1. Speckle-illumination spatial frequency domain imaging with a stereo laparoscope for profile-corrected optical property mapping

Abstract

We introduce a compact, two-camera laparoscope that combines active stereo depth estimation and speckle-illumination spatial frequency domain imaging (si-SFDI) to map profile-corrected, pixel-level absorption and reduced scattering optical properties in tissues with complex geometries. Our approach uses multimode fiber-coupled laser illumination to generate high-contrast speckle patterns, requiring only two images for optical property estimation. We demonstrate 3D profilometry using active stereo from low-coherence RGB laser flood illumination, which corrects for measured intensity variations caused by object height and surface angle differences. Validation against conventional SFDI in phantoms and an in-vivo human finger study showed good agreement, with profile-correction improving accuracy for complex geometries. This stereo-laparoscopic implementation of si-SFDI provides a simple method to obtain accurate optical property maps through a laparoscope, potentially offering quantitative endogenous contrast for minimally invasive surgical guidance.

Abstract

We analyze JWST MIRI/MRS IFU observations of three Seyferts and showcase the intriguing polycyclic aromatic hydrocarbon (PAH) emission characteristics in regions of $\sim 500\,\rm pc$ scales over or around their active galactic nuclei (AGN). Combining the model predictions and the measurements of PAH features and other infrared emission lines, we find that the central regions containing a high fraction of neutral PAHs with small sizes, e.g., those in ESO137-G034, are in highly heated environments, due to collisional shock heating, with hard and moderately intense radiation fields. Such environments are proposed to be associated with inhibited growth or preferential erosion of PAHs, decreasing the average PAH size and the overall abundance of PAHs. We additionally find that the central regions containing a high fraction of ionized PAHs with large sizes, e.g., those in MCG-05-23-016, are likely experiencing severe photo-ionization because of the radiative effects from the radiative shock precursor besides the AGN. The severe photo-ionization can contribute to the ionization of all PAHs and further destruction of small PAHs. Overall, different Seyferts, even different regions in the same galaxy, e.g., those in NGC\,3081, can contain PAH populations of different properties. Specifically, Seyferts that exhibit similar PAH characteristics to ESO137-G034 and MCG-05-23-016 also tend to have similar emission line properties to them, suggesting that the explanations for PAH characteristics of ESO137-G034 and MCG-05-23-016 may also apply generally. Abstract

Utilizing JWST MIRI/MRS IFU observations of the kiloparsec scale central regions, we showcase the diversity of ionized gas distributions and kinematics in six nearby Seyfert galaxies included in the GATOS survey. Specifically, we present spatially resolved flux distribution and velocity field maps of six ionized emission lines covering a large range of ionization potentials ($15.8-97.1$ eV). Based on these maps, we showcase the evidence of ionized gas outflows in the six targets, and find some highly disturbed regions in NGC\,5728, NGC\,5506, and ESO137-G034. We propose AGN-driven radio jets plausibly play an important role in triggering these highly disturbed regions. With the outflow rates estimated based on [Ne~{\footnotesize V}] emission, we find the six targets tend to have ionized outflow rates converged to a narrower range than previous finding. Abstract

Aims: Though best known for its role in oxidative DNA damage repair, apurinic/apyrimidinic endonuclease 1 (APE1) is a multifunctional protein that regulates multiple host responses during oxidative stress, including the reductive activation of transcription factors. As knockout of the APE1-encoding gene, Apex1, is embryonically lethal, we sought to create a viable model with generalized inhibition of APE1 expression. Results: A hypomorphic (HM) mouse with decreased APE1 expression throughout the body was generated using a construct containing a neomycin resistance (NeoR) cassette knocked into the Apex1 site. Offspring were assessed for APE1 expression, breeding efficiency, and morphology with a focused examination of DNA damage in the stomach. Heterozygotic breeding pairs yielded 50% fewer HM mice than predicted by Mendelian genetics. APE1 expression was reduced up to 90% in the lungs, heart, stomach, and spleen. The HM offspring were typically smaller, and most had a malformed tail. Oxidative DNA damage was increased spontaneously in the stomachs of HM mice. Further, all changes were reversed when the NeoR cassette was removed. Primary gastric epithelial cells from HM mice differentiated more quickly and had more evidence of oxidative DNA damage after stimulation with Helicobacter pylori or a chemical carcinogen than control lines from wildtype mice. Innovation: A HM mouse with decreased APE1 expression throughout the body was generated and extensively characterized. Conclusion: The results suggest that HM mice enable studies of APE1's multiple functions throughout the body. Abstract

Urinary tract infections (UTIs) are a common condition that can lead to serious complications including kidney injury, altered mental status, sepsis, and death. Laboratory tests such as urinalysis and urine culture are the mainstays of UTI diagnosis, whereby a urine specimen is collected and processed to reveal its cellular and chemical composition. This process requires precise specimen collection, handling infectious human waste, controlled urine storage, and timely transportation to modern laboratory equipment for analysis. Holographic lens free imaging (LFI) can measure large volumes of urine via a simple and compact optical setup, potentially enabling automatic urine analysis at the patient bedside. We introduce an LFI system capable of resolving important urine clinical biomarkers such as red blood cells, white blood cells, crystals, casts, and E. Coli in urine phantoms. This approach is sensitive to the particulate concentrations relevant for detecting several clinical urine abnormalities such as hematuria, pyuria, and bacteriuria. We show bacteria concentrations across eight orders of magnitude can be estimated by analyzing LFI measurements. LFI measurements of blood cell concentrations are relatively insensitive to changes in bacteria concentrations of over seven orders of magnitude. Lastly, LFI reveals clear differences between UTI-positive and UTI-negative urine from human patients. Together, these results show promise for LFI as a tool for urine screening, potentially offering early, point-of-care detection of UTI and other pathological processes.

Abstract

During the second world war, Canada made several important contributions to the wartime work of the Manhattan Project. The three main contributions were: establishing a domestic nuclear research laboratory in Montreal to investigate heavy water reactors, creating supply chains to provide uranium oxide, heavy water and polonium to the Manhattan Project, and the direct contributions of several Canadians living the United States. These wartime efforts helped establish a legacy of nuclear research in Canada which has persisted to the present day.

Abstract

Abstract Introduction: Alleviation of symptom severity for major depressive disorder (MDD) is known to be associated with a lagged improvement of functioning. Pharmacotherapy guidelines support algorithms for MDD treatment. However, it is currently unclear whether concordance with guidelines influences functional recovery. A guideline concordance algorithm (GCA-8) was used to measure this pathway in a naturalistic clinical setting. Methods: Data from 1403 adults (67% female, 84% non-Hispanic/Latino White, mean age of 43 years) with nonpsychotic MDD from the Penn State Psychiatry Clinical Assessment and Rating Evaluation System registry (visits from 02/01/2015 to 04/13/2021) were evaluated. Multivariable linear regression measured associations between GCA-8 and World Health Organization Disability Assessment Schedule 2.0 (WHODAS) scores. Mediation by MDD symptom severity using the Patient Health Questionnaire depression module (PHQ-9) was also evaluated. Results: This study found a statistically significant improvement in WHODAS scores (functioning) between baseline and final measures (−2 points, P < .001) within one year. A one standard deviation increase in the GCA-8 score was associated with a 0.48-point reduction in mean disability score (total effect; P = .02) with significant mediation by the change in MDD symptom severity (coefficient = −0.51, P < .001) and a nonsignificant natural direct effect of the GCA-8 independent of PHQ-9 change (coefficient = −0.02, P = .92). Conclusions: Higher pharmacotherapy guideline concordance is associated with better functioning for MDD patients; this association likely occurs through improvement in MDD symptom severity rather than directly.

14. Rapid tissue oxygenation mapping from snapshot structured-light images with adversarial deep learning

Abstract

Spatial frequency domain imaging (SFDI) is a powerful technique for mapping tissue oxygen saturation over a wide field of view. However, current SFDI methods either require a sequence of several images with different illumination patterns or, in the case of single snapshot optical properties (SSOP), introduce artifacts and sacrifice accuracy. To avoid this tradeoff, we introduce OxyGAN: a data-driven, content-aware method to estimate tissue oxygenation directly from single structured light images using end-to-end generative adversarial networks. Conventional SFDI is used to obtain ground truth tissue oxygenation maps for ex vivo human esophagi, in vivo hands and feet, and an in vivo pig colon sample under 659 nm and 851 nm sinusoidal illumination. We benchmark OxyGAN by comparing to SSOP and to a two-step hybrid technique that uses a previously-developed deep learning model to predict optical properties followed by a physical model to calculate tissue oxygenation. When tested on human feet, a cross-validated OxyGAN maps tissue oxygenation with an accuracy of 96.5%. When applied to sample types not included in the training set, such as human hands and pig colon, OxyGAN achieves a 93.0% accuracy, demonstrating robustness to various tissue types. On average, OxyGAN outperforms SSOP and a hybrid model in estimating tissue oxygenation by 24.9% and 24.7%, respectively. Lastly, we optimize OxyGAN inference so that oxygenation maps are computed ~10 times faster than previous work, enabling video-rate, 25Hz imaging. Due to its rapid acquisition and processing speed, OxyGAN has the potential to enable real-time, high-fidelity tissue oxygenation mapping that may be useful for many clinical applications.

Abstract

Spatial Frequency Domain Imaging can map tissue scattering and absorption properties over a wide field of view, making it useful for clinical applications such as wound assessment and surgical guidance. This technique has previously required the projection of fully-characterized illumination patterns. Here, we show that random and unknown speckle illumination can be used to sample the modulation transfer function of tissues at known spatial frequencies, allowing the quantitative mapping of optical properties with simple laser diode illumination. We compute low- and high-spatial frequency response parameters from the local power spectral density for each pixel and use a look-up-table to accurately estimate absorption and scattering coefficients in tissue phantoms, in vivo human hand, and ex vivo swine esophagus. Because speckle patterns can be generated over a large depth of field and field of view with simple coherent illumination, this approach may enable optical property mapping in new form-factors and applications, including endoscopy.

Abstract

We present a deep learning framework for wide-field, content-aware estimation of absorption and scattering coefficients of tissues, called Generative Adversarial Network Prediction of Optical Properties (GANPOP). Spatial frequency domain imaging is used to obtain ground-truth optical properties from in vivo human hands, freshly resected human esophagectomy samples and homogeneous tissue phantoms. Images of objects with either flat-field or structured illumination are paired with registered optical property maps and are used to train conditional generative adversarial networks that estimate optical properties from a single input image. We benchmark this approach by comparing GANPOP to a single-snapshot optical property (SSOP) technique, using a normalized mean absolute error (NMAE) metric. In human gastrointestinal specimens, GANPOP estimates both reduced scattering and absorption coefficients at 660 nm from a single 0.2/mm spatial frequency illumination image with 58% higher accuracy than SSOP. When applied to both in vivo and ex vivo swine tissues, a GANPOP model trained solely on human specimens and phantoms estimates optical properties with approximately 43% improvement over SSOP, indicating adaptability to sample variety. Moreover, we demonstrate that GANPOP estimates optical properties from flat-field illumination images with similar error to SSOP, which requires structured-illumination. Given a training set that appropriately spans the target domain, GANPOP has the potential to enable rapid and accurate wide-field measurements of optical properties, even from conventional imaging systems with flat-field illumination.

Abstract

Abstract Volumetric muscle loss (VML) is associated with persistent functional impairment due to a lack of de novo muscle regeneration. As mechanisms driving the lack of regeneration continue to be established, adjunctive pharmaceuticals to address the pathophysiology of the remaining muscle may offer partial remediation. Studies were designed to evaluate the tolerance and efficacy of two FDA‐approved pharmaceutical modalities to address the pathophysiology of the remaining muscle tissue after VML injury: (1) nintedanib (an anti‐fibrotic) and (2) combined formoterol and leucine (myogenic promoters). Tolerance was first established by testing low‐ and high‐dosage effects on uninjured skeletal muscle mass and myofiber cross‐sectional area in adult male C57BL/6J mice. Next, tolerated doses of the two pharmaceutical modalities were tested in VML‐injured adult male C57BL/6J mice after an 8‐week treatment period for their ability to modulate muscle strength and whole‐body metabolism. The most salient findings indicate that formoterol plus leucine mitigated the loss in muscle mass, myofiber number, whole‐body lipid oxidation, and muscle strength, and resulted in a higher whole‐body metabolic rate (p ≤ 0.016); nintedanib did not exacerbate or correct aspects of the muscle pathophysiology after VML. This supports ongoing optimization efforts, including scale‐up evaluations of formoterol treatment in large animal models of VML.

Abstract

Utilizing JWST MIRI/Medium Resolution Spectrograph integral field unit observations of the kiloparsec-scale central regions, we showcase the diversity of ionized gas distributions and kinematics in six nearby Seyfert galaxies included in the GATOS survey. Specifically, we present spatially resolved flux distribution and velocity field maps of six ionized emission lines covering a large range of ionization potentials (15.8–97.1 eV). Based on these maps, we showcase the evidence of ionized gas outflows in the six targets, and find some highly disturbed regions in NGC 5728, NGC 5506, and ESO137-G034. We propose active galactic nucleus (AGN)-driven radio jets plausibly play an important role in triggering these highly disturbed regions. With the outflow rates estimated based on [Ne V ] emission, we find the six targets tend to have ionized outflow rates converged to a narrower range than the previous finding. These results have an important implication for the outflow properties in AGN of comparable luminosity.

Abstract

This project investigates how the goals of microenterprise entrepreneurs affect their use of communication genres. Although slide-based business pitches are key for traditional entrepreneurs, microenterprise entrepreneurs have little interest in investment. Therefore, acquiring customers through short elevator pitches takes this central position. This article also explores the social justice dimensions of microenterprise acceleration, finding that such organizations can provide important services in combating inequality. This project uses writing, activity, and genre research as a theoretical framework, and the research site is a microenterprise accelerator in Tacoma, Washington called Spaceworks Tacoma, which supports both lower-income and Black owners of small businesses. 26. Changes in Polar Lipid Composition in Balsam Fir during Seasonal Cold Acclimation and Relationship to Needle Abscission

Abstract

Needle abscission in balsam fir has been linked to both cold acclimation and changes in lipid composition. The overall objective of this research is to uncover lipid changes in balsam fir during cold acclimation and link those changes with postharvest abscission. Branches were collected monthly from September to December and were assessed for cold tolerance via membrane leakage and chlorophyll fluorescence changes at −5, −15, −25, −35, and −45 °C. Lipids were extracted and analyzed using mass spectrometry while postharvest needle abscission was determined gravimetrically. Cold tolerance and needle retention each significantly (p < 0.001) improved throughout autumn in balsam fir. There were concurrent increases in DGDG, PC, PG, PE, and PA throughout autumn as well as a decrease in MGDG. Those same lipids were strongly related to cold tolerance, though MGDG had the strongest relationship (R2 = 55.0% and 42.7% from membrane injury and chlorophyll fluorescence, respectively). There was a similar, albeit weaker, relationship between MGDG:DGDG and needle retention (R2 = 24.3%). Generally, a decrease in MGDG:DGDG ratio resulted in better cold tolerance and higher needle retention in balsam fir, possibly due to increased membrane stability. This study confirms the degree of cold acclimation in Nova Scotian balsam fir and presents practical significance to industry by identifying the timing of peak needle retention. It is suggested that MGDG:DGDG might be a beneficial tool for screening balsam fir genotypes with higher needle retention characteristics.

Abstract

A novel carbon-free silicon-doped niobium suboxide was synthesized and tested as an electrocatalyst support for fuel cell applications. The promising Nb0·24O0·46Si0.30 (NbOS) metal oxide support exhibits a low structural band-gap with significantly high electronic conductivity. Furthmore, the NbOS support shows high corrosion-resistance in acidic media at high potentials. Pt nanoparticles were dispersed over the NbOS support (Pt/NbOS) to create an electrocatalyst with excellent electroactivity towards the oxygen reduction reaction (ORR). Likewise, in situ electrochemical analysis, and impedance spectroscopy (EIS) confirmed remarkable durability of the Pt/NbOS catalyst when subjected to harsh accelerated stress testing (AST) condtions. The enhanced stability and electrocatalytic activity and of Pt/NbOS is attributed to the strong metal-support interaction (SMSI) between the Pt nanoparticles (NPs) and the conductive NbOS support, which were confirmed via XRD and XPS measurements.

Robert Oppenheimer’s thoughts, concerns, and intentions regarding the U.S. pursuit of thermonuclear weapons. This paper provides context surrounding the 1949 recommendation of the Atomic Energy Commission’s General Advisory Committee, chaired by Oppenheimer, against a crash program for hydrogen bombs, a recommendation that would be used against him in the 1954 decision to revoke his security clearance. An appendix reproduces the April 5, 2022, letter from Los Alamos National Laboratory directors to the secretary of energy. Abstract

The escalation of militarized interstate disputes (MIDs) into interstate wars has been studied extensively with opportunity and willingness frameworks. In this article, we conceptualize and operationalize proto-insurgencies as a civil war equivalent to MIDs. Just as most MIDs do not escalate into interstate war, most proto-insurgencies never make the key tactical transitions needed to produce the onset of civil war. We separately examine proto-insurgency formation and proto-insurgency escalation to onset. We find that proto-insurgency formation is associated with political persecution, such as denial of access to justice, regime corruption, and mid-range levels of repression. Our theoretical focus is on how state repressive violence creates diffusion effects that push proto-insurgencies toward the tactical transitions that enable the escalation to civil war. Proto-insurgency escalation to civil war onset is driven by high levels of state repressive violence directed against proto-insurgency and its area of origin. This leads to the dispersal of proto-insurgents and the displacement of surrounding populations, inducing the spread of state repression to a larger number of administrative units. This creates the foundation for guerrilla warfare and establishes the conditions for rebels to offer sanctuary to displaced persons in return for active support, marking the onset of civil war. Abstract

The objective of this study was to compare the effectiveness of a second tumor necrosis factor inhibitor (TNFi) versus a non‐TNFi biologic following discontinuation of a TNFi for patients with polyarticular‐course juvenile idiopathic arthritis (pJIA). Using the Childhood Arthritis and Rheumatology Research Alliance Registry, patients with pJIA who started receiving a second biologic following a first TNFi were identified. Patients were required to have no active uveitis on the index date and a visit six months after the index date. Outcome measures included Clinical Juvenile Arthritis Disease Activity Score with a maximum of 10 active joints (cJADAS10), cJADAS10 inactive disease (ID; ≤2.5) and cJADAS10 minimal disease activity (MiDA; ≤5). Multiple imputation was used to account for missing data. Adjusted odds ratios (aORs) were calculated using propensity score quintiles to compare outcomes at six months following second biologic initiation. There were 216 patients included, 84% initially received etanercept, and most patients stopped receiving it because of its ineffectiveness (74%). A total of 183 (85%) started receiving a second TNFi, and 33 (15%) started receiving a non‐TNFi. Adalimumab was the most common second biologic received (71% overall, 84% of second TNFi), and tocilizumab was the most common non‐TNFi second biologic received (9% overall, 58% of non‐TNFi). There was no difference between receiving TNFi versus non‐TNFi in cJADAS10 ID (29% vs 25%; aOR 1.23, 95% confidence interval [CI] 0.47–3.20) or at least MiDA (43% vs 39%; aOR 1.11, 95% CI 0.47–2.62) at six months. Most patients with pJIA started receiving TNFi rather than non‐TNFi as their second biologic, and there were no differences in disease activity at six months.

Abstract

We combine neutron scattering (INS) and NMR/NQR nuclear spin lattice relaxation rate data to deduce the existence of a new contribution to the magnetic response (dynamic susceptibility) in cuprate superconductors. This contribution, which has yet to be observed with INS, is shown to embody the magnetic pseudogap effects. As such, it explains the long-standing puzzle of pseudogap effects missing from cuprate INS data, dominated by stripe fluctuations, for the dynamic susceptibility at low energies. For La_(1.86)Sr_(0.14)CuO_(4) and YBa_(2)Ba_(3)O_(6.5), the new term is the chief contributor to nuclear spin lattice relaxation at T >> T_(c)., Comment: 11 pages, 2 figures.
