Abstract Land plants, especially those with significant woody biomass, represent the largest source of biomass on Earth, making the biodegradation of lignocellulosic materials critical to understanding the global carbon cycle. Cellulose, a major component of lignocellulose, is notoriously resistant to degradation due to its highly crystalline structure. While the degradation of cellulose by terrestrial microbes has been extensively studied, the mechanisms of cellulose degradation in deep-sea environments remain largely unexplored. The deep-sea ecosystem depends on organic matter, such as cellulose, that is synthesized in terrestrial environments and surface waters and descends to the deep sea. Recent studies suggest that a significant amount of cellulose is likely to reach the deep sea. Here, we present an in-depth study of cellulases from a novel deep-sea γ-proteobacterial strain TOYAMA8, isolated from Toyama Bay, Japan, using Surface Pitting Observation Technology (SPOT), a highly sensitive assay for enzymatic cellulose hydrolysis. The cellulases of strain TOYAMA8 show similarities to those of a previously reported deep-sea cellulolytic microbe, Marinagarivorans cellulosilyticus strain GE09. Genomic and transcriptomic analyses of these strains reveal novel cellulase genes and mechanisms that differ from terrestrial counterparts, shedding light on the unique adaptations of deep-sea microbes to recalcitrant biomass. In particular, these strains produce high-molecular-weight cellulases with unique domain architectures, likely optimized for membrane anchoring, which prevents enzyme diffusion and ensures efficient localized activity. Our findings provide critical insights into the microbial cellulose degradation in the deep sea, highlighting its role in the fate of organic carbon and the potential for biotechnological applications in biorefineries.

ABSTRACTDegradation of the appearance of rice grains due to high temperatures during ripening is a serious problem being worsened by global warming. QTLs for tolerance to high temperature during ripening must be identified in order to develop new cultivars efficiently. Here, we developed recombinant inbred lines (RILs) from crosses between the high-temperature-tolerant ‘Genkitsukushi’ and the high-temperature-sensitive ‘Tsukushiroman’. The occurrence of white-back grains is significantly lower in ‘Genkitsukushi’ than in ‘Tsukushiroman’. We evaluated their high-temperature tolerance and performed quantitative trait locus (QTL) analysis. A QTL for white-back grains on chromosome (chr.) 6 and a large-effect QTL on chr. 8 were identified in the two-year experiments. The QTL on chr. 8 was close to one we previously detected using RILs derived from ‘Chikushi 52’ × ‘Tsukushiroman’. The results indicate that the QTL on chr. 6 is unique to ‘Genkitsukushi’ × ‘Tsukushiroman’, and that on chr. 8 could be identical between ‘Genkitsukushi’ and ‘Chikushi 52’. Backcrossed lines which harbor the QTL region on chr. 8 of ‘Genkitsukushi’ in the background of ‘Tsukushiroman’ verified its effects. We also analyzed the marker genotypes near the QTLs on chr. 6 and 8 in relatives of ‘Genkitsukushi’. We discuss the interaction between these QTLs and their relationship with high-temperature tolerance.

The Bering Sea is the second largest marginal sea in the North Pacific and is one of the areas with highest biological productivity in high-latitude waters. The continental shelf of the Bering Sea hosts large populations of marine mammals and fishery resources. However, the smaller organisms in benthic ecosystems, including meiofauna, have been largely overlooked in this area, despite their potential importance in ecosystem functioning and the resultant biogeochemical cycles. This study analyzed spatial differences in the total abundance and community structure of the metazoan meiofauna at five stations around the Bering Canyon, located at the southeastern margin of the Bering Sea. Their association with environmental factors in sediments was also studied. The results confirmed that the investigated stations had meiofaunal standing stocks that were comparable to those of other Arctic seas. Among the investigated sediment biological and geochemical parameters (total organic carbon, median grain size, prokaryotic cell numbers, etc.), multivariate analyses showed that the C/N of organic matter in sediments was the main factor associated with meiofaunal community structure.

We, herein, report a 61-year-old male patient with amyotrophic lateral sclerosis (ALS) complicated pneumomediastinum while using mechanical insufflation-exsufflation (MI-E) after recovery from riluzole (RZ)-induced interstitial lung disease (RZ-ILD). After the treatment of RZ-ILD, he required non-invasive mechanical ventilation (NIV) at minimal pressure settings and MI-E to manage ALS-related breathing and airway-clearance issues, respectively. After a while, he developed progressive worsening dyspnoea, and chest computed tomography revealed extensive pneumomediastinum that had spread to the area surrounding the oesophagus, the retrosternal space, and the pericardial space. He was treated with immediate discontinuation of MI-E; however, he had to keep using NIV to support his severe respiratory muscle involvement. Pneumomediastinum gradually reduced in size and no recurrence of pneumomediastinum occurred. The clinical course of our patient suggests that excessive coughing associated with MI-E combined with his previous RZ-ILD, which potentially renders his lungs vulnerable to airway pressure, may have been the aetiological factors for secondary pneumomediastinum, i.e. barotrauma. Clinicians should be aware of the risk of pneumomediastinum while using MI-E in patients with ALS, who have other pre-existing risk factors for pneumomediastinum, such as drug-induced ILD in our case.

Background/Aim: Capecitabine plus oxaliplatin (CapeOX) therapy is used as an adjuvant chemotherapy regimen for patients with colorectal cancer (CRC). Although oxaliplatin induces thrombocytopenia, the risk factors for thrombocytopenia in oxaliplatin-treated patients with CRC are not well established. We aimed to investigate the risk factors for thrombocytopenia in CapeOX-treated patients with CRC. In addition, we evaluated platelet counts and noninvasive liver fibrosis indices, specifically the aspartate aminotransferase-to-platelet ratio index (APRI) and the fibrosis-4 index (FIB-4), during CapeOX therapy in these patients. Patients and Methods: Between July 2017 and June 2020, we enrolled CapeOX-treated patients with high-risk stage II or stage III CRC at seven hospitals collaborating with the Division of Oncology, Aichi Prefectural Society of Hospital Pharmacists (Aichi prefecture, Japan). In this retrospective study, we investigated patients' backgrounds, laboratory data, concomitant medications, number of cycles of CapeOX and oxaliplatin, cumulative dose of oxaliplatin, and administration period. The cut-off values were calculated using receiver operating characteristic analysis of platelet counts and APRI and FIB-4 scores. Results: Fiftyfive patients without thrombocytopenia and 44 patients with thrombocytopenia were enrolled. During CapeOX therapy, the thrombocytopenia group showed a significant decrease in platelet count and a significant increase in APRI and FIB- 4 scores compared to the non-thrombocytopenia group. Baseline albumin level ≤3.5 g/dl and platelet count ≤238 x 103/μl were independently associated with ≥grade 2 thrombocytopenia in CapeOX-treated patients. Conclusion: Baseline albumin level and platelet count may be useful for predicting thrombocytopenia in CapeOX-treated patients with high-risk stage II or stage III CRC. [ABSTRACT FROM AUTHOR]

Radiotherapy combined with temozolomide (TMZ+RT) is the primary treatment for high-grade glioma. TMZ is classified as a moderate emetic risk agent and, thus, supportive care for nausea and vomiting is important. In Nagoya University Hospital, all patients are treated with a 5-hydroxy-tryptamine 3 receptor antagonist (5-HT3RA) for the first 3 days. The daily administration of 5-HT3RA is resumed after the 4th day based on the condition of patients during TMZ+RT. Therefore, the present study investigated risk factors for nausea and vomiting in patients requiring the daily administration of 5-HT3RA. Patients with high-grade glioma who received TMZ+RT between January 2014 and December 2019 at our hospital were included. Patients were divided into two groups: a control group (patients who did not resume 5-HT3RA) and resuming 5-HT3RA group (patients who resumed 5-HT3RA after the 4th day), and both groups were compared to identify risk factors for nausea and vomiting during TMZ+RT. There were 78 patients in the control group (68%) and 36 in the resuming 5-HT3RA group (32%). A multivariate analysis of patient backgrounds in the two groups identified age <18 years, PS 2 or more, and occipital lobe tumors as risk factors for nausea and vomiting. Nausea and vomiting were attenuated in 30 patients (83%) in the resuming 5-HT3RA group following the resumption of 5-HT3RA. The results obtained highlight the importance of extracting patients with these risk factors before the initiation of therapy and the early resumption or daily administration of 5-HT3RA according to the condition of each patient. Carotenoids are used commercially for dietary supplements, cosmetics, and pharmaceuticals because of their antioxidant activity. In this study, colored microorganisms were isolated from deep sea sediment that had been collected from Suruga Bay, Shizuoka, Japan. One strain was found to be a pure yellow carotenoid producer, and the strain was identified as Sphingomonas sp. (Proteobacteria) by 16S rRNA gene sequence analysis; members of this genus are commonly isolated from air, the human body, and marine environments. The carotenoid was identified as nostoxanthin ((2,3,2′,3′)-β,β-carotene-2,3,2′,3′-tetrol) by mass spectrometry (MS), MS/MS, and ultraviolet-visible absorption spectroscopy (UV-Vis). Nostoxanthin is a poly-hydroxy yellow carotenoid isolated from some photosynthetic bacteria, including some species of Cyanobacteria. The strain Sphingomonas sp. SG73 produced highly pure nostoxanthin of approximately 97% (area%) of the total carotenoid production, and the strain was halophilic and tolerant to 1.5-fold higher salt concentration as compared with seawater. When grown in 1.8% artificial sea salt, nostoxanthin production increased by 2.5-fold as compared with production without artificial sea salt. These results indicate that Sphingomonas sp. SG73 is an efficient producer of nostoxanthin, and the strain is ideal for carotenoid production using marine water because of its compatibility with sea salt.

High-temperature stress during the grain-filling stage reduces grain quality of rice, and this is a serious problem in Japan, especially in the Kyushu region. To solve this problem, various heat-tolerant cultivars have been bred, such as ‘Nikomaru’, ‘Kumasannochikara’, ‘Genkitsukushi’, ‘Sagabiyori’, and ‘Otentosodachi’. When cultivated under high temperature after flowering, these heat-tolerant cultivars had lower percentages of chalky grains than in the heat-sensitive cultivar ‘Hinohikari’. All the heat-tolerant cultivars markedly decreased the nonstructural carbohydrate content in the stem under the high temperature compared to control condition during early grain-filling stage, which is considered to be a common trait of heat tolerance. Notably, ‘Sagabiyori’, ‘Genkitsukushi’, and ‘Nikomaru’ maintained a nucellar epidermis at 17 days after flowering (DAF) under high temperature, whereas the nucellar epidermis disappeared in ‘Hinohikari’. In addition, the expression of AGPS2b, thought to be a rate-limiting enzyme in starch synthesis, in ‘Kumasannochikara’, ‘Otentosodachi’, and ‘Nikomaru’ did not decrease under high temperature, whereas ‘Hinohikari’, ‘Sagabiyori’, and ‘Genkitsukushi’ could not maintain expression of the gene at 17 DAF. Moreover, the expression of Amy3E, a starch-degradation-related gene considered to induce grain chalkiness, in ‘Kumasannochikara’ at 17 DAF was not increased by high temperature. These results suggest that the heat-stress-tolerant cultivars have various mechanisms for dealing with high-temperature stress.

An 85-year-old man complained of a 2-month history of pain on the left side of his face. Brain computed tomography (CT) and magnetic resonance imaging/magnetic resonance angiography did not clearly show any intracranial abnormality and only showed fluid effusion in his left sphenoid sinus. Filamentous fungi were detected from the left sphenoid sinus specimen. The isolate was Scedosporium apiospermum. He was empirically treated with voriconazole, to which the isolate was susceptible. His consciousness decreased rapidly. Urgent 3D-CT angiography revealed an intracranial aneurysm near the left sphenoid sinus. Despite urgent coil embolization, the aneurysm ruptured, and he died.

Ten-Koh is a 23.5 kg, low-cost satellite developed to conduct space environment effects research in low-Earth orbit (LEO). Ten-Koh was developed primarily by students of the Kyushu Institute of Technology (Kyutech) and launched on 29 October 2018 on-board HII-A rocket F40, as a piggyback payload of JAXA’s Greenhouse gas Observing Satellite (GOSAT-2). The satellite carries a double Langmuir probe, CMOS-based particle detectors and a Liulin spectrometer as main payloads. This paper reviews the design of the mission, specifies the exact hardware used, and outlines the implementation and operation phases of the project. This work is intended as a reference that other aspiring satellite developers may use to increase their chances of success. Such a reference is expected to be particularly useful to other university teams, which will likely face the same challenges as the Ten-Koh team at Kyutech. Various on-orbit failures of the satellite are also discussed here in order to help avoid them in future small spacecraft. Applicability of small satellites to conduct space-weather research is also illustrated on the Ten-Koh example, which carried out simultaneous measurements with JAXA’s ARASE satellite.

Rice productivity is related to the ability of plants to adapt to heat stress. The heat-tolerant cultivars ‘Nikomaru’ and ‘Chikushi 64’ and heat-sensitive cultivar ‘Hinohikari’ were grown at 30ºC and 25ºC for 49 days after flowering. At 30ºC, only a few white immature kernels were produced in ‘Nikomaru’ and ‘Chikushi 64’, but about 22% of grains had immature kernels in ‘Hinohikari’. The high temperature(30ºC) caused no significant changes in grain dry weight, water content, and the NMR T1 value during the early ripening stage in ‘Nikomaru’ and ‘Chikushi 64’. It also did not affect grain development, especially with respect to the nucellar epidermis, in ‘Nikomaru’ and ‘Chikushi 64’, but caused clear cessation of development of the nucellar epidermis at 14 days after flowering in ‘Hinohikari’. In addition, high temperature decreased the amylose content and increased hardnessvs. adhesion ratio of cooked rice in both ‘Nikomaru’ and ‘Chikushi 64’ resulting a softer, less sticky texture, but not in ‘Hinohikari’. The maximum viscosity and breakdown values were increased, and final viscosity decreased at 30ºC in all three cultivars. These results suggested that starch in the endosperm of grains changed from a fluid state to a doughy state more slowly in ‘Nikomaru’ and ‘Chikushi 64’ than in ‘Hinohikari’, in which the water content and NMR relaxation time decreased, and transported assimilates accumulated slowly during grain development.
