Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

Summary: We show that the non-canonical nucleobase 2,6-diaminopurine (D) spontaneously base pairs with uracil (U) in water and the solid state without the need to be attached to the ribose-phosphate backbone. Depending on the reaction conditions, D and U assemble in thermodynamically stable hydrated and anhydrated D-U base-paired cocrystals. Under UV irradiation, an aqueous solution of D-U base-pair undergoes photochemical degradation, while a pure aqueous solution of U does not. Our simulations suggest that D may trigger the U photodimerization and show that complementary base-pairing modifies the photochemical properties of nucleobases, which might have implications for prebiotic chemistry.

A plug-flow fixed-bed cell for synchrotron powder X-ray diffraction (PXRD) and X-ray absorption fine structure (XAFS) idoneous for the study of heterogeneous catalysts at high temperature, pressure and under gas flow is designed, constructed and demonstrated. The operating conditions up to 1000°C and 50 bar are ensured by a set of mass flow controllers, pressure regulators and two infra-red lamps that constitute a robust and ultra-fast heating and cooling method. The performance of the system and cell for carbon dioxide hydrogenation reactions under specified temperatures, gas flows and pressures is demonstrated both for PXRD and XAFS at the P02.1 (PXRD) and the P64 (XAFS) beamlines of the Deutsches Elektronen-Synchrotron (DESY).

Importance It is unknown whether coronary revascularization, when added to optimal medical therapy, improves prognosis in patients with stable ischemic heart disease (SIHD) at increased risk of cardiovascular events owing to moderate or severe ischemia. Objective To describe baseline characteristics of participants enrolled and randomized in the International Study of Comparative Health Effectiveness With Medical and Invasive Approaches (ISCHEMIA) trial and to evaluate whether qualification by stress imaging or nonimaging exercise tolerance test (ETT) influenced risk profiles. Design, Setting, and Participants The ISCHEMIA trial recruited patients with SIHD with moderate or severe ischemia on stress testing. 6. Scaling carbon fluxes from eddy covariance sites to globe: synthesis and evaluation of the FLUXCOM approach

- Abstract
FLUXNET comprises globally distributed eddy-covariance-based estimates of carbon fluxes between the biosphere and the atmosphere. Since eddy covariance flux towers have a relatively small footprint and are distributed unevenly across the world, upscaling the observations is necessary to obtain global-scale estimates of biosphere–atmosphere exchange. Based on cross-consistency checks with atmospheric inversions, sun-induced fluorescence (SIF) and dynamic global vegetation models (DGVMs), here we provide a systematic assessment of the latest upscaling efforts for gross primary production (GPP) and net ecosystem exchange (NEE) of the FLUXCOM initiative, where different machine learning methods, forcing data sets and sets of predictor variables were employed. Spatial patterns of mean GPP are consistent across FLUXCOM and DGVM ensembles (R2>0.94at 1∘spatial resolution) while the majority of DGVMs show, for 70 % of the land surface, values outside the FLUXCOM range. Global mean GPP magnitudes for 2008–2010 from FLUXCOM members vary within 106 and 130 PgC yr−1with the largest uncertainty in the tropics. Seasonal variations in independent SIF estimates agree better with FLUXCOM GPP (mean global pixel-wiseR2∼0.75) than with GPP from DGVMs (mean global pixel-wiseR2∼0.6). Seasonal variations in FLUXCOM NEE show good consistency with atmospheric inversion-based net land carbon fluxes, particularly for temperate and boreal regions (R2>0.92). Interannual variability of global NEE in FLUXCOM is underestimated compared to inversions and DGVMs. The FLUXCOM version which also uses meteorological inputs shows a strong co-variation in interannual patterns with inversions (R2=0.87for 2001–2010). Mean regional NEE from FLUXCOM shows larger uptake than inversion and DGVM-based estimates, particularly in the tropics with discrepancies of up to several hundred grammes of carbon per square metre per year. These discrepancies can only partly be reconciled by carbon loss pathways that are implicit in inversions but not captured by the flux tower measurements such as carbon emissions from fires and water bodies. We hypothesize that a combination of systematic biases in the underlying eddy covariance data, in particular in tall tropical forests, and a lack of site history effects on NEE in FLUXCOM are likely responsible for the too strong tropical carbon sink estimated by FLUXCOM. Furthermore, as FLUXCOM does not account forCO2fertilization effects, carbon flux trends are not realistic. Overall, current FLUXCOM estimates of mean annual and seasonal cycles of GPP as well as seasonal NEE variations provide useful constraints of global carbon cycling, while interannual variability patterns from FLUXCOM are valuable but require cautious interpretation. Exploring the diversity of Earth observation data and of machine learning concepts along with improved quality and quantity of flux tower measurements will facilitate further improvements of the FLUXCOM approach overall.

Cubic silicon nitride (γ-Si3N4) is superhard and one of the hardest materials after diamond and cubic boron nitride (cBN), but has higher thermal stability in an oxidizing environment than diamond, making it a competitive candidate for technological applications in harsh conditions (e.g., drill head and abrasives). Here, we report the high-pressure synthesis and characterization of the structural and mechanical properties of a γ-Si3N4/Hf3N4 ceramic nanocomposite derived from single-phase amorphous silicon (Si)–hafnium (Hf)–nitrogen (N) precursor. The synthesis of the γ-Si3N4/Hf3N4 nanocomposite is performed at ~20 GPa and ca. 1500 ℃ in a large volume multi anvil press. The structural evolution of the amorphous precursor and its crystallization to γ-Si3N4/Hf3N4 nanocomposites under high pressures is assessed by the in situ synchrotron energy-dispersive X-ray diffraction (ED-XRD) measurements at ~19.5 GPa in the temperature range of ca. 1000–1900 ℃. The fracture toughness (KIC) of the two-phase nanocomposite amounts ~6/6.9 MPa·m1/2 and is about 2 times that of single-phase γ-Si3N4, while its hardness of ca. 30 GPa remains high. This work provides a reliable and feasible route for the synthesis of advanced hard and tough γ-Si3N4-based nanocomposites with excellent thermal stabililty.

Developing cost-effective and reliable solid-state sodium batteries with superior performance is crucial for stationary energy storage. A key component in facilitating their application is a solid-state electrolyte with high conductivity and stability. Herein, we employed aliovalent cation substitution to enhance ionic conductivity while preserving the crystal structure. Optimized substitution of Y3+ with Zr4+ in Na5YSi4O12 introduced Na+ ion vacancies, resulting in high bulk and total conductivities of up to 6.5 and 3.3 mS cm−1, respectively, at room temperature with the composition Na4.92Y0.92Zr0.08Si4O12 (NYZS). NYZS shows exceptional electrochemical stability (up to 10 V vs. Na+/Na), favorable interfacial compatibility with Na, and an excellent critical current density of 2.4 mA cm−2. The enhanced conductivity of Na+ ions in NYZS was elucidated using solid-state nuclear magnetic resonance techniques and theoretical simulations, revealing two migration routes facilitated by the synergistic effect of increased Na+ ion vacancies and improved chemical environment due to Zr4+ substitution. NYZS extends the list of suitable solid-state electrolytes and enables the facile synthesis of stable, low-cost Na+ ion silicate electrolytes.

It represents an original contribution to the existing body of English textbooks of public policy – a young but dynamically developing discipline. It builds on the author's many years of experience researching and teaching the discipline in Europe. In the first theoretical part, the chapters are ordered from the most general topics (definition of key terms, the field's value fundamentals, contemporary forms of governance) to more specific texts on policy actors, institutions and instruments. The second part (co-authored with Veronika Rudolfová) is devoted to case studies of the pension system reforms. These studies are well-integrated with the theoretical part, which makes them more intelligible for the reader while shedding light on the different theories'potential of application in research practice. Background With multifaceted imaging capabilities, cardiovascular magnetic resonance (CMR) is playing a progressively increasing role in the management of various cardiac conditions. A global registry that harmonizes data from international centers, with participation policies that aim to be open and inclusive of all CMR programs, can support future evidence-based growth in CMR. Methods The Global CMR Registry (GCMR) was established in 2013 under the auspices of the Society for Cardiovascular Magnetic Resonance (SCMR). The GCMR team has developed a web-based data infrastructure, data use policy and participation agreement, data-harmonizing methods, and site-training tools based on results from an international survey of CMR programs. Results At present, 17 CMR programs have established a legal agreement to participate in GCMR, amongst them 10 have contributed CMR data, totaling 62,456 studies. There is currently a predominance of CMR centers with more than 10 years of experience (65%), and the majority are located in the United States (63%). The most common clinical indications for CMR have included assessment of cardiomyopathy (21%), myocardial viability (16%), stress CMR perfusion for chest pain syndromes (16%), and evaluation of etiology of arrhythmias or planning of electrophysiological studies (15%) with assessment of cardiomyopathy representing the most rapidly growing indication in the past decade. Most CMR studies involved the use of gadolinium-based contrast media (95%). Conclusions We present the goals, mission and vision, infrastructure, preliminary results, and challenges of the GCMR. We report the experimental discovery of a new structural phase of well-known orthorhombic R2BaCuO5 (R = Sm and Eu), exhibiting a tetragonal crystal structure with space group P4∕mbm. The high-pressure tetragonal phase is isostructural with the brown phase R2BaCuO5 (R = La, Pr, and Nd). In this structure, the Cu ions form an isolated square planar environment, contrary to the orthorhombic phase, where the Cu ions are located in a distorted square pyramid. Magnetization and specific heat measurements reveal the long-range antiferromagnetic order of the Cu2+ and/or Sm3+ moments for the Sm-sample, with the magnetic specific heat accounting for only 35% of the magnetic entropy. Interestingly, the Eu-sample remains paramagnetic down to the lowest temperature. The high Curie-Weiss temperature of −140 K and magnetic entropy of 3% of the expected value indicates that the system is highly frustrated. We estimated the isothermal entropy change and investigated the magnetocaloric effect for Eu2BaCuO5, and the maximum entropy change detected at a field of 70 kOe at 3 K reaches 5.6 J kg-1K−1.

Abstract Polyanion‐type phosphate materials, such as M3V2(PO4)3 (M = Li/Na/K), are promising as insertion‐type negative electrodes for monovalent‐ion batteries including Li/Na/K‐ion batteries (lithium‐ion batteries (LIBs), sodium‐ion batteries (SIBs), and potassium‐ion batteries (PIBs)) with fast charging/discharging and distinct redox peaks. However, it remains a great challenge to understand the reaction mechanism of materials upon monovalent‐ion insertion. Here, triclinic Mg3V4(PO4)6/carbon composite (MgVP/C) with high thermal stability is synthesized via ball‐milling and carbon‐thermal reduction method and applied as a pseudocapacitive negative electrode in LIBs, SIBs, and PIBs. In operando and ex situ studies demonstrate the guest ion‐dependent reaction mechanisms of MgVP/C upon monovalent‐ion storage due to different sizes. MgVP/C undergoes an indirect conversion reaction to form Mg0, V0, and Li3PO4 in LIBs, while in SIBs/PIBs the material only experiences a solid solution with the reduction of V3+ to V2+. Moreover, in LIBs, MgVP/C delivers initial lithiation/delithiation capacities of 961/607 mAh g−1 (30/19 Li+ ions) for the first cycle, despite its low initial Coulombic efficiency, fast capacity decay for the first 200 cycles, and limited reversible insertion/deinsertion of 2 Na+/K+ ions in SIBs/PIBs. This work reveals a new pseudocapacitive material and provides an advanced understanding of polyanion phosphate negative material for monovalent‐ion batteries with guest ion‐dependent energy storage mechanisms.

Manipulations and exploration of large data sets are common problems. This paper aims at adapting a fisheye technique that uses hyperbolic geometry. Unlike the classical approach, the hyperbolic space used in our research is tiled. The tiling method proposed supplies a code for data that ensures the neighbourhood between data. The difficulty lies in finding how to distribute data through the tiling. We give some heuristics to better target suitable data sets. Our research results in defining an interactive tool for manipulating such data sets: the hyperbolic palette. To illustrate the various possibilities of this tool, a prototype application was developed. Its goal is to provide a new palette to select a colour out of 16 million options. We show in this paper why such an application fulfills our heuristics to use an hyperbolic palette.

20. Does Participation in Farmer Field School Extension Program Improve Crop Yields? Evidence from Smallholder Tea Production Systems in Kenya

Agricultural Extension services are among the most important rural services in developing countries. The services are considered to be a key driver of technological change and productivity growth in agriculture. In Kenya, like in the rest of the developing economies, agricultural extension has largely been delivered through supply–driven approaches. Due to perceived low impact of agricultural extension, the country is implementing the National Extension Policy (NEP) which advocates for demand–driven extension and participation of other players. Using the case of the smallholder tea sub-sector, this paper examines the effects the FFS extension on tea crop yields in Kenya. The FFS system uses participatory approaches including the demonstration of best sustainable practices in the farms and farmers learn by doing. Data for the study was collected from a sample of 525 farm households in Western Kenya using a multi stage random sampling procedure and analyzed using the propensity score matching (PSM) model which controls for self-selection endogeneity. The results show that participation in FFS extension increases annual tea yields by an average of 471.70 kgs per acre (p = 0.009) while the farmer–funded train and visit system has no influence on crop yields. A part from showing the contribution of FFS to crop yields, the paper demonstrates that the supply–driven extension models including T&V are necessary to stimulate demand in the initial stages of implementing the FFS models. Based on the findings, investments to enhance FFS access among smallholder farmers are recommended.

Recent work on mid-infrared (MIR) detection through the process of non-degenerate two-photon absorption (NTA) in semiconducting materials has shown that wide-field MIR imaging can be achieved with standard Si cameras. While this approach enables MIR imaging at high pixel densities, the low nonlinear absorption coefficient of Si prevents fast NTA-based imaging at lower illumination doses. Here, we overcome this limitation by using InGaAs as the photosensor. Taking advantage of the much higher nonlinear absorption coefficient of this direct bandgap semiconductor, we demonstrate high-speed MIR imaging up to 500 fps with under 1 ms exposure per frame, enabling 2D or 3D mapping without pre- or post-processing of the image.

Since its launch just over a decade ago by the cryptocurrency Bitcoin, the distributed ledger technology (DLT) blockchain has followed a breathtaking trajectory into manifold application spaces. This study aper analyses how key factors underpinning the success of this ground-breaking “Internet of value” technology, such as staking of collateral (“skin in the game”), competitive crowdsourcing, crowdfunding, and prediction markets, can be applied to substantially innovate the legacy organization of science, research, and technology development (RTD). Here, we elaborate a highly integrative, community-based strategy where a token-based crypto-economy supports finding best possible consensus, trust, and truth by adding unconventional elements known from reputation systems, betting, secondary markets, and social networking. These tokens support the holder’s formalized reputation and are used in liquid-democracy style governance and arbitration within projects or community-driven initiatives. This participatory research model serves as a solid basis for comprehensively leveraging collective intelligence by effectively incentivizing contributions from the crowd, such as intellectual property work, validation, assessment, infrastructure, education, assessment, governance, publication, and promotion of projects. On the analogy of its current blockbusters like peer-to-peer structured decentralized finance (“DeFi”), blockchain technology can seminally enhance the efficiency of science and RTD initiatives, even permitting to fully stage operations as a chiefless decentralized autonomous organization (DAOs).

We outline the concept of an open technology platform that builds upon a publicly accessible library of fluidic designs, manufacturing processes, and experimental characterization, as well as virtualization by a “digital twin” based on modeling, simulation, and cloud computing. Backed by the rapidly emerging Web3 technology “Blockchain,” we significantly extend traditional approaches to effectively incentivize broader participation by an interdisciplinary “value network” of diverse players. Ranging from skilled individuals (the “citizen scientist” and the “garage entrepreneur”) and more established research institutions to companies with their infrastructures, equipment, and services, the novel platform approach enables all stakeholders to jointly contribute to value creation along more decentralized supply chain designs including research and technology development (RTD). A blockchain-enabled token economy efficiently leverages the “Wisdom of the Crowds” and secures “trust” and transparency by reputation systems requiring “skin in the game” from contributors. Prediction markets are created for guiding decision making, planning, and allocation of funding; competitive parallelization of work and its validation from independent participants substantially enhances quality, credibility, and speed of project outcomes in the real world along the entire path from RTD, fabrication, and testing to eventual commercialization. This novel, Blockchain-backed, open platform concept can be led by a corporation, academic entity, a loosely organized group, or even “chieflessly” within a smart-contract encoded Decentralized Autonomous Organization (DAO). The proposed strategy is particularly attractive for highly interdisciplinary fields like microfluidic Lab-on-a-Chip systems in the context of manifold applications in the Life Sciences. As an exemplar, we outline the centrifugal “Lab-on-a-Disc” technology. Rather than engaging in all sub-disciplines themselves, many smaller, highly innovative actors can focus on strengthening the product component distinguishing their unique selling point (USP), e.g., a particular bioassay, detection scheme, or application scenario. In this effort, system integrators access underlying commons like fluidic design, manufacture, instrumentation, and software from a more resilient and diversified supply chain, e.g., based on a verified pool of community-endorsed or certified providers.

An overwhelming majority of experts has been flagging for decades that “Saving the Planet” requires immediate, persistent and drastic action to curb a variety of catastrophic risks over the 21st century. However, despite compelling evidence and a range of suggested solutions, transnational coordination of effective measures to protect our biosphere continues to fall short. To remedy, we propose a novel platform for addressing the central issue of affording trust, transparency and truth while minimizing administrative overheads. This will empower an even loosely organized, global grass-roots community to coordinate a large-scale project on a shared goal (“Commons”) spanning the digital and real world. The Web3 concept is based on the swiftly emerging “Blockchain” and related cryptographic, distributed and permissionless technologies. “Wisdom of the crowds” mechanisms involving competitive parallelization and prediction markets are enabled by formalized reputation and staking to incentivize high-quality work, fair validation and best management practice. While these mechanisms have been (mostly separately) applied to science, business, governance, web, sensor, information and communication technologies (ICT), our integrative approach around Blockchain-enabled ‘operating principles and protocols’ sets the basis for designing novel forms of potentially crowdfunded Decentralized Autonomous Organizations (DAOs).

Imaging is valuable in determining the presence, extent, and severity of myocardial ischemia and the severity of obstructive coronary lesions in patients with chronic chest pain in the setting of high probability of coronary artery disease. Imaging is critical for defining patients best suited for medical therapy or intervention, and findings can be used to predict long-term prognosis and the likely benefit from various therapeutic options. Chest radiography, radionuclide single photon-emission CT, radionuclide ventriculography, and conventional coronary angiography are the imaging modalities historically used in evaluating suspected chronic myocardial ischemia. Stress echocardiography, PET, cardiac MRI, and multidetector cardiac CT have all been more recently shown to be valuable in the evaluation of ischemic heart disease. Other imaging techniques may be helpful in those patients who do not present with signs classic for angina pectoris or in those patients who do not respond as expected to standard management. The ACR Appropriateness Criteria((R)) are evidence-based guidelines for specific clinical conditions that are reviewed every 2 years by a multidisciplinary expert panel. The guideline development and review include an extensive analysis of current medical literature from peer-reviewed journals and the application of a well-established consensus methodology (modified Delphi) to rate the appropriateness of imaging and treatment procedures by the panel. In those instances in which evidence is lacking or not definitive, expert opinion may be used to recommend imaging or treatment. [ABSTRACT FROM AUTHOR]

Cells receive a multitude of signals from the environment, but how they process simultaneous signaling inputs is not well understood. Response to infection, for example, involves parallel activation of multiple Toll-like receptors (TLRs) that converge on the nuclear factor κB (NF-κB) pathway. Although we increasingly understand inflammatory responses for isolated signals, it is not clear how cells process multiple signals that co-occur in physiological settings. We therefore examined a bacterial infection scenario involving co-stimulation of TLR4 and TLR2. Independent stimulation of these receptors induced distinct NF-κB dynamic profiles, although surprisingly, under co-stimulation, single cells continued to show ligand-specific dynamic responses characteristic of TLR2 or TLR4 signaling rather than a mixed response, comprising a cellular decision that we term “non-integrative” processing. Iterating modeling and microfluidic experiments revealed that non-integrative processing occurred through interaction of switch-like NF-κB activation, receptor-specific processing timescales, cell-to-cell variability, and TLR cross-tolerance mediated by multilayer negative feedback.

Na3Ni2BiO6 with Honeycomb structure suffers from poor cycle stability when applied as cathode material for sodium-ion batteries. Herein, the strategy to improve the stability is to substitute Ni and Bi with inactive Ti. Monoclinic Na3Ni2-xBi1-yTix+yO6 powders with different Ti content were successfully synthesized via sol gel method, and 0.3 mol of Ti was determined as a maximum concentration to obtain a phase-pure compound. A solid-solution in the system of O3-NaNi0.5Ti0.5O2 and O3-Na3Ni2BiO6 is obtained when this critical concentration is not exceeded. The capacity of the first desodiation process at 0.1 C of Na3Ni2BiO6 (~93 mAh g−1) decreases with the increasing Ti concentration to ~77 mAh g−1 for Na3Ni2Bi0.9Ti0.1O6 and to ~82 mAh g−1 for Na3Ni0.9Bi0.8Ti0.3O6, respectively. After 100 cycles at 1 C, a better electrochemical kinetics is obtained for the Ti-containing structures, where a fast diffusion effect of Na+-ions is more pronounced. As a result of in operando synchrotron radiation diffraction, during the first sodiation (O1-P3-O’3-O3) the O’3 phase, which is formed in the Na3Ni2BiO6 is fully or partly replaced by P’3 phase in the Ti substituted compounds. This leads to an improvement in the kinetics of the electrochemical process. The pathway through prismatic sites of Na+-ions in the P’3 phase seems to be more favourable than through octahedral sites of O’3 phase. Additionally, at high potential, a partial suppression of the reversible phase transition P3-O1-P3 is revealed.
