The lipophile extract of Lonchocarpus nicou roots afforded the new pyranochalcone 3-O-methylabyssinone A as well as the new rotenoids 7′-hydroxytephrosin, and 7′-nor-6′-oxo-2′,3′-dehydrorotenone, both compounds occurring with the known 7′-hydroxydeguelin and 7′-nor-6′-oxo-2′,3′-dehydro-12aβ-hydroxyrotenone. Furthermore, two rotenoid epoxides previously reported as resulting from the direct oxidative conversion of rotenone and 12aβ-hydroxyrotenone, respectively, were isolated for the first time from a plant source. All the structures were established on the basis of UV, MS, and NMR data.

The greatest concentration of pans in southern Africa occurs in the western Free State province, South Africa. A feature of many Free State pans is their fringing lunettes, located on the southern and south-eastern margins. Lunette dunes associated with pans in the neighbouring and presently drier Kalahari region show depositional ages, determined by optically stimulated luminescence (OSL) dating, primarily in the Holocene. However, to date, the precise timing of Free State lunette accumulation has not been investigated. The morphology, sedimentology and age of lunettes at five pan sites in the western Free State panfield are reported here. The lunettes form distinct topographic features, with heights up to 5 m above the pan floor, and all have been dissected by gully erosion. Sediment in the sand size class dominates in the lunettes, often overlying clay-rich basal or pan floor sediments. The intra- and intersite data consistency of 46 OSL ages is interpreted as reflecting regional causal factors responsible for lunette accretion, with phases of lunette building at 12—10 ka, 5.5—3 ka, 2—1 ka and 0.3—0.07 ka ago. These are in good agreement with the findings from pan-fringing lunettes in the southwest Kalahari and consistent with established records of palaeocirculation and wind direction over central southern Africa during the late Pleistocene. Lunettes in the western Free State are currently not in a major accretion phase. They are subject to degradation by localized fluvial erosion, with sediment being recycled into the pans.

A lipophile extract of Lonchocarpus nicou roots afforded two likely O -substituted isomers of the known tubaic acids. Spectroscopic analysis assigned to the former, named (−)-rotoic acid, the new 4-(2,3-dihydro-6,7-dimethoxychromon-3-oxy) tubaic acid structure which differs from (−)-deguoic acid, the latter, by the β-tubaic acid part. Biogenetically, the two compounds could be considered as resulting from the parent rotenone and deguelin, respectively, by oxidative ring-C cleavage.

Lonchocarpus nicou roots lipophile extract yielded, among isoflavonoids, two coumaronochromones isomers. Spectroscopic analysis assigned the structure of nor-dehydrodeguelin to the former and (−)-nor-dehydrorotenone to the latter, the two products only differing by the cyclization of the C 5 side chain. Biogenetically, the two compounds could be considered as resulting either from cyclization of the corresponding 2′-hydroxyisoflavones and/or from oxidative ring-B contraction of the dehydrorotenoids parents.

A series of 4-hydroxy-6-methoxyaurones and 4,6-dimethoxyaurones has been synthesised and tested for their binding affinity toward the nucleotide-binding domain of P-glycoprotein, an ABC (ATP-Binding Cassette) transporter which mediates the resistance of cancer cells to chemotherapy. These compounds differ from each other by the nature of the substituent on the aurone B-ring. The binding affinity seems to be linked to the nature of the substituent, as well as to the presence or the absence of a hydroxy group at position 4. We report the synthesis of 5-hydroxy-7-methoxy-2-phenyl-4-quinolones and their biological activity as antitumor agents. These molecules were initially evaluated for their ability to induce cell cycle arrest in the G2/M phase. Compounds that showed significant G2/M cell cycle arrest were tested for antiproliferative activity using both the MTT assay and the NCI in vitro 60 cell line human tumor screen. The 5-hydroxy-7-methoxy-2-phenyl-4-quinolone (3a) and 2-(3-fluorophenyl)-5-hydroxy-7-methoxy-4-quinolone (3f) were the most active in the cell cycle arrest test whereas 3f was found to be the most active in the MTT assay. In terms of structural requirements, we found that the presence of a 5-hydroxyl group, a 7-methoxy group, and an unsubstituted N1 were essential for the antimitotic activity. In accordance with the literature, a fluoro group at the 3'- or 2'-position and a methoxy or a chloro group at the 3'-position were found to be highly advantageous for both the cell cycle arrest and the antiproliferative activities.

3,5-Dimethoxyaniline was acetylated and found to be a key intermediate for the synthesis of a variety of biologically active aminoflavones and quinolones.
