Abstract: OBJECTIVES: To evaluate the association between the pre-extubation sum of eye and motor components of the Glasgow Coma Score (GCS-EM) and odds of extubation failure in patients with acute brain injury being liberated from mechanical ventilation. DESIGN: Secondary analysis of a prospective, multicenter observational study (ClinicalTrials.gov identifier NCT03400904). SETTING: Sixty-three hospital sites worldwide, with patient recruitment from January 2018 to November 2020. PATIENTS: One thousand one hundred fifty-two critically ill patients with acute brain injury, with a median age of 54 years, of whom 783 (68.0%) were male, 559 (48.5%) had traumatic brain injury, and 905 (78.6%) had a GCS-EM greater than 8 before extubation (scores range from 2 to 10). INTERVENTIONS: None. MEASUREMENTS AND MAIN RESULTS: GCS-EM was computed in intubated patients on the day of extubation. The main outcome was extubation failure, defined as unplanned reintubation within 5 days of extubation. Analyses used multilevel logistic regression with adjustment for patient characteristics and a random intercept for hospital site. In the primary analysis, GCS-EM was not associated with extubation failure (odds ratio, 1.07 per additional point; 95% CI, 0.87-1.31). Findings were consistent in sensitivity analyses that: 1) used different adjustment covariates, 2) included a verbal estimate to derive an overall GCS, 3) accounted for missing data, 4) considered a 2-day time interval to define extubation failure, 5) accounted for competing risks, and 6) used a propensity score-based model. There was no association between GCS-EM and extubation outcome in subgroups defined by brain injury diagnosis or age. CONCLUSIONS: In this large, contemporary, multicenter cohort of patients with acute brain injury, we found no association between the GCS-EM and odds of extubation failure. Abstract: Purpose: The use of arterial partial pressure of carbon dioxide (PaCO2) as a target intervention to manage elevated intracranial pressure (ICP) and its effect on clinical outcomes remain unclear. We aimed to describe targets for PaCO2 in acute brain injured (ABI) patients and assess the occurrence of abnormal PaCO2 values during the first week in the intensive care unit (ICU). The secondary aim was to assess the association of PaCO2 with in-hospital mortality. Methods: We carried out a secondary analysis of a multicenter prospective observational study involving adult invasively ventilated patients with traumatic brain injury (TBI), subarachnoid hemorrhage (SAH), intracranial hemorrhage (ICH), or ischemic stroke (IS). PaCO2 was collected on day 1, 3, and 7 from ICU admission. Normocapnia was defined as PaCO2 > 35 and to 45 mmHg; mild hypocapnia as 32–35 mmHg; severe hypocapnia as 26–31 mmHg, forced hypocapnia as < 26 mmHg, and hypercapnia as > 45 mmHg. Results: 1476 patients (65.9% male, mean age 52 ± 18 years) were included. On ICU admission, 804 (54.5%) patients were normocapnic (incidence 1.37 episodes per person/day during ICU stay), and 125 (8.5%) and 334 (22.6%) were mild or severe hypocapnic (0.52 and 0.25 episodes/day). Forced hypocapnia and hypercapnia were used in 40 (2.7%) and 173 (11.7%) patients. PaCO2 had a U-shape relationship with in-hospital mortality with only severe hypocapnia and hypercapnia being associated with increased probability of in-hospital mortality (omnibus p value = 0.0009). Important differences were observed across different subgroups of ABI patients. Conclusions: Normocapnia and mild hypocapnia are common in ABI patients and do not affect patients' outcome. Extreme derangements of PaCO2 values were significantly associated with increased in-hospital mortality.

Abstract: Background: There is insufficient evidence to guide ventilatory targets in acute brain injury (ABI). Recent studies have shown associations between mechanical power (MP) and mortality in critical care populations. We aimed to describe MP in ventilated patients with ABI, and evaluate associations between MP and clinical outcomes. Methods: In this preplanned, secondary analysis of a prospective, multi-center, observational cohort study (ENIO, NCT03400904), we included adult patients with ABI (Glasgow Coma Scale ≤ 12 before intubation) who required mechanical ventilation (MV) ≥ 24 h. Using multivariable log binomial regressions, we separately assessed associations between MP on hospital day (HD)1, HD3, HD7 and clinical outcomes: hospital mortality, need for reintubation, tracheostomy placement, and development of acute respiratory distress syndrome (ARDS). Results: We included 1217 patients (mean age 51.2 years [SD 18.1], 66% male, mean body mass index [BMI] 26.3 [SD 5.18]) hospitalized at 62 intensive care units in 18 countries. Hospital mortality was 11% (n = 139), 44% (n = 536) were extubated by HD7 of which 20% (107/536) required reintubation, 28% (n = 340) underwent tracheostomy placement, and 9% (n = 114) developed ARDS. The median MP on HD1, HD3, and HD7 was 11.9 J/min [IQR 9.2–15.1], 13 J/min [IQR 10–17], and 14 J/min [IQR 11–20], respectively. MP was overall higher in patients with ARDS, especially those with higher ARDS severity. After controlling for same-day pressure of arterial oxygen/fraction of inspired oxygen (P/F ratio), BMI, and neurological severity, MP at HD1, HD3, and HD7 was independently associated with hospital mortality, reintubation and tracheostomy placement. The adjusted relative risk (aRR) was greater at higher MP, and strongest for: mortality on HD1 (compared to the HD1 median MP 11.9 J/min, aRR at 17 J/min was 1.22, 95% CI 1.14–1.30) and HD3 (1.38, 95% CI 1.23–1.53), reintubation on HD1 (1.64; 95% CI 1.57–1.72), and tracheostomy on HD7 (1

Abstract: Background: In acute brain injury (ABI), the effects of hypoxemia as a potential cause of secondary brain damage and poor outcome are well documented, whereas the impact of hyperoxemia is unclear. The primary aim of this study was to assess the episodes of hypoxemia and hyperoxemia in patients with ABI during the intensive care unit (ICU) stay and to determine their association with in-hospital mortality. The secondary aim was to identify the optimal thresholds of arterial partial pressure of oxygen (PaO2) predicting in-hospital mortality. Methods: We conducted a secondary analysis of a prospective multicenter observational cohort study. Adult patients with ABI (traumatic brain injury, subarachnoid aneurysmal hemorrhage, intracranial hemorrhage, ischemic stroke) with available data on PaO2 during the ICU stay were included. Hypoxemia was defined as PaO2 < 80 mm Hg, normoxemia was defined as PaO2 between 80 and 120 mm Hg, mild/moderate hyperoxemia was defined as PaO2 between 121 and 299 mm Hg, and severe hyperoxemia was defined as PaO2 levels ≥ 300 mm Hg. Results: A total of 1,407 patients were included in this study. The mean age was 52 (±18) years, and 929 (66%) were male. Over the ICU stay, the fractions of patients in the study cohort who had at least one episode of hypoxemia, mild/moderate hyperoxemia, and severe hyperoxemia were 31.3%, 53.0%, and 1.7%, respectively. PaO2 values below 92 mm Hg and above 156 mm Hg were associated with an increased probability of in-hospital mortality. Differences were observed among subgroups of patients with ABI, with consistent effects only seen in patients without traumatic brain injury. Conclusions: In patients with ABI, hypoxemia and mild/moderate hyperoxemia were relatively frequent. Hypoxemia and hyperoxemia during ICU stay may influence in-hospital mortality. However, the small number of oxygen values collected represents a major limitation of the study.

Abstract: Purpose: Neurocritical care patients receive prolonged invasive mechanical ventilation (IMV), but there is poor specific information in this high-risk population about the liberation strategies of invasive mechanical ventilation. Methods: ENIO (NCT03400904) is an international, prospective observational study, in 73 intensive care units (ICUs) in 18 countries from 2018 to 2020. Neurocritical care patients with a Glasgow Coma Score (GCS) ≤ 12, receiving IMV ≥ 24 h, undergoing extubation attempt or tracheostomy were included. The primary endpoint was extubation failure by day 5. An extubation success prediction score was created, with 2/3 of patients randomly allocated to the training cohort and 1/3 to the validation cohort. Secondary endpoints were the duration of IMV and in-ICU mortality. Results: 1512 patients were included. Among the 1193 (78.9%) patients who underwent an extubation attempt, 231 (19.4%) failures were recorded. The score for successful extubation prediction retained 20 variables as independent predictors. The area under the curve (AUC) in the training cohort was 0.79 95% confidence interval (CI95) [0.71–0.87] and 0.71 CI95 [0.61–0.81] in the validation cohort. Patients with extubation failure displayed a longer IMV duration (14 [7–21] vs 6 [3–11] days) and a higher in-ICU mortality rate (8.7% vs 2.4%). Three hundred and nineteen (21.1%) patients underwent tracheostomy without extubation attempt. Patients with direct tracheostomy displayed a longer duration of IMV and higher in-ICU mortality than patients with an extubation attempt (success and failure). Conclusions: In neurocritical care patients, extubation failure is high and is associated with unfavourable outcomes. A score could predict extubation success in multiple settings. However, it will be mandatory to validate our findings in another prospective independent cohort.

Abstract: The shrinking kinetics of poly(N-isopropylacrylamide) (PNIPA) gels has been studied for two types of PNIPA gels prepared by (i) copolymerization of constituent monomer and cross-linker (monomer cross-linked gels) and (ii) γ-ray irradiation in the PNIPA solutions(polymer cross-linked gels) in order to investigate the role of cross-linking on shrinking kinetics. The shrinking kinetics of the monomer cross-linked gels is quite similar to that of the polymer cross-linked gels. For example, the rapid shrinking is attained by simply lowering the cross-linking density for both types of gels with a skin formation with skin thickness of ca. 3 μm. On the other hand, a significant difference was found when the microscopic structure and the dynamics were investigated by small-angle neutron scattering (SANS) and static/dynamic light scattering (SLS/DLS). The degree of built-in inhomogeneities and dynamic fluctuations were evaluated as a function of the cross-linking degree and the gel preparation temperature by intensity decomposition methods for both types of gels. It is concluded from the SANS and SLS/DLS results that the monomer cross-linked gels have extra built-in inhomogeneities due to the spatial distribution of cross-links in addition to the frozen concentration fluctuations inherent in polymer gels.

23. Swelling Behavior and Microstructure of Poly(12-acryloyloxydodecanoic acid-co-acrylic acid) Gels in the Ethanol/Water System

Abstract: The swelling behavior and hierarchical structures of poly(acrylic acid) (AA) homopolymer gels and poly(12-acryloyloxydodecanoic acid (ADA)-co-AA) gels carrying ionizable groups as well as hydrophobic groups have been investigated by equilibrium swelling measurement, dynamic light scattering (DLS), and small-angle neutron scattering (SANS). The equilibrium swelling degree (Sv) and the structure factors of the copolymer gels were obtained as a function of the ethanol (EtOH)/water (W) solvent composition, x (= vol % of EtOH). Sv showed a convex-upward function of x. That is, the gels swelled with increasing x until x = 50−60 vol % for AA gels and x = 70−80 vol % for ADA/AA gels and then decreased gradually. This phenomenon is explained by hydrophobic association of long alkyl chains of ADA and a cosolvency effect of poly(AA) in EtOH and W. The DLS results also indicated that the major component of light scattering was frozen inhomogeneities. SANS intensity functions, I(q), for the copolymer gels when x = 25 vol % showed the presence of an ordered structure in water with a long spacing of ca. 46 Å. These experimental pieces of evidence suggest a strong correlation between the macroscopic properties, such as Sv, and the microscopic structure as well as the dynamics. The diffusion coefficients were obtained by DLS as well as SANS, of which physical meanings are discussed by comparing the x dependence of Sv.

Abstract: The roles of hydrophobic ionizable side chains in amphiphilic copolymer gels consisting of 12-acryloyloxydodecanoic acid (ADA, hydrophobic) and acrylic acid (AA, hydrophilic) (ADA/AA) have been investigated by small-angle neutron scattering (SANS). The equilibrium swelling ratio and the structure factor of copolymer gels were obtained as a function of the solvent composition of two mixed solvent systems consisting of water and organic solvents. Propionic acid and 1-propanol were chosen because these solvents have similar values of log P, where P is the partition equilibrium coefficient with respect to octanol/water. This choice made it possible to suppress hydrophobicity effects and to illuminate hydrogen-bonding effects. The ADA/AA gels exhibited different swelling behaviors and disorganization processes of organized structures depending on the kind of mixed solvents in the concentration range of the organic component from x = 2−15 mol %, where x is the mol % of organic solvent in water. SANS results indicated the presence of an organized structure for the ADA/AA gel in water (x = 0 mol %) and up to x = 5 mol % with a long spacing of ca. 55 Å. The organized structure was disorganized by further increase of x, depending on the kind of solvent. The roles of acidity as well as hydrophobicity on organization/disorganization of hydrophobically associated structure were discussed from the viewpoints of swelling behavior and microscopic structure.

Abstract: The 4 202 353 bp genome of the alkaliphilic bacterium Bacillus halodurans C-125 contains 4066 predicted protein coding sequences (CDSs), 2141 (52.7%) of which have functional assignments, 1182 (29%) of which are conserved CDSs with unknown function and 743 (18. 3%) of which have no match to any protein database. Among the total CDSs, 8.8% match sequences of proteins found only in Bacillus subtilis and 66.7% are widely conserved in comparison with the proteins of various organisms, including B.subtilis. The B. halodurans genome contains 112 transposase genes, indicating that transposases have played an important evolutionary role in horizontal gene transfer and also in internal genetic rearrangement in the genome. Strain C-125 lacks some of the necessary genes for competence, such as comS, srfA and rapC, supporting the fact that competence has not been demonstrated experimentally in C-125. There is no paralog of tupA, encoding teichuronopeptide, which contributes to alkaliphily, in the C-125 genome and an ortholog of tupA cannot be found in the B.subtilis genome. Out of 11 sigma factors which belong to the extracytoplasmic function family, 10 are unique to B. halodurans, suggesting that they may have a role in the special mechanism of adaptation to an alkaline environment.

Abstract: HepG2 cells, known to support the replication and virion formation of hepatitis B virus (HBV), were transfected with a cosmid constructed to contain 12 tandem head-to-tail repeats of the HBV genome for effective HBV genome expression. We detected previously identified RNAs of 3.3, 2.3, and 2.0 kilobases (kb) that code for core antigen, large surface antigen, and middle/major surface antigen, respectively. We also detected four additional RNAs of 2.1, 1.7, 1.1, and 0.7 kb [the lengths exclude the poly(A) tail]. S1 mapping and nucleotide sequencing data showed that the 2.1-kb RNA is a spliced RNA whose 5' and 3' ends are identical to those of the 3.3-kb RNA. The results suggest that the 2.1-kb RNA codes for an altered core antigen lacking the last amino acid, cysteine, and that expression of the 3.3-kb pregenomic RNA is regulated, at least in part, by splicing. The map positions of the 1.7- and 1.1-kb RNAs suggest that they code for the carboxyl-terminal portions of the putative polymerase, whereas the 0.7-kb RNA codes for the X protein.

Abstract: Background: There is insufficient evidence to guide ventilatory targets in acute brain injury (ABI). Recent studies have shown associations between mechanical power (MP) and mortality in critical care populations. We aimed to describe MP in ventilated patients with ABI, and evaluate associations between MP and clinical outcomes. Methods: In this preplanned, secondary analysis of a prospective, multi-center, observational cohort study (ENIO, NCT03400904), we included adult patients with ABI (Glasgow Coma Scale ≤ 12 before intubation) who required mechanical ventilation (MV) ≥ 24h. Using multivariable log binomial regressions, we separately assessed associations between MP on hospital day (HD)1, HD3, HD7 and clinical outcomes: hospital mortality, need for reintubation, tracheostomy placement, and development of acute respiratory distress syndrome (ARDS). Results: We included 1217 patients (mean age 51.2years [SD 18.1], 66% male, mean body mass index [BMI] 26.3 [SD 5.18]) hospitalized at 62 intensive care units in 18 countries. Hospital mortality was 11% (n = 139), 44% (n = 536) were extubated by HD7 of which 20% (107/536) required reintubation, 28% (n = 340) underwent tracheostomy placement, and 9% (n = 114) developed ARDS. The median MP on HD1, HD3, and HD7 was 11.9J/min [IQR 9.2-15.1], 13J/min [IQR 10-17], and 14J/min [IQR 11-20], respectively. MP was overall higher in patients with ARDS, especially those with higher ARDS severity. After controlling for same-day pressure of arterial oxygen/fraction of inspired oxygen (P/F ratio), BMI, and neurological severity, MP at HD1, HD3, and HD7 was independently associated with hospital mortality, reintubation and tracheostomy placement. The adjusted relative risk (aRR) was greater at higher MP, and strongest for: mortality on HD1 (compared to the HD1 median MP 11.9J/min, aRR at 17J/min was 1.22, 95% CI 1.14-1.30) and HD3 (1.38, 95% CI 1.23-1.53), reintubation on HD1 (1.64; 95% CI 1.57-1.72), and tracheostomy on HD7 (1.53; 95%CI 1.18-1.99). MP was associated with the development of moderate-severe ARDS on HD1 (2.07; 95% CI 1.56-2.78) and HD3 (1.76; 95% CI 1.41-2.22). Conclusions: Exposure to high MP during the first week of MV is associated with poor clinical outcomes in ABI, independent of P/F ratio and neurological severity. Potential benefits of optimizing ventilator settings to limit MP warrant further investigation.

Abstract: Purpose: Neurocritical care patients receive prolonged invasive mechanical ventilation (IMV), but there is poor specific information in this high-risk population about the liberation strategies of invasive mechanical ventilation. Methods: ENIO (NCT03400904) is an international, prospective observational study, in 73 intensive care units (ICUs) in 18 countries from 2018 to 2020. Neurocritical care patients with a Glasgow Coma Score (GCS) ≤ 12, receiving IMV ≥ 24h, undergoing extubation attempt or tracheostomy were included. The primary endpoint was extubation failure by day 5. An extubation success prediction score was created, with 2/3 of patients randomly allocated to the training cohort and 1/3 to the validation cohort. Secondary endpoints were the duration of IMV and in-ICU mortality. Results: 1512 patients were included. Among the 1193 (78.9%) patients who underwent an extubation attempt, 231 (19.4%) failures were recorded. The score for successful extubation prediction retained 20 variables as independent predictors. The area under the curve (AUC) in the training cohort was 0.79 95% confidence interval (CI95) [0.71–0.87] and 0.71 CI95 [0.61–0.81] in the validation cohort. Patients with extubation failure displayed a longer IMV duration (14 [7–21] vs 6 [3–11] days) and a higher in-ICU mortality rate (8.7% vs 2.4%). Three hundred and nineteen (21.1%) patients underwent tracheostomy without extubation attempt. Patients with direct tracheostomy displayed a longer duration of IMV and higher in-ICU mortality than patients with an extubation attempt (success and failure). Conclusions: In neurocritical care patients, extubation failure is high and is associated with unfavourable outcomes. A score could predict extubation success in multiple settings. However, it will be mandatory to validate our findings in another prospective independent cohort.

Abstract: An international workshop on "Adapting Agriculture to Climate Change and Air Pollution" took place at Nanjing University of Information Science and Technology, Nanjing, China, during 23-27 October, 2023. Experts working in various multi-disciplinary areas of agroecosystem and environmental research gathered for academic communication and discussions. Two discussion groups focused on "agriculture under air pollution and climate change: current challenges and priorities for the future" and "adapting agriculture to air pollution and climate change: current status and next steps." Insights derived from the discussions are summarized in this article and include opinions about current issues, knowledge gaps' identification, and potential priorities and actions that could be taken. The first discussion mainly addresses ozone impact estimates; ozone metrics for impact and risk assessments; ozone monitoring; air pollution impacts and policy; and the pivotal role of agriculture and consumer choices. The second discussion covers adaptation and mitigation; greenhouse gases and energy efficiency; concerns about the link between adaptation and mitigation; local food, planetary-health diets and carbon footprint; irrigation and climate change adaptation; scientific evidence and policy-making; air pollution and crop adaptation; machine learning and crop modeling; and challenges faced by smallholder farmers and large-scale enterprises. 39. Sublethal chemical stimulation of arthropod parasitoids and parasites of agricultural and environmental importance.

Abstract: An increasing number of studies have reported stimulation of various organisms in the presence of environmental contaminants. This has created a need to critically evaluate sublethal stimulation and hormetic responses of arthropod parasitoids and parasites following exposure to pesticides and other contaminants. Examining this phenomenon with a focus on arthropods of agricultural and environmental importance serves as the framework for this literature review. This review shows that several pesticides, with diverse chemical structures and different modes of action, applied individually or in combination at sublethal doses, commonly stimulate an array of arthropod parasitoids and parasites. Exposure at sublethal doses can enhance responses related to physiology (e.g., respiration, total lipid content, and total protein content), behavior (e.g., locomotor activity, antennal drumming frequency, host location, and parasitization), and fitness (longevity, growth, fecundity, population net and gross reproduction). Concordantly, the parasitic potential (e.g., infestation efficacy, parasitization rate, and parasitoid/parasite emergence) can be increased, and as a result host activities inhibited. There is some evidence illustrating hormetic dose-responses, but the relevant literature commonly included a limited number and range of doses, precluding a robust differentiation between sub- and superNOAEL (no-observed-adverse-effect level) stimulation. These results reveal a potentially significant threat to ecological health, through stimulation of harmful parasitic organisms by environmental contaminants, and highlight the need to include sublethal stimulation and hormetic responses in relevant ecological pesticide risk assessments. Abstract: A coronary artery fistula usually originates in the right coronary artery and often opens into the right ventricle. In approximately 50% of cases with a main pulmonary artery opening, aberrant blood vessels originate from both coronary arteries. Only a few cases of both coronary and bronchial artery-pulmonary artery fistulas have been reported. The patient was an 83-year-old man. Echocardiography showed severe aortic stenosis, while coronary angiography revealed aberrant vessels from both coronary arteries to the pulmonary artery. The right heart catheterization revealed a 26% left-to-right shunt ratio and a pulmonary/body blood flow ratio (Qp/Qs) of 1.36. MDCT scan confirmed that the aberrant vascular plexus originating from both coronary arteries was connected to the bronchial artery. We performed surgery on the patient, replacing the aortic valve and resecting the coronary arteriovenous fistulas. On the 11th postoperative day, the shunt had disappeared, as evidenced by a 1.2% left-toright shunt ratio and a Qp/Qs of the right heart catheterization of 1.02. The patient progressed uneventfully and was discharged on the 25th postoperative day.

41. Elevated ozone alters long-chain fatty acids in leaves of Japanese white birch saplings.

Abstract: Long-chain fatty acids (LCFAs) in leaves have attracted attention as nutritious phytochemicals and olfactory signals that influence the behavior and growth of herbivorous insects. In recognition of the negative effects of increasing tropospheric ozone (O 3 ) levels on plants, LCFAs can be altered through peroxidation by O 3 . However, how elevated O 3 changes the amount and composition of LCFAs in field-grown plants is still unknown. We investigated palmitic, stearic, oleic, linoleic, linolenic LCFAs in the two leaf types (spring and summer) and two stages (early and late stage after expansion) of Japanese white birch (Betula platyphylla var. japonica) after a multi-year O 3 exposure on the field. Summer leaves exhibited a distinct composition of LCFAs under elevated O 3 at the early stage, whereas both stages of spring leaves did not exhibit significant changes in LCFAs composition by elevated O 3 . In the spring leaves, the amounts of saturated LCFAs significantly increased at the early stage, however, the amount of total, palmitic, and linoleic acids at the late stage were significantly decreased by elevated O 3 . Summer leaves had a lower amount of all LCFAs at both leaf stages. Regarding the early stage of summer leaves, the lower amount of LCFAs under elevated O 3 was possibly due to O 3 -suppressed photosynthesis in the current spring leaves. Furthermore, the decrease ratio of spring leaves over time was significantly increased by elevated O 3 in all LCFAs, whereas summer leaves did not exhibit such an effect. 42. Plant-insect communication in urban forests: Similarities of plant volatile compositions among tree species (host vs. non-host trees) for alder leaf beetle Agelastica coerulea.

Abstract: Behavior of insects, such as pollination and grazing, is usually determined by biogenic volatile organic compounds (BVOCs). However, particularly in O 3 -polluted urban forests, the BVOCs-based plant-insect communication can be disrupted by the reaction of O 3 with leaf-emitted BVOCs, such as between Japanese white birch (Betula platyphylla var. japonica) and a leaf beetle (Agelastica coerulea). To understand plant-insect communication in O 3 -polluted environments, it is necessary to identify chemical species of BVOCs that contribute to attractiveness toward insects but are diminished by elevated O 3 . In this study, we conducted olfactory response tests and gas chromatography mass spectrometry (GC-MS) analyses to clarify whether there is a similarity of BVOC components among Betulaceae host trees that can explain the attraction of the stenophagous insect A. coerulea. The olfactory response tests indicated that Betulaceae host trees attract A. coerulea via leaf-emitted BVOCs, while there was no preference of the leaf beetles to non-host trees (Sorbus commixta and Morus bombycis). However, GC-MS analyses indicated that the composition of BVOC blends considerably differed among Betulaceae host trees, although alders (Alnus hirsuta and A. japonica) had a similar composition of BVOC blend in each season (June and September) during which the adult leaf beetle is active. A distinct characteristic of the emission from B. platyphylla was that 2-carene and limonene, which are O 3 -reactive species, were emitted with a high monoterpene ratio irrespective of the season. Thus, these volatiles and the blend could be expected to lead the disrupted communication found between B. platyphylla and A. coerulea under elevated O 3 in previous field studies. In addition, our results indicated that A. coerulea is attracted to more than one blend within Betulaceae host trees, suggesting that grazing damages can be affected by different host preferences and O 3 reactivity with specific BVOCs in the field. 43. Ozone disrupts the communication between plants and insects in urban and suburban areas: an updated insight on plant volatiles.

Abstract: Plant-insect interactions are basic components of biodiversity conservation. To attain the international Sustainable Development Goals (SDGs), the interactions in urban and in suburban systems should be better understood to maintain the health of green infrastructure. The role of ground-level ozone (O 3 ) as an environmental stress disrupting interaction webs is presented. Ozone mixing ratios in suburbs are usually higher than in the center of cities and may reduce photosynthetic productivity at a relatively higher degree. Consequently, carbon-based defense capacities of plants may be suppressed by elevated O 3 more in the suburbs. However, contrary to this expectation, grazing damages by leaf beetles have been severe in some urban centers in comparison with the suburbs. To explain differences in grazing damages between urban areas and suburbs, the disruption of atmospheric communication signals by elevated O 3 via changes in plant-regulated biogenic volatile organic compounds and long-chain fatty acids are considered. The ecological roles of plant volatiles and the effects of O 3 from both a chemical and a biological perspective are presented. Abstract: Japanese elm (Ulmus davidiana var. japonica) is a native species in cool-temperate forests in Japan. We investigated growth, physiological reactions, and leaf defense capacity of Japanese elm seedlings under nitrogen (N) loading (45.3 kg N ha -1 year -1 ) and seasonal insect dynamics in a free-air ozone (O 3 )-enriched environment (about 54.5 nmol O 3 mol -1 ) over a growing season. Higher leaf N content and lower condensed tannin content in the presence of N loading and lower condensed tannin content in elevated O 3 were observed, suggesting that both N loading and elevated O 3 decreased the leaf defense capacity and that N loading further enhanced the leaf quality as food resource of insect herbivores. Two major herbivores were observed on the plants, elm leaf beetle (Pyrrhalta maculicollis) and elm sawfly (Arge captiva). The peak number of observed insects was decreased by N loading. Visible foliar injury caused by N loading might directly induce the reduction of number of the observed elm sawfly individuals. While elevated O 3 slightly suppressed the chemical defense capacity, significantly lower number of elm leaf beetle was observed in elevated O 3 . We conclude that N loading and elevated O 3 can alter not only the leaf defense capacity of Japanese elm seedlings but also the dynamics of elm leaf beetle and sawfly herbivores.

Abstract: Subseafloor pelagic sediments with high concentrations of organic matter form habitats for diverse microorganisms. Here, we determined depth profiles of genes for SSU rRNA, mcrA, dsrA and amoA from just beneath the seafloor to 363.3 m below the seafloor (mbsf) using core samples obtained from the forearc basin off the Shimokita Peninsula. The molecular profiles were combined with data on lithostratigraphy, depositional age, sedimentation rate and pore-water chemistry. The SSU rRNA gene tag structure and diversity changed at around the sulfate-methane transition zone (SMTZ), whereas the profiles varied further with depth below the SMTZ, probably in connection with the variation in pore-water chemistry. The depth profiles of diversity and abundance of dsrA, a key gene for sulfate reduction, suggested the possible niche separations of sulfate-reducing populations, even below the SMTZ. The diversity and abundance patterns of mcrA, a key gene for methanogenesis/anaerobic methanotrophy, suggested a stratified distribution and separation of anaerobic methanotrophy and hydrogenotrophic or methylotrophic methanogensis below the SMTZ. Abstract: In bacteriophage lambda, formation of a transcriptional anti-termination complex involving the elongating RNA polymerase is mediated by the interaction of boxB RNA with the RNA-binding domain of the N protein (N peptide). In an attempt to understand the spatial requirements for boxB/N peptide interaction within the anti-termination complex, the effects of changes in the distance between boxA and boxB RNA, the length of the boxB stem, and the distance between the N peptide and remainder of the N protein were examined using a bacterial reporter system. It was found that the requirements for boxB stem length and the distance between N peptide and the remainder of N were optimized and strict. In contrast, replacement of the boxB/N interaction by heterologous RNA-peptide interactions appeared to relax the strict requirement for RNA stem length and the orientation of the RNA-binding peptide, presumably due to the absence of the cooperative interaction between boxB/N and the host factor NusA. In addition, the decrease in activity upon stem lengthening could be partially suppressed by simultaneous lengthening of the RNA spacer. A further understanding of the structural organization of the anti-termination complex may provide insights into how functional ribonucleoprotein complexes may be engineered.

Abstract: Detection and enumeration of microbial life in natural environments provide fundamental information about the extent of the biosphere on Earth. However, it has long been difficult to evaluate the abundance of microbial cells in sedimentary habitats because non-specific binding of fluorescent dye and/or auto-fluorescence from sediment particles strongly hampers the recognition of cell-derived signals. Here, we show a highly efficient and discriminative detection and enumeration technique for microbial cells in sediments using hydrofluoric acid (HF) treatment and automated fluorescent image analysis. Washing of sediment slurries with HF significantly reduced non-biological fluorescent signals such as amorphous silica and enhanced the efficiency of cell detachment from the particles. We found that cell-derived SYBR Green I signals can be distinguished from non-biological backgrounds by dividing green fluorescence (band-pass filter: 528/38 nm (center-wavelength/bandwidth)) by red (617/73 nm) per image. A newly developed automated microscope system could take a wide range of high-resolution image in a short time, and subsequently enumerate the accurate number of cell-derived signals by the calculation of green to red fluorescence signals per image. Using our technique, we evaluated the microbial population in deep marine sediments offshore Peru and Japan down to 365 m below the seafloor, which provided objective digital images as evidence for the quantification of the prevailing microbial life. Our method is hence useful to explore the extent of sub-seafloor life in the future scientific drilling, and moreover widely applicable in the study of microbial ecology.

Abstract: Clarifying the participation of oxidative stress among possible contributing factors in potassium bromate (KBrO(3))-induced carcinogenesis is of importance from the perspective of human health protection. In the present study, utilizing the antioxidative effects of alpha-tocopherol (alpha-TP) or sodium ascorbic acid (SAA) to attenuate oxidative stress, alterations in bromodeoxyuridine labeling indices (BrdU-LIs) and reporter gene mutations in kidneys of male and female gpt delta rats given KBrO(3) were examined. Five male and female gpt delta rats in each group were given KBrO(3) at a concentration of 500ppm in the drinking water for 9 weeks, with 1% of alpha-TP or SAA administered in the diet from 1 week prior to the KBrO(3) treatment until the end of the experiment. Increases in 8-hydroxydeoxyguanosine levels in kidney DNA of both sexes of rats given KBrO(3) were significantly inhibited by SAA, but not alpha-TP. While BrdU-LIs in the proximal tubules of female rats were also significantly reduced by SAA, those in the males and gpt mutant frequencies in kidney DNA of both sexes were not affected by SAA or alpha-TP. Immunohistochemical and Western blot analyses for alpha(2u)-globulin strongly suggested that induction of cell proliferation observed in the males might primarily result from accumulation of this protein, independent of oxidative stress. The overall data indicated that while oxidative stress well correlates with induction of cell proliferation in females, its role in males and in generation of in vivo mutagenicity by KBrO(3) in both sexes is limited.

Abstract: Almost 10,000 single nucleotide polymorphisms (SNPs) had been identified in the RT1 complex, the major histocompatibility complex of the rat, but less than approximately 0.5% have been characterized. In the context of the incomplete characterization of most SNPs, simple sequence length polymorphism (SSLP) marker development is still valuable for understanding the involvement of genes in the RT1 in controlling disease susceptibility, since SSLPs are user-friendly and cost-effective genetic markers in rat genome analysis. In this study, we developed a set of 67 SSLP markers, including 57 novel markers, to cover the entire RT1 complex and then created genetic profiles across 67 rat strains. These markers are located almost every 50 kb in the RT1 complex and show comparable polymorphism; the average number of alleles was 8.04 +/- 3.44 and the average polymorphic rate was 71 +/- 23%. Interestingly, markers failing to amplify polymerase chain reaction products were highly observed in all strains except for BN/SsNHsd, which suggests the existence of highly variable genomic sequences or genomic rearrangements in the RT1 region across rat strains. Based on the phylogenic tree and individual genotyping data, we identified 28 SSLP marker haplotypes in the RT1 region that roughly consisted of three genomic regions. These findings provided new insight into the genomic organization of the RT1 complex and we recognized the need of additional RT1 genome sequences in different strains. Owing to the accuracy and ease of determination, PCR-based SSLP genotyping could replace serological typing in genetic analyses and characterization of rat major histocompatibility.

Abstract: Renin, a major regulatory component of the renin-angiotensin system, plays a pivotal role in regulating blood pressure and electrolyte homeostasis and is predominantly expressed in the kidney. Several cAMP-responsive elements have been identified within renin gene promoters. Here, we study how 2 such elements, renin proximal promoter element-2 (RP-2) and overlapping cAMP and negative regulatory elements (CNRE), affect the transcriptional regulation of renin. We generated Tg mice (TgM) bearing BACs containing either WT or mutant RP-2 or CNRE, integrated at single chromosomal loci. Analysis of the TgM revealed that RP-2 was essential to basal promoter activity in the kidney, while renin mRNA levels did not significantly change in any tissues tested in the CNRE mutant TgM. To evaluate the physiological significance of these mutations, we used the BAC Tg to rescue hypotensive Renin-null mutant mice. As predicted, no renin expression was observed in the kidneys of RP-2 mutant/Renin-null compound mice, whereas renin expression in CNRE mutant compound mice was indistinguishable from that in control mice. Consistent with this, RP-2 mutant animals were hypotensive, while CNRE mutants had normal blood pressure. Thus, transcriptional regulation of renin expression via RP-2 but not CNRE is critical for blood pressure regulation by this gene.
