The "common sense" intervention by toxicology journal editors regarding proposed European Union endocrine disrupter regulations ignores scientific evidence and well-established principles of chemical risk assessment. In this commentary, endocrine disrupter experts express their concerns about a recently published, and is in our considered opinion inaccurate and factually incorrect, editorial that has appeared in several journals in toxicology. Some of the shortcomings of the editorial are discussed in detail. We call for a better founded scientific debate which may help to overcome a polarisation of views detrimental to reaching a consensus about scientific foundations for endocrine disrupter regulation in the EU. © 2013 Bergman et al.; licensee BioMed Central Ltd.

Abstract The cerebral cortex extends axonal projections to several subcortical brain regions, including the striatum, thalamus, superior colliculus, and pontine nuclei. Experimental tract-tracing studies have shown that these subcortical projections are topographically organized, reflecting the spatial organization of sensory surfaces and body parts. Several public collections of mouse- and rat- brain tract-tracing data are available, with the Allen mouse brain connectivity atlas being most prominent. There, a large body of image data can be inspected, but it is difficult to combine data from different experiments and compare spatial distribution patterns. To enable co-visualization and comparison of topographical organization in mouse brain cortico-subcortical projections across experiments, we represent axonal labelling data as point data in a common 3D brain atlas space. We here present a collection of point-cloud data representing spatial distribution of corticostriatal, corticothalamic, corticotectal, and corticopontine projections in mice and exemplify how these spatially integrated point data can be used as references for experimental investigations of topographic organization in transgenic mice, and for cross-species comparison with corticopontine projections in rats.

Introduction Despite technological developments and intensified care, pregnancies in women with pre-existing diabetes are still considered high-risk pregnancies. The rate of adverse outcomes in pregnancies affected by diabetes in Denmark is currently unknown, and there is a limited understanding of mechanisms contributing to this elevated risk. To address these gaps, the Danish Diabetes Birth Registry 2 (DDBR2) was established. The aims of this registry are to evaluate maternal and fetal-neonatal outcomes based on 5 years cohort data, and to identify pathophysiology and risk factors associated with short-term and long-term outcomes of pregnancies in women with pre-existing diabetes.Methods and analysis The DDBR2 registry is a nationwide 5-year prospective cohort with an inclusion period from February 2023 to February 2028 of pregnancies in women with all types of pre-existing diabetes and includes registry, clinical and questionnaire data and biological samples of mother–partner–child trios. Eligible families (parents age ≥18 years and sufficient proficiency in Danish or English) can participate by either (1) basic level data obtained from medical records (mother and child) and questionnaires (partner) or (2) basic level data and additional data which includes questionnaires (mother and partner) and blood samples (all). The primary maternal outcome is Hemoglobin A1c (HbA1c) levels at the end of pregnancy and the primary offspring endpoint is the birth weight SD score. The DDBR2 registry will be complemented by genetic, epigenetic and metabolomic data as well as a biobank for future research, and the cohort will be followed through data from national databases to illuminate possible mechanisms that link maternal diabetes and other parental factors to a possible increased risk of adverse long-term child outcomes.Ethics and dissemination Approval from the Ethical Committee is obtained (S-20220039). Findings will be sought published in international scientific journals and shared among the participating hospitals and policymakers.Trial registration number NCT05678543.

Abstract Self-harm among adolescents has increased in many countries, but few studies have examined possible explanations. One explanation could be the changes in the way adolescents socialize and use of social media. We explored the relationship between past year self-harm and time spent on social media, employing data from a nationwide cross-sectional survey among students in grades 8 through 11 in Norway (N = 37,268). The association was estimated in logistic regression models and we adjusted for identified confounders and stratified on gender, age group and depressive symptoms. A total of 16.1% of the study population reported to have self-harmed in the past year. This proportion was elevated among those spending more than 3 h daily on social media (unadjusted OR = 2.74 (CI 2.58.–2.90)). Adjustment for confounders modified the association (OR = 1.49 (CI 1.39–1.60)). In stratified analyses, adjusted OR did not differ significantly by gender or age The association between time spent on social media and self-harm was weaker among adolescents with severe depressive symptoms (adjusted OR = 1.38 (CI 1.22–1.55)), than among those with mild or no symptoms (adjusted OR = 1.70 (CI 1.56–1.86)). Risk of self-harm was elevated among those who spent 3 or more hours daily on social media, also after controlling for other factors. Further studies are needed to explore the nature and underlying mechanisms of this association. Strengthening the evidence will help informing the development of adequate measures to prevent self-harm.

This study investigates the challenges and potential of conventional injection molding for producing thick-walled optical components. The research primarily focuses on optimizing process parameters and mold design to enhance product quality. The methods include software simulations and experimental validation using polycarbonate test samples (optical lenses). Significant parameters such as melt temperature, mold temperature, injection pressure, and packing pressure were varied to assess their impact on geometric accuracy and visual properties. The results show that lower melt temperatures and higher mold temperatures significantly reduce the occurrence of dimensional defects. Additionally, the design of the gate system was found to be crucial in minimizing defects and ensuring uniform material flow. Effective packing pressure was essential in reducing volumetric shrinkage and sink marks. Furthermore, we monitored the deviation between the predicted and actual defects relative to the thickness of the sample wall. After optimization, the occurrence of obvious defects was eliminated across all sample thicknesses (lenses), and the impact of the critical defect, the sink mark on the planar side of the lens, was minimized. These findings demonstrate the substantial potential of conventional injection molding to produce high-quality thick-walled parts when these parameters are precisely controlled. This study provides valuable insights for the efficient design and manufacturing of optical components, addressing the growing demand for high-performance thick-walled plastic products.

This work is concerned with forming, specifically deep drawing, and its influence on the micro-mechanical properties of sheet metal. In practice, there are several applications in which fractions can occur due to weak spots in the deep-drawn sheet metal, especially after long-term use. The deep drawing process was carried out on BUP–600 machines using the LHD (Limiting Dome Height) method, which uses a forming tool with a diameter of 100 mm and bead groove. Sheet metals X8CrMnNi19-6-3 (1.4376) with thicknesses of 1, 1.5, and 3 mm were selected for this process. To study the effect of a lubricant on the formability of the sheet metal, deep drawing without and with a lubricant was compared. An FEM analysis was conducted to identify critical points in the deep drawing process, and the results were later compared with real results. The analysis was conducted using the AutoForm program. The micro-mechanical properties of these points were subsequently examined. The specified points on the formed part showed significant differences in their micro-mechanical properties, suggesting a higher strength but also less resistance to fractures. The difference in micro-mechanical properties (indentation and Vickers hardness) in points that were not deep-drawn and points located in critical areas was up to 86%. Significant changes in behavior were found in the indentation modulus and plastic/elastic deformation work as well. This study demonstrates the significant effect of the use of a lubricant in achieving the deep drawing of the sheet metal. The application of a lubricant resulted in a 33% increase in drawing range compared to drawing without lubrication. This study has a significant influence on the deep drawing of sheet metals in practice, showing the fundamental influence of the lubricant on the drawing process and also showing the problem of critical points that need to be eliminated.

This research investigates the micro-mechanical and tribological properties of injection-molded parts made from polypropylene. The tribological properties of polymers are a very interesting area of research. Understanding tribological processes is very crucial. Considering that the mechanical and tribological properties of injected parts are not uniform at various points of the part, this research was conducted to explain the non-homogeneity of properties along the flow path. Non-homogeneity can be influenced by numerous factors, including distance from the gate, mold and melt temperature, injection pressure, crystalline structure, cooling rate, the surface of the mold, and others. The key factor from the micro-mechanical and tribological properties point of view is the polymer morphology (degree of crystallinity and size of the skin and core layers). The morphology is influenced by polymer flow and the injection molding process conditions. Gained results indicate that the indentation method was sufficiently sensitive to capture the changes in polypropylene morphology, which is a key parameter for the resulting micro-mechanical and tribological properties of the part. It was proven that the mechanical and tribological properties are not equal in varying regions of the part. Due to cooling and process parameters, the difference in the indentation modulus in individual measurement points was up to 55%, and the tribological properties, in particular the friction coefficient, showed a difference of up to 20%. The aforementioned results indicate the impact this finding signifies for injection molding technology in technical practice. Tribological properties are a key property of the part surface and, together with micro-mechanical properties, characterize the resistance of the surface to mechanical failure of the plastic part when used in engineering applications. A suitable choice of gate location, finishing method of the cavity surface, and process parameters can ensure the improvement of mechanical and tribological properties in stressed regions of the part. This will increase the stiffness and wear resistance of the surface.

Neuroscientists employ a range of methods and generate increasing amounts of data describing brain structure and function. The anatomical locations from which observations or measurements originate represent a common context for data interpretation, and a starting point for identifying data of interest. However, the multimodality and abundance of brain data pose a challenge for efforts to organize, integrate, and analyze data based on anatomical locations. While structured metadata allow faceted data queries, different types of data are not easily represented in a standardized and machine-readable way that allow comparison, analysis, and queries related to anatomical relevance. To this end, three-dimensional (3D) digital brain atlases provide frameworks in which disparate multimodal and multilevel neuroscience data can be spatially represented. We propose to represent the locations of different neuroscience data as geometric objects in 3D brain atlases. Such geometric objects can be specified in a standardized file format and stored as location metadata for use with different computational tools. We here present the Locare workflow developed for defining the anatomical location of data elements from rodent brains as geometric objects. We demonstrate how the workflow can be used to define geometric objects representing multimodal and multilevel experimental neuroscience in rat or mouse brain atlases. We further propose a collection of JSON schemas (LocareJSON) for specifying geometric objects by atlas coordinates, suitable as a starting point for co-visualization of different data in an anatomical context and for enabling spatial data queries.

Abstract Shift current photovoltaic devices are potential candidates for future cheap, sustainable, and efficient electricity generation. In the present work, we calculate the solar-generated shift current and efficiencies in 326 different 2D materials obtained from the computational database C2DB. We apply, as metrics, the efficiencies of monolayer and multilayer samples. The monolayer efficiencies are generally found to be low, while the multilayer efficiencies of infinite stacks show great promise. Furthermore, the out-of-plane shift current response is considered, and material candidates for efficient out-of-plane shift current devices are identified. Among the screened materials, MXY Janus and MX2 transition metal dichalchogenides (TMDs) constitute a prominent subset, with chromium based MXY Janus TMDs holding particular promise. Finally, in order to explain the band gap dependence of the PV efficiency, a simple gapped graphene model with a variable band gap is established and related to the calculated efficiencies.

This work is dedicated to the design of flexible composite elements, specifically leaf springs. The design of these flexible composite elements took in consideration the technologies, materials and intermediate goods that are available and useable in laboratory manufacturing and the possibility for the transfer of gained knowledge to industrial practice. This work deals with individual types of materials and their processability and usability for the manufacturing of composite products exposed to cyclic stress. The impact of the designed lay-up diagrams and cyclic load boundary on the fatigue behaviour of manufactured specimens was used to evaluate the effect of cyclic stressing. Based on this assessment, a conclusion and recommendation were formulated for the serial manufacturing of flexible composite elements.

The modification of polymer materials’ useful properties can be applicable in many industrial areas due to the ability to make commodity and technical plastics (plastics that offer many benefits, such as processability, by injection molding) useful in more demanding applications. In the case of injection-molded parts, one of the most suitable methods for modification appears to be high-energy irradiation, which is currently used primarily for the modification of mechanical and thermal properties. However, well-chosen doses can effectively modify the properties of the surface layer as well. The purpose of this study is to provide a complex description of high-energy radiation’s (β radiation) influence on the useful properties of injection-molded parts made from common polymers. The results indicate that β radiation initiates the cross-linking process in material and leads to improved mechanical properties. Besides the cross-linking process, the material also experiences oxidation, which influences the properties of the surface layer. Based on the measured results, the main outputs of this study are appropriately designed regression models that determine the optimal dose of radiation.

Brain atlases are widely used in neuroscience as resources for conducting experimental studies, and for integrating, analyzing, and reporting data from animal models. A variety of atlases are available, and it may be challenging to find the optimal atlas for a given purpose and to perform efficient atlas-based data analyses. Comparing findings reported using different atlases is also not trivial, and represents a barrier to reproducible science. With this perspective article, we provide a guide to how mouse and rat brain atlases can be used for analyzing and reporting data in accordance with the FAIR principles that advocate for data to be findable, accessible, interoperable, and re-usable. We first introduce how atlases can be interpreted and used for navigating to brain locations, before discussing how they can be used for different analytic purposes, including spatial registration and data visualization. We provide guidance on how neuroscientists can compare data mapped to different atlases and ensure transparent reporting of findings. Finally, we summarize key considerations when choosing an atlas and give an outlook on the relevance of increased uptake of atlas-based tools and workflows for FAIR data sharing.

BackgroundTumor necrosis factor (TNF) is pathologically elevated in human abdominal aortic aneurysms (AAA). Non-selective TNF inhibition-based therapeutics are approved for human use but have been linked to several side effects. Compounds that target the proinflammatory soluble form of TNF (solTNF) but preserve the immunomodulatory capabilities of the transmembrane form of TNF (tmTNF) may prevent these side effects. We hypothesize that inhibition of solTNF signaling prevents AAA expansion.MethodsThe effect of the selective solTNF inhibitor, XPro1595, and the non-selective TNF inhibitor, Etanercept (ETN) was examined in porcine pancreatic elastase (PPE) induced AAA mice, and findings with XPro1595 was confirmed in angiotensin II (ANGII) induced AAA in hyperlipidemic apolipoprotein E (Apoe) –/– mice.ResultsXPro1595 treatment significantly reduced AAA expansion in both models, and a similar trend (p = 0.06) was observed in PPE-induced AAA in ETN-treated mice. In the PPE aneurysm wall, XPro1595 improved elastin integrity scores. In aneurysms, mean TNFR1 levels reduced non-significantly (p = 0.07) by 50% after TNF inhibition, but the histological location in murine AAAs was unaffected and similar to that in human AAAs. Semi-quantification of infiltrating leucocytes, macrophages, T-cells, and neutrophils in the aneurysm wall were unaffected by TNF inhibition. XPro1595 increased systemic TNF levels, while ETN increased systemic IL-10 levels. In ANGII-induced AAA mice, XPro1595 increased systemic TNF and IL-5 levels. In early AAA development, proteomic analyses revealed that XPro1595 significantly upregulated ontology terms including “platelet aggregation” and “coagulation” related to the fibrinogen complex, from which several proteins were among the top regulated proteins. Downregulated ontology terms were associated with metabolic processes.ConclusionIn conclusion, selective inhibition of solTNF signaling reduced aneurysm expansion in mice, supporting its potential as an attractive treatment option for AAA patients.

Today, more and more importance is given to the improvement of polymer materials’ wear resistance, i.e., their micro-mechanical and tribological properties, which could widen their application in practice. The properties of materials can be modified by several methods, among them exposure to electron radiation. This study focuses on the effect of varying radiation intensity (15 kGy to 99 kGy), depth of penetration, and subsequent structure modification of injection-molded polypropylene on tribological and micro-mechanical properties. Electron radiation influences the structure of individual layers, thus improving or degrading their properties. Hence, the depth of penetration can be examined by instrumented hardness tests and scratch tests. Due to irradiation, surface properties and wear resistance increased by up to 105% (from 38 MPa to 78 MPa). As the results show, an increase in mechanical properties was recorded in the direction towards the center of the sample (from 72 MPa to 82 MPa). Micro-mechanical tests were also confirmed by the observation of cross-linking changes (gel test) as well as crystallinity increases (wide-angle X-ray diffraction and microtome cuts). This finding could have a significant effect on the manufacturing and subsequent modification of injection-molded polypropylene parts, which opens new possibilities in practice for this material. The increased surface wear resistance enables the use of parts for which the durability and abrasion resistance of the surface are demanded, especially in applications facing exposure to long-term cyclical loads (e.g., gears).

Nowadays, technical practice puts emphasis on improving selected material properties of polymers which could lead to new applications. Material properties can be modified in numerous ways, among which is radiation treatment. This study looks into the influence of beta radiation on several properties of polyamide 6, e.g., indentation hardness, modulus and creep. Main changeable parameters were the concentration of triallyl isocyanurate (TAIC), which promotes cross-linking, and intensity of radiation. The concentration was in the range from 2 to 6 wt.%, while the radiation dose was 0, 66, 99 and 132 kGy. The treated materials were measured for indentation hardness, modulus and creep. Degree of cross-linking was verified by thermo-mechanical analysis (TMA), while degradation processes was investigated by Fourier-transform infrared spectroscopy (FTIR). The results indicate that electron radiation positively affects the tested material properties. The best results were seen in polyamide with 6 wt.% of TAIC, which demonstrated a 38% improvement in mechanical properties after exposure to 132 kGy. This improvement in properties affects the final parts and their application (e.g., in the automotive industry—engine parts; in electrical engineering—insulation of wires and cables; and in industry—pipes for underfloor heating, etc.).

Cross-linking is a process in which polymer chains are associated through chemical bonds. The cross-linking level can be adjusted by the irradiation dosage and often by means of a cross-linking booster. The polymer additional cross-linking influences the surface nano and micro layers in the way comparable to metals during the thermal and chemical-thermal treatments. Polybutylene terephthalate (PBT) can be found in a group of structural polymers, which are often used in industry, especially in automotive. Applying the technology of electron radiation induces a creation of 3D network structure, which improves the local mechanical properties. These were later measured by a depth sensing indentation (DSI) test. This state of the art method is based on immediate detection of indentation depth in relation to applied force. The creation of 3D network caused an increase in nano-mechanical properties values, such as indentation hardness and indentation modulus, in comparison to the virgin material. The indentation hardness rose by 80%, while the indentation modulus elevated by 62%. The selected structural materials, e.g. PBT, were modified by the electron irradiation in a positive way and as such could be moved to a group of high performance materials.

Cross-linking is a process in which polymer chains are associated through chemical bonds. Radiation, which penetrated through specimens and reacted with the cross-linking agent, gradually formed cross-linking (3D net), first in the surface layer and then in the total volume, which resulted in considerable changes in specimen behaviour. Creep value is an important parameter that describes the behaviour of a material for the duration of its exposure to long term stress. Most of the technical parts used in industry are subjected to the long term stress for their whole life cycle. This can lead to material creep, which directly results in the transformation of dimensions. To eliminate this problem, a number of construction materials was chosen and subsequently irradiated by a source of electrons. This created a 3D network within the polymer structure, which led to an increase of the micro-mechanical and micro-creep properties. Evaluation of these modifications was done by state of the art device Micro-combi tester manufactured by Anton Paar. This device lowers the time required to measure the creep by standard technology and it fluently records the changes of indentation depth in relation to applied force. This dependence is then used to calculate the creep values. Due to the electron irradiation, a 40 % increase was reached in creep resistance; therefore the useful properties of selected construction materials were improved.

This research paper is concerned with the experimental study of high-feed end milling of 1.4541 (X6CrNiTi18-10) stainless steel with replaceable cermet plates. Several machining operations were performed under different cutting conditions. The variable values were depth of cut, feed per tooth and cutting speed. The results were analyzed, and cutting forces were evaluated for dependence on cutting conditions (cutting speed, depth of cut, feed per tooth). The obtained data were statistically processed and plotted in graphs. It was found that the percentage distribution of cutting forces changed as the tool load increased. The ratio of forces acting in individual axes also changed with varying trends. An increasing trend was recorded in the x and y axes, while a decreasing trend was recorded in the z axis. Measured change, approximately 10%, can no longer be neglected as it can significantly influence the clamping stability of a part.

Background The objective was to evaluate predictive performance and optimal decision threshold of the Kryptor soluble fms‐like tyrosine kinase‐1 (sFlt‐1)/placental growth factor (PlGF) ratio when implemented for routine management of women presenting with symptoms of preeclampsia. Methods and Results Observational retrospective study of a cohort of 501 women with suspected preeclampsia after 20 weeks of gestation. Women referred to maternity ward for observation of preeclampsia had an sFlt‐1/PlGF ratio test included in routine diagnostic workup. Maternal and offspring characteristic data included maternal risk factors, outcomes, delivery mode, and indication for suspected preeclampsia. Biochemical measurements to determine sFlt‐1/PlGF ratio were performed using the BRAHMS/Kryptor sFlt‐1/PlGF ratio immunoassays. Results were analyzed by area under receiver‐operating characteristic curve. Preeclampsia occurred in 150 of 501 (30%) of symptomatic women with an sFlt‐1/PlGF ratio determined before the time of diagnosis. Area under receiver‐operating characteristic curve for diagnosis of early‐onset preeclampsia within 1 and 4 weeks was 0.98 (95% CI, 0.96–1.00) and 0.95 (95% CI, 0.92–0.98), respectively. For late‐onset preeclampsia, predictive performance within 1 and 4 weeks was lower: 0.90 (95% CI, 0.85–0.94) and 0.85 (95% CI, 0.80–0.90), respectively. The optimal single sFlt‐1/PlGF ratio threshold for all preeclampsia and late‐onset preeclampsia within 1 and 4 weeks was 66. The negative and positive predictive values for ruling out and ruling in developing preeclampsia within 1 week were 96% and 70%, respectively. Conclusions The Kryptor sFlt‐1/PlGF ratio is a useful clinical tool ruling out and in preeclampsia within 1 week. Prediction within 4 weeks is superior for early‐onset preeclampsia. A single decision threshold of 66 is indicated for use in clinical routine.

This study describes the influence of polymer flow length on mechanical properties of tested polymer, specifically polycarbonate. The flow length was examined in a spiral shaped mould. The mould cavity’s surface was machined by several methods, which led to differing roughness of the surface. The cavity was finished by milling, grinding and polishing. In order to thoroughly understand the influence of the mould surface quality on the flow length, varying processing parameters, specifically the pressure, were used. The polymer part was divided into several segments, in which the micro-mechanical properties, such as hardness and indentation modulus were measured. The results of this study provide interesting data concerning the flow length, which was up to 3% longer for rougher surfaces, but shorter in cavities with polished surface. These results are in disagreement with the commonly practiced theory, which states that better surface quality leads to greater flow length. Furthermore, evaluation of the micro-mechanical properties measured along the flow path demonstrated significant variance in researched properties, which increased by 35% (indentation hardness) and 86% by indentation modulus) in latter segments of the spiral in comparison with the gate.

This study focuses on the problematic of polyamide 6 containing various concentrations of cross-linking agent that was exposed to electron radiation. It is important to improve the material properties of polymers as much as possible. This endeavor can be realized by numerous methods, one of which is radiation exposure. This study investigates the effect of electron beam radiation in doses ranging from 66 to 132 kGy on the micro-mechanical properties of polymers, specifically polyamide 6 filled with 1, 3 and 5 wt.% of cross-linking agent triallyl isocyanurate (TAIC). The changes in the material brought by the radiation exposure were quantified by measurements of indentation hardness and modulus, which were the main measured micro-mechanical properties. Furthermore, thermo-mechanical analysis (TMA) was chosen to confirm the results of the material cross-linking, while the effect of degradation was investigated by Fourier-transform infrared spectroscopy (FTIR). In pursuit of complete evaluation, the topography of the test subject’s surface was explored by atomic force microscopy (AFM). The optimal concentration/radiation ratio was found in polyamide 6 enriched by 5 wt.% concentration of TAIC, which was irradiated by 132 kGy. Material treated in such a way had its indentation hardness by 33% and indentation modulus improved by 26% in comparison with the untreated material. These results were subsequently confirmed by the TMA and FTIR methods.

Background: The issue of endocrine disrupting chemicals (EDCs) is receiving wide attention from both the scientific and regulatory communities. Recent analyses of the EDC literature have been criticized for failing to use transparent C exposures to adverse health or environmental outcomes. Systematic review methodologies are ideal for addressing this issue as they provide transparent and consistent approaches to study selection and evaluation. Objective methods are needed for integrating the multiple streams of evidence (epidemiology, wildlife, laboratory animal, in vitro, and in silico data) that are relevant in assessing EDCs. Methods: We have developed a framework for the systematic review and integrated assessment (SYRINA) of EDC studies. The framework was designed for use with the International Program on Chemical Safety (IPCS) and World Health Organization (WHO) definition of an EDC, which requires appraisal of evidence regarding 1) association between exposure and an adverse effect, 2) association between exposure and endocrine disrupting activity, and 3) a plausible link between the adverse effect and the endocrine disrupting activity. Results: Building from existing methodologies for evaluating and synthesizing evidence, the SYRINA framework includes seven steps: 1) Formulate the problem; 2) Develop the review protocol; 3) Identify relevant evidence; 4) Evaluate evidence from individual studies; 5) Summarize and evaluate each stream of evidence; 6) Integrate evidence across all streams; 7) Draw conclusions, make recommendations, and evaluate uncertainties. The proposed method is tailored to the IPCS/WHO definition of an EDC but offers flexibility for use in the context of other definitions of EDCs. Conclusions: When using the SYRINA framework, the overall objective is to provide the evidence base needed to support decision making, including any action to avoid/minimise potential adverse effects of exposures. This framework allows for the evaluation and synthesis of evidence from multiple evidence streams. Finally, a decision regarding regulatory action is not only dependent on the strength of evidence, but also the consequences of action/inaction, e.g. limited or weak evidence may be sufficient to justify action if consequences are serious or irreversible.

BACKGROUND:Gestational diabetes mellitus (GDM) is a common pregnancy complication associated with adverse outcomes including preeclampsia, caesarean section, macrosomia, neonatal morbidity and future development of type 2 diabetes in both mother and child. Current selective screening strategies rely on clinical risk factors such as age, family history of diabetes, macrosomia or GDM in a previous pregnancy, and they possess a relatively low specificity. Here we hypothesize that novel first trimester protein predictors of GDM can contribute to the current selective screening strategies for early and accurate prediction of GDM, thus allowing for timely interventions. METHODS:A proteomics discovery approach was applied to first trimester sera from obese (BMI ≥27 kg/m2) women (n = 60) in a nested case-control study design, utilizing tandem mass tag labelling and tandem mass spectrometry. A subset of the identified protein markers was further validated in a second set of serum samples (n = 210) and evaluated for their contribution as predictors of GDM in relation to the maternal risk factors, by use of logistic regression and receiver operating characteristic analysis. RESULTS:Serum proteomic profiling identified 25 proteins with significantly different levels between cases and controls. Three proteins; afamin, serum amyloid P-component and vitronectin could be further confirmed as predictors of GDM in a validation set. Vitronectin was shown to contribute significantly to the predictive power of the maternal risk factors, indicating it as a novel independent predictor of GDM. CONCLUSIONS:Current selective screening strategies can potentially be improved by addition of protein predictors.

This study describes the effect of electron radiation on the nano-mechanical properties of surface layers of selected polyamide (PA) types. Electron radiation initiates the cross-linking of macromolecules in the polyamide structure, leading to the creation of a 3D network which fundamentally changes the properties of the tested polymers. Selected types of polyamide (PA 6, PA 66 and PA 9T) were exposed to various intensities of electron radiation (33 kGy, 66 kGy, 99 kGy, 132 kGy, 165 kGy and 198 kGy). The cross-linked polyamides’ surface properties were measured by means of the modern nano-indentation technique (Depth Sensing Indentation; DSI), which operates on the principle of the immediate detection of indenter penetration depth in dependence on the applied load. The evaluation was preformed using the Oliver–Pharr method. The effect of electron radiation on the tested polyamides manifested itself in the creation of a 3D network, which led to an increase of surface layer properties, such as indentation hardness, elastic modulus, creep and temperature resistance, by up to 93%. The increase of temperature and mechanical properties substantially broadens the field of application of these materials in technical practice, especially when higher temperature resistance is required. The positive changes to the nano-mechanical properties as well as mechanical and temperature capabilities instigated by the cross-linking process were confirmed by the gel volume test. These measurements lay the foundation for a detailed study of this topic, as well as for a more effective means of modifying chosen properties of technical polyamide products by radiation.

This study’s goal was to describe the influence of a wide range of ionizing beta radiation upon the changes in surface layer mechanical properties and structural modifications of selected types of polymer. Radiation crosslinking is a process whereby the impingement of high-energy electrons adjusts test sample structures, thus enhancing the useful properties of the material, e.g., hardness, wear-resistance, and creep, in order that they may function properly during their technical use. The selected polymers tested were polyolefin polymers like polyethylene (Low-density polyethylene LDPE, High-density polyethylene HDPE). These samples underwent exposure to electron radiation of differing dosages (33, 66, 99, 132, 165, and 198 kGy). After the crosslinking process, the samples underwent testing of the nano-mechanical properties of their surface layers. This was done by means of a state-of-the-art indentation technique, i.e., depth-sensing indentation (DSI), which detects the immediate change in the indentation depth associated with the applied force. Indeed, the results indicated that the optimal radiation dosage increased the mechanical properties by up to 57%; however, the beneficial levels of radiation varied with each material. Furthermore, these modifications faced examination from the structural perspective. For this purpose, a gel test, Raman spectroscopy, and crystalline portion determination by X-ray all confirmed the assumed trends.

Introduction: Corpus luteum (CL) produces progesterone (P4) and 17-OH progesterone (17-OH P4) during the luteal phase. Contrary to P4, 17-OH P4 is not supplied as part of the luteal phase support following IVF-treatment. Therefore, measuring endogenous serum 17-OH P4 levels may more accurately reflect the CL function compared to monitoring serum P4 concentrations.Objective: To explore the correlation between mid-luteal serum 17-OH P4 levels and live birth rates and to explore the possible daytime variations in mid-luteal serum 17-OH P4.Design: Prospective cohort study.Patients: 614 women undergoing IVF-treatment and fresh embryo transfer.Intervention: All patients had serum 17-OH P4 measured 7 days after oocyte pick-up (OPU+7). Furthermore, on OPU+7, seven patients underwent repeated blood sampling during daytime to clarify the endogenous daytime secretory pattern of 17-OH P4.Outcome measure: Live birth rate.Secondary outcome measure: Daytime variation in serum 17-OH P4 levels.Results: The highest chance of a live birth was seen with mid-luteal 17-OH P4 between 6.0 and 14.0 nmol/l. The chance of a live birth was reduced below (RD −10%, p = 0.07), but also above the optimal range for 17-OH P4 (RD −12%, p = 0.04). Patients with diminished CL-function (17-OH P4 < 6 nmol/l) displayed clinically stable 17-OH P4 values, whereas patients with 17-OH P4 levels >6 nmol/l showed random 17-OH P4 fluctuations during daytime.Conclusion: The association between 17-OH P4 and reproductive outcomes is non-linear, and the negative effect of excessive CL-secretion seems to be just as strong as the negative effect of a reduced CL-function during the peri-implantation period.

In experimental neuroscientific research, anatomical location is a key attribute of experimental observations and critical for interpretation of results, replication of findings, and comparison of data across studies. With steadily rising numbers of publications reporting basic experimental results, there is an increasing need for integration and synthesis of data. Since comparison of data relies on consistently defined anatomical locations, it is a major concern that practices and precision in the reporting of location of observations from different types of experimental studies seem to vary considerably. To elucidate and possibly meet this challenge, we have evaluated and compared current practices for interpreting and documenting the anatomical location of measurements acquired from murine brains with different experimental methods. Our observations show substantial differences in approach, interpretation and reproducibility of anatomical locations among reports of different categories of experimental research, and strongly indicate that ambiguous reports of anatomical location can be attributed to missing descriptions. Based on these findings, we suggest a set of minimum requirements for documentation of anatomical location in experimental murine brain research. We furthermore demonstrate how these requirements have been applied in the EU Human Brain Project to optimize workflows for integration of heterogeneous data in common reference atlases. We propose broad adoption of some straightforward steps for improving the precision of location metadata and thereby facilitating interpretation, reuse and integration of data.

Calls for implementing new models of development cooperation have been on the increase including a call for increased use of horizontal development cooperation mechanisms such as South-South Cooperation (SSC) as a way to enhance aid effectiveness in the health sector of developing countries. In this policy and practice paper, we review recent experiences in application of SSC initiatives to two public health situations in Africa to demonstrate the veracity of this new paradigm. Our review highlight the immense benefits associated with the use of SSC for health and provide evidence for increasing use of horizontal development coordination mechanisms to strengthen public health services delivery and socioeconomic development among African countries. Opportunities for SSC among African countries include in the areas of disease prevention and control, production of medical products and essential medicines, harmonization of regulatory processes, and health workforce development among others. However, pitfalls such as poor coordination, inadequate political commitment, lack of conducive policy environments, language barrier and inadequate financing opportunities for SSC initiatives present major dilemma for the use of SSC mechanisms. We conclude that the need for a paradigm shift from vertical to horizontal development cooperation needs no further proof but a call to action. We call on the concerned stakeholders to support the establishment of a systematic approach for use of SSC mechanisms in the health sector of Africa, designation of an African Centre of Excellence for SSC in public health and development of a regional mechanism for monitoring and evaluation of SSC initiatives in Africa.

Antecedentes: La aterosclerosis es una arteriopatía inflamatoria, crónica y progresiva que conlleva a disfunción del endotelio vascular, con estenosis y obstrucción de los vasos sanguíneos. A nivel coronario causa infarto agudo de miocardio y angina de pecho, uno de los principales motivos de consulta en las salas de urgencia. La inflamación es un factor emergente que actualmente es un campo activo de investigación científica en la etiopatogenia de la lesión aterosclerótica. Objetivos: Cuantificar los niveles basales de citocinas proinflamatorias en pacientes con aterosclerosis y correlacionar la concentración de estas con comorbilidades de la población en estudio. Métodos: Se realizó estudio descriptivo y analítico transversal en 31 pacientes mayores de 35 años, con diagnóstico presuntivo de cardiopatía isquémica en dos centros de hemodinámia en la ciudad de Barranquilla. Se cuantificó interleucina 6 (IL-6), factor de necrosis tumoral alfa (TNF-α), proteína quimioatrayente de monocitos 1 (MCP-1) y factor de crecimiento de endotelio vascular (VEGF), utilizando el kit comercial Milliplex® para Luminex y siguiendo las especificaciones del fabricante. El análisis estadístico se realizó usando el software Minitab 18®, Excel 2010® de Microsoft. Resultados: Todos los pacientes presentaban enfermedad coronaria aterosclerótica severa con más de 70 % de oclusión en uno o más vasos. La concentración media de IL-6 fue de 7584 pg/mL (con intervalo de confianza de 95 % [95 %] = 0-845.32); 67 % (20 pacientes) tenía concentración de MCP-1 > 10 000 pg/mL. La concentración media de TNF-α fue de 38.16 pg/mL, IC 95 % = 16.01-23.72; todos los pacientes tuvieron una concentración de VEGF

ABSTRACT The ability to change cell morphology is an advantageous characteristic adopted by multiple pathogenic bacteria in order to evade host immune detection and assault during infection. Uropathogenic Escherichia coli (UPEC) exhibits such cellular dynamics and has been shown to transition through a series of distinct morphological phenotypes during a urinary tract infection. Here, we report the first systematic spatio-temporal gene expression analysis of the UPEC transition through these phenotypes by using a flow chamber-based in vitro infection model that simulates conditions in the bladder. This analysis revealed a novel association between the cell division gene damX and reversible UPEC filamentation. We demonstrate a lack of reversible bacterial filamentation in a damX deletion mutant in vitro and absence of a filamentous response by this mutant in a murine model of cystitis. While deletion of damX abrogated UPEC filamentation and secondary surface colonization in tissue culture and in mouse infections, transient overexpression of damX resulted in reversible UPEC filamentation. In this study, we identify a hitherto-unknown damX-mediated mechanism underlying UPEC morphotypical switching. Murine infection studies showed that DamX is essential for establishment of a robust urinary tract infection, thus emphasizing its role as a mediator of virulence. Our study demonstrates the value of an in vitro methodology, in which uroepithelium infection is closely simulated, when undertaking targeted investigations that are challenging to perform in animal infection models. IMPORTANCE Urinary tract infections (UTIs) are most often caused by uropathogenic Escherichia coli (UPEC) and account for a considerable health care burden. UPEC exhibits a dynamic lifestyle in the course of infection, in which the bacterium transiently adopts alternative morphologies ranging from rod shaped to coccoid and filamentous, rendering it better at immune evasion and host epithelium adhesion. This penchant for morphotype switching might in large measure account for UPEC’s success as a pathogen. In aiming to uncover genes underlying the phenomenon of UPEC morphotype switching, this study identifies damX, a cell division gene, as a mediator of reversible filamentation during UTI. DamX-mediated filamentation represents an additional pathway for bacterial cell shape control, an alternative to SulA-mediated FtsZ sequestration during E. coli uropathogenesis, and hence represents a potential target for combating UTI.

The main advantages of Thermoplastic Polyester Elastomers (TPE-E) are their elastomer properties as well as their ability to be processed in the same way as thermoplastic polymers (e.g., injection moulding, compression moulding and extrusion). However, TPE-Es’ properties, mainly their mechanical properties and thermal characteristics, are not as good as those of elastomers. Because of this TPE-Es are often modified with the aim of improving their properties and extending their range of application. Radiation cross-linking using accelerated electron beams is one of the most effective ways to change virgin polymers’ properties significantly. Their electrical (that is to say permittivity and resistivity measurements), mechanical (that is, tensile and impact tensile tests), as well as surface (that is, nano-indentation) properties were measured on modified/cross-linked TPE-E specimens with and/or without a cross-linking agent at irradiation doses of 0, 33, 66, 99, 132, 165 and 198 kGy. The data acquired from these procedures show significant changes in the measured properties. The results of this study allow the possibility of determining the proper processing parameters and irradiation doses for the production of TPE-E products which leads to the enlargement of their application in practice.

This article deals with the study of the utilisation of irradiated HDPE products after their end-of-life cycle. Today, polymer waste processing is a matter of evermore intensive discussion. Common thermoplastic waste recycling—especially in the case of wastes with a defined composition—is generally well-known—and frequently used. On the contrary, processing cross-linked plastics is impossible to do in the same way as with virgin thermoplastics—mainly due to the impossibility of remelting them. The possibility of using waste in the form of grit or a powder, made from cross-linked High Density PolyEthylene (rHDPEx) products, after their end-of-life cycle, as a filler for virgin Low Density PolyEthylene (LDPE) was tested in a matrix. It was found that both the mechanical behaviour and processability of new composites with an LDPE matrix, with rHDPEx as a filler, depend—to a high degree—on the amount of the filler. The composite can be processed up to 60% of the filler content. The Polymer Mixture Fluidity dropped significantly, in line with the amount of filler, while the mechanical properties, on the other hand, predominantly grew with the increasing amount of rHDPEx.

This article deals with the influence of electron-beam radiation on the micro-mechanical, thermo-mechanical, and structural properties of selected polymers. In the search for the desired improvement of polymers, it is possible to use, inter alia, one particular possible modification—Namely, crosslinking—Which is a process during which macromolecular chains start to connect to each other and, thus, create the spatial network in the structure. In the course of the treatment of the ionizing radiation, two actions can occur: crosslinking and scission of macromolecules, or degradation. Both these processes run in parallel. Using the crosslinking technology, standard and technical polymers can acquire the more “expensive” high-tech polymeric material properties and, thus, replace these materials in many applications. The polymers that were tested were selected from across the whole spectra of thermoplastics, ranging from commodity polymers, technical polymers, as well as high-performance polymers. These polymers were irradiated by different doses of beta radiation (33, 66, 99, 132, 165, and 198 kGy). The micro-mechanical and thermo-mechanical properties of these polymers were measured. When considering the results, it is obvious that irradiation acts on each polymer differently but, always when the optimal dose was found, the mechanical properties increased by up to 36%. The changes of micro-mechanical and thermo-mechanical properties were confirmed by structural measurement when the change of the micro-hardness and modulus corresponded to the crystalline phase change as determined by X-ray and gel content.

Some polymers need a cross-linking agent for the controlled cross-linking process of polymers with a tendency to degradation during the radiation cross-linking process. While, on the other hand, other polymers do not need a cross-linking agent—predominantly there are cross-linking polymers. The Thermo-Plastic Elastomer (TPE) that was used belongs to this group of predominantly cross-linking polymers; however, this agent is added because of faster reaction times and smaller irradiation doses. Microindentation–tensile and tensile impact tests were carried out on a thermoplastic sample—with, and without, a cross-linking agent. Small changes were measured between these materials at low radiation doses, (up to 66 kGy); nevertheless, at higher doses, the influence of the cross-linking agent on the mechanical properties is significant.

Background: Over 1800 food contact chemicals (FCCs) are known to migrate from food contact articles used to store, process, package, and serve foodstuffs. Many of these FCCs have hazard properties of concern, and still others have never been tested for toxicity. Humans are known to be exposed to FCCs via foods, but the full extent of human exposure to all FCCs is unknown., Objective: To close this important knowledge gap, we conducted a systematic overview of FCCs that have been monitored and detected in human biomonitoring studies according to a previously published protocol., Methods: We first compared the more than 14,000 known FCCs to five biomonitoring programs and three metabolome/exposome databases. In a second step, we prioritized FCCs that have been frequently detected in food contact materials and systematically mapped the available evidence for their presence in humans., Results: For 25% of the known FCCs (3601), we found evidence for their presence in humans. This includes 194 FCCs from human biomonitoring programs, with 80 of these having hazard properties of high concern. Of the 3528 FCCs included in metabolome/exposome databases, most are from the Blood Exposome Database. We found evidence for the presence in humans for 63 of the 175 prioritized FCCs included in the systematic evidence map, and 59 of the prioritized FCCs lack hazard data., Significance: Notwithstanding that there are also other sources of exposure for many FCCs, these data will help to prioritize FCCs of concern by linking information on migration and biomonitoring. Our results on FCCs monitored in humans are available as an interactive dashboard (FCChumon) to enable policymakers, public health researchers, and food industry decision-makers to make food contact materials and articles safer, reduce human exposure to hazardous FCCs and improve public health., Impact Statement: We present systematically compiled evidence on human exposure to 3601 food contact chemicals (FCCs) and highlight FCCs that are of concern because of their known hazard properties. Plastics entering the marine environment primarily originate from land-based sources, prompting significant attention on single-use plastic packaging. However, fishing plastic waste also contributes substantially to marine plastic pollution, though it is often overlooked in the literature due to the challenges in pinpointing pollution sources. This study addresses this key knowledge gap by synthesizing existing literature to explore and document the knowns and known unknowns surrounding fishing plastic waste's environmental, health, and socio-economic impacts. Through the development of a causal loop diagram, the study offers a preliminary understanding of the issue, serving as a foundation for a deeper exploration of the complexities within the fishing industry's plastic waste dynamics. Finally, the study highlights that short-sighted views and approaches are likely to lead to systemic failures. Food contact materials (FCMs) and food contact articles are ubiquitous in today's globalized food system. Chemicals migrate from FCMs into foodstuffs, so called food contact chemicals (FCCs), but current regulatory requirements do not sufficiently protect public health from hazardous FCCs because only individual substances used to make FCMs are tested and mostly only for genotoxicity while endocrine disruption and other hazard properties are disregarded. Indeed, FCMs are a known source of a wide range of hazardous chemicals, and they likely contribute to highly prevalent non-communicable diseases. FCMs can also include non-intentionally added substances (NIAS), which often are unknown and therefore not subject to risk assessment. To address these important shortcomings, we outline how the safety of FCMs may be improved by (1) testing the overall migrate, including (unknown) NIAS, of finished food contact articles, and (2) expanding toxicological testing beyond genotoxicity to multiple endpoints associated with non-communicable diseases relevant to human health. To identify mechanistic endpoints for testing, we group chronic health outcomes associated with chemical exposure into Six Clusters of Disease (SCOD) and we propose that finished food contact articles should be tested for their impacts on these SCOD. Research should focus on developing robust, relevant, and sensitive in-vitro assays based on mechanistic information linked to the SCOD, e.g., through Adverse Outcome Pathways (AOPs) or Key Characteristics of Toxicants. Polyethylene (PE) is the most widely used type of plastic food packaging, in which chemicals can potentially migrate into packaged foods. The implications of using and recycling PE from a chemical perspective remain underexplored. This study is a systematic evidence map of 116 studies looking at the migration of food contact chemicals (FCCs) across the lifecycle of PE food packaging. It identified a total of 377 FCCs, of which 211 were detected to migrate from PE articles into food or food simulants at least once. These 211 FCCs were checked against the inventory FCCs databases and EU regulatory lists. Only 25% of the detected FCCs are authorized by EU regulation for the manufacture of food contact materials. Furthermore, a quarter of authorized FCCs exceeded the specific migration limit (SML) at least once, while one-third (53) of non-authorised FCCs exceeded the threshold value of 10 μg/kg. Overall, evidence on FCCs migration across the PE food packaging lifecycle is incomplete, especially at the reprocessing stage. Food packaging is important for today's globalized food system, but food contact materials (FCMs) can also be a source of hazardous chemicals migrating into foodstuffs. Assessing the impacts of FCMs on human health requires a comprehensive identification of the chemicals they contain, the food contact chemicals (FCCs). We systematically compiled the "database on migrating and extractable food contact chemicals" (FCCmigex) using information from 1210 studies. We found that to date 2881 FCCs have been detected, in a total of six FCM groups (Plastics, Paper & Board, Metal, Multi-materials, Glass & Ceramic, and Other FCMs). 65% of these detected FCCs were previously not known to be used in FCMs. Conversely, of the more than 12'000 FCCs known to be used, only 1013 are included in the FCCmigex database. Plastic is the most studied FCM with 1975 FCCs detected. Our findings expand the universe of known FCCs to 14,153 chemicals. This knowledge contributes to developing non-hazardous FCMs that lead to safer food and support a circular economy.

Chemicals can migrate from polyethylene terephthalate (PET) drink bottles to their content and recycling processes may concentrate or introduce new chemicals to the PET value chain. Therefore, even though recycling PET bottles is key in reducing plastic pollution, it may raise concerns about safety and quality. This study provides a systematic evidence map of the food contact chemicals (FCCs) that migrate from PET drink bottles aiming to identify challenges in closing the plastic packaging loop. The migration potential of 193 FCCs has been investigated across the PET drink bottles lifecycle, of which 150 have been detected to migrate from PET bottles into food simulants/food samples. The study reveals that much research has focused on the migration of antimony (Sb), acetaldehyde and some well-known endocrine-disrupting chemicals (EDCs). It indicates and discusses the key influential factors on FCCs migration, such as physical characteristics and geographical origin of PET bottles, storage conditions, and reprocessing efficiency . Although, safety and quality implications arising from the recycling of PET bottles remain underexplored, the higher migration of Sb and Bishphenol A has been reported in recycled (rPET) compared to virgin PET. This is attributed to multiple contamination sources and the variability in the collection, sorting, and decontamination efficiency. Better collaboration among stakeholders across the entire PET bottles lifecycle is needed to ensure sustainable resource management and food contact safety of rPET., (Copyright © 2022 Elsevier B.V. Food packaging is of high societal value because it conserves and protects food, makes food transportable and conveys information to consumers. It is also relevant for marketing, which is of economic significance. Other types of food contact articles, such as storage containers, processing equipment and filling lines, are also important for food production and food supply. Food contact articles are made up of one or multiple different food contact materials and consist of food contact chemicals. However, food contact chemicals transfer from all types of food contact materials and articles into food and, consequently, are taken up by humans. Here we highlight topics of concern based on scientific findings showing that food contact materials and articles are a relevant exposure pathway for known hazardous substances as well as for a plethora of toxicologically uncharacterized chemicals, both intentionally and non-intentionally added. We describe areas of certainty, like the fact that chemicals migrate from food contact articles into food, and uncertainty, for example unidentified chemicals migrating into food. Current safety assessment of food contact chemicals is ineffective at protecting human health. In addition, society is striving for waste reduction with a focus on food packaging. As a result, solutions are being developed toward reuse, recycling or alternative (non-plastic) materials. However, the critical aspect of chemical safety is often ignored. Developing solutions for improving the safety of food contact chemicals and for tackling the circular economy must include current scientific knowledge. This cannot be done in isolation but must include all relevant experts and stakeholders. Therefore, we provide an overview of areas of concern and related activities that will improve the safety of food contact articles and support a circular economy. Our aim is to initiate a broader discussion involving scientists with relevant expertise but not currently working on food contact materials, and decision makers and influencers addressing single-use food packaging due to environmental concerns. Ultimately, we aim to support science-based decision making in the interest of improving public health. Notably, reducing exposure to hazardous food contact chemicals contributes to the prevention of associated chronic diseases in the human population.

Regular physical activity has multifarious benefits for physical and mental health, and music has been found to exert positive effects on physical activity. Summative literature reviews and conceptual models have hypothesized potential benefits and salient mechanisms associated with music listening in exercise and sport contexts, although no large-scale objective summary of the literature has been conducted. A multilevel meta-analysis of 139 studies was used to quantify the effects of music listening in exercise and sport domains. In total, 598 effect sizes from four categories of potential benefits (i.e., psychological responses, physiological responses, psychophysical responses, and performance outcomes) were calculated based on 3,599 participants. Music was associated with significant beneficial effects on affective valence ( g = 0.48, CI [0.39, 0.56]), physical performance ( g = 0.31, CI [0.25, 0.36]), perceived exertion ( g = 0.22, CI [0.14, 0.30]), and oxygen consumption ( g = 0.15, CI [0.02, 0.27]). No significant benefit of music was found for heart rate ( g = 0.07, CI [-0.03, 0.16]). Performance effects were moderated by study domain (exercise > sport) and music tempo (fast > slow-to-medium). Overall, results supported the use of music listening across a range of physical activities to promote more positive affective valence, enhance physical performance (i.e., ergogenic effect), reduce perceived exertion, and improve physiological efficiency. Background: Food contact articles (FCAs) are manufactured from food contact materials (FCMs) that include plastics, paper, metal, glass, and printing inks. Chemicals can migrate from FCAs into food during storage, processing, and transportation. Food contact materials' safety is evaluated using chemical risk assessment (RA). Several challenges to the RA of FCAs exist., Objectives: We review regulatory requirements for RA of FCMs in the United States and Europe, identify gaps in RA, and highlight opportunities for improving the protection of public health. We intend to initiate a discussion in the wider scientific community to enhance the safety of food contact articles., Discussion: Based on our evaluation of the evidence, we conclude that current regulations are insufficient for addressing chemical exposures from FCAs. RA currently focuses on monomers and additives used in the manufacture of products, but it does not cover all substances formed in the production processes. Several factors hamper effective RA for many FCMs, including a lack of information on chemical identity, inadequate assessment of hazardous properties, and missing exposure data. Companies make decisions about the safety of some food contact chemicals (FCCs) without review by public authorities. Some chemical migration limits cannot be enforced because analytical standards are unavailable., Conclusion: We think that exposures to hazardous substances migrating from FCAs require more attention. Background: The European Food Safety Authority recently concluded that the exposure of small children (1-3 y old) to brominated diphenyl ether (BDE)-99 may exceed acceptable levels defined in relation to neurodevelopmental toxicity in rodents. The flame retardant BDE-209 may release BDE-99 and other lower brominated BDEs through biotic and abiotic degradation, and all age groups are exposed not only to BDE-209 and -99 but also to a cocktail of BDE congeners with evidence of neurodevelopmental toxicity. The possible risks from combined exposures to these substances have not been evaluated., Objectives: We performed a congener-specific mixture risk assessment (MRA) of human exposure to combinations of BDE-209 and other BDEs based on estimated exposures via diet and dust intake and on measured levels in biologic samples., Methods: We employed the Hazard Index (HI) method by using BDE congener-specific reference doses for neurodevelopmental toxicity., Results: Our HI analysis suggests that combined exposures to polybrominated diphenyl ethers (PBDEs) may exceed acceptable levels in breastfeeding infants (0-3 mo old) and in small children (1-3 y old), even for moderate (vs. high) exposure scenarios. Our estimates also suggest that acceptable levels of combined PBDEs may be exceeded in adults whose diets are high in fish. Small children had the highest combined exposures, with some estimated body burdens that were similar to body burdens associated with developmental neurotoxicity in rodents., Conclusions: Our estimates corroborate reports from several recent epidemiological studies of associations between PBDE exposures and neurobehavioral outcomes, and they support the inclusion of BDE-209 in the persistent organic pollutant (POP) convention as well as the need for strategies to reduce exposures to PBDE mixtures, including maximum residue limits for PBDEs in food and measures for limiting the release of PBDEs from consumer waste. https://doi.org/10.1289/EHP826.
