Alpha elastic scattering angular distributions of the 106Cd(alpha,alpha)106Cd reaction were measured at three energies around the Coulomb barrier to provide a sensitive test for the alpha + nucleus optical potential parameter sets. Furthermore, the new high precision angular distributions, together with the data available from the literature were used to study the energy dependence of the locally optimized {\alpha}+nucleus optical potential in a wide energy region ranging from E_Lab = 27.0 MeV down to 16.1 MeV. The potentials under study are a basic prerequisite for the prediction of alpha-induced reaction cross sections and thus, for the calculation of stellar reaction rates used for the astrophysical gamma process. Therefore, statistical model predictions using as input the optical potentials discussed in the present work are compared to the available 106Cd + alpha cross section data.

The cross sections for the elastic scattering reactions {112,124}Sn(a,a){112,124}Sn at energies above and below the Coulomb barrier are presented and compared to predictions for global alpha-nucleus potentials. The high precision of the new data allows a study of the global alpha-nucleus potentials at both the proton and neutron-rich sides of an isotopic chain. In addition, local alpha-nucleus potentials have been extracted for both nuclei, and used to reproduce elastic scattering data at higher energies. Predictions from the capture cross section of the reaction 112Sn(a,g)116Te at astrophysically relevant energies are presented and compared to experimental data., Comment: 20 pages, 10 figures, accepted for publication in Phys. The elastic scattering cross section of$^{92}$Mo($\alpha$,$\alpha$)$^{92}$Mo has been measured at energies of $E_{\rm{c.m.}} \approx$ 13, 16, and 19 MeV in a wide angular range. The real and imaginary parts of the optical potential for the system $^{92}$Mo - $\alpha$ have been derived at energies around and below the Coulomb barrier. The result fits into the systematic behavior of $\alpha$-nucleus folding potentials. The astrophysically relevant $^{96}$Ru($\gamma$,$\alpha$)$^{92}$Mo reaction rates at $T_9=2.0$ and $T_9=3.0$ could be determined to an accuracy of about 16 % and are compared to previously published theoretical rates., Comment: accepted for Physical Review C

The role of natural killer group 2D (NKG2D) in peripheral T cells as a costimulatory receptor is well established. However, its contribution to T cell thymic education and functional imprint is unknown. Here, we report significant changes in development, receptor signaling, transcriptional program, and function in T cells from mice lacking NKG2D signaling. In C57BL/6 (B6) and OT-I mice, we found that NKG2D deficiency results in Vβ chain usage changes and stagnation of the double-positive stage in thymic T cell development. We found that the expression of CD5 and CD45 in thymocytes from NKG2D deficient mice were reduced, indicating a direct influence of NKG2D on the strength of T cell receptor (TCR) signaling during the developmental stage of T cells. Depicting the functional consequences of NKG2D, peripheral OT-I NKG2D-deficient cells were unresponsive to ovalbumin peptide stimulation. Paradoxically, while αCD3/CD28 agonist antibodies led to phenotypic T cell activation, their ability to produce cytokines remained severely compromised. We found that OT-I NKG2D-deficient cells activate STAT5 in response to interleukin-15 but were unable to phosphorylate ERK or S6 upon TCR engagement, underpinning a defect in TCR signaling. Finally, we showed that NKG2D is expressed in mouse and human thymic T cells at the double-negative stage, suggesting an evolutionarily conserved function during T cell development. The data presented in this study indicate that NKG2D impacts thymic T cell development at a fundamental level by reducing the TCR threshold and affecting the functional imprint of the thymic progeny. In summary, understanding the impact of NKG2D on thymic T cell development and TCR signaling contributes to our knowledge of immune system regulation, immune dysregulation, and the design of immunotherapies. For the determination of the $^{144}Sm-\alpha$ optical potential we measured the angular distribution of $^{144}Sm(\alpha,\alpha)^{144}Sm$ scattering at the energy $E_{lab} = 20 MeV$ with high accuracy. 10. Memory CD4 T cell-derived IL-2 synergizes with viral infection to exacerbate lung inflammation.

Defining the most penetrating correlates of protective memory T cells is key for designing improved vaccines and T cell therapies. Here, we evaluate how interleukin (IL-2) production by memory CD4 T cells, a widely held indicator of their protective potential, impacts immune responses against murine influenza A virus (IAV). Unexpectedly, we show that IL-2-deficient memory CD4 T cells are more effective on a per cell basis at combating IAV than wild-type memory cells that produce IL-2. Improved outcomes orchestrated by IL-2-deficient cells include reduced weight loss and improved respiratory function that correlate with reduced levels of a broad array of inflammatory factors in the infected lung. Blocking CD70-CD27 signals to reduce CD4 T cell IL-2 production tempers the inflammation induced by wild-type memory CD4 T cells and improves the outcome of IAV infection in vaccinated mice. Finally, we show that IL-2 administration drives rapid and extremely potent lung inflammation involving NK cells, which can synergize with sublethal IAV infection to promote acute death. These results suggest that IL-2 production is not necessarily an indicator of protective CD4 T cells, and that the lung environment is particularly sensitive to IL-2-induced inflammation during viral infection.

IL-2 deficient cells are more effective than wild-type memory cells on a per cell basis at combating IAV and drive tempered early innate inflammatory responses. Our studies define a clear and surprising role for IL-2 as a cytokine adjuvant within the lung that can synergize with virus driven acute inflammatory responses to cause morbidity during sublethal respiratory viral infection.

- Abstract
BACKGROUND The potential benefit of dual antiplatelet therapy beyond 1 year after a myocardial infarction has not been established. We investigated the efficacy and safety of ticagrelor, a P2Y(12) receptor antagonist with established efficacy after an acute coronary syndrome, in this context. METHODS We randomly assigned, in a double-blind 1: 1: 1 fashion, 21,162 patients who had had a myocardial infarction 1 to 3 years earlier to ticagrelor at a dose of 90 mg twice daily, ticagrelor at a dose of 60 mg twice daily, or placebo. All the patients were to receive low-dose aspirin and were followed for a median of 33 months. The primary efficacy end point was the composite of cardiovascular death, myocardial infarction, or stroke. The primary safety end point was Thrombolysis in Myocardial Infarction (TIMI) major bleeding. RESULTS The two ticagrelor doses each reduced, as compared with placebo, the rate of the primary efficacy end point, with Kaplan-Meier rates at 3 years of 7.85% in the group that received 90 mg of ticagrelor twice daily, 7.77% in the group that received 60 mg of ticagrelor twice daily, and 9.04% in the placebo group (hazard ratio for 90 mg of ticagrelor vs. placebo, 0.85; 95% confidence interval [CI], 0.75 to 0.96; P = 0.008; hazard ratio for 60 mg of ticagrelor vs. placebo, 0.84; 95% CI, 0.74 to 0.95; P = 0.004). Rates of TIMI major bleeding were higher with ticagrelor (2.60% with 90 mg and 2.30% with 60 mg) than with placebo (1.06%) (P < 0.001 for each dose vs. placebo); the rates of intracranial hemorrhage or fatal bleeding in the three groups were 0.63%, 0.71%, and 0.60%, respectively. CONCLUSIONS In patients with a myocardial infarction more than 1 year previously, treatment with ticagrelor significantly reduced the risk of cardiovascular death, myocardial infarction, or stroke and increased the risk of major bleeding. Alpha elastic scattering angular distributions of the Cd-106(alpha, alpha)Cd-106 reaction were measured at three energies around the Coulomb barrier to provide a sensitive test for the alpha + nucleus optical potential parameter sets. Furthermore, the new high precision angular distributions, together with the data available from the literature were used to study the energy dependence of the locally optimized alpha + nucleus optical potential in a wide energy region ranging from E-Lab = 27.0 MeV down to 16.1 MeV. The potentials under study are a basic prerequisite for the prediction of alpha-induced reaction cross sections and thus, for the calculation of stellar reaction rates used for the astrophysical gamma process. Therefore, statistical model predictions using as input the optical potentials discussed in the present work are compared to the available Cd-106 + alpha cross section data. (C) 2015 Elsevier B.V. The U-233(d,pf)U-234 reaction has been studied with high energy resolution. The observed fission resonances in the excitation energy range of 4.75 less than or equal to E* less than or equal to 5.40 MeV were described as members of rotational bands with rotational parameters characteristic to the hyperdeformed nuclear shape ((h) over bar(2)/2 theta = 2.1 +/- 0.2 keV). Information on the K values of the bands has been obtained from fission fragment angular distribution measurements. The level density of the most strongly excited J = 3 states has been compared to the prediction of the back-shifted Fermi-gas formula and the energy of the ground state in the third minimum has been estimated to be E-III = 3.1 +/- 0.4 MeV

The excitation energy spectrum of fission resonances has been measured with high energy resolution using the U-235 (d,pf) reaction in order to study hyperdeformed (HD) rotational bands and HD nuclear shapes. The moments of inertia of the rotational bands and the energy of the ground state in the third minimum were determined. Another signature of these highly deformed states, their enhanced a decay, was also observed. By studying cold or compact fission in the Th-232(n,f) reaction around HD resonances, we obtained data for heavy clustering.

Sharp transmission resonances have been observed in the U-235(d, Pf) U-236 reaction. The shapes of the resonances as well as the angular distributions of the associated fission fragments are analyzed together with the data obtained previously for U-234. The resonances are interpreted as being hyperdeformed rotational bands with (h) over bar(2)/2 theta = 1.8(-0.7)(+0.5) and 1.6(-0.4)(+1.0) keV and K values of 1, 2, and 4 for U-234 and U-236, respectively.

Dimeric IgA (dIgA) can move through cells via the IgA/IgM polymeric immunoglobulin receptor (PIGR), which is expressed mainly on mucosal epithelia. Here, we studied the ability of dIgA to target commonly mutated cytoplasmic oncodrivers. Mutation-specific dIgA, but not IgG, neutralized KRASG12D within ovarian carcinoma cells and expelled this oncodriver from tumor cells. dIgA binding changed endosomal trafficking of KRASG12D from accumulation in recycling endosomes to aggregation in the early/late endosomes through which dIgA transcytoses. dIgA targeting of KRASG12D abrogated tumor cell proliferation in cell culture assays. In vivo , KRASG12D-specific dIgA1 limited the growth of KRASG12D-mutated ovarian and lung carcinomas in a manner dependent on CD8+ T cells. dIgA specific for IDH1R132H reduced colon cancer growth, demonstrating effective targeting of a cytoplasmic oncodriver not associated with surface receptors. dIgA targeting of KRASG12D restricted tumor growth more effectively than small-molecule KRASG12D inhibitors, supporting the potential of this approach for the treatment of human cancers. - Abstract
Ultraviolet-B light (UV-B) regulates the expression of genes in a wavelength- and fluence rate-dependent fashion. A signaling pathway consisting of CONSTITUTIVE PHOTOMORPHOGENESIS 1 (COP1) and UV RESISTANCE LOCUS 8 (UVR 8) mediates responsiveness to longer wavelength, low intensity UV-B light-activating, for example, HY5 gene expression. By contrast, transcription of another group of genes, including ANAC13, modulated by shorter wavelength, higher intensity UV-B is controlled by a yet unknown and largely COP1-independent signaling cascade. Here we provide evidence by promoter deletion analysis, and characterization of genetic mutants displaying aberrant expression patterns, that two cis-regulatory elements, designated MRE(ANAC13) and UVBox(ANAC13), are required for maximal UV-B induction of the ANAC13 gene in transgenic plants. These elements are located in the proximal 150-bp region of the ANAC13 promoter. They show no significant similarity to each other; the putative MRE(ANAC13) (-AACCTT-) is closely related to MRE(CHS) (-AACCTA-) found in the CHALCONE SYNTHASE (CHS) gene, whereas UVBox(ANAC13) (with core sequence CAAG) represents a novel cis-regulatory element. The novel UVBox(ANAC13) sequence is significantly enriched in the promoter region of a subset of UV-B-induced genes with similar activation properties as ANAC13. In addition, we demonstrate that expression of a chimeric gene containing only the dimerized 12-mer containing UVBox(ANAC13) fused to a minimal CaMV35S promoter/luciferase reporter is (i) efficiently induced by shorter wavelength, higher intensity UV-B, but (ii) does not respond either to longer wavelength UV-B and red light or (iii) to abscisic acid treatment and osmotic, salt, heat and cold stresses.
