Magnetic materials are essential for energy generation and information devices, and they play an important role in advanced technologies and green energy economies. Currently, the most widely used magnets contain rare earth (RE) elements. An outstanding challenge of notable scientific interest is the discovery and synthesis of novel magnetic materials without RE elements that meet the performance and cost goals for advanced electromagnetic devices. Here, we report our discovery and synthesis of an RE-free magnetic compound, Fe 3 CoB 2 , through an efficient feedback framework by integrating machine learning (ML), an adaptive genetic algorithm, first-principles calculations, and experimental synthesis. Magnetic measurements show that Fe 3 CoB 2 exhibits a high magnetic anisotropy ( K 1 = 1.2 MJ/m 3 ) and saturation magnetic polarization ( J s = 1.39 T), which is suitable for RE-free permanent-magnet applications. Our ML-guided approach presents a promising paradigm for efficient materials design and discovery and can also be applied to the search for other functional materials.

The genome is the ultimate architect of the brain. Its evolutionary variations build the neural circuits that endow each species with its innate senses and behaviors. A central question for neuroscience and translational medicine is whether neural circuits from two species can be made to function in an intact brain. Here, we establish genetic tools and use blastocyst complementation to selectively build and test interspecies neural circuits in rat-mouse brains. Despite ∼10-20 million years of evolution and prominent differences in brain size and cellular composition, rat pluripotent stem cells injected into mouse blastocysts widely populate and persist in the mouse brain. Unexpectedly, the mouse niche reprograms the birthdates of cortical and hippocampal rat neurons, where they also form synaptically active rat-mouse circuits. By genetically disabling host olfactory circuitry, we show that rat neurons restore synaptic information flow from the nose to the cortex. Rat neurons can also rescue a primal olfactory behavior (food-seeking), though less than mouse controls. By enabling a mouse to sense the world with rat neurons, we highlight the power of interspecies neural blastocyst complementation to uncover mechanisms of neural circuit development and evolution, and to inform efforts to rescue neural circuits affected by injury or disease.

Interspecies chimera formation with human pluripotent stem cells (PSCs) holds great promise to generate humanized animal models and provide donor organs for transplant. However, the approach is currently limited by low levels of human cells ultimately represented in chimeric embryos. Different strategies have been developed to improve chimerism by genetically editing donor human PSCs. To date, however, it remains unexplored if human chimerism can be enhanced in animals through modifying the host embryos. Leveraging the interspecies PSC competition model, here we discovered retinoic acid-inducible gene I (RIG-I)-like receptor (RLR) signaling, an RNA sensor, in “winner” cells plays an important role in the competitive interactions between co-cultured mouse and human PSCs. We found that genetic inactivation ofDdx58/Ifih1-Mavs-Irf7axis compromised the “winner” status of mouse PSCs and their ability to outcompete PSCs from evolutionarily distant species during co-culture. Furthermore, by usingMavs-deficient mouse embryos we substantially improved unmodified donor human cell survival. Comparative transcriptome analyses based on species-specific sequences suggest contact-dependent human-to-mouse transfer of RNAs likely plays a part in mediating the cross-species interactions. Taken together, these findings establish a previously unrecognized role of RNA sensing and innate immunity in “winner” cells during cell competition and provides a proof-of-concept for modifying host embryos, rather than donor PSCs, to enhance interspecies chimerism.

SUMMARYFaithful embryogenesis requires precise coordination between embryonic and extraembryonic tissues. Although stem cells from embryonic and extraembryonic origins have been generated for several mammalian species(Bogliotti et al., 2018; Choi et al., 2019; Cui et al., 2019; Evans and Kaufman, 1981; Kunath et al., 2005; Li et al., 2008; Martin, 1981; Okae et al., 2018; Tanaka et al., 1998; Thomson et al., 1998; Vandevoort et al., 2007; Vilarino et al., 2020; Yu et al., 2021b; Zhong et al., 2018), they are grown in different culture conditions with diverse media composition, which makes it difficult to study cross-lineage communication. Here, by using the same culture condition that activates FGF, TGF-β and WNT signaling pathways, we derived stable embryonic stem cells (ESCs), extraembryonic endoderm stem cells (XENs) and trophoblast stem cells (TSCs) from all three founding tissues of mouse and cynomolgus monkey blastocysts. This allowed us to establish embryonic and extraembryonic stem cell co-cultures to dissect lineage crosstalk during early mammalian development. Co-cultures of ESCs and XENs uncovered a conserved and previously unrecognized growth inhibition of pluripotent cells by extraembryonic endoderm cells, which is in part mediated through extracellular matrix signaling. Our study unveils a more universal state of stem cell self-renewal stabilized by activation, as opposed to inhibition, of developmental signaling pathways. The embryonic and extraembryonic stem cell co-culture strategy developed here will open new avenues for creating more faithful embryo models and developing more developmentally relevant differentiation protocols.

Understanding blastocyst formation and implantation is critical for improving farm animal reproduction but is hampered by a limited supply of embryos. We developed an efficient method to generate bovine blastocyst-like structures (termed blastoids) via the assembly of trophoblast stem cells and expanded potential stem cells. Bovine blastoids resemble blastocysts in morphology, cell composition, single-cell transcriptomes, and represent an accessible in vitro model for studying bovine embryogenesis.

Limited access to embryos has hampered the study of human embryogenesis and disorders that occur during early pregnancy. Human pluripotent stem cells provide an alternative means to study human development in a dish1–7. Recent advances in partial embryo models derived from human pluripotent stem cells have enabled human development to be examined at early post-implantation stages8–14. However, models of the pre-implantation human blastocyst are lacking. Starting from naive human pluripotent stem cells, here we developed an effective three-dimensional culture strategy with successive lineage differentiation and self-organization to generate blastocyst-like structures in vitro. These structures—which we term ‘human blastoids’—resemble human blastocysts in terms of their morphology, size, cell number, and composition and allocation of different cell lineages. Single-cell RNA-sequencing analyses also reveal the transcriptomic similarity of blastoids to blastocysts. Human blastoids are amenable to embryonic and extra-embryonic stem cell derivation and can further develop into peri-implantation embryo-like structures in vitro. Using chemical perturbations, we show that specific isozymes of protein kinase C have a critical function in the formation of the blastoid cavity. Human blastoids provide a readily accessible, scalable, versatile and perturbable alternative to blastocysts for studying early human development, understanding early pregnancy loss and gaining insights into early developmental defects. An in vitro culture strategy enables the generation of blastocyst-like structures termed human blastoids from naive human pluripotent stem cells, providing a model for studying human embryogenesis.

Cell competition involves a conserved fitness-sensing process during which fitter cells eliminate neighbouring less-fit but viable cells1. Cell competition has been proposed as a surveillance mechanism to ensure normal development and tissue homeostasis, and has also been suggested to act as a barrier to interspecies chimerism2. However, cell competition has not been studied in an interspecies context during early development owing to the lack of an in vitro model. Here we developed an interspecies pluripotent stem cell (PSC) co-culture strategy and uncovered a previously unknown mode of cell competition between species. Interspecies competition between PSCs occurred in primed but not naive pluripotent cells, and between evolutionarily distant species. By comparative transcriptome analysis, we found that genes related to the NF-κB signalling pathway, among others, were upregulated in less-fit ‘loser’ human cells. Genetic inactivation of a core component (P65, also known as RELA) and an upstream regulator (MYD88) of the NF-κB complex in human cells could overcome the competition between human and mouse PSCs, thereby improving the survival and chimerism of human cells in early mouse embryos. These insights into cell competition pave the way for the study of evolutionarily conserved mechanisms that underlie competitive cell interactions during early mammalian development. Suppression of interspecies PSC competition may facilitate the generation of human tissues in animals. Primed pluripotent stem cells from distant species compete with each other, and inactivation of NF-κB signalling in normally outcompeted human cells improves their survival and chimerism in mouse embryos.

New magnetic materials for energy and information-processing applications are of paramount importance in view of significant global challenges in environmental and information security. The discovery and design of materials requires efficient computational and experimental approaches for high throughput and efficiency. When increasingly powerful computational techniques are combined with special non-equilibrium fabrication methods, the search can uncover metastable compounds with desired magnetic properties. Here we review recent results on novel Fe-, Co- and Mn-rich magnetic compounds with high magnetocrystalline anisotropy, saturation magnetization, and Curie temperature created by combining experiments, adaptive genetic algorithm searches, and advanced electronic-structure computational methods. We discuss structural and magnetic properties of such materials including Co– and/or Fe–X compounds (X = N, Si, Sn, Zr, Hf, Y, C, S, Ti, or Mn), and their prospects for practical applications.

The recent development of stem-cell-based embryo models open new avenues for studying early development in vitro, including humans1. Here we report a protocol for the generation of human blastocyst-like structures from naïve human pluripotent stem cells (hPSCs). These structures, termed human blastoids, provide an accessible, scalable, versatile, and manipulable 3D model to study early human development, understand early pregnancy loss, and gain insights into early developmental defects.

An open-access database specialized for magnetic compounds, as well as for magnetic clusters, is developed with a focus on magnets free from rare earths. Data-intensive methods are used to facilitate the theoretical and experimental design and discovery of new magnetic materials. The utility of the datasets for computational screening, machine-learning modeling, and experimental fabrication is discussed.

We apply first-principles calculations to metastable B-doped Fe-Ni compounds to compute their magnetic properties. We focus on crystal structures with tetragonal and orthorhombic lattices. Boron atoms, doped at interstitial sites, help in stabilizing noncubic Fe-Ni structures. At the same time, the dopants improve the magnetic properties, such as magnetocrystalline anisotropy, compared to FeNi alloys in the tetragonal $L{1}_{0}$-ordered structures. The calculated magnetocrystalline anisotropy constants of $1--2 \mathrm{MJ}/{\mathrm{m}}^{3}$ along with sufficient magnetic polarization saturation $(\ensuremath{\ge}1 \mathrm{T})$ show the potential of our metastable Fe-Ni-B compounds for replacing rare-earth-based magnets in permanent-magnet applications.

Summary Dynamic pluripotent stem cell (PSC) states are in vitro adaptations of pluripotency continuum in vivo. Previous studies have generated a number of PSCs with distinct properties. To date, however, no known PSCs have demonstrated dual competency for chimera formation and direct responsiveness to primordial germ cell (PGC) specification, a unique functional feature of formative pluripotency. Here, by modulating fibroblast growth factor (FGF), transforming growth factor β (TGF-β), and WNT pathways, we derived PSCs from mice, horses, and humans (designated as XPSCs) that are permissive for direct PGC-like cell induction in vitro and are capable of contributing to intra- or inter-species chimeras in vivo. XPSCs represent a pluripotency state between naive and primed pluripotency and harbor molecular, cellular, and phenotypic features characteristic of formative pluripotency. XPSCs open new avenues for studying mammalian pluripotency and dissecting the molecular mechanisms governing PGC specification. Our method may be broadly applicable for the derivation of analogous stem cells from other mammalian species.

Embryonic stem cells (ESCs) are derived from the inner cell mass of preimplantation blastocysts. From agricultural and biomedical perspectives, the derivation of stable ESCs from domestic ungulates is important for genomic testing and selection, genome engineering, and modeling human diseases. Cattle are one of the most important domestic ungulates that are commonly used for food and bioreactors. To date, however, it remains a challenge to produce stable pluripotent bovine ESC lines. Employing a culture system containing fibroblast growth factor 2 and an inhibitor of the canonical Wnt-signaling pathway, we derived pluripotent bovine ESCs (bESCs) with stable morphology, transcriptome, karyotype, population-doubling time, pluripotency marker gene expression, and epigenetic features. Under this condition bESC lines were efficiently derived (100% in optimal conditions), were established quickly (3–4 wk), and were simple to propagate (by trypsin treatment). When used as donors for nuclear transfer, bESCs produced normal blastocyst rates, thereby opening the possibility for genomic selection, genome editing, and production of cattle with high genetic value.

In vivo genome editing represents a powerful strategy for both understanding basic biology and treating inherited diseases. However, it remains a challenge to develop universal and efficient in vivo genome-editing tools for tissues that comprise diverse cell types in either a dividing or non-dividing state. Here, we describe a versatile in vivo gene knock-in methodology that enables the targeting of a broad range of mutations and cell types through the insertion of a minigene at an intron of the target gene locus using an intracellularly linearized single homology arm donor. Asa proof-of-concept, we focused on a mouse model of premature-aging caused by a dominant point mutation, which is difficult to repair using existing in vivo genome-editing tools. Systemic treatment using our new method ameliorated aging-associated phenotypes and extended animal lifespan, thus highlighting the potential of this methodology for a broad range of in vivo genome-editing applications.

Human pluripotent stem (PS) cells can be isolated from preimplantation embryos or by reprogramming of somatic cells or germline progenitors. Human PS cells are considered the "holy grail" of regenerative medicine because they have the potential to form all cell types of the adult body. Because of their similarity to humans, nonhuman primate (NHP) PS cells are also important models for studying human biology and disease, as well as for developing therapeutic strategies and test bed for cell replacement therapy. This chapter describes adjusted methods for cultivation of PS cells from different primate species, including African green monkey, rhesus monkey, chimpanzee, and human. Supplementation of E8 medium and inhibitors of the Tankyrase and GSK3 kinases to various primate PS cell media reduce line-dependent predisposition for spontaneous differentiation in conventional PS cell cultures. We provide methods for basic characterization of primate PS cell lines, which include immunostaining for pluripotency markers such as OCT4 and TRA-1-60, as well as in vivo teratoma formation assay. We provide methods for generating alternative PS cells including region-selective primed PS cells, two different versions of naïve-like cells, and recently reported extended pluripotent stem (EPS) cells. These derivations are achieved by acclimation of conventional PS cells to target media, episomal reprogramming of somatic cells, or resetting conventional PS cells to a naïve-like state by overexpression of KLF2 and NANOG. We also provide methods for isolation of PS cells from human blastocysts. We describe how to generate interspecies primate-mouse chimeras at the blastocyst and postimplantation embryo stages. Systematic evaluation of the chimeric competency of human and primate PS cells will aid in efforts to overcome species barriers and achieve higher grade chimerism in postimplantation conceptuses that could enable organ-specific enrichment of human xenogeneic PS cell derivatives in large animals such as pigs and sheep.

We provide an avenue to achieve optimized magnetic properties without relying on rare-earth elements. In particular, we focus on transition-metal nanoparticles with magnetic ordering that is rarely seen in bulk. By using first-principles real-space pseudopotential calculations, we show that Mn can exhibit ferromagnetic behavior similar to Fe when Mn is placed on the surface of a FeCo cluster. This ferromagnetic behavior of surface Mn is in sharp contrast with Mn bulk and Mn-based alloys, which are either antiferromagnetic or paramagnetic. The local magnetic moment carried by a surface Mn atom is larger than those of Fe and Co, leading to an increase of the total magnetic moment. We find that surface-Mn-induced magnetic enhancement is robust in ternary MnFeCo clusters with different sizes and various chemical compositions. Our theoretical findings provide a useful guide for designing new nanoparticles with a large magnetic moment.

Necroptosis, a form of regulated necrosis, might be a potential mechanism of delayed paraplegia; therefore, its role in transient spinal cord ischaemia was investigated by immunohistochemical analysis of necroptosis related protein receptor interacting protein kinase (RIP) 1, RIP3, and cellular inhibitor of apoptosis protein (cIAP) 1/2.This study used rabbit normothermic (n = 24) and hypothermic (n = 24) transient spinal cord ischaemia models and sham controls (n = 6). Neurological function was assessed according to a modified Tarlov score at 8 h, 1, 2, and 7 days after reperfusion (n = 6 each). Morphological changes in the spinal cord were examined using haematoxylin and eosin staining in the sham, 2, and 7 day groups. Western blot and histochemical analyses of RIP1, RIP3, and cIAP1/2, and double label fluorescent immunocytochemical studies of RIP3 and cIAP1/2 were performed at 8 h, 1, and 2 days after reperfusion (n = 6 each).There were significant differences in neurological function between the normothermic and hypothermic groups (median scores 0 and 5 at 7 days, p = .023). In the normothermic group, most motor neurons were lost seven days after reperfusion (p = .046 compared with sham), but they were preserved in the hypothermic group. Western blot analysis revealed the upregulation of RIP1, RIP3, and cIAP1/2 at 8 h in the normothermic group (RIP1, p = .032; RIP3, p .001; cIAP1/2, p = .041 compared with sham), and the overexpression of RIP3 was prolonged for two days. In the hypothermic group, the expression of these proteins was not observed. The double label fluorescent immunocytochemical study revealed the induction of RIP3 and cIAP1/2 in the same motor neurons.These data suggest that transient normothermic ischaemia induces necroptosis, a potential factor in delayed motor neuron death, and that hypothermia may inhibit necroptosis.

The development of spinal cord injury is believed to be related to the vulnerability of spinal motor neurons to ischemia. However, the mechanisms underlying this vulnerability have not been fully investigated. Previously, we reported that spinal motor neurons are lost likely due to autophagy and that local hypothermia prevents such spinal motor neuron death. Therefore, we investigated the role of autophagy in normothermic and hypothermic spinal cord ischemia using an immunohistochemical analysis of Beclin 1 (BCLN1; B-cell leukemia 2 protein [Bcl-2] interacting protein), Bcl-2, and γ-aminobutyric acid type-A receptor-associated protein (GABARAP), which are considered autophagy-related proteins.We used rabbit normothermic and hypothermic transient spinal cord ischemia models using a balloon catheter. Neurologic function was assessed according to the Johnson score, and the spinal cord was removed at 8 hours and 1, 2, and 7 days after reperfusion, and morphologic changes were examined using hematoxylin and eosin staining. A Western blot analysis and histochemical study of BCLN1, Bcl-2, and GABARAP, and double-labeled fluorescent immunocytochemical studies were performed.There were significant differences in the physiologic function between the normothermic model and hypothermic model after the procedure (P .05). In the normothermic model, most of the motor neurons were selectively lost at 7 days of reperfusion (P .001 compared with the sham group), and they were preserved in the hypothermic model (P = .574 compared with the sham group). The Western blot analysis revealed that the sustained expression of the autophagy markers, BCLN1 and GABARAP, was observed (P .001 compared with the sham group) and was associated with neuronal cell death in normothermic ischemic conditions. In hypothermic ischemic conditions, the autophagy inhibitory protein Bcl-2 was powerfully induced (P .001 compared with the sham group) and was associated with blunted expression of BCLN1 and GABARAP and neuronal cell survival. The double-label fluorescent immunocytochemical study revealed that immunoreactivitiy for BCLN1, Bcl-2, and GABARAP was induced in the same motor neurons.These data suggest that the prolonged induction of autophagy might be a potential factor responsible for delayed motor neuron death, and the induction of the autophagy inhibitory protein Bcl-2 using hypothermia might limit autophagy and protect against delayed motor neuron death.

Large cutaneous ulcers are, in severe cases, life threatening(1,2). As the global population ages, non-healing ulcers are becoming increasingly common(1,2). Treatment currently requires the transplantation of pre-existing epithelial components, such as skin grafts, or therapy using cultured cells(2). Here we develop alternative supplies of epidermal coverage for the treatment of these kinds of wounds. We generated expandable epithelial tissues using in vivo reprogramming of wound-resident mesenchymal cells. Transduction of four transcription factors that specify the skin-cell lineage enabled efficient and rapid de novo epithelialization from the surface of cutaneous ulcers in mice. Our findings may provide a new therapeutic avenue for treating skin wounds and could be extended to other disease situations in which tissue homeostasis and repair are impaired.

We present a real-space pseudopotential method for calculating magnetocrystalline anisotropy within relativistic density-functional theory. The spin-orbit interaction, a fundamental origin of magnetocrystalline anisotropy, is incorporated with norm-conserving pseudopotentials expressed on a real-space grid. We demonstrate the utility of our method by calculating the magnetocrystalline anisotropy constant, ${K}_{1}$, of ${\mathrm{YCo}}_{5}$, which is a prototype compound with large ${K}_{1}$. Our calculated ${K}_{1}$ agrees with previous theoretical work. We show that our formalism also works for describing magnetocrystalline anisotropy in other transition-metal compounds, such as ${\mathrm{Mn}}_{2}\mathrm{Ga}$ and FeNi, which have modest values for ${K}_{1}$. Our real-space pseudopotential method is well suited for a parallel computing environment and is an efficacious approach to solving the relativistic Kohn-Sham equations, which include spin-orbit effects as well as noncollinear magnetism.

We investigate the magnetocrystalline anisotropy of ${\mathrm{YCo}}_{5}$ and ${\mathrm{ZrCo}}_{5}$ compounds from first-principles calculations using a real-space formalism of pseudopotentials. We study not only the experimentally observed phases but also other possible crystal structures in order to examine the impact of structural differences on the magnetic properties. Our results indicate that it may be difficult to enhance magnetocrystalline anisotropy constant ${K}_{1}$ and saturation magnetization ${M}_{s}$ simultaneously in ${\mathrm{YCo}}_{5}$ compounds in a hexagonal crystal family. We find that ${\mathrm{ZrCo}}_{5}$ compounds have moderate ${K}_{1}$ and sufficient ${M}_{s}$, which can be much better than those of conventional ferrite magnets. We expect that ${\mathrm{ZrCo}}_{5}$ compounds, consisting of affordable elements, are promising rare-earth-free materials that will be useful for permanent magnet applications.

Using a real-space implementation of pseudopotentials within the density-functional theory, we show that the magnetization of a ${\mathrm{Co}}_{3}\mathrm{N}$ cluster with a recently discovered atomic structure is significantly affected by nitrogen dopants. In a ${\mathrm{Co}}_{3}\mathrm{N}$ cluster with the hexagonal $P{6}_{3}/mmc$ structure, N dopants promote spin polarization for the Co-$3d$ electrons and the dopants themselves make additional contributions to net magnetic moment. These two factors enhance the total magnetic moment of a hexagonal ${\mathrm{Co}}_{3}\mathrm{N}$ cluster, which can be as strong as bulk iron. In contrast, N dopants in a ${\mathrm{Co}}_{3}\mathrm{N}$ cluster with the rhombohedral $R\overline{3}c$ structure degrade magnetic moment and the dopants are magnetically ``inert,'' which results in lower total magnetic moments in rhombohedral ${\mathrm{Co}}_{3}\mathrm{N}$ clusters. These changes in magnetic moment originate from differences in the orbital hybridization between the Co-$3d$ and N-$2p$ states. We also examine how the magnetization of a ${\mathrm{Co}}_{3}\mathrm{N}$ cluster depends on a N content. We find that the total magnetic moment of a hexagonal ${\mathrm{Co}}_{3}{\mathrm{N}}_{1+x}$ cluster with $\ensuremath{-}0.15\ensuremath{\le}x\ensuremath{\le}0.15$ is tunable and can be enhanced further by controlling the amount of nitrogen dopants.

Compared to the early stage (2011-2012) after the Great East Japan Earthquake, no significant changes were seen in the total number of case of tuberculosis (TB), and pulmonary TB cases in the mid-term stage (2013-2014). However, the number of patients with latent tuberculosis infection (LTBI) decreased significantly in the coastal region of Northern Miyagi, which was ravaged by a post-quake tsunami. We analyzed the data of all TB patients in this region in the early (2011- 2012) and the late (2015- 2016) stages following the Great East Japan Earthquake and compared it with the pre-disaster data. The number of cases of total TB, extra pulmonary TB and LTBI significantly in comparison to the initial stage after the disaster decreased, but there was a significantly higher prevalence of TB and the LTBI compared to pre-disaster. These results suggest that continued health activities and an improved transportation system reduced the prevalence of total TB after the Great East Japan Earthquake. Thus, these health activities should be maintained.

At our clinic, a successfully fertilized human oocyte is defined as one with two polar bodies and two pronuclei (PN) 18 h after ICSI, but when some oocytes do not form PN. Recently, a successful pregnancy and live birth following artificial oocyte activation (AOA; using Ca2+ ionophore) of ICSI-treated one-day-old unfertilized oocytes was reported. In this study, we performed AOA on PN-lacking unfertilized oocytes, 18 h after ICSI (defined as Day 1-AOA, since ICSI=Day 0). The abilities of these oocytes to form PN and develop were compared with those of oocytes fertilized by ICSI (defined as No-AOA). Piezo-ICSI was performed on 168 oocytes, 50 of which failed to fertilize (0PN) but were subsequently artificially activated. 2PN formation rate was significantly lower in Day 1-AOA, and the rates of 1PN and ≥4PN were significantly higher than in No-AOA. We investigated the expression of focal adhesion kinase (FAK) in mouse cumulus-oocyte complexes (COCs), as well as the role of FAK phosphorylation at Tyr397 during oocyte maturation. The effect of inhibiting FAK phosphorylation at Tyr397 during in vitro maturation (IVM) on subsequent fertilization and preimplantation embryo development was also examined. Western blotting analyses revealed that total and Tyr397-phosphorylated FAK were expressed in vivo in both cumulus cells and oocytes. Immunocytochemical studies localized this kinase throughout the cytoplasm of cumulus cells and oocytes; in particular, Tyr397-phosphorylated FAK tended to accumulate in regions where cumulus cells contact each other. Interestingly, the in vivo level of Tyr397 phosphorylation in cumulus cells was significantly lower after compared to before cumulus expansion. Addition of FAK inhibitor 14, which specifically blocks phosphorylation at Tyr397, stimulated oocyte meiotic maturation and cumulus expansion during IVM in the absence of follicle-stimulating hormone (FSH). Reverse-transcriptase PCR showed that the mRNA expression of hyaluronan synthase 2 (Has2), a marker of cumulus expansion, was significantly induced in cumulus cells. Subsequent in vitro fertilization and culture showed that more oocytes developed to the blastocyst stage when they were treated with FAK inhibitor 14 during IVM, although the blastocyst total cell number was lower than in oocytes stimulated with FSH. These results indicate that FAK is involved in the maturation of COCs; specifically, phosphorylation at Tyr397 may regulate cumulus expansion via the expression of Has2 mRNA in cumulus cells, which could affect the developmental competence of oocytes.

We perform ab initio many-body Green's function calculations to investigate the quasiparticle energies and optical properties of diamond polytypes that have been predicted to be producible via a pressure-induced structural phase transition from carbon nanotube solids. We find, through quasiparticle band-structure calculations within the $GW$ approximation, that the band gaps of two hexagonal $(2H$- and $4H$-type) polytypes of diamond differ significantly from that of cubic diamond as well as from that of the crystalline $s{p}^{3}$ carbon phase with a body-centered-tetragonal structure, called bct ${\mathrm{C}}_{4}$. We also examine the dielectric functions of three polytypes of diamond (cubic, $2H$, and $4H)$ by employing the $GW$ plus Bethe-Salpeter equation approach. The calculated optical absorption spectra are found to be distinct from each other. The lattice mismatches of carbon layers of these diamond polytypes are very small and the total-energy differences are also small. Our work opens up the possibility of fabricating diamond superlattices with various electronic and optoelectronic properties by utilizing and controlling the different stacking sequences of carbon layers.

We propose a systematic approach to obtain various forms of insulating titanium oxynitrides Ti$_{n}$N$_{2}$O$_{2n-3}$ and we conduct a detailed study on its $n=2$ case, Ti$_{2}$N$_{2}$O. We study the energetics and the electronic structures of Ti$_{2}$N$_{2}$O and compare these results with those of pristine and nitrogen-doped TiO$_{2}$ within the framework of the density-functional theory (DFT) and the GW approximation. We find that Ti$_{2}$N$_{2}$O is semiconducting with the calculated band-gap of 1.81 eV, which is significantly smaller than those of pristine TiO$_{2}$ rutile (3.14 eV) or anatase (3.55 eV). Furthermore, the reduction of the band-gap of Ti$_{2}$N$_{2}$O is realized not by lowering of the conduction-band minimum but by rising the valence-band maximum. Therefore the proposed Ti$_{2}$N$_{2}$O has suitable band-edge alignment for water-splitting photocatalysis. Finally, total energy calculations indicate that Ti$_{2}$N$_{2}$O is potentially easier to synthesize than nitrogen-doped TiO$_{2}$. During the early stage (2011-2012) after the Great East Japan Earthquake, the total number of tuberculosis (TB), pulmonary TB and latent tuberculosis infection (LTBI) patients significantly increased only in the coastal region of northern Miyagi, which was ravaged by a post-quake tsunami. Here, we investigated the midterm-stage follow-up (2013-2014) of TB infection in this region. Although the number of total TB, pulmonary TB, and LTBI cases had significantly decreased, their prevalence had not returned to the levels recorded in the pre-disaster period. These results suggest that the influence of the Great East Japan Earthquake remains. Thus, we believe that it is necessary to continue strict monitoring of TB in the coastal region of northern Miyagi.

Interspecies blastocyst complementation enables organ-specific enrichment of xenogenic pluripotent stem cell (PSC) derivatives. Here, we establish a versatile blastocyst complementation platform based on CRISPR-Cas9-mediated zygote genome editing and show enrichment of rat PSC-derivatives in several tissues of gene-edited organogenesis-disabled mice. Besides gaining insights into species evolution, embryogenesis, and human disease, interspecies blastocyst complementation might allow human organ generation in animals whose organ size, anatomy, and physiology are closer to humans. To date, however, whether human PSCs (hPSCs) can contribute to chimera formation in non-rodent species remains unknown. We systematically evaluate the chimeric competency of several types of hPSCs using a more diversified clade of mammals, the ungulates. We find that naive hPSCs robustly engraft in both pig and cattle pre-implantation blastocysts but show limited contribution to post-implantation pig embryos. Instead, an intermediate hPSC type exhibits higher degree of chimerism and is able to generate differentiated progenies in post-implantation pig embryos.

A single mouse blastomere from an embryo until the 8-cell stage can generate an entire blastocyst. Whether laboratory-cultured cells retain a similar generative capacity remains unknown. Starting from a single stem cell type, extended pluripotent stem (EPS) cells, we established a 3D differentiation system that enabled the generation of blastocyst-like structures (EPS-blastoids) through lineage segregation and self-organization. EPS-blastoids resembled blastocysts in morphology and cell-lineage allocation and recapitulated key morphogenetic events during preimplantation and early postimplantation development in vitro. Upon transfer, some EPS-blastoids underwent implantation, induced decidualization, and generated live, albeit disorganized, tissues in utero. Single-cell and bulk RNA-sequencing analysis revealed that EPS-blastoids contained all three blastocyst cell lineages and shared transcriptional similarity with natural blastocysts. We also provide proof of concept that EPS-blastoids can be generated from adult cells via cellular reprogramming. EPS-blastoids provide a unique platform for studying early embryogenesis and pave the way to creating viable synthetic embryos by using cultured cells.

We examine the structural stability and magnetization for nickel clusters containing up to 500 atoms by performing first-principles calculations based on pseudopotential in real space computed within density-functional theory. After structural relaxation, Ni clusters in this size range favor either an fcc structure, which is a crystal structure in bulk, or an icosahedral structure, which is expected for small clusters. The calculated total magnetic moments per atom of energetically stable clusters agree well with experiment, wherein the moments decrease nonmonotonically toward the bulk value as the cluster size increases. We analyze the spatial distribution of the local magnetic moment, which explains why the magnetic moments of Ni clusters are enhanced compared to their bulk value.

We use a real-space implementation of pseudopotentials within the density functional theory to investigate the structural and magnetic properties of cobalt clusters with up to 365 atoms. We find from structural optimization that hcp and icosahedral clusters are lower in energy than bcc and fcc clusters. The calculated magnetic moments generally decrease as a function of increasing cluster size. For clusters of several hundred atoms the bulk limit becomes apparent. However, the decrease is not monotonic. It depends on the details of the interior structure of the cluster and the corresponding surface geometry. By analyzing the detailed evolution of the local magnetic moment, we find the spin moment is bulklike in the cluster interior and increases in the vicinity of the surface and can be correlated with coordination. The calculated behavior accounts for the observed variations in the measured moments.

On March 11, 2011, the Great East Japan Earthquake struck off the northeast coast of Japan. Within an hour of the earthquake, devastating tsunamis swept over the coastal region of the Miyagi Prefecture, facing Pacific Ocean. Accordingly, more than 400,000 residents were forced to stay at evacuation shelters. We investigated the changes in tuberculosis prevalence after the disaster. Annual data for all tuberculosis patients between April 1, 2009 and March 31, 2013 were extracted from the database of the Miyagi Prefectural Government. In the coastal region of Northern Miyagi, the number of tuberculosis patients increased in the post-disaster period (p < 0.001, 9.6 vs.19.1 per 100,000 people), compared to the pre-disaster period. In contrast, its prevalence did not change in the inland region of Northern Miyagi and the coastal and inland regions of Southern Miyagi. Importantly, in the inland and coastal regions of Northern Miyagi, the number of patients with latent tuberculosis infection (LTBI) increased in the post-disaster period (p < 0.001). Furthermore, in the coastal shelters, 11 evacuees with the history of contacting tuberculosis patients were diagnosed with LTBI, whereas no cases of LTBI patients were observed in the inland shelters. Thus, staying in the coastal shelters was a risk factor for contracting tuberculosis (OR: 19.31, 95% CI: 1.11-334.80); indeed, twice as many evacuees visited each coastal shelter on April 1, 2011, compared to the inland region. We should prepare the shelters to avoid overcrowding, and long-term observation is required to detect the prevalence of tuberculosis infection.

We have previously reported unique localization of TINAGL1 to the trophectoderm (TE) of mouse blastocysts. TINAGL1 was found to be upregulated in implantation-competent blastocysts after estrogen treatment using progesterone-treated delayed-implantation models. Moreover, colocalization of TINAGL1 and extracellular matrix (ECM) protein laminin 1 was detected in the Reichert membrane on embryonic days 6.5 and 7.5. Although these data suggested a role for TINAGL1 in the embryo development at postimplantation, its relevance to other ECM proteins during preimplantation development is not clear. In this study, we examined the expression of TINAGL1 and its relevance to other ECM proteins fibronectin (FN) and collagen type IV (ColIV) duringin vivodevelopment of preimplantation embryos, particularly at blastocyst stage in detail. Localizations of TINAGL1, FN, and ColIV were similar. In 1-cell to 8-cell embryos, they were expressed in cytoplasm of blastomeres, and in morulae they were localized in the outer cells. FN and ColIV were expressed primarily on outer surface of the cells. In blastocysts, FN and ColIV were distributed in the cytoplasm of TE, but, just prior to implantation, they became localized uniquely to the blastocoelic surface of TE. Inin vitrofertilized (IVF) blastocysts, expression levels of TINAGL1 and FN were lower than inin vivoblastocysts. These results suggest that, during preimplantation development, TINAGL1 may be involved in roles of structural matrix proteins, whose expression in blastocysts may be affected byin vitroculture.

Several protein tyrosine kinases (PTKs) are identified as follicle survival factors that suppress apoptosis in granulosa cells. Focal adhesion kinase (FAK/PTK2) interacts with numerous signaling partners and is important for cell adhesion, survival and other vital processes in which FAK autophosphorylation at Y397 (pY397 FAK) is critical for activating signaling pathways. Despite its important roles in apoptosis, the expression and function of FAK in the ovaries remain unknown. Here, we describe FAK expression, including pY397 FAK, in normal healthy mouse ovaries and its association with follicular development and/or atresia. Normal healthy mouse ovaries were used for western blot (n > 60) and immunohistochemical (n > 180) analyses. Western blot results in immature and mature mice revealed that total FAK and pY397 FAK were highly expressed in the ovary and immunohistochemistry results in 3-week-old mice showed they were localized to granulosa cells of ovarian follicles, especially preantral follicles. In 3-week-old mice treated with 5 IU pregnant mare serum gonadotropin (for obtaining homogenous populations of growing or atretic follicles), western blotting revealed that follicular atresia progression involved decreased phosphorylation of Y397 at 72 and 96 h after treatment, particularly in granulosa cells of atretic follicles, as shown by immunohistochemistry results at 72 h after treatment. Moreover, immunostaining patterns of FAK and cleaved caspase-3 were negatively correlated in serial sections of 3-week-old mouse ovaries. These results suggest that FAK is most active in ovarian follicle granulosa cells and that its phosphorylation at Y397 is histologically meaningful in follicular development in normal healthy ovaries.

We study pressure-induced structural phase transition of carbon nanotube solids using the constant-pressure tight-binding molecular-dynamics method. We find that the bundle with armchair nanotube transforms into a new phase consisting of so-called zigzag graphene nanoribbons and sp 3 carbon atoms by applying the pressure of 10 GPa. Under higher pressure of 30 GPa, anisotropic sp 3 phases possessing high hardness comparable to cubic diamond are obtained from armchair nanotube solid, whereas amorphous diamond phases are obtained from the bundle with chiral nanotubes. Using the density-functional theory, we also study electronic structure of a new sp 3 phase obtained in the molecular-dynamics simulation.

Summary Current genome-editing systems generally rely on inducing DNA double-strand breaks (DSBs). This may limit their utility in clinical therapies, as unwanted mutations caused by DSBs can have deleterious effects. CRISPR/Cas9 system has recently been repurposed to enable target gene activation, allowing regulation of endogenous gene expression without creating DSBs. However, in vivo implementation of this gain-of-function system has proven difficult. Here, we report a robust system for in vivo activation of endogenous target genes through trans -epigenetic remodeling. The system relies on recruitment of Cas9 and transcriptional activation complexes to target loci by modified single guide RNAs. As proof-of-concept, we used this technology to treat mouse models of diabetes, muscular dystrophy, and acute kidney disease. Results demonstrate that CRISPR/Cas9-mediated target gene activation can be achieved in vivo , leading to measurable phenotypes and amelioration of disease symptoms. This establishes new avenues for developing targeted epigenetic therapies against human diseases. Video Abstract

Tubulointerstitial nephritis antigen-like 1 (Tinagl1, also known as adrenocortical zonation factor 1 [AZ-1] or lipocalin 7) has been cloned from mouse adrenocortical cells and is known to be closely associated with zonal differentiation of adrenocortical cells. In cell culture systems, TINAGL1 is a matricellular protein that interacts with both structural matrix proteins and cell surface receptors. However, the physiological roles of TINAGL1 and regulation of its expression are still not clearly understood. In the present study, the expression and localization of TINAGL1 in peri-implantation mouse embryos was examined. During preimplantation, the expression of both Tinagl1 mRNA and TINAGL1 protein was increased just prior to implantation. In blastocysts, TINAGL1 expression was localized to the trophectoderm. Using a progesterone-treated, delayed-implantation model, TINAGL1 was found to be upregulated in implantation-competent blastocysts after estrogen treatment. During postimplantation, TINAGL1 expression was restricted to extraembryonic regions. Marked expression was detected in the Reichert membrane on Embryonic Days 6.5 (E6.5) and E7.5. Colocalization of laminin 1 and TINAGL1 was also examined. Using an anti-LAMA1 antibody, colocalization of LAMA1 and TINAGL1 was observed in postimplantation embryos. Colocalization was also detected in the Reichert membrane. Immunoprecipitation analysis determined that LAMA1 and TINAGL1 interact in embryos on E7.5. These results demonstrate that after implantation, TINAGL1 is an extraembryonic tissue-specific protein. In particular, TINAGL1 is a novel component of the Reichert membrane that interacts with laminin 1. These results suggest that TINAGL1 most likely plays a physical and physiological role in embryo development at postimplantation.
