Housing is a significant determinant of health and is widely accepted as a key solution to address some of the health disparities that exist among the homeless. It is estimated that 150 million people worldwide are homeless, and approximately 1.8 billion lack adequate housing. However, understanding of how housing has a positive impact on the health of the homeless remains unclear and underdeveloped. This systematic review investigates intervention studies that report on the physical and mental health effects of housing homeless persons. A search of PubMed, PsycINFO, EBSCOHost-Academic Search Complete and the Cochrane Library was conducted for peer-reviewed articles published in English from 1999 to 2020 that had a combination of at least one housing intervention and health outcome, with a homeless sample. Three previous reviews and 24 studies were included for analysis. Most of the studies (n = 20) encompassed permanent supportive housing interventions that emphasised placing homeless people with mental illness directly into affordable housing with access to support services. The primary health outcomes reported were general physical and mental health, well-being, and quality of life. Despite inconsistent findings and significant issues identified in the reviewed literature, housing (in the short term) improves some aspects of health in homeless populations with human immunodeficiency virus, anxiety and depression.

Several lines of research suggest that autism is a neurological phenomenon, but the precise associations between neurological activity and the key diagnostic symptoms of autism are yet to be completely clarified. This study examined EEG connectivity and Sensory Features (SF) in a sample of young autistic males by examining bi-directional neural connectivity between separate brain regions as the key potential correlate of SF. Forty male autistic participants aged between 6 and 17 years, with an IQ of at least 70, underwent EEG measurements of their Frontal, Occipital and Temporal region responses to low-, medium-, and high-intensity audiovisual stimulus conditions. Investigation of the neurological underpinnings of the diagnostic symptoms for Autism Spectrum Disorder (ASD) represents a potential pathway towards a biomarker for this disorder. One of the key symptoms of ASD is Sensory Features (SF), which refers to the difficulties that autistic people experience with particular kinds of environmental stimuli. Studies using eeg measures of neural connectivity across various regions of the brain hold promise in identifying how the autistic brain reacts to its environment. This commentary identifies several ‘participant’ and ‘measurement’ methodological issues that need to be adequately addressed in SF-eeg connectivity studies, and applies these comments to a sample of five previous studies. Recommendations are made for future research procedures.

Headaches have been studied and treated since nearly 7,000 BC because of their significant global impact. Current headache treatment modalities are various and have a wide variety of targets, but medications are the most common. Since conventional medical treatments have several side effects, alternative remedies such as osteopathic manipulative treatment (OMT) should be considered. OMT can assist in the management of various health conditions, such as low back pain, neck pain, and headaches. The purpose of this scoping review is to evaluate recent findings regarding the efficacy of OMT modalities in the management of headaches such as tension-type headaches (TTH) and migraines. This study was designed as a scoping review to gather evidence on the efficacy of OMT modalities in the management of headaches. Following PRISMA guidelines, four databases were used to search for articles published between 2010 and 2022 that reported the use of OMT and manual therapy for TTH and migraines. Databases used include Embase, PubMed, Medline, and Web of Science. The following keywords were used: treatment, therapy, Headache, migraine, craniosacral, muscle energy, myofascial release, trigger point, osteopathic, and manipulation. The initial search yielded 473 unique articles after removing duplicates. After screening based on the inclusion and exclusion criteria, and after further analysis, 15 articles were selected. Data reports of OMT and manual therapy efficacy and/or effectiveness in treating TTH and migraine were analyzed. Articles included were randomized control studies (13 of 15, 86.6%), one pilot study (one of 15, 6.7%), and one case series (one of 15, 6.7%), which were divided into TTH (nine of 15, 60%) and Migraine Headaches (six of 15, 40%). All articles reported significant headache improvement in at least one measurement. Of all treatments analyzed, single technique interventions (seven of 15, 47%) and multiple technique interventions (eight of 15, 53%) were identified. Among the techniques used, Myofascial Release was the most common (nine of 15, 60%). The articles presented provide evidence of the significant benefits of manual therapy. Because of the limitations of traditional medicine, OMT can be used either as an alternative or adjuvant therapy for headaches. Evidence suggests the positive impact it can provide on headache management, but the number of randomized control trials and population samples should be increased to support its recommendation. This demonstrates how different osteopathic techniques can provide therapeutic effects on TTH, MH, and potentially other types of headaches. A preference for myofascial release was observed, which can be due to the fast relief from the physiologic effect on tissue movement. This review study demonstrates the benefits OMT has on decreasing headache frequency, intensity, and duration in TTH and migraines. OMT has shown to be beneficial, especially for patients seeking alternative non-pharmaceutical and non-invasive treatments. Further studies are needed to evaluate the effects of different OMT techniques, and different combinations of treatments, on other types of headaches.

In order to evaluate the effects of specific forms of childhood maltreatment (CM) upon adult C-reactive protein (CRP) concentrations, and to further describe the potentially confounding role that recent life stress and depression hold in that relationship, 221 participants from rural Australia (M age = 44yr, SD = 17.8yr) completed self-report questionnaires and provided a blood sample. There were no sex differences in any variables across the 91 males and 130 females, but depression status did confound the association between global CM and CRP. The specific aspect of CM was identified as physical and mental health abuse, and this was significantly associated with CRP level in participants with depressive symptoms and those without. There was no significant confound from recent life stressors. Results hold implications for the diagnosis of CM-related CRP elevation and (potentially) depression.

Purpose Hormonal and inflammatory factors have been suggested as potentially influencing depressive state and depressive symptoms, but rarely compared for their relative contribution to these states and to specific depressive symptoms. This study examined cortisol:C-reactive protein (CRP) ratio, plus cortisol and CRP separately, as correlates of global depression and fatigue-related depression. Patients and methods One hundred and twenty-six community volunteers from rural Australia provided saliva and serum samples, and also completed a depression inventory. Results There was a significant correlation between cortisol:CRP ratio and depression-related fatigue, and this resolved to the effects of CRP rather than cortisol. Most of the variance in this association came from patients who were "depressed", and there were no significant gender associations. Conclusion Inflammation, rather than HPA-axis activity, was associated with depression-related fatigue, supporting a model that places inflammation as a contributor to one of the major symptoms and predictors of depression. Individualization of therapy for depression-related fatigue in chronically stressed or physically ill patients might benefit from future research into cytokine therapy.

Water shortage and soil degradation are common environmental stressors encountered in the Mediterranean area. We evaluated how different soil moisture levels, dependent on distance from drip irrigation points, impact on the biological, chemical and physical properties of citrus soil under organic and inorganic fertilization. We measured soil physicochemical properties, basal soil respiration, soil microbial biomass carbon, soil microbial community structure (phospholipid fatty acid assay), bacterial load (16S rRNA gene abundance), enzymatic activities (urease, dehydrogenase, β-glucosidase and acid phosphatase) and abundance of microbial nitrogen cyclers (quantitative PCR). A field experiment was established in an orange orchard (Citrus sinensis) in southeast Spain and eighteen soil samples were taken from each plot to compare the impacts of soil moisture: near (wet, w) or away (dry, d) from drip-irrigation points, in plots with inorganic fertilizers under intensive ploughing (PI) or organic fertilization (OA). The results showed that changes in microbial properties and soil microbial indexes were strongly associated with soil moisture content under both organic and inorganic fertilization, and with organic carbon content. Soil moisture influenced soil aggregation, basal soil respiration, phosphatase activity, bacterial and fungal load (PLFAs) and the abundances of bacterial N cycling genes, including nifH (nitrogen fixation) nirS/K and nosZ genes (denitrification) and amoA-B (bacterial nitrification). The potential for N fixation and denitrification, two microbial processes that are crucial for determining the amount of N in the soil, were improved by increased soil moisture in the proximity of the drip irrigation. Soil OC and total N, which are higher under organic fertilization than under inorganic fertilization, were also shown to be highly correlated with the abundance of the N cycling genes. By controlling irrigation doses and applying organic amendments, it may be possible to increase the microbial abundance and function in soil and support greater fertility of soils.

Non-canonical AUX/IAA protein, OsIAA29, and ZmMPR-1 homologues, OsMRPLs, are part of an auxin-related signalling cascade operating in the dorsal aleurone during early rice grain development. Endosperm of rice and other cereals accumulates high concentrations of the predominant in planta auxin, indole-3-acetic acid (IAA) during early grain development. However, IAA signalling and function during endosperm development are poorly understood. Here, we report that OsYUC12 (an auxin biosynthesis gene) and OsIAA29 (encoding a non-canonical AUX/IAA) are both expressed exclusively in grains, reaching a maximum 5-6 days after pollination. OsYUC12 expression is localised in the aleurone, sub-aleurone and embryo, whereas OsIAA29 expression is restricted to a narrow strip in the dorsal aleurone, directly under the vascular bundle. Although rice has been reported to lack endosperm transfer cells (ETCs), this region of the aleurone is enriched with sugar transporters and is likely to play a key role in apoplastic nutrient transfer, analogous to ETCs in other cereals. OsIAA29 has orthologues only in grass species; expression of which is also specific to early grain development. OsYUC12 and OsIAA29 are temporally co-expressed with two genes (AL1 and OsPR602) previously linked to the development of dorsal aleurone or ETCs. Also up-regulated at the same time is a cluster of MYB-related genes (designated OsMRPLs) homologous to ZmMRP-1, which regulates maize ETC development. Wheat homologues of ZmMRP-1 are similarly expressed in ETCs. Although previous work has suggested that other cereals do not have orthologues of ZmMRP-1, our work suggests OsIAA29 and OsMRPLs and their homologues in other grasses are part of an auxin-regulated, conserved signalling network involved in the differentiation of cells with ETC-like function in developing cereal grains.

Autism Spectrum Disorder (ASD) and depression are often co-occurring in young people. However, despite the association between these two disorders, and the fact that females have higher prevalence for depression than males in the general population, there is little reported evidence regarding the correlates of depression in young autistic females. Several physiological (age, menarche, HPA-axis responses), psychological (social anxiety), and environmental or genetic (mothers' depression) factors were tested for their contribution to depression severity in a sample of 53 autistic girls aged 6yr to 17yr. Depression scores were collected from the girls' self-ratings and also from the ratings their mothers gave them. Regression results indicated that girls' social anxiety, age, and mothers' depression were common significant contributors to both sets of depression scores, but with different effects. Autistic girls' self-reports of their depression were significantly associated with their HPA-axis responses but not with their menarche status. Implications for research and clinical settings are discussed.

Background Interprofessional education initiatives can be used as effective means to influence students' perceptions of their own and others' roles and interactions as health professionals. There is a need to better understand how interprofessional education learning outcomes are appreciated by students. Aim The aims of this study were to describe and compare evaluation feedback from students in undergraduate nursing and medicine programmes about the learning outcomes of an interprofessional education initiative. Methods A mixed methods pre-post-study design was used to collect data using evaluation questions about the initiative and two interprofessional learning outcomes: communication and teamwork. Ratings were analysed with a two-way repeated measures analysis of variance or a t-test. Written responses were analysed using a general inductive approach. Results Data from 30 nursing students and 12 medical students were analysed. A noteworthy finding was a significantly higher average rating for nursing students than medicine students before and after the session for the statement about valuing interprofessional learning. Three themes represented comments from both groups: positive experiences, relevance to practice, and learning design issues. Discussion Findings were interpreted to indicate that students from both programmes valued the learning outcomes session and was greater at the end of the session. Notably, the perceived value of interprofessional learning was higher for nursing students than medical students before and after the session. This finding may be related to the familiarity nursing students may have with the teaching methods used in the initiative. Further exploration of this finding is needed so educators can better understand how they can provide optimal learning experiences for all students who participate in interprofessional education. Conclusion Nursing and medicine students appear to value the interprofessional learning outcomes in an undergraduate health professional initiative. However, differences between these two groups of students are an area for further exploration.

Background Individual Burden of Illness for Depression (IBI-D) represents a method for assessing the degree to which depressive symptomatology reduces functioning. Although the IBI-D has been shown to provide more information than measures of depressive symptomatology alone, previous studies have relied upon participant self-reports, and have not included an objective measure of physiological functioning. Such an index might add to the validity and informational value of IBI-D data. Methods Responses to scales measuring depressive symptoms, functional impairment due to mental and physical health, and saliva samples were collected from 111 community volunteers from rural Australia. Saliva was assayed for cortisol and C-Reactive Protein concentrations, and the ratio of these was used as an indicator of the balance between the Hypothalamic-Pituitary-Adrenal axis and the inflammatory response from the immune system. Results Principal Components Analysis (PCA) produced two- and three-factor solutions from the psychological and biological data, and were used to form weighted models of two IBI-D equations. Stepwise regression analysis indicated that the addition of the biological index to the second IBI-D made a significant extra contribution to variance in depression score. Limitations No longitudinal data were collected, participants were from a single geo-cultural region, and were self-selecting. Clinician interviews might augment participant self-reports. Conclusions Valid assessment of the self-reported symptoms of depression provides one aspect of diagnostic information, but the addition of biological information can further inform clinicians and researchers about the effect that these symptoms have upon individual patient functioning.

Background The Cortisol Awakening Response (CAR) is sometimes dysregulated in young people with Autism Spectrum Disorder (ASD), but previous findings are mostly based upon group mean data and do not report individual responses. In addition, investigation of the correlates of CAR dysregulation has been limited. Methods To provide insight into the individual profiles and correlates of the CAR in young males with ASD, 32 high-functioning male participants with ASD aged between 9 yr and 18 yr completed several measures of anxiety and mood, and provided saliva samples at waking and 30 min later for calculation of the CAR. Results Although group mean data showed an expected CAR profile, over half of the participants had a dysregulated CAR. There was a significant interaction between cortisol concentrations at waking and 30 min later and CAR presence/absence, suggestive of the presence of hyper- and hypo-cortisolism. Unlike previous data regarding CAR and mood states in young females with ASD, there were no significant associations between anxiety or depression and CAR dysregulation in this sample of boys with ASD. Conclusions The use of the CAR in research and clinical settings must be accompanied by an awareness of the likelihood of individual variability.

Children with Autism Spectrum Disorder (ASD) also often suffer from elevated stress and anxiety. These states can be measured via reports of behaviour (from self or others) or from physiological measures of stress, including the symptoms of Generalised Anxiety Disorder (GAD) and salivary cortisol, respectively. The use of these measures assumes a degree of reliability over time so that data from a specific period may be generalised to other periods. To measure the test-retest reliability of salivary cortisol and self-ratings of GAD in 27 high-functioning boys with ASD (M age = 12.1 yr., SD = 2.8 yr), samples were collected a mean of 2.4 yr. (SD = 0.5 yr) apart and analysed to test for the presence of a significant correlation within each variable over time. Results indicated that, although the concentrations of salivary cortisol increased over the period of the study, there was a significant correlation between the two measures of cortisol. GAD scores also showed a significant correlation over the period of the study. These findings suggest that both GAD and salivary cortisol data collected from boys with ASD may be reasonably inferred to represent fairly stable phenomena over time in research and clinical settings.

This study was designed to investigate the association between the short (s) versus non-short (non-s) 5HTTLPR variants, presence of childhood stressors and recent life stressors, and depression, and to do so at two levels that would expose the associations between total depression scores and also individual depression items.Materials and methods: Two hundred and forty-nine volunteers from one of the Australian Electoral Office electorates covering a large rural land area completed a series of questionnaires about childhood and recent life stress and depression, and provided a buccal cell sample for genotyping the 5-HTTLPR polymorphism into s versus non-s carriers.Results: Although there were no significant differences in the depression scores of the s-carriers versus the non-s carriers, each subtype of the 5-HTTLPR polymorphism showed different patterns of association between childhood stress and depression symptoms, and between recent life stress and depression symptoms.Conclusion: Individualization of therapy for depression may be achieved through consideration of the specific associations that patients exhibit between life stress, 5-HTTLPR polymorphism, and depression symptomatology. Traditional intensive agricultural practices negatively impact soil quality, leading to increased erosion in the Mediterranean region. In contrast, sustainable management practices that have utilized organic fertilization over a decade improved soil quality in the Spanish “Pago Casa Gran” vineyard, Valencia. Grape prunings, combined with sheep manure or leguminous cover, have improved soil chemical and biological parameters, such as organic carbon, nitrogen content, respiration, PLFA, total DNA and aggregation, however, there has been a lack of information on their effect on specific functional microbial groups. The nitrogen cycling community of the soil was investigated by quantification of genes involved in key pathways, in particular nitrogen fixation, denitrification and nitrification. The abundances of bacterial nifH, nosZ, nirS and nirK genes significantly increased under a decade of organic fertilization when compared to inorganic fertilization, and was linked to increased soil organic carbon. The abundance of nifH was lower where fertilizers rich in ammonia and nitrate were used and its increase under organic fertilization was more related to the availability of organic carbon than to the nature of the organic amendment. Archaeal amoA abundance did not correlate with the type of fertilization and the bacterial amoA abundance was more dependent on the availability of soluble nitrogen than on the type of management. An increase in nosZ under organic fertilization may suggest a greater abundance of denitrifiers with the ability to reduce nitrous oxide to nitrogen gas, reducing greenhouse gas emission. Our results indicate that soil microbial communities involved in biological nitrogen cycling, in particular nitrogen fixation and denitrification, are more abundant under management practices that include organic fertilization when compared to traditional agricultural practices. It may therefore be recommended that organic fertilization be utilized in agricultural systems to assist with sustaining healthy soils.

Some prostate cancer (PCa) patients become clinically anxious or depressed after diagnosis and treatment. Some also show the physiological signs of chronic stress. However, there are currently no data describing how these particular patients might be identified at intake. This study tested the individual and combined predictive power of a psychological factor and a genetic factor as potential predictors of anxiety, depression, and chronic stress in a sample of PCa patients. Ninety-five PCa patients completed psychological inventories for anxiety, depression, and psychological resilience (PR) and also gave a saliva sample for cortisol and a mouthwash sample for genetic testing for the presence of the BDNF Val66Met polymorphism. High PR patients had significantly lower anxiety and depression than low PR patients, but showed no significant differences in their salivary cortisol. Carriers of the Met allele of the BDNF Val66Met polymorphism had significantly higher salivary cortisol concentrations than patients who did not carry this allele. Each of these two factors may provide valuable information regarding the vulnerability of PCa patients to anxiety, depression, or chronic stress. Suggestions are made for their inclusion in clinical settings.

SUMMARY A series of studies were undertaken to optimize the propagation of hemorrhagic enteritis virus (HEV) in specific-pathogen-free (SPF) chickens. A total of 562 SPF chickens were orally inoculated with an Australian avirulent HEV isolate of turkey origin at 9, 14, 21, or 28 days of age with 5, 6, 7, or 8 log 10 genomic copies (GC), while 102 chickens served as uninfected controls. No clinical signs were observed in infected chickens. There was an inoculum-dose–dependent increase in the relative spleen and liver weight (P < 0.01). Relative spleen weight 7 days post-HEV inoculation was up to 85% higher in chickens that were inoculated with 6 to 7 GC compared with controls, with no further increase at higher doses. Relative liver weight increased up to 14% in chickens inoculated with 6 GC, with no further increase. Birds inoculated with a 7 GC dose had a higher frequency of HEV DNA-positive birds (77% to 86%) than birds inoculated with lower doses (33% to 59%; P < 0.01). The most efficient dose for live...

Productivity in agricultural ecosystems is often largely dependent on the input of nitrogen fertilizers such as urea, nitrate, and ammonia, despite the high financial costs and potential detrimental effects on the environment. Soil enrichment with organic matter, such as plant compost, straw, or manure, can enhance soil organic carbon and improve soil quality and increase aggregate stability, biological activity, and microbial diversity, including nitrogen cycles. The abundance of the nifH gene, thus nitrogen cycles, was found to be enhanced when organic fertilization was used and correlated mainly to the availability of organic carbon, potentially due to the presence of fuels required to run the energetically expensive nitrogen fixation process. On the other hand, readily available ammonia and nitrate, which are often associated with inorganic fertilization, tend to suppress the soil potential for nitrogen fixation. The impacts of fertilizers on denitrifying microbial communities are complex, due to the great diversity of the denitrifiers and the variation in their abundance in different environments. However, in general, the use of organic fertilizers increases denitrification potential and activity in soils when compared to inorganic fertilization. This is particularly important for increasing the abundance of those denitrifiers containing the nosZ genes and capable of the final step of the denitrification process, removing the potent greenhouse gas nitrous oxide by its conversion to dinitrogen. Bacterial and archaeal nitrifiers react differently to variations in soil conditions and to different fertilization management strategies. It is important to note that nitrogen fertilizers in access might turn into an ecosystem hazard where nitrifying microbes convert them to nitrate. This survey of the literature suggests that the addition of organic matter to agricultural soils, even where inorganic nitrogen fertilizers are used, enhances the soil potential for nitrogen cycling and soil sustainability.

Although previous data indicate that dyadic coping is associated with Hypothalamic-Pituitary-Adrenal (HPA-axis and C-reactive protein (CRP) separately, no study has reported on the ratio between these two systems and dyadic coping, despite this index of physiological homeostasis being associated with physical health and depression. Forty-eight community volunteers who were either married (n = 36) or cohabiting (n = 12) provided saliva and serum samples, and also completed a dyadic coping inventory. There was a significant inverse correlation between cortisol:CRP ratio and dyadic coping, but only for married participants. One of the six dyadic coping items, related to being able to calmly discuss something within a dyad, was the key factor in the association between dyadic coping and cortisol:CRP ratio. These findings provide some initial support for the influence of the way that spouses interact to solve problems and the balance between their HPA and immune systems.

Background Autistic girls’ change in salivary cortisol concentrations from morning to afternoon were compared with their parents’ own anxiety states as potential correlates of the autistic girls’ self-ratings of their anxiety and those given by their parents about their daughters’ anxiety. Methods Thirty-three autistic girls (6 yr to 10 yr, M = 8.21 yr, SD = 1.29 yr) and 19 autistic adolescent females (11 yr to 15 yr, M = 12.74 yr, SD = 1.52 yr) completed anxiety inventories about themselves and provided saliva samples in the morning and afternoon. Their mothers also rated their daughters’ anxiety and their own anxiety. Results There were no significant differences between mothers’ and daughters’ ratings of the latter’s GAD. The autistic girls’ self-ratings of their anxiety were more strongly influenced by their mothers’ anxiety in the younger girls, but more responsive to their own chronic stress state (i.e., cortisol) in the adolescent girls. Conclusions Mothers’ evaluations of their autistic daughters’ anxiety appear to be strongly influenced by their own anxiety. There was also some indication that autistic girls may disguise their anxiety.

Small molecular weight oxygen free radical species (ROS) involved in oxidative stress can cause damage to cellular macromolecules including proteins, DNA and lipids. One of the most important enzymes involved in ROS detoxification is glutathione peroxidase (GPx). Here we study the age-related expression of GPx isoenzymes in various parts of the rat kidney with and without exposure to external oxidative stress. These results are correlated to the age dependent changes in the expression of the chaperone, Hsp-70. Protein and mRNA expression of GPx1 and GPx4 was studied in different regions of the kidney in ageing rats in the presence and absence of the external stressor 0.2 mM H2O2. Protein levels were examined by Western blot analysis following detection with appropriate antibodies and mRNA levels were analysed by quantitative reverse transcription polymerase chain reaction (qRT-PCR) using appropriate primer sequences. mRNA expression for the chaperone Hsp70 was investigated in parallel. After reaching a peak at maturity (12 weeks), GPx1 protein and mRNA levels decreased with age under both control and stress conditions, and were higher in the cortex than in the outer and inner medulla. GPx4 protein and mRNA levels showed few comparable age-related changes. By contrast with the observed age-related decrease in GPx1 expression, chaperone Hsp-70 mRNA expression greatly increased with age. These findings suggest that the age-related decline in GPx1 expression in the cortex may be partly offset by a reciprocal change in Hsp-70 expression. These results are consistent with the oxidative stress theory of ageing.

Co-composting for the remediation treatment of aged polycyclic aromatic hydrocarbons (PAHs) in field contaminated soil was examined in this study. Two PAH contaminated soils collected from a manufacturing gas plant site with high PAH concentrations (184.00 ± 3.99 mg kg−1 and 490.0 ± 43.53 mg kg−1) were co-composted with cattle manure and wheat straw for up to 56 days in laboratory microcosms. A range of optimised co-composting treatments including addition of Triton-X100 surfactant and a biological surfactant from the Australian Red Ash tree (Alphitonia excelsa) were tested. There was no significant decrease in the PAH concentrations in any of the different co-composting treatments for either of the soils, despite an 87% decrease in a phenanthrene spike (100 mg kg−1), and evidence of PAH microbial catabolic activity. Non-exhaustive extraction using hydroxypropl-β-cyclodextrin (HP-β-CD) and linear regression modelling confirmed that PAH bioaccessibility constrained PAH co-composting remediation in these soils. The results demonstrate that co-composting alone is not an effective remediation strategy for certain aged PAH contaminated soils. For these soils some form of pre-composting chemical or engineering treatment is required to overcome bioaccessibility limitations in successful remediation.

Although EEG connectivity data are often used to build models of the association between overt behavioural signs of Autism Spectrum Disorder (ASD) and underlying brain connectivity indices, use of a large number of possible connectivity methods across studies has produced a fairly inconsistent set of results regarding this association. To explore the level of agreement between results from five commonly-used EEG connectivity models (i.e., Coherence, Weighted Phased Lag Index- Debiased, Phase Locking Value, Phase Slope Index, Granger Causality), a sample of 41 young males with ASD provided EEG data under eyes-opened and eyes-closed conditions. There were relatively few statistically significant and/or meaningful correlations between the results obtained from the five connectivity methods, arguing for a re-estimation of the methodology used in such studies so that specific connectivity methods may be matched to particular research questions regarding the links between neural connectivity and overt behaviour within this population.

Haemorrhagic enteritis virus (HEV) causes clinical haemorrhagic enteritis in young poults and/or subclinical immunosuppression which is often associated with colibacillosis. This disease is controlled with live vaccines worldwide, however, importation of HEV vaccines or cells that support HEV propagation are not permitted in Australia. A major experiment in isolators was conducted to test the safety and efficacy of a putative HEV vaccine. The study had a factorial design with four factors namely vaccination age (28 and 42 days of age), vaccine dose (0, 105 , 106 , 107 genomic copies of HEV vaccine), challenge with HEV (yes, no) and vaccination-challenge interval (7, 21 or 42 days). A total of 315 poults were used providing 6-8 birds per treatment combination. Turkey growth rate, mortality, pathological findings, anti-HEV antibodies and viral load were examined. Vaccination lead to significant increases in anti HEV antibody over the following 2-4 weeks. Overall, vaccination with 106 and 107 was protective against increase in relative splenic weight and splenic viral load in challenged birds. Clinical haemorrhagic enteritis was not induced by any treatment but there was an increased incidence of airsacculitis in groups receiving either HEV vaccine or challenge virus compared to the negative control birds (25.8-29.3% vs. 9.4%, P

Children with Autism Spectrum Disorder (ASD) show dysregulation of the expected Hypothalamus-Pituitary-Adrenal (HPA) axis and elevated cortisol responses to stress and response patterns, but little has been reported regarding their recovery from stress in terms of cortisol concentrations. This response was investigated in a sample of 32 young males with ASD aged between 9 and 18 years (M = 14.3 yr, SD = 2.7 yr), using a standardised experimental protocol combined with individualised stressor and non-stressor tasks. Results indicated that about half of the sample demonstrated unexpected HPA axis response patterns, and that recovery from stress cortisol concentrations were significantly associated with a single symptom of Social Phobia and Morning cortisol. These findings suggest that one of the key diagnostic criteria for ASD may be strongly influential in the HPA axis responses of boys with ASD and that training regimesto assist them to form less fearful associations with their non-ASD peers may be central to the academic and social progress of these boys.

Autism spectrum disorder (ASD) is a neurodevelopmental condition affecting about 1 in 100 children and is currently incurable. ASD represents a challenge to traditional methods of assessment and diagnosis, and it has been suggested that direct measures of brain activity and connectivity between brain regions during demanding tasks represents a potential pathway to building more accurate models of underlying brain function and ASD. One of the key behavioural diagnostic indicators of ASD consists of sensory features (SF), often characterised by over- or under-reactivity to environmental stimuli. SF are associated with behavioural difficulties that impede social and education success in these children as well as anxiety and depression. This review examines the previous literature on the measurement of EEG connectivity and SF observed in individuals with ASD.

The comparative strength of the 5-HTTLPR polymorphism as a 'predictor' of depression after major stress, versus the 'protective' effect of psychological resilience (PR) against depression after major stress, was tested in a homogeneous sample of older men who had all received a diagnosis and treatment for prostate cancer. Results supported the association between PR and lower depression after stress, but did not support the association between the 5-HTTLPR and elevated depression after stress. Examination of PR at scale, factor, and item level identified the specific PR-related behaviour that was the most powerful predictor of low depression. These data suggest that the carriage of the short form of the 5-HTTLPR may negate the protective effect of PR against depression in these men, or that PR may nullify the depression vulnerability of this form of the 5-HTTLPR. These findings may explain some of the 'null' findings regarding the link between the 5-HTTLPR and depression in the wider literature by arguing for an interaction between these two factors in the association between major stress and depression.

ObjectiveTo investigate the effect of chronic stress as measured in cortisol concentrations upon the association between psychological resilience (PR) and depression in prostate cancer (PCa) patients.MethodsA total of 104 men with PCa completed inventories on PR, depression, and background factors, plus gave a sample of their saliva for cortisol assay.ResultsThe inverse correlation between PR and depression was present only for PCa patients with low or moderate concentrations of salivary cortisol (when classified as more than 1.0 SD below the mean vs within 1.0 SD of the group mean) but not for those men whose cortisol was >1.0 SD from the group mean. Specific PR factors and behaviours that made the greatest contribution to depression were identified for the low and moderate cortisol groups.ConclusionsThese results suggest that there are particular aspects of PR that are most strongly related to depression, but that PR's inverse association with depression may be absent in participants with extreme chronic physiological stress.

This study aimed to investigate the feasibility of propagating and titrating hemorrhagic enteritis virus (HEV) in chicken embryos. A total of 308 embryonated eggs were used. At 10 days of embryonic age, eggs were inoculated via allantoic sac or chorioallantoic membrane routes with non-heat-treated (live) HEV or heat-treated (dead) HEV or served as negative controls. Allantoic fluid retrieved at 0, 1, 3, 5, and 7 days postinoculation (dpi) was tested for HEV by quantitative PCR. Inoculation with HEV did not cause visible growth impairment or lesions in the chicken embryos. Overall, there was no difference in postinoculation mortality rates among groups sham-inoculated (6/30, 20.0%) or inoculated with live (34/252, 13.4%) or dead (3/ 26, 6.9%) HEV (El virus de la enteritis hemorrágica de los pavos puede ser titulado pero no propagado en embriones de pollo. Este estudio tuvo como objetivo investigar la viabilidad de propagar y titular al virus de la enteritis hemorrágica de los pavos (con las siglas en inglés HEV) en embriones de pollo. Se utilizaron un total de 308 huevos embrionados. A los 10 días de edad embrionaria, los huevos se inocularon por vía saco alantoideo o por la membrana corioalantoidea con el virus de la enteritis hemorrágica sin tratamiento térmico (vivo) o con un virus de la enteritis hemorrágica con tratamiento térmico (muerto) y algunos sirvieron como controles negativos. El fluido alantoideo recuperado a los cero, uno, tres, cinco y siete días después de la inoculación se analizó para detectar el VHE mediante un método cuantitativo de PCR. La inoculación con el virus de la enteritis hemorrágica no causó daños visibles en el crecimiento ni lesiones en los embriones de pollo. Molecular methods have been used in the study of Azospirillum and other related PGPRs to carry out gene functional analysis, create gene knockouts, generate genetically engineered strains, and carry out gene expression studies. Genetic transformation has routinely been carried out using conjugation, while chromosomal modifications have been performed using unstable, suicide plasmids, or more stable, broad host-range vectors. Gene expression studies are often carried out using promoter-bound reporter genes; however, quantitative methods such as reverse transcribed polymerase chain reaction can now be used to directly study gene expression. In this chapter we describe the common types of vectors used in Azospirillum, as well as methods for transformation and mutagenesis. We also describe the use of promoter-bound reporter genes and the applications of quantitative RT-PCR for Azospirillum gene expression studies. Methods for the isolation of DNA and RNA from Azospirillum for use in molecular and gene expression studies are also described.

To measure the prevalence and severity of Generalised Anxiety Disorder (GAD), hypo- and hypercortisolaemia, and their association in a sample of prostate cancer (PCa) patients, 97 Australian PCa patients completed a background questionnaire and the GAD-7, and provided a sample of saliva collected 30-45 min after waking. The mean GAD7 score was 9.67 (SD = 3.09), and prevalence rates for current anxiety were higher than those reported for non-PCa males of a similar age. Mean salivary cortisol concentrations (30.78 nmol/L, SD = 13.97 nmol/L) were also higher than for age-comparative non-PCa men. There was a significant inverse correlation between GAD and cortisol (r = -. 209, p < .05), and four subgroups of GAD-cortisol patients were able to be identified, with evidence of both hyper- and hypocortisolaemia. These findings provide initial neurobiological evidence of the chronic and profound nature of stress experienced by PCa patients, and also suggest a possible measure that might be used to identify most at-risk PCa patients.

Spermatogonial stem cells hold enormous potential in mammalian reproductive medicine through the preservation of gametes, the restoration of fertility, enhancement of germ-lineage genetic manipulation and the improvement in our understanding of stem cell biology. Here we describe the protein profiles of the membrane compartment of bovine testicular cell isolates which were enriched for germ cells using differential plating. The isolated cells were characterised with antibodies to UCHL1 (previously known as PGP9.5) for type A spermatogonia; DDX4 (previously known as VASA) for germ cells and vimentin for Sertoli cells. Ultracentrifugation techniques were used to specifically isolate cell membranes, with membrane protein identifications significantly increased when compared to whole cell lysates. We utilised the filter-aided sample preparation protocol for improved digestion efficiency of membrane proteins. Using ESI-LC-MS/MS, we compared the proteins present in two cell populations. A total of 1,387 proteins were identified in bovine testis cell isolates, of which 39% were membrane-associated. A total of 64 proteins were differentially expressed in the non-adhered fraction (enriched for undifferentiated germ cells), of which 16 were unique to this cell population and the remaining 48 showed a two-fold change as judged by spectral counting. This analysis revealed a number of candidate germ cell markers including the known markers, DDX4 and UCHL1. The proteomic profiles generated in this study support and complement transcriptomics studies and reinforce the potential of proteomics in identifying and characterising the protein effectors of self-renewal and/or differentiation in stem cells. Adhesion molecules are expressed by both adult and embryonic stem cells, with different classes of adhesion molecules involved in cell-membrane and intercellular contacts. In this study the expression of the adhesion molecule claudin-8 (CLDN8), a tight-junction protein, was investigated as a potential marker for undifferentiated spermatogonia in the bovine testis. We found that CLDN8 was expressed by both spermatogonia and a subset of Sertoli cells in the bovine testis. We also showed co-expression of GFRα1 in testis cells with CLDN8 and with Dolichos biflorus agglutinin–fluorescein isothiocyanate (DBA–FITC) staining. We observed co-enrichment of spermatogonia and CLDN8-expressing Sertoli cells in DBA–FITC-assisted magnetic-activated cell sorting (MACS), an observation supported by results from fluorescence-activated cell sorting analysis, which showed CLDN8-expressing cells were over-represented in the MACS-positive cell fraction, leading to the hypothesis that CLDN8 may play a role in the spermatogonial stem-cell niche.

Nonviral reprogramming of bovine embryonic and adult fibroblasts was undertaken using electroporation of a polycistronic plasmid carrying human reprogramming factors Oct4, Sox2, Lin28, and Nanog (Addgene Plasmid #20922). Because of difficulties encountered in reliably and reproducibly reprogramming bovine cells to a pluripotent state, several small-molecule combinations have been tested to support reprogramming. Previously, a combination of three molecules, (sodium butyrate, PD0325901 and SB431542; NaB-PD-SB) in induced pluripotent stem (iPS) medium [Minimum Essential Medium Alpha, 20% FBS, 1x insulin-transferrin-selenium (ITS), 2 mM Glutamax (Gibco, Grand Island, NY, USA), 100 μM nonessential amino acids (NEAA), 50 U mol–1 penicillin, 50 mg mL–1 streptomycin, 0.1 mM β-mercaptoethanol, 4 ng mL–1 human leukaemia inhibitory factor, and 10 ng mL–1 basic fibroblast growth factor] was found to accelerate the reprogramming process of bovine iPS cells, with colonies observed at 12 days post-electroporation, as opposed to 21 days without the addition of the small-molecule cocktail. The addition of 1 mM valproic acid (VPA), a histone deacetylase (HDAC) inhibitor, in combination with the other small-molecule cocktail (NaB-PD-SB), was found to improve the reprogramming efficiency of bovine adult and embryonic fibroblasts, with colonies observed at Day 10 post-electroporation, and the average number of colonies present per 10-cm dish, rising from an average of 5 colonies observed in the NaB-PD-SB cocktail to 27 with the addition of VPA. Colonies grown in the presence of VPA had a consistent morphology, forming compact domed colonies consisting of small round cells with well-defined borders. It was observed that colonies growing in the presence of VPA tended to have a better defined border and grew larger in size than those grown in the presence of NaB-PD-SB alone. Colonies began to differentiate after 21 days and were no longer alkaline phosphatase positive after this time. Cells were either harvested for mRNA extraction or differentiated into embryoid bodies (EB). Expression of pluripotency genes, Oct4, Sox2, Klf4, Nanog, and Alkaline Phosphatase were significantly increased in the presence of VPA compared to nonreprogrammed somatic bovine fibroblasts, with expression profiles similar to those grown in the NaB-PD-SB cocktail. Embryoid bodies were analysed for gene expression of different germ layer markers, FOXA2 (endoderm), Nestin and TUBB3 (ectoderm), and Desmin (mesoderm) using quantitative RT–PCR. For both EBs derived from the NaB-PD-SB cocktail and those with VPA, at least one marker from each germ layer was present, demonstrating the potential potency of these cells. At least a 10-fold increase in expression of these was observed in comparison to somatic fibroblast cells. It is apparent that the addition of small molecules can assist in the reprogramming of bovine iPS cells, and addition of VPA to the cocktail results in more consistent putative bovine iPS colonies. Further work is needed to identify the causes for early differentiation of colonies in order to obtain fully reprogrammed pluripotent stem cells.

The radioimmunoassay described measures TSH in dried whole blood spots collected from neonates onto filter paper Guthrie cards. Microgranular cellulose is added to the precipitating reagent at the critical separation stage of the assay to overcome imprecision caused by the presence of the filter paper sample disc in the tube. The method was developed for a regional neonatal screening unit and has been found to be very reliable during ten months' routine use. It was required to be as precise, sensitive, accurate, rapid, simple, and inexpensive as possible and suitable for use with automatic diluting equipment in order to process large numbers of samples. Other methods were examined for their suitability and found not to fulfil one or more of the above criteria.
