Abstract

Adenoviruses have long been identified as good candidates for use as viral vectors in gene therapy and as vaccines. These viruses can infect multiple cell types, while in division or in quiescence, and are relatively easy to manipulate so that parts of their genome can be replaced with exogenous genes. Progressive safety improvements in replication-deficient adenoviral vectors have been achieved with the second and third generation, and ending with the gutless adenoviral vectors. Adenoviral vectors are immunogenic and can act as adjuvants. Nonetheless, the potency of human recombinant adenoviral vaccines was below expectations in clinical trials mainly because of the pre-existing adenoviral immunity found in the general population. This drawback can however become advantageous in animal health, as no previous immunity to human adenoviral vectors exists in animals. Other viral vectors viruses are used as vaccine, but adenoviruses remain the most employed and promising recombinant vector in veterinary medicine. In this chapter, we review the generation of adenoviral vectors, the immune response they trigger, and their advantages and disadvantages for veterinary use in terms of safety and efficacy. This chapter also describes how recombinant adenoviral vectors can be integrated as tools for vaccination and immunomodulation in veterinary medicine.

Abstract

Objective: To estimate the test-retest and inter-rater reliability of the new Spanish abbreviated version of the Luria Neuropsychological Diagnosis (DNA-2) battery for older adults. Method: A total of thirty cognitively healthy volunteers were examined in this study. The participants completed a comprehensive standardized assessment, encompassing cognitive and functional performance. Intraclass correlation coefficients (ICC) were used to examine test-retest and inter-rater reliability. One month was allowed between administrations. Furthermore, correlations between Luria DNA-2 (total and domain subscores) and other classical cognitive measures were explored. Results: The test-retest reliability on the overall Luria DNA-2 score was high (ICC=.834, 95% CI [.680,.917], p <.001). Furthermore, the inter-rater reliability for the total score demonstrated an excellent concordance between administrators (ICC=.990, 95% CI [.979,.995], p <.001). Positive and significant correlations were observed between Luria DNA-2 (both total and domain subscores) and the Addenbrooke's Cognitive Examination (ACE-III; ρ =.857, p <.001). Conclusions: This study supports the adequate reliability of the Luria DNA-2, as an abbreviated neuropsychological battery, for assessing cognitive performance in Spaniards aged 55 years and older. Future studies should continue to explore the psychometric properties of the Luria DNA-2, particularly those related to its diagnostic validity for early detection of cognitive impairment. Abstract

Dementia remains an underdiagnosed syndrome, and there is a need to improve the early detection of cognitive decline. This narrative review examines the role of neuropsychological assessment in the characterization of cognitive changes associated with dementia syndrome at different states. The first section describes the early indicators of cognitive decline and the major barriers to their identification. Further, the optimal cognitive screening conditions and the most widely accepted tests are described. The second section analyzes the main differences in cognitive performance between Alzheimer's disease and other subtypes of dementia. Finally, the current challenges of neuropsychological assessment in aging/dementia and future approaches are discussed. Essentially, we find that current research is beginning to uncover early cognitive changes that precede dementia, while continuing to improve and refine the differential diagnosis of neurodegenerative disorders that cause dementia. However, neuropsychology faces several barriers, including the cultural diversity of the populations, a limited implementation in public health systems, and the adaptation to technological advances. Nowadays, neuropsychological assessment plays a fundamental role in characterizing cognitive decline in the different stages of dementia, but more efforts are needed to develop harmonized procedures that facilitate its use in different clinical contexts and research protocols. Abstract

RESUMO: Trata-se de um relato de experiência vinculada à experiência profissional e acadêmica no Amentúrate Programa de Mentorias Universitárias. Programa do departamento de Educação da Universidade da Cantábria - UC ? Santander ? Espanha, que oferece a estudantes dos anos finais do ensino fundamental ou ensino médio com altas habilidades ou superdotação ? AH/SD, uma experiência de mentoria universitária. A mentoria é entendida como uma relação didática, relativamente estável ao longo do tempo, entre um mentor experiente (professor) e um mentee (pupilo,estudante AH/SD) menos experiente. O programa está no seu quarto ano de funcionamento e já atendeu mais de 90 estudantes da região. Destacamos a importância do Relato de Experiência ? RE, como mais uma possibilidade de criação de narrativa científica, especialmente no campo das pesquisas capazes de englobar processos e produções subjetivas, como é o caso da psicologia. Nesse sentido, e considerando que essa experiência compõe uma trajetória acadêmica de uma doutoranda, foi possível identificar a repetição de um discurso científico sobre a educação de superdotados, no que se refere a quatro questões: a) dificuldade em nomear e conceituar o fenômeno; b) maior participação masculina no programa; c) pouca ou nenhuma participação de estrangeiros como latinos ou africanos no programa e (d) estratégias educativas mais desafiadoras, complexas e que promovam a autonomia dos estudantes. Essas questões nos colocam frente a barreiras sociais e ideológicas, como o sexismo, o racismo e o classicismo que interferem no bem-estar e na qualidade de vida da pessoa superdotada. A invisibilidade de mulheres com AH/SD em programas de desenvolvimento de talentos nos provoca a questionar: por onde andam essas mulheres? ABSTRACT: This is an experience report linked to professional and academic experience in the Amentúrate University Mentorship Program. Program of the Department of Education at the University of Cantabria - UC ? Santander ? Spain, which offers gifted students in the final years of elementary school or high school a mentoring experience at the university. Mentoring is understood as a didactic relationship, relatively stable over time, between an experienced mentor (teacher) and a less experienced mentee (gifted student). The program is in its fourth year of operation and has already served more than 90 students in the region. We highlight the importance of the Experience Report, as one more possibility of creating a scientific narrative, especially in the field of research capable of encompassing subjective processes and productions, as is the case of psychology. In this sense, and considering that, this experience makes up the academic trajectory of a doctoral student, it was possible to identify the repetition of a scientific discourse on gifted education, with regard to four issues: a) difficulty in naming and conceptualizing the phenomenon; b) greater male participation in the program; c) little or no participation of foreigners such as Latinos or Africans in the program and (d) more challenging, complex educational strategies that promote student autonomy. These issues place us in the face of social and ideological barriers, such as sexism, racism and classicism that interfere with the well-being and quality of life of gifted people. The invisibility of gifted women in talent development programs makes us a question: where are these women?

Abstract

RESUMEN: Los estudios que investigan Inteligencia Emocional en alumnos con altas capacidades son escasos y presentan resultados contradictorios. Algunas investigaciones consideran que los estudiantes con altas capacidades son grupos vulnerables y, según algunos informes, están más involucrados en situaciones de acoso como víctimas y/o acosadores (Casino-Garcia et al., 2019). Los diferentes modelos de vulnerabilidad muestran la importancia de identificar no sólo los factores de riesgo, sino también los factores de protección que permiten diseñar intervenciones preventivas más eficaces (Ezpeleta, 2005). Dentro de esta perspectiva, este trabajo presenta los resultados de la aplicación del cuestionario de Inteligencia Emocional TMSS-24 en una población de estudiantes con altas capacidades. Cabe señalar que estos resultados forman parte de una investigación más amplia que investiga la calidad de vida y el apoyo social percibido de jóvenes (de entre 12 y 17 años) con altas capacidades que participan en Amentúrate, un programa de mentorías realizado en la Universidad de Cantabria. En este sentido, los datos y reflexiones aquí publicados deben entenderse como parciales y son iniciativas exploratorias para comprender el universo de este alumnado en la región Cántabra. Los resultados mostraron que los participantes presentaban, en general, una buena Inteligencia Emocional. En relación con las diferencias de género, no se encontraron diferencias significativas, pero destaca que las chicas obtuvieron mejores puntuaciones que los chicos en cuanto a la claridad de sus sentimientos y a la capacidad de reparación sobre los mismos. Para terminar, se discuten los resultados, se proponen aportes teóricos y prácticos a la literatura y se sugieren implicaciones para padres y docentes. ABSTRACT: There are few studies researching the Emotional Intelligence of gifted students and their results are contradictory. Some researchers consider that gifted students are vulnerable and, according to some reports, are more involved in bullying situations as victims and/or bullies (Casino-Garcia et al., 2019). The different vulnerability models show the importance of identifying not only the risk factors, but also the protective factors that allow the design of more effective preventive interventions (Ezpeleta, 2005). From this perspective, this paper presents the results of the administration of the TMSS-24 Emotional Intelligence questionnaire in a population of gifted students. It should be noted that these results are part of a broader research where we assess the quality of life and perceived social support of gifted students (between 12 and 17 years old) who participate in Amentúrate, a mentoring program carried out at the University of Cantabria. Thus, the data and reflections showed must be understood as partial and exploratory initiatives to understand gifted students in the Cantabrian region. The results revealed that pour participants have, in general, a good Emotional Intelligence. In relation to gender differences, no significant differences were found, but it stands out that the girls obtained higher scores than the boys in terms of the clarity of their feelings and the ability to repair them. Finally, the results are discussed, theoretical and practical contributions to the literature are proposed, and implications for parents and teachers are suggested.

Nos adentramos en el análisis de un derecho relativamente reciente: el derecho a la adaptación de jornada de progenitores y cuidadores. Este derecho ha surgido como una opción a investigar para avanzar hacia una igualdad más efectiva entre hombres y mujeres. Se reconoce que, históricamente, las mujeres han llevado la carga principal de las responsabilidades familiares, lo que ha generado desigualdades en el ámbito laboral y social. El presente trabajo se enfoca en el estudio de la conciliación como una estrategia para abordar las brechas de género, centrándose en la evolución normativa de las fórmulas flexibles de trabajo y la adaptación de la jornada laboral. Abstract

[EN] African swine fever virus (ASFV) causes a devastating hemorrhagic disease with worldwide circulation and no widely available therapeutic prevention. The infectious particle has a multilayered architecture that is articulated upon an endoplasmic reticulum (ER)-derived inner envelope. This membrane acts as docking platform for the assembly of the outer icosahedral capsid and the underlying core shell, a bridging layer required for the formation of the central genome-containing nucleoid. While the details of outer capsid assembly are relatively well understood, those of core formation remain unclear. Here we report the functional characterization of pEP84R, a transmembrane polypeptide embedded in the inner envelope that surrounds the viral core. Using an ASFV recombinant inducibly expressing the EP84R gene, we show that absence of pEP84R results in the formation of non-infectious core-less icosahedral particles displaying a significant DNA-packaging defect. Concomitantly, aberrant core shell-like structures formed by co-assembly of viral polyproteins pp220 and pp62 are mistargeted to non-ER membranes, as also occurs when these are co-expressed in the absence of other viral proteins. Interestingly, co-expression of both polyproteins with pEP84R led to the formation of ER-targeted core shell-like assemblies and co-immunoprecipitation assays showed that pEP84R binds to the N-terminal region of pp220. Altogether, these results indicate that pEP84R plays a crucial role in core assembly by targeting the core shell polyproteins to the inner viral envelope, which enables subsequent genome packaging and nucleoid formation. These findings unveil a key regulatory mechanism for ASFV morphogenesis and identify a relevant novel target for the development of therapeutic tools against this re-emerging threat., [ES] El virus de la peste porcina africana es un virus complejo de ADN de múltiples capas que produce una enfermedad altamente letal que actualmente afecta a los cerdos domésticos y salvajes en África, Europa, Asia, Oceanía y, más recientemente, América. A pesar de una extensa investigación, todavía faltan vacunas ampliamente disponibles o cualquier otra estrategia antiviral y quedan por abordar cuestiones relevantes sobre la replicación del ASFV. El presente estudio caracteriza la proteína de membrana viral pEP84R como un componente clave para el ensamblaje del virión. Utilizando enfoques genéticos, bioquímicos y microscópicos, proporcionamos evidencia de que pEP84R actúa como un enlace molecular que une el núcleo interno que contiene el genoma con las capas virales externas. Estos hallazgos amplían significativamente nuestra comprensión de los mecanismos subyacentes a la formación de la partícula viral infecciosa, que pueden ser relevantes para la investigación de vacunas y antivirales.

Abstract

Mercury is a very dangerous toxic metal that bioaccumulates very easily in organisms, and it migrates through the food web. The specimens studied in this study were Scomber colias. In the results obtained for the years 1973, 1992, and 2021, the concentration of mercury has been decreasing considerably over the decades, starting with 0.23 ± 0.04 mg/kg in 1973, and having less than half in 2021 with 0.11 ± 0.01 mg/kg. This may be due to the laws imposed by the countries against pollution and to a greater extent in the elimination of mercury; thanks to these measures it has been possible to reduce the concentration of mercury by half in S. colias. Otherwise, the conservation and availability of historical collections of living beings should be considered an unvalued source of information which could help to monitor legislation measures launched to ameliorate the human impact on the natural environment.

39. Religiosas y el aggiornamento conciliar en España: hacia una renovación identitaria (1962-1965)

Abstract

The purpose of this article is to analyse the aggiornamento of Vatican II during Franco’s regime. In particular, the focus of attention is on women religious and their identity crisis after the ecclesiastical call to renewal. In this way, during the council’s years, it is perceived how the nun archetype of nationalcatholicism coexisted with another one promoted by Vatican II, which was characterized by an open position with new models of being a woman and religious. Therefore, the categories of modernity, gender and religion will be interwoven in the study in order to approach the complexity of this process.; El objetivo de este artículo es profundizar en el aggiornamento del Vaticano II durante el franquismo. Especialmente, el foco de interés se centra en las religiosas y el giro identitario que asumieron tras la llamada eclesiástica a la modernización. De esta manera, durante los años conciliares, se percibe cómo el arquetipo monjil del nacionalcatolicismo comienza a convivir con otro impulsado desde el Vaticano II, más abierto y en consonancia con la nueva doctrina conciliar sobre las mujeres y la vida consagrada. Se entrecruzarán, por tanto, las categorías de modernidad, género y religión para un acercamiento a la complejidad de este proceso.

Abstract

Bluetongue virus (BTV) is an arbovirus transmitted by the bite of infected Culicoides midges that affects domestic and wild ruminants producing great economic losses. The infection induces an IFN response, followed by an adaptive immune response that is essential in disease clearance. BTV can nonetheless impair IFN and humoral responses. The main goal of this study was to gain a more detailed understanding of BTV pathogenesis and its effects on immune cell populations. To this end, we combined flow cytometry and transcriptomic analyses of several immune cells at different times post-infection (pi). Four sheep were infected with BTV serotype 8 and blood samples collected at days 0, 3, 7 and 15pi to perform transcriptomic analysis of B-cell marker+, CD4+, CD8+, and CD14+ sorted peripheral mononuclear cells. The maximum number of differentially expressed genes occurred at day 7pi, which coincided with the peak of infection. KEGG pathway enrichment analysis indicated that genes belonging to virus sensing and immune response initiation pathways were enriched at day 3 and 7 pi in all 4 cell population analyzed. Transcriptomic analysis also showed that at day 7pi T cell exhaustion pathway was enriched in CD4+ cells, while CD8+ cells downregulated immune response initiation pathways. T cell functional studies demonstrated that BTV produced an acute inhibition of CD4+ and CD8+ T cell activation at the peak of replication. This coincided with PD-L1 upregulation on the surface of CD4+ and CD8+ T cells as well as monocytes. Taken together, these data indicate that BTV could exploit the PD1/PD-L1 immune checkpoint to impair T cell responses. These findings identify several mechanisms in the interaction between host and BTV, which could help develop better tools to combat the disease.
