Investigations into cholinesterase inhibition have received attention from researchers in recent years for the treatment of Alzheimer’s disease. Cholinesterase enzymes, namely, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), hold pivotal significance in Alzheimer’s disease (AD) treatment. In this study, we utilized the ethanolic extract of Astragalus crenatus followed by liquid chromatography–electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) to separate and identify at least 21 compounds in the extract. Rosmarinic acid exhibited the highest concentration (96.675 ± 1.3 mg/g extract), succeeded by hesperidin (79.613 ± 1.2 mg/g extract), hesperetin (75.102 ± 1.4 mg/g extract), rutin (68.156 ± 1.6 mg/g extract), chlorogenic acid (67.645 ± 1.5 mg/g extract), fisetin (66.647 ± 2.3 mg/g extract), and hyperoside (63.173 ± 1.5 mg/g extract). A. crenatus extract efficiently inhibited both AChE and BChE activities in a dosage-dependent manner. Molecular docking was employed to scrutinize the anticholinesterase mechanisms of the identified phytocompounds. Notably, a network pharmacology analysis was executed for the most efficacious compound. Based on binding energies, hesperidin emerged as the most potent inhibitor against both AChE and BChE, exhibiting scores of −10.5 Kcal/mol and −9.8 Kcal/mol, respectively. Due to its dual inhibition of AChE and BChE activities, hesperidin from Astragalus crenatus holds promise for the development of novel therapeutics aimed at neurological disorders, particularly AD.

This study aimed to investigate the chemical composition and antidiabetic properties of cultivated Hyoscyamus albus L. The ethanol extract was analyzed using LC-MS/MS, and 18 distinct phenolic compounds were identified. Among these, p-coumaric acid (6656.8 ± 3.4 µg/g), gallic acid (6516 ± 1.7 µg/g), luteolin (6251.9 ± 1.3 µg/g), apigenin (6209.9 ± 1.1 µg/g), and rutin (5213.9 ± 1.3 µg/g) were identified as the most abundant polyphenolic molecules. In the in vitro antidiabetic experiment, the ability of the plant extract to inhibit α-glucosidase and α-amylase activities was examined. The results indicated that the extract from H. albus L. exhibited a higher inhibitory effect on α-amylase compared to α-glucosidase, with an IC50 of 146.63 ± 1.1 µg/mL and 270.43 ± 1.1 µg/mL, respectively. Docking simulations revealed that luteolin, fisetin, and rutin exhibited the most promising inhibitory activity against both enzymes, as indicated by their high contrasting inhibition scores. To further investigate the in vivo antidiabetic effects of H. albus L., an experiment was conducted using STZ-induced diabetic mice. The results demonstrated that the plant extract effectively reduced the levels of cholesterol and triglycerides. These findings suggest that H. albus L. may have therapeutic potential for managing hyperlipidemia, a common complication associated with diabetes. This highlights its potential as a natural remedy for diabetes and related conditions.

Fruit and vegetable production across the Mediterranean generates large volumes of by-products that can potentially lessen the livestock feeds deficit in some regions due to climate change effects and other socio-economic factors. However, the utilisation of agro-industrial by-products to their full potential is hindered by several factors such as variability in composition, limited development of preserving technologies and the presence of undesirable substances that may represent a risk for the animal or the final consumer. This work was conducted to study the nutritive value of a range of different alternative by-products from the Spanish agro-industry, assessing the potential to replace conventional ingredients often used in diets for small ruminants and the risk associated with the presence of pesticide residues and minerals. A total of 15 agro-industrial by-products commonly available in different regions of Spain were tested: three different types of apple pomace from three different varieties of apples (Perico, Regona, and Solarina), lemon pulp, avocado seed, broccoli, watermelon silage, olive cake, avocado cake, and six types of silages made with straw and a combination of different by-products (80/20 on fresh matter basis) from the intensive greenhouse horticulture: cucumber, tomato, pepper-cucumber mix (Pep-Cuc silage), aubergine-tomato mix (Aub-Tom silage), tomato-pepper-watermelon mix (Tom-Pep-Wat silage), and watermelon-melon-tomato mix (Wat-Mel-Tom silage). The study involved chemical characterization, including minerals and pesticides residues, and in vitro assessment of the ability to partially replace (10%, 20%, 30%, 50% and 100%) conventional ingredients in a diet for adult sheep (oat hay, barley and sunflower meal), depending on their chemical composition. The inclusion of by-products up to 30% replacement had not effect or improved some of the fermentation parameters and cucumber silage and apple pomace could successfully replace 100% of oat hay

Respiration chambers are the gold standard technique for measuring methane in ruminants provided that their gas recovery rates are close to 100%. The determination of the gas recovery rate of respiration chamber facilities is a central prerequisite to assess the accuracy of the methane emission quantification. However, data of recovery tests are seldom reported. This paper presents data from gas recovery tests applied to an experimental facility of four open-circuit respiration chambers designed to measure methane emissions from goats. The experimental facility is located at Thiverval-Grignon, France. The recovery test was assessed by placing a known source of methane emission at six locations in each chamber successively. For each chamber, the gas from the chamber and the ambient air were continuously sampled by a Multi-Gas Analyser 3500 gas analyser provided with a multiport unit that switches the sampling between the pipe from chamber and from the ambient air every 90 s. The analyser determines the concentration (ppm) of methane by infrared. The data were further imported in an R script for calculation of the methane recovery percentage. These data are useful resources for illustrating the protocol to assess the accuracy of respiration chambers.

The monthly variations in metal concentration of Cadmium (Cd), Zinc (Zn) and Chromium (Cr) were determined in tissues of Mytilus galloprovincialis and seawater obtained from the mussel farm installed along the Al Hoceima coasts, during the sampling period of 2018. The heavy metals encountered in the bivalves were higher than those observed in their environment with a decreasing order of Zn > Cr > Cd. The results also showed a decreasing trend for the metals studied during the summer sampling campaign. The maximum values were obtained during the wet periods. The monthly variations in trace metals observed in the mussel tissues were due to environmental parameters and the physiological state of the bivalves. Our results revealed that the average concentrations of pollutants found in M. galloprovincialis were below the limits allowed by the Food and Agriculture Organization of the United Nations (FAO) which are 2.0, 1000 and 1.0 mg/kg, for Cd, Zn and Cr, respectively in molluscan shellfish for human consumption. Therefore, the trace elements present in the tissues of Al Hoceima shore mussels do not represent a risk to human health associated with their consumption, even during the period of their maximum bioaccumulation. Our work highlights the potential use of Mytilus galloprovincialis in the Biomonitoring of metallic pollutants in this region of the northern Moroccan coasts.

Newborn ruminants possess an underdeveloped rumen which is colonized by microorganisms acquired from adult animals and the surrounding environment. This microbial transfer can be limited in dairy systems in which newborns are separated from their dams at birth. This study explores whether the direct inoculation of fresh or autoclaved rumen fluid from adult goats to newborn kids has a beneficial effect on rumen microbial development and function.

Dried orange pulp (DOP) can be incorporated into ruminant diets, but no reports have considered this strategy during the entire lactation period in goats. Two experiments were performed using lactating Payoya goats. In experiment 1, to study the effect, over 180 days, of DOP on milk yield and composition, blood metabolites and economic values, 44 primiparous goats were allocated into three groups: control diet (concentrate plus lucerne) and DOP40 and DOP80 diets, in which DOP replaced 40% and 80%, respectively, of the cereals. Nutrient digestibility and rumen fermentation were also studied (experiment 2). The DOP diets did not affect milk yield and composition. DOP triggered lower intake and digestibility of ether extract and crude protein. Ruminal fermentation was unaffected by DOP, except for a decrease in butyrate for DOP80. The energy balance was unaltered by diet while the balance and retention of nitrogen decreased. Regarding plasma biochemistry, DOP supplementation caused changes that could indicate an improvement in hepatic function and reduced muscular damage and oxidative muscular stress. Moreover, DOP80 provided a profit increase of EUR 3.27/goat. In conclusion, the partial replacement of cereals by DOP is a profitable and healthy nutritional strategy in dairy goats and is suitable for the entire lactation period without compromising productivity.

Plant-derived compounds have recently been gaining popularity as skincare factors due to their ability to absorb ultraviolet radiations and their anti-inflammatory, and antioxidant properties. In this light, this work aimed to evaluate in vitro the pharmacological activities of the butanolic extract prepared from the aerial parts of Astragalus gombiformis Pomel, an endemic species to southern Algeria. The sun protection factor was used to assess the photoprotective effect (SPF), the protein denaturation method to determine the anti-inflammatory activity, and brine shrimp nauplii and OxHLIA assay, respectively, to assess the cytotoxicity and antioxidant capacity of A. gombiformis. In addition, LC–ESI–MS analysis was employed for the characterization of the phenolic constituents of A. gombiformis. The results showed that A. gombiformis had high capacity for absorbing UV radiations with an SPF of 37.78 ± 0.85 and significant anti-inflammatory activity with a percentage inhibition of 75.38% which is close to that of diclofenac and ketoprofen. In addition, A. gombiformis was found to have effective cytotoxicity against Artemia nauplii with a DC50 value of about 44.7 µg/mL, but a weak hemolytic effect against human erythrocytes. LC–ESI–MS results detected the presence of 17 phenolic compounds with a predominance of cirsiliol, silymarin, quercitrin (quercetin-3-O-rhamnoside), and kaempferol. Taken together, these results suggest that A. gombiformis extract could be used as a skincare agent in cosmetic formulations, providing excellent antioxidant and anti-inflammatory protection, allowing the treatment of skin conditions, as well as a pharmaceutical agent with multidimensional applications.

The research about the role of saliva in ruminants has been mainly focused on its buffering capacity together with facilitation of the rumination process. However, the role of salivary bioactive components on modulating the activity of the rumen microbiota has been neglected until recently. This study developed an in vitro approach to assess the impact of different components in saliva on rumen microbial fermentation. Four different salivary fractions were prepared from four goats: (i) non-filtrated saliva (NFS), (ii) filtrated through 0.25 µm to remove microorganisms and large particles (FS1), (iii) centrifuged through a 30 kDa filter to remove large proteins, (FS2), and (iv) autoclaved saliva (AS) to keep only the minerals. Two experiments were conducted in 24 h batch culture incubations with 6 ml of total volume consisting of 2 ml of rumen fluid and 4 ml of saliva/buffer mix. In Experiment 1, the effect of increasing the proportion of saliva (either NFS or FS1) in the solution (0%, 16%, 33% and 50% of the total volume) was evaluated. Treatment FS1 promoted greater total volatile fatty acids (VFA) (+8.4%) and butyrate molar proportion (+2.8%) but lower NH-N concentrations than NFS fraction. Replacing the bicarbonate buffer solution by increasing proportions of saliva resulted in higher NH-N, total VFA (+8.0%) and propionate molar proportion (+11%). Experiment 2 addressed the effect of the different fractions of saliva (NFS, FS1, FS2 and AS). Saliva fractions led to higher total VFA and NH-N concentrations than non-saliva incubations, which suggests that the presence of some salivary elements enhanced rumen microbial activity. Fraction FS1 promoted a higher concentration of total VFA (+7.8%) than the other three fractions, and higher propionate (+26%) than NFS and AS. This agrees with findings from Experiment 1 and supports that ‘microbe-free saliva’, in which large salivary proteins are maintained, boosts rumen fermentation. BACKGROUND: Meeting the energy and nitrogen (N) requirements of high-performing ruminants at the same time as avoiding digestive disturbances (i.e. rumen acidosis) is a key priority in ruminant nutrition. The present study evaluated the effect of a cereal ammoniation treatment, in which barley grains are combined with urea and enzymes that catalyze the conversion of urea to ammonia to optimize rumen function. Twelve rumen cannulated sheep were randomly divided into two groups and fed a diet containing 60% of ammoniated barley (AMM) or untreated barley supplemented with urea (CTL) to investigate the impact on rumen fermentation and feed utilization. RESULTS: AMM had higher total N content and effective rumen degradable N than untreated barely. AMM sheep had a consistently higher rumen pH throughout the day (6.31 versus 6.03) and tended to have a lower post-prandial ammonia peak and higher acetate molar proportion (+5.1%) than CTL sheep. The rumen environment in AMM sheep favored the colonization and utilization of agro-industrial by-products (i.e. orange pulp) by the rumen microbes leading to a higher feed degradability. AMM sheep also had higher total tract apparent N digestibility (+21.7%) and urinary excretion of purine derivatives (+34%), suggesting a higher N uptake and microbial protein synthesis than CTL sheep. CONCLUSION: The inclusion of AMM in the diet of ruminants represents a valid strategy for maintaining rumen pH within a physiological range and improving N utilization by the rumen microbes, which could have positive effects on the health and productivity of animals in intensive production systems. These findings warrant further studies under conventional farm conditions. © 2021 The Authors. Condensed tannins are polyphenolic compounds which can exert beneficial effects in ruminants. They have the ability to bind proteins and decrease their degradation. They have also been reported to reduce methanogenesis and improve ruminants performances. The present work aimed to study the effect of purified condensed tannins from Acacia horrida extract on fermentation parameters, gas and methane production. In this context, seven substrates were selected: four Acacia species (Acacia pycnantha, Acacia dealbata, Acacia horrida and Acacia cyanophylla) and three plant by-products (date palm leaves, grenade peel and artichoke stems). Biological activity of tannins was evaluated by the incubation of the substrates in vitro with Polyethylene glycol (PEG). Tannins decreased concentration of all fermentation parameters (gas (p < 0.0001), methane (CH) (p= 0.0983), ammonia (N-NH) (p= 0.0382), Volatile Fatty Acids (VFA) (p= 0.0009), acetate (p <0.0001), propionate (p= 0.1024), butyrate (p= 0.0373), isobutyrate (p= 0.0046), valerate (p < 0.0001), isovalerate (p= 0.0032)). Substrates were also characterized by their content in main nutrients (dry matter (DM), organic matter (OM), ash, crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF),acid detergent lignin (ADL)) and in secondary metabolites (total phenols (TP), total tannins (TT) and condensed tannins (CT)). All substrates presented high CP content except by-products (> 100 g/kg DM) and moderate cell-wall components. CT content was comprised between 120.5 and 680.4 g/kg DM in plants and between 23.1 and 170.4 g/kg DM in plant by-products. A. horrida presented the highest biological activity. Thus, their CT were extracted and purified on a sephadex LH-20 column. Purified tannins from A. horrida were incubated with two different forages alfalfa hay and barley straw at three different concentrations: 50, 100, 150 mg/g DM. Dairy goat production systems in developed countries are experiencing an intensification process in terms of higher farm size, electronic identification, reproductive intensification, genetic selection and milking automation. This new situation generates “big data” susceptible to be used to aid farmers during the decision making process. This case study describes how the farm management can be improved by the use of the “Eskardillo” a tool with a smart-phone terminal which relies on three principles: i) systematic individual data recording (milking control, productivity, genetic merit, morphology, phylogeny, etc.), ii) big data processing and interpretation and iii) interactive feedback to the farmer to optimize farm management. This study evaluated the effectiveness of the Eskardillo tool by monitoring the productive parameters from 2013 to 2016 in 12 conventional Murciano-Granadina dairy goat farms which implemented the Eskardillo (ESK) in late 2014. Moreover, 12 conventional farms without Eskardillo were also monitored as control farms (CTL). Results demonstrated that ESK farms were able to better monitor the productivity and physiological stage of each animal and Eskardillo allowed selecting animals for breeding, replacement or culling according to each animal's records. As a result, goats from ESK farms decreased their unproductive periods such as the first partum age (−30 days), and the dry period length (−20 days) without negatively affecting milk yield per lactation. This study revealed an acceleration in the milk yield in ESK farms since this innovation was implemented (+26 kg / lactation per year) in comparison to the situation before (+7.3) or in CTL farms (+6.1). Data suggested that this acceleration in milk yield in ESK farms could rely on i) a greater genetic progress as a result of a more knowledgeable selection of high merit goats, ii) the implementation of a more effective culling off strategy based on the production, reproductive and health records

BACKGROUND: The rumen microbiota has been used as inoculum for in vitro studies and as a probiotic to improve productivity in young animals. However, great variability across studies has been noted depending on the inoculum considered. The present study aims to assess the relevance of different factors (microbial fraction, collection time, donor animal diet, fermentation substrate and inoculum preservation method) to maximize the rumen inoculum activity and set the standards for further in vitro and in vivo applications. RESULTS: Rumen inoculum sampled at 3 h after feeding led to greater microbial growth and activity [+12% volatile fatty acid (VFA), +17% ammonia] compared to before feeding. Similar results were noted when rumen liquid or rumen content were used as inocula. Rumen inoculum adapted to concentrate diets increased microbial activity (+19% VFA) independently of the substrate used in vitro. Freezing-thawing the inoculum, in comparison to fresh inoculum, decreased microbial activity (−14% VFA, −96% ammonia), anaerobic fungi and protozoa, with holotrichs protozoa being particularly vulnerable. Inoculum lyophilization had a stronger negative effect on microbial activity (−51% VFA) and delayed re-activation of the microbes, leading to lower levels of methanogens and anaerobic fungi, as well as almost complete wipe out of rumen protozoa. CONCLUSIONS: Fresh rumen fluid sampled at 3 h after feeding from donor animals that were fed concentrate diets should be chosen when the aim is to provide the most diverse and active rumen microbial inoculum. © 2018 Society of Chemical Industry.

Heavy metal concentrations of Cd, Cr, Cu, Fe, Ni, Zn, Co, and Pb were investigated in soft tissues of Mytilus galloprovincialis coming from an aquaculture farm in Cala Iris sea of AlHoceima. Mytilus galloprovincialis were collected monthly during the period January to December 2016. The seasonal variations were affected significantly the concentrations of metals (Cd, Fe and Cr) in M. galloprovincialis. The highest heavy metal concentrations were recorded in winter (0.89 mg/kg, 673.2 and 3.330 mg/kg; for Cd, Fe and Cr, respectively) and the lowest values were founded in summer for Cd (0.646 mg/kg), and in autumn for Fe (340.1 mg/kg) and Cr (1.959 mg/kg). A significant effect of seasons on metal concentrations can be attributed to a number of biological and environmental inter-related factors. Data from this study may provide information on the use of M. galloprovincialis as a bioindicator for heavy metals pollution in the Cala Iris Sea.

This work aimed to gain insight into the transition from milk to solid feeding at weaning combining genomics and metabolomics on rumen contents from goat kids treated with a methanogenic inhibitor (bromochloromethane, BCM). Sixteen goats giving birth to two kids were used. Eight does were treated (D+) with BCM after giving birth and over 2 months. One kid per doe in both groups was treated with BCM (k+) for 3 months while the other was untreated (k-). Rumen samples were collected from kids at weaning (W) and 1 (W + 1) and 4 (W + 4) months after and from does at weaning and subjected to 16S pyrosequencing and metabolomics analyses combining GC/LC-MS. Results from pyrosequencing showed a clear effect of age of kids, with more diverse bacterial community as solid feed becomes more important after weaning. A number of specific OTUs were significantly different as a result of BCM treatment of the kid at W while at W + 1 and W + 4 less OTUs were significantly changed. At W + 1, Prevotella was increased and Butyrivibrio decreased in BCM treated kids. At W + 4 only the effect of treating mothers resulted in significant changes in the abundance of some OTUs: Ruminococcus, Butyrivibrio and Prevotella. The analysis of the OTUs shared by different treatments revealed that kids at weaning had the largest number of unique OTUs compared with kids at W + 1 (137), W + 4 (238), and does (D) (23). D + k+ kids consistently shared more OTUs with mothers than the other three groups at the three sampling times. The metalobomic study identified 473 different metabolites. In does, lipid super pathway included the highest number of metabolites that were modified by BCM, while in kids all super-pathways were evenly affected. The metabolomic profile of samples from kids at W was different in composition as compared to W + 1 and W + 4, which may be directly ascribed to the process of rumen maturation and changes in the solid diet. The objective of this study was to examine the effects of four nitrocompounds (Nitrophenol, 5-Nitrobenzimidazol and two synthetic nitrocompounds ABLE 244 and ABLE 245) on methane production and fermentation characteristics using in vitro rumen batch culture. 0, 2, 8 or 12 µM of each nitrocompound were incubated. The higher concentrations of Nitrophenol and 5-Nitrobenzimidazol produced 60% less CH (P < 0.05) compared to controls, while two synthetic nitrocompounds ABLE 244 and ABLE 245 had no effect on CH production. Quantification of fermentation end-products indicated that fermentation efficiencies were not compromised by the nitro-treatments.

Hay una inconsistencia de la perspectiva analítica losó ca de frente a la estructura cerebral; ésta es más que regresión hacia los átomos del conocimiento, que para los analíticos clásicos está en el exterior. A la arquitectura cognitiva cerebral se la puede vislumbrar como un todo sintético que conjunta las dimensiones internas de la naturaleza humana con la externa, es decir que toma en cuenta tanto la exterioridad como la naturalidad y parte de ambas; en efecto el ordenamiento inteligente sabe bordar lo instintivo y límbico con los datos que le da la sensorialidad y a su vez lo instintivo-límbico en lo lingüístico-lógico y lo sensorial en lo lingüístico-lógico para la plani cación y la decisión, elementos muy propios y al mismo tiempo simultáneos en la estructura cerebral. Por lo tanto, el absolutizar la analítica es realmente una reducción del conocimiento, más aún, la reducción del signi cado de los contenidos a los hechos y a la materia, es un contradictorio cerebral, por la naturaleza misma de su estructura que no parcializa ni reduce nada como si es el caso de lo analítico. El empirismo clásico y el del siglo pasado delante de los estudios de la estructura y funcionamiento del cerebro, resulta ser parcial, pues poseen una estructura reduccionista que se asimila totalitaria. No es que sea inválido totalmente, sino que es parcialmente válido, es decir que, si se analizan los contenidos desde la sensorialidad cerebral, es más o menos coherente, pero actualmente, quedarse sólo en ese campo e interpretación, e impertinencia cognitiva.

The aim of this work was to evaluate the effect of feeding management during the first month of life (natural with the mother, NAT, or artificial with milk replacer, ART) on the rumen microbial colonization and the host innate immune response. Thirty pregnant goats carrying two fetuses were used. At birth one kid was taken immediately away from the doe and fed milk replacer (ART) while the other remained with the mother (NAT). Kids from groups received colostrum during first 2 days of life. Groups of four kids (from ART and NAT experimental groups) were slaughtered at 1, 3, 7, 14, 21 and 28 days of life. On the sampling day, after slaughtering, the rumen content was sampled and epithelial rumen tissue was collected. Pyrosequencing analyses of the bacterial community structure on samples collected at 3, 7, 14 and 28 days showed that both systems promoted significantly different colonization patterns (P = 0.001). Diversity indices increased with age and were higher in NAT feeding system. Lower mRNA abundance was detected in TLR2, TLR8 and TLR10 in days 3 and 5 compared to the other days (7, 14, 21 and 28). Only TLR5 showed a significantly different level of expression according to the feeding system, presenting higher mRNA abundances in ART kids. PGLYRP1 showed significantly higher abundance levels in days 3, 5 and 7, and then experienced a decline independently of the feeding system. These observations confirmed a highly diverse microbial colonisation from the first day of life in the undeveloped rumen, and show that the colonization pattern substantially differs between preruminants reared under natural or artificial milk feeding systems. However, the rumen epithelial immune development does not differentially respond to distinct microbial colonization patterns.
