1. Multiple Emission Lines of Hα Emitters at z ∼ 2.3 from the Broad- and Medium-band Photometry in the ZFOURGE Survey

Abstract

We present a multiple emission line study of ∼1300 H α emitters (HAEs) at z ∼ 2.3 in the ZFOURGE survey. In contrast to the traditional spectroscopic method, our sample is selected based on the flux excess in the ZFOURGE K _s broadband data relative to the best-fit stellar continuum. Using the same method, we also extract the strong diagnostic emission lines for these individual HAEs: [O iii ] λ λ 4959, 5007 and [O ii ] λ λ 3726, 3729. Our measurements demonstrate good consistency with those obtained from spectroscopic surveys. We investigate the relationship between the equivalent widths (EWs) of these emission lines and various galaxy properties, including stellar mass, stellar age, star formation rate, specific star formation rate, and ionization state (O32). We have identified a discrepancy between HAEs at z ∼ 2.3 and typical local star-forming galaxies observed in the Sloan Digital Sky Survey, suggesting the evolution of lower gas-phase metallicity ( Z ) and higher ionization parameters ( U ) with redshift. Notably, we have observed a significant number of low-mass HAEs exhibiting exceptionally high EW _[O _iii _] . Their galaxy properties are comparable to those of extreme objects, such as extreme O3 emitters and Ly α emitters at z ≃ 2–3. Considering that these characteristics may indicate potential strong Lyman continuum leakage, higher-redshift analogs of the low-mass HAEs could be significant contributors to the cosmic reionization. Further investigations of this particular population are required to gain a clearer understanding of galaxy evolution and cosmic reionization.

3. Serum IgG4 levels at diagnosis can predict unfavorable outcomes of untreated patients with IgG4-related disease

Abstract

Abstract The outcomes of patients with immunoglobulin G4 (IgG4)-related disease (IgG4-RD) who are not treated are unclear. This study aimed to clarify these outcomes and identify the factors related to them. We retrospectively evaluated various clinical features including laboratory data and involved organs at diagnosis in 107 patients with IgG4-RD, who were followed up for more than 6 months, at a single center in Japan. We compared the clinical features of the 27 untreated patients with those of the 80 patients treated with glucocorticoid. The patient outcomes were investigated, and logistic regression analysis was performed to identify factors related to them. The patients comprised 73 men and 34 women (median age 67 years). The untreated patients had significantly lower IgG4-RD responder index (9 vs. 12) and fewer affected organs (1 vs. 3) than did those treated with glucocorticoid. Of these 27 patients, 8 experienced deterioration of IgG4-RD after the diagnosis. In the age- and sex-adjusted logistic regression analysis, serum IgG4 elevation (per 100 mg/dL, odds ratio 1.194, 95% confidence interval 1.017–1.402) was the only significant factor related to disease deterioration in untreated patients with IgG4-RD, whereas not serum IgG4 levels (per 100 mg/dL, odds ratio 0.995, 95% confidence interval 0.921–1.075) but history of allergy (OR 3.134, 95% confidence interval 1.094–8.977, P = 0.033) related to deterioration in patients who underwent treatment. Serum IgG4 levels may be a useful predictor of unfavorable outcomes in untreated patients with IgG4-RD, who tend to have fewer affected organs and lower IgG4-RD responder index.

Abstract

We have performed intensive follow-up observations of a Type IIn/Ia-CSM supernova (SN IIn/Ia-CSM), 2020uem, with photometry, spectroscopy, and polarimetry. In this paper, we report on the results of our observations focusing on optical/near-infrared (NIR) photometry and spectroscopy. The maximum V -band magnitude of SN 2020uem is less than −19.5 mag. The light curves decline slowly with a rate of ∼0.75 mag/100 days. In the late phase (≳300 days), the light curves show accelerated decay (∼1.2 mag/100 days). The optical spectra show prominent hydrogen emission lines and broad features possibly associated with Fe-peak elements. In addition, the H α profile exhibits a narrow P-Cygni profile with an absorption minimum of ∼100 km s ^−1 . SN 2020uem shows a higher H α /H β ratio (∼7) than those of SNe IIn, which suggests a denser circumstellar medium (CSM). The NIR spectrum shows the Paschen and Brackett series with a continuum excess in the H and Ks bands. We conclude that the NIR excess emission originates from newly formed carbon dust. The dust mass ( M _d ) and temperature ( T _d ) are derived to be ( M _d , T _d ) ∼ (4−7 × 10 ^−5 M _⊙ , 1500–1600 K). We discuss the differences and similarities between the observational properties of SNe IIn/Ia-CSM and those of other SNe Ia and interacting SNe. In particular, spectral features around ∼4650 Å and ∼5900 Å of SNe IIn/Ia-CSM are more suppressed than those of SNe Ia; these lines are possibly contributed, at least partly, by Mg i ] and Na i , and may be suppressed by high ionization behind the reverse shock caused by the massive CSM.

5. Multi-dimensional Transcriptional Remodeling by Physiological Insulin In Vivo

Abstract

Summary: Regulation of gene expression is an important aspect of insulin action but in vivo is intertwined with changing levels of glucose and counter-regulatory hormones. Here we demonstrate that under euglycemic clamp conditions, physiological levels of insulin regulate interrelated networks of more than 1,000 transcripts in muscle and liver. These include expected pathways related to glucose and lipid utilization, mitochondrial function, and autophagy, as well as unexpected pathways, such as chromatin remodeling, mRNA splicing, and Notch signaling. These acutely regulated pathways extend beyond those dysregulated in mice with chronic insulin deficiency or insulin resistance and involve a broad network of transcription factors. More than 150 non-coding RNAs were regulated by insulin, many of which also responded to fasting and refeeding. Pathway analysis and RNAi knockdown revealed a role for lncRNA Gm15441 in regulating fatty acid oxidation in hepatocytes. Altogether, these changes in coding and non-coding RNAs provide an integrated transcriptional network underlying the complexity of insulin action. : Batista et al. demonstrate potent transcriptional remodeling by physiological insulin action in skeletal muscle and liver, involving interrelated networks of protein-coding genes, transcription factors, and long non-coding RNAs (lncRNAs). From an array of metabolically sensitive lncRNAs, Gm15441 is identified as a regulator of fatty acid oxidation in hepatocytes. Abstract

An extended version of cross-bar type addressing technique is developed for three-port electrostatic micro shutters arranged in an arrayed format. A microelectromechanical systems (MEMS) shutter blade suspended by a pair of torsion beams works as a movable electrode that is either attracted upwards to the cover plate to close the aperture or retracted downwards into the through-hole to open it. Tri-state positioning of the shutter—i.e., open, rest, and close—is controlled by the hysteresis loop of the electrostatic pull-in and release behavior using the combination of the voltages applied to the shutter, the cover, and the substrate. Random access addressing of the shutters is demonstrated by a control system composed of MATLAB-coded Arduino electronics. The shutter array developed in this work is for a sub-cluster of a reconfigurable shutter array under development for a multi-object galactic astronomy.

Abstract

Background The effects of empagliflozin in patients with chronic kidney disease who are at risk for disease progression are not well understood. The EMPA-KIDNEY trial was designed to assess the effects of treatment with empagliflozin in a broad range of such patients. Methods We enrolled patients with chronic kidney disease who had an estimated glomerular filtration rate (eGFR) of at least 20 but less than 45 ml per minute per 1.73 m2 of body-surface area, or who had an eGFR of at least 45 but less than 90 ml per minute per 1.73 m2 with a urinary albumin-to-creatinine ratio (with albumin measured in milligrams and creatinine measured in grams) of at least 200. Patients were randomly assigned to receive empagliflozin (10 mg once daily) or matching placebo. The primary outcome was a composite of progression of kidney disease (defined as end-stage kidney disease, a sustained decrease in eGFR to Results A total of 6609 patients underwent randomization. During a median of 2.0 years of follow-up, progression of kidney disease or death from cardiovascular causes occurred in 432 of 3304 patients (13.1%) in the empagliflozin group and in 558 of 3305 patients (16.9%) in the placebo group (hazard ratio, 0.72; 95% confidence interval [CI], 0.64 to 0.82; PConclusions Among a wide range of patients with chronic kidney disease who were at risk for disease progression, empagliflozin therapy led to a lower risk of progression of kidney disease or death from cardiovascular causes than placebo. Abstract

The formation of massive stars in dense and cold molecular clouds is a fundamental problem in star formation. In this work, we studied three compact massive star-forming regions, M8E, RAFGL 6366S, and IRAS 18317−0513, to investigate the environment of massive star formation, specifically the possibility of a local feedback process. Our new mid-infrared observations using miniTAO/MAX38 at 31 and 37 μm resolved individual objects in each region and allowed us to obtain their individual luminosities and masses. Together with existence/absence of ultra-compact H ii regions, it is suggested that less-massive objects are more evolved than the more-massive objects in two out of the three regions. Because the objects that are more massive evolve faster, those that are less massive form earlier. This formation trend of local mass-sequential star was first suggested in 0.1 pc-scale massive star-forming regions. Therefore, feedback, such as outflows, jets, or radiative heating, from previously formed lower-mass young stellar objects might affect the environment of parental clouds and lead to next-generation massive young stellar objects.

Abstract

The brain is now recognized as an insulin-sensitive tissue; however, the role of changing insulin concentrations in the peripheral circulation in gene expression in the brain is largely unknown. Here, we performed a hyperinsulinemic-euglycemic clamp on 3-month-old male C57BL/6 mice for 3 h. We show that, in comparison with results in saline-infused controls, increases in peripheral insulin within the physiological range regulate expression of a broad network of genes in the brain. Insulin regulates distinct pathways in the hypothalamus (HTM), hippocampus, and nucleus accumbens. Insulin shows its most robust effect in the HTM and regulates multiple genes involved in neurotransmission, including upregulating expression of multiple subunits of GABA-A receptors, Na+ and K+ channels, and SNARE proteins; differentially modulating glutamate receptors; and suppressing multiple neuropeptides. Insulin also strongly modulates metabolic genes in the HTM, suppressing genes in the glycolysis and pentose phosphate pathways, while increasing expression of genes regulating pyruvate dehydrogenase and long-chain fatty acyl-CoA and cholesterol biosynthesis, thereby rerouting of carbon substrates from glucose metabolism to lipid metabolism required for the biogenesis of membranes for neuronal and glial function and synaptic remodeling. Furthermore, based on the transcriptional signatures, these changes in gene expression involve neurons, astrocytes, oligodendrocytes, microglia, and endothelial cells. Thus, peripheral insulin acutely and potently regulates expression of a broad network of genes involved in neurotransmission and brain metabolism. Dysregulation of these pathways could have dramatic effects in normal physiology and diabetes.

Abstract

Reduced glucose uptake into the skeletal muscle is an important pathophysiological abnormality in type 2 diabetes, and is caused by impaired translocation of glucose transporter 4 (GLUT4) to the skeletal muscle cell surface. Here, we show a xanthene derivative, DS20060511, induces GLUT4 translocation to the skeletal muscle cell surface, thereby stimulating glucose uptake into the tissue. DS20060511 induced GLUT4 translocation and stimulated glucose uptake into differentiated L6-myotubes and into the skeletal muscles in mice. These effects were completely abolished in GLUT4 knockout mice. Induction of GLUT4 translocation by DS20060511 was independent of the insulin signaling pathways including IRS1-Akt-AS160 phosphorylation and IRS1-Rac1-actin polymerization, eNOS pathway, and AMPK pathway. Acute and chronic DS20060511 treatment attenuated the glucose intolerance in obese diabetic mice. Taken together, DS20060511 acts as a skeletal muscle-specific GLUT4 translocation enhancer to facilitate glucose uptake. Further studies of DS20060511 may pave the way for the development of novel antidiabetic medicines., Furuzono et al. identify DS20060511, a skeletal muscle cell-specific GLUT4 translocation enhancer. They find that DS20060511 have remarkable effects on lowering blood glucose and enhancing skeletal muscle glucose uptake via GLUT4, highlighting its potential as antidiabetic medicine.

Abstract

The outcomes of patients with immunoglobulin G4 (IgG4)-related disease (IgG4-RD) who are not treated are unclear. This study aimed to clarify these outcomes and identify the factors related to them. We retrospectively evaluated various clinical features including laboratory data and involved organs at diagnosis in 107 patients with IgG4-RD, who were followed up for more than 6 months, at a single center in Japan. We compared the clinical features of the 27 untreated patients with those of the 80 patients treated with glucocorticoid. The patient outcomes were investigated, and logistic regression analysis was performed to identify factors related to them. The patients comprised 73 men and 34 women (median age 67 years). The untreated patients had significantly lower IgG4-RD responder index (9 vs. 12) and fewer affected organs (1 vs. 3) than did those treated with glucocorticoid. Of these 27 patients, 8 experienced deterioration of IgG4-RD after the diagnosis. In the age- and sex-adjusted logistic regression analysis, serum IgG4 elevation (per 100 mg/dL, odds ratio 1.194, 95% confidence interval 1.017–1.402) was the only significant factor related to disease deterioration in untreated patients with IgG4-RD, whereas not serum IgG4 levels (per 100 mg/dL, odds ratio 0.995, 95% confidence interval 0.921–1.075) but history of allergy (OR 3.134, 95% confidence interval 1.094–8.977, P = 0.033) related to deterioration in patients who underwent treatment. Serum IgG4 levels may be a useful predictor of unfavorable outcomes in untreated patients with IgG4-RD, who tend to have fewer affected organs and lower IgG4-RD responder index.

Abstract

Previous studies have shown that insulin and IGF-1 signaling in the brain, especially the hypothalamus, is important for regulation of systemic metabolism. Here, we develop mice in which we have specifically inactivated both insulin receptors (IRs) and IGF-1 receptors (IGF1Rs) in the hippocampus (Hippo-DKO) or central amygdala (CeA-DKO) by stereotaxic delivery of AAV-Cre into IR(lox/lox)/IGF1R(lox/lox) mice. Consequently, both Hippo-DKO and CeA-DKO mice have decreased levels of the GluA1 subunit of glutamate AMPA receptor and display increased anxiety-like behavior, impaired cognition, and metabolic abnormalities, including glucose intolerance. Hippo-DKO mice also display abnormal spatial learning and memory whereas CeA-DKO mice have impaired cold-induced thermogenesis. Thus, insulin/IGF-1 signaling has common roles in the hippocampus and central amygdala, affecting synaptic function, systemic glucose homeostasis, behavior, and cognition. In addition, in the hippocampus, insulin/IGF-1 signaling is important for spatial learning and memory whereas insulin/IGF-1 signaling in the central amygdala controls thermogenesis via regulation of neural circuits innervating interscapular brown adipose tissue.

Abstract

Objectives In IgG4-related dacryoadenitis and/or sialadenitis (IgG4-DS), involvement of two or more sets of lacrimal glands (LGs) and/or major salivary glands (MSGs) is regarded as a specific finding with diagnostic significance. This study aimed to clarify the influence of this factor on the overall clinical picture of IgG4-DS. Methods We retrospectively reviewed the medical records of 130 patients with IgG4-related disease, 97 of whom were diagnosed with IgG4-DS. We determined their clinical features according to the presence/absence of involvement of ≥2 sets of LGs and/or MSGs and compared the results with those obtained in 33 DS-limited patients. Results The IgG4-DS patients comprised 60 men and 37 women (median age 65 years). The median serum IgG4 level at diagnosis was 548 mg/dL. The patients with involvement of ≥2 sets (n = 44) had significantly more affected organs, lower serum C3 and C4 levels, and a tendency to have higher serum IgG levels and IgG4-RD responder index than did those without it (n = 53). In the 33 DS-limited patients, these two groups had no significant differences in clinical features. Conclusions Involvement of ≥2 sets of LGs and/or MSGs suggests greater systemic disease activity mainly reflected by involvement of more organs.

Abstract

The Simultaneous-color Wide-field Infrared Multi-object Spectrograph (SWIMS) is one of the 1st generation facility instruments for the University of Tokyo Atacama Observatory (TAO) 6.5 m telescope currently being constructed at the summit of Cerro Chajnantor (5,640 m altitude) in northern Chile. SWIMS has two optical arms, the blue arm covering 0.9–1.4 µm and the red 1.4–2.5 µm, by inserting a dichroic mirror into the collimated beam, and thus is capable of taking images in two filter-bands simultaneously in imaging mode, or whole nearinfrared (0.9–2.5 µm) low-to-medium resolution multi-object spectra in spectroscopy (MOS) mode, both with a single exposure. SWIMS was carried into Subaru Telescope in 2017 for performance evaluation prior to completion of the construction of the 6.5 m telescope, and successfully saw the imaging first light in May 2018 and MOS first light in Jan 2019. After three engineering runs including the first light observations, SWIMS has been accepted as a new PI instrument for Subaru Telescope from the semester S21A until S22B. In this paper, we report on details of on-sky performance of the instrument evaluated during the engineering observations for a total of 7.5 nights.

Abstract

Since the University of Tokyo Atacama Observatory (TAO) is located in extremely high altitude (5,640 m.a.s.l.), safety management is one of the most important issues for the project. We have developed safety management program for work at the site in order to prevent medical illness for humans. In this program all staff have to take medical examinations such as Electrocardiogram and Hipobaria before their work starts. The results are reviewed by medical doctor. Only authorized staff can be permitted to work at the site. During stay in site, all staff need to always use oxygen supply because there is only half of the oxygen at the site. It is also important to understand physical workload at the site. Our safety staff reviews it and determines necessary resting time for each worker, e.g. great load works need to work 50 minutes and rest 10 minutes. In addition to low air pressure, very low temperature, extremely high UV radiation, and extremely dry atmosphere should be concerned. Our program requests all staff to use winter clothes, sunblock and UV protection sunglasses and a lot of potable water consumption. Keep communication is also very important to secure the safety. Normal telephone communication is not available as well as internet connectivity. We have established satellite communication as well as UHF internal communication for general works and safety coordination with other observatories in Atacama area.

Abstract

The telescope of the University of Tokyo Atacama Observatory has a primary mirror with a diameter in 6.5m. In order to fabricate the reflecting film initially on the mirror surface and to maintain its optical performance over a long period, a mirror{coating facility will be installed in operation building beside enclosure of the telescope at the summit of Co. Chajnantor (5,640m). The facility consists of mirror coating chamber, cleaning unit for stripping off the old film and clean-up the mirror, and a cart with a lifter for handling the primary mirror cell. Almost all equipment, including the main chamber, was completed by early 2020 and engaged as a mirror coating facility. In order to optimize the coating parameters, comprehensive performance tests were carried out (without primary mirror cell which plays a role of a part of chamber). The evaluation items are how long it takes to reach the target vacuum level and parameters of the current, voltage, and application time during ion bombardment and aluminization. Through iterating test, we were able to obtain each parameter that ultimately met the requirements of the TAO telescope mirror.

Abstract

The Near-Infrared Cross-dispersed Echelle spectrograph (NICE) is a first light instrument for the TAO 6.5 m telescope. The instrument covers a wavelength range of 0.9 to 2.4 µm and has a resolving power of λ/∆λ ~2,600. NICE was first used on the 1.5 m infrared telescope at the National Astronomical Observatory of Japan from 2001 to 2005 and on the 1.6 m Pirka telescope in Japan from 2009 to 2018. We are now upgrading the cryogenics and computer system for the installation of NICE on the TAO telescope. Here we report on the current status and future schedules of instrumentation updates, control system, and modified specifications of NICE for its transfer to the TAO 6.5 m telescope.

Abstract

The fabrication of the new medium-resolution grisms for MOIRCS onboard Subaru 8.2-m Telescope is presented. Our new grisms feature the state-of-the-art gratings that have very high efficiency and wide-spectral coverage manufactured by LightSmyth for the pulse-compression technology. The grating has the peak efficiency of over 96% and can cover the whole H-band wavelength range with over 90%. This is the first-time astronomical application case for the LightSmyth grating. We manufactured the custom-made H-band grism as well as the catalog product J-band grism with a bit less ideal performance. The cooling test of the grisms was done successfully, confirming the stability of the wave-front error over the cooling cycle. On-sky performance test of the new grisms were achieved in July 2020, and we have confirmed the high sensitivity as well as the amazing flatness of the throughput of these grisms. Though we cofirmed the existence of some faint ghosts of the 0-th order light as anticipated, the effect of them are confirmed to be negligible. Abstract

Results of a conceptual design study of ULTAIMTE-Wide Field Imager (WFI) is presented. ULTIMATE-WFI is a near-infrared wide-field imager for the ground-layer adaptive optics system of the Subaru telescope (ULTIMATESubaru) which realizes a 0. 002 seeing size over 200diameter at the Cassegrain focus utilizing a deformable 2ndry mirror. WFI has a 15. 07×15. 07 FoV with a wavelength coverage of 0.9–2.5µm. The FoV is covered by four identical optics, each having a square field lens with 226mm on a side. Its effective FoV is 7. 02 on a side, and is covered by a HAWAII-4RG array detector with a pixel scale of 0. 0011/pix. Effective FoV will be 14. 04×14. 04 or 2070 in total. Spot sizes at a detector plane are less than 0. 001 over the wavelength coverage. Due to the large FoV, vignetting by the telescope structure occurs and an additional cold stop is necessary to block their thermal emission, which causes ~80% vignetting at the edge of the FoV. All the optics are contained in a cylindrical structure to be installed on the Cassegrain focus of the telescope, and kept under cryogenic temperature except for the field lenses. Abstract

Ice rich permafrost is observed at Chajnantor volcano (5,640m a.s.l.) on the University of Tokyo Atacama Observatory (TAO) site. Presence or absence of the permafrost is considered to be requested quite different engineering skills for their infrastructures. Lower altitude boundary is reported to be above 5,079m a.s.l. and maximum active (thawing) layer is 14cm. Minimal seasonal temperature variation, small active layer thickness as the consequences of low numbers of thawing and freezing degree days. Diurnal amplitude results in freeze-thaw cycles only near the surface. Severe frost shattering occurs near the ground surface, producing a dusty, fine-material horizon called a hyper-cryogenic layer. Abstract

We are developing an image-slicer type integral field unit (IFU), SWIMS-IFU, for SWIMS (Simultaneous-color Wide-field Infrared Multi-object Spectrograph), a near-infrared instrument for TAO 6.5 m telescope. SWIMS- IFU divides a field-of-view of 16:006 12:008 into 26 slices with a width of 0:005, which is the largest FoV among near-infrared IFUs on 8 m class telescopes. It is also capable of obtaining entire near-infrared spectra from 0.9 to 2.5 m with R1000 with a single exposure. Because of limitations of space in SWIMS, SWIMS-IFU should fit in a volume of 170 x 220 x 60 mm3, which results in small and complicatedly aligned mirror facets. To reduce alignment procedures, we adopt an ultra-precision cutting technique to fabricate mirror arrays monolithically. We have completed one of the mirror arrays, the slit-mirror array which consists of 26 spherical mirror facets, and confirmed both their surface roughness and shape errors satisfy the requirements. We also have fabricated a prototype of the pupil-mirror array including some elliptical mirror facets and confirmed that the elliptical mirrors have enough surface qualities and produce better image quality than spherical ones by a pinhole imaging test.

Abstract

The University of Tokyo Atacama Observatory Project is to construct a 6.5 m infrared-optimized telescope at the summit of Co. Chajnantor (5640 m altitude) in northern Chile. The telescope optics uses a Ritchey-Chretien type layout, with an under-sized secondary mirror to reduce stray light caused by thermal emission from the telescope structure. The primary mirror is a F/1.25 lightweight borosilicate glass (Ohara E6) mirror with honeycomb structure, which is developed by Steward Observatory Richard F. Caris Mirror Lab. The telescope has two Nasmyth foci and two folded-Cassegrain foci, which can be switched by rotating a tertiary mirror. The final focal ratio is 12.2 with a field of view of 25 arcmin in diameter. The telescope mount is a tripod-disk alt-azimuth mount. Both the azimuth and elevation axes are supported by and run on hydrostatic bearings, and they are driven by friction drives with servo motors, which are controlled by the telescope control system. It also controls the hexapod mount of the secondary mirror and the pneumatic actuators of the primary mirror support to keep good image quality during the observation. An off-axis Shack-Hartmann sensor installed in each focus measures the wavefront aberration of the telescope optics, then the misalignment between the secondary and primary mirrors is corrected by adjusting the hexapod mount while other aberrations are corrected by the deformation of the primary mirror. The force distribution of the actuators for correction will be calculated by fitting the wave-front errors with a series of bending modes of the primary mirror.

Institute of Astronomy, Graduate School of Science, the University of Tokyo is promoting the University of Tokyo Atacama Observatory Project, which is to construct an infrared-optimized 6.5m telescope at the summit of Co. Chajnantor (5640m altitude) in northern Chile. The high altitude and dry climate (PWV-0.5mm) realize transparent atmosphere in the infrared wavelength. The project is now approaching the final phase of the construction. Production of major components are almost completed: Production and preassembly test of a telescope mount and dome enclosure have been completed in Japan, and they are being transported to Chile. Three mirrors, the 6.5m primary, 0.9m secondary, and 1.1m-0.75m tertiary mirrors and their support systems have been all completed and tested in the USA. An aluminizing chamber have been fabricated in China, and its tests have been carried out in Japan. Development of two facility instruments, SWIMS and MIMIZUKU, are also completed. They were transported to the Subaru telescope, successfully saw the first light in 2018, and are confirmed to have the performance as designed. On-site construction work at the summit is now underway. Expansion of a summit access road from the ALMA concession was completed in 2019. Installation of foundation will follow, and then erection of the dome enclosure and a control building. The construction works are delayed by COVID-19, and we expect to complete the dome enclosure by Q3 of 2021. The telescope will be installed inside the dome and see the engineering first light by early 2022.

Abstract

Reduced glucose uptake into the skeletal muscle is an important pathophysiological abnormality in type 2 diabetes, and is caused by impaired translocation of glucose transporter 4 (GLUT4) to the skeletal muscle cell surface. We found a novel xanthene compound, DS20060511, which induces GLUT4 translocation to the skeletal muscle cell surface, thereby stimulating glucose uptake into the skeletal muscle. DS20060511 induced GLUT4 translocation and glucose uptake into differentiated L6-miytubes and into the skeletal muscles of live mice. These effects were completely abolished in GLUT4 knockout mice. Induction of GLUT4 surface translocation by DS20060511 was independent of the insulin signaling pathways including IRS1-Akt-AS160 phosphorylation and IRS1-Rac1-actin polymerization, eNOS pathway and AMPK pathway. Acute and chronic DS20060511 treatment attenuated the glucose intolerance in obese diabetic mice. Taken together, DS20060511 acts as a skeletal muscle specific-GLUT4 translocation enhancer to facilitate glucose utilization. Further studies with DS20060511 would help to develop a novel antidiabetic medicine.

Abstract

Galaxies change their properties as they assemble into clusters. In order to understand the physics behind that, we need to go back in time and observe directly what is occurring in galaxies as they fall into a cluster. We have conducted a narrow-band and $J$-band imaging survey on a cluster CL1604-D at $z=0.923$ using a new infrared instrument SWIMS installed at the Subaru Telescope. The narrow-band filter, NB1261, matches to H$\alpha$ emission from the cluster at $z=0.923$. Combined with a wide range of existing data from various surveys, we have investigated galaxy properties in and around this cluster in great detail. We have identified 27 H$\alpha$ emitters associated with the cluster. They have significant overlap with MIPS 24$\mu$m sources and are located exclusively in the star forming regime on the rest-frame $UVJ$ diagram. We have identified two groups of galaxies near the cluster in the 2D spatial distribution and the phase-space diagram, which are likely to be in-falling to the cluster main body. We have compared various physical properties of star forming galaxies, such as specific star formation rates (burstiness) and morphologies (merger) as a function of environment; cluster center, older group, younger group, and the field. As a result, a global picture has emerged on how the galaxy properties are altered as they assemble into a denser region. This includes the occurrence of mergers, enhancement of star formation activity, excursion to the dusty starburst phase, and eventual quenching to a passive phase., Comment: 19 pages, 15 figures. Abstract

Radar and optical simultaneous observations of meteors are important to understand the size distribution of the interplanetary dust. However, faint meteors detected by high power large aperture radar observations, which are typically as faint as 10 mag. in optical, have not been detected until recently in optical observations, mainly due to insufficient sensitivity of the optical observations. In this paper, two radar and optical simultaneous observations were organized. The first observation was carried out in 2009 to 2010 using Middle and Upper Atmosphere Radar (MU radar) and an image-intensified CCD camera. The second observation was carried out in 2018 using the MU radar and a mosaic CMOS camera, Tomo-e Gozen, mounted on the 1.05-m Kiso Schmidt Telescope. In total, 331 simultaneous meteors were detected. The relationship between radar cross sections and optical V-band magnitudes was well approximated by a linear function. A transformation function from the radar cross section to the V-band magnitude was derived for sporadic meteors. The transformation function was applied to about 150,000 meteors detected by the MU radar in 2009--2015, large part of which are sporadic, and a luminosity function was derived in the magnitude range of $-1.5$ to $9.5$ mag. The luminosity function was well approximated by a single power-law function with the population index of $r = 3.52{\pm}0.12$. Abstract

Complications of diabetes affect tissues throughout the body, including the central nervous system. Epidemiological studies show that diabetic patients have an increased risk of depression, anxiety, age-related cognitive decline, and Alzheimer’s disease. Mice lacking insulin receptor (IR) in the brain or on hypothalamic neurons display an array of metabolic abnormalities; however, the role of insulin action on astrocytes and neurobehaviors remains less well studied. Here, we demonstrate that astrocytes are a direct insulin target in the brain and that knockout of IR on astrocytes causes increased anxiety- and depressive-like behaviors in mice. This can be reproduced in part by deletion of IR on astrocytes in the nucleus accumbens. At a molecular level, loss of insulin signaling in astrocytes impaired tyrosine phosphorylation of Munc18c. This led to decreased exocytosis of ATP from astrocytes, resulting in decreased purinergic signaling on dopaminergic neurons. These reductions contributed to decreased dopamine release from brain slices. Central administration of ATP analogs could reverse depressive-like behaviors in mice with astrocyte IR knockout. Thus, astrocytic insulin signaling plays an important role in dopaminergic signaling, providing a potential mechanism by which astrocytic insulin action may contribute to increased rates of depression in people with diabetes, obesity, and other insulin-resistant states.

Here we show that mice with an adipose-tissue-specific knockout of the microRNA (miRNA)-processing enzyme Dicer (ADicerKO), as well as humans with lipodystrophy, exhibit a substantial decrease in levels of circulating exosomal miRNAs. Transplantation of both white and brown adipose tissue-brown especially-into ADicerKO mice restores the level of numerous circulating miRNAs that are associated with an improvement in glucose tolerance and a reduction in hepatic Fgf21 mRNA and circulating FGF21. This gene regulation can be mimicked by the administration of normal, but not ADicerKO, serum exosomes. Expression of a human-specific miRNA in the brown adipose tissue of one mouse in vivo can also regulate its 3' UTR reporter in the liver of another mouse through serum exosomal transfer. Thus, adipose tissue constitutes an important source of circulating exosomal miRNAs, which can regulate gene expression in distant tissues and thereby serve as a previously undescribed form of adipokine.

Abstract

Summary Insulin and IGF1 signaling are important for adipose tissue development and function; however, their role in mature adipocytes is unclear. Mice with a tamoxifen-inducible knockout of insulin and/or IGF1 receptors (IR/IGF1R) demonstrate a rapid loss of white and brown fat due to increased lipolysis and adipocyte apoptosis. This results in insulin resistance, glucose intolerance, hepatosteatosis, islet hyperplasia with hyperinsulinemia, and cold intolerance. This phenotype, however, resolves over 10–30 days due to a proliferation of preadipocytes and rapid regeneration of both brown and white adipocytes as identified by mTmG lineage tracing. This cycle can be repeated with a second round of receptor inactivation. Leptin administration prior to tamoxifen treatment blocks development of the metabolic syndrome without affecting adipocyte loss or regeneration. Thus, IR is critical in adipocyte maintenance, and this loss of adipose tissue stimulates regeneration of brown/white fat and reversal of metabolic syndrome associated with fat loss.

Abstract

Background IgG4-related dacryoadenitis/sialadenitis (IgG4-DS) is a major organ manifestation of IgG4-related disease (IgG4-RD) and is notable for its lack of a marked male predominance [1]. In IgG4-DS, involvement of two or more sets of lacrimal glands (LGs) and/or major salivary glands (MSGs) is regarded as a specific finding with diagnostic significance [2]. However, the influence of the presence of this finding on clinical features of IgG4-DS has not been well evaluated. Objectives This study aimed to clarify the influence of the involvement of ≥ 2 sets of LGs and/or MSGs on clinical features of IgG4-DS. Methods We retrospectively reviewed the medical records of 130 patients with IgG4-RD at a single center in Japan. Among them, 97 patients were diagnosed with IgG4-DS. We divided the 97 patients into Group A (n=44) with involvement of ≥ 2 sets of LGs and/or MSGs and Group B (n=53) with involvement of one set of LGs or MSGs or unilateral gland involvement. We compared the clinical features of Group A and B. Moreover, 33 IgG4-DS patients without extra-lacrimal/salivary gland lesions (DS-limited patients) were divided into Group a (n=14) and b (n=19) according to the presence/absence of the involvement of ≥ 2 sets of LGs and/or MSGs respectively, and their clinical features were also compared. Results The IgG4-DS patients comprised 60 men and 37 women (mean age 64.1 years). The mean follow-up period was 61.6 months, and the mean serum IgG4 level at diagnosis was 757 mg/dL. Group A had significantly more affected organs (3.5 ± 1.6 vs 2.7 ± 1.6, P=0.010), lower serum C3 and C4 levels (83.6 ± 29.5 vs 95.0 ± 25.2, P=0.018, and 16.5 ± 9.8 vs 21.4 ± 9.5, P=0.034, respectively), and a tendency to have higher serum IgG levels (2,509 ± 1,066 vs 2,165 ± 911, P=0.075) and IgG4-RD responder index (14.7 ± 6.7 vs 12.2 ± 6.5, P=0.050) than did Group B. In the 33 DS-limited patients, compared with Group b, Group a had no significant differences in clinical features including the proportion of patients with serum IgG4 elevation [≥ upper limit of normal (ULN), 85.7% vs 78.9%, P=1.000; ≥ 2× ULN, 64.3% vs 47.4%, P=0.482; ≥ 5× ULN, 28.6% vs 21.1%, P=0.695]. Conclusion Involvement of two or more sets of lacrimal glands and/or major salivary glands suggests greater systemic disease activity mainly due to involvement of more organs. Background IgG4-related periaortitis/periarteritis (IgG4-PA) is a major organ manifestation of IgG4-related disease (IgG4-RD) [1]. In the affected aorta/artery, arteriosclerotic changes are frequently observed [2]. However, the influence of arteriosclerosis on IgG4-PA and aneurysmal changes of the affected lesions has not been well evaluated. Objectives This study aimed to clarify the relationship of arteriosclerosis with periaortic/periarterial lesions and their aneurysmal changes. Methods We retrospectively investigated the medical data, including the presence of IgG4-PA at diagnosis, new development of IgG4-PA during the clinical course, arteriosclerotic and aneurysmal changes of the affected lesions, and classic risk factors for arteriosclerosis in 130 patients with IgG4-RD at a single center in Japan. The relationship of arteriosclerosis with the periaortic/periarterial lesions and their aneurysmal changes was statistically analyzed. Results Of the 130 patients with IgG4-RD, 44 comprising 39 men and 5 women (mean age 67.4 years) were diagnosed with IgG4-PA. The mean follow-up period was 72.8 months, and the mean serum IgG4 level at diagnosis was 851 mg/dL. Abstract

Impaired insulin signaling in brain leads to both metabolic and behavioral abnormalities. To understand the role of insulin on gene expression in the brain, we performed hyperinsulinemic-euglycemic clamps using physiological doses of insulin in C57BL/6 mice, and after 3 hours collected different brain regions and analyzed their transcriptomes by RNA Seq. In response to peripheral insulin, 1851 genes were up- or down-regulated in the hypothalamus by at least 50% (all P < 0.01). Other regions showed different responses with 205 genes significantly regulated by ≥ 50% in hippocampus and 131 regulated in nucleus accumbens. Insulin upregulated the pathway for very long chain fatty acyl-CoA synthesis in all three regions, but most insulin responsive genes were region specific. Thus, in hypothalamus, insulin modulated metabolism by suppressing the glycolysis and pentose phosphate pathway, while increasing expression of genes in the pyruvate dehydrogenase complex and cholesterol biosynthesis. Conversely, mice with streptozotocin diabetes displayed decreased expression of genes involved in the regulation of pyruvate dehydrogenase complex, very long-chain fatty acyl-CoA and cholesterol biosynthesis in hypothalamus, all of which were normalized by exogenous insulin. In addition, insulin regulated many genes in the hypothalamus involved in neurotransmission, including enhancing the expression of multiple GABA-A receptor subunits and suppressing many neuropeptides. Based on the transcriptional signature, changes in gene expression by insulin involve neurons, astrocytes, oligodendrocytes, microglia and endothelial cells. In summary, peripherally administered insulin rapidly and potently regulates expression of genes involved in neurotransmission, neuromodulation and brain metabolism, especially in the hypothalamus. Abstract

We present properties of two types of bulges (classical- and pseudo- bulges) in 20 luminous infrared galaxies (LIRGs) observed in the near infrared of the $H$, $K_s$ and 1.91$\mu$m narrow-band targeting at the hydrogen Pa$\alpha$ emission line by the University of Tokyo Atacama Observatory (TAO) 1.0 m telescope. To classify the two types of bulges, we first perform a two-dimensional bulge-disk decomposition analysis in the $K_\mathrm{s}$-band images. The result shows a tentative bimodal distribution of S\'ersic indices with a separation at $\log(n_b)\sim0.5$, which is consistent with that of classical and normal galaxies. We next measure extents of the distribution of star forming regions in Pa$\alpha$ emission line images, normalized with the size of the bulges, and find that they decrease with increasing S\'ersic indices. These results suggest that star-forming galaxies with classical bulges have compact star forming regions concentrated within the bulges, while those with pseudobulges have extended star forming regions beyond the bulges, suggesting that there are different formation scenarios at work in classical and pseudobulges., Comment: 10 pages, 25 figures, 1 tables, accepted for publication in PASJ

Abstract

We report herein the synthesis and structure-activity relationships (SAR) of a series of pyridazine derivatives with the activation of glucose transporter type 4 (GLUT4) translocation. Through a cell-based phenotype screening in L6-GLUT4-myc myoblasts and functional glucose uptake assays, lead compound 1a was identified as a functional small molecule. After further derivatization, the thienopyridazine scaffold as the central ring (B-part) was revealed to have potent GLUT4 translocation activities. Consequently, we obtained promising compound 26b, which showed a significant blood glucose lowering effect in the severe diabetic mice model (10-week aged db/db mice) after oral dosing even at 10 mg/kg, implying that our pyridazine derivatives have potential to become novel therapeutic agents for diabetes mellitus.

Abstract

Novel compounds based on 1a were synthesized with the focus of obtaining agonists acting upon peripheral BRS-3. To identify potent anti-obesity compounds without adverse effects on the central nervous system (CNS), a carboxylic acid moiety and a labile carboxylic ester with an antedrug functionality were introduced. Through the extensive synthetic exploration and the pharmacokinetic studies of intravenous administration in mice, the ester 2b was selected owing to its most suitable pharmacological profile. In the evaluation of food intake suppression in C57BL/6N mice, 2b showed significant in vivo efficacy and no clear adverse effects on blood pressure change in dogs administered the compound by intravenous infusion.

Abstract

Tomo-e Gozen (Tomo-e) is a wide field optical camera for the Kiso 1.05 m f/3.1 Schmidt telescope operated by the University of Tokyo. Tomo-e is equipped with 84 chips of front-illuminated CMOS image sensors with a microlens array. The field of view is about 20 square degrees and maximum frame rate is 2 fps. The CMOS sensor has 2160x1200 pixels and a size of pixel is 19 microns, which is larger than those of other CMOS sensors. We have evaluated performances of the CMOS sensors installed in Tomo-e. The readout noise is 2.0 e- in 2 fps operations when an internal amplifier gain is set to 16. The dark current is 0.5 e-/sec/pix at room temperature, 290K, which is lower than a typical sky background flux in Tomo-e observations, 50 e-/sec/pix. The efficiency of the camera system peaks at approximately 0.7 in 500 nm.
