Abstract Peyronie’s disease (PD) has detrimental effects on the quality of life, mental health, sexual functioning and several other aspects that increase the risk of relationship problems. However, no study to date has assessed the risk of relationship separation in med with PD. Herein, we utilized data from Swedish national registers to examine the risk of relationship separation in men with PD. We conducted a matched cohort study on men born 1933–1992, followed from 1997 to 2013. PD was defined as a physician-assigned diagnosis according to the International Classification of Diseases, Tenth version. Each man with PD (n = 8020) was matched with 10 comparison men. We defined relationship separation as (1) ever separated, and (2) separation rate. We used log-linear regression to estimate the risk ratio, and rate ratio of relationship separation. We adjusted for matching variables (birth year and country of birth), and an indicator of each follow-up year. We found that men with PD had a 13% increased risk of relationship separation (risk ratio 1.13, 95% confidence interval [CI] 1.08–1.17). The rate of relationship separation events, measured on a yearly basis, was increased by 18% (rate ratio 1.18, CI 1.12–1.24), and remained similar when adjusting for follow-up year and socio-economic status.

The aim of this extended review essay is to discuss the potential relevance of degrowth-aligned social-ecological transformation for the specific context of Central and Eastern Europe (CEE). We frame this discussion around three recent books which we consider especially useful for this debate: The Future is Degrowth by Schmelzer et al. (2022, in Czech 2023) for an up-to-date and comprehensive overview of the concept of degrowth; Marx in the Anthropocene by Saito (2023) for an ecologically grounded debate on anticapitalist strategies stemming from writings of late Marx; and The Political Economy of Middle Class Politics and the Global Crisis in Eastern Europe by Gagyi (2021) that empirically analyses the specific position of the CEE semiperiphery and its implications for a radical social-ecological transformation. We introduce and interlink the main ideas of these books and discuss their implications for the degrowth movement in the CEE context. We argue that to deeply transform our socio-metabolic relation with nature, it is crucial to cultivate and expand spaces of reproductive autonomy, and link them to struggles of labour and social movements. We conclude by emphasising the role of internationalism from below.

Abstract We construct the teleparallel dynamics for extended geometry where the structure algebra is (an extension of) an untwisted affine Kac-Moody algebra. This provides a geometrisation of the Geroch symmetry appearing on dimensional reduction of a gravitational theory to two dimensions. The formalism is adapted to the underlying tensor hierarchy algebra, and will serve as a stepping stone towards the geometrisation of other infinite-dimensional, e.g. hyperbolic, symmetries.

The management of Invasive Alien Species (IAS) is often hindered by ecological, social and economic factors, resulting in inadequate biodiversity protection and inefficient use of public money. A clear example of such inefficient management in aquatic ecosystems is the European catfish Silurus glanis L. in southern Europe. Native to central Eurasia, S. glanis is an emblematic and controversial freshwater fish, being the subject of extensive and profitable trophy angling in central Europe and of commercial fishing in eastern Europe. Concurrently, in western and southern Europe where it was introduced in the XIX century, S. glanis is considered a problematic invader. The lack of comprehensive information on S. glanis invasive populations has limited effective management, which is critical to successfully control the spread and minimize negative impacts on native ecosystems and species. LIFE PREDATOR, started in September 2022 with a budget of € 2.85 million and a consortium of six partners from three countries, aims at developing a multidisciplinary and transnational approach to control established populations of S. glanis, and prevent further spreading and future introductions in southern European lakes and reservoirs. The project will develop and test an early warning system based on eDNA and citizen science and identify the most effective and selective capture techniques to reduce the abundance of catfish, particularly in Natura 2000 lakes, actively involving anglers and professional fishermen on this. Massive raising awareness campaigns will be conducted targeting anglers but also the general public, and protocols and best practices will be transferred to management authorities. For the long-term sustainability of the project, a South European Management Group will be created. Additionally, in northern Italy, where the catfish invasion is more advanced, a local circular economy will be implemented, involving the increase in fishing pressure by encouraging catfish consumption as food.

ABSTRACT Heterobasidion annosum sensu lato comprises some of the most devastating pathogens of conifers. Exploring virocontrol as a potential strategy to mitigate economic losses caused by these fungi holds promise for the future. In this study, we conducted a comprehensive screening for viruses in 98 H. annosum s.l. specimens from different regions of Czechia aiming to identify viruses inducing hypovirulence. Initial examination for dsRNA presence was followed by RNA-seq analyses using pooled RNA libraries constructed from H. annosum and Heterobasidion parviporum, with diverse bioinformatic pipelines employed for virus discovery. Our study uncovered 25 distinct ssRNA viruses, including two ourmia-like viruses, one mitovirus, one fusarivirus, one tobamo-like virus, one cogu-like virus, one bisegmented narna-like virus and one segment of another narna-like virus, and 17 ambi-like viruses, for which hairpin and hammerhead ribozymes were detected. Coinfections of up to 10 viruses were observed in six Heterobasidion isolates, whereas another six harbored a single virus. Seventy-three percent of the isolates analyzed by RNA-seq were virus-free. These findings show that the virome of Heterobasidion populations in Czechia is highly diverse and differs from that in the boreal region. We further investigated the host effects of certain identified viruses through comparisons of the mycelial growth rate and proteomic analyses and found that certain tested viruses caused growth reductions of up to 22% and significant alterations in the host proteome profile. Their intraspecific transmission rates ranged from 0% to 33%. Further studies are needed to fully understand the biocontrol potential of these viruses in planta.IMPORTANCEHeterobasidion annosum sensu lato is a major pathogen causing significant damage to conifer forests, resulting in substantial economic losses. This study is significant as it explores the potential of using viruses (virocontrol) to combat these fungal pathogens. By identifying and characterizing a diverse array of viruses in H. annosum populations from Czechia, the research opens new avenues for biocontrol strategies. The discovery of 25 distinct ssRNA viruses, some of which reduce fungal growth and alter proteome profiles, suggests that these viruses could be harnessed to mitigate the impact of Heterobasidion. Understanding the interactions between these viruses and their fungal hosts is crucial for developing effective, environmentally friendly methods to protect conifer forests and maintain ecosystem health. This study lays the groundwork for future research on the application of mycoviruses in forest disease management.

The ecological and evolutionary consequences of ploidy-driven trait variation, particularly for autopolyploids, remain poorly understood. Saxifraga oppositifolia L. (purple saxifrage), is a highly successful arctic-alpine plant species with significant genetic and phenotypic variation potentially related to autopolyploidy. Over the past decade, we have accumulated knowledge of S. oppositifolia through student projects at the University Centre in Svalbard. Building on this knowledge, we initiated the long-term “SO-field study” in 2018 including 768 permanently tagged plants screened for ploidy level, trait variation and environmental associations. Here we summarize main findings from unpublished student projects, consolidating and extending these findings with the available data from the ongoing SO-field study, containing 63% diploids, 16% triploids, and 21% tetraploids. Our results support that Svalbard is a mixed-ploidy contact zone, where ploidy is strongly associated with certain traits, including growth form, leaf mass area, flower investment, seed production and habitat. Autopolyploidy has significantly influenced both ecology and evolution of S. oppositifolia and needs to be accounted for when autopolyploids are used as a study species or evaluated for management purposes. Our findings suggest considerable re-interpretation of former research and pave the way for new inquiries and hypotheses to be tested within this framework.

Abstract Darier disease (DD) is a rare monogenetic skin disorder with limited data on its potential association with neurological disorders. This study aimed to investigate the association between DD and neurological disorders, specifically Parkinson's disease, dementias, and epilepsy. Using Swedish national registers in a period spanning between 1977 and 2013, 935 individuals with DD were compared with up to 100 comparison individuals each, randomly selected from the general population based on birth year, sex, and county of residence at the time of the first diagnosis of DD. Individuals with DD had increased risks of being diagnosed with Parkinson's disease (RR 2.1, CI 1.1; 4.4), vascular dementia (RR 2.1, CI 1.0; 4.2), and epilepsy, (RR 2.5, CI 1.8; 3.5). No association of DD with other dementias were detected. This study demonstrates a new association between DD and neurodegenerative disorders and epilepsy, underlining the need for increased awareness, interdisciplinary collaboration, and further research to understand the underlying mechanisms. Early identification and management of neurological complications in DD patients could improve treatment strategies and patient outcomes. The findings also highlight the role of SERCA2 in the pathophysiology of neurological disorders, offering new targets for future research and potentials for novel treatments.

Missed and unfinished nursing care in hospitals depends on the quality of human resources. This paper aims to analyze the influence of selected work characteristics on missing and incomplete nursing care in inpatient wards of Czech hospitals. The relationship between the rate of missed and unfinished nursing care and selected work characteristics was studied using the Czech version of the standardized questionnaire, the MISSCARE Survey. The study was conducted from September 26, 2021, to October 15, 2021. Controlled interviews with 1,205 nurses working in ward blocks in Czech hospitals were used. The research results showed statistically significant connections between selected work characteristics and missed and unfinished nursing care. Nurses with the highest level of education (Master, Ph.D.), nurses with specialized education, nurses with the lowest number of working hours (less than 30 hours per week), and nurses with the highest number of years of work experience (21 years and over) show a significantly lower rate of missed nursing care. Nurses from surgical departments, specialist nurses, and nurses with the highest education (Master, Ph.D.) report statistically significantly lower unfinished care levels. In contrast, nurses from regional/district hospitals, practical nurses/nursing assistants, and nurses with the lowest education (secondary school of nursing) report significantly higher unfinished nursing care levels. The information obtained can be used to improve nursing processes in the identified weak parts, strategic planning of nursing care, and sufficient personnel. European catfish is a large-bodied apex predator, a key species in native areas, but invasive in others where it negatively impacts local aquatic fauna necessitates catfish regulation. However, traditional ichthyological methods face challenges in capturing it. The study presents a detailed description of the efficient long-line method, refined through 48 sampling campaigns across twelve European water bodies. This method proves cost-effective and technically undemanding, requiring an average of 5.6 bait fish to catch one European catfish per day. The long-lines outperform other techniques, with the highest Biomass per unit effort (BPUE) of 6.205 kg of catfish per man-hour and minimal by-catch (0.276 kg per man-hour). In contrast, fyke nets, the second most efficient method, achieve a BPUE of 0.621 kg of catfish per man-hour with 3.953 kg of by-catch per man-hour. To optimize long-line catches, a 15 m distance between branch lines and regular relocation is recommended. Live fish is the most effective bait with no significant differences observed among species. However, earthworms, a less controversial alternative, are also efficient, especially for smaller catfish. Our recapture approach using various ichthyological methods revealed no hook avoidance behavior by catfish after a previous catch or avoidance by a certain part of the population. The long-line method is suitable for population regulation, scientific research, and conservation efforts and is the most effective means of capturing live European catfish.

Considering that food is a critical factor for a healthy population, social media analysis of discussions about this area is very important to understanding people’s behavior. This research aimed to identify the main areas of communication about healthy food, including sentiment identification, on the social network Twitter (now X). We achieved this aim by analyzing 828,963 Tweets posted from 1 April 2018 to 17 April 2023 based on a dataset that included all Tweets containing the hashtag “#healthyfood” or the phrase “healthy food.” This study extends previous research that focused only on hashtags and did not include all messages about healthy food. The results identified five main communication areas, namely (1) Inspiring, (2) Access to healthy food for children and youth, (3) Expenditure on healthy food, (4) Homemade food, and (5) Vegan/vegetarian diet, of which the second and third areas were not identified in previous research due to selective analysis focusing only on hashtags. These two communities had the highest negative sentiment of the top 10 communities, suggesting that these issues require attention.

Cilj istraživanja: Cilj ovog istraživanja jest prikazati rezultate artroskopskog zahvata za kirurško liječenje stražnjeg sindroma sraza lakta te usporediti dobivene rezultate s rezultatima objavljenima u literaturi. Ispitanici i metode: Pretraživanjem operacijskih protokola Klinike za ortopediju KBC-a Zagreb pronađeno je da je u osmogodišnjem razdoblju načinjeno deset uzastopnih artroskopija lakta zbog sindroma stražnjeg sraza lakta. Svu medicinsku dokumentaciju koja je bila dostupna u elektroničkoj i papirnatoj bazi podataka Klinike sustavno je pregledao liječnik koji nije sudjelovao u liječenju, a na pregledu provedenom u svrhu istraživanja procijenio je funkcionalni status operiranog lakta koristeći standardizirane upitnike. Duljina praćenja bolesnika prosječno je iznosila 23,89 (raspon 6 – 67) mjeseci. Rezultati: Artroskopski je zahvat u svih bolesnika protekao bez komplikacija, a one nisu zabilježene niti tijekom poslijeoperacijskog razdoblja. Značajna poboljšanja funkcije lakta zapažena su u kategorijama boli, opsega pokreta te mogućnosti izvođenja svakodnevnih aktivnosti. Medijan vrijednosti ukupnog rezultata koristeći upitnik kojeg su sastavili Timmerman i Andrews prije učinjenog zahvata iznosio je 155 bodova, a nakon zahvata 195. Medijan ostvarenog broja bodova bolesnika koristeći modificirani Andrewsov upitnik o funkciji lakta iznosio je 100 bodova. Funkcija lakta nakon zahvata ocijenjena je kao izvrsna u 88,89% bolesnika, a svi su se bolesnici uspješno vratili sportskim aktivnostima kojima su se bavili prije zahvata. Zaključak: Rezultati ovog istraživanja, kao i spoznaje iz literature, pokazuju da se stražnji sindrom sraza lakta može uspješno i sigurno liječiti artroskopskim zahvatom. Tim će se zahvatom u većini slučajeva značajno smanjiti bol, dobit će se bolji opseg pokreta u laktu te će se omogućiti ponovno bavljenje sportom i to će sve biti praćeno malim brojem komplikacija, kao i rijetkom potrebom za reoperacijom.

This paper introduces a newly designed reactive power control method for single-phase photovoltaic (PV) inverters. The control focuses on easy application and autonomous actions. The regulation is designed with regard to the effective network operation and the saving of reactive power with the functionality of voltage control and optimization of active losses. This paper proves the easy application of the newly proposed control design using the implementation of a control algorithm into a dynamic photovoltaic power plant (PVP) model and supply point with PSCAD software. The long-term benefit of this control is illustrated by the MATLAB/Simulink case study of a one-week simulation over the CIGRE LV European benchmark four-wire network using data from smart meters and measured PVP generation, which was provided by the Distribution System Operator (DSO). The case study showed the benefits of the newly designed regulation in terms of saving reactive energy supply from the upstream system by up to 64% (the regulation contributes to the self-sufficiency of the LV network in terms of reactive power), a positive effect on the reduction of active losses by up to 1.5% and a simultaneous improvement of the voltage profile at the nodes of the modelled network is also observed. The case study also reveals the risks of autonomous control of $Q = f(V)$ single phase inverter on the voltage unbalance depending on its definition.

Abstract Background Reactive arthritis and septic arthritis rarely present concomitantly in the same joint and patient. Reactive arthritis presenting after coronavirus disease 2019 is also exceedingly rare, with less than 30 cases reported thus far. Less common pathogens such as Clostridium difficile have been reported to cause reactive arthritis, especially in patients with a positive human leukocyte antigen B27, and therefore should be considered in diagnostic algorithms. The aim of this case report is to highlight the difficulties and precautions in discerning and diagnosing patients presenting with sudden swelling of the knee. Case presentation We report the case of a 70-year-old Caucasian male with a recent history of coronavirus disease 2019 upper respiratory infection and diarrhea and negating trauma, who presented with a swollen and painful knee. Pain and swelling worsened and inflammatory parameters increased after an intraarticular corticosteroid injection. The patient was therefore treated with arthroscopic lavage and intravenous antibiotics for suspected septic arthritis. Synovial fluid and synovium samples were taken and sent for microbiological analysis. Synovial fluid cytology showed increased leukocytes at 10,980 × 106/L, while polymerase chain reaction and cultures came back sterile. Clostridium difficile toxin was later detected from a stool sample and the patient was treated with oral vancomycin. The patient was tested for the presence of human leukocyte antigen B27, which was positive. We present a review of the literature about the challenges of distinguishing septic from reactive arthritis, and about the mechanisms that predispose certain patients to this rheumatological disease. Conclusions It is still a challenge to differentiate between septic and reactive arthritis of the knee, and it is even more challenging to identify the exact cause of reactive arthritis. This case report of a human leukocyte antigen-B27-positive patient highlights the necessity of contemplating different, less common causes of a swollen knee joint as a differential diagnosis of an apparent septic infection, especially in the coronavirus disease 2019 era. Treating the patient for septic arthritis prevented any possible complications of such a condition, while treating the C. difficile infection contributed to the substantial relief of symptoms.

Aim: The period after the First World War was very challenging for countries across the whole of Europe. The health condition and health literacy of inhabitants in the First Czechoslovak Republic were poor. Design: Historical research. Methods: Direct, indirect, and progressive methods were chosen to describe community care and public health care. Archival sources were found in selected archives in the Czech and Slovak republics. Results: It is evident that many changes related to preventing infectious and non-infectious diseases. Ethnic and socioeconomic diversity was a factor that influenced the health condition and knowledge of the population, and the success of preventive measures. Therefore, the involvement of professional and voluntary organizations, and government institutions was essential. Public health care arose from the idea that a healthy population was the basis of the newly-formed republic. Conclusion: The literature and archival sources discovered show the importance of the health condition of the population to the newly-formed Czechoslovak state. To improve the health condition of the population, interventions were realized in cooperation with various organizations. Concurrently, there was an appreciable effort to enhance the health literacy of the population.

Objectives: Elderly patients with hematologic malignancies face the highest risk of severe COVID-19 outcomes. The infection's impact on different age groups remains unstudied in detail. Methods: We analyzed elderly patients (age groups: 65-70, 71-75, 76-80, and >80 years old) with hematologic malignancies included in the EPICOVIDEHA registry between January 2020 and July 2022. Univariable and multivariable Cox regression models were conducted to identify factors influencing death in COVID-19 patients with hematological malignancy. Results: The study included data from 3,603 elderly patients (aged 65 or older) with hematological malignancy, with a majority being male (58.1%) and a significant proportion having comorbidities. The patients were divided into four age groups, and the analysis assessed COVID-19 outcomes, vaccination status, and other variables in relation to age and pandemic waves. The 90-day survival rate for patients with COVID-19 was 71.2%, with significant differences between groups. The pandemic waves had varying impacts, with the first wave affecting patients over 80 years old, the second being more severe in 65-70, and the third being the least severe in all age groups. Factors contributing to 90-day mortality included age, comorbidities, lymphopenia, active malignancy, acute leukemia, less than three vaccine doses, severe COVID-19, and using only corticosteroids as treatment. Conclusion: These data underscore the heterogeneity of elderly hematological patients, highlight the different impacts of COVID-19 waves and the pivotal importance of vaccination, and may help in planning future healthcare efforts.

This paper outlines the purposeful adaptation and utilization of the design thinking process in an innovation case involving market segmentation in social network research. Based on a case study, this paper combines the design thinking process with systems approach methods to foster innovation in social network analyses. The paper details the entire process, from the initial stages to the development of a viable solution defined in the final assignment for programmers. The case study emphasizes the effective use of systems thinking tools and demonstrates the value of combining these two approaches to meet the needs of the innovation process. The paper aims to narrate the entire process and highlight critical points in a real-world case study. The focus was on the challenge of creating a market segmentation tool for researchers and marketers in the realm of social network analysis.

Summary: Background: The COVID-19 pandemic heightened risks for individuals with hematological malignancies due to compromised immune systems, leading to more severe outcomes and increased mortality. While interventions like vaccines, targeted antivirals, and monoclonal antibodies have been effective for the general population, their benefits for these patients may not be as pronounced. Methods: The EPICOVIDEHA registry (National Clinical Trials Identifier, NCT04733729) gathers COVID-19 data from hematological malignancy patients since the pandemic's start worldwide. It spans various global locations, allowing comprehensive analysis over the first three years (2020–2022). Findings: The EPICOVIDEHA registry collected data from January 2020 to December 2022, involving 8767 COVID-19 cases in hematological malignancy patients from 152 centers across 41 countries, with 42% being female. Over this period, there was a significant reduction in critical infections and an overall decrease in mortality from 29% to 4%. However, hospitalization, particularly in the ICU, remained associated with higher mortality rates. Factors contributing to increased mortality included age, multiple comorbidities, active malignancy at COVID-19 onset, pulmonary symptoms, and hospitalization. On the positive side, vaccination with one to two doses or three or more doses, as well as encountering COVID-19 in 2022, were associated with improved survival. Interpretation: Patients with hematological malignancies still face elevated risks, despite reductions in critical infections and overall mortality rates over time. Hospitalization, especially in ICUs, remains a significant concern. The study underscores the importance of vaccination and the timing of COVID-19 exposure in 2022 for enhanced survival in this patient group. Ongoing monitoring and targeted interventions are essential to support this vulnerable population, emphasizing the critical role of timely diagnosis and prompt treatment in preventing severe COVID-19 cases. Abstract Background There is a conceptual overlap between negative and depressive symptoms, requiring further exploration to advance the understanding of negative symptoms. The aim of this study was to examine psychometric properties of the Clinical Assessment Interview for Negative Symptoms (CAINS) in patients with depression, and to explore the relationship between the negative and affective symptoms domains. Methods Fifty-one patients with a depressive episode were included and interviewed with the CAINS and the Brief Psychiatric Rating Scale—Expanded (BPRS-E). Self-reported depressive symptoms were collected with the Montgomery-Asberg Depression Rating Scale (MADRS-S). Inter-rater agreement, internal consistency and validity measures were examined, as were correlations between negative and affective symptoms. Results The intraclass correlation for the CAINS motivation and pleasure subscale (CAINS-MAP) was 0.98 (95% CI 0.96–0.99) and that for the expressional subscale (CAINS-EXP) was 0.81 (95% CI 0.67–0.89). Cronbach’s alpha was 0.71 (95% CI 0.57–0.82) for the CAINS-MAP and 0.86 (95% CI 0.79–0.92) for the CAINS-EXP. Background. Infectious complications during induction chemotherapy of acute myeloid leukaemia are very common. Prophylactic use of antibiotics however is an ongoing challenge in this situation due to bacterial multi-drug resistance. The aim of this study was to provide a comprehensive overview of the incidence of infectious complications in patients with AML undergoing induction therapy using the "7+3" protocol without routine antibiotic prophylaxis at one clinical site providing specialised haematological care in the Czech Republic, over a period of 15 years. The study also evaluates the aetiological spectrum of causative agents and the development of antibiotic resistance in the context of the use of the various classes of antibiotics. The analysis includes evaluation of the importance of risk factors for infectious complications and their impact on treatment of the underlying disease. The data are compared with published figures for similar cohorts of patients. Patients and Methods. This study presents a retrospective analysis of infectious complications in 242 patients with acute myeloid leukaemia undergoing the first cycle of induction therapy without routine antibiotic prophylaxis in one clinical site in Czech Republic during years 2006-2020. Results. A total of 363 febrile episodes (FE) were recorded. At least 1 FE during the induction was detected in 229 (94.6%) patients. Clinically defined infection was the cause in 96 (26.4%) FEs and blood stream infection in 69 (19.0%) FEs. Both complications occurred simultaneously in 29 (8.0%) FEs. 169 (46.6%) FEs were evaluated as fever of unknown origin (FUO). The achievement of complete remission had a significant effect on the duration of the FE (6 vs. 9 days, P=0.0005) and on the overall survival duration (79.3 vs. 6.5 months, P

Abstract Bacterial growth rate (µ) depends on the protein synthesis capacity of the cell and thus on the number of active ribosomes and their translation elongation rate. The relationship between these fundamental growth parameters have only been described for few bacterial species, in particular Escherichia coli. Here, we analyse the growth-rate dependency of ribosome abundance and translation elongation rate for Corynebacterium glutamicum, a gram-positive model species differing from E. coli by a lower growth temperature optimum and a lower maximal growth rate. Unlike metals, which have constant fatigue parameters, polymers can exhibit frequency-dependent fatigue parameters, significantly affecting the vibration resilience of 3D-printed polymer structures. This manuscript presents a study utilizing a novel vibration-fatigue testing methodology to characterize the frequency dependence of polymer material fatigue parameters under constant temperature conditions. In this investigation, 3D-printed PLA samples with frequency-tunable geometry were experimentally tested on an electro-dynamical shaker with a random vibration profile. Using the validated numerical models, the estimation of vibration-fatigue life was obtained and compared to the experimental results. Performing the numerical minimization of estimated and actual fatigue lives, the frequency-dependent fatigue parameters were assessed. In particular, the results indicate that the tested samples exhibit varying fatigue parameters within the loading frequency range of 250–750 Hz. Specifically, as the loading frequency increases, the fatigue exponent increases and fatigue strength decreases. These findings confirm the frequency dependence of fatigue parameters for 3D-printed polymer structures, underscoring the necessity of experimental characterization to reliably estimate the vibration-fatigue life of 3D-printed polymer structures. The utilization of the introduced approach therefore enhances the vibration resilience of the 3D-printed polymer mechanical component.

Xylem sap proteomics provides crucial insights into plant defense and root-to-shoot communication. This study highlights the sensitivity and reproducibility of xylem sap proteome analyses, using a single plant per sample to track over 3000 proteins in two model crop plants, Solanum tuberosum and Hordeum vulgare. By analyzing the flg22 response, we identified immune response components not detectable through root or shoot analyses. Notably, we discovered previously unknown elements of the plant immune system, including calcium/calmodulin-dependent kinases and G-type lectin receptor kinases. Despite similarities in the metabolic pathways identified in the xylem sap of both plants, the flg22 response differed significantly: S. tuberosum exhibited 78 differentially abundant proteins, whereas H. vulgare had over 450. However, an evolutionarily conserved overlap in the flg22 response proteins was evident, particularly in the CAZymes and lipid metabolism pathways, where lipid transfer proteins and lipases showed a similar response to flg22. Additionally, many proteins without conserved signal sequences for extracellular targeting were found, such as members of the HSP70 family. Interestingly, the HSP70 response to flg22 was specific to the xylem sap proteome, suggesting a unique regulatory role in the extracellular space similar to that reported in mammalians.

Fe78B15Sn7 and (Fe3Co1)78B15Sn7 amorphous metallic alloys were prepared using the method of planar flow casting. The amorphous nature of ribbons containing 7 at. % Sn was verified by X-ray diffraction. The resulting chemical composition was checked by flame atomic absorption spectroscopy and by mass spectrometry with inductively coupled plasma. The microstructure of the as-quenched metallic glasses was investigated by 57-Fe and 119-Sn Mössbauer spectrometry. The experiments were performed with transmission geometry at 300 K, 100 K, and 4.2 K, and in an external magnetic field of 6 T. The replacement of a quarter of the Fe by Co did not cause significant modifications of the hyperfine interactions in the 57-Fe nuclei. The observed minor variations in the local magnetic microstructure were attributed to alterations in the topological short-range order. However, the in-field 57-Fe Mössbauer spectra indicated a misalignment of the partial magnetic moments. On the other hand, the presence of Co considerably affected the local magnetic microstructure of the 119-Sn nuclei. This was probably due to the higher magnetic moment of Co, which induces transfer fields and polarization effects on the diamagnetic Sn atoms.

BackgroundHailey-Hailey disease (HHD) is a rare autosomal dominant skin disease caused by mutations in the ATP2C1 gene, which encodes the secretory Ca2+/Mn2+-ATPase (SPCA1) pump in the Golgi apparatus. Although ATP2C1 is ubiquitously expressed in the body, possible extracutaneous manifestations of HHD are unknown. However, dysfunction of the Golgi apparatus not specifically coupled to ATP2C1 has been associated with heart disease.ObjectiveTo investigate the association between HHD and common heart disease in a Swedish, population-based cohort.MethodsWe conducted a population-based cohort study based on a linkage of Swedish nationwide registers to investigate the relationship between HHD and heart disease. We have been granted ethical approval from the Swedish Ethical Review Authority to conduct this study. The patients in this manuscript have given written informed consent to the publication of their case details. A total of 342 individuals with an ICD-10 diagnosis of HHD (Q82.8E) were identified and matched with randomly selected comparison individuals without HHD on a 1:100 ratio. Furthermore, in a separate clinical cohort we matched 23 HHD patients for age, sex, and BMI with control subjects to examine electrocardiogram parameters, electrolytes, and cardiovascular biomarkers.ResultsCompared with individuals without HHD, individuals with HHD had an excess risk of arrhythmia (RR 1.4, CI 1.0-2.0), whereas no increased risks of myocardial infarction (RR 1.1, CI 0.6-1.7) or heart failure (RR 1.0, CI 0.6-1.6; Table 1) were found. We found no difference in ECG parameters, cardiovascular biomarkers, and electrolytes in the clinical subset.ConclusionThis study reveals that HHD is associated with an increased risk of arrhythmia and represents the first data of any extracutaneous comorbidity in HHD. Thus, HHD may be a systemic disease. Our findings also shed light on the importance of the Golgi apparatus' Ca2+/Mn2+ homeostasis in common heart disease.

ABSTRACT In this study, we investigated the impact of zinc oxide (ZnO) and zinc oxide nanoparticles (ZnONPs) on Escherichia coli. These compounds are commonly used as fertilizers and feed additives and may have unintended consequences on bacteria. We conducted phenotypic and multi-omics analyses of E. coli exposed to sub-lethal concentrations of ZnO and ZnONPs for 40 sub-culturings and 20 sub-culturings with zinc followed by 20 sub-culturings without zinc in an attempt to reverse zinc effects. An extended treatment with ZnO for 40 sub-culturings had the greatest impact leading to bacterial resistance to aminoglycosides, cephalosporins, and sulfonamides while the treatment with ZnONPs for 40 sub-culturings led to the elevated MIC to chloramphenicol only. Cells exposed to these treatments were thicker and had retarded growth in elevated temperatures. Importantly, zinc withdrawal reversed most phenotypic changes. Zinc exposure caused dramatic changes in cell transcripts and proteins with a role in antibiotic response, heat stress, growth regulation, cell shape, and biofilm formation. An extended zinc exposure led to overall major alterations in E. coli biology and resulted in multi-drug resistance.IMPORTANCEA long-term exposure of bacteria to zinc oxide and zinc oxide nanoparticles leads to major alterations in bacterial morphology and physiology. These included biochemical and physiological processes promoting the emergence of strains with multi-drug resistance and virulence traits. After the removal of zinc pressure, bacterial phenotype reversed back to the original state; however, certain changes at the genomic, transcriptomic, and proteomic level remained. Why is this important? The extensive and intensive use of supplements in animal feed effects the intestinal microbiota of livestock and this may negatively impact the health of animals and people. Therefore, it is crucial to understand and monitor the impact of feed supplements on intestinal microorganisms in order to adequately assess and prevent potential health risks.

Background Medication administration errors (MAE) are a worldwide issue affecting the safety of hospitalized patients. Through the early identification of potential causes, it is possible to increase the safety of medication administration (MA) in clinical nursing. The study aimed to identify potential risk factors affecting drug administration in inpatient wards in the Czech Republic. Material and Methods A descriptive correlation study through a non-standardized questionnaire was used. Data were collected from September 29 to October 15, 2021, from nurses in the Czech Republic. For statistical analysis, the authors used SPSS vers. 28 (IBM Corp., Armonk, NY, USA). Results The research sample consisted of 1205 nurses. The authors found that there was a statistically significant relationship between nurse education (p = 0.05), interruptions, preparation of medicines outside the patient rooms (p < 0.001), inadequate patient identification (p < 0.01), large numbers of patients assigned per nurse (p < 0.001), use of team nursing care and administration of generic substitution and an MAE. Conclusions The results of the study point to the weaknesses of medication administration in selected clinical departments in hospitals. The authors found that several factors, such as high patient ratio per nurse, lack of patient identification, and interruption during medication preparation of nurses, can increase the prevalence of MAE. Nurses who have completed MSc and PhD education have a lower incidence of MAE. More research is needed to identify other causes of medication administration errors. Improving the safety culture is the most critical challenge for today’s healthcare industry. Education for nurses can be an effective way to reduce MAEs by enhancing their knowledge and skills, mainly focusing on increasing adherence to safe medication preparation and administration and a better understanding of medication pharmacodynamics. Abstract We provide, through the framework of extended geometry, a geometrisation of the duality symmetries appearing in magical supergravities. A new ingredient is the general formulation of extended geometry with structure group of non-split real form. A simple diagrammatic rule for solving the section constraint by inspection of the Satake diagram is derived.

Abstract We formalise the teleparallel version of extended geometry (including gravity) by the introduction of a complex, the differential of which provides the linearised dynamics. The main point is the natural replacement of the two-derivative equations of motion by a differential which only contains terms of order 0 and 1 in derivatives. Second derivatives arise from homotopy transfer (elimination of fields with algebraic equations of motion). The formalism has the advantage of providing a clear consistency relation for the algebraic part of the differential, the “dualisation”, which then defines the dynamics of physical fields. It remains unmodified in the interacting BV theory, and the full non-linear models arise from covariantisation. A consequence of the use of the complex is that symmetry under local rotations becomes as good as manifest, instead of arising for a specific combination of tensorial terms, for less obvious reasons. We illustrate with a derivation of teleparallel Ehlers geometry, where the extended coordinate module is the adjoint module of a finite-dimensional simple Lie group.

This study explores the pyrolysis process applied to various non-utilized waste materials, specifically focusing on separated plastics from municipal waste, wood waste (including pallets and window frames), paper rejects, and automotive carpets. Different combinations of these waste materials were subjected to pyrolysis, a process involving high-temperature treatment (600 °C) in a nitrogen atmosphere. The resulting products, including biochar, gas, and liquid fractions, as well as the residual waste materials, underwent comprehensive analysis. The evaluation of pyrolysis products emphasizes their quality, energy content, and potential applications. Notably, the pyrolysis gas derived from the combination of separated municipal plastics and waste wood exhibited the highest calorific value at 49.45 MJ/m3. Additionally, Mixture 2, consisting of plastic and wood waste, demonstrated the highest calorific value for the pyrolysis condensate, reaching 30.62 MJ/kg. Moreover, Mixture 3, benefiting from biochar utilization as a sorbent, displayed the highest iodine value at 90.01 mg/g.

Studie analyzuje dva koncepty přístup k pravdě tak, jak je konstituje Heidegger – jako αληθεια a veritas. Jejich analýzu promítá do procesu definování ve vědách o výchově a vzdělávání. Ukazuje, že představa definice, jako přesného a trvalého vymezení určitého fenoménu či kategorie může být praktickou aktivitou, ale je jen málo vědecká. Na příkladu postupné proměny sebe definování konceptu informační gramotnosti studie dochází k závěru, že definice není trvalým vymezením, ale spíše praktickým komunikačním předporozuměním, které můžeme klást před sebe jako podnět k hlubší analýze. Definice mají (stejně jako pravda) rozměr dynamicky se měnící struktury reagující na prostředí, do nějž jsou zasazovány. Tento aspekt je nutné zásadním způsobem zohledňovat při budování konceptů filosofie výchovy i pedagogiky.

Hailey-Hailey disease is a rare hereditary skin disease caused by mutations in the ATP2C1 gene encoding the secretory pathway Ca2+/Mn2+-ATPase 1 (SPCA1) protein. Extracutaneous manifestations of Hailey-Hailey disease are plausible but still largely unknown. The aim of this study was to explore the association between Hailey-Hailey disease and diabetes. A population-based cohort study of 347 individuals with Hailey-Hailey disease was performed to assess the risks of type 1 diabetes and type 2 diabetes, using Swedish nationwide registries. Pedigrees from 2 Swedish families with Hailey-Hailey disease were also investigated: 1 with concurrent type 1 diabetes and HLA-DQ3, the other with type 2 diabetes. Lastly, a clinical cohort with 23 individuals with Hailey-Hailey disease and matched healthy controls was evaluated regarding diabetes. In the register data males with Hailey-Hailey disease had a 70% elevated risk of type 2 diabetes, whereas no excess risk among women could be confirmed. In both pedigrees an unusually high inheritance for diabetes was observed. In the clinical cohort, individuals with Hailey-Hailey disease displayed a metabolic phenotype indicative of type 2 diabetes. Hailey-Hailey disease seems to act as a synergistic risk factor for diabetes. This study indicates, for the first time, an association between Hailey-Hailey disease and diabetes and represents human evidence that SPCA1 and the Golgi apparatus may be implicated in diabetes pathophysiology.

Fish are an important component of aquatic ecosystems, thus representative and reliable assessments of their population variables are essential for a variety of ecological applications, management and conservation. Determining Fish Density per actual Spatial Unit (volume or area, FDSU) as a measure of absolute fish quantity is of particular interest. Gillnets are undoubtedly one of the most common and important methods for assessing fish populations in large lentic waters. However, direct calculating of FDSU from gillnet catches is impossible because of the passive nature of this method, and to date there is no reliable model for calculating FDSU from gillnet catches. This weakness largely limits the use of gillnet data for applications requiring FDSU estimates.The aim of this study was to calibrate gillnet catches using FDSU obtained by active methods (beach seine nets and hydroacoustics) to develop a tool for assessing FDSU from gillnet catches. To achieve this goal, we compared gillnet biomass to fish biomass estimated from the active methods, both of which cover similar spatiotemporal niches. This comparison was performed using a statistical approach based on the recognition of non-negligible random measurement error in both the explanatory (active methods) and response (gillnets) variables.We found a strong positive linear relationship between fish biomasses sampled with gillnets and with active methods. The slope of the fitted linear model was similar when comparing gillnets with the two active methods. The statistical method used allowed for the inclusion of error in the biomass estimates with gillnets and active methods, refining the credible intervals of the estimated relationship. The effect of gillnet effort on model accuracy was simulated to show how increased effort narrows the credible interval. Finally, comparison with previously published relationships revealed a large but explainable discrepancy between our model and previous models.Our study showed that conversion of gillnet biomass to biomass per actual spatial unit is possible. The effective sampling area of one square meter of gillnet was determined to be 8 m2 of waterbody surface area when European standard 12 mesh-sizes gillnets are used, and 5 m2 when four larger meshes are added to the European standard gillnets. Our model further stressed the impact of increased sampling effort on reducing estimation variability and shows that the model may be dependent on the fish community.

Aim: Increased cultural diversity due to global migration and on-going associated healthcare disparities has highlighted the importance of culturally competent care. We sought to summarize existing knowledge and identify gaps in research linked to the cultural challenges encountered by nurses when caring for patients from different cultures. Design: A scoping review. Methods: Informed by PRISMA-ScR guidelines, we searched PubMed, ScienceDirect, EBSCO Host, Web of Science, and Google Scholar using relevant MeSH keywords for articles published from 2010-2020. We then used a 3-step data screening and extraction process to manage retrieved articles. Results: Initially 80 studies were identified, six of which met inclusion criteria. Thematic analysis yielded five themes: lack of cultural knowledge, language barriers, micro-racism, lack of time to attend training and provide culturally competent care, and recommendations to improve care delivery. Conclusion: Nurses should be proactive and advocate for the necessary education and training to provide culturally competent care so critical to providing high-quality healthcare. More research, interventional and effectiveness in particular, is needed to address common challenges such as micro-racism and to facilitate the World Health Organization and National Academy of Medicine recommendations directed at providing culturally appropriate care to promote health equity.

Abstract Background The knowledge of how the separate Attention-Deficit/Hyperactivity Disorder (ADHD) subdimensions (impulsivity, hyperactivity, and inattention) are associated with nonsuicidal self-injury (NSSI) and suicidal behavior (SB) is limited. The objective of this study was to investigate the associations of childhood ADHD subdimensions with NSSI and SB in children at risk of neurodevelopmental disorders (NDDs; including ADHD). Methods The sample (N = 391) included twin pairs where at least one twin screened positive for at least one NDD or common comorbidity at age 9 or 12. Data on ADHD subdimensions was collected through a telephone interview with a caregiver/legal guardian at age 9 or 12, and data on NSSI and SB was collected through an in-person clinical assessment at age 15. The associations between the ADHD subdimensions and NSSI or SB were tested in three different models: (1) univariable, (2) together with the other ADHD subdimensions, and (3) in a confounder-adjusted model including other NDD symptoms in addition to ADHD subdimensions, for NSSI and SB separately. Results A total of 32 (8.2%) adolescents reported life-time engagement of NSSI, and 18 (4.6%) SB. Childhood impulsivity was associated with SB and childhood inattention with NSSI, in all models. Hyperactivity was not meaningfully associated with any of the outcomes. Conclusion Impulsivity and inattention, but not hyperactivity, may be of particular importance in understanding SB and NSSI. Brief screening for impulsivity and inattention in childhood could facilitate detection of children vulnerable to NSSI and SB and indicate valuable information for preventive and intervention strategies.

Solar parks are currently considered a new source of energy generation and one of the methods for reducing the usage of fossil fuels. The studies related to the influence of solar parks on vegetation structure are not yet sufficient. The aim of this paper was to evaluate the structure of vegetation biodiversity within a solar park. The vegetation assessment took place in the solar park, located in the cadastral territory of Tišnov (Czech Republic). A total of 85 taxa of vascular plants were found. The vegetation of the solar park is species-rich and significantly fragmented. Perennial grasses and perennial dicots dominate between the panels, whereas annual grasses and annual dicots have more coverage under the photovoltaic (PV) panels. The influence of the solar park microclimate is seen in a higher representation of species with the indication of characteristics mid-continental, and species with the indication of characteristics mid-ocean are more represented among within the PV panels. The vegetation of the solar park is noteworthy from the perspective of biological importance in the agricultural landscape and has a substantial potential to perform ecosystem functions.

In the field of energy networks, for their effective functioning, it is necessary to distribute the required load between all online generating units in a proper way to cover the demand. The load schedule is obtained by solving the so-called Economic Dispatch Problem (EDP). The EDP can be solved in many ways, resulting in a power distribution plan between online generating units in the network so that the resulting price per unit of energy is minimal. This article focuses on designing a distributed gradient algorithm for solving EDP, supplemented by models of renewable sources, Battery Energy Storage System (BESS), variable fuel prices, and consideration of multiple uncertainties at once. Specifically, these are: time-variable transport delays, noisy gradient calculation, line losses, and drop-off packet representations. The algorithm can thus be denoted as robust, which can work even in unfavorable conditions commonly found in real applications. The capabilities of the presented algorithm will be demonstrated and evaluated on six examples.

BackgroundArtificial intelligence (AI) has advanced substantially in recent years, transforming many industries and improving the way people live and work. In scientific research, AI can enhance the quality and efficiency of data analysis and publication. However, AI has also opened up the possibility of generating high-quality fraudulent papers that are difficult to detect, raising important questions about the integrity of scientific research and the trustworthiness of published papers. ObjectiveThe aim of this study was to investigate the capabilities of current AI language models in generating high-quality fraudulent medical articles. We hypothesized that modern AI models can create highly convincing fraudulent papers that can easily deceive readers and even experienced researchers. MethodsThis proof-of-concept study used ChatGPT (Chat Generative Pre-trained Transformer) powered by the GPT-3 (Generative Pre-trained Transformer 3) language model to generate a fraudulent scientific article related to neurosurgery. GPT-3 is a large language model developed by OpenAI that uses deep learning algorithms to generate human-like text in response to prompts given by users. The model was trained on a massive corpus of text from the internet and is capable of generating high-quality text in a variety of languages and on various topics. The authors posed questions and prompts to the model and refined them iteratively as the model generated the responses. The goal was to create a completely fabricated article including the abstract, introduction, material and methods, discussion, references, charts, etc. Once the article was generated, it was reviewed for accuracy and coherence by experts in the fields of neurosurgery, psychiatry, and statistics and compared to existing similar articles. ResultsThe study found that the AI language model can create a highly convincing fraudulent article that resembled a genuine scientific paper in terms of word usage, sentence structure, and overall composition. The AI-generated article included standard sections such as introduction, material and methods, results, and discussion, as well a data sheet. It consisted of 1992 words and 17 citations, and the whole process of article creation took approximately 1 hour without any special training of the human user. However, there were some concerns and specific mistakes identified in the generated article, specifically in the references. ConclusionsThe study demonstrates the potential of current AI language models to generate completely fabricated scientific articles. Although the papers look sophisticated and seemingly flawless, expert readers may identify semantic inaccuracies and errors upon closer inspection. We highlight the need for increased vigilance and better detection methods to combat the potential misuse of AI in scientific research. At the same time, it is important to recognize the potential benefits of using AI language models in genuine scientific writing and research, such as manuscript preparation and language editing.

Apps that target posttraumatic stress are rarely evaluated and long-term examination of symptom change is rare. In a waitlist-controlled randomized controlled trial, we found that the Swedish version of the self-management app PTSD Coach confers benefits on posttraumatic stress and depressive symptoms after three months use. Here, we aimed to evaluate between-group effects on functional disability as well as within-group changes on mental health, somatic illness and functional disability after access to the Swedish PTSD Coach app during 9 months. In addition, we described negative effects, helpfulness and satisfaction with the app. Among the 179 trauma-exposed adults (92 % women) randomized to instant access or delayed access to PTSD Coach, symptoms of posttraumatic stress, depression, somatic illness and functional disability decreased and were maintained within 3 to 9 months of app access. Posttraumatic stress continued to improve during follow-up. PTSD Coach was considered slightly to moderately helpful and satisfactory and 43 % reported any negative effect related to using the app. PTSD Coach is an effective self-management intervention for trauma-related distress. Future research should investigate mechanisms of change, as well as individual characteristics that predict symptom reduction after access to PTSD Coach in order to inform clinical practice.

Background: Patients with Philadelphia-negative chronic myeloproliferative neoplasms (MPN) typically incur high rates of infections and both drugs and comorbidities may modulate infection risk. Objectives: The present study aims to assess the effect of immunosuppressive agents on clinical outcomes of MPN patients affected by the coronavirus disease 2019 (COVID-19). Design: This is an observational study. Methods: We specifically searched and analyzed MPN patients collected by EPICOVIDEHA online registry, which includes individuals with hematological malignancies diagnosed with COVID-19 since February 2020. Results: Overall, 398 patients with MPN were observed for a median of 76 days [interquartile range (IQR): 19–197] after detection of SARS-CoV2 infection. Median age was 69 years (IQR: 58–77) and 183 individuals (46%) had myelofibrosis (MF). Overall, 121 patients (30%) of the whole cohort received immunosuppressive therapies including steroids, immunomodulatory drugs, or JAK inhibitors. Hospitalization and consecutive admission to intensive care unit was required in 216 (54%) and 53 patients (13%), respectively. Risk factors for hospital admission were identified by multivariable logistic regression and include exposure to immunosuppressive therapies [odds ratio (OR): 2.186; 95% confidence interval (CI): 1.357–3.519], age ⩾70 years, and comorbidities. The fatality rate was 22% overall and the risk of death was independently increased by age ⩾70 years [hazard ratio (HR): 2.191; 95% CI: 1.363–3.521], previous comorbidities, and exposure to immunosuppressive therapies before the infection (HR: 2.143; 95% CI: 1.363–3.521). Conclusion: COVID-19 infection led to a particularly dismal outcome in MPN patients receiving immunosuppressive agents or reporting multiple comorbidities. Therefore, specific preventive strategies need to be tailored for such individuals. Plain language summary EPICOVIDEHA registry reports inferior outcomes of COVID-19 in patients with Philadelphia-negative chronic myeloproliferative neoplasms receiving immunosuppressive therapies. Patients with Philadelphia-negative chronic myeloproliferative neoplasms (MPN) incur high rates of infections during the course of their disease. The present study was aimed at assessing which patient characteristics predicted a worse outcome of SARS-COV-2 infection in individuals with MPN. To pursue this objective, the researchers analyzed the data collected by EPICOVIDEHA, an international online registry, which includes individuals with hematological malignancies diagnosed with COVID-19 since February 2020. The database provided clinical data of 398 patients with MPN incurring COVID-19: Patients were mostly elderly (median age was 69 years); Forty-six percent of them were affected by myelofibrosis, which is the most severe MPN; Moreover, 32% were receiving immunosuppressive therapies (JAK inhibitors, such as ruxolitinib, steroids, or immunomodulatory IMID drugs, such as thalidomide) before COVID-19. Hospitalization was required in 54% of the patients, and the risk of being hospitalized for severe COVID-19 was independently predicted by Older age; Comorbidities; Exposure to immunosuppressive therapies. Overall, 22% of MPN patients deceased soon after COVID-19 and the risk of death was independently increased over twofold by Older age; Comorbidities; Exposure to immunosuppressive therapies before the infection. In conclusion, COVID-19 infection led to a particularly dismal outcome in MPN patients receiving immunosuppressive agents, including JAK inhibitors, or reporting multiple comorbidities. Therefore, specific preventive strategies need to be tailored for such individuals.

Abstract Extended geometry is based on an underlying tensor hierarchy algebra. We extend the previously considered L ∞ structure of the local symmetries (the diffeomorphisms and their reducibility) to incorporate physical fields, field strengths and Bianchi identities, and identify these as elements of the tensor hierarchy algebra. This requires considering the dual complex. We show how local invariance under the compact subgroup locally defined by a generalised metric arises as a “dual gauge symmetry” associated with a certain torsion Bianchi identity, generalising Lorentz invariance in the teleparallel formulation of gravity. The analysis is performed for a large class of finite-dimensional structure groups, with E 5 as a detailed example. The continuation to infinite-dimensional cases is discussed.

Abstract Background Many regulatory circuits in plants contain steps of targeted proteolysis, with the ubiquitin proteasome system (UPS) as the mediator of these proteolytic events. In order to decrease ubiquitin-dependent proteolysis, we inducibly expressed a ubiquitin variant with Arg at position 48 instead of Lys (ubK48R). This variant acts as an inhibitor of proteolysis via the UPS, and allowed us to uncover processes that are particularly sensitive to UPS perturbation. Results Expression of ubK48R during germination leads to seedling death. We analyzed the seedling transcriptome, proteome and metabolome 24 h post ubK48R induction and confirmed defects in chloroplast development. We found that mutations in single genes can suppress seedling lethality, indicating that a single process in seedlings is critically sensitive to decreased performance of the UPS. Suppressor mutations in phototropin 2 (PHOT2) suggest that a contribution of PHOT2 to chloroplast protection is compromised by proteolysis inhibition. Conclusions Overall, the results reveal protein turnover as an integral part of a signal transduction chain that protects chloroplasts during development.

Ensuring a functioning critical infrastructure system is crucial for many industries, including energy. Due to the transformation of the energy sector, it is necessary to take into account the impact of the induced changes on critical infrastructure. The presented paper focuses on the assessment of the effects of small dispersed photovoltaic power plants (PV), which are gaining in popularity both in households and in other buildings. Specifically, it is an impact assessment within the emergence of island operation in the selected area with the aim of restoring power to the facilities included in the critical infrastructure system as soon as possible. The specificity of the considered small photovoltaic power plants is their autonomy and impossibility of control from the superior control system. The presented case study compares possible approaches to power recovery on the one hand and the impact of different levels of dispersed PV on the other. The results provide important conclusions for the expected development of the critical infrastructure system in connection with the increase in autonomous distributed electricity sources.
