Currently the demonstration of the presence of sites contaminated by organic compounds, and the importance of decontamination needs no introduction. Indeed, human activity is generating more and more contaminated soils and polycyclic aromatic hydrocarbons (PAH) are amongst the contaminants most frequently encountered. Although natural attenuation is often depreciated by people because associated with inaction of contaminated land owners, it might represent a valuable alternative to classical treatments such as physico-chemical treatments. Indeed, this technic aims at reducing the concentration of pollutants while minimizing human intervention and therefore costs. A misunderstanding of this method and a small amount of data available does not encourage its development in France. This present work proposes the results obtained during a long-term natural attenuation assessment in real climatic conditions. This study was performed on seventy-two off ground plots filled with contaminated soils from different former industrial sites (coking industry, manufactured gas plants) whose initial concentration of PAH varies between 380 and 2077 mg/kg. The impact diagnosis was made on this device after ten years under natural attenuation. This diagnosis included leached water analysis, soil PAHs concentrations, as well as evaluation of vegetation cover quality and quantity.

International audience; Le caractère épisodique de la pollution est rarement pris en compte dans les études de toxicité lorsqu'il s'agit d'évaluer la durée de vie des composants organiques soumis à des expositions toxiques. Cependant, l'exposition à la toxicité des composants en pratique met en évidence des niveaux d'exposition fluctuants au cours du temps. C'est dans l'optique d'exhiber la spécificité des durées de vie des composants exposés par intermittence à la toxicité que les expériences ont été menées au sein du LIEBE dans le cadre de la thèse de Beaunoir (2001) encadrée par J.-F. Masfaraud. Une étude menée au préalable nous permet d'affirmer que les modèles usuels de durées de vie accélérée ne sont pas adaptés à ce type de données. L'étude met en oeuvre un succession de périodes d'exposition à un composant toxique (le chrome) et de repos. Ainsi le composant biologique (ceriodaphnia dubia) est soumis dans un premier temps à un stress puis dans un second temps à une période de repos. Supposant sa durée de vie exponentielle, nous avons adapté le modèle de durée de vie accéléré (Bagdonavicius et al., 2000) soumis à un schéma de stress semblable à celui étudié dans Balakhrishnan (2009) mais dans le cas de censures par intervalles. L'estimateur des paramètres de deux lois exponentielles en jeu est obtenu par un maximum de vraisemblance, le calcul de sa loi permet ensuite d'obtenir un intervalle de confiance. Ce modèle est ensuite appliqué aux données réelles dans le cas premier d'une seule séquence exposition/repos.

International audience; In the assessment of genotoxic risk factors in the environment, the measurement of DNA adducts in aquatic organisms and in plants may have considerable implications. Using 32P-postlabelling, we have detected DNA adducts in the liver of carp (Chondrostoma nasus) from the River Rhône (France), both downstream and upstream from a polychlorinated biphenyl incineration plant. Some of the DNA adducts were specific to downstream fish, suggesting a differential pattern of exposure. We have also detected DNA damage in needles in a declining spruce forest. We found that, in the declining forest, the amounts of DNA adducts increase in relation to the degree of damage to the needles whereas, in a healthy forest, the levels of DNA adducts were low. We have also found DNA adducts in the leaves of hops grown in fields where heptachlor residues persisted.

The 7-ethoxyresorufine O-deethylase activity is a cytochrome P450-dependant monooxygenase whose induction in fish liver is considered as a biological marker of exposure to environmental contaminants such as polycyclic aromatic hydrocarbons (PAH), some polyhalogenated biphenyls, furans or dioxins. Since the EROD activity is catalysed by a cytochrome P450 (1A1 form) that is involved in the activation of PAH to genotoxic metabolites, the EROD activity level may be significant on a toxicological basis. This hypothesis has been tested in the rainbow trout (Oncorhynchus mykiss) exposed to the model PAH benzo(a)pyrene (B(a)P). The hepatic microsomal EROD activity level was found to be correlated to the level of DNA adducts generated by incubating microsomes with B(a)P and DNA. Following an intraperitoneal to (B(a)P, an increase in hepatic microsomal EROD activity level and a formation of liver DNA adducts were simultaneously observed. However, in a primary culture of trout hepatocytes exposed to B(a)P, a pre-exposure to an EROD inducer (β-naphthoflavone) failed to increase the DNA adduct level. Thus, the generalization of this result is questionable, since a variety of factors are involved in the formation and the persistence of DNA adducts. This difficulty has been especially noticed during a field study with nase (Chondrostoma nasus) from the river Rhône. Finally, EROD activity and DNA adducts should be considered as two complementary biological markers, accounting for the contamination of the environment respectively by inducer and genotoxic compounds., L'activité 7-éthoxyrésorufine o-dééthylase (EROD) est une monooxygénase à cytochrome p450 dont l'induction au niveau hépatique chez les poissons constitue un marqueur biologique de la contamination des milieux aquatiques par des composés tels que les hydrocarbures aromatiques polycycliques (HAP) certains composésbiphényles, furanes ou dioxines polyhalogénés. Le niveau de l'activité EROD pourrait être significatif sur un plan toxicologique car elle est catalysée par un cytochrome p450 (forme 1A1) impliqué dans l'activation des HAP en métabolites génotoxiques.
