader applications in assessing CL properties, thereby facilitating future research on the material characteristics and safety of various CLs, including decorative lenses. However, the multi-instillation of these three eye drops causes corneal damage, and the FL drops have the disadvantage of low ocular bioavailability. To overcome these problems, we prepared fixed-combination eye drops based on FL nanoparticles (FL-NPs) and BF/LV solution (nFBL-FC), and evaluated the corneal toxicity and transcorneal penetration of the nFBL-FC eye drops.Methods: FL powder was mixed in 2-hydroxypropyl-β-cyclodextrin solution containing benzalkonium chloride, mannitol and methylcellulose, and milled with a Bead Smash 12 (5500 rpm for 30 s× 30 times). The BF/LV solution was then added to the milled-dispersions to be used as nFBL-FC. The FL, BF and LV concentrations were measured by HPLC methods, and transcorneal penetration was evaluated in rabbits.Results: The FL particle size in nFBL-FC was 40– 150 nm, with only 0.0018% in liquid form. No aggregation of FL particles in the nFBL-FC was observed for 1 month. Abstract

Introduction Pathological changes of severe chronic allergic conjunctivitis are driven not only via acquired immunity but also via innate immunity. Type 2 immune response‐initiating cytokines may play some roles as innate immunity‐dependent components of the ocular surface inflammation. To investigate the involvement of type 2 immune response‐initiating cytokines in innate immunity‐dependent, papain‐induced conjunctival inflammation model using IL‐25‐, IL‐33‐, and TSLP receptor (TSLPR)‐knockout (KO) mice with reference to basophils and ILC2. Methods Papain‐soaked contact lenses (papain‐CLs) were installed in the conjunctival sacs of C57BL/6‐IL‐25 KO, IL‐33 KO, TSLPR KO, Rag2 KO, Bas‐TRECK, and wild‐type mice and their eyes were sampled at day 5. The eosinophil and basophil infiltration in papain‐CL model was evaluated histologically and cytokine expression was examined. To clarify the roles of basophils and ILC2, basophil/ILC2‐depletion experiments were carried out. Results Papain‐induced conjunctival inflammation exhibited eosinophil infiltration and upregulation of Th2 cytokine expression. Reduction of eosinophil and basophil infiltration and attenuated Th2 cytokine expression were observed in the papain‐CL model using IL‐33 KO and TSLPR KO mice. Depletion of basophils or ILC2s in the conjunctivae of the papain‐CL model reduced eosinophil infiltration. Conclusions Innate immunity‐driven type 2 immune responses of the ocular surface are dependent on IL‐33, TSLP, basophils, and ILC2. These components may be possible therapeutic targets for refractory allergic keratoconjunctivitis.

Abstract

Nasopharyngeal carcinoma (NPC), a cancerous tumor that occurs in the back of the nasal cavity, is not easily accessible for clinical evaluation. Computed tomography (CT) provides a method for the examination of this area, but the interpretation of these findings remains a challenge. To evaluate the usefulness of CT in the diagnosis and staging of nasopharyngeal carcinoma, 61 patients were examined by CT. In 29 of those patients, the NPC had invaded the surrounding bone. In four cases, the standard classification system did not indicate bone involvement, but such involvement was observed on the CT. Bone involvement was more readily observed on the CT than conventional radiographs. Computed tomography is clearly useful in the diagnosis of nasopharyngeal carcinoma and the evaluation of spread and bone involvement. We have introduced a zero-calorie sweet sugar, D-psicose (D-allulose), a rare sugar that has been proven to have strong antihyperglycemic and antihyperlipidemic effects, and could be used as a replacement of natural sugar for the obese and diabetic subjects.Aim: Above mentioned efficacy of D-psicose (D-allulose) has been confirmed in our previous studies on type 2 diabetes mellitus (T2DM) model Otsuka Long-Evans Tokushima Fatty (OLETF) rats with short-term treatment. In this study we investigated the long-term effect of D-psicose in preventing the commencement and progression of T2DM with the mechanism of preservation of pancreatic β-cells in OLETF rats.Methods: Treated OLETF rats were fed 5% d-psicose dissolved in water and control rats only water. Nondiabetic control rats, Long-Evans Tokushima Otsuka (LETO), were taken as healthy control and fed water. To follow the progression of diabetes, periodic measurements of blood glucose, plasma insulin, and body weight changes were continued till sacrifice at 60 weeks. Periodic in vivo body fat mass was measured. Abstract

Claudins are members of a large family of transmembrane proteins, which are essential for the formation of tight junctions and have a significant effect on the biological behavior of tumor progression. Previous studies have demonstrated that several claudins show aberrant expression patterns in numerous types of cancer. The present study investigated the expression and localization of claudin-3 and claudin-7 in human colorectal adenocarcinoma cell lines and tissues. The protein expression levels of claudin-3 and claudin-7 were determined using immunocytochemical and immunohistochemical staining. Claudin-3, but not claudin-7, exhibited nuclear localization in the human colorectal adenocarcinoma Caco-2 and SW620 cell lines. Surgically resected colorectal adenocarcinoma tissue specimens were obtained, and the associations between the expression of claudin-3 or claudin-7 and various clinicopathological parameters were analyzed. The membranous expression rates of claudin-3 and claudin-7 were 58.0 and 50.0%, while their nuclear expression rates were 22.0 and 2.0%, respectively. The membranous expression of claudin-3 and claudin-7 was not associated with any clinicopathological factors, whereas the nuclear expression of claudin-3 was associated with histological type and was significantly increased in colorectal mucinous adenocarcinomas compared with that in well- to moderately-differentiated colorectal adenocarcinomas (P<0.01). However, no associations were observed between the nuclear expression of claudin-7 and any clinicopathological parameter. In conclusion, the nuclear expression of claudin-3 in colorectal mucinous adenocarcinoma may be involved in the biological transformation of tumors. The results from the present study indicated that claudin-3 is an important protein associated with histological type and has potential as a prognostic marker. Although the mechanisms underlying the nuclear localization of claudin-3 in tumorigenesis have not yet been elucidated in detail, the present results indicated the potential of claudin-3 as a histopathological biomarker for colorectal adenocarcinomas.
