Abstract

The effect of the addition of Fe0 and Fe3+ on the formation of expanded clay aggregates was studied using iron-free kaolin as an aluminosilicates source. Likewise, the incorporation of cork powder as a source of organic carbon and Na2CO3 as a flux in the mixtures was investigated in order to assess its effect in combination with the iron phases. An experimental protocol, statistically supported by a mixture experiments/design of experiments approach, was applied to model and optimize the bloating index, density, absorption capacity, and mechanical strength. The process of expansion and pore generation and the associated decrease in density required the addition of iron, such that the optimum mixtures of these properties presented between 25 and 40 wt.% of Fe0 or Fe3+, as well as the incorporation of 3.5–5 wt.% of organic carbon. The addition of Fe3+ produced a greater volumetric expansion (max. 53%) than Fe0 (max. 8%), suggesting that the formation of the FeO leading to this phenomenon would require reducing and oxidizing conditions in the former and the latter, respectively. The experimental and model-estimated results are in good agreement, especially in the aggregates containing Fe0. This reinforces the application of statistical methods for future investigations. As the photoinitiator in CXL must diffuse into the cornea, a depth dependence on CXL efficacy is expected. We used custom-built Second Harmonic Generation (SHG) microscopy and quantifying routines to measure the order of collagen fibers throughout the cornea following CXL.

36. Dynamic changes of the extracellular matrix during corneal wound healing

Abstract

After the animals received both general and ocular topical anesthesia, the central cornea of the left eye was burned by placing an 8-mm diameter filter paper soaked in 0.5 N NaOH for 60 s. The eyes were evaluated under a surgical microscope at 1, 3, and 6 months after burning. At each time point, the clinical conditions of the burned and control corneas were observed. The arrangement of collagen fibers in the corneal stroma was visualized by Picrosirius-red staining, Gomori's silver impregnation and transmission electronic microscopy. Corneal light transmittance was also measured. Myofibroblasts presence was analyzed by immunohistochemistry. mRNA expression levels of collagen types I and III, lumican, decorin, keratocan and alpha-smooth muscle actin were determined by quantitative real-time polymerase chain reaction. One month after alkali burn, the ECM was disorganized and filled with lacunae containing different types of cells and collagen type III fibers in the wound area. Corneal opacities were present with attendant loss of light transmittance. Collagen and proteoglycan mRNA expression levels were up-regulated. After three months, wound healing progress was indicated by reduced corneal opacity, increased light transmittance, reorganization of collagen fibers and only collagen type I expression levels were at control levels. After six months, the wound area ECM morphology was similar to controls, but transmittance values remained low, denoting incomplete restoration of the stromal architecture. This multidisciplinary study of the stromal wound healing process revealed changes in corneal transmittance, collagen organization, myofibroblasts presence and ECM composition at 1, 3, and 6 months after alkali burning. Documenting wound resolution during the six-month period provided reliable information that can be used to test new therapies.

Abstract

Within a circular economy approach, this study investigates the environmental impact of lightweight aggregates (LWAs) produced starting from different mixes of different clays with brewery sludge and cattle bone flour ash (CBA), used as poring and fertilizing agents, respectively. The environmental impact was evaluated by means of release tests, insulation capacity, carbon footprint (CFP), and particulate matter emission during pellet firing. Release tests representative of LWAs realistic application showed very high release of phosphate and satisfactory release of potassium. The thermal insulation of the LWAs was tested by thermal imaging camera and resulted highly variable depending on the composition, with the mix containing CBA performing best. This latter composition leads also to the smallest CO2 equivalent emission, due to the calorific power of CBA, allowing lower consumption of fossil fuels during the LWA production. Finally, total particulate emissions during the thermal treatment resulted similar in terms of mass for all mixes, while differences in terms of particle morphology and composition occurred. Samples containing residue resulted with a quite good release behavior, CFP, and insulation properties, but higher emission of particles, particularly when glass is added. [ABSTRACT FROM AUTHOR]

Abstract

La esclerosis múltiple (EM) es una enfermedad neurodegenerativa autoinmune adquirida, caracterizada por un proceso crónico de desmielinización inflamatoria que afecta de forma específica al sistema nervioso central (SNC) [1-6]. Es mediada por el ataque del sistema inmune a través de células T, contra antígenos del SNC. Hay destrucción o daño de la vaina de mielina (desmielinización), pérdida de oligodendrocitos y neuronas lo que lleva al aumento de la disfuncionalidad neurológica. Con el objetivo de estudiar más a fondo los mecanismos de la EM y sus posibles tratamientos, se han desarrollado diversos modelos animales, fundamentalmente en ratones. La encefalitis autoinmune experimental (EAE) que se trata de un desorden autoinmune inflamatorio del sistema nervioso central cuyas características y evolución clínica se relacionan con lo descrito en los pacientes con EM [20]. La EAE puede ser inducida en algunas cepas de ratones por inmunización con péptidos que contienen secuencias concretas de derivados de la proteína de mielina básica (MBP), glicoproteína de la mielina de oligodendrocitos (MOG) y la proteína proteolipídica de la mielina (PLP) [21, 22]. De los diferentes tipos de inducción de EAE, los más comunes se basan en la inyección de un péptido, MOG35-55, que es considerado el mejor antígeno causante de EAE, crónica y no remitente, ya que aproximadamente el 50% de los pacientes son positivos para el anticuerpo anti-MOG [23]. En la EAE al igual que en la EM, la edad, el género, los factores ambientales y cambios hormonales también influyen en la susceptibilidad, desarrollo y severidad de la enfermedad.

42. Detection by HRM and COLD-PCR-HRM BRAF mutational status in paraffin blocks of melanoma patients

Abstract

Resumen del póster presentado a la European Human Genetics Conference celebrada en Nuremberg (Alemania) del 23 al 26 de junio de 2012., Over 50% of melanoma tumors have BRAF oncogene mutation. In 2011 we have obtained the first results of clinical trials of targeted therapy with molecules that act on BRAF signaling pathways (Ras/Raf/MEK/ERK). Vemurafenib and GSK2118436 showed an increase in overall survival and disease-free in p.V600E BRAF carriers patients. In the coming months, these molecules will be approved for clinical use in Europe. The application of analytical methods of high sensitivity in determining the mutational status of BRAF will be crucial to the imminent clinical application of new drugs, as the results will select patients eligible for or untreated. We have studied the 11 and 15 exons of BRAF by HRM.sequencing, and COLD-PCR-HRM-sequencing in 20 paraffin blocks of melanoma, previously studied by Sanger sequencing. Mutation was detected p.V600E of exon 15 in 9 of them for HRM, and 11 by COLD-HRM, compared to 8 previously detected by sequencing. The sensitivity of such methods as HRM and COLD-PCR-HRM is greater than sequencing. These results should be compared with those obtained by the propose clinical diagnostic tests (eg cobas 4800 BRAF V600 Mutation Test Roche©). Our previous results show the necessary standardization of the methods of detection of somatic mutations, especially in cases where the determined result to apply the therapy to the patient.

45. Oleanolic acid controls allergic and inflammatory responses in experimental allergic conjunctivitis

Abstract

Pollen is the most common aeroallergen to cause seasonal conjunctivitis. The result of allergen exposure is a strong Th2-mediated response along with conjunctival mast cell degranulation and eosinophilic infiltration. Oleanolic acid (OA) is natural a triterpene that displays strong anti-inflammatory and immunomodulatory properties being an active anti-allergic molecule on hypersensitivity reaction models. However, its effect on inflammatory ocular disorders including conjunctivits, has not yet been addressed. Hence, using a Ragweed pollen (RWP)-specific allergic conjunctivitis (EAC) mouse model we study here whether OA could modify responses associated to allergic processes. We found that OA treatment restricted mast cell degranulation and infiltration of eosinophils in conjunctival tissue and decreased allergen-specific Igs levels in EAC mice. Th2-type cytokines, secreted phospholipase A2 type-IIA (sPLA2-IIA), and chemokines levels were also significantly diminished in the conjunctiva and serum of OA-treated EAC mice. Moreover, OA treatment also suppressed RWP-specific T-cell proliferation. In vitro studies, on relevant cells of the allergic process, revealed that OA reduced the proliferative and migratory response, as well as the synthesis of proinflammatory mediators on EoL-1 eosinophils and RBL-2H3 mast cells exposed to allergic and/or crucial inflammatory stimuli such as RWP, sPLA2-IIA or eotaxin. Taken together, these findings demonstrate the beneficial activity of OA in ocular allergic processes and may provide a new intervention strategy and potential therapy for allergic diseases. © 2014 Córdova et al.

Abstract

BACKGROUND: Non-Hodgkin's lymphomas are a heterogeneous group of neoplasms arising from the lymphopoietic system including a wide range of subtypes of either B-cell or T-cell lymphomas. The few established risk factors for the development of these neoplasms include viral infections and immunological abnormalities, but their etiology remains largely unknown. Evidence suggests that certain medical conditions may be linked, through immunosuppression, to the risk of non-Hodgkin's lymphoma. Multiple myeloma is a neoplasm of plasma cells that accounts for approximately 15% of lymphopoietic cancers. Increases in the incidence of non-Hodgkin's lymphoma and multiple myeloma in the past implicate environmental factors as potential causal agents. DESIGN AND METHODS: In the European Prospective Investigation into Cancer and Nutrition (EPIC), 1,213 histologically confirmed incident cases of non-Hodgkin's lymphoma and multiple myeloma (594 men; 619 women) were identified during a follow-up of 8.5 years. Cox proportional hazard models were used to explore the association between self-reported diabetes, diagnosed after 30 years of age, and the risk of non-Hodgkin's lymphoma overall and multiple myeloma and various lymphoma subtypes. RESULTS: We found no association between a personal history of diabetes and the risk of non-Hodgkin's lymphoma overall in men (HR: 1.28, 95% CI: 0.89-1.84), in women (HR: 0.71, 95% CI: 0.41- 1.24), or in men and women combined (HR: 1.09, 95% CI: 0.80-1.47). Among the B-non-Hodgkin's lymphoma subtypes, we observed a statistically significant increased risk of B-cell chronic lymphocytic leukemia (HR: 2.0, 95% CI: 1.04-3.86) in men, but not in women (HR: 1.07, 95% CI: 0.33-3.43). CONCLUSIONS: This prospective study did not provide evidence for a role of self-reported diabetes in the etiology of non-Hodgkin's lymphoma overall or multiple myeloma. We found an increased risk of B-cell chronic lymphocytic leukemia among men with diabetes, but not among women. We hypothesize that diabetes may not play a causal role in the etiology of B-cell chronic lymphocytic leukemia, though the underlying pathogenic mechanisms of both disorders may include shared genetic, host and/or environmental susceptibility factors.

Abstract

Fundamento: En el caso de las enfermedades raras existen dificultades para utilizar herramientas como los registros de enfermedades. El objetivo de este trabajo es describir la situación de los registros sobre enfermedades raras en España. Métodos: Se utiliza la información proporcionada por dos directorios de registros sanitarios españoles elaborados por la Agencia Española de Evaluación de Tecnologías Sanitarias, realizando un estudio descriptivo transversal. Los registros identificados en estos directorios se han clasificado como: 1) «Específicos de enfermedades raras», 2) «No específicos pero con información de enfermedades raras», y 3) «No informativos sobre enfermedades raras». Resultados: En el directorio del año 2000 aparecían 82 registros, de los que el 15,8% se clasificaron en el grupo 1, mientras que en la revisión de 2005 se identificaron 107 registros, 16,8% en el grupo 1. Las principales áreas temáticas de los registros sanitarios del grupo 2 fueron el cáncer, la mortalidad, la psiquiatría y la nefrología. En los directorios no se ha hallado ningún registro genérico de enfermedades raras. Conclusiones: Aunque escasos, existen registros sanitarios en España con información sobre enfermedades raras. Se han identificado áreas temáticas con carencias de registros y también registros o sistemas de información no identificados en las fuentes utilizadas. Es necesario continuar los esfuerzos por mejorar la información disponible sobre las enfermedades raras. Background: The use of tools such as disease registries poses a problem in the case of rare diseases. This study is aimed at describing the current situation concerning rare disease registries in Spain. Methods: The information provided by two Spanish health registries directories prepared by the Spanish Health Assessment Technologies Agencies are employed, a descriptive cross-sectional study being conducted. The registries identified in these directories has been classified as: 1) «Specific rare diseases» 2) «Unspecific but with information on rare disease» and 3) «Non-informative regarding rare diseases». Results: The 2000 directory listed 82 registries, 15.8% of which were classified under Group 1, whilst a total of 107 registries, 16.8% in Group 1, were identified in the 2005 review. The main health registries in Group 2, by topic, were: cancer, mortality, psychiatry and nephrology. No general rare disease registries were found in the directories. Conclusions: Although few in number, health registries do exist in Spain including information on rare diseases. Areas have been identified by topic lacking registries and also information systems or registries unidentified in the sources used. Continuing efforts must be made to improve the information available on rare diseases.

Abstract

This study investigates the effects of sieved wastes generated from the brewing industry on lightweight aggregates manufactured with clay. Sludge from a wastewater treatment plant, bagasse and diatomaceous earth were used to obtain the samples. These wastes are usually dumped in landfills, but the current increase in restrictions on dumping and interest in improving the environment make our proposal for gaining value from these wastes a significant contribution. Laboratory tests show that the new aggregate has low bulk density and increased water absorption and porosity. The thermographic camera results provide evidence that new aggregates have significant insulating properties and are suitable for use on green roofs. [ABSTRACT FROM AUTHOR]
