This study evaluated enamel mineral content and surface microhardness before and after bleaching treatment using 10% carbamide peroxide (CP) containing calcium (Ca) or amorphous calcium phosphate (ACP). Thirtysix bovine slabs were randomly allocated into 3 groups (n = 12) according to bleaching treatment: G1 Opalescence PF 10% (CP), G2 NiteWhite ACP (CP+ACP), and G3 Opalescence PF (10%) with calcium (CP+CA). The bleaching agent was applied on enamel surface for 6 h/day over a period of 21 days. Enamel surface was evaluated by Knoop microhardness (KNH) and micro energydispersive Xray fluorescence spectrometry (μEDXRF) at baseline and at after bleaching treatment. Data were statistically analyzed by repeated measures ANOVA and Tukey's test (α = 0.05). There was a significant decrease in microhardness after bleaching treatments for all study groups, but no difference between bleaching gels. There was no difference in the Ca/P ratio measured by μEDXRF for all groups at the study times, but the mean value was lower in group CP+CA than in group CP+ACP. Group CP was similar to both CP+ACP and CP+CA. It can be concluded that enamel microhardness decreased after the bleaching process, regardless of the presence of calcium or ACP, but there was no significant change in the Ca/P ratio of enamel after bleaching for each tested gel. This indicates that the bleaching gels have erosive potential, causing softening of enamel without promoting surface loss, regardless of the presence of calcium of ACP ions.

Vaccine control and prevention of porcine reproductive and respiratory syndrome (PRRS), the most important disease of swine, is difficult to achieve. However, the discovery of broadly neutralizing antibody activity against porcine reproductive and respiratory syndrome virus (PRRSV) under typical field conditions opens the door to new immunologic approaches for robust protection. We show here that passive administration of purified immunoglobulins with neutralizing antibodies reduced PRRSV2 infection by up to 96%, and PRRSV1 infection by up to 87%, whereas immune immunoglobulins lacking neutralizing activity had no effect on viral infection. Hence, immune competence of passive immunoglobulin transfer was associated specifically with antibody neutralizing activity. Current models of PRRSV infection implicate a minor envelope glycoprotein (GP) complex including GP2, GP3, and GP4, as critical to permissive cell infection. However, conserved peptides comprising the putative cell attachment structure did not attenuate neutralization or viral infection. The results show that immunological approaches aimed at induction of broadly neutralizing antibodies may substantially enhance immune protection against PRRSV. The findings further show that naturally occurring viral isolates are able to induce protective humoral immunity against unrelated PRRSV challenge, thus removing a major conceptual barrier to vaccine development., (Copyright © 2018 Elsevier B.V. The improvement of agronomic practices and the use of high technology in field crops contributes for significant increases in maize productivity, and may have altered the dynamics of nutrient uptake and partition by the plant. Official recommendations for fertilizer applications to the maize crop in Brazil and in many countries are based on critical soil nutrient contents and are relatively outdated. Since the factors that interact in an agricultural production system are dynamic, mathematical modeling of the growth process turns out to be an appropriate tool for these studies. Agricultural modeling can expand our knowledge about the interactions prevailing in the soil-plant-atmosphere system. The objective of this study is to propose a methodology for characterizing the micronutrient composition of different organs and their extraction, and export during maize crop development, based on modeling nutrient uptake, crop potential evapotranspiration and micronutrient partitioning in the plant, considering the production environment. This preliminary characterization study (experimental growth analysis) considers the temporal variation of the micronutrient uptake rate in the aboveground organs, which defines crop needs and the critical nutrient content of the soil solution. The methodology allowed verifying that, initially, the highest fraction of dry matter, among aboveground organs, was assigned to the leaves. After the R 1 growth stage, the largest part of dry matter was partitioned to the stalk, which in this growth stage is the main storage organ of the maize plant. During the reproductive phase, the highest fraction of dry matter was conferred to the reproductive organs, due to the high demand for carbohydrates for grain filling. The micronutrient (B, Cu, Fe, Mn, and Zn) content follows a power model, with higher values for the initial growth stages of development and leveling off to minimum values at the R 6 growth stage. The proposed model allows to verify that fertilizer recommendations should be related to the temporal variability of micronutrient absorption rates, in contrast to the classic recommendation based on the critical soil micronutrient content. The maximum micronutrient absorption rates occur between the reproductive R 4 and R 5 growth stages. These evaluations allowed to predict the maximum micronutrient requirements, considered equal to respective stalk sap concentrations.

Decision support for nutrient application remains an enigma if based on soil nutrient analysis. If the crop could be used as an auxiliary indicator, the plant nutrient status during different growth stages could complement the soil test, improving the fertilizer recommendation. Nutrient absorption and partitioning in the plant are here studied and described with mathematical models. The objective of this study considers the temporal variation of the nutrient uptake rate, which should define crop needs as compared to the critical content in soil solution. A uniform maize crop was grown to observe dry matter accumulation and nutrient content in the plant. The dry matter accumulation followed a sigmoidal model and the macronutrient content a power model. The maximum nutrient absorption occurred at the R4 growth stage, for which the sap concentration was successfully calculated. It is hoped that this new approach of evaluating nutrient sap concentration will help to develop more rational ways to estimate crop fertilizer needs. This new approach has great potential for on-the-go crop sensor-based nutrient application methods and its sensitivity to soil tillage and management systems need to be examined in following studies. If mathematical model reflects management impact adequately, resources for experiments can be saved.

Background: The manufacturing process of islets includes a culture step which was originally introduced to ease the logistics of procedures in preparing the graft and transplant recipient. It has been suggested that culture at room temperature has an advantage over culture at 37 °C, in part by reducing immunogenicity via preferential elimination of contaminating cells (such as passenger leukocytes) within islets. We investigated this using islets isolated from pancreata of adult pigs., Methods: Porcine islets were isolated from three donors and cultured at 37 °C for 1 day, and then under three different conditions: 37 °C for 6 days (condition A); 22 °C for 6 days (condition B); or 22 °C for 5 days followed by 37 °C for 1 day (condition C). Recovery was assessed by DNA measurement, viability by oxygen consumption rate normalized for DNA (OCR/DNA), and gene expression by RT-PCR for a series of 9 lymphocyte markers, 11 lymphokines and chemokines, and 14 apoptotic and stress markers., Results: Post-culture islet recoveries were similar for the three culture conditions. Average OCR/DNA values were 129-159 nmol/min·mgDNA before culture, and 259-291, 204-212, and 207-228 nmol/min·mgDNA, respectively, for culture under conditions A, B, and C, respectively. Irrespective of culture condition, examined gene expression in all three series of lymphocyte markers, lymphokines and chemokines, and apoptotic and stress markers manifested a statistically significant decrease upon culture for 7 days. This decrease was most dramatic for condition A: in particular, most of lymphocyte markers showed a >10-fold reduction and also six markers in the lymphokine and chemokine series; these reductions are consistent with the elimination of immune cells present within islets during culture. The reduction was less for apoptotic and stress markers. For culture under condition B, the reduction in gene expression was less, and culture under condition C resulted in gene expression levels similar to those under condition A: this indicates that 24 h at 37 °C is sufficient to re-equilibrate gene expression levels from those in islets cultured at 22 °C to those in islets cultured at 37 °C. Results were consistent among the preparations from the three donors., Conclusions: Culture of porcine islets at 37 °C provides benefits over culture at 22 °C with respect to OCR/DNA outcomes and reduced expression of genes encoding lymphocyte markers, lymphokines and chemokines, and markers for apoptosis and stress., (© 2013 John Wiley & Sons A/S.)

Background: Reports indicate that human and canine patients with atopic dermatitis (AD) have reduced production of several skin antimicrobial peptides, but more recent data have called those results into question., Hypothesis/objectives: To compare the mRNA expression of seven antimicrobial peptide genes in lesional and adjacent nonlesional skin biopsy specimens from dogs with AD with those from normal dogs and from dogs experiencing other inflammatory skin conditions., Animals: Normal dogs and patients with AD or other inflammatory skin conditions were enrolled with owner permission and approval of the Institutional Animal Care and Use Committee., Methods: Transcripts were measured by quantitative RT-PCR using a standard curve assessment., Results: Normal transcript levels for all seven antimicrobial peptides varied depending on the body site assessed. Transcripts for secretory leukocyte proteinase inhibitor (SLPI) and skin-derived antileucoproteinase (SKALP; also known as elafin) were typically ~10-fold greater in number than transcripts for the canine β-defensins (CBD)-1, -102, -103, -122 and -124. Transcripts for SKALP, SLPI, CBD-1, CBD-103 and CBD-122 were lower in both lesional and adjacent nonlesional skin from dogs with AD in comparison to normal skin. Transcripts were reduced to a similar extent versus normal dogs in skin of dogs with inflammatory skin conditions from both lesional and nonlesional biopsies, except for CBD-122, which was reduced only in lesional skin. Compared with normal dog skin, transcripts for CBD-102 and CBD-124 were unaffected in dogs with AD., Conclusions and Clinical Importance: Both SKALP and SLPI may be important contributors to skin innate immunity, but their decreased expression in AD patients does not account for increased skin infections compared with other skin conditions., (© 2013 The Authors. Objective: To assess the seasonal variation of Aedes aegypti and Aedes albopictus and to estimate the association between the species abundance and abiotic factors., Methods: Tire-traps were kept for a period of 15 days monthly in an urban area of the city of Potim in the state of São Paulo, Southeastern Brazil, from November 2002 to October 2003. Samples of immature forms were followed up under laboratory conditions for 29 days and species were identified. The association between abundance and temperature, rainfall and air relative humidity was tested using Spearman correlations (r s). The descriptive statistics were presented by the average and the standard error (alpha=0.05)., Results: From a total of 20,727 immatures collected, 95.3% were Ae. aegypti and 4.7%, Ae. albopictus. Ae. aegypti was found throughout the whole year while Ae. albopictus was found from November to July. The association between these species and abiotic factors was significant for Ae. aegypti (p=0.04) as for maximum temperature and for Ae. albopictus as for maximum temperature (p=0.01) and rainfall (p=0.02)., Conclusions: Both species presented seasonal variation. However, Ae. aegypti was found throughout the whole year showing a decrease between April and May and June and July. Ae. albopictus, less abundant, was found only between November, 2002 and July, 2003 with a peak in April, 2003. The higher abundance of Ae. aegypti compared to Ae. albopictus in an urban area showed Ae. aegypti has greater capacity of colonizing tires. Such a breeding site in the environment may be important to the maintenance and abundance of Ae. aegypti.
