Context Eutrophication caused extensive macroalgal blooms in the Peel–Harvey Estuary, Western Australia, in the 1970s. Nutrient inputs were reduced and an artificial channel was constructed in 1994 to increase marine flushing. Aims This study examines benthic mollusc populations in the estuary in 1978, 2000 and 2020, to determine what changes have occurred in the estuaries over time. Methods Quantitative samples were made at 10 sites in autumn and spring of each year; physical and chemical parameters were measured in 2000 and 2020. Key results Species composition was stable, dominated by Arthritica semen and Hydrococcus brazieri; however, there have been substantial changes in abundance of these and less common species. Conclusions The exact cause(s) of density changes could not be determined, but it is likely to be due to a combination of factors. Implications Further changes in mollusc assemblages in south-western Australian estuaries are expected as the climate warms and dries and the estuaries are stressed by human population growth.

Trace element supplementation to the freshwater environment can influence the plankton density and species diversity, contributing to the nutrition of aquaculture species, especially during the juvenile stage. An experiment was conducted under laboratory conditions to evaluate the effects of supplementing different mixtures of manganese, silica and phosphorus on the plankton density and species diversity and their impact on cultured juvenile marron (Cherax cainii, Austin and Ryan, 2002). Manganese, silica and phosphorus in concentrations of 0.0024, 0.41, 0.05 mg*L-1; 0.0041, 0.82, 0.12 mg*L-1; and 0.0058, 1.26, 0.25 mg*L-1 respectively termed as low, medium and high were supplemented to tank water containing a phytoplankton density of 3.77 ± 0.16 × 106 cells*L-1 and 292.9 ± 17.6 individuals*L-1 of zooplankton, and plankton growth was observed every 24 h for 6 days. Afterwards, a 3-month trial was conducted studying the effects of these trace element concentrations and resulting plankton densities on marron growth, survival, moulting, gut microbiota and health indices. Silica supplementation at high concentration increased the diatom abundance, silica and phosphorus supplementation at higher concentration that resulted in a significant increase in plankton density and species diversity, leading to improved marron health indices than the control and the tanks receiving a low concentration. Marron-specific growth rate, weight gain and dissolved copper concentration in haemolymph were significantly higher in tanks with higher supplementation and higher plankton density. Marron survival, moult interval and total haemocyte count were not affected by the supplementation. Marron gut microbiota at higher trace element concentration supplementation showed a significant increase in abundance of phosphate solubilizing bacteria.

The abundance and diversity of the plankton community represents the health of the aquatic ecosystem, and plays an important role in the growth of cultured animals under aquaculture conditions. The temporal variations of plankton abundance, taxonomic composition, diversity, evenness and species richness were studied in three old and three new semi-intensive marron (Cherax cainii, Austin and Ryan, 2002) ponds. Water parameters such as temperature, dissolved oxygen, pH, turbidity, TAN, nitrite, nitrate and reactive phosphate were recorded, and plankton samples were collected every two months, for one year of juvenile production cycle. A total of twenty-six phytoplankton and seven zooplankton genera were recorded. Chlorophyceae was the dominant class of phytoplankton throughout the year, followed by Trebouxiophyceae. Rotifera comprised 49.8% of the total zooplankton community (individuals L−1), the largest proportion of any group. Temporal variations impacted the plankton abundance and community structure, and plankton abundance were more abundant during summer. The pond age did not influence the phytoplankton abundance, whereas zooplankton abundance was higher in older ponds.

In the present study, we investigated the possibility of identifying the source oils of exposed fish using ratios of bicyclic sesquiterpane (bicyclane) chemical biomarkers. In the event of an oil spill, identification of source oil(s) for assessment, or for litigation purposes, typically uses diagnostic ratios of chemical biomarkers to produce characteristic oil "fingerprints." Although this has been applied in identifying oil residues in sediments, water, and sessile filtering organisms, so far as we are aware this has never been successfully demonstrated for oil-exposed fish. In a 35-day laboratory trial, juvenile Lates calcarifer (barramundi or Asian seabass) were exposed, via the diet (1% w/w), to either a heavy fuel oil or to Montara, an Australian medium crude oil. Seasonal variations can affect the concentration of trace elements, and the change in their concentrations can affect the natural productivity of freshwater aquaculture ponds. Hence, we studied the seasonal variations of the 12 pre‐selected trace elements (Co, Cu, Fe, Mn, Zn, Se, Ca, Mg, P, S, Al, Si) and their relationships with primary and secondary productivity in two aged ponds, stocked with three different life stages of marron (Cherax cainii), for a period of 1 year. Trace element analysis was performed by using (Agilent, ICP‐OES). Except Co and Se, all trace elements, and primary and secondary productivity were influenced by seasonal variation. The pond age significantly influenced the concentrations of some trace elements. On a seasonal basis, trace elements were positively correlated with the plankton abundance, species diversity, and wet and dry plankton weights. Seasonal variations and pond age affected the dissolved concentrations of trace elements and plankton productivity.

International audience; Wetlands and their biodiversity are constantly threatened by contaminant pollution from urbanisation. Despite evidence suggesting that snakes are good bioindicators of environmental health, the bioaccumulation of contaminants in reptiles is poorly researched in Australia. We conducted the first broad-scale analysis of 17 metals and trace elements, 21 organochlorine pesticides, and 14 polycyclic aromatic hydrocarbons in the sediments (4 samples per site, December 2018) from four wetlands along an urban gradient in Perth, Western Australia, and from the livers (5 livers per site, February-April 2019) of western tiger snakes Notechis scutatus occidentalis captured at those sites. All 17 metals and trace elements were detected in the sediments of wetlands as well as 16 in the livers of tiger snakes. Arsenic, Cu, Hg, Pb, Se, and Zn were at concentrations exceeding government trigger values in at least one sediment sample. Two organochlorine pesticides and six of seven polycyclic aromatic hydrocarbons were detected in the sediments of a single wetland, all exceeding government trigger values, but were not detected in tiger snakes. Metals and trace elements were generally in higher concentration in sediments and snake livers from more heavily urbanised wetlands. The least urbanised site had some higher concentrations of metals and trace elements, possibly due to agriculture contaminated groundwater. Concentrations of nine metals and trace elements in snake livers were statistically different between sites. Arsenic, Cd, Co, Hg, Mo, Sb, and Se near paralleled the pattern of contamination measured in the wetland sediments; this supports the use of high trophic wetland snakes, such as tiger snakes, as bioindicators of wetland contamination. Contamination sources and impacts on these wetland ecosystems and tiger snakes are discussed herein.

Zeolite is known to uptake toxic metals and filter nitrogenous waste from aquaculture effluents. The present study aimed to investigate the impacts of zeolite in three different applications namely, dietary zeolite (DZ), suspended zeolite (SZ) in the water column, and a combination of both (DZSZ) relative to unexposed freshwater crayfish, marron (control). At the end of the 56-days trial, the impact was assessed in terms of characterization of microbial communities in the culture environment and the intestine of marron. Alongside the microbial communities, the innate immune response of marron was also evaluated. The 16S rRNA data showed that marrons exposed to the suspended zeolite had a significant increase of bacterial diversity in the gut, including the restoration of marron core operational taxonomic units (OTUs), relative to other forms of exposures (DZ, DZSZ) and the control. Suspended zeolite alone also increased the number of unshared OTUs and genera, and improved predicted metabolic functions for the biosynthesis and digestion of proteins, amino acids, fatty acids, and hormones. In the tank sediment, the shift of microbial communities was connected more strongly with the time of experiment than the type of zeolite exposure. In the second case, only control marron had a different microbial ordination in terms of rare taxa present in the community. Nevertheless, the modulation in the gut environment was found more prominent in DZ, relative to modulation in the tank sediments. The taxa-environment correlation identified Rhodoferax as the most potential bacteria in removing nitrogenous waste from the rearing environment. Further analysis showed that SZ resulted in the upregulation of genes associated with the innate immune response of marron. Overall results suggest that SZ can be used to enrich microbial communities in the gut and tank sediments and better immune performance of marron.

The gut microbiome of fish contains core taxa whose relative abundances are modulated in response to diet, environmental factors, and exposure to toxicogenic chemicals, influencing the health of the host fish. Recent advances in genomics and metabolomics have suggested the potential of microbiome analysis as a biomarker for exposure to toxicogenic compounds. In this 35-day laboratory study, 16S RNA sequencing and multivariate analysis were used to explore changes in the gut microbiome of juvenile Lates calcarifer exposed to dietary sub-lethal doses of three metals: vanadium (20 mg/kg), nickel (480 mg/kg), and iron (470 mg/kg), and to two oils: bunker C heavy fuel oil (HFO) (1% w/w) and Montara, a typical Australian medium crude oil (ACO) (1% w/w). Diversity of the gut microbiome was significantly reduced compared to negative controls in fish exposed to metals, but not petroleum hydrocarbons. The core taxa in the microbiome of negative control fish comprised phyla Proteobacteria (62%), Firmicutes (7%), Planctomycetes (3%), Actinobacteria (2%), Bacteroidetes (1%), and others (25%). Differences in the relative abundances of bacterial phyla of metal-exposed fish were pronounced, with the microbiome of Ni-, V-, and Fe-exposed fish dominated by Proteobacteria (81%), Firmicutes (68%), and Bacteroidetes (48%), respectively. The genus Photobacterium was enriched proportionally to the concentration of polycyclic aromatic hydrocarbons (PAHs) in oil-exposed fish. The probiotic lactic acid bacterium Lactobacillus was significantly reduced in the microbiota of fish exposed to metals. Transcription of cytokines IL-1, IL-10, and TNF-a was significantly upregulated in fish exposed to metals but unchanged in oil-exposed fish compared to negative controls. However, IL-7 was significantly downregulated in fish exposed to V, Ni, Fe, and HFOs. Fish gut microbiome exhibits distinctive changes in response to specific toxicants and shows potential for use as biomarkers of exposure to V, Ni, Fe, and to PAHs present in crude oil.

Urbanisation alters landscapes, introduces wildlife to novel stressors, and fragments habitats into remnant ‘islands’. Within these islands, isolated wildlife populations can experience genetic drift and subsequently suffer from inbreeding depression and reduced adaptive potential. The Western tiger snake (Notechis scutatus occidentalis) is a predator of wetlands in the Swan Coastal Plain, a unique bioregion that has suffered substantial degradation through the development of the city of Perth, Western Australia. Within the urban matrix, tiger snakes now only persist in a handful of wetlands where they are known to bioaccumulate a suite of contaminants, and have recently been suggested as a relevant bioindicator of ecosystem health. Here, we used genome-wide single nucleotide polymorphism (SNP) data to explore the contemporary population genomics of seven tiger snake populations across the urban matrix. Specifically, we used population genomic structure and diversity, effective population sizes (Ne), and heterozygosity-fitness correlations to assess fitness of each population with respect to urbanisation. We found that population genomic structure was strongest across the northern and southern sides of a major river system, with the northern cluster of populations exhibiting lower heterozygosities than the southern cluster, likely due to a lack of historical gene flow. We also observed an increasing signal of inbreeding and genetic drift with increasing geographic isolation due to urbanisation. Effective population sizes (Ne) at most sites were small (< 100), with Ne appearing to reflect the area of available habitat rather than the degree of adjacent urbanisation. This suggests that ecosystem management and restoration may be the best method to buffer the further loss of genetic diversity in urban wetlands. If tiger snake populations continue to decline in urban areas, our results provide a baseline measure of genomic diversity, as well as highlighting which ‘islands’ of habitat are most in need of management and protection.

The uptake of metals into the aragonite lattice of the fish otolith (ear-bone) has been used for decades as a historical record of exposure to metals in polluted environments. The relative abundance of two metals in particular, Ni and V, are used in forensic chemical analysis of crude oils to assist in confirming its origin. In this study we investigate the potential for metal accumulation in otoliths to act as a biomarker of exposure to crude oil. Using a 33-day static-renewal laboratory trial design, 56 juvenile Lates calcarifer (commonly known as Asian seabass or barramundi) were fed diets enriched with V (20 mg/kg), Ni (500 mg/kg), Fe (500 mg/kg), and two crude oils with distinctly different metals profiles: a heavy fuel oil (1% w/w) and a typical Australian medium crude (1% w/w). Fish exposed to crude oils showed Ba and Al retained in otoliths in a dose-dependent manner, but fish fed V-, Ni- and Fe-enriched diets showed no metal increase in otoliths, indicating that V, Ni and Fe are not incorporated into the otolith of L. calcarifer via dietary exposure. For crude oils, incorporation into otolith for many metals is likely limited due to porphyrin casing reducing their bioavailability. Principal components analysis (PCA) and subsequent linear discriminatory analysis (LDA) of selected otolith metals demonstrated that, even despite large variability in the metal abundances detected in otolith between individuals within the test groups (cv = 1.00), it is possible to discriminate between fish exposed to different crude oils using multivariate analysis of their otolith microchemistry.

We examine here the effects on evaporative water loss, at and below thermoneutrality, of perturbing the evaporative environment for the red-capped parrot (Purpureicephalus spurius) by modifying the ambient relative humidity or the diffusive properties of the ambient environment using a helium‑oxygen mix (helox). We found that evaporative water loss did not change with relative humidity at an ambient temperature of 30 °C, but there was a negative relationship for evaporative water loss with relative humidity at 20 and 25 °C. The evaporative water loss per water vapour pressure deficit between the bird and its ambient environment was not constant with relative humidity, as would be expected for a physical effect (slope = 0); rather there was a significant positive relationship with relative humidity at ambient temperatures of 25 and 30 °C. Consequently, we conclude that the red-capped parrot can physiologically control its EWL over a range of relative humidities. For the first time for a bird species, we also confirmed EWL control using a second methodology to perturb the evaporative environment, and demonstrated that a more diffusive helox atmosphere has no effect on evaporative water loss of live birds, but evaporative water loss was higher for dead birds in helox compared to air. Our results for evaporative water loss and other physiological variables for red-capped parrots are consistent with the hypothesis that evaporative water loss is under physiological control.

Wetland snakes, as top predators, are becoming globally recognised as bioindicators of wetland contamination. Livers are the traditional test organ for contaminant exposure in organisms, but research is moving towards a preference for non-lethal tissue sampling. Snake scales can be used as an indicator of exposure, as many metals bind to the keratin. We used laser ablation with inductively coupled plasma-atomic emission spectroscopy and mass spectrometry (LA-ICP-MS) to quantify the concentrations of 19 metals and metalloids (collectively referred to 'metals' hereafter) in Western tiger snake (Notechis scutatus occidentalis) scales from four wetlands along an urban gradient, and compared them to concentrations measured in captive tiger snake scales. We conducted repeat measures to determine the concentration accuracy of each metal using LA-ICP-MS. Concentrations in wild Western tiger snake scales were significantly higher than in reference tiger snake scales for most metals analysed, suggesting accumulation from environmental exposure. We compared the scale concentrations to sediment concentrations of sampled wetlands, and found inter-site differences between mean concentrations of metals in scales parallel patterns recorded from sediment. Four metals (Mn, As, Se, Sb) had strong positive correlations with liver tissue contents suggesting scale concentrations can be used to infer internal concentrations. By screening for a larger suite of metals than we could using traditional digestive methods, we identified additional metals (Ti, V, Sr, Cs, Tl, Th, U) that may be accumulating to levels of concern in tiger snakes in Perth, Western Australia. This research has progressed the use of LA-ICP-MS for quantifying a suite of metals available in snake scales, and highlights the significance of using wetland snake scales as a non-lethal indicator of environmental contamination.

Urban ecosystems and remnant habitat 'islands' therein, provide important strongholds for many wildlife species including those of conservation significance. However, the persistence of these habitats can be undermined if their structure and function are too severely disrupted. Urban wetlands, specifically, are usually degraded by a monoculture of invasive vegetation, disrupted hydrology, and chronic-contamination from a suite of anthropogenic pollutants. Top predators-as bioindicators-can be used to assess and monitor the health of these ecosystems. We measured eight health parameters (e.g., parasites, wounds and scars, tail loss and body condition) in a wetland top predator, the western tiger snake, Notechis scutatus occidentalis. For three years, snakes were sampled across four wetlands along an urban gradient. For each site, we used GIS software to measure the area of different landscapes and calculate an urbanisation-landscape score. Previously published research on snake contamination informed our calculations of a metal-pollution index for each site. We used generalised linear mixed models to assess the relationship between all health parameters and site variables. We found the metal-pollution index to have the most significant association with poor body condition. Although parasitism, tail loss and wounds differed among sites, none of these parameters influenced body condition. Additionally, the suite of health parameters suggested differing health status among sites; however, our measure of contemporary landscape urbanisation was never a significant predictor variable. Our results suggest that the health of wetland predators surrounding a rapidly growing city may be offset by higher levels of environmental pollution.

The release of petroleum hydrocarbons into the environment from natural seeps, well blowouts, pipeline leaks, shipping accidents and deliberate tank washing poses an ongoing threat to marine ecosystems. Distinguishing the source of oil contamination in exposed biota can be relatively straightforward if samples of the oil are available but, in their absence, such discrimination in fish poses a major challenge. The use of physiological and behavioral biomarker analysis provides a useful tool to describe sub-lethal effects of toxicant exposure. In this study we describe the responses of 12 biomarkers in Lates calcarifer (Asian seabass) following a 33-day dietary exposure (1%w/w) to heavy fuel oil (HFO) and to Montara, a typical Australian medium crude oil (MCO). Principal components analysis was used to differentiate between fish exposed to HFO from those exposed to MCO. Inferences can be made about the composition of an oil from the biomarker profiles produced in exposed fish.

Many cities worldwide are established adjacent to estuaries and their catchments resulting in estuarine contamination due to intense anthropogenic activities. The aim of this study was to evaluate if fish living in an estuarine urban waterway were affected by contamination, via the measurement of a suite of biomarkers of fish health. Black bream (Acanthopagrus butcheri) were sampled in a small urban embayment and a suite of biomarkers of fish health measured. These were condition factor (CF), liver somatic index (LSI), gonadosomatic index (GSI), hepatic EROD activity, polycyclic aromatic hydrocarbon (PAH) biliary metabolites, serum sorbitol dehydrogenase (s-SDH) and branchial enzymes cytochrome C oxidase (CCO), and lactate dehydrogenase (LDH) activities. The biomarkers of exposure EROD activity, and pyrene- and B(a)P-type biliary metabolites confirmed current or recent exposure of the fish and that fish were metabolizing contaminants. Relative to a reference site, LSI was higher in fish collected in the urban inlet as was the metabolic enzyme LDH activity. CF, GSI, s-SDH, CCO, and naphthalene-type metabolites were at similar levels in the urban inlet relative to the reference site. PAH biliary metabolite ratios of high-molecular-weight to low-molecular-weight suggest that fish from the urban inlet were exposed to pyrogenic PAHs, likely from legacy contamination and road runoff entering the embayment. Similarly, the sediment PAH ratios and the freshness indices suggested legacy contamination of a pyrogenic source, likely originating from the adjacent historic gasworks site and a degree of contamination of petrogenic nature entering the inlet via storm water discharge. Biomarkers of exposure and effect confirmed that black bream collected in the Claisebrook Cove inlet, Western Australia, are currently exposed to contamination and are experiencing metabolic perturbations not observed in fish collected at a nearby reference site.

Hepatic gene expression and liver histology were examined in sand flathead (Platycephalus bassensis) from six locations in Port Phillip Bay, Victoria, Australia. Four sets of genes including thyroid-related genes (D1, D2, TTR, TRα and TRβ), metal metabolism-related genes (MT, MTF1, TF, Ferritin and FPN1), apoptosis-related genes (Diablo/SMAC1, Diablo/SMAC2 and CYP1A) and an endoplasmic reticulum stress biomarker gene (GRP78) were examined in female flathead using qRT-PCR. TRβ and Diablo/SMAC1 gene expression was significantly up-regulated in fish from all polluted sites compared to those from a reference site. The transcripts of TRα and FPN1 were significantly higher in flathead from Corio Bay, while the hepatic mRNA of TTR and GRP78 were significantly lower in those fish. Positive correlations were observed between Diablo/SMAC1 and CYP1A, D2 and TRβ, TRα and TRβ. This study demonstrates that application of pathway-based biomarker genes and histopathology can provide comprehensive information on the impact of environmental pollutants on fish.

Activities of hepatic catalase (CAT) and superoxide dismutase (SOD) increased at the highest dose relative to the carrier control group. Lipid peroxidation (LPO), serum-sorbitol dehydrogenase (s-SDH) and DNA damage in blood cells were higher for all BaP doses when compared to the carrier control group. At high dosage, the production of BaP metabolites was paralleled by induced activity of the antioxidant enzyme SOD, and high levels of DNA damage in blood cells. In a similar way, high LPO was concomitant to elevated s-SDH in the bloodstream, suggesting that lipid peroxidation caused the loss of membrane integrity and leakage of s-SDH from hepatocytes into the bloodstream. These biomarkers were also positively co-correlated. The results demonstrate the potential use of a suite of biomarkers for tambaqui living in contaminated tropical aquatic environments. In particular, we recommend the analysis of DNA damage in blood cells, as this was highly correlated with all other biomarkers. The conversion of ethoxyresorufin to the fluorescent product resorufin by the enzyme ethoxyresorufin- O -deethylase (EROD) enables researchers to rapidly measure the upregulation of this protein in response to exposure to a range of organic contaminants e.g. PAH, PCBs. The EROD activity assay has been widely used to examine the effects of such pollutants on fish taxa in both laboratory and field studies. This review is intended to provide fundamental information for researchers using this EROD activity as an endpoint, including methods used in the assay, the species studied to date, the background EROD levels which would be expected for these species and when available, the EROD activity induction potential for these species. While the focus in on Australian studies, many species listed in this review have a worldwide distribution and the information presented may be extend to other bioindicator fish species. Common shortfalls in the published literature leads to recommendations: it is recommended to have multiple laboratory control or field reference groups as basal EROD activity might vary with biotic and/or abiotic factors. The use of native species over introduced species offers no advantages, with EROD activity induction potential (x-fold) being similar for native or introduced fish species. Similarly, in laboratory studies, EROD activity induction potential is comparable for lab/hatchery-reared fish to the activity observed in field-caught animals. Because EROD activity is often reported to reduce at high concentrations of toxicants, laboratory studies should use a carefully considered range of contaminant concentrations rather than a single exposure concentration, as a single exposure concentration may be on the decreasing side of the response curve. The measurement of serum sorbitol dehydrogenase (SDH) activity in conjunction with EROD activity is recommended to insure liver functions are not jeopardized by high contaminant levels. In field studies, and depending on the variability observed in EROD activity within a given fish species, a number of 8–13 non-reproductively active fish, preferably of similar age and sex, per site is recommended to improve the chances of detecting a significant difference in EROD activity, if one does exist. Finally, an inter-continental comparison of EROD activity induction potential (expressed as x-fold relative to a control or reference group) suggests that highly inducible species can be found on all continents, with reported EROD activity induction as high as 135-fold in exposed groups but more commonly less than 5-fold relative to laboratory control or field reference groups.

Tiger snakes (Notechis scutatus) in wetlands of South-West Western Australia (SW WA) are commonly parasitised by the nematode Ophidascaris pyrrhus. Host-parasite interactions are complex and can potentially be impacted by factors such as urbanisation or climate. We assessed whether urbanisation, distance to wetland sites, and climatic factors have influenced parasitism in tiger snakes from specimens collected over the last century. We dissected 91 museum specimens of tiger snakes across SW WA and counted gastrointestinal nematodes. Binomial generalised linear modelling, with presence/absence of nematodes as a response variable, was used to determine which factors were driving infection. Model selection using AICc values showed that proximity to wetlands, rainfall and topographic wetness were most strongly associated with the probability of infection of snakes by nematodes. We also found a slight positive correlation between nematode abundance and annual mean maximum temperature. We found no significant influence of distance to urban centre on nematode burdens; however, our results suggest that water-related variables are a key driver of nematode parasitism in tiger snakes in SW WA. Imposex is a condition in which females of dioecious marine snails develop rudimentary male characteristics. It is caused by tributyltin (TBT) used as an antifoulant in vessel paints since the late 1960s. Following the 2008 international ban on TBT, a decreasing rate of imposex has been observed worldwide. In Western Australia, imposex surveys of the whelk Thais orbita up to 2011 in the Perth metropolitan area suggested a decreasing trend but a significant recovery has not been confirmed. Collection of T. orbita from 11 sites in 2019 demonstrated a virtually complete recovery from imposex. Although it is generally accepted that male snails are not affected by TBT, the historical data set allowed confirmation that the male penis is of similar size in T. orbita collected at heavily affected sites and at relatively unaffected sites. Similarly, imposexed female snails had similar shell lengths to female snails at non-impacted sites.

There is limited information on the impacts of anthropogenic noise on dolphin behaviour, making assessment and mitigation of impacts from anthropogenic noises difficult. As the use of echolocation and other vocalizations are of vital importance for cetaceans, it is important to better understand the potential impact of anthropogenic acoustic disturbance. The small Indo-Pacific bottlenose dolphin (Tursiops aduncus) community that inhabits the Fremantle Inner Harbour regularly travels through an area where impact and vibratory pile driving occurred during wharf upgrading. The overall aim of this study was to measure the noise energy created by pile driving associated with wharf construction activities in the Fremantle Inner Harbour and to determine whether the reduced detection of dolphins within the vicinity of the wharf was associated with pile driving activities. Measuring noise was accomplished with noise loggers and a hand-held digital acoustic recorder, under water, allowing identification of signals produced by impact and vibratory pile drivers and calculating the energy of recorded noise. Dolphin detections in the Inner Harbour were conducted by examination of high-definition video recordings. The association of pile driving and dolphin detections was assessed using Generalized Estimating Equations (GEEs), using observations before and during pile driving. The final model indicated that there was a difference in detections between the two treatments, with more dolphin detections observed when there was no pile driving activity taking place (mean = 0.26 +/- 0.03 SE) than during pile driving (mean = 0.18 +/- 0.04 SE). Significant efforts are invested in field studies using fish, and it is important to optimize the number of organisms collected to evaluate the possible impacts of contamination. This paper provides ecotoxicologists with the approximate numbers of fish needed to identify statistically significant differences among samples using physiological indices and biochemical markers of fish health. The numbers of fish to collect are reported for ethoxyresorufin-o-deethylase (EROD); ethoxycoumarin-o-deethylase (ECOD), serum sorbitol dehydrogenase (sSDH), stress proteins, gonadosomatic index, liver somatic index, condition factor, and biliary metabolites of polycyclic aromatic hydrocarbons. The number of fish to collect was as few as four for ECOD activity (with a power of 80%), but as high as 106 for CF (with a power of 95%). Achieving statistical significance between sites does not help in the interpretation of the biological significance of a parameter, but well-planned field samplings will maximize the chances of correctly identifying areas of concern.

Black bream (Acanthopagrus butcheri) were collected from an artificial inlet, Claisebrook Cove, Western Australia. Claisebrook Cove is adjacent to an historic contaminated site that was remediated during the 1990s. It was later identified as a priority area due to elevated levels of sediment contaminants including Zn, Cu, and Pb. Black bream were collected from this cove in 2005 and 2012 and their otoliths were analysed by laser ablation inductively coupled plasma mass spectrometry of the most recent growth zone. Levels of Zn and Mn, which are metabolically regulated, did not correlate with sediment contamination. However, reduction in sediment Cu levels over time coincided with reduced Cu otolith levels from 2005 to 2012. Results indicate that the elemental composition of the marginal edge of Black bream otoliths can identify bioavailable contaminants in an urban estuary and, with monitoring, can be utilized to establish long-term trends.

Concentrations of PAHs, OCs, PCBs and trace metals were determined in the white muscle of sand flathead Platycephalus bassensis collected at 6 locations in Port Phillip Bay during 2015. No PAHs, OCs or PCBs were detected in the white muscle of sand flathead at any of the locations, however measurable levels of As, Cu, Hg, Se and Zn were detected at all sites. Only As and Hg exhibited regional difference in white muscle concentrations, with As present only in a non-toxic organic form and Hg measured at levels that are comparable to levels reported in reference sites in other studies. All contaminants detected in the white muscle of sand flathead collected in Port Phillip Bay in 2015 were below Australian Food Standards guideline values, and by world standards, the Port Phillip Bay sand flathead population is considered minimally contaminated. Furthermore, tissue contaminant concentrations appear to be decreasing over time.

Synthetic-based drilling muds (SBMs) offer excellent technical characteristics while providing improved environmental performance over other drilling muds. The low acute toxicity and high biodegradability of SBMs suggest their discharge at sea would cause minimal impacts on marine ecosystems, however, chronic toxicity testing has demonstrated adverse effects of SBMs on fish health. Sparse environmental monitoring data indicate effects of SBMs on bottom invertebrates. However, no environmental toxicity assessment has been performed on fish attracted to the cutting piles. SBM formulations are mostly composed of synthetic base oils, weighting agents, and drilling additives such as emulsifiers, fluid loss agents, wetting agents, and brine. The present study aimed to evaluate the impact of exposure to individual ingredients of SBMs on fish health. To do so, a suite of biomarkers [ethoxyresorufin-O-deethylase (EROD) activity, biliary metabolites, sorbitol dehydrogenase (SDH) activity, DNA damage, and heat shock protein] have been measured in pink snapper (Pagrus auratus) exposed for 21 days to individual ingredients of SBMs. The primary emulsifier (Emul S50) followed by the fluid loss agent (LSL 50) caused the strongest biochemical responses in fish. The synthetic base oil (Rheosyn) caused the least response in juvenile fish. The results suggest that the impact of Syndrill 80:20 on fish health might be reduced by replacement of the primary emulsifier Emul S50 with an alternative ingredient of less toxicity to aquatic biota. The research provides a basis for improving the environmental performance of SBMs by reducing the environmental risk of their discharge and providing environmental managers with information regarding the potential toxicity of individual ingredients.

Hsp70 protein in three tissue types (gill, liver, and muscle) from black bream (Acanthopagrus butcheri) collected in a highly variable estuarine environment was investigated to determine which tissue provides better intersite discrimination. The usefulness of hsp70 expression to identify anthropogenic stress under field conditions was evaluated. Intersite differences were detected in hsp70 levels in gill and white muscle of black bream while liver showed no spatial difference. There was high interfish variability in hsp70 levels in each tissue group. A post hoc power analysis of the datasets for each tissue found that in black bream, white muscle provided the best discriminatory power to elucidate spatial variability. Only 11 fish per site are required to identify significant intersite differences in white muscle whereas for gill and liver tissues 14 and 21 fish per site, respectively, would be required. Because of high intertissue and interindividual variability, field measurement of hsp70 should be complemented by evidence of changes in other biomarkers of fish health.

Separate laboratory experiments were conducted to examine if incorporation of Zn into the otoliths of juvenile pink snapper ( Pagrus auratus Forster) was related to levels in the food and/or water. In the first experiment, fish were fed a regular diet (600 mg Zn kg − 1 dw, control group) or a Zn-enriched diet (6000 mg Zn kg − 1 dw or 9000 mg Zn kg − 1 dw) for 35 days. In the second experiment, fish were exposed to waterborne Zn concentrations of − 1 (control), 50 µg L − 1 , 100 µg L − 1 and 200 µg L − 1 for 35 days. The sagittal otoliths were analysed using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Juvenile fish exposed to higher concentrations of waterborne Zn did not display increased Zn levels in their otoliths. However, Zn levels in the otoliths of fish consuming the Zn-enriched diet were significantly higher relative to control fish. This study clearly demonstrated that dietary Zn was the major source of Zn incorporated into the otoliths by this marine fish.

Otoliths of black bream (Acanthopagrus butcheri) collected from the Swan River Estuary were analysed by Laser Ablation Inductively Coupled Plasma Mass Spectrometry (LA-ICP-MS) to measure concentrations of 14 trace metals. Trace metal concentrations in the otoliths may be related to the environmental exposure history of fish to contamination. The following metal isotopes were investigated: aluminium (27Al), calcium (44Ca), manganese (55Mn), iron (57Fe), copper (65Cu), zinc (66Zn), strontium (88Sr), cadmium (111Cd), tin (120Sn), barium (138Ba), mercury (202Hg), lead (208Pb) and the metalloids arsenic (75As, 77As) and selenium (82Se). Significant differences in otolith trace metal composition were found between sampling sites. Lead and 57Fe were consistently lower in downstream fish relative to upstream fish, while 88Sr varied with the salinity gradient in the urban estuary. Lead and 57Fe followed similar patterns within the otoliths, and appeared to provide the best discriminatory power for relating otolith metal concentration to the environmental history of the fish.

The value of serum sorbitol dehydrogenase (s-SDH) activity to diagnose chemically induced liver damage in the fish, black bream (Acanthopagrus butcheri), was assessed by intraperitoneal injection with pentachlorophenol sodium salt. Day 3 post-injection s-SDH activity levels were elevated in fish injected at the highest dosage (30 mg Kg−1) compared to control and other treatment groups. Control fish had s-SDH activity levels within the range of 15 to 88 mU mL−1 serum, while at the highest Na-PCP treatment s-SDH activity was 2.5 to 3.5 times higher on day 3 relative to control fish with levels returning to control levels by the 6th day following injection. An increase in the liver somatic index was not identified until day 6 post-injection and histological damage (hyperplasia) was not detected until the 9th day following injection in the high dosage treatment group. Biomarkers of fish health are recognised as valuable biomonitoring tools that inform on the impact of pollution on biota. The integration of a suite of biomarkers in a statistical analysis that better illustrates the effects of exposure to xenobiotics on living organisms is most informative; however, most published ecotoxicological studies base the interpretation of results on individual biomarkers rather than on the information they carry as a set. To compare the interpretation of results from individual biomarkers with an interpretation based on multivariate analysis, a case study was selected where fish health was examined in two species of fish captured in two ports located in Western Australia. The suite of variables selected included chemical analysis of white muscle, body condition index, liver somatic index (LSI), hepatic ethoxyresorufin-O-deethylase activity, serum sorbitol dehydrogenase activity, biliary polycyclic aromatic hydrocarbon metabolites, oxidative DNA damage as measured by serum 8-oxo-dG, and stress protein HSP70 measured on gill tissue. Statistical analysis of individual biomarkers suggested little consistent evidence of the effects of contaminants on fish health. However, when biomarkers were integrated as a set by principal component analysis, there was evidence that the health status of fish in Fremantle port was compromised mainly due to increased LSI and greater oxidative DNA damage in fish captured within the port area relative to fish captured at a remote site. The conclusions achieved using the integrated set of biomarkers show the importance of viewing biomarkers of fish health as a set of variables rather than as isolated biomarkers of fish health.

The blue colour observed in the serum of female blue-throated wrasse Notolabrus letricus was related to the presence of biliverdin. The biliverdin is probably stored in the female's serum until deposited in the skin upon transition to the male reproductive status, resulting in the brilliant blue colouration typical of males of this protogynous labrid species.
