Abstract: Global changes push to set up strategies able to mitigate and adapt agricultural and forest crops to environmental variability, and the sustainable intensification of production processes under agricultural and forestry systems is one of the approaches mainly supported. In Italy biomass and biogas are the renewable energy sources that have shown the greatest potential for growth in recent years. In this context, during the XV National Congress held in Bolzano in February 2018, the Italian Association of Agricultural Scientific Societies has promoted an analysis about potential and limits of the sustainable intensification of agricultural and forestry systems for bioenergy production. This document reports the outcome, in the form of a commented discussion on the main evidences and proposals from technical-scientific and operational points of view.

2. Fluoxetine accelerates epileptogenesis and magnifies disease severity in a rat model of acquired epilepsy

Abstract: OBJECTIVE: Many people with epilepsy experience comorbid anxiety and depression, and antidepressants remain a primary treatment for this. Emerging evidence suggests that these agents may modulate epileptogenesis to influence disease severity. Here, we assessed how treatment with the selective serotonin reuptake inhibitor (SSRI) antidepressant fluoxetine impacts epileptogenic, behavioral, and pathological sequelae following status epilepticus. METHODS: Male Wistar rats received kainic acid to induce status epilepticus (SE) or vehicle (sham). Animals then received either fluoxetine (10 mg/kg/day) or vehicle for 8 weeks via subcutaneous osmotic pump. Video-electroencephalography was recorded continuously until behavioral testing at day 56, including assessments of anxiety- and depression-like behavior and spatial cognition. Postmortem immunocytochemistry studies examined mossy fiber sprouting. RESULTS: Fluoxetine treatment significantly accelerated epileptogenesis following SE, reducing the average period to the first spontaneous seizure (from 32 days [vehicle] to 6 days [fluoxetine], p < .01). Also, fluoxetine exposure magnified the severity of the resultant epilepsy, increasing seizure frequency compared to vehicle (p < .01). Exposure to fluoxetine was associated with improved anxiety- and depression-like behaviors but significantly worsened cognition. Mossy fiber sprouting was more pronounced in fluoxetine-treated rats compared to vehicle (p < .0001). SIGNIFICANCE: Our studies demonstrate that, using a model exhibiting spontaneous seizures, epileptogenesis is accelerated and magnified by fluoxetine, an effect that may be related to more severe pathological neuroplasticity. The differential influence of fluoxetine on behavior indicates that different circuitry and mechanisms are responsible for these comorbidities. These findings suggest that caution should be exercised when prescribing SSRI antidepressants to people at risk of developing epilepsy.

Abstract: To guarantee a safe compliance with the ambitious targets of acid gas (HCl, SO2) removal set by recent European regulations, existing waste-To-energy (WtE) plants equipped with single-stage acid gas treatment systems might take into consideration the installation of an additional treatment stage as retrofit. Multi-stage treatment systems can achieve higher acid gas removal efficiency and flexibility to cope with highly variable inlet pollutant concentrations, resulting from variability in waste composition. From a sustainability viewpoint, the choice of the most suited retrofitting option has to be driven not only by considerations on performance at the WtE plant but also by considerations on the indirect environmental burdens related to the material and energy consumption and waste generation of the selected options. The present study analyses the full environmental profile of two competitive dry retrofitting solutions (the lowtemperature injection of hydrated lime and the high-Temperature injection of calcined dolomite) by considering their life cycle impacts associated with the supply of reactants and the disposal of process residues. Modelling of the material balance of the alternatives as a function of pollutant load in the inlet flue gas allowed estimating the effect of flue gas composition and mode of operation of the two-stage system on the overall environmental impact. Both retrofitting options were found to reduce the life cycle impacts of the base case, but the use of hydrated lime showed the greatest potential. The generation of process residues constitutes a relevant fraction of the overall impacts. An alternative scenario for the management of process residues was evaluated to quantify the reduction of impact achievable with a commercially available valorisation route.

Abstract: Pluripotent stem cells (PSCs) have the capacity to give rise to all cell types of the adult body and to expand rapidly while retaining genome integrity, representing a perfect tool for regenerative medicine. PSCs are obtained from preimplantation embryos as embryonic stem cells (ESCs), or by reprogramming of somatic cells as induced pluripotent stem cells (iPSCs). Understanding the metabolic requirements of PSCs is instrumental for their efficient generation, expansion and differentiation. PSCs reshape their metabolic profile during developmental progression. Fatty acid oxidation is strictly required for energy production in naive PSCs, but becomes dispensable in more advanced, or primed, PSCs. Other metabolites directly affect proliferation, differentiation or the epigenetic profile of PSCs, showing how metabolism plays an instructive role on PSC behaviour. Developmental progression of pluripotent cells can be paused, both in vitro and in vivo, in response to hormonal and metabolic alterations. Such reversible pausing has been recently linked to mammalian target of rapamycin activity, lipid metabolism and mitochondrial activity. Finally, metabolism is not simply regulated by exogenous stimuli or nutrient availability in PSCs, as key pluripotency regulators, such as Oct4, Stat3 and Tfcp2l1, actively shape the metabolic profile of PSCs.

Abstract: The setting up of the ARGO detector at the YangBaJing Cosmic Ray Laboratory (4300 m a.s.l., Tibet, P.R. China) has been completed during the last spring (2007). It consists of a central carpet made of 130 identical sub-units of 12 RPCs each (a "cluster"), covering a surface of about 5800 m2 with 93% active area, and a guard ring of 24 further clusters of the same type surrounding the central carpet with a lower sampling density. Signals are picked up by external electrodes of small size, thus allowing the sampling of EAS with high space-time granularity. Shower events are detected at a trigger rate of about 4 kHz. Events with a few particles detected by a single cluster are counted in scaler mode on a time base of 500 ms. The intrinsic modularity of the ARGO detector allowed us to collect data even during the setting-up period, using only the central carpet (or even part of it). Some preliminary results from the analysis of events collected in a few months of data taking are presented. © 2008 Elsevier B.V. All rights reserved.

Abstract: Following the completion of the 5700m 2 RPC carpet-like AS array at YangBaJing this year, a further plan based on the ARGO Carpet is raised. It characterizes with its high altitude site, its full-coverage detector array and the multi-parameter measurements. Apart from studies on TeV and sub-TeV energy range, UHE Gamma-ray sources and the Knee Physics will become its characteristic subjects. In the first phase of the plan, the Yangbajing Super Complex Array (YSCA) will include a 10 4 m 2 RPC Carpet, five indoor μ -detectors (∼170 m 2 each), four big outdoor μ -detectors (∼432 m 2 each) and a traditional field scintillation detector array surrounding the Carpet. Monte-Carlo study shows that, with such an array in YBJ (4300m a.s.l.), γ -ray induced air showers can be separated from proton induced air showers clearly using only the observed electron and μ data event by event. To assist the classification of UHE air showers induced by different primary nuclei groups without severe interference by Composition/Model Entanglement, a second phase plan having a Central Burst Detector Array (CBDA) and some pulse shape detectors is schemed.

Abstract: To verify whether it is possible, in people with diabetes mellitus (DM) considered at very high cardiovascular (CV) risk, stratify this risk better and identify significant modifiable risk factor (including lifestyle habits) to help patients and clinicians improve CV prevention. People with DM and microvascular diseases or one or more CV risk factors (hypertension, hyperlipidemia, smoking, poor dietary habits, overweight, physical inactivity) included in the Risk and Prevention study were selected. We considered the combined endpoint of non-fatal acute myocardial infarction and stroke and CV death. A multivariate Cox proportional analysis was carried out to identify relevant predictors. We also used the RECPAM method to identify subgroups of patients at higher risk. In our study, the rate of major CV events was lower than expected (5 % in 5 years). Predictors of CV events were age, male, sex, heart failure, previous atherosclerotic disease, atrial fibrillation, insulin treatment, high HbA1c, heart rate and other CV diseases while being physically active was protective. RECPAM analysis indicated that history of atherosclerotic diseases and a low BMI defined worse prognosis (HR 4.51 95 % CI 3.04–6.69). Among subjects with no previous atherosclerotic disease, men with HbA1c more than 8 % were at higher CV risk (HR 2.77; 95 % CI 1.86–4.14) with respect to women. In this population, the rate of major CV events was lower than expected. This prediction model could help clinicians identify people with DM at higher CV risk and support them in achieving goals of physical activity and HbA1c.

Abstract: Fibroleiomyoma is the most common benign tumour of the uterus and sometimes it may also be present during pregnancy. The purpose of this study was to report 11 pregnant pazients with fibroleiomyomas larger than 5 cm submitted to myomectomy during cesarean section. No complications were observed and blood transfusion was given to only two patients with thrombocytopenia. This retrospective investigation would indicate that myomectomy during cesarean section can be performed without significant morbidity by expert surgeon and adopting careful and rigorous technique. © Copyright 2012, CIC Edizioni Internazionali.

Abstract: Introduction. During routine vaginal ultrasound in postmenopausal women it is frequently to find fluid collections in uterine cavity. The aim of this study is to analyze the clinical significance of this find, using as diagnostic methods hysteroscopy and endometrial biopsy. Materials and methods. Twenty postmenopausal patients (mean age 60.6±8.9 years) with endometrial fluid collections diagnosed by intravaginal ultrasound have been studied. Endometrial thickness was less than 5 mm in 16 patients, greater than 5 mm in the other 4 patients. All patients underwent to hysteroscopy and endometrial biopsy. Results. Histological examination of patients with endometrial ultrasound 5 mm deposed for endometrial hyperplasia. At ultrasound follow-up performed after one month, three months and six months from hysteroscopy, patients that showed endometrial atrophy had no more fluid collections, respectively, in 37.5%, in 43.75% and in 56, 25% of cases; in patients with endometrial hyperplasia, however, the fluid collection remained. Conclusion. Ultrasonographic finding of endometrial fluid collections when endometrium had not preneoplastic characteristics can presumably be attributed to impaired motility of the uterus and / or cervical stenosis. © Copyright 2012, CIC Edizioni Internazionali.

Abstract: The ARGO-YBJ array for EAS detection at high altitude (4300 m a.s.l.) is the largest RPC-based detector in operation at present (active area of 6700 m 2 out of a total instrumented surface of about 10 4 m 2 ). The constant monitoring of the main operational and environmental parameters is essential for both keeping the detector status under control and understanding the detector behaviour more deeply. Exploiting the information provided by the ARGO-YBJ Detector Control System, the present status of the detector is described and some correlations between the environmental and the operational parameters of the ARGO-YBJ RPCs are shown. This study was performed using the monitored data from the complete ARGO-YBJ array which has been in operation since October 2007. Preliminary results on the observations of the Moon shadow and of γ -ray point sources are also presented.

Abstract: The ARGO-YBJ detector at the YangBaJing Cosmic Ray Observatory (4300 m a.s.l., Tibet, P.R. China) has been put into operation in the full congration since November 2007. It is the rst EAS detector combining a very high mountain altitude with a full coverage detection surface. The high time-space granularity combined with the full coverage make ARGO-YBJ a unique device to study the EAS characteristics. In this paper we report a few selected results in Gamma-Ray Astronomy and Cosmic Ray Physics

Abstract: ARGO-YBJ is the first EAS detector combining a very high mountain altitude (4300 m a.s.l.) to a "full coverage" detection surface. These features allow ARGO-YBJ to work with an energy threshold as low as a few hundreds GeV. The high duty cycle and the large field of view (~2 sr) make ARGO-YBJ suitable to monitor the gamma ray sky searching for unknown sources and unexpected events, like Active Galactic Nuclei flaring episodes or high energy Gamma Ray Bursts. In this paper we present the first observations of ARGO-YBJ concerning gamma ray astronomy, in particular the detection of the Crab Nebula and the blazar Markarian 421 during the 2006 and 2008 outbursts, and the results of a search for Gamma Ray Bursts emission in the GeV energy range using the scaler mode technique.

Abstract: We report on the search for Gamma Ray Bursts (GRBs) in the energy range 1-100 GeV in coincidence with the prompt emission detected by satellites using the Astrophysical Radiation with Ground-based Observatory at YangBaJing (ARGO-YBJ) air shower detector. Thanks to its mountain location (Yangbajing, Tibet, P.R. China, 4300 m a.s.l.), active surface (about 6700 m**2 of Resistive Plate Chambers), and large field of view (about 2 sr, limited only by the atmospheric absorption), the ARGO-YBJ air shower detector is particularly suitable for the detection of unpredictable and short duration events such as GRBs. The search is carried out using the "single particle technique", i.e. counting all the particles hitting the detector without measurement of the energy and arrival direction of the primary gamma rays. Between 2004 December 17 and 2009 April 7, 81 GRBs detected by satellites occurred within the field of view of ARGO-YBJ (zenith angle < 45 deg). It was possible to examine 62 of these for >1 GeV counterpart in the ARGO-YBJ data finding no statistically significant emission. With a lack of detected spectra in this energy range fluence upper limits are profitable, especially when the redshift is known and the correction for the extragalactic absorption can be considered. The obtained fluence upper limits reach values as low as 10**{-5} erg cm**{-2} in the 1-100 GeV energy region. Besides this individual search for a higher energy counterpart, a statistical study of the stack of all the GRBs both in time and in phase was made, looking for a common feature in the GRB high energy emission. Abstract: Background Trials have shown a beneficial effect of n-3 polyunsaturated fatty acids in patients with a previous myocardial infarction or heart failure. We evaluated the potential benefit of such therapy in patients with multiple cardiovascular risk factors or atherosclerotic vascular disease who had not had a myocardial infarction. Methods In this double-blind, placebo-controlled clinical trial, we enrolled a cohort of patients who were followed by a network of 860 general practitioners in Italy. Eligible patients were men and women with multiple cardiovascular risk factors or atherosclerotic vascular disease but not myocardial infarction. Patients were randomly assigned to n-3 fatty acids (1 g daily) or placebo (olive oil). The initially specified primary end point was the cumulative rate of death, nonfatal myocardial infarction, and nonfatal stroke. At 1 year, after the event rate was found to be lower than anticipated, the primary end point was revised as time to death from cardiovascular causes or admission to the hospital for cardiovascular causes. Results Of the 12,513 patients enrolled, 6244 were randomly assigned to n-3 fatty acids and 6269 to placebo. With a median of 5 years of follow-up, the primary end point occurred in 1478 of 12,505 patients included in the analysis (11.8%), of whom 733 of 6239 (11.7%) had received n-3 fatty acids and 745 of 6266 (11.9%) had received placebo (adjusted hazard ratio with n-3 fatty acids, 0.97; 95% confidence interval, 0.88 to 1.08; P=0.58). The same null results were observed for all the secondary end points. Conclusions In a large general-practice cohort of patients with multiple cardiovascular risk factors, daily treatment with n-3 fatty acids did not reduce cardiovascular mortality and morbidity. Abstract: The Pierre Auger Observatory is a facility built to detect air showers produced by cosmic rays above 1017 eV. During clear nights with a low illuminated moon fraction, the UV fluorescence light produced by air showers is recorded by optical telescopes at the Observatory. To correct the observations for variations in atmospheric conditions, atmospheric monitoring is performed at regular intervals ranging from several minutes (for cloud identification) to several hours (for aerosol conditions) to several days (for vertical profiles of temperature, pressure, and humidity). In 2009, the monitoring program was upgraded to allow for additional targeted measurements of atmospheric conditions shortly after the detection of air showers of special interest, e. g., showers produced by very high-energy cosmic rays or showers with atypical longitudinal profiles. The former events are of particular importance for the determination of the energy scale of the Observatory, and the latter are characteristic of unusual air shower physics or exotic primary particle types. The purpose of targeted (or "rapid") monitoring is to improve the resolution of the atmospheric measurements for such events. In this paper, we report on the implementation of the rapid monitoring program and its current status. The rapid monitoring data have been analyzed and applied to the reconstruction of air showers of high interest, and indicate that the air fluorescence measurements affected by clouds and aerosols are effectively corrected using measurements from the regular atmospheric monitoring program. We find that the rapid monitoring program has potential for supporting dedicated physics analyses beyond the standard event reconstruction.

Abstract: Neuropsychological and activation studies on the neural correlates of abstract and concrete words have produced contrasting results. The present study explores the anatomical substrates of abstract/concrete words in 22 brain-damaged patients with a single vascular lesion either in the right or left hemisphere. One hundred and twenty (60 concrete and 60 abstract) noun triplets were used for a semantic similarity judgment task. We found a significant interaction in word type × group since left temporal brain-damaged patients performed significantly better with concrete than abstract words. Lesion mapping of patients with predominant temporal damage showed that the left superior and middle temporal gyri and the insula were the areas of major overlapping, while the anterior portion of the left temporal lobe was generally spared. Errors on abstract words mainly concerned (although at a non-significant level) semantically associate targets, while in the case of concrete words, coordinate targets were significantly more impaired than associate ones. Our results suggest that the left superior and middle temporal gyri and the insula are crucial regions in processing abstract words. They also confirm the hypothesis of a semantic similarity vs. associative organization of concrete and abstract concepts

Abstract: Neuropsychological and activation studies on the neural correlates of abstract and concrete words have produced contrasting results. The present study explores the anatomical substrates of abstract/concrete words in 22 brain-damaged patients with a single vascular lesion either in the right or left hemisphere. One hundred and twenty (60 concrete and 60 abstract) noun triplets were used for a semantic similarity judgment task. We found a significant interaction in word type × group since left temporal brain-damaged patients performed significantly better with concrete than abstract words. Lesion mapping of patients with predominant temporal damage showed that the left superior and middle temporal gyri and the insula were the areas of major overlapping, while the anterior portion of the left temporal lobe was generally spared. Errors on abstract words mainly concerned (although at a non-significant level) semantically associate targets, while in the case of concrete words, coordinate targets were significantly more impaired than associate ones. Our results suggest that the left superior and middle temporal gyri and the insula are crucial regions in processing abstract words. They also confirm the hypothesis of a semantic similarity vs. associative organization of concrete and abstract concepts.

Abstract: The essay "Antagonistic Fashion. The Last Generations after the Japanese Post-atomic Wave" deals with the latest radical trends and current inclinations labelled as anti-pop, anti-form, anti-glam, in a word, anti-fashion. The prefix “anti” is to be intended as a banner of contrast and opposition in respect to mainstream fashion canons. Drawing from a methodological frame rooted in the phenomenology of styles, from which she borrows a historical-critical structure usually applied to the arts, the author marks the trends of some designers’ latest generations (such as Carol Christian Poell, Maurizio Altieri, Issei Fujita, Boris Bidjan Saberi, Damir Doma) who are on the same wavelength as that of the research started by Yohji Yamamoto, Rei Kawakubo and Issey Miyake.

Abstract: TFEB, a bHLH-leucine zipper transcription factor belonging to the MiT/TFE family, globally modulates cell metabolism by regulating autophagy and lysosomal functions. Remarkably, loss of TFEB in mice causes embryonic lethality due to severe defects in placentation associated with aberrant vascularization and resulting hypoxia. However, the molecular mechanism underlying this phenotype has remained elusive. By integrating in vivo analyses with multi-omics approaches and functional assays, we have uncovered an unprecedented function for TFEB in promoting the formation of a functional syncytiotrophoblast in the placenta. Our findings demonstrate that constitutive loss of TFEB in knock-out mice is associated with defective formation of the syncytiotrophoblast layer. Indeed, using in vitro models of syncytialization, we demonstrated that TFEB translocates into the nucleus during syncytiotrophoblast formation and binds to the promoters of crucial placental genes, including genes encoding fusogenic proteins (Syncytin-1 and Syncytin-2) and enzymes involved in steroidogenic pathways, such as CYP19A1, the rate-limiting enzyme for the synthesis of 17β-Estradiol (E2). Conversely, TFEB depletion impairs both syncytial fusion and endocrine properties of syncytiotrophoblast, as demonstrated by a significant decrease in the secretion of placental hormones and E2 production. Notably, restoration of TFEB expression resets syncytiotrophoblast identity. Our findings identify that TFEB controls placental development and function by orchestrating both the transcriptional program underlying trophoblast fusion and the acquisition of endocrine function, which are crucial for the bioenergetic requirements of embryonic development., (© 2024. 46. Mitochondrial mechanotransduction through MIEF1 coordinates the nuclear response to forces.

Abstract: Tissue-scale architecture and mechanical properties instruct cell behaviour under physiological and diseased conditions, but our understanding of the underlying mechanisms remains fragmentary. Here we show that extracellular matrix stiffness, spatial confinements and applied forces, including stretching of mouse skin, regulate mitochondrial dynamics. Actomyosin tension promotes the phosphorylation of mitochondrial elongation factor 1 (MIEF1), limiting the recruitment of dynamin-related protein 1 (DRP1) at mitochondria, as well as peri-mitochondrial F-actin formation and mitochondrial fission. Strikingly, mitochondrial fission is also a general mechanotransduction mechanism. Indeed, we found that DRP1- and MIEF1/2-dependent fission is required and sufficient to regulate three transcription factors of broad relevance-YAP/TAZ, SREBP1/2 and NRF2-to control cell proliferation, lipogenesis, antioxidant metabolism, chemotherapy resistance and adipocyte differentiation in response to mechanical cues. This extends to the mouse liver, where DRP1 regulates hepatocyte proliferation and identity-hallmark YAP-dependent phenotypes. Abstract: Embryonic stem cells (ESCs) preserve the unique ability to differentiate into any somatic cell lineage while maintaining their self-renewal potential, relying on a complex interplay of extracellular signals regulating the expression/activity of pluripotency transcription factors and their targets. Leukemia inhibitory factor (LIF)-activated STAT3 drives ESCs' stemness by a number of mechanisms, including the transcriptional induction of pluripotency factors such as Klf4 and the maintenance of a stem-like epigenetic landscape. However, it is unknown if STAT3 directly controls stem-cell specific non-coding RNAs, crucial to balance pluripotency and differentiation. Applying a bioinformatic pipeline, here we identify Lncenc1 in mouse ESCs as an STAT3-dependent long non-coding RNA that supports pluripotency. Lncenc1 acts in the cytoplasm as a positive feedback regulator of the LIF-STAT3 axis by competing for the binding of microRNA-128 to the 3'UTR of the Klf4 core pluripotency factor mRNA, enhancing its expression. Abstract: Objective: Many people with epilepsy experience comorbid anxiety and depression, and antidepressants remain a primary treatment for this. Emerging evidence suggests that these agents may modulate epileptogenesis to influence disease severity. Here, we assessed how treatment with the selective serotonin reuptake inhibitor (SSRI) antidepressant fluoxetine impacts epileptogenic, behavioral, and pathological sequelae following status epilepticus., Methods: Male Wistar rats received kainic acid to induce status epilepticus (SE) or vehicle (sham). Animals then received either fluoxetine (10 mg/kg/day) or vehicle for 8 weeks via subcutaneous osmotic pump. Video-electroencephalography was recorded continuously until behavioral testing at day 56, including assessments of anxiety- and depression-like behavior and spatial cognition. Postmortem immunocytochemistry studies examined mossy fiber sprouting., Results: Fluoxetine treatment significantly accelerated epileptogenesis following SE, reducing the average period to the first spontaneous seizure (from 32 days [vehicle] to 6 days [fluoxetine], p < .01). Also, fluoxetine exposure magnified the severity of the resultant epilepsy, increasing seizure frequency compared to vehicle (p < .01). Exposure to fluoxetine was associated with improved anxiety- and depression-like behaviors but significantly worsened cognition. Mossy fiber sprouting was more pronounced in fluoxetine-treated rats compared to vehicle (p < .0001)., Significance: Our studies demonstrate that, using a model exhibiting spontaneous seizures, epileptogenesis is accelerated and magnified by fluoxetine, an effect that may be related to more severe pathological neuroplasticity. The differential influence of fluoxetine on behavior indicates that different circuitry and mechanisms are responsible for these comorbidities. These findings suggest that caution should be exercised when prescribing SSRI antidepressants to people at risk of developing epilepsy., (© 2024 The Author(s). Abstract: The high mutation rate of SARS-CoV-2 leads to the emergence of multiple variants, some of which are resistant to vaccines and drugs targeting viral elements. Targeting host dependency factors, e.g. cellular proteins required for viral replication, would help prevent the development of resistance. However, it remains unclear whether different SARS-CoV-2 variants induce conserved cellular responses and exploit the same core host factors. To this end, we compared three variants of concern and found that the host transcriptional response was conserved, differing only in kinetics and magnitude. Clustered regularly interspaced short palindromic repeats screening identified host genes required for each variant during infection. Most of the genes were shared by multiple variants. We validated our hits with small molecules and repurposed the US Food and Drug Administration-approved drugs. All the drugs were highly active against all the tested variants, including new variants that emerged during the study (Delta and Omicron). Mechanistically, we identified reactive oxygen species production as a key step in early viral replication. Antioxidants such as N-acetyl cysteine (NAC) were effective against all the variants in both human lung cells and a humanized mouse model. Abstract: Embryonic stem cells (ESCs) are defined as stem cells with self-renewing and differentiation capabilities. These unique properties are tightly regulated and controlled by complex genetic and molecular mechanisms, whose understanding is essential for both basic and translational research. A large number of studies have mostly focused on understanding the molecular mechanisms governing pluripotency and differentiation of ESCs, while the regulation of proliferation has received comparably less attention. Here, we investigate the role of ZZZ3 (zinc finger ZZ-type containing 3) in human ESCs homeostasis. We found that knockdown of ZZZ3 negatively impacts ribosome biogenesis, translation, and mTOR signaling, leading to a significant reduction in cell proliferation. This process occurs without affecting pluripotency, suggesting that ZZZ3-depleted ESCs enter a "dormant-like" state and that proliferation and pluripotency can be uncoupled also in human ESCs., Competing Interests: Declaration of interests The authors declare no competing interests., (Copyright © 2024 The Author(s).
