13. Use of Nature-Based Schoolyards Predicts Students' Perceptions of Schoolyards as Places to Support Learning, Play, and Mental Health

- Abstract
Although green schoolyards provide many benefits to children and communities, little research explores students' perceptions of these benefits and how natural elements (e.g. gardens, trees) and associated use shape student experience. We examine the relationship between nature-based attributes, nature-based activities, and teacher-led activities with students' perceptions of schoolyards' benefits to self (e.g. feeling calm, good place to learn) and to community (e.g. contributions to cleaner water, increased biodiversity). We measured 199 3rd -6th grade students' perceptions of schoolyards' benefits from 9 schools in Raleigh, North Carolina. Overall, students viewed schoolyards positively, and recognized more benefits to self than community. We found that the relationship between the presence of nature-based attributes and students' positive feelings was limited. Instead, we found that nature-based activities and teacher-led activities had a positive relationship with students' positive feelings. Our findings suggest that school greening efforts need to be paired with policies and curricular practices to encourage interaction with natural elements to ensure benefits are fully realized by students.

26. The Flipped Classroom in ESL Teacher Education: An Example from CALL

- Abstract
The flipped classroom is one of many technology-enhanced teaching strategies. In this approach, students are responsible for initial learning at home (often via instructional videos) and class time is used for problem-solving and activities to deepen understanding. Although research on and use of the flipped classroom in language education is growing, little work has examined its use in teacher education, particularly for language teachers. To address this gap, this study examined the flipped classroom through the eyes of pre-service language teachers to reveal what hinders them from or encourages them to adopt this approach. Data were collected from students in a Computer-Assisted Language Learning (CALL) course; they experienced two flipped class sessions (complementing the traditional instructor-led sessions) and completed a survey about their experiences. Semi-structured interviews were also conducted with a sub-set of students to examine their perceptions in greater depth. Three major themes emerged regarding benefits of the flipped classroom: learner autonomy, learning by doing with support, and preventing cognitive overload. Four challenges emerged: learners' technology access and technical ability, technical support for instructors, ambiguous student responsibility, and an inability to provide immediate clarification. Three additional notable themes emerged: heightened awareness of peers in the classroom, different reactions to content-oriented versus technically-oriented instructional videos, and student workload. These themes are discussed in detail, along with suggestions for teacher training and professional development. Also considered is the need to establish guidelines for best practices in flipped classrooms and to develop high-quality approaches to flipping without a dependence on instructional videos.

- Abstract
Human observations can only capture a portion of ongoing classroom social activity, and are not ideal for understanding how children's interactions are spatially structured. Here we demonstrate how social interaction can be investigated by modeling automated continuous measurements of children's location and movement using a commercial system based on radio frequency identification. Continuous location data were obtained from 16 five-year-olds observed during three 1-h classroom free play observations. Illustrative coordinate mapping indicated that boys and girls tended to cluster in different physical locations in the classroom, but there was no suggestion of gender differences in children's velocity (i.e., speed of movement). To detect social interaction, we present the radial distribution function, an index of when children were in social contact at greater than chance levels. Rank-order plots indicated that children were in social contact tens to hundreds of times more with some peers than others. We illustrate the use of social ties (higher than average levels of social contact) to visualize the classroom network. Analysis of the network suggests that transitivity is a potential lens through which to examine male, female, and mixed-sex cliques. The illustrative findings suggest the validity of the new measurement approach by re-examining well-established gender segregation findings from a new perspective.

Background: The purpose of this qualitative study was to evaluate the effects of curricular changes made to foster graduate students' understanding of how they can be catalysts of change. For more than a decade, the concept of the citizen nurse has been woven into coursework in nursing education, thus indicating an evaluation process was needed. Method: Ten oral history interviews were completed with current students or alumni. Data were analyzed for codes, allowing the final themes to emerge. Results: Two main themes emerged. Students reported (1) having a new understanding of what it means to be a nurse and (2) feeling more prepared to take action. Need for admission to the neonatal intensive care unit (NICU) confers an increased risk of hearing loss in the newborn and of later neurodevelopmental impairment. In this retrospective longitudinal case-controlled study, we assess how the degree of prematurity, measured via gestational age, birth weight, and z-scores, in 138 infants admitted to the NICU are associated with permanent childhood hearing loss (PCHI) and 2-year developmental outcomes. Logistic regression analyses, Kruskal–Wallis analysis of variance, and Chi-squared tests were used. Independent of prematurity, PCHI and NICU admission were predictive of poor developmental outcomes. Twenty-one (47%) children with PCHI had a moderate-to-severe developmental delay, compared to three (7%) matched controls. Days in the NICU but not z-scores predicted PCHI. Z-score was not prognostic of moderate or severe developmental impairment in children with PCHI. The odds ratio of moderate-to-severe neurodevelopmental impairment with PCHI was high, at 12.48 [95% CI = 3.37–46.40]. Children with PCHI were significantly more likely to have cerebral palsy than their matched counterparts (30% vs. 2%). These findings challenge the conventional focus on gestational age and birth weight on neurodevelopmental outcomes for children with PCHI and NICU admission. Objectives Neutrophils play a key role in ANCA-associated vasculitis, both as targets of autoimmunity and as facilitators of vascular damage. In granulomatosis with polyangiitis (GPA), the data regarding the production of reactive oxygen species (ROS) in neutrophils are unclear. Further, recent data suggests that ROS production could have an anti-inflammatory effect through the regulation of inflammasomes and IL-1–related cytokines. We aimed to analyse ROS production in neutrophils from patients with GPA and investigate its association with IL-1–related cytokines and the autoantigen PR3. Methods Seventy-two GPA patients with disease flare were included in the NEUTROVASC prospective cohort study. ROS production in whole blood of patients with active GPA was evaluated and compared with that in the same patients in remission or healthy controls. Associations between ROS production, PR3 membrane expression on neutrophils, serum levels of IL-1–related cytokines as well as inflammasome-related proteins were analysed. Results We observed a robust defect in ROS production by neutrophils from patients with active GPA compared with healthy controls, independent of glucocorticoid treatment. Serum levels of IL-1–related cytokines were significantly increased in GPA patients, particularly in patients with kidney involvement, and levels of these cytokines returned to normal after patients achieved remission. Further, inflammasome-related proteins were significantly dysregulated in the cytosol of neutrophils as well as the serum from GPA patients. Background and Purpose: Pulmonary arterial hypertension (PAH), a rare fatal disorder characterised by inflammation, vascular remodelling and vasoconstriction. Current vasodilator therapies reduce pulmonary arterial pressure but not mortality. The G‐protein coupled formyl peptide receptors (FPRs) mediates vasodilatation and resolution of inflammation, actions possibly beneficial in PAH. We investigated dilator and anti‐inflammatory effects of the FPR biased agonist compound 17b in pulmonary vasculature using mouse precision‐cut lung slices (PCLS). Experimental Approach: PCLS from 8‐week‐old male and female C57BL/6 mice, intrapulmonary arteries were pre‐contracted with 5‐HT for concentration–response curves to compound 17b and 43, and standard‐of‐care drugs, sildenafil, iloprost and riociguat. Compound 17b‐mediated relaxation was assessed with FPR antagonists or inhibitors and in PCLS treated with TNF‐α or LPS. Cytokine release from TNF‐α‐ or LPS‐treated PCLS ± compound 17b was measured. Key Results: Compound 17b elicited concentration‐dependent vasodilation, with potencies of iloprost > compound 17b = riociguat > compound 43 = sildenafil. Compound 17b was inhibited by the FPR1 antagonist cyclosporin H but not by soluble guanylate cyclase, nitric oxide synthase or cyclooxygenase inhibitors. Under inflammatory conditions, the efficacy and potency of compound 17b were maintained, while iloprost and sildenafil were less effective. Additionally, compound 17b inhibited secretion of PAH‐relevant cytokines via FPR2. Conclusions and Implications: Vasodilation to compound 17b but not standard‐of‐care vasodilators, is maintained under inflammatory conditions, with additional inhibition of PAH‐relevant cytokine release. This provides the first evidence that targeting FPR, with biased agonist, simultaneously targets vascular function and inflammation, supporting the development of FPR‐based pharmacotherapy to treat PAH. In the still‐face episode of the Face‐to‐Face/Still‐Face (FFSF), parents are asked to become unresponsive. However, infant–parent interaction may be irrepressible, and there is some evidence that interaction during the still‐face is associated with attachment outcome. To explore these questions, we independently coded the continuous affective valence (negative to positive) of seventy‐three 6‐month‐old infants (45 males; 36 Hispanic/Latinx; 38 White, 3 Black/African‐American, 2 Asian, 30 multiracial) and their parents (Mage = 36 years; 5 males; 30 Hispanic/Latinx; 65 White, 3 Black/African‐American, 2 Asian, 2 unknown) during the FFSF and assessed attachment at 15 months with the Strange Situation Procedure (n = 66). There was a mean positive correlation between moment‐to‐moment parent and infant affective valence, indicating synchronous affective interaction during the still‐face (d = 0.63). Higher levels of affect interaction during the still‐face episode were detected in infants later classified as disorganised compared to securely attached (d = 0.97). Findings underscore the importance of testing for still‐face interaction and suggest that this interaction may be an unappreciated predictor of infant attachment outcomes. [ABSTRACT FROM AUTHOR]

Common sense and recent research make it clear that schools cannot be successful without strong principals. Strong principals help improve student performance in many ways, from shaping a school's mission and culture to hiring, developing, and retaining its teachers. Even so, principals often get short shrift in today's debates about human capital in public education. Compared to the large body of research and policy attention given to teachers, the field knows surprisingly little about the principal workforce. This knowledge gap means that it often is hard for school districts and states to make strategic decisions to improve their principal workforces. Where do most principals come from? How long do they stay? How well do they perform? How many will need to be hired and developed in the future? Few districts or states know the answers. To address these and other important questions about their principals, states need, among other things, to build detailed longitudinal data systems like the ones they use to track teachers and students. But in some places those types of systems are still a long way off. In the meantime, system leaders can examine the administrative data they already have to paint a basic picture of their principal workforce, one that can help prompt deeper questions and discussions about the challenges and opportunities they face. This "Principal Concerns" brief offers an example of this type of analysis for Wisconsin. Why should Wisconsin be concerned about its principal workforce? After all, by some measures, the state's schools are doing well. Wisconsin's NAEP scores, for example, are consistently higher than the national average. Yet there is still much work to be done to ensure that all students achieve at high levels, and strong leadership is key to that success. Under the state's recently revamped accountability system, 266 schools across the state are not meeting performance expectations. In Milwaukee Public Schools, the state's largest school system, only 21 percent of schools met or exceeded the state's expectations. (Contains 3 figures and 11 footnotes.)

50. Investigating the role of beta1 integrin in hepatic stellate cell activation and liver fibrosis

Liver fibrosis is characterised by progressive deposition of type 1 collagen-rich extracellular matrix in response to iterative liver injury. The progressive accumulation of scar tissue within the liver predisposes to the development of hepatocellular carcinoma, and ultimately leads to organ failure. Liver fibrosis is an increasing cause of morbidity and mortality. However, current treatment options are limited, and the only curative option for end stage liver disease is transplantation. Anti-fibrotic agents are urgently needed to halt, or reverse, the fibrotic process; however, to date, they have remained frustratingly elusive. The primary cell type responsible for laying down the pathological fibrotic matrix is the hepatic stellate cell (HSCs). In the healthy liver, HSCs are quiescent vitamin A storing cells, however, in response to liver injury they are activated into proliferative, migratory and contractile myofibroblasts. A number of cytokines and transcription factors are implicated in this activation process; in addition, biomechanical forces have emerged as an important regulator. The integrins are a family of cell surface receptors, which are predominately involved in mediating cellular interactions with the microenvironment. The work presented in this thesis demonstrates that beta1 integrin (Itgb1) plays an important role in HSC activation through regulation of the actin myosin cytoskeleton, and thereby, the cell’s ability to sense and respond to changes in the biomechanical microenvironment. By identifying and investigating downstream effectors of Itgb1 in HSC activation, the group I p21-activated kinases (Paks) were discovered as potential therapeutic targets in liver fibrosis. In addition, alpha11 integrin was identified as a fibroblast-specific partner of Itgb1 in HSCs, which crucially, may allow HSC-specific targeting of Itgb1. Taken together, the data presented in this thesis suggest that disrupting Itgb1 signalling in HSC activation may be a novel therapeutic avenue for liver fibrosis. In particular, pharmacological inhibition of the downstream effectors, the group I Paks, has shown promise as an anti-fibrotic both in vitro and in vivo.
