Background: Health consciousness in daily life is very important because lifestyle habits, such as dietary and exercise habits, sleep, and smoking, are involved in the development and progression of lifestyle-related diseases, such as cancer, heart diseases, stroke, liver diseases, kidney diseases, and diabetes. Purpose: In this study, we surveyed to see if the health consciousness increased and what behaviors in daily life changed in students of a nursing school (Seiwa Nursing College, Tokyo, Japan) after they learned medical care knowledge required to become a nurse. Results: After learning medical care knowledge for 3 years, the students were found to have a higher level of health consciousness in the third year than they did in the first year. They applied the medical care knowledge they learned in daily life. All data were collected in 2019. Conclusions: The medical care knowledge learned in a nursing school was demonstrated to serve as information to increase the health consciousness.

[Purpose] It has been reported that exercise affects skeletal muscle in the chronic obstructive pulmonary disease (COPD) disease model. In this study, we examined the effects of neuromuscular electrical stimulation (NMES) in skeletal muscle on alveoli and cytokines. [Materials and Methods] We used twenty wild-type mice, randomly divided into three groups: Group A: Control (non-COPD, non-amyotrophia, non-NMES), Group B: COPD, amyotrophia with NMES and Group C: COPD, amyotrophia without NMES. Among those, a group of mice with ages from 12 to 14 weeks were used to create a chronic obstructive pulmonary disease (COPD) model, a group of mice with ages from 15 to 16 weeks was used to create a disuse syndrome by hind limb suspension, and a group of mice with ages from 17 to 28 weeks (12 weeks) were used to implement NMES. In this study, we used the real-time PCR method to assess the mRNA expression levels. We also conducted morphological analysis, assessed macrophage expression level by staining (general staining and immunostaining), and employed spirometry. [Results] Our study results showed significant decreases in Interleukin-6 (IL-6) levels in the lungs and muscle RING-finger protein-1 (MuRF1) in the muscles. Moreover, the pulmonary stromal macrophage marker (F4/80) and the protease marker (MMP12) showed significantly decreased expression, while no change was observed in the morphological of the alveolar spaces (mean linear intercept). [Conclusion] On the basis of these findings, our study reveals that NMES affects cytokines and macrophages in COPD skeletal muscle atrophy.

Activity-dependent reorganizations of central neuronal synapses are thought to play important roles in learning and memory. Although the precise mechanisms of how neuronal activities modify synaptic connections in neurons remain to be clarified, the activity-induced neuronal presynaptic proteins such as synaptotagmin1 may contribute to the onset of synaptic remodeling. To understand better the physiological roles of synaptotagmin1, we first examined the prolonged effects of neuronal stimulation capable of inducing synaptotagmin1 on the distribution of a postsynaptic proteins (PSD) protein Homer1c by immunostaining. Previously we found that glutamate stimulation induced other postsynaptic proteins, such as postsynaptic density-95 (PSD95), a biphasic change with an initially diffuse distribution after 30 min to 1 hr, followed by reassembly to more than the original level after 4–8 hr, suggesting that glutamate stimulation induces a global biphasic alteration in synaptic structures. To dissect further the functions of synaptotagmin1 in the activity-induced synaptic remodeling, short hairpin RNA (shRNA) vectors that specifically block the expression of endogenous synaptotagmin1 were constructed. When the shRNA of synaptotagmin1 was introduced to the neurons, the activity-induced changes were almost completely suppressed. We found that synaptotagmin1 contributes to the postsynaptic remodeling in a retrograde manner. Our data indicate that synaptotagmin1 regulates the activity-induced biphasic changes of post- and presynaptic sites. © 2013 Wiley Periodicals, Inc.

Background The role of the piriform cortex (PC) in olfactory information processing remains largely unknown. The anterior part of the piriform cortex (APC) has been the focus of cortical-level studies of olfactory coding, and associative processes have attracted considerable attention as an important part in odor discrimination and olfactory information processing. Associational connections of pyramidal cells in the guinea pig APC were studied by direct visualization of axons stained and quantitatively analyzed by intracellular biocytin injection in vivo. Results The observations illustrated that axon collaterals of the individual cells were widely and spatially distributed within the PC, and sometimes also showed a long associational projection to the olfactory bulb (OB). The data showed that long associational axons were both rostrally and caudally directed throughout the PC, and the intrinsic associational fibers of pyramidal cells in the APC are omnidirectional connections in the PC. Within the PC, associational axons typically followed rather linear trajectories and irregular bouton distributions. Quantitative data of the axon collaterals of two pyramidal cells in the APC showed that the average length of axonal collaterals was 101 mm, out of which 79 mm (78% of total length) were distributed in the PC. The average number of boutons was 8926 and 7101, respectively, with 79% of the total number of boutons being distributed in the PC. The percentage of the total area of the APC and the posterior piriform cortex occupied by the average distribution region of the axon collaterals of two superficial pyramidal (SP) cells was about 18 and 5%, respectively. Conclusion Our results demonstrate that omnidirectional connection of pyramidal cells in the APC provides a substrate for recurrent processes. These findings indicate that the axon collaterals of SP cells in the PC could make synaptic contacts with all granule cells in the OB. This study provides the morphological evidence for understanding the mechanisms of information processing and associative memory in the APC. Although anatomical knowledge is vital in nursing practice, it is considered to be difficult to understand. Therefore, we attempted to arrange anatomical knowledge based on the framework of nursing to enable students to learn anatomical knowledge. Anatomical knowledge associated with eight lifestyle issues were considered to be necessary by nursing and anatomy researchers were recorded, and concordance rates were examined. Therefore, we confirmed the validity of the learning content. However, results suggested that a prospective, multifaceted study is required.

This study aimed to evaluate changes in glucose metabolism at the early stage and onset of diabetes in Otsuka Long-Evans Tokushima Fatty (OLETF) rats. Specifically, after the oral administration of [1, 2, 3-13C]glucose, the levels of exhaled 13CO2, which most likely originated from pyruvate decarboxylation and tricarboxylic acid, were measured. Eight OLETF rats and eight control rats (Long-Evans Tokushima Otsuka [LETO]) were administered 13C-glucose. Three types of 13C-glucose breath tests were performed thrice in each period at 2-week intervals. [3-13C]glucose results in a 13C isotope at position 1 in the pyruvate molecule, which provides 13CO2. The 13C at carbons 1 and 2 of glucose is converted to 13C at carbons 2 and 1 of acetate, respectively, which produce 13CO2. Based on metabolic differences of the labeled sites, glucose metabolism was evaluated using the results of three breath tests. The increase in 13CO2 excretion in OLETF rats was delayed in all three breath tests compared to that in control rats, suggesting that OLETF rats had a lower glucose metabolism than control rats. In addition, overall glucose metabolism increased with age in both groups. The utilization of [2-13C]glucose was suppressed in OLETF rats at 6-12 weeks of age, but they showed higher [3-13C]glucose oxidation than control rats at 22-25 weeks of age. In the [1-13C]glucose breath test, no significant differences in the area under the curve until 180 minutes (AUC180) were observed between OLETF and LETO rats of any age. Glucose metabolism kinetics were different between the age groups and two groups of rats; however, these differences were not significant based on the overall AUC180 of [1-13C]glucose. We conclude that breath 13CO2 excretion is reduced in OLETF rats at the primary stage of prediabetes, indicating differences in glucose oxidation kinetics between OLETF and LETO rats.

To understand the functional organization of the piriform cortex (PC), the axon collaterals of three pyramidal cells in layer IIb of the anterior PC and one pyramidal cell in layer IIb of the posterior PC were labeled and quantitatively analyzed by intracellular biocytin injection in the guinea pig. Single pyramidal cells in the anterior and posterior PCs have widely distributed axon collaterals, which exhibit little tendency for patchy concentrations inside as well as outside the PC. The total lengths of the axon collaterals of the three fully analyzed pyramidal cells ranged from 68 to 156 mm, more than 50% of which were distributed in the PC. The total number of boutons of the three cells ranged from 6000 to 14,000, 5000-7000 of which were distributed in the PC. It was estimated that individual pyramidal cells in layer IIb form synaptic contacts with 2200 to 3000 other pyramidal cells in the PC, indicating that single pyramidal cells in layer IIb receive input from a large number of other pyramidal cells. This high connectivity of the network of pyramidal cells in the PC can be regarded as the neural network operating parallel distributed processing, which may play an important role in experience-induced enhancement in odorant discrimination in the PC.

To study the various types of neurons in layer IIa in the piriform cortex (PC) and the spatial distribution of their axons, axon collaterals of three neurons in layer IIa were labeled and quantitatively analyzed by intracellular injection of biocytin in the guinea pig. Individual neurons have highly distributed axon collaterals, which display a little tendency toward patchy concentrations inside as well as outside the PC. One semilunar cell in the posterior PC had 54-mm-long axon collaterals and 4,200 boutons, out of which 2,100 (49% of the total number of boutons) were distributed in the PC. One semilunar-pyramidal transitional cell in the posterior PC had 256-mm-long axon collaterals and 23,000 boutons, out of which 16,100 (70% of the total number of boutons) and 4,000 (18% of the total number of boutons) were respectively distributed in all layers and in layer Ia of the PC. One multipolar cell in the posterior PC had 188-mm-long axon collaterals and 18,000 boutons, out of which 13,700 (78% of the total number of boutons) were distributed in the PC. Our results revealed that the connection patterns of individual cells in layer IIa have most of the features required for an associative neural network, which may function as a content-addressable memory for the association of odor stimuli.

The aim of the present postnatal developmental study was to determine densities of unique genital corpuscles (GCs) in glans penis of developing and aged rats. GCs were identified as corpuscular endings consisting of highly branched and coiled axons with many varicosities, which were immunoreactive for protein gene product 9.5. In addition, GCs were immunoreactive for calcitonin gene-related peptide and substance P, but not for vasoactive intestinal polypeptide and neuropeptide Y. GCs were not found in the glans penis of 1 week old rats. Densities of GCs were low at 3 weeks, significantly increased at 5 and 10 weeks, reached the peak of density at 40 weeks, and tended to decrease at 70 and 100 weeks. Sizes of GCs were small in 3 weeks old rats, increased at 5 and 10 weeks, reached the peak-size at 40 weeks and reduced in size at 70 and 100 weeks. Considering sexual maturation of the rat, the results reveal that GCs of the rat begins to develop postnatal and reaches to the peak of their development after puberty and continues to exist until old age, in contrast to prenatal and early postnatal development of other sensory receptors of glabrous skin.

Sub-harmonic vibration of clamped-free thin cylindrical shells immersed in fluid under seismic excitation was investigated by numerical simulation. The conditions when the sub-harmonic vibration occurs under narrow-band random waves were compared with those under sisusoidal wave excitations : the critical pressures under narrow-band random waves were higher than those under sinusoidal waves. It was also found that the frequency componets other than the dominant frequency of a narrow-band random wave have a negligible effect on the occurrence of the sub-harmonic vibration. These two findings provide the basis for a simple equation for predicting the critical pressure under narrow-band random waves.

The distribution, morphological features, and postnatal development of parvalbumin (PV) immunoreactive neurons in the main olfactory bulb (MOB) of the house musk shrew, Suncus murinus, were studied to report for the first time on PV positive bulbar interneurons in the order Insectivora. In adult animals, PV neurons are distributed in the glomerular layer (GL), the external plexiform layer (EPL), the internal plexiform layer (IPL) and the granule cell layer (GCL) of the MOB. These neurons were identified as a subpopulation of periglomerular cells and perinidal cells [Alonso et al., 1995] in the GL and at the GL-EPL border, respectively, and as bipolar and multipolar neurons in the EPL and four types of the interneurons (horizontal cells, Cajal cells, Golgi cells, and bitufted cells) in the layers deeper than the mitral cell layer. During development of PV neurons, neurons exhibiting extremely faint PV immunoreactivity first appeared in the GCL at postnatal day 14 and increased markedly in number and intensity of their PV immunoreactivity from postnatal days 14 to 28. At postnatal day 21, PV neurons were identified as periglomerular cells in the GL, perinidal cells at the GL-EPL border, and morphologically unidentifiable neurons in the EPL, IPL and GCL. At postnatal day 28, PV neurons exhibited a nearly adult pattern with respect to distribution and structural features. The present results strongly suggest that a wide variety of PV positive neurons in the MOB of the house musk shrew may develop postnatally.

Layer 10 neurons of the chick tectum were morphologically investigated. The layer 10 neurons displayed heterogeneous immunoreactivities to calcium-binding proteins (CaBPs). Calbindin (CB)-immunoreactive (ir) neurons had pyramidal or round somata, primarily found in layers 5, 9, and 13. Parvalbumin (PV)-ir neurons were of various shapes with small to large somata (109.7 ± 48.6 μm 2 ) that were located mainly in layers 4 and 10. Calretinin (CR)-ir neurons had small to middle-sized somata (79.3 ± 9.7 μm 2 ) located primarily in layers 10 and 13, and most of them were similar to typical radial cells in size and shape. Two distinct types of neurons that projected to the nucleus geniculatus lateralis, pars ventralis (GLv) and ventral thalamus were demonstrated in layer 10. Type 1 cells had small to middle-sized somata (74.3 ± 33 μm 2 ), and each cell had a single apical dendrite that ramified into bush-like branches in layer 7. These cells corresponded to CR-ir neurons and radial cells in size and shape. Type 2 cells had larger somata (124.7 ± 52.6 μm 2 ), and their shapes were pyramidal, polygonal, or oval. They had multiple obliquely ascending dendrites that ramified into bush-like branches in layer 7. These cells often appeared similar to PV-ir neurons.

An upright stainless steel circular cylinder with a mass attached at midlength is excited seismically in the horizontal direction using a vibration table. A well-known seismic wave called EI Centro 1940 NS is applied to excite the test specimen. Fourier components over 10Hz of the seismic wave are cut off and the duration is shortened to 30%, 40%, or 50%. The eccentricity of the center of gravity of the attached mass is varied and the amplitude of the modified El Centro 1940 NS is increased incrementally. Parametric excitation is observed for each combination of eccentricity of the attached mass and time scale of the seismic wave.

Cylindrical shells subjected to lateral loads buckle in shear or bending mode.s The bending buckling mode is a diamond-shaped pattern or an elephant-foot bulge at the edge. The effects of geometrical imperfections and combined loading on buckling loads and postbuckling behaviour are investigated by developing a special purpose finite element analysis program using the 8-node isoparametric shell element. Four types of imperfections are considered and the relationships between the imperfection amplitude and the bucking load are obtained. Three types of loading are taken into consideration. For short cylindrical shells, shear buckling is dominant but elephant-foot bulges appear with an increase in bending moments and axial loads. Effects of axial loads on shear buckling and elephant-foot bulges are calculated. Postbuckling behaviour which is closely related to imperfection sensitivity is also investigated.

Cadavers in gross anatomy laboratories at most medical schools are conventionally embalmed in formaldehyde solution, which is carcinogenic to humans. Medical students and instructors are thus exposed to formaldehyde vapors emitted from cadavers during dissection. To reduce high formaldehyde concentrations in the breathing zone above cadavers being examined by anatomy medical students provisionally, dissection beds were located under existing admission ports on the ceiling to supply cooled fresh air from the admission port blowing downward on to the cadaver. In all cases, compared to normal condition, the downward flow of cooled fresh air from an admission port reduced formaldehyde concentrations by 0.09-0.98 ppm and reduced to 12.6-65.4% in the air above a cadaver in the breathing zone of students. The formaldehyde concentrations above cadavers under admission ports were not more than the formaldehyde concentrations between beds representing the indoor formaldehyde concentrations. Although the application of an existing admission port on the ceiling in this study did not remove formaldehyde, the downflow of cooled fresh air using this system reduced the formaldehyde concentration in the air above cadavers being attended by anatomy students during dissections. These results suggest the need for reducing formaldehyde levels in gross anatomy laboratories using fundamental countermeasures in order to satisfy the guidelines of 0.08 ppm established by the World Health Organization and the Japan Ministry of Health, Labor and Welfare.

Cadavers for gross anatomy laboratories are typically embalmed in formaldehyde. Thus, medical students and instructors are exposed to formaldehyde vapors emitted from cadavers during dissection. In an attempt to improve the dissection environment, we examined indoor formaldehyde concentrations in a gross anatomy laboratory. Air samples were taken from 20, 110, 160, and 230 cm above the floor between dissection beds to represent areas near the floor, in the breathing zone of sitting students, in the breathing zone of standing students, and near the ceiling, respectively. Formaldehyde vapors were thoroughly diffused from the floor to the ceiling, suggesting that medical students are exposed to similar concentrations of formaldehyde based on distance from the floor. Computational fluid dynamics showed that cadavers are warmed by overhead fluorescent lights and the body heat of anatomy students, and indicated that the diffusion of formaldehyde vapors is increased by lighting and the body temperature of students. Computational fluid dynamics showed that gentle convection from anatomy students and cadavers carry formaldehyde vapors upward; downward flow near admission ports diffuse formaldehyde vapors from the ceiling to the floor in the anatomy laboratory.

The displacement functions of an axisymmetric shell with eccentrically attached masses are expanded using the free vibration modes of the axisymmetric shells without attached masses. These base vectors for expansion are calculated using a conical shell finite element. The expanded functions are used to express the strain energy and kinetic energy. The contribution of the rocking motion of the attached masses to the kinetic energy is considered. Also the effects of contained liquid are included. The strain energy and kinetic energy are substituted into Lagrange's equations to obtain natural frequencies, vibration modes and response for base excitation. Analytical results using the present method show good agreement with analytical results obtained by other methods or experimental results.

The parametric resonance of an axisymmetric shell-lumped weight system subjected to periodic acceleration in the radial direction is analyzed. Symmetric and asymmetric free-vibration modes of the system are derived using the free-vibration modes of the axisymmetric shell without lumped weight. The aforementioned eigenmodes are calculated by employing a conical shell finite element. Modal Mathieu equations of the system are derived, and the parametric resonance regions of the system are calculated using Bolotin's method, considering the effect of translational motion of the lumped weight in the radial direction. Also, a circular cylindrical shell with a lumped weight is experimentally investigated using a vibration table. Numerical results of the parametric resonance regions of the system show good agreement with experimental results.

Cadavers for gross anatomy laboratories are conventionally embalmed by formaldehyde (FA) solution in most medical schools. Thus, medical students and instructors are exposed to FA vapors emitted from cadavers during dissection. As a basic survey for the improvement of the dissection environment, we examined FA concentration in the gross anatomy laboratory during the 2006 academic year at the Faculty of Medicine of Toho University. Air samples were taken from 20 cm above a cadaver as breathing zone, and above a desk between cadavers as indoor FA concentration. FA concentrations in the breathing zone were ranged from 0.24 to 3.04 (mean 1.71) ppm during systematic anatomy, and from 0.72 to 1.60 (mean 1.16) ppm during neuroanatomy, and indoor FA concentration ranged from 048 to 1.11 (mean 0.76) ppm and from 0.21 to 0.23 (mean 0.22) ppm, respectively. These results showed that medical students and instructors are exposed to higher concentrations of FA than allowed by the guidelines of the Japan Ministry of Health, Labor and Welfare, and suggested the need to reduce FA levels in the gross anatomy laboratory.

In order to investigate converging projections originating from adjacent populations of cortical neurons, injections of two different anterograde tracers, biotinylated dextranamine (BDA) and Phaseolus vulgaris leucoagglutinin (PHA-L), were made in close proximity. When the two injection sites were separated by around 500 μm and the time between injections was 1–4 h, BDA-labeling of neuronal elements was found not only at the BDA injection site but also at the PHA-L injection site. This false-positive BDA labeling of neurons at the PHA-L injection site was so intense that labeled axons could be traced, both into the neighboring cortical gray matter and into white matter. Increasing the separation distance to 1000 μm resulted in much fewer falsely positive labeled neurons at the PHA-L injection site. Even more effective was extending the time interval between the two injections. Thus, if the BDA injection preceded the PHA-L injection by more than 12 h, virtually no false-positive labeling was associated with the PHA-L injection site. These procedures may be applied to other combinations of anterograde tracers, such as BDA with tetramethylrhodamine-conjugated dextran amine.

The colocalization of serotonin and aromatic L-amino acid decarboxylase (AADC) in the avian pancreatic polypeptide-containing PP-cells and glucagon-storing A-cells of the chicken endocrine pancreas was investigated using combined pre-embedding immuno-peroxidase and post-embedding immunogold electron microscopic immunocytochemistry. The avian pancreatic polypeptide-immunoreactive cells manifested by the labeling of immunogold particles on secretory granules were also immunoreactive with antisera directed against serotonin and AADC, an enzyme involved in the synthesis of serotonin. In PP-cells immunoreactivity against the anti-serotonin serum was stronger in secretory granules than in the cytoplasmic matrix, whereas immunoreaction with the anti-AADC serum was observed to be more intense in the cytoplasmic matrix. Immunoreactions with the serotonin and AADC antisera were also found in secretory granules of glucagon-storing A-cells. These results indicate that serotonin is co-stored within secretory granules of both A- and PP-cells, and that AADC is localized within secretory granules of A-cells, and may be present in the cytoplasmic matrix of PP-cells. It is probable that serotonin is synthesized and released simultaneously with secretory granules from both A- and PP-cells of the chicken endocrine pancreas.

Buckling failures are often observed in tanks subjected to seismic load. In this paper, dynamic buckling is studied, using small, plastic cylindrical tanks made of polyester sheets. Static buckling tests on the fluid-filled tanks subjected to lateral load, and dynamic buckling tests under harmonic excitation using a shaking table are carried out, and the buckling modes are compared. The stress resulting from the frequency response (the circumferential wave numbers n=1 for horizontal excitation and n=0 for vertical excitation) is calculated using an axi-symmetric shell finite element program, which includes the effect of fluid structure interaction. Static buckling stress criteria are applied to the above calculations in order to predict the occurrence of buckling. The predictions are compared with the test results and they prove to be fairly satisfactory in the restricted region the natural frequency of the fundamental mode.

An approximate method is proposed for the analysis of liquid containers considering the effects of the compressibility of the liquid. This method is based on the expansion of the vibration mode of compressible fluid-structure interaction using the coupled modes of incompressible fluid-structure system. Further simplified equations are derived assuming that the vibration modes are not changed when fluid compressibility is included. This paper also shows a technique to avoid the drawback of the mass matrix becoming the full matrix when fluid-structure interaction is considered. It consists of solving simultaneous equations utilizing the sparsity of fluid stiffness matrix without composing the full matrix in the subspace interation method for eigenvalues. Calculated natural frequencies and frequency response spectra, including compressibility of fluid, agree well with experimental results.
