5. Two-Step Monte Carlo Tree Search for Optimal Design of High-Frequency Toroidal Inductors in Power Electronics Circuits

Abstract

This study devised a novel optimization technique for toroidal inductors utilizing a two-step Monte Carlo tree search (MCTS) algorithm. The proposed method integrates a global search based on analytical formulas with a local search involving 3-D finite element analysis (FEA). By employing this approach, the optimal core materials, sizes, number of turns, and windings are determined to enhance efficiency and reduce the overall size of the inductors. The proposed method was applied to optimize inductors in a 6.78 MHz electrically coupled power transfer system. The results of the study demonstrate that the optimized inductor, identified through the proposed method, not only achieved superior efficiency but also minimized computational costs. Subsequent system measurements confirmed that the optimized inductor improved system efficiency by over 1.8% compared with inductors designed through empirical methods.

Purpose: Elastomeric infusion pumps are widely used in colorectal cancer chemotherapy. However, no studies to date have investigated patient preferences regarding different infusion pump types. Patients and Methods: Twenty patients with unresectable colorectal cancer undergoing chemotherapy were initially treated with a portable hard-shelled continuous infusion pump, followed by a soft-shelled continuous infusion pump. The respondents used a numerical rating scale (0– 10) to rate their comfort when using each pump, their ease of carrying it, the pump size and shape, its weight, their ease of reading its memory, and their overall satisfaction with it. Finally, 13 patients (65%) indicated that they would prefer a soft-shelled pump for their next treatment, while only one patient (5%) preferred a hard-shelled alternative. A preference for soft-shelled pump was observed, particularly in female patients and those with a body mass index of < 22 kg/m2. Conclusion: The selection of portable elastomeric infusion pumps should consider the preferences of patients with colorectal cancer, as these devices have the potential to enhance their quality of life. Abstract

Japanese Brown cattle is the second most popular breed among Wagyu breeds and raised mainly in Kumamoto and Kochi Prefectures. Typical coat color of the Kumamoto sub‐breed is solid brown, but individuals with diluted coat color are sometimes born. We previously detected four SNPs in PMEL gene and identified p.L18del as the causative polymorphism of this diluted phenotype. The current study examined the association between the SNPs in PMEL gene and carcass traits of the Kumamoto sub‐breed. Our association analysis revealed that p.L18del had significant effects on BMS (p = 0.0263), meat brightness (p = 0.0179), meat firmness (p = 0.0102), and meat texture (p = 0.0252) and that del allele of this SNP might be useful to improve these carcass traits. 37. Segment Switching Method of Three-Phase Wireless Power Transmitter for AGV

Abstract

This article proposes a segment-switching method of a three-phase wireless power transmitter for automatic guided vehicles (AGV). The method includes the segmented coil structure, the segment-switching control, and circuit configuration with resonant devices. The segmented coil structure ensures that mutual inductance between the transmitter phase coils and the receiver coil is in a three-phase balance. The segment-switching is conducted in each phase; depending on the receiver coil position, the segment of each phase is changed independently and sequentially. The mutual inductances between the transmitter phase coils vary significantly depending on the switching state. The proposed circuit configuration has resonant devices to cancel the variable mutual inductances for effective operation. The receiver position is estimated by the unbalanced current control. Experiments confirm that the method can perform a power transfer of 300W with a slight variation to a moving AGV, equipping the receiver at 30m/min as well as an AGV at a standstill.

38. Homogenized Finite Element Analysis for Multi-Turn Coils Considering Parasitic Capacitances

Abstract

This study presents a novel homogenized finite element (FE) method that can consider parasitic capacitances of the multi-turn coils. In this method, a total capacitance, describing the parasitic capacitances, is parallel to the homogenized region, and the resonant peak is identified only by solving the electrostatic and magnetostatic FE analysis. The proposed method is applied to the solenoid and planar coil models, and it is confirmed that the impedance characteristics including the resonant peaks can be represented. Moreover, the computing cost is shown to be significantly lower than that of the conventional full-wave FE analysis.

43. Insulin-Deficient Diabetic Condition Upregulates the Insulin-Secreting Capacity of Human Induced Pluripotent Stem Cell-Derived Pancreatic Endocrine Progenitor Cells After Implantation in Mice

Abstract

The host environment is a crucial factor for considering the transplant of stem cell–derived immature pancreatic cells in patients with type 1 diabetes. Here, we investigated the effect of insulin (INS)-deficient diabetes on the fate of immature pancreatic endocrine cell grafts and the underlying mechanisms. Human induced pluripotent stem cell–derived pancreatic endocrine progenitor cells (EPCs), which contained a high proportion of chromogranin A⁺ NK6 homeobox 1⁺ cells and very few INS⁺ cells, were used. When the EPCs were implanted under the kidney capsule in immunodeficient mice, INS-deficient diabetes accelerated increase in plasma human C-peptide, a marker of graft-derived INS secretion. The acceleration was suppressed by INS infusion but not affected by partial attenuation of hyperglycemia by dapagliflozin, an INS-independent glucose-lowering agent. Immunohistochemical analyses indicated that the grafts from diabetic mice contained more endocrine cells including proliferative INS-producing cells compared with that from nondiabetic mice, despite no difference in whole graft mass between the two groups. These data suggest that INS-deficient diabetes upregulates the INS-secreting capacity of EPC grafts by increasing the number of endocrine cells including INS-producing cells without changing the graft mass. These findings provide useful insights into postoperative diabetic care for cell therapy using stem cell–derived pancreatic cells.

Abstract

The differentiation of pancreatic endocrine cells from human pluripotent stem cells has been thoroughly investigated for their application in cell therapy against diabetes. Although non-endocrine cells are inevitable contaminating by-products of the differentiation process, a comprehensive profile of such cells is lacking. Therefore, we characterized non-endocrine cells in iPSC-derived pancreatic islet cells (iPIC) using single-cell transcriptomic analysis. We found that non-endocrine cells consist of (1) heterogeneous proliferating cells, and (2) cells with not only pancreatic traits but also liver or intestinal traits marked by FGB or AGR2. Non-endocrine cells specifically expressed FGFR2, PLK1, and LDHB. We demonstrated that inhibition of pathways involving these genes selectively reduced the number of non-endocrine cells in the differentiation process. These findings provide useful insights into cell purification approaches and contribute to the improvement of the mass production of endocrine cells for stem cell-derived cell therapy for diabetes.
