Five sherds of green glazed pottery excavated at Canosa (Apulia) in Italy have been analysed by scanning electron microscopy. The aim of the investigation was to determine the chemical composition of the glazes and thus obtain information on the methods used in their production. The glazes were all of the high-lead type, coloured green by the addition of copper. Intermediate layers, observed at the interface between the glaze and body and giving the appearance of an applied white slip, were the result of the crystallization of lead feldspar from the molten glaze. Non-calcareous clays were used in the production of the pottery bodies. Concentration profiles from the glaze exterior to the body suggested that the glazes were produced by applying a suspension consisting of lead oxide plus silica to the bodies. On the basis of the glaze and body compositions, it is suggested that the Canosa glazed pottery was produced locally. [ABSTRACT FROM AUTHOR]

Aims: Allergen immunotherapy aims to induce tolerance, which persists after its discontinuation, to targeted allergens. However, concern exists regarding the use of subcutaneous immunotherapy with whole extracts due to frequently reported events of anaphylactic reactions. Materials & methods: In this pharmacovigilance study, the authors assessed the safety of subcutaneous immunotherapy with the monomeric allergoid Lais-in using a database of adverse reactions documented in real-world postmarketing reports from 2010 to 2020. Results & conclusion: The results showed that more than 171,916 doses of Lais-in were administered from 2010 to 2020, resulting in five adverse drug reactions. Nonserious adverse events, including hives, eye irritation and skin reactions, were reported. These data indicate that monomeric allergoids prevent serious reactions to subcutaneous immunotherapy.

An experimental nano-filled coating, based on a fluorine resin containing SiO 2 nano-particles, was applied on calcareous stones, representative of materials used in buildings and monuments of the Mediterranean basin; for comparison purposes, two commercial products were applied on the same substrates. The efficacy of the protective treatments was assessed by analyzing different characteristics of the three experimental/commercial products, i.e., color changes and permeability to water vapor to evaluate the treatments' harmlessness; capillary water absorption and water stone contact angle to evaluate the protection against water ingress; oleophobicity of the treated surfaces and the behavior under staining by acrylic blue-colored spray paint and felt-tip marker to verify the anti-graffiti action. Finally, the properties of the treated stone surfaces were analyzed also after the application of pancreatin, used to simulate bird excreta (guano). The protective coatings were found to promote graffiti removal, reducing also the detrimental effects due to simulated guano. The experimental nano-filled product, in addition, was able to provide outstanding performance but using smaller amounts of product in comparison to commercial systems.

Titanium dioxide based nanocomposites for stone coating have been found to be promising in laboratory conditions to obtain manifold protective actions against pollution and weathering affecting the outdoor built heritage. Lasting performances in real conditions of these multifunctional coatings have been scarcely examined, although this is a key issue in evaluating their potential for applications in a real building context and their optimization. This paper illustrates a field study aimed at investigating simultaneous hydrophobic and self-cleaning effectiveness, on the medium-long run, of TiO 2 NPs/fluoropolymer coatings applied on a limestone. The samples coated with the nanocomposites were exposed for one year in an urban environment and their surface was monitored. Hydrophobic properties were checked through contact angle measurements and a capillary water absorption test, while self-cleaning efficiency was evaluated by a photodegradation test of Rhodamine B. Optical microscopy observations and colour measurements were also performed. In addition, the contents of Ti and water-soluble ions on the sample surfaces were determined by X-ray Fluorescence and ion chromatography, respectively. The overall findings showed that TiO 2 NPs did not affect the ability of the polymer to protect the stone surface against water penetration. The coatings were able to preserve the surfaces from dirt. However, photocatalytic efficiency progressively decreased, due to the loss of the photocatalyst from the coating surface, which may be attributed to a polymer modification by ageing. The embedding of nanosized titania within the polymer limited the adsorption and accumulation of soluble salt ions on the coated surface, which may increase the stone damage risk. The study supports that TiO 2 NPs embedded in a fluoropolymer host matrix to appropriate amounts may be a suitable option to obtain stone coatings with both barrier effects against water penetration into the stone and photocatalytic ability, and provides useful knowledge for the improvement of these nanocomposites., (Copyright © 2018 Elsevier B.V. We describe a family carrying a γ-globin gene deletion associated with an increase of Hb A2 level beyond the normal range. The family included the proband, his sister and their father, all with increased Hb A2 and normal Hb F levels. The proband and his sister showed borderline values of mean corpuscular volume (MCV) and reduced values of mean corpuscular hemoglobin (Hb) (MCH). The proband was referred to our Medical Genetics Service for preconception counseling together with his partner, a typical β-thalassemia (β-thal) carrier. The results were negative for the most frequent α-thalassemia (α-thal) mutations, and had no significant sequence variations of the coding sequences and promoter of the β- and δ-globin genes. Quantitative analysis by multiplex ligation-dependent probe amplification (MPLA) of the β-globin gene cluster detected a heterozygous deletion, ranging between 2.1 and 4.7 kb, in the proband, his sister and the father. The deletion involved the (G)γ gene and (G)γ-(A)γ intergenic region, whereas the 3' region of the (A)γ gene was preserved. A subsequent gap-polymerase chain reaction (gap-PCR) showed that a hybrid (GA)γ fusion gene was present. The deletion segregated with the elevation of Hb A2. The MLPA analysis of the β-globin gene cluster in 150 control alleles excluded a common polymorphism. Despite stronger evidence being needed, the described family suggests a possible role of this γ-globin gene deletion in contributing to Hb A2 elevation, possibly by altering the transcription regulation of the cluster. We propose γ-globin gene dosage analysis to be performed in patients with unexplained elevated Hb A2 levels.

A previously unreported β chain variant, Hb Belluno [β111(G13)Val→Gly;β133(H11)Val→Val (HBB: c.335T > G;402G > C)], was incidentally discovered in a woman suffering from diabetes, during glycated hemoglobin (Hb A1c) assay. Its presence was suspected because of a small abnormal peak with a retention time just shorter than that of normal Hb A1c. Standard high performance liquid chromatography (HPLC), capillary zone electrophoresis (CZE) and agarose gel electrophoresis did not allow to separate the variant from Hb A. The reversed phase HPLC of globin chains showed the presence of a heterozygous β-globin variant amounting to approximately 43.5% of the total β chains. Later, this variant was found in five other members of the same family and DNA sequencing analysis confirmed a β-globin gene mutation. The variant is clinically silent in all patients and showed a slight instability with both heat and isopropanol tests. The other three mutations at this locus also affect stability. Hemoglobin (Hb) variants may invalidate the results of Hb A1c analysis and could result in mismanagement of diabetes. A comment alerting the requesting clinician to the presence of the Hb variant must be appended to the Hb A1c result. Additionally, many Hb variants can be chromatographically and/or electrophoretically silent. Therefore, when the clinician suspects a variant Hb, it is not sufficient to get a negative response from an HPLC screening test to rule it out. A dialogue with the pathologist is essential, involving exchange of information and sharing a diagnostic work-up including surveys to assess Hb stability and oxygen affinity, as much as DNA sequencing.

Objectives: Mutations in the SLC26A4 gene (7q22.3-7q31.1) are considered one of the most common causes of genetic hearing loss. There are two clinical forms related to these mutations: syndromic and non-syndromic deafness. The first one is named Pendred Syndrome (PS) when deafness is associated with thyroid goiter; the second is called DFNB4, when no other symptoms are present. Both are transmitted as an autosomal recessive trait, but simple heterozygotes can develop both forms of deafness. Actually it is thought that Pendred Syndrome occurs when both alleles of SLC26A4 gene are mutated; DFNB4 seems due to monoallelic mutations. PS and DFNB4 can be associated with inner ear malformations. In most of the cases (around 80%), these consist in Enlarged Vestibular Aqueduct (EVA). EVA can also be present without SLC26A4 mutations. Understanding the role of new SLC26A4 variants should facilitate clinical assessment, as well as diagnostic and therapeutic approaches. This investigation aims to detect and report genetic causes of two unrelated Italian boys with hearing loss., Methods: Patients and family members underwent clinical, audiological and genetic evaluations. To identify genetic mutations, DNA sequencing of SLC26A4 gene (including all 21 exons, exon-intron boundaries and promoter region) was carried out., Results: Both probands were affected by congenital, progressive and fluctuating mixed hearing loss. Temporal bone imaging revealed a bilateral EVA with no other abnormalities in both cases. Probands were heterozygotes for previously undescribed mutations in the SLC26A4 gene: R409H/IVS2+1delG (proband 1) and L236P/K590X (proband 2). No other mutations were detected in GJB2, GJB6 genes or mitochondrial DNA (mit-DNA)., Conclusions: The IVS2+1delG and K590X mutations have not yet been described in literature but there is some evidence to suggest that they have a pathological role. The results underlined the importance of considering the complete DNA sequencing of the SLC26A4 gene for differential molecular diagnosis of deafness, especially in those patients affected by congenital, progressive and fluctuating mixed hearing loss with bilateral EVA., (Copyright © 2012 Elsevier Ireland Ltd. Polysensitization is quite frequent in allergic children and may cause difficulties for the allergist in prescribing allergen-specific immunotherapy. This study aimed at evaluating the clinical effectiveness of 1 year of sublingual immunotherapy (SLIT) in a cohort of Italian allergic children with polysensitization. This open study was performed on 51 polysensitized children (34 boys; mean age, 11.8 years; range, 5.2-17.7 years) with allergic rhinitis and/or mild to moderate asthma. All of them were treated with SLIT for 1 year. The kind and the number of prescribed allergen extracts, the type of diagnosis, the severity of symptoms, and the use of drugs were evaluated at baseline and after 1 year. The adverse events to SLIT were also evaluated. Forty-two children were treated with a single extract, four with two different extracts and three with a mix of allergens. SLIT treatment induced a significant reduction in the number of sensitizations (p = 0.018); significant improvement of allergic rhinitis classification and severity; significant reduction of ocular, nasal, and bronchial symptoms (p < 0.01 for all); and drugs use (p < 0.01 for all drugs). No systemic reactions to SLIT were observed. This open study provides evidence that polysensitization is not an obstacle for prescribing SLIT in polysensitized children. Indeed, SLIT efficacy on clinical parameters is significant after 1 year and the therapy is safe.

Hemophilia A is an X-linked recessive disorder resulting from deficiency of Factor VIII (F8C), an important protein in blood coagulation. A large number of disease producing mutations have been reported in the F8C gene. However, a comprehensive analysis of mutations is difficult to conduct due to the large gene size, its many scattered exons, and the high frequency of de novo mutations. In this study, we performed analysis using PCR, Conformation Sensitive Gel Electrophoresis (CSGE), Denaturing High Performance Liquid Chromatography (DHPLC) and direct sequencing. We found seven novel mutations causing severe, moderate and mild Hemophilia A: IVS14-1G>A, G458V, T1695S, L1758P, Q2311P, 1441delT, 1269-1271insA. At least four variants detected by DHPLC (IVS14-1G>A, Q2311P,&#95;R698W and D1241Q) were not detectable by CSGE., (Copyright 2002 Wiley-Liss, Inc.)
