1. Uncovering large inconsistencies between machine learning derived gridded settlement datasets

Abstract

High-resolution human settlement maps provide detailed delineations of where people live and are vital for scientific and practical purposes, such as rapid disaster response, allocation of humanitarian resources, and international development. The increased availability of high-resolution satellite imagery, combined with powerful techniques from machine learning and artificial intelligence, has spurred the creation of a wealth of settlement datasets. However, the precise agreement and alignment between these datasets is not known. Here we quantify the overlap of high-resolution settlement map for 42 African countries developed by Google (Open Buildings), Meta (High Resolution Population Maps) and GRID3 (Geo-Referenced Infrastructure and Demographic Data for Development). Across all studied countries we find large disagreement between datasets on how much area is considered settled. We demonstrate that there are considerable geographic and socio-economic factors at play and build a machine learning model to predict for which areas datasets disagree. It it vital to understand the shortcomings of AI derived high-resolution settlement layers as international organizations, governments, and NGOs are already experimenting with incorporating these into programmatic work. 4. Elucidating electrochemical nitrate and nitrite reduction over atomically-dispersed transition metal sites.

Abstract

Electrocatalytic reduction of waste nitrates (NO3-) enables the synthesis of ammonia (NH3) in a carbon neutral and decentralized manner. Atomically dispersed metal-nitrogen-carbon (M-N-C) catalysts demonstrate a high catalytic activity and uniquely favor mono-nitrogen products. However, the reaction fundamentals remain largely underexplored. Herein, we report a set of 14; 3d-, 4d-, 5d- and f-block M-N-C catalysts. The selectivity and activity of NO3- reduction to NH3 in neutral media, with a specific focus on deciphering the role of the NO2- intermediate in the reaction cascade, reveals strong correlations (R=0.9) between the NO2- reduction activity and NO3- reduction selectivity for NH3. Moreover, theoretical computations reveal the associative/dissociative adsorption pathways for NO2- evolution, over the normal M-N4 sites and their oxo-form (O-M-N4) for oxyphilic metals. This work provides a platform for designing multi-element NO3RR cascades with single-atom sites or their hybridization with extended catalytic surfaces.

Abstract

The organization and evolution of science has recently become itself an object of scientific quantitative investigation, thanks to the wealth of information that can be extracted from scientific documents, such as citations between papers and co-authorship between researchers. However, only few studies have focused on the concepts that characterize full documents and that can be extracted and analyzed, revealing the deeper organization of scientific knowledge. Unfortunately, several concepts can be so common across documents that they hinder the emergence of the underlying topical structure of the document corpus, because they give rise to a large amount of spurious and trivial relations among documents. To identify and remove common concepts, we introduce a method to gauge their relevance according to an objective information-theoretic measure related to the statistics of their occurrence across the document corpus. After progressively removing concepts that, according to this metric, can be considered as generic, we find that the topic organization displays a correspondingly more refined structure., Comment: Main + SI. Abstract

Sepsis is a complex disorder caused by a dysregulated host response to infection, with high levels of morbidity and mortality. Treatment aimed to modulate immune response and maintain vascular function is still one of the major clinical challenges. This study was designed to test the effect of the small molecule 1-Piperidine Propionic Acid (1-PPA) as molecular targeted agent to block protease-activated receptor 2 (PAR2), one of the major modulators of inflammatory response in LPS-induced experimental endotoxemia. In the THP-1 cell line, LPS-induced cytokine expression was inhibited by 1-PPA in a dose-dependent manner. In LPS-injected mice, treatment with 1-PPA was effective in reducing mortality and sepsis-related symptoms and improved cardiac function parameters. After 6 h from LPS injection, a significant decrease in IL-6, IL-1β, and IL-10 was observed in the lung tissue of 1-PPA-treated mice, compared to controls. In these mice, a significant decrease in vasoactive molecules, especially kininogen-1, was also observed, mainly in the liver. Histopathological analysis confirmed typical features of sepsis in different organs and these findings were markedly reduced in mice treated with 1-PPA. Abstract

We provide an up-to-date view on the knowledge management system ScienceWISE (SW) and address issues related to the automatic assignment of articles to research topics. So far, SW has been proven to be an effective platform for managing large volumes of technical articles by means of ontological concept-based browsing. However, as the publication of research articles accelerates, the expressivity and the richness of the SW ontology turns into a double-edged sword: a more fine-grained characterization of articles is possible, but at the cost of introducing more spurious relations among them. In this context, the challenge of continuously recommending relevant articles to users lies in tackling a network partitioning problem, where nodes represent articles and co-occurring concepts create edges between them. In this paper, we discuss the three research directions we have taken for solving this issue: i) the identification of generic concepts to reinforce inter-article similarities; ii) the adoption of a bipartite network representation to improve scalability; iii) the design of a clustering algorithm to identify concepts for cross-disciplinary articles and obtain fine-grained topics for all articles., Comment: 6 pages; 5 figures

Simple Summary: This review highlights SerpinB3's multifaceted roles in liver disease, from fibrosis, carcinogenesis and immune modulation to cell death protection. In different types of cancer its overexpression correlates with tumor aggressiveness; however, in acute oxidative stress conditions, SerpinB3 promotes cell survival. Novel therapeutic strategies targeting SerpinB3 through its upstream regulators are under development, while its therapeutic potential in acute medical conditions has also been proposed. SerpinB3, a member of the serine-protease inhibitor family, has emerged as a crucial player in various physiological and pathological processes. Initially identified as an oncogenic factor in squamous cell carcinomas, SerpinB3's intricate involvement extends from fibrosis progression and cancer to cell protection in acute oxidative stress conditions. This review explores the multifaceted roles of SerpinB3, focusing on its implications in fibrosis, metabolic syndrome, carcinogenesis and immune system impairment. Furthermore, its involvement in tissue protection from oxidative stress and wound healing underscores its potential as diagnostic and therapeutic tool. Recent studies have described the therapeutic potential of targeting SerpinB3 through its upstream regulators, offering novel strategies for cancer treatment development. Abstract

Transition‐metal nitrogen‐doped carbons (TM‐N‐C) are emerging as a highly promising catalyst class for several important electrocatalytic processes, including the electrocatalytic CO2 reduction reaction (CO2RR). The unique local environment around the singly dispersed metal site in TM‐N‐C catalysts is likely to be responsible for their catalytic properties, which differ significantly from those of bulk or nanostructured catalysts. However, the identification of the actual working structure of the main active units in TM‐N‐C remains a challenging task due to the fluctional, dynamic nature of these catalysts, and scarcity of experimental techniques that could probe the structure of these materials under realistic working conditions. This issue is addressed in this work and the local atomistic and electronic structure of the metal site in a Co–N–C catalyst for CO2RR is investigated by employing time‐resolved operando X‐ray absorption spectroscopy (XAS) combined with advanced data analysis techniques. This multi‐step approach, based on principal component analysis, spectral decomposition and supervised machine learning methods, allows the contributions of several co‐existing species in the working Co–N–C catalysts to be decoupled, and their XAS spectra deciphered, paving the way for understanding the CO2RR mechanisms in the Co–N–C catalysts, and further optimization of this class of electrocatalytic systems. Abstract

Summary: Background: SerpinB3 is a cysteine protease inhibitor involved in liver disease progression due to its proinflammatory and profibrogenic properties. The polymorphic variant SerpinB3‐PD (SB3‐PD), presents a substitution in its reactive centre loop, determining the gain of function. Aims: To disclose the clinical characteristics of a cohort of patients with cirrhosis in relation to the presence of SB3‐PD and to assess the effect of this genetic variant on fibrogenic and inflammatory cytokines in vitro. Methods: We assessed SB3 polymorphism in 90 patients with cirrhosis, prospectively followed up in our referral centre. We used HepG2 and HuH‐7 cells transfected to overexpress either wild‐type SB3 (SB3‐WT) or SB3‐PD to assess their endogenous effect, while LX2 and THP‐1 cells were treated with exogenous SB3‐WT or SB3‐PD proteins. Results: Patients carrying SB3‐PD had more severe portal hypertension and higher MELD scores, than patients carrying SB3‐WT. In multivariate analysis, SB3‐PD was an independent predictor of cirrhosis complications. Patients with SB3‐PD polymorphism presented with more severe liver fibrosis and inflammatory features. Hepatoma cells overexpressing SB3‐PD showed higher TGF‐β1 expression than controls. The addition of recombinant SB3‐PD induced an up‐regulation of TGF‐β1 in LX2 cells and a more prominent inflammatory profile in THP‐1 cells, compared to the effect of SB3‐WT protein. Conclusions: The polymorphic variant SB3‐PD is highly effective in determining activation of TGF‐β1 and inflammation in vitro. Patients with cirrhosis who carry SB3‐PD polymorphism may be more prone to develop severe liver disease progression. Abstract

Although Italy was one of the first European countries to tolerate the existence of a fascist party, the democratic governments of its immediate postwar engaged in a genuine and important reflection on the means to counter the re-emergence of fascism. Thus Italy too, like its European counterparts, sought to conceive of itself as a militant democracy. This article aims to illuminate its efforts and its approach, and also to cast light on the evident limits and contradictions. Looking at the Italian case while observing how other European countries tackled the re-emergence of fascism in the same period will make clear the intrinsic difficulties of turning a democracy into a militant one only a short time after the fall of an authoritarian regime and in the aftermath of an inevitably problematic transition.
